Studies of wave-current interactions are vital for the safe design of structures.Regular waves in the presence of uniform,linear shear,and quadratic shear currents are explored by the High-Level Green-Naghdi model in ...Studies of wave-current interactions are vital for the safe design of structures.Regular waves in the presence of uniform,linear shear,and quadratic shear currents are explored by the High-Level Green-Naghdi model in this paper.The five-point central difference method is used for spatial discretization,and the fourth-order Adams predictor-corrector scheme is employed for marching in time.The domain-decomposition method is applied for the wave-current generation and absorption.The effects of currents on the wave profile and velocity field are examined under two conditions:the same velocity of currents at the still-water level and the constant flow volume of currents.Wave profiles and velocity fields demonstrate substantial differences in three types of currents owing to the diverse vertical distribution of current velocity and vorticity.Then,loads on small-scale vertical cylinders subjected to regular waves and three types of background currents with the same flow volume are investigated.The maximum load intensity and load fluctuation amplitude in uniform,linear shear,and quadratic shear currents increase sequentially.The stretched superposition method overestimates the maximum load intensity and load fluctuation amplitude in opposing currents and underestimates these values in following currents.The stretched superposition method obtains a poor approximation for strong nonlinear waves,particularly in the case of the opposing quadratic shear current.展开更多

Some bacterial and fungal microorganisms as a direct-fed microbial(DFM) can be the most suitable solutions. Microorganisms that are commonly used in DFM for ruminants may be classified mainly as lactic acid producing bacteria(LAB), lactic acid utilizing bacteria(LUB), or other microorganism's species like Lactobacillus, Bifidobacterium, Enterococcus, Streptococcus, Bacillus, Propionibacterium, Megasphaera elsdenii and Prevotellabryantii, in addition to some fungal species of yeast such as Saccharomyces and Aspergillus. A definitive mode of action for bacterial or fungal DFM has not been established; although a variety of mechanisms have been suggested. Bacterial DFM potentially moderate rumen conditions, and improve weight gain and feed efficiency. Fungal DFM may reduce harmful oxygen from the rumen, prevent excess lactate production, increase feed digestibility, and alter rumen fermentation patterns. DFM may also compete with and inhibit the growth of pathogens, immune system modulation, and modulate microbial balance in the gastrointestinal tract. Improved dry matter intake, milk yield, fat corrected milk yield and milk fat content were obtained with DFM administration. However, the response to DFM is not constant; depending on dosages, feeding times and frequencies, and strains of DFM. Direct-fed microbials(DFM) are considered as a promising technique to improve animal productivity without affecting animal health or harming the environment.The potential of three bacterial DFM to reduce methane(CH4)e...Direct-fed microbials(DFM) are considered as a promising technique to improve animal productivity without affecting animal health or harming the environment.The potential of three bacterial DFM to reduce methane(CH4)emissions,modulate ruminal fermentation,milk production and composition of primiparous dairy cows was examined in this study.As previous reports have shown that DFM respond differently to different diets,two contrasting diets were used in this study.Eight lactating primiparous cows were randomly divided into two groups that were fed a corn silage-based,high-starch diet(HSD) or a grass silage-based,high-fiber diet(HFD).Cows in each dietary group were randomly assigned to four treatments in a 4 × 4 Latin square design.The bacterial DFM used were selected for their proven CH4-reducing effect in vitro.Treatments included control(without DFM) and 3 DFM treatments: Propionibacterium freudenreichii 53-W(2.9 × 10^10 colony forming units(CFU)/cow per day),Lactobacillus pentosus D31(3.6 × 10^11 CFU/cow per day) and Lactobacillus bulgaricus D1(4.6 × 10^10 CFU/cow per day).Each experimental period included 4 weeks of treatment and 1 week of wash-out,with measures performed in the fourth week of the treatment period.Enteric CH4 emissions were measured during 3 consecutive days using respiration chambers.Rumen samples were collected for ruminal fermentation parameters and quantitative microbial analyses.Milk samples were collected for composition analysis.Body weight of cows were recorded at the end of each treatment period.Irrespective of diet,no mitigating effect of DFM was observed on CH4 emissions in dairy cows.In contrast,Propionibacterium increased CH4 intensity by 27%(g CH4/kg milk) in cows fed HSD.There was no effect of DFM on other fermentation parameters and on bacterial,archaeal and protozoal numbers.Similarly,the effect of DFM on milk fatty acid composition was negligible.Propionibacterium and L.pentosus DFM tended to increase body weight gain with HSD.We conclude that,contrary to the effect previously observed in vitro,bacterial DFM Propionibacterium freudenreichii 53-W,Lactobacillus pentosus D31 and Lactobacillus bulgaricus D1 did not alter ruminal fermentation and failed to reduce CH4 emissions in lactating primiparous cows fed high-starch or high-fiber diets.展开更多
Objective: To evaluate the ability of Bacillus spp. as direct-fed microbials(DFM) to biodegrade al atoxin B1(AFB1) by using an in vitro digestive model simulating in vivo conditions.Methods: Sixty-nine Bacillus isolat...Objective: To evaluate the ability of Bacillus spp. as direct-fed microbials(DFM) to biodegrade al atoxin B1(AFB1) by using an in vitro digestive model simulating in vivo conditions.Methods: Sixty-nine Bacillus isolates were obtained from intestines, and soil samples were screened by using a selective media method against 0.25 and 1.00 μg/m L of AFB1 in modii ed Czapek-Dox medium. Plates were incubated at 37 °C and observed every two days for two weeks. Physiological properties of the three Bacillus spp. candidates were characterized biochemically and by 16 S r RNA sequence analyzes for identii cation. Tolerance to acidic p H, osmotic concentrations of Na Cl, bile salts were tested, and antimicrobial sensitivity proi les were also determined. Bacillus candidates were individually sporulated by using a solid fermentation method and combined. Spores were incorporated into 1 of 3 experimental feed groups: 1) Negative control group, with unmedicated starter broiler feed without AFB1; 2) Positive control group, with negative control feed contaminated with 0.01% AFB1; 3) DFM treated group, with positive control feed supplemented with 109 spores/g. After digestion time(3:15 h), supernatants and digesta were collected for high-performance liquid chromatography l uorescence detection analysis by triplicate.Results: Three out of those sixty-nine DFM candidates showed ability to biodegrade AFB1 in vitro based on growth as well as reduction of l uorescence and area of clearance around each colony in modii ed Czapek-Dox medium which was clearly visible under day light after 48 h of evaluation. Analysis of 16S-DNA identii ed the strains as Bacillus amyloliquefaciens, Bacillus megaterium and Bacillus subtilis. The three Bacillus strains were tolerant to acidic conditions(p H 2.0), tolerant to a high osmotic pressure(Na Cl at 6.5%), and were able to tolerate 0.037% bile salts after 24 h of incubation. No signii cant dif erences(P > 0.05) were observed in the concentrations of AFB1 in neither the supernatants nor digesta samples evaluated by highperformance liquid chromatography with l uorescence detection between positive control or DFM treated groups. Conclusions: In vitro digestion time was not enough to confirm biodegradation of AFB1. Further studies to evaluate the possible biodegradation ef ects of the BacillusDFM when continuously administered in experimentally contaminated feed with AFB1, are in progress.展开更多

On the other hand, methanogenesis in rumen, which accounts for a significant loss of ruminant energy and increased greenhouse gas in environment, is of great concern, therefore, use of DFM for improving productivity without compromising the animal health and ecological sustainability is encouraged. The present study was conducted to investigate the methane reducing potential of bacteriocinogenic strain Pediococcus pentosaceus-34. Since, the culture showed no hemolysis on blood agar and DNase activity, hence, it was considered to be avirulent in nature, a prerequisite for any DFM. The culture also showed tolerance to pH 5.0 for 24 h with 0.5% organic acid mixture, whereas when given a shock for 2 h at different p H and organic acids concentrations, it showed growth at pH 3.0 and 4.0 with 0.1 and 1.0% organic acids, respectively, as having good animal probiotics attributes. The total gas production was significantly(P〈0.05) higher in live pedicoccal culture(LPC) and dead pedicoccal culture(DPC) both with wheat straw, when compared to the control. In sugarcane bagasse, gas production was significantly lower(P〈0.05) with LPC compared to the control and DPC both. Methane was reduced by the inclusion of LPC in sugarcane bagasse(0.07 mL CH4 mg–1 dry matter digestibility) with no effect on other rumen fermentation parameters. However, with wheat straw and LPC total gas, in vitro dry matter digestibility, total volatile fatty acids increased significantly but no reduction in methane production was observed in comparison to the control. Therefore, further research is warranted in this direction, if the bacteriocinogenic strains can be used as DFM for ruminants to improve the ruminant productivity.展开更多

Thirty-two Hu lambs with inihtial weight of 22.20 ( ±0.75 ) kg were randomly assigned to one of four treatments: basal diet without additive (control), added with yeast culture at 15 g/head/d (YEC), YEC plus Bacillus licheniformis preparation at 2. 3 g/head/d (YBL) or plus Clostridium butyricum preparation at 2. 3 g/head/d (YCB). The feeding trial lasted 75 d with 15 d for adaptation. Feed intake was not influenced (P 〉0. 05) by treatment. Average daily gain of growing lambs was 102, 114, 90, and 89 g/d in control, YEC, YBL, and YCB, respectively, with no significant difference (P 〉 0.05) among treatments, but the carcass weight of YEC lambs was significantly higher (P 〈 0.05) than that of other treatments. Total volatile fatty acids and acetate to propionate ratio in the rumen were unaffected, although the butyric acid concentration was higher ( P 〈 0.05 ) in the ru men fluid of YCB lambs compared with YEC lambs and slightly higher ( P 〉 0. 05 ) than in controls and YBL lambs. Solid-associated fungi population relative to total rumen bacteria 16S ribosomal DNA was significantly lower ( P 〈 0. 05 ) in YBL lambs (3.55) compared with those on YCB (23.12). There was little difference in blood glucose and plasma urea-N concentrations among the treatments. Blood concentrations of creatinine and globulin were significantly higher (P 〈0.05) in YBL lambs, compared with the control and YEC-fed animals, and no difference with YCB lambs. Total protein and triglycerides in blood were significantly (P 〈 0.05) higher in YBL lambs, compared with controls. These serum biochemical parameters suggest that treatment increased amounts of absorbable protein but not efficiency of protein utilization and in YBL and YCB lambs. The results indicated that yeast culture improve growth performance, while little advantage could be expected from combining yeast culture with either the B. licheniformis preparation or C. butyricum preparation. More research using lower doses of B. licheniformis prepara- tion or C. butyricum preparation in combination with yeast culture is warranted.展开更多

In Exp. 1, 400 steers (BW = 348 kg) were assigned to treatments: <strong>CON</strong> = lactose carrier only, <strong>BOV</strong> =<em> P. freudenreichii </em>(NP24) +<em> L. acidophilus</em> (NP51), <strong>BOVD</strong> = <em>P. freudenreichii</em> (NP24) +<em> L. acidophilus</em> (NP51), and <strong>COMB</strong> = BOV fed for the first 101 d on feed, followed by BOVD for the final 28 d prior to harvest. In Exp. 2 (n = 1800;BW = 354 kg) and Exp. 3 (n = 112;BW = 397 kg), steers were utilized in a randomized complete block design and assigned to DFM treatments using low dose and high dose, respectively. Fecal samples were collected prior to harvest and analyzed for <em>E. coli</em> serogroups. In Exp. 1, DFM reduced (P < 0.01) the concentration of<em> E. coli</em> O157. Prevalence of O157 was reduced by BOVD supplementation in Exp. 2 and 3 (P < 0.01 and P = 0.08, respectively), and concentration of <em>E. coli</em> O157 in positive samples was reduced in both experiments where enumeration was performed (P ≤ 0.02). Weighted mean differences across the three experiments were equal to a 33% reduction in the prevalence of E. coli O157:H7 in BOVD treated cattle. A significant reduction in prevalence of O26, O45, O103, and O121 was observed in Exp. 2 (P ≤ 0.03). These results indicate that high levels of <em>L. acidophilus</em> (NP51) may represent an effective pre-harvest food safety intervention to reduce fecal shedding of several <em>E. coli</em> serogroups.展开更多

Gas explosion in confined space often leads to significant pressure oscillation.It is widely recognized that structural damage can be severe when the oscillation frequency of the load resonates with the natural vibrat...Gas explosion in confined space often leads to significant pressure oscillation.It is widely recognized that structural damage can be severe when the oscillation frequency of the load resonates with the natural vibration frequency of the structure.To reveal the oscillation mechanism of gas explosion load,the experiment of gas explosion was conducted in a large-scale confined tube with the length of 30 m,and the explosion process was numerically analyzed using FLACS.The results show that the essential cause of oscillation effect is the reflection of the pressure wave.In addition,due to the difference in the propagation path of the pressure wave,the load oscillation frequency at the middle position of the tunnel is twice that at the end position.The average sound velocity can be used to calculate the oscillation frequency of overpressure accurately,and the error is less than 15%.The instability of the flame surface and the increase of flame turbulence caused by the interaction between the pressure wave and the flame surface are the main contributors to the increase in overpressure and amplitude.The overpressure peaks calculated by the existing flame instability model and turbulence disturbance model are 31.7%and 34.7%lower than the numerical results,respectively.The turbulence factor model established in this work can describe the turbulence enhancement effect caused by flame instability and oscillatory load,and the difference between the theoretical and numerical results is only 4.6%.In the theoretical derivation of the overpressure model,an improved model of dynamic turbulence factor is established,which can describe the enhancement effect of turbulence factor caused by flame instability and self-turbulence.Based on the one-dimensional propagation theory of pressure wave,the oscillatory effect of the load is derived to calculate the frequency and amplitude of pressure oscillation.The average error of amplitude and frequency is less than 20%.展开更多

In rock drilling and blasting,the misfire of electronic detonators will not only affect the rock fragmentation result but also bring serious potential safety hazards to engineering construction.An accurate and compreh...In rock drilling and blasting,the misfire of electronic detonators will not only affect the rock fragmentation result but also bring serious potential safety hazards to engineering construction.An accurate and comprehensive understanding of the failure mechanisms of electronic detonators subjected to impact loading is of great significance to the reliability design and field safety use of electronic detonators.The spatial distribution characteristics and failure modes of misfired electronic detonators under different application scenarios are statistically analysed.The results show that under high impact loads,electronic detonators will experience failure phenomena such as rupture of the fuse head,fracture of the bridge wire,falling off of the solder joint,chip module damage and insufficient initiation energy after deformation.The lack of impact resistance is the primary cause of misfire of electronic detonators.Combined with the underwater impact resistance test and the impact load test in the adjacent blasthole on site,the formulas of the impact failure probability of the electronic detonator under different stress‒strength distribution curves are deduced.The test and evaluation method of the impact resistance of electronic detonators based on stress‒strength interference theory is proposed.Furthermore,the impact failure model of electronic detonators considering the strength degradation effect under repeated random loads is established.On this basis,the failure mechanism of electronic detonators under different application environments,such as open-pit blasting and underground blasting,is revealed,which provides scientific theory and methods for the reliability analysis,design and type selection of electronic detonators in rock drilling and blasting.展开更多

The demand response(DR)market,as a vital complement to the electricity spot market,plays a key role in evoking user-side regulation capability to mitigate system-level supply‒demand imbalances during extreme events.Wh...The demand response(DR)market,as a vital complement to the electricity spot market,plays a key role in evoking user-side regulation capability to mitigate system-level supply‒demand imbalances during extreme events.While the DR market offers the load aggregator(LA)additional profitable opportunities beyond the electricity spot market,it also introduces new trading risks due to the significant uncertainty in users’behaviors.Dispatching energy storage systems(ESSs)is an effective means to enhance the risk management capabilities of LAs;however,coordinating ESS operations with dual-market trading strategies remains an urgent challenge.To this end,this paper proposes a novel systematic risk-aware coordinated trading model for the LA in concurrently participating in the day-ahead electricity spot market and DR market,which incorporates the capacity allocation mechanism of ESS based on market clearing rules to jointly formulate bidding and pricing decisions for the dual market.First,the intrinsic coupling characteristics of the LA participating in the dual market are analyzed,and a joint optimization framework for formulating bidding and pricing strategies that integrates ESS facilities is proposed.Second,an uncertain user response model is developed based on price‒response mechanisms,and actual market settlement rules accounting for under-and over-responses are employed to calculate trading revenues,where possible revenue losses are quantified via conditional value at risk.Third,by imposing these terms and the capacity allocation mechanism of ESS,the risk-aware stochastic coordinated trading model of the LA is built,where the bidding and pricing strategies in the dual model that trade off risk and profit are derived.The simulation results of a case study validate the effectiveness of the proposed trading strategy in controlling trading risk and improving the trading income of the LA.展开更多
A conceptual model of intermittent joints is introduced to the cyclic shear test in the laboratory to explore the effects of loading parameters on its shear behavior under cyclic shear loading.The results show that th...A conceptual model of intermittent joints is introduced to the cyclic shear test in the laboratory to explore the effects of loading parameters on its shear behavior under cyclic shear loading.The results show that the loading parameters(initial normal stress,normal stiffness,and shear velocity)determine propagation paths of the wing and secondary cracks in rock bridges during the initial shear cycle,creating different morphologies of macroscopic step-path rupture surfaces and asperities on them.The differences in stress state and rupture surface induce different cyclic shear responses.It shows that high initial normal stress accelerates asperity degradation,raises shear resistance,and promotes compression of intermittent joints.In addition,high normal stiffness provides higher normal stress and shear resistance during the initial cycles and inhibits the dilation and compression of intermittent joints.High shear velocity results in a higher shear resistance,greater dilation,and greater compression.Finally,shear strength is most sensitive to initial normal stress,followed by shear velocity and normal stiffness.Moreover,average dilation angle is most sensitive to initial normal stress,followed by normal stiffness and shear velocity.During the shear cycles,frictional coefficient is affected by asperity degradation,backfilling of rock debris,and frictional area,exhibiting a non-monotonic behavior.展开更多
Accurate and efficient prediction of the distribution of surface loads on buildings subjected to explosive effects is crucial for rapidly calculating structural dynamic responses,establishing effective protective meas...Accurate and efficient prediction of the distribution of surface loads on buildings subjected to explosive effects is crucial for rapidly calculating structural dynamic responses,establishing effective protective measures,and designing civil defense engineering solutions.Current state-of-the-art methods face several issues:Experimental research is difficult and costly to implement,theoretical research is limited to simple geometries and lacks precision,and direct simulations require substantial computational resources.To address these challenges,this paper presents a data-driven method for predicting blast loads on building surfaces.This approach increases both the accuracy and computational efficiency of load predictions when the geometry of the building changes while the explosive yield remains constant,significantly improving its applicability in complex scenarios.This study introduces an innovative encoder-decoder graph neural network model named BlastGraphNet,which uses a message-passing mechanism to predict the overpressure and impulse load distributions on buildings with conventional and complex geometries during explosive events.The model also facilitates related downstream applications,such as damage mode identification and rapid assessment of virtual city explosions.The calculation results indicate that the prediction error of the model for conventional building tests is less than 2%,and its inference speed is 3-4 orders of magnitude faster than that of state-of-the-art numerical methods.In extreme test cases involving buildings with complex geometries and building clusters,the method achieved high accuracy and excellent generalizability.The strong adaptability and generalizability of BlastGraphNet confirm that this novel method enables precise real-time prediction of blast loads and provides a new paradigm for damage assessment in protective engineering.展开更多
Non-seismically designed(NSD)beam-column joints are susceptible to joint shear failure under seismic loads.Although significant research is available on the seismic behavior of such joints of planar frames,the informa...Non-seismically designed(NSD)beam-column joints are susceptible to joint shear failure under seismic loads.Although significant research is available on the seismic behavior of such joints of planar frames,the information on the seismic behavior of joints of space frames(3D joints)is insufficient.The 3D joints are subjected to bi-directional excitation,which results in an interaction between the shear strength obtained for the joint in the two orthogonal directions separately.The bi-directional seismic behavior of corner reinforced concrete(RC)joints is the focus of this study.First,a detailed finite element(FE)model using the FE software Abaqus,is developed and validated using the test results from the literature.The validated modeling procedure is used to conduct a parametric study to investigate the influence of different parameters such as concrete strength,dimensions of main and transverse beams framing into the joint,presence or absence of a slab,axial load ratio and loading direction on the seismic behavior of joints.By subjecting the models to different combinations of loads on the beams along perpendicular directions,the interaction of the joint shear strength in two orthogonal directions is studied.The comparison of the interaction curves of the joints obtained from the numerical study with a quadratic(circular)interaction curve indicates that in a majority of cases,the quadratic interaction model can represent the strength interaction diagrams of RC beam to column connections with governing joint shear failure reasonably well.展开更多
To address the problems of low accuracy by the CONWEP model and poor efficiency by the Coupled Eulerian-Lagrangian(CEL)method in predicting close-range air blast loads of cylindrical charges,a neural network-based sim...To address the problems of low accuracy by the CONWEP model and poor efficiency by the Coupled Eulerian-Lagrangian(CEL)method in predicting close-range air blast loads of cylindrical charges,a neural network-based simulation(NNS)method with higher accuracy and better efficiency was proposed.The NNS method consisted of three main steps.First,the parameters of blast loads,including the peak pressures and impulses of cylindrical charges with different aspect ratios(L/D)at different stand-off distances and incident angles were obtained by two-dimensional numerical simulations.Subsequently,incident shape factors of cylindrical charges with arbitrary aspect ratios were predicted by a neural network.Finally,reflected shape factors were derived and implemented into the subroutine of the ABAQUS code to modify the CONWEP model,including modifications of impulse and overpressure.The reliability of the proposed NNS method was verified by related experimental results.Remarkable accuracy improvement was acquired by the proposed NNS method compared with the unmodified CONWEP model.Moreover,huge efficiency superiority was obtained by the proposed NNS method compared with the CEL method.The proposed NNS method showed good accuracy when the scaled distance was greater than 0.2 m/kg^(1/3).It should be noted that there is no need to generate a new dataset again since the blast loads satisfy the similarity law,and the proposed NNS method can be directly used to simulate the blast loads generated by different cylindrical charges.The proposed NNS method with high efficiency and accuracy can be used as an effective method to analyze the dynamic response of structures under blast loads,and it has significant application prospects in designing protective structures.展开更多

文摘Studies of wave-current interactions are vital for the safe design of structures.Regular waves in the presence of uniform,linear shear,and quadratic shear currents are explored by the High-Level Green-Naghdi model in this paper.The five-point central difference method is used for spatial discretization,and the fourth-order Adams predictor-corrector scheme is employed for marching in time.The domain-decomposition method is applied for the wave-current generation and absorption.The effects of currents on the wave profile and velocity field are examined under two conditions:the same velocity of currents at the still-water level and the constant flow volume of currents.Wave profiles and velocity fields demonstrate substantial differences in three types of currents owing to the diverse vertical distribution of current velocity and vorticity.Then,loads on small-scale vertical cylinders subjected to regular waves and three types of background currents with the same flow volume are investigated.The maximum load intensity and load fluctuation amplitude in uniform,linear shear,and quadratic shear currents increase sequentially.The stretched superposition method overestimates the maximum load intensity and load fluctuation amplitude in opposing currents and underestimates these values in following currents.The stretched superposition method obtains a poor approximation for strong nonlinear waves,particularly in the case of the opposing quadratic shear current.

文摘This study establishes and validates a method for the precise quantification of aquatic microbial loads using microbial diversity absolute quantitative sequencing.By adding synthetic spike-in DNA to water samples from the Dahei River prior to DNA extraction and 16S rRNA gene sequencing,it generates standard curves to convert sequencing data into absolute microbial copy numbers.The method,which is proved highly accurate(R^(2)>0.99),reveals a clear contrast between the river sites:the upstream community has not only a significantly higher total microbial load but also a completely different makeup of species compared to the downstream site.This approach effectively overcomes the limitations of relative abundance analysis,providing a powerful tool for environmental monitoring,and proposes key steps for future standardization to ensure data comparability and integration.

文摘Direct-fed microbials(DFMs)are feed additives containing live naturally existing microbes that can benefit animals’health and production performance.Due to the banned or strictly limited prophylactic and growth promoting usage of antibiotics,DFMs have been considered as one of antimicrobial alternatives in livestock industry.Microorganisms used as DFMs for ruminants usually consist of bacteria including lactic acid producing bacteria,lactic acid utilizing bacteria and other bacterial groups,and fungi containing Saccharomyces and Aspergillus.To date,the available DFMs for ruminants have been largely based on their effects on improving the feed efficiency and ruminant productivity through enhancing the rumen function such as stabilizing ruminal pH,promoting ruminal fermentation and feed digestion.Recent research has shown emerging evidence that the DFMs may improve performance and health in young ruminants,however,these positive outcomes were not consistent among studies and the modes of action have not been clearly defined.This review summarizes the DFM studies conducted in ruminants in the last decade,aiming to provide the new knowledge on DFM supplementation strategies for various ruminant production stages,and to identify what are the potential barriers and challenges for current ruminant industry to adopt the DFMs.Overall literature research indicates that DFMs have the potential to mitigate ruminal acidosis,improve immune response and gut health,increase productivity(growth and milk production),and reduce methane emissions or fecal shedding of pathogens.More research is needed to explore the mode of action of specific DFMs in the gut of ruminants,and the optimal supplementation strategies to promote the development and efficiency of DFM products for ruminants.
文摘For many years, ruminant nutritionists and microbiologists have been interested in manipulating the microbial ecosystem of the rumen to improve production efficiency of different ruminant species. Removal and restriction of antibiotics subtherapeutic uses from ruminant diets has amplified interest in improving nutrient utilization and animal performance and search for more safe alternatives. Some bacterial and fungal microorganisms as a direct-fed microbial(DFM) can be the most suitable solutions. Microorganisms that are commonly used in DFM for ruminants may be classified mainly as lactic acid producing bacteria(LAB), lactic acid utilizing bacteria(LUB), or other microorganism's species like Lactobacillus, Bifidobacterium, Enterococcus, Streptococcus, Bacillus, Propionibacterium, Megasphaera elsdenii and Prevotellabryantii, in addition to some fungal species of yeast such as Saccharomyces and Aspergillus. A definitive mode of action for bacterial or fungal DFM has not been established; although a variety of mechanisms have been suggested. Bacterial DFM potentially moderate rumen conditions, and improve weight gain and feed efficiency. Fungal DFM may reduce harmful oxygen from the rumen, prevent excess lactate production, increase feed digestibility, and alter rumen fermentation patterns. DFM may also compete with and inhibit the growth of pathogens, immune system modulation, and modulate microbial balance in the gastrointestinal tract. Improved dry matter intake, milk yield, fat corrected milk yield and milk fat content were obtained with DFM administration. However, the response to DFM is not constant; depending on dosages, feeding times and frequencies, and strains of DFM. Nonetheless, recent studies have supported the positive effects of DFM on ruminant performance.

文摘Direct-fed microbials(DFM) are considered as a promising technique to improve animal productivity without affecting animal health or harming the environment.The potential of three bacterial DFM to reduce methane(CH4)emissions,modulate ruminal fermentation,milk production and composition of primiparous dairy cows was examined in this study.As previous reports have shown that DFM respond differently to different diets,two contrasting diets were used in this study.Eight lactating primiparous cows were randomly divided into two groups that were fed a corn silage-based,high-starch diet(HSD) or a grass silage-based,high-fiber diet(HFD).Cows in each dietary group were randomly assigned to four treatments in a 4 × 4 Latin square design.The bacterial DFM used were selected for their proven CH4-reducing effect in vitro.Treatments included control(without DFM) and 3 DFM treatments: Propionibacterium freudenreichii 53-W(2.9 × 10^10 colony forming units(CFU)/cow per day),Lactobacillus pentosus D31(3.6 × 10^11 CFU/cow per day) and Lactobacillus bulgaricus D1(4.6 × 10^10 CFU/cow per day).Each experimental period included 4 weeks of treatment and 1 week of wash-out,with measures performed in the fourth week of the treatment period.Enteric CH4 emissions were measured during 3 consecutive days using respiration chambers.Rumen samples were collected for ruminal fermentation parameters and quantitative microbial analyses.Milk samples were collected for composition analysis.Body weight of cows were recorded at the end of each treatment period.Irrespective of diet,no mitigating effect of DFM was observed on CH4 emissions in dairy cows.In contrast,Propionibacterium increased CH4 intensity by 27%(g CH4/kg milk) in cows fed HSD.There was no effect of DFM on other fermentation parameters and on bacterial,archaeal and protozoal numbers.Similarly,the effect of DFM on milk fatty acid composition was negligible.Propionibacterium and L.pentosus DFM tended to increase body weight gain with HSD.We conclude that,contrary to the effect previously observed in vitro,bacterial DFM Propionibacterium freudenreichii 53-W,Lactobacillus pentosus D31 and Lactobacillus bulgaricus D1 did not alter ruminal fermentation and failed to reduce CH4 emissions in lactating primiparous cows fed high-starch or high-fiber diets.

文摘Objective: To evaluate the ability of Bacillus spp. as direct-fed microbials(DFM) to biodegrade al atoxin B1(AFB1) by using an in vitro digestive model simulating in vivo conditions.Methods: Sixty-nine Bacillus isolates were obtained from intestines, and soil samples were screened by using a selective media method against 0.25 and 1.00 μg/m L of AFB1 in modii ed Czapek-Dox medium. Plates were incubated at 37 °C and observed every two days for two weeks. Physiological properties of the three Bacillus spp. candidates were characterized biochemically and by 16 S r RNA sequence analyzes for identii cation. Tolerance to acidic p H, osmotic concentrations of Na Cl, bile salts were tested, and antimicrobial sensitivity proi les were also determined. Bacillus candidates were individually sporulated by using a solid fermentation method and combined. Spores were incorporated into 1 of 3 experimental feed groups: 1) Negative control group, with unmedicated starter broiler feed without AFB1; 2) Positive control group, with negative control feed contaminated with 0.01% AFB1; 3) DFM treated group, with positive control feed supplemented with 109 spores/g. After digestion time(3:15 h), supernatants and digesta were collected for high-performance liquid chromatography l uorescence detection analysis by triplicate.Results: Three out of those sixty-nine DFM candidates showed ability to biodegrade AFB1 in vitro based on growth as well as reduction of l uorescence and area of clearance around each colony in modii ed Czapek-Dox medium which was clearly visible under day light after 48 h of evaluation. Analysis of 16S-DNA identii ed the strains as Bacillus amyloliquefaciens, Bacillus megaterium and Bacillus subtilis. The three Bacillus strains were tolerant to acidic conditions(p H 2.0), tolerant to a high osmotic pressure(Na Cl at 6.5%), and were able to tolerate 0.037% bile salts after 24 h of incubation. No signii cant dif erences(P > 0.05) were observed in the concentrations of AFB1 in neither the supernatants nor digesta samples evaluated by highperformance liquid chromatography with l uorescence detection between positive control or DFM treated groups. Conclusions: In vitro digestion time was not enough to confirm biodegradation of AFB1. Further studies to evaluate the possible biodegradation ef ects of the BacillusDFM when continuously administered in experimentally contaminated feed with AFB1, are in progress.

文摘Direct-fed microbials(DFM), generally regarded as safe status, are successfully used in improving rumen ecology, gastro-intestinal health, feed efficiency, milk production and growth rate in ruminants. On the other hand, methanogenesis in rumen, which accounts for a significant loss of ruminant energy and increased greenhouse gas in environment, is of great concern, therefore, use of DFM for improving productivity without compromising the animal health and ecological sustainability is encouraged. The present study was conducted to investigate the methane reducing potential of bacteriocinogenic strain Pediococcus pentosaceus-34. Since, the culture showed no hemolysis on blood agar and DNase activity, hence, it was considered to be avirulent in nature, a prerequisite for any DFM. The culture also showed tolerance to pH 5.0 for 24 h with 0.5% organic acid mixture, whereas when given a shock for 2 h at different p H and organic acids concentrations, it showed growth at pH 3.0 and 4.0 with 0.1 and 1.0% organic acids, respectively, as having good animal probiotics attributes. The total gas production was significantly(P〈0.05) higher in live pedicoccal culture(LPC) and dead pedicoccal culture(DPC) both with wheat straw, when compared to the control. In sugarcane bagasse, gas production was significantly lower(P〈0.05) with LPC compared to the control and DPC both. Methane was reduced by the inclusion of LPC in sugarcane bagasse(0.07 mL CH4 mg–1 dry matter digestibility) with no effect on other rumen fermentation parameters. However, with wheat straw and LPC total gas, in vitro dry matter digestibility, total volatile fatty acids increased significantly but no reduction in methane production was observed in comparison to the control. Therefore, further research is warranted in this direction, if the bacteriocinogenic strains can be used as DFM for ruminants to improve the ruminant productivity.

文摘The effects of yeast culture and directfed microbes on the growth performance of weaner lambs was examined. Thirty-two Hu lambs with inihtial weight of 22.20 ( ±0.75 ) kg were randomly assigned to one of four treatments: basal diet without additive (control), added with yeast culture at 15 g/head/d (YEC), YEC plus Bacillus licheniformis preparation at 2. 3 g/head/d (YBL) or plus Clostridium butyricum preparation at 2. 3 g/head/d (YCB). The feeding trial lasted 75 d with 15 d for adaptation. Feed intake was not influenced (P 〉0. 05) by treatment. Average daily gain of growing lambs was 102, 114, 90, and 89 g/d in control, YEC, YBL, and YCB, respectively, with no significant difference (P 〉 0.05) among treatments, but the carcass weight of YEC lambs was significantly higher (P 〈 0.05) than that of other treatments. Total volatile fatty acids and acetate to propionate ratio in the rumen were unaffected, although the butyric acid concentration was higher ( P 〈 0.05 ) in the ru men fluid of YCB lambs compared with YEC lambs and slightly higher ( P 〉 0. 05 ) than in controls and YBL lambs. Solid-associated fungi population relative to total rumen bacteria 16S ribosomal DNA was significantly lower ( P 〈 0. 05 ) in YBL lambs (3.55) compared with those on YCB (23.12). There was little difference in blood glucose and plasma urea-N concentrations among the treatments. Blood concentrations of creatinine and globulin were significantly higher (P 〈0.05) in YBL lambs, compared with the control and YEC-fed animals, and no difference with YCB lambs. Total protein and triglycerides in blood were significantly (P 〈 0.05) higher in YBL lambs, compared with controls. These serum biochemical parameters suggest that treatment increased amounts of absorbable protein but not efficiency of protein utilization and in YBL and YCB lambs. The results indicated that yeast culture improve growth performance, while little advantage could be expected from combining yeast culture with either the B. licheniformis preparation or C. butyricum preparation. More research using lower doses of B. licheniformis prepara- tion or C. butyricum preparation in combination with yeast culture is warranted.
文摘Three experiments were conducted to evaluate direct-fed microbial (<strong>DFM</strong>) supplementation on live performance, carcass characteristics, and fecal shedding of <em>E. coli</em> in feedlot steers. In Exp. 1, 400 steers (BW = 348 kg) were assigned to treatments: <strong>CON</strong> = lactose carrier only, <strong>BOV</strong> =<em> P. freudenreichii </em>(NP24) +<em> L. acidophilus</em> (NP51), <strong>BOVD</strong> = <em>P. freudenreichii</em> (NP24) +<em> L. acidophilus</em> (NP51), and <strong>COMB</strong> = BOV fed for the first 101 d on feed, followed by BOVD for the final 28 d prior to harvest. In Exp. 2 (n = 1800;BW = 354 kg) and Exp. 3 (n = 112;BW = 397 kg), steers were utilized in a randomized complete block design and assigned to DFM treatments using low dose and high dose, respectively. Fecal samples were collected prior to harvest and analyzed for <em>E. coli</em> serogroups. In Exp. 1, DFM reduced (P < 0.01) the concentration of<em> E. coli</em> O157. Prevalence of O157 was reduced by BOVD supplementation in Exp. 2 and 3 (P < 0.01 and P = 0.08, respectively), and concentration of <em>E. coli</em> O157 in positive samples was reduced in both experiments where enumeration was performed (P ≤ 0.02). Weighted mean differences across the three experiments were equal to a 33% reduction in the prevalence of E. coli O157:H7 in BOVD treated cattle. A significant reduction in prevalence of O26, O45, O103, and O121 was observed in Exp. 2 (P ≤ 0.03). These results indicate that high levels of <em>L. acidophilus</em> (NP51) may represent an effective pre-harvest food safety intervention to reduce fecal shedding of several <em>E. coli</em> serogroups.

文摘Ice-breaking methods have become increasingly significant with the ongoing development of the polar regions.Among many ice-breaking methods,ice-breaking that utilizes a moving load is unique compared with the common collision or impact methods.A moving load can generate flexural-gravity waves(FGWs),under the influence of which the ice sheet undergoes deformation and may even experience structural damage.Moving loads can be divided into above-ice loads and underwater loads.For the above-ice loads,we discuss the characteristics of the FGWs generated by a moving load acting on a complete ice sheet,an ice sheet with a crack,and an ice sheet with a lead of open water.For underwater loads,we discuss the influence on the ice-breaking characteristics of FGWs of the mode of motion,the geometrical features,and the trajectory of motion of the load.In addition to discussing the status of current research and the technical challenges of ice-breaking by moving loads,this paper also looks ahead to future research prospects and presents some preliminary ideas for consideration.

文摘Gas explosion in confined space often leads to significant pressure oscillation.It is widely recognized that structural damage can be severe when the oscillation frequency of the load resonates with the natural vibration frequency of the structure.To reveal the oscillation mechanism of gas explosion load,the experiment of gas explosion was conducted in a large-scale confined tube with the length of 30 m,and the explosion process was numerically analyzed using FLACS.The results show that the essential cause of oscillation effect is the reflection of the pressure wave.In addition,due to the difference in the propagation path of the pressure wave,the load oscillation frequency at the middle position of the tunnel is twice that at the end position.The average sound velocity can be used to calculate the oscillation frequency of overpressure accurately,and the error is less than 15%.The instability of the flame surface and the increase of flame turbulence caused by the interaction between the pressure wave and the flame surface are the main contributors to the increase in overpressure and amplitude.The overpressure peaks calculated by the existing flame instability model and turbulence disturbance model are 31.7%and 34.7%lower than the numerical results,respectively.The turbulence factor model established in this work can describe the turbulence enhancement effect caused by flame instability and oscillatory load,and the difference between the theoretical and numerical results is only 4.6%.In the theoretical derivation of the overpressure model,an improved model of dynamic turbulence factor is established,which can describe the enhancement effect of turbulence factor caused by flame instability and self-turbulence.Based on the one-dimensional propagation theory of pressure wave,the oscillatory effect of the load is derived to calculate the frequency and amplitude of pressure oscillation.The average error of amplitude and frequency is less than 20%.

文摘Determining earth pressure on jacked pipes is essential for ensuring lining safety and calculating jacking force,especially for deep-buried pipes.To better reflect the soil arching effect resulting from the excavation of rectangular jacked pipes and the distribution of the earth pressure on jacked pipes,we present an analytical solution for predicting the vertical earth pressure on deep-buried rectangular pipe jacking tunnels,incorporating the tunnelling-induced ground loss distribution.Our proposed analytical model consists of the upper multi-layer parabolic soil arch and the lower friction arch.The key parameters(i.e.,width and height of friction arch B and height of parabolic soil arch H 1)are determined according to the existing research,and an analytical solution for K l is derived based on the distribution characteristics of the principal stress rotation angle.With consideration for the transition effect of the mechanical characteristics of the parabolic arch zone,an analytical solution for soil load transfer is derived.The prediction results of our analytical solution are compared with tests and simulation results to validate the effectiveness of the proposed analytical solution.Finally,the effects of different parameters on the soil pressure are discussed.

文摘In rock drilling and blasting,the misfire of electronic detonators will not only affect the rock fragmentation result but also bring serious potential safety hazards to engineering construction.An accurate and comprehensive understanding of the failure mechanisms of electronic detonators subjected to impact loading is of great significance to the reliability design and field safety use of electronic detonators.The spatial distribution characteristics and failure modes of misfired electronic detonators under different application scenarios are statistically analysed.The results show that under high impact loads,electronic detonators will experience failure phenomena such as rupture of the fuse head,fracture of the bridge wire,falling off of the solder joint,chip module damage and insufficient initiation energy after deformation.The lack of impact resistance is the primary cause of misfire of electronic detonators.Combined with the underwater impact resistance test and the impact load test in the adjacent blasthole on site,the formulas of the impact failure probability of the electronic detonator under different stress‒strength distribution curves are deduced.The test and evaluation method of the impact resistance of electronic detonators based on stress‒strength interference theory is proposed.Furthermore,the impact failure model of electronic detonators considering the strength degradation effect under repeated random loads is established.On this basis,the failure mechanism of electronic detonators under different application environments,such as open-pit blasting and underground blasting,is revealed,which provides scientific theory and methods for the reliability analysis,design and type selection of electronic detonators in rock drilling and blasting.

文摘With increasing water depth,marine drilling conductors exhibit higher slenderness ratios,significantly reducing their resistance to environmental loads in Arctic waters.These conductors,when subjected to combined wind,current,and ice loads,may experience substantial horizontal displacements and bending moments,potentially compromising off-shore operational safety and wellhead stability.Additionally,soil disturbance near the mudline diminishes the conductor’s bearing capacity,potentially rendering it inadequate for wellhead support and increasing operational risks.This study introduces a static analysis model based on plastic hinge theory to evaluate conductor survivability.The conductor analysis divides the structure into three segments:above waterline,submerged,and embedded below mudline.An idealized elastic-plastic p-y curve model characterizes soil behavior beneath the mudline,while the finite difference method(FDM)analyzes the conductor’s mechanical response under complex pile-head boundary conditions.Numerical simulations using ABAQUS validate the plastic hinge approach against conventional methods,confirming its accuracy in predicting structural performance.These results provide valuable insights for optimizing installation depths and bearing capacity designs of marine drilling conductors in ice-prone regions.

文摘The demand response(DR)market,as a vital complement to the electricity spot market,plays a key role in evoking user-side regulation capability to mitigate system-level supply‒demand imbalances during extreme events.While the DR market offers the load aggregator(LA)additional profitable opportunities beyond the electricity spot market,it also introduces new trading risks due to the significant uncertainty in users’behaviors.Dispatching energy storage systems(ESSs)is an effective means to enhance the risk management capabilities of LAs;however,coordinating ESS operations with dual-market trading strategies remains an urgent challenge.To this end,this paper proposes a novel systematic risk-aware coordinated trading model for the LA in concurrently participating in the day-ahead electricity spot market and DR market,which incorporates the capacity allocation mechanism of ESS based on market clearing rules to jointly formulate bidding and pricing decisions for the dual market.First,the intrinsic coupling characteristics of the LA participating in the dual market are analyzed,and a joint optimization framework for formulating bidding and pricing strategies that integrates ESS facilities is proposed.Second,an uncertain user response model is developed based on price‒response mechanisms,and actual market settlement rules accounting for under-and over-responses are employed to calculate trading revenues,where possible revenue losses are quantified via conditional value at risk.Third,by imposing these terms and the capacity allocation mechanism of ESS,the risk-aware stochastic coordinated trading model of the LA is built,where the bidding and pricing strategies in the dual model that trade off risk and profit are derived.The simulation results of a case study validate the effectiveness of the proposed trading strategy in controlling trading risk and improving the trading income of the LA.

文摘A conceptual model of intermittent joints is introduced to the cyclic shear test in the laboratory to explore the effects of loading parameters on its shear behavior under cyclic shear loading.The results show that the loading parameters(initial normal stress,normal stiffness,and shear velocity)determine propagation paths of the wing and secondary cracks in rock bridges during the initial shear cycle,creating different morphologies of macroscopic step-path rupture surfaces and asperities on them.The differences in stress state and rupture surface induce different cyclic shear responses.It shows that high initial normal stress accelerates asperity degradation,raises shear resistance,and promotes compression of intermittent joints.In addition,high normal stiffness provides higher normal stress and shear resistance during the initial cycles and inhibits the dilation and compression of intermittent joints.High shear velocity results in a higher shear resistance,greater dilation,and greater compression.Finally,shear strength is most sensitive to initial normal stress,followed by shear velocity and normal stiffness.Moreover,average dilation angle is most sensitive to initial normal stress,followed by normal stiffness and shear velocity.During the shear cycles,frictional coefficient is affected by asperity degradation,backfilling of rock debris,and frictional area,exhibiting a non-monotonic behavior.

文摘Accurate and efficient prediction of the distribution of surface loads on buildings subjected to explosive effects is crucial for rapidly calculating structural dynamic responses,establishing effective protective measures,and designing civil defense engineering solutions.Current state-of-the-art methods face several issues:Experimental research is difficult and costly to implement,theoretical research is limited to simple geometries and lacks precision,and direct simulations require substantial computational resources.To address these challenges,this paper presents a data-driven method for predicting blast loads on building surfaces.This approach increases both the accuracy and computational efficiency of load predictions when the geometry of the building changes while the explosive yield remains constant,significantly improving its applicability in complex scenarios.This study introduces an innovative encoder-decoder graph neural network model named BlastGraphNet,which uses a message-passing mechanism to predict the overpressure and impulse load distributions on buildings with conventional and complex geometries during explosive events.The model also facilitates related downstream applications,such as damage mode identification and rapid assessment of virtual city explosions.The calculation results indicate that the prediction error of the model for conventional building tests is less than 2%,and its inference speed is 3-4 orders of magnitude faster than that of state-of-the-art numerical methods.In extreme test cases involving buildings with complex geometries and building clusters,the method achieved high accuracy and excellent generalizability.The strong adaptability and generalizability of BlastGraphNet confirm that this novel method enables precise real-time prediction of blast loads and provides a new paradigm for damage assessment in protective engineering.
文摘Non-seismically designed(NSD)beam-column joints are susceptible to joint shear failure under seismic loads.Although significant research is available on the seismic behavior of such joints of planar frames,the information on the seismic behavior of joints of space frames(3D joints)is insufficient.The 3D joints are subjected to bi-directional excitation,which results in an interaction between the shear strength obtained for the joint in the two orthogonal directions separately.The bi-directional seismic behavior of corner reinforced concrete(RC)joints is the focus of this study.First,a detailed finite element(FE)model using the FE software Abaqus,is developed and validated using the test results from the literature.The validated modeling procedure is used to conduct a parametric study to investigate the influence of different parameters such as concrete strength,dimensions of main and transverse beams framing into the joint,presence or absence of a slab,axial load ratio and loading direction on the seismic behavior of joints.By subjecting the models to different combinations of loads on the beams along perpendicular directions,the interaction of the joint shear strength in two orthogonal directions is studied.The comparison of the interaction curves of the joints obtained from the numerical study with a quadratic(circular)interaction curve indicates that in a majority of cases,the quadratic interaction model can represent the strength interaction diagrams of RC beam to column connections with governing joint shear failure reasonably well.

文摘To address the problems of low accuracy by the CONWEP model and poor efficiency by the Coupled Eulerian-Lagrangian(CEL)method in predicting close-range air blast loads of cylindrical charges,a neural network-based simulation(NNS)method with higher accuracy and better efficiency was proposed.The NNS method consisted of three main steps.First,the parameters of blast loads,including the peak pressures and impulses of cylindrical charges with different aspect ratios(L/D)at different stand-off distances and incident angles were obtained by two-dimensional numerical simulations.Subsequently,incident shape factors of cylindrical charges with arbitrary aspect ratios were predicted by a neural network.Finally,reflected shape factors were derived and implemented into the subroutine of the ABAQUS code to modify the CONWEP model,including modifications of impulse and overpressure.The reliability of the proposed NNS method was verified by related experimental results.Remarkable accuracy improvement was acquired by the proposed NNS method compared with the unmodified CONWEP model.Moreover,huge efficiency superiority was obtained by the proposed NNS method compared with the CEL method.The proposed NNS method showed good accuracy when the scaled distance was greater than 0.2 m/kg^(1/3).It should be noted that there is no need to generate a new dataset again since the blast loads satisfy the similarity law,and the proposed NNS method can be directly used to simulate the blast loads generated by different cylindrical charges.The proposed NNS method with high efficiency and accuracy can be used as an effective method to analyze the dynamic response of structures under blast loads,and it has significant application prospects in designing protective structures.
