Background:Therapeutic responses of breast cancer vary among patients and lead to drug resistance and recurrence due to the heterogeneity.Current preclinical models,however,are inadequate for predicting individual pat...Background:Therapeutic responses of breast cancer vary among patients and lead to drug resistance and recurrence due to the heterogeneity.Current preclinical models,however,are inadequate for predicting individual patient responses towards different drugs.This study aimed to investigate the patient-derived breast cancer culture models for drug sensitivity evaluations.Methods:Tumor and adjacent tissues from female breast cancer patients were collected during surgery.Patient-derived breast cancer cells were cultured using the conditional reprogramming technique to establish 2D models.The obtained patient-derived conditional reprogramming breast cancer(CRBC)cells were subsequently embedded in alginate-gelatin methacryloyl hydrogel microspheres to form 3D culture models.Comparisons between 2D and 3D models were made using immunohistochemistry(tumor markers),MTS assays(cell viability),flow cytometry(apoptosis),transwell assays(migration),and Western blotting(protein expression).Drug sensitivity tests were conducted to evaluate patient-specific responses to anti-cancer agents.Results:2D and 3D culture models were successfully established using samples from eight patients.The 3D models retained histological and marker characteristics of the original tumors.Compared to 2D cultures,3D models exhibited increased apoptosis,enhanced drug resistance,elevated stem cell marker expression,and greater migration ability—features more reflective of in vivo tumor behavior.Conclusion:Patient-derived 3D CRBC models effectively mimic the in vivo tumor microenvironment and demonstrate stronger resistance to anti-cancer drugs than 2D models.These hydrogel-based models offer a cost-effective and clinically relevant platform for drug screening and personalized breast cancer treatment.展开更多

BACKGROUND The unique survival environment of Helicobacter pylori(H.pylori)presents challenges in establishing appropriate in vitro culture conditions.The fastidious nature of H.pylori and its relatively low growth ra...BACKGROUND The unique survival environment of Helicobacter pylori(H.pylori)presents challenges in establishing appropriate in vitro culture conditions.The fastidious nature of H.pylori and its relatively low growth rate emphasize the need for customized culture conditions,which are essential for both research and clinical applications.AIM To identify the optimal liquid culture medium for H.pylori using both reference and clinical strains.METHODS Nine H.pylori strains were incubated in 10 different broth media commonly used for bacterial cultures under microaerophilic conditions.Bacterial concentrations were estimated using serial dilutions and the pour plate method.A range of H.pylori initial inoculum concentrations was tested for each strain.For growth evaluation,optical density at 600 nm was measured at 24,48,and 72 hours.The growth trends of H.pylori were compared among strains and media.RESULTS All H.pylori strains grew successfully over time,regardless of the initial inoculum concentration.The specific growth trends and the broth that yielded the highest optical density value at each time point varied with strain.The reference strains(H.pylori 26695,ATCC 43504,and J99)consistently exhibited robust growth,whereas the clinical strains displayed distinct media preferences.Overall,bacterial growth was higher in chopped meat carbohydrate broth,Columbia broth,and fastidious anaerobe broth than in Brucella broth after 72 hours.CONCLUSION Chopped meat carbohydrate broth was the most effective for H.pylori liquid culture,and Columbia broth and fastidious anaerobe broth also supported greater growth than Brucella broth.展开更多
Chengdu teahouses,as core public spaces in marketplace society,have undergone transformative reconstruction-from“containers of everyday life”to“containers of commercial traffic and digital flows”-during the proces...Chengdu teahouses,as core public spaces in marketplace society,have undergone transformative reconstruction-from“containers of everyday life”to“containers of commercial traffic and digital flows”-during the process of modernization.Employing spatial archaeology as a methodology,combined with fieldwork and analysis of historical documents,this study systematically examines the diachronic evolution of architectural forms,functional orientations,and social networks within Chengdu teahouses.The study reveals the logic of spatial reconstruction under the interplay of multiple forces,including cultural heritage preservation,capital-driven development,and technological intervention.The findings identify three paradigms of spatial transformation in teahouses.First,heritage specimenization,which reinforces the continuity of collective memory through symbolic extraction but risks diminishing the vitality of everyday social interactions.Second,consumption upgrading,which caters to the demands of emerging groups through iterative business models yet necessitates vigilance against spatial differentiation eroding marketplace inclusivity.Third,digital parasitism,which expands communicative dimensions through technological empowerment but confronts the risk of flattening localized knowledge.These paradigms reflect both adaptive responses of traditional spaces to contemporary pressure and the tension of reconstruction imposed by instrumental rationality on marketplace networks.The study demonstrates that spatial transformation in Chengdu teahouses is not unidirectional alienation but rather a multifaceted configuration where the continuity of tradition coexists with innovative practices amid functional diversification.This research advocates for striking a balance between the preservation of traditional spaces and modern renewal and explores organic integration approaches for traditional and modern elements,thereby providing a theoretical framework and practical insights for the transformation of traditional public spaces.展开更多

文摘This study focuses on the deepening paths of ideological and political construction of professional courses in higher vocational colleges,taking the layout design course of the digital media technology major as the research object,and explores the integration mechanism of excellent traditional Chinese culture and professional teaching.The curriculum’s ideological and political reform is advanced through three major measures:Reconstructing the role of teachers,innovating teaching methods,and building a cultural infiltration system.Combined with the three-dimensional assessment mechanism of“professional+culture+value”and the transformation of teaching achievements into competitions,it breaks through the limitations of traditional skill training and systematically solves the problems of formalization tendency and insufficient value guidance existing in the practice of curriculum ideology and politics.It responds to the era proposition of“promoting the creative transformation of excellent traditional Chinese culture,”and provides an operable solution for higher vocational colleges to break the predicament of“emphasizing skills while neglecting culture.”
文摘The rise of creative industries in this century has aroused strong interest in the concept“cultural capital”in academia.Cultural capital can be understood as various forms of cultural resources that achieve significant appreciation in existing economic,political,and cultural structures through active and rapid reproduction,thereby influencing changes in existing economic,political,and cultural structures,including production relations with capitalist characteristics.The capitalization of culture negates the original ecological value of culture,denies human needs for culture,undermines the true value of cultural exchange and creation,and can easily lead to the formation of cultural hegemony.However,the process of cultural capitalization and the changes in cultural values within it indicate that the capitalization of culture is to some extent an inevitable historical development.The new cultural capital has demonstrated tremendous power in promoting the development of contemporary creative industries and producing cultural masterpieces.Of course,this first requires it to refuse to become a tool for economic capital and digital technology.

文摘BACKGROUND The unique survival environment of Helicobacter pylori(H.pylori)presents challenges in establishing appropriate in vitro culture conditions.The fastidious nature of H.pylori and its relatively low growth rate emphasize the need for customized culture conditions,which are essential for both research and clinical applications.AIM To identify the optimal liquid culture medium for H.pylori using both reference and clinical strains.METHODS Nine H.pylori strains were incubated in 10 different broth media commonly used for bacterial cultures under microaerophilic conditions.Bacterial concentrations were estimated using serial dilutions and the pour plate method.A range of H.pylori initial inoculum concentrations was tested for each strain.For growth evaluation,optical density at 600 nm was measured at 24,48,and 72 hours.The growth trends of H.pylori were compared among strains and media.RESULTS All H.pylori strains grew successfully over time,regardless of the initial inoculum concentration.The specific growth trends and the broth that yielded the highest optical density value at each time point varied with strain.The reference strains(H.pylori 26695,ATCC 43504,and J99)consistently exhibited robust growth,whereas the clinical strains displayed distinct media preferences.Overall,bacterial growth was higher in chopped meat carbohydrate broth,Columbia broth,and fastidious anaerobe broth than in Brucella broth after 72 hours.CONCLUSION Chopped meat carbohydrate broth was the most effective for H.pylori liquid culture,and Columbia broth and fastidious anaerobe broth also supported greater growth than Brucella broth.

文摘Chengdu teahouses,as core public spaces in marketplace society,have undergone transformative reconstruction-from“containers of everyday life”to“containers of commercial traffic and digital flows”-during the process of modernization.Employing spatial archaeology as a methodology,combined with fieldwork and analysis of historical documents,this study systematically examines the diachronic evolution of architectural forms,functional orientations,and social networks within Chengdu teahouses.The study reveals the logic of spatial reconstruction under the interplay of multiple forces,including cultural heritage preservation,capital-driven development,and technological intervention.The findings identify three paradigms of spatial transformation in teahouses.First,heritage specimenization,which reinforces the continuity of collective memory through symbolic extraction but risks diminishing the vitality of everyday social interactions.Second,consumption upgrading,which caters to the demands of emerging groups through iterative business models yet necessitates vigilance against spatial differentiation eroding marketplace inclusivity.Third,digital parasitism,which expands communicative dimensions through technological empowerment but confronts the risk of flattening localized knowledge.These paradigms reflect both adaptive responses of traditional spaces to contemporary pressure and the tension of reconstruction imposed by instrumental rationality on marketplace networks.The study demonstrates that spatial transformation in Chengdu teahouses is not unidirectional alienation but rather a multifaceted configuration where the continuity of tradition coexists with innovative practices amid functional diversification.This research advocates for striking a balance between the preservation of traditional spaces and modern renewal and explores organic integration approaches for traditional and modern elements,thereby providing a theoretical framework and practical insights for the transformation of traditional public spaces.
文摘In recent years,the production-oriented approach has been applied in the field of teaching Chinese as a foreign language,providing a new perspective for language and cultural instruction.Currently,several issues exist in cultural teaching,such as the lack of in-depth cultural input,insufficient training in cross-cultural communication skills,and the over-stylization of cultural teaching in the classroom.The production-oriented approach offers a solution to these challenges.This paper seeks to introduce the production-oriented method into the teaching of Chinese culture as a foreign language,using silk culture as a case study for teaching design.The aim is to implement cultural teaching through a new instructional model and to promote the spread of Chinese silk culture.
文摘An earlier version of this paper has been presented online at the annual Conference of Canadian Sociological Association,during the Congress of Humanities and Social Sciences in Toronto in June 2025.It explores how rap music in Quebec,particularly through the work of the rap artist Webster and his collective Limoilou Starz(LS),serves as a tool for meaning-making,political expression,and resistance against racial profiling and police brutality experienced by Black youth in Quebec City.The study focuses on the song“SPVQ”(Service de police de la Ville de Québec)as a case study to analyze the encoding and reception of anti-racist messages within rap culture.Webster draws from a global tradition of socio-political rap-like that of African-American rappers KRS-One,LL Cool J,and Tupac-while anchoring his critique in the local realities of Limoilou,a marginalized,racially diverse district of Quebec City.Through a narrative,figurative,semantic,and ideological analysis of the song,the paper reveals how Webster articulates themes of police brutality,structural discrimination,economic marginalization,and resistance.The rapper’s message is both a form of testimony and a civic intervention.His broader activism is analyzed through ethnographic techniques-including participant observation and interviews during workshops,media appearances,and online campaigns.It translates these messages into tangible social practices.To explain the persistence of these injustices,the paper situates the issue within Quebec’s interculturalism framework,which ostensibly promotes dialogue and integration but often masks or even reinforces structural racism.This model maintains a symbolic majority/minority duality and conditions inclusion on conformity to a dominant White Francophone identity,thus rendering racialized youth as perpetual outsiders.
文摘Chinese incense culture represents a distillation of historical practices and artistic traditions surrounding the use of aromatic materials.As incense becomes increasingly integrated into contemporary households,its multifaceted significance warrants scholarly exploration.This paper traces the historical evolution of Chinese incense culture-from ancient rituals to its zenith in the Song Dynasty-and examines its modern-day relevance.We identify cultural preservation,health and wellness applications,socioeconomic impacts,and philosophical enrichment as key domains where incense culture exerts positive effects.Findings underscore its role in enhancing life aesthetics,promoting mental well-being,supporting rural economies,and bridging tradition with modernity.The study advocates for further interdisciplinary research to address challenges in heritage conservation and sustainable innovation.
文摘Integrating Chinese culture into classroom teaching is a requirement for the ideological and political construction of courses in institutions of higher learning.Due to its disciplinary attributes,the Comprehensive English course bears the mission of fostering cultural confidence and telling Chinese stories well in English.To fulfill this mission and responsibility of Comprehensive English in terms of Chinese culture,this paper proposes practical paths for integrating Chinese culture into the course.It suggests focusing on textbook texts to explore cultural elements,expanding resources to enhance the representational ability of cultural content,demonstrating diverse cultural investigation methods through in-depth teaching,innovating tasks to improve the communication power of Chinese culture,and emphasizing evaluation to pay attention to the effect of Chinese culture learning.By doing so,Chinese culture can be integrated into teaching,and students’ability to tell Chinese stories well in English can be enhanced.
文摘Using Xiamen as an example,this study investigates the practical route of incorporating Minnan culture into high school English reading instruction.It does this by combining the materials of Compulsory 2 UNIT4 History and Traditions,Compulsory 2 UNIT1 Cultural Heritage,and Compulsory 3 UNIT3 Diverse Cultures,and by creating a series of English reading lessons with a Minnan cultural theme.To enhance students’English reading skills and cultural literacy,southern Fujian cultural elements like Nanyin,lacquer thread carving,and Pok Pok were incorporated into the classroom through the use of games,pre-reading activities,reading activities,and post-reading activities.The results of the study show that the integration of southern Fujian culture into English reading teaching is feasible and effective,can stimulate students’learning interest,enhance the sense of cultural identity,and provide practical references for cultural integration in English teaching.
文摘This research examines the use of digital learning to preserve and promote Cambodian culture in universities.As benefits and challenges of globalization and modernization abound,digital learning along with the quickening pace of social advancement provides new opportunities for students and faculty to sustain cultural heritage,enhance student engagement,and create inclusive education.This study explores two questions:1)how does foreign culture appear in Cambodian English textbooks and 2)how might digital learning be used to co-decolonize in a way creates gender and cultural inclusivity?The study adopts an analysis-by-synthesis method and dissects and synthesizes the various components—technology infrastructure,faculty preparedness,student engagement,and cultural content—how they interacted with one another,and how they interacted with each representative group of stakeholders.The results indicate that digital tools provide an opportunity for both higher cultural awareness and accurate representation,and gender equity.Challenges exist in accessibility to the technology infrastructure and the authentic cultural content in the digital space.The study concludes with a discussion of how to scale faculty professional development,expand the curriculum,and collaborate regionally in interoperable scaffolding in order preserve Cambodia’s cultural identity in the quickly evolving landscape of education.

文摘In order to further explore the rich connotation of Huai'an historical and cultural space and the Grand Canal culture,the synchronicity of urban spatial pattern and historical buildings of Huai'an was studied under the guidance of synchronic theory and method.It is found that there are the identity and reality law of the development and change of Huai'an historical and cultural space and the Grand Canal cultural system,and it is deduced that there is a spiraling systematic structure chain between the historical and cultural space of Huai'an and the Grand Canal culture,which provides a theoretical reference for promoting the deepening construction of regional culture,the Grand Canal cultural belt and national cultural parks.

文摘Since the second half of the 20th century,the concept of global green development has gained increasing global attention,with the harmonious coexistence between humans and nature emerging as a critical issue.The cultural landscape of Jingmai Mountain old tea forest,as a typical representative of ecological wisdom in agricultural civilization,showcases a sustainable practice in tea production and livelihood system.Employing a combination of field investigation and literature analysis,this study systematically examines the ecological wisdom of the“forest-tea coexistence”system in Jingmai Mountain from 5 dimensions:tea forest cultivation,land use,village construction,residential buildings,and ethnic culture.The results indicate that Jingmai Mountain has established an integrated system of“ecological technology-spatial planning-cultural institution”for synergistic development:a multi-layered planting structure and vertical gradient development mode have enabled efficient resource utilization;The village layout of“Following the mountain terrain and close to the tea forest”and the climate-adaptive architecture reflect the harmonious unity of living environment and the natural environment;the belief in“Tea Spirit”and community-based governance mechanism ensure the institutionalization and sustainability of ecological protection.This system transforms traditional ecological knowledge into modern practices,providing important reference for the protection of agricultural cultural heritage and serving as a model for ecological protection and rural revitalization in the context of global green development.

文摘The Bashu region is not only the cultural center of southwestern China but also an important cultural corridor for the interaction among various ethnic groups.Under the influence of multi-ethnic interactions,geographical conditions,and historical changes,its music culture has developed a distinct characteristic of symbiotic integration,reflecting a cultural pattern of“diversity within unity.”Bashu music not only inherits the aesthetic system of Han culture but also deeply incorporates musical elements from minority ethnic groups,such as Tibetan,Qiang,Yi,and Miao.It showcases cultural characteristics that are diverse in tone,variable in rhythm,and complex in musical form.Music,as a medium of cultural identity,reflects the inclusiveness and sustainability of Bashu culture.In the context of globalization,studying Bashu music culture from the perspective of musical geography not only helps enhance our understanding of the Bashu music culture integration mechanism but also provides a theoretical reference for the holistic study of China’s multi-ethnic music culture.This research holds significant practical importance for the construction of ethnic cultural identity,the protection of intangible cultural heritage,and cultural exchanges in the context of the Belt and Road Initiative.
