Laser elktacytometry is a technique widely used for measuring the deformability of red blood cells(erythrocytes)in blood samples in vitro.In ektacytometer,a flow of highly diluted suspension of erythrocytes in variabl...Laser elktacytometry is a technique widely used for measuring the deformability of red blood cells(erythrocytes)in blood samples in vitro.In ektacytometer,a flow of highly diluted suspension of erythrocytes in variable shear stress conditions is iluninated with a laser beam to obtain a diffraction pattern.The diffraction pattern provides information about the shapes(shear induced elongations)of the cells under investigation.This paper is dedicated to developing the technique of laser ektacytometry s0 that it would enable one to measure the distrilbution of the erythrocytes in deformability.We discuss the problem of calibration of laser elktacytometer and test a novel data processing algorithm allowing to determine the parameters of the distribution of ery-throcytes deformability.Experimentally,we examined 12 specimens of blood of rats under the action of 4 shear stresses.Analysis of the data shows that in conditions of a limited range of digitizing the diffraction patterns,the measurement errors for the mean deformability,deform-ability scatter and the skewness of erythrocytes distribution in deformability by our method are respectively 15%,20%and 20%.展开更多

This damage ranges from slight morphological alteration to the destruction of red b...It has been well documented that shear forces effective above a certain magnitude under flow conditions causes damage in blood cells. This damage ranges from slight morphological alteration to the destruction of red blood cells (RBC). The hemolytic trauma to RBC can easily be detected by measuring free hemoglobin level in plasma, however there are no standardized protocols to quantitate subhemolytic trauma. Ektacytometry has been used to study the alterations in RBC mechanical properties induced by the application of shear stress at various levels. Additionally, a protocol for measuring the hemolytic threshold as an indicator of subhemolytic damage to RBC has been developed based on ektacytometry. These standardized protocols may find applications in the pre-clinical and clinical evaluation of artificial organs and biomedical devices contacting with blood.展开更多

The measurements of deformation index, orientation index, electrophoresis mobility etc. were performed for more than 72 h in the process of reticulocytes turning into red blood cells in vivo. There were obvious changes in the micro- rheological characteristics of reticulocytes in the course of turning into erythrocytes. The present study is significant in clinic for studying erythrocytes' microrheological characteristics when there are a lot of reticulocytes in blood, and also important in basic theorem for studying reticulocytes microrheological characteristics. It makes up a deficiency in the study on microrheological characteristics of reticulocytes turning into new RBCs from reticulocytes during reticulocytes life span.展开更多
