B501 promoted the production of artemisinin in Artemisia annua L. hairy root culture. When hairy roots of 22-day-old cultures (later growth phase) were exposed to the elicitor (20 mg/L) for 4 d, the maximum content of artemisinin reached 1.15 mg/g, a 64.29% increment over the control. The electron X-ray microanalysis disclosed the rapid accumulation of Ca 2+ in the elicited cortical cells of hairy root. The electronic microscope observation revealed the high electron density area in vacuole of elicited cells. During the first day of elicitation the peroxidase activity of hairy roots was improved sharply. Some cellular morphological changes including cell shrinkage, condensation of cytoplasm and nuclear fragmentation, coincident with the appearance of DNA ladders, were observed after the third day of elicitation. It was suggested that the oligosaccharide elicitor triggered the programmed cell death, which may provide the substance or chemical signal for artemisinin biosynthesis. 展开更多

Requirements elicitation is a fundamental phase of software development in which an analyst discovers the needs of different stakeholders and transforms them into requirements.This phase is cost-and time-intensive,and a project may fail if there are excessive costs and schedule overruns.COVID-19 has affected the software industry by reducing interactions between developers and customers.Such a lack of interaction is a key reason for the failure of software projects.Projects can also fail when customers do not know precisely what they want.Furthermore,selecting the unsuitable elicitation technique can also cause project failure.The present study,therefore,aimed to identify which requirements elicitation technique is the most cost-effective for large-scale projects when time to market is a critical issue or when the customer is not available.To that end,we conducted a systematic literature review on requirements elicitation techniques.Most primary studies identified introspection as the best technique,followed by survey and brainstorming.This finding suggests that introspection should be the first choice of elicitation technique,especially when the customer is not available or the project has strict time and cost constraints.Moreover,introspection should also be used as the starting point in the elicitation process of a large-scale project,and all known requirements should be elicited using this technique. 展开更多

The main factor 4 biotic elicitors: control (E<sub>1</sub>), phenylalanine (E<sub>2</sub>), yeast (E<sub>3</sub>) and (E4), E2 + F3 Whereas, the sub-main factor, four fertilizers: (F1) NPK, (F2) 1/2NPK + PGPB and humic acid + PGPB (F<sub>3</sub>), and moringa dry leaves extract. + PGPB (F<sub>4</sub>). Statistical analysis for collected data revealed significant promotion for growth traits leading to significant increment biomass yield, secondary metabolites production and quality. Total phenolics, total flavonoids and essential oil its terpenes contents in which solitary, E2, E3 acted positive significant impact while E4exhiboted significant positive impact over E1 whereas, F1-4 achieved significant increment in which, biofertilser f4 > f3 > f2 over NPK biofertilser, while paired E1-4 with F1-4 performed E4E4 exceeded E3F3 exceeded E3F2 that exceeded E1F1. Therefore, multi-repeating elicitation with E2, 3, 4 coupled with Biofertilizers F2, 4 could be considered as eco-friendly innovative reliable practical application for sustainable improvement and sustainable use that exceeded significantly over traditional agriculture NPK alone or 1/2 NPK-PGPB for <em>R. officinals</em>. 展开更多

Rare earth elements(REEs)have been widely applied as efficient elicitors to improve production of biologically important secondary metabolites in plants.However,their effects on microbial secondary metabolites were seldom reported.In this study,lanthanum(LaCl3)was used as an elicitor in mycelium cultures of Shiraia bambusicola to stimulate the production of hypocrellin A(HA),a promising photosensitizer for anticancer photodynamic therapy.La^3+at 1.0 g/L not only increases HA content by 2.50-fold in mycelia,but stimulates HA exudation to the medium with the highest production(225.05 mg/L)on day 6.Further analysis shows that La^3+can induce the intracellular ROS(reactive oxygen species)accumulation with 2.33-fold higher contents of H2O2 in mycelium.HA production induced by La^3+treatment can be suppressed by ROS scavengers.Further analysis reveals that the elicitation of La^3+on HA accumulation is mediated by ROS generation to enhance expression of HA biosynthetical genes.The expression of ATP-binding cassette transporter gene(SbABC)for HA exudation is also regulated partially via ROS generated by NADPH oxidase.These results indicate that the enhanced HA production by La^3+might be mediated by ROS signaling pathway.Our results provide a basis for understanding REE elicitation on fungal metabolites and a practical biotechnological process for enhanced production of HA. 展开更多

In decision modeling with influence diagrams, the most challenging task is probability elicitation from domain experts. It is usually very difficult for experts to directly assign precise probabilities to chance nodes. In this paper, we propose an approach to elicit probability effectively by using the concept of interval probability (IP). During the elicitation process, a group of experts assign intervals to probabilities instead of assigning exact values. Then the intervals are combined and converted into the point valued probabilities. The detailed steps of the elicitation process are given and illustrated by constructing the influence diagram for employee recruitment decision for a China’s IT Company. The proposed approach provides a convenient and comfortable way for experts to assess probabilities. It is useful in influence diagrams modeling as well as in other subjective probability elicitation situations. 展开更多

Local Shear Stresses Elicit Different Mechanical Response and Gene Expression from Complex Stresses

It is well established that living cells and tissues respond to mechanical forces such as flow-related shear stresses in blood or interstitial space and complex tractional stresses at cell-matrix contacts and cell-cell contacts.However,how different modes of forces impact mechanical and biological responses is elusive.Here we describe a strategy of using the three-dimensional magnetic twisting cytometry(3D MTC)technology to apply forces in any desired directions to the same living cell.We reveal that for a fixed stress amplitude and frequency,a live cell exhibits mechanical anisotropy and responds to a local shear stress differently from responding to a local complex stress by stretching chromatin and upregulating gene transcription to different levels,extending our previous finding on force-induced direct gene activation.This finding highlights the importance of force modes in impacting cellular mechanical and biological responses in living cells and tissues and may have implications in tissue patterning and embryonic development. 展开更多

Most software systems have different stakeholders with a variety of concerns.The process of collecting requirements from a large number of stakeholders is vital but challenging.We propose an efficient,automatic approach to collecting requirements from different stakeholders’responses to a specific question.We use natural language processing techniques to get the stakeholder response that represents most other stakeholders’responses.This study improves existing practices in three ways:Firstly,it reduces the human effort needed to collect the requirements;secondly,it reduces the time required to carry out this task with a large number of stakeholders;thirdly,it underlines the importance of using of data mining techniques in various software engineering steps.Our approach uses tokenization,stop word removal,and word lemmatization to create a list of frequently accruing words.It then creates a similarity matrix to calculate the score value for each response and selects the answer with the highest score.Our experiments show that using this approach significantly reduces the time and effort needed to collect requirements and does so with a sufficient degree of accuracy. 展开更多

In order to develop quality software that meets the originals needs of its users, it is necessary to perform the Requirements Engineering, so that the software context to be developed is identified, examined and specified properly. However, there is a problem that is increasingly in debate: the difficulty in understanding and establishing the purpose of the software to be developed, as pointed out by important researches in the area, such as the Chaos Report, which indicates that only 29% of software projects are successful, and the Software Engineering Institute, which points out software requirements as a critical factor for the success of software engineering and that deficiencies in this dimension are the main causes of software project failures. This article presents a proposal to address this problem through the use of the Hermeneutical Elicitation of Requirements, which is the conceptual adequacy of some hermeneutical methods in a technical approach that assists the requirements engineer to conceive better of the software requirements. In this way, the software engineer will be better able to develop the software to better meet the needs of its end users and sponsors. 展开更多

In this study, exogenous pre-harvest treatment of tomato plants with MeJA (Methyl Jasmonate) and chitosan was used to elicit lycopene biosynthesis and stimulate plant growth. Lycopene is a bright red carotene and carotenoid pigment found in tomatoes, carrots and other red fruits and vegetables. It is a powerful antioxidant that protects cells from damage and blocks cancerous cell growth. Many studies suggest eating lycopene-rich foods may be directly linked to reducing risk of several cancers, heart disease and stroke, hence there is a need to enhance its levels in tomatoes. Tomato seeds were soaked in MeJA and chitosan solutions of varying concentrations and then planted. The resulting plants were irrigated with the solutions used to soak the seeds, accordingly. The heights of the plants were recorded on a tri-weekly basis and HPLC (High Performance Liquid Chromatography) was used to analyze the effects of the MeJA and chitosan solutions on lycopene yields in the tomato fruits. Analysis of chromatograms obtained from tomato samples in the MeJA and chitosan-treated groups showed enhanced levels of lycopene. Plant growth was not significantly affected by treatment ofMeJA. Chitosan, however, noticeably increased plant growth over 7 weeks. 展开更多

Biosynthesis of peanut stilbenes, including resveratrol as the secondary metabolites, could be enhanced by subjecting the kernels to germination and wound-stress. Investigations of the bio-elicited peanut sprout powder (BPSP) addressed on characterization of the comprising stilbenes and effectiveness in longevity extension deserves intensive research. In this study, peanut kernels were subjected to germination and wound-stress in preparation of BPSP. The methanol extracts of BPSP were medium pressure liquid chromatographic (MPLC) fractionated and semi-preparative HPLC recovered and followed by instrumental identification and biological activity determinations of the isolated stilbenes. In longevity experiments, 16 female 11-mon-old BALB/c mice and both genders of 12-mon-old ICR mice were daily fed with BPSP supplemented diets at doses of 0, 0.1 and 0.5 g BPSP/kg bw for 750 and 762 days, respectively. Based on chemical characterization, enriched quantity of stilbenes in the BPSP up to ca. 1% (w/w) was detected. Two new stilbene compounds, namely, 4, 5’-dihydroxy-6’’-hydroxymethyl, 6’’-methylpyrano [2’’, 3’’: 3’, 4’] stilbene and 3, 4, 5’-trihydroxy-6’’, 6’’-dimethylpyrano [2’’, 3’’: 3’, 4’]stilbene along with 5 known stilbenes were isolated. The 7 stilbenes exhibited potent antioxidative and antiglycative activities and varied with structure-activity nature. Based on the resultant survival curves and average lifespans of both mouse models, basal diets supplemented with BPSP are effective to extend mouse longevity by a dose dependent manner. It is of merit to demonstrate that peanut kernels as a potent producer could be bio-elicited to biosynthesize a broad spectrum of bioactive stilbenes to prepare BPSP which is effective to extend mouse longevity as science-evidenced by the two long-term animal experiments. 展开更多

A challenge for many School-Based Family Counseling(SBFC)practitioners,child psychotherapists and researchers are finding ways to give voice to children and eliciting trustworthy and detailed narratives that could serve as resource for understanding the needs of young clients in the context of all their interpersonal networks.Children are often reticent when asked to self-disclose and tell their stories during consultation.The purpose of the present study was to examine the utility of five sequential steps constituting the Collage Life Story Elicitation Technique(CLET)for scaffolding storytelling among children in middle childhood(aged 9-12 years).Using the CLET for data collection and conducting an interpretive analysis,the researchers explored the performance of 38 middle-childhood children living in three different settings.Findings suggest that the five sequential steps of the CLET adequately and satisfactorily combine to stimulate and elicit rich data and help children to construct their narratives and represent the challenges they face in everyday living.We discuss the application of CLET in SBFC practice as tool when screening and intervention planning for children’s perspectives pertaining to a range of topics regarding each of the four quadrants as proposed in the SBFC metamodel. 展开更多

With a growing number of foreign language studies on proficiency outcomes,it is imperative to address the challenge of measuring students’proficiency development in a language program where standardized proficiency testing is not readily available.This article reports administering a Chinese elicited imitation test(EIT)by an instructor to track students’global oral proficiency development in a small language program in a mid-size U.S.public university.The test results from the EIT of second language(L2)Chinese suggest that this tool can provide the instructor with valuable insights into students’oral proficiency.This study also discusses the potential practical value of using this EIT in a language program with limited resources for standardized proficiency assessment.The hope is that this study will encourage language educators who are not already doing so to start using empirical evidence from a valid and reliable proficiency measurement tool to reflect on,improve,and guide their instructional practices. 展开更多

A challenge for cancer vaccines is to elicit immune responses of sufficient magnitude to control malignant tumor growth and spread.In this study,we iteratively screened a panel of 22 lipid-phase vaccine adjuvants in mice for the elicitation of neoantigen-specific CD8^(+)T cell responses,using an integrated peptide-lipid nanoparticle approach.CL401,a dual Toll-like receptor 2/7(TLR2/7)adjuvant rapidly induced neoantigen-specific T cell responses and improved lymphatic drainage and uptake of the particle.Additional rounds of in vivo screening identified complementary adjuvants which targeted TLR4(3D6A-PHAD adjuvant),TLR8(motolimod),and inflammasome(QS-21)pathways and synergized to enhance cytokine secretion in antigen presenting cells and vaccine-elicited neoantigen-specific CD8^(+)T cells.Co-delivery of adjuvants and antigens led to effective immune responses which regressed large established tumors,synergized with immune checkpoint blockade,and inhibited lung nodules in an experimental metastasis model,without overt toxicity or reactogenicity.We conclude that iterative adjuvant screening,performed in mice in vivo,can identify useful adjuvant combinations that hold potential for therapeutic cancer vaccine research. 展开更多

近10年来,情绪加工中大脑与心脏的复杂生理交互成为研究热点。本研究采用网络生理学方法,分析Dreamer数据库中414条视觉情绪诱发期间的脑电和心电信号间的时间延迟稳定性(TDS)量化指标,探究视觉情绪刺激下的脑心交互。研究揭示了情绪处... 近10年来,情绪加工中大脑与心脏的复杂生理交互成为研究热点。本研究采用网络生理学方法,分析Dreamer数据库中414条视觉情绪诱发期间的脑电和心电信号间的时间延迟稳定性(TDS)量化指标,探究视觉情绪刺激下的脑心交互。研究揭示了情绪处理时大脑半球间的不对称连接,特别是大脑右半球在交互中的主导地位。脑电分析强调了低频段(δ、θ、α)在情绪信息传输中的核心作用,其中额叶区域的δ-θ耦合对情绪调节尤为关键。在高效价情绪状态下δ-θ耦合的%TDS值(0.78±0.05)显著高于低效价状态(0.65±0.04)。此外,低效价脑心交互的平均连接强度最高达(0.68±0.06),而高效价下降至最低(0.59±0.03)。这些发现不仅增进了对情绪加工中大脑皮层与心脏同步机制的理解,而且丰富了神经生理学与情绪科学的知识体系。 展开更多
