In this study,we constructed dual-transgene vectors(pDT1,pDT7,and pDT7G) that simultaneously co-expressed two genes in plants.ACTIN2 and UBQ10 promoters were used to control the expression of these two genes.The 4...In this study,we constructed dual-transgene vectors(pDT1,pDT7,and pDT7G) that simultaneously co-expressed two genes in plants.ACTIN2 and UBQ10 promoters were used to control the expression of these two genes.The 4×Myc.3×HA,and 3×Flag reporter genes allowed for the convenient identification of a tunable co-expression system in plants,whereas the dexamethasone(Dex) inducible reporter gene C-terminus of the glucocorticoid receptor(cGR) provided Dex-dependent translocation of the fusion gene between the nucleus and cytoplasm.The function of pDT vectors was validated using four pairwise genes in Nicotiana benthamiana or Anihidopsis thaliana.The co-expression efficiency of two genes from the pDT1 and pDT7 G vectors was 35%and 42%,respectively,which ensured the generation of sufficient transgenic materials.These pDT vectors are simple,reliable,efficient,and time-saving tools for the co-expression of two genes through a single transformation event and can be used in the study of protein-protein interactions or multi-component complexes.展开更多

The...This study aimed to construct the dual-gene expression vector p Hsa-miR16-siRNA which can express human miR-16 and HBV X siRNA, and examine its regulatory effect on HBV gene expression in the HepG2.2.15 cell line. The expression vectors siR-1583 and pHsa-miR16-siRNA were designed and constructed. Hep G2.2.15 cells were transfected with the empty vector, siR-1583, pmiR-16 and pHsa-miR16-siRNA, respectively. ELISA was performed to measure the expression of HBsAg and HBeAg in the culture supernatant 48 and72 h post transfection. Fluorescence quantitative PCR was used to measure the HBV mRNA degradation efficiency and HBV DNA copy number. The results showed that the expression of HBV genes was significantly inhibited in Hep G2.2.15 cells transfected with siR-1583, pmiR-16 and pHsa-miR16-siRNA, respectively, when compared with that in cells transfected with the empty vectors, with the inhibitory effect of pHsa-miR16-siRNA being the most significant. ELISA showed that the inhibitory rates of HBs Ag and HBeAg in pHsa-miR16-siRNA transfected cells were correspondingly 87.3% and 85.0% at 48 h, and 88.6% and 86.5% at 72 h post transfection(P〈0.01 vs. control group). RT-PCR showed that the level of HBV mRNA decreased by 80.2%(t=–99.22, P〈0.01), the genomic HBV DNA by 92.8%(t=–73.06, P〈0.01), and the supernatant of HBV DNA copy number by 89.8%(t=–47.13, P〈0.01) in pHsa-miR16-siRNA transfected group. It was suggested that the dual-gene expression vector pHsa-miR16-siRNA can inhibit the replication of HBV more efficiently than a single-gene expression vector.展开更多
In this paper, responses of a new dual-induction resistivity logging-while-drilling (LWD) tool in 3D inhomogeneous formation models are simulated by the vectorfinite element method (VFEM), the influences of the bo...In this paper, responses of a new dual-induction resistivity logging-while-drilling (LWD) tool in 3D inhomogeneous formation models are simulated by the vectorfinite element method (VFEM), the influences of the borehole, invaded zone, surroundingstrata, and tool eccentricity are analyzed, and calibration loop parameters and calibrationcoefficients of the LWD tool are discussed. The results show that the tool has a greater depthof investigation than that of the existing electromagnetic propagation LWD tools and is moresensitive to azimuthal conductivity. Both deep and medium induction responses have linearrelationships with the formation conductivity, considering optimal calibration loop parametersand calibration coefficients. Due to the different depths of investigation and resolution, deepinduction and medium induction are affected differently by the formation model parameters,thereby having different correction factors. The simulation results can provide theoreticalreferences for the research and interpretation of the dual-induction resistivity LWD tools.展开更多
The wind retrieval performance of HY-2 A scanning scatterometer operating at Ku-band in HH and VV polarizations has been well evaluated in the wind speed range of 0–25 m s^-1.In order to obtain more accurate ocean wi...The wind retrieval performance of HY-2 A scanning scatterometer operating at Ku-band in HH and VV polarizations has been well evaluated in the wind speed range of 0–25 m s^-1.In order to obtain more accurate ocean wind field,a potential extension of dual-frequency(C-band and Ku-band)polarimetric measurements is investigated for both low and very high wind speeds,from 5 to 45 m s^-1.Based on the geophysical model functions of C-band and Ku-band,the simulation results show that the polarimetric measurements of Ku-band can improve the wind vector retrieval over the entire scatterometer swath,especially in nadir area,with the wind direction root-mean-square error(RMSE)less than 12?in the wind speed range of 5–25 m s^-1.Furthermore,the results also show that C-band cross-polarization plays a very important role in improving the wind speed retrieval,with the wind speed retrieval accuracy better than 2 m s^-1 for all wind conditions(0–45 m s^-1).For extreme winds,the C-band HH backscatter coefficients modeled by CMOD5.N(H)and the ocean co-polarization ratio model at large incidence are used to retrieve sea surface wind vector.This result reveals that there is a big decrease of wind direction retrieval RMSE for extreme wind fields,and the retrieved result of C-band HH polarization is nearly the same as that of C-band VV polarization for low-to-high wind speed(5–25 m s^-1).Thus,to improve the wind retrieval for all wind conditions,the dual-frequency polarimetric scatterometer with C-band and Ku-band horizontal polarization in inner beam,and C-band horizontal and Ku-band vertical polarization in outer beam,can be used to measure ocean winds.This study will contribute to the wind retrieval with merged satellites data and the future spaceborne scatterometer.展开更多

The method modulates vector signal on +3 rd...To generate high-frequency radio frequency(RF) vector signals, a vector signal generation method by optical frequency sextupling using a dual-parallel modulator is proposed. The method modulates vector signal on +3 rd order optical sideband and local oscillator(LO) signal on-3 rd order sideband using the intermodulation process in the DPMZM. After suppressing of the optical carrier and other sidebands through proper adjustment for modulator biases and modulation index, a frequency sextupled millimeter-wave vector signal can be generated after photodetection. The frequency sextupling will lower the bandwidth of the modulator, the local oscillator and the driving circuits. In addition, the phase of generated signal is not distorted after detection, and the power fading after fiber transmission can be avoided. In the simulation, a 500-MSym/s QPSK signal at 60 GHz is generated by 10-GHz drive signal. After travelling over fiber with length of 20/30/40-km, receiver power penalty keeps below 2.5 dB.展开更多
