The infiltration of immune cells into the hepatocellular carcinoma microenvironment is the main reason why hepatocellular carcinoma patients are prone to carcinoma recurrence and the disease are incurable.Notably,the infiltration of Treg cells is the main trigger.Dahuang Zhechong pill(DHZCP)is a traditional Chinese herbal compound successful in the treatment of hepatitis and hepatocellular carcinoma.DHZCP can heal and nourish while slowing the onset of the disease,thereby strengthening the body’s immune function.It can localize tumors and ultimately achieve the goal of eliminating tumors.In this study,an orthotopic liver cancer model of mice was used to explore the mechanism of DHZCP enhancing anti-tumor immunity,which showed more Th1 cells in the peripheral blood and spleen after DHZCP treatment,while more IFN-γwas secreted to activate CD8^(+)T cells and Treg cell production was inhibited,thereby suppressing the growth of HCC.Finally,we also analyzed the potential components of DHZCP from the perspective of modern targets using network pharmacology methods and experimental results. 展开更多

AIM:To develop a high performance liquid chromatography(HPLC)coupled with electrospray ionization mass spectrometry(ESI-MS)and ultraviolet(UV)detector method for the acid-alkaline simultaneous determination of ten bioactive compounds,and analyze the effect of compatible medicinal plants on the concentration of components in Dahuang Fuzi Tang(DFT).METHOD:The chromatographic separation was performed on a Hypersil BDS C18 analytical column by gradient elution with acetonitrile and formate buffer(containing 0.15%formic acid,V/V)at 25°C with a flow rate of 1.0 mL·min–1 and UV detection at 280 nm.Four of the ten compounds in DFT were identified and their MS fragments were elucidated by HPLC-ESI-MS,and the contents of the six compounds were determined by HPLC-UV.RESULTS:All calibration curves showed good linear regression(r2≥0.9990).The limits of detection and limits of quantification were 0.021–0.155μg·mL–1 and 0.076–0.520μg·mL–1,respectively.Overall precision RSD(intra-day and inter-day)were less than 2.96%,and the average recoveries were 98.35%–101.45%,with RSD ranging from 1.54%to 3.01%for the analytes.CONCLUSION:The developed method can be applied for the quality control and provide analytical evidence on the chemical basis and combinational principles of DFT. 展开更多

Objective:To investigate the protective effect of Dahuang Fuzi Decoction(DHFZD),a traditional Chinese prescription,at alleviating sepsis-induced inflammation and gut barrier damage in rats.Methods:Forty clean-grade male Sprague-Dawley rats were divided randomly into three groups:normal control group(NCG,n?10),model control group(MCG,n?15)and DHFZD-treated group(DHFZDG,n?15).NCG rats were sham operated on and used as the controls,whereas MCG and DHFZDG rats were used to replicate the rat sepsis model using cecal ligation and puncture(CLP).The DHFZDG rats received DHFZD by gavage(4.5 mg/g of body weight)2 h prior to CLP and after its successful induction,while the NCG and MCG rats received equivalent amounts of sterilized water by gavage.All rat groups were starved and had free access to water.At 24 h post-experimental set up,the mortality of rats in each group was recorded,and peritoneal inflammation assessment and pathological changes related to the intestinal mucosal injury index(IMII)in the surviving rats were evaluated.D-lactic acid,tumor necrosis factor(TNF)-a,interleukin(IL)-6 and IL-10 peripheral blood concentrations,along with secretory immunoglobulin A(sIgA)in the intestinal mucosa were evaluated by enzyme-linked immunosorbent assays.Gut microbes were detected using 16S rRNA gene sequencing.Results:DHFZD reduced sepsis-related mortality in the rats.Moreover,it alleviated peritoneal inflammation and pathological changes according to the IMII.DHFZD reduced serum procalcitonin,TNF-a and IL-6 concentrations,but not the IL-10 concentration.It also reduced serum D-lactic acid and increased sIgA concentrations in intestinal mucosa.Notably,DHFZDG restored gut microbiota diversity and regulated the decrease in Bacteroidetes induced by sepsis,compared with the MCG rats.Conclusion:DHFZDG may play a protective role in sepsis by alleviating sepsis-induced inflammation and gut barrier damage in rats. 展开更多

[Methods]86 cases of acute pancreatitis were randomly divided into the control group( 43 cases) and the treatment group( 43 cases). The control group was given saline enema and routine treatment of western medicine,while the treatment group was treated with Dahuang( Rhubarb) decoction retention enema and routine treatment,2 times/d. After 7 days of treatment,two groups of clinical curative effects were compared. And changes of IL-6,hs-CRP,endotoxin and amylase levels before and after treatment in two groups were compared. [Results] Abdominal pain,abdominal distension,exhaust defecation,bowel sounds in treatment group patients were obviously improved. After treatment,the decline levels of serum IL-6,hsCRP,endotoxin and amylase in the treatment group were significantly better than those in the control group. [Conclusions] Dahuang( Rhubarb) retention enema can significantly improve the clinical curative effect of severe acute pancreatitis,and its mechanism may be related to the inhibition of inflammatory reaction. 展开更多

OBJECTIVE: To explore the material basis of the difference of efficacy of Dahuang(Radix Et Rhizoma Rhei Palmati)-Taoren(Semen Persicae)(DT) drugs with different proportions. METHODS: Samples of different ratios of Dahuang(Radix et Rhizoma Rhei Palnati, DH) to Taoren(Semen Persicae, TR)(Group A 1∶1, B 2∶3, C 3∶2) were analyzed based on gas chromatography time-of-flight mass spectrometry untargeted metabolomics technique. RESULTS: A total of 240 primary metabolites were detected. Forty-one differential metabolites involved nine differential metabolic pathways, of which four were closely related to the efficacy of DT in the treatment of heat and blood stasis syndrome. These pathways included the biosynthesis of amino acid(phenylalanine tyrosine and tryptophan), flavonoids, unsaturated fatty acids, and the glycolysis/glycogenesis pathway. CONCLUSION: There are significant differences in the efficacy of different ratios of DT drugs, and their optimal ratio for the treatment of heat and blood stasis syndrome should be 1∶1. 展开更多

Methods: The main targets of the active ingredients of RERR and SP were filtered based on the traditional Chinese medicine system pharmacology analysis platform. Cytoscape 3.2.1 was applied to build the ingredient-target network of RERR and SP for AIO. Results: Fifteen active components were predicted from the RERR and SP herb pair, such as aloe-emodin, catechin, rhein, gibberellin (GA) 119, GA120 and GA121. These components were applied to 59 targets mainly involved in many biological processes such as signal transduction, anti-apoptosis, and inflammatory response involved in activating the immune effect. Conclusion: This study proposes the system pharmacology method and identifies the potent combination therapeutic mechanism of RERR and SP for AIO. OBJECTIVE:To investigate the effects of a modified Dahuang Zhechong Pill(MDZP) on the angiogene sis of rhesus choroid-retina endothelial(RF/6A cells and its preliminary mechanism.METHODS:A 3-(4,5-dimethylthiazol-2-yl)-5-(3-car boxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazol ium(MTS) method was used to assess the effect o a MDZP on RF/6A cell proliferation induced by vas cular endothelial growth factor(VEGF).Transwell in serts were used to assess the effect of the MDZP on RF/6A cell migration.Matrigel was used to asses the effect of the MDZP on the tube formation of RF 6A cells.Western blotting and quantitative re al-time reverse transcription polymerase chain reac tion(RT-PCR) were used to detect the protein and mRNA expression,respectively,of VEGF and matri metalloproteinase-2(MMP-2) in RF/6A cells treatedwith the MDZP.RESULTS:RF/6A cell proliferation induced by VEGF was inhibited by 0.2 mg/mL MDZP.At 0,12.5,25 and 50 mg/mL MDZP,the number of cells that migrated through Transwell membranes was 73.33± 4.51,61.33±4.04,28.67±6.66 and 17.67±4.16,respectively,and the number of tubes formed in Matrigel was 20.33±0.58,13.33±1.53,11.00±1.00 and 1.33±0.58,respectively.At 100 and 200 mg/mL MDZP,the protein and mRNA expression of VEGF and MMP-2 were inhibited in RF/6A cells.At 400 mg/mL MDZP,the expression of VEGF mRNA and MMP-2 protein were inhibited in RF/6A cells.CONCLUSIONS:MDZP inhibits the angiogenesis of RF/6A cells via the suppression of proliferation,migration and tube formation of RF/6A cells.Inhibition of the protein and mRNA expression of VEGF and MMP-2 in RF/6A cells may be an important mechanism. 展开更多

Objective: To investigate the mechanism of inhibiting the secretion of “lithogenic bile” and treating gallbladder distention with the TCM prescription Dahuang Lingxian. Methods: Forty SPF-grade SD male mice were randomly divided into blank control group, model group, ursodeoxycholic acid group and TCM group, 10 mice in each group. The blank control group was kept normally, and the model group, ursodeoxycholic acid group and TCM group were established as gallbladder distention models. The blank control group and the model group were given 1.5 ml of saline by gavage daily;the ursodeoxycholic acid group was given 100 mg/kg/d of a mixture of saline and ursodeoxycholic acid capsules by gavage daily;the TCM group was given 39 g/kg/d of aqueous decoction of Dahuang Lingxian Prescription by gavage daily. After 6 weeks of continuous intervention, all mice were executed and the lithogenesis rate, ultrastructure and mRNA expression were observed. Results: The lithogenesis rate of mice in the TCM group was significantly reduced (P β1/Smads pathway was significantly improved (P < 0.05), which could achieve the same therapeutic effect as ursodeoxycholic acid capsules. Conclusion: Dahuang Lingxian Prescription can reduce the secretion of “lithogenic bile” by inhibiting the inflammatory reaction and fibrosis of biliary system. Dahuang Lingxian Prescription has certain advantages in the treatment of gallbladder distention. 展开更多

Objective:To observe the effect of lipopolysaccharide(LPS)on cytoskeleton and the effect of Dahuang Lingxianfang on nF-KB/MAPK signaling pathway.Methods:Biliary epithelial cells of each group were stained with photolipin fluorescent staining,and the arrangement of cytoskeleton was observed under laser confocal microscope.Western blotting was used to detect the expression of f-actin.Results:After LPS intervention,the biliary epithelial cells showed nuclear shrinkage or damage,and the skeleton was broken or lumped.The cytoskeleton was partially repaired after the intervention of pathway blocking preparation combined with RHUbarb Lingxianfang.All the other groups had different degree of cytoskeleton fracture.Compared with normal group,the expression of F-actin protein in LPS group was decreased(P<0.05);Compared with LPS group,the expression of F-actin in LPS+TCM group,LPS+PDTC+TCM group,LPS+SB203580+TCM group and LPS+PDTC+SB203580+TCM group was significantly increased(P<0.05);Compared with traditional Chinese medicine group,the expression of F-actin in LPS+PDTC+traditional Chinese medicine group,LPS+SB203580+traditional Chinese medicine group and LPS+PDTC+SB203580+traditional Chinese medicine group had no significant difference(P>0.05).Conclusion:RHUbarb Lingxianfang can restore the sequence of biliary epithelial cytoskeleton and protect its microfilament structure under inflammation,and its mechanism may be related to the regulation of NF-KB/MAPK signaling pathway. 展开更多

Objective:To explore the mechanism of action of Dahuang Lingxian Formula in alleviating the inflammatory response of bile duct cells in LPS-induced intrahepatic bile duct inflammation model rats based on IL-6/JAK/STAT3 signaling pathway.Methods:Fifty SD rats were randomly divided into five groups,blank group,model group,choling tablets(0.5 g/kg),and low and high concentration groups(2.4 g/kg and 4.8 g/kg)of Dahuang Lingxian Formula,ten rats in each group.Except for the blank group,the rats in each group were injected with 1.25 mg/kg LPS at the common bile duct at one time to construct an animal model of intrahepatic bile duct infection.After gavage on day 8,liver tissues were taken from rats at the hepatic hilum,and the histopathological changes of the hepatic hilum and biliary tree were observed by HE staining.The expression levels of serum glutamic alanine transaminase(ALT),glutamic oxalacetic transaminase(AST),malondialdehyde(MDA)and superoxide dismutase(SOD)were measured by biochemical method.The expression levels of interleukin 6(IL-6),Janus protein tyrosine kinase 2(JAK2),signal transducer and activator of transcription 3(STAT3)in rat serum were measured by enzyme-linked immunosorbent assay(ELISA).Protein immunoblotting(WB)and real-time fluorescence quantitative PCR(RT-qPCR)were used to detect the expression levels of IL-6,JAK2,STAT3 protein and mRNA in biliary tree tissues.Results:①Compared with the blank group,the structures such as interlobular bile ducts in the hepatic sinusoids and portal duct area of the model rats were destroyed,and inflammatory cells infiltrated around them.The expression of ALT,AST,MDA,IL-6,JAK2 and STAT3 in the serum increased significantly,the expression level of SOD decreased,and the expression levels of IL-6,JAK2 and STAT3 proteins and mRNA increased.②Compared with the model group,the degree of liver pathological damage in rats in the Chiling Ning tablet group and the low and high concentration groups of Dahuang Lingxian Formula were improved,which could significantly reduce the expression levels of ALT,AST,MDA,IL-6,JAK2,STAT3 and up-regulate SOD in serum,and down-regulate the expression of IL-6,JAK2,STAT3 protein and mRNA,with the best effect in the high concentration group of Dahuang Lingxian Formula.③Compared with the choling tablet group,the rats in the low and high concentration groups of Dahuang Lingxian Formula tended to normalize the degree of liver pathological damage,without obvious inflammatory cell infiltration,and the expression levels of ALT,AST,MDA,IL-6,JAK2,STAT3 and the expression levels of IL-6,JAK2,STAT3 protein and mRNA in serum were reduced,and the expression levels of SOD were increased,with the best effect of Dahuang Lingxian Formula The treatment effect was best in the high concentration group.Conclusion:The mechanism may be related to the down-regulation of IL-6/JAK/STAT3 signaling pathway activation,and the best therapeutic effect was achieved by the high concentration group of Dahuang Lingxian Formula. 展开更多

[Objectives]To treat patients with Lower-Jiao Obstruction Syndrome caused by severe pneumonia complicated with heart failure using the retention enema with Modified Dahuang Mudan Decoction,and to observe the effect and safety of this treatment on inflammatory indicators,cardiac function,gastrointestinal function and multiple organ failure in patients with Lower-Jiao(lower energizer)Obstruction Syndrome caused by severe pneumonia complicated with heart failure.[Methods]54 patients with Lower-Jiao Obstruction Syndrome caused by severe pneumonia complicated with heart failure diagnosed in Nanchong Chinese Medicine Hospital from January 2020 to December 2021 were randomly divided into experimental group(n=27)and control group(n=27).The control group was given conventional treatment combined with 0.9%sodium chloride injection(100 mL)+glycerine enema(40 mL).The treatment group was treated with Modified Dahuang Mudan Decoction on the basis of conventional treatment for 7 d.The efficacy indicators and adverse events such as white blood count(WBC),C-reactive protein(CRP),procalcitonin(PCT),N-terminal pro-brain natriuretic peptide(NT-proBNP),Lausanne Intestinal Failure Estimation(LIFE)and multiple organ dysfunction score(MODS)were observed.[Results](i)There was no significant difference in WBC,PCT,CRP and NT-proBNP between the two groups before treatment.All indicators after treatment were significantly lower than those before treatment in both groups(P<0.05).The levels of WBC,PCT and CRP in the treatment group were lower than those in the control group after treatment(P<0.05),and there was no significant difference in NT-proBNP between the two groups after treatment(P>0.05),but the difference in NT-proBNP before and after treatment in the treatment group was higher than that in the control group.(ii)The LIFE of the two groups decreased significantly after treatment,and the improvement of LIFE of the treatment group was better than that of the control group(P<0.05).(iii)The MODS of the two groups decreased significantly after treatment(P<0.05),and the degree of decrease after treatment in the treatment group was higher than that in the control group(P<0.05).(iv)There was no significant difference in all-cause mortality between the two groups(P>0.05).(v)No serious adverse events were found in both groups during the treatment period.[Conclusions]The conventional treatment,combined with the retention enema with Modified Dahuang Mudan Decoction could further reduce the inflammatory indicators,improve cardiac function,improve intestinal function and organ function,and the safety was good. 展开更多

Objective:To study the effect of Dahuang Zhechong pill combined with antiviral therapy on serum virus replication indexes as well as immunity and inflammation indexes in patients with chronic hepatitis B cirrhosis.Methods: A total of 104 patients with chronic hepatitis B cirrhosis who were treated in our hospital between May 2013 and April 2016 were selected and randomly divided into combined treatment group and western medicine treatment group who accepted Dahuang Zhechong pill combined with entecavir treatment and entecavir monotherapy respectively. Before and after treatment, the HBV-DNA loads, liver fibrosis index and immune cell levels as well as inflammatory cytokine levels were determined.Results:HBV-DNA loads of both groups of patients at 48 weeks after treatment were significantly lower than those before treatment, and the HBV-DNA loads were not significantly different between the two groups after treatment;serum HA, LN, PC-III, IV-C, IL-15, IL-16 and TGF-β1 levels as well as peripheral blood Treg cell levels of both groups at 24 weeks and 48 weeks after treatment were significantly lower than those before treatment while peripheral blood NK cell levels were higher than those before treatment, and serum HA, LN, PC-III, IV-C, IL-15, IL-16 and TGF-β1 levels as well as peripheral blood Treg cell levels of combined treatment group were significantly lower than those of western medicine treatment group while peripheral blood NK cell levels were higher than those of western medicine treatment group. Conclusion:Dahuang Zhechong pill combined with antiviral treatment of chronic hepatitis B cirrhosis can regulate the immune response and inflammatory reaction without affecting the inhibiting effect of antiviral drugs on viral replication. 展开更多

Objective To explore the preventive and therapeutic effects of Dahuang Zhechong Pill(DZP)on pulmonary fibrosis and the underlying mechanisms.Methods The first key rate-limiting enzyme hexokinase 2(HK2)of glycolysis was silenced and over-expressed through small interfering RNA and lentivirus using lung fibroblast MRC-5 cell line,respectively.The cell viability,migration,invasion and proliferation were detected by cell counting kit-8,wound healing assay,transwell assay,and flow cytometry.The mRNA and protein expression levels of HK2 were detected by RT-PCR and Western blotting,respectively.The contents of glucose,adenosine triphosphate(ATP)and lactate in MRC-5 cells were determined by enzyme-linked immunosorbnent assay(ELISA).Then,the relationship between miR-29b-2-5p and HK2 was explored by luciferase reporter gene assay.Pulmonary fibrosis cell model was induced by transforming growth factor-β1(TGF-β1)in MRC-5 cells,and the medicated serum of DZP(DMS)was prepared in rats.MRC-5 cells were divided into control,TGF-β1,TGF-β1+10%DMS,TGF-β1+10%DMS+miR-29b-2-5p inhibitor,TGF-β1+10%DMS+inhibitor negative control,TGF-β1+10%DMS+miR-29b-2-5p mimic and TGF-β1+10%DMS+mimic negative control groups.After miR-29b-2-5p mimics and inhibitors were transfected into MRC-5 cells,all groups except control and model group were treated with DMS.The effect of DMS on MRC-5 cells were detected using aforementioned methods and immunofluorescence.Similarly,the contents of glucose,ATP and lactate in each group were measured by ELISA.Results The mRNA and protein expressions of HK2 in MRC-5 cells were successfully silenced and overexpressed through si-HK2-3 and lentiviral transfection,respectively.After silencing HK2,the mRNA and protein expressions of HK2 were significantly decreased(P<0.01),and the concentrations of glucose,ATP and lactate were also significantly decreased(P<0.05).The proliferation,migration and invasion of MRC-5 cells were significantly declined(P<0.05 or P<0.01),while the apoptosis of MRC-5 cells was significantly increased(P<0.01).After overexpressing HK2,the mRNA and protein expressions of HK2 were significantly increased(P<0.05),and the concentrations of glucose,ATP and lactate were also significantly increased(P<0.05 or P<0.01).The proliferation,migration and invasion of MRC-5 cells were significantly increased(P<0.05 or P<0.01),while the apoptosis of MRC-5 cells was significantly decreased(P<0.05).The relative luciferase activity of 3’UTR-WT+hsa-miR-29b-2-5p transfected with HK2 was significantly decreased(P<0.01).After miR-29b-2-5p mimic and inhibitor were transfected into the MRC-5 cells,DMS intervention could significantly reduce the concentration of glucose,ATP and lactate,and the mRNA and proteins expressions of HK2,phosphofructokinase and pyruvate kinase isoform M2(P<0.05 or P<0.01).The proliferation,migration and invasion of MRC-5 cells were alleviated(P<0.05 or P<0.01),and the deposition of fibronectin,α-smooth muscle actin,and collagen I were significantly decreased(P<0.05 or P<0.01).Conclusions Glycolysis is closely related to pulmonary fibrosis.DZP reduced glycolysis and inhibited fibroblasts’excessive differentiation and abnormal collagen deposition through the miR-29b-2-5p/HK2 pathway,which played a role in delaying the process of pulmonary fibrosis. 展开更多

Cancer still has elevated morbidity and mortality,which undoubtedly impacts the life quality of affected individuals.Remarkable advances have been made in cancer therapy,although the toxicities of traditional therapies remain an obvious challenge.Dahuang Zhechong Pill(DHZCP),developed by Zhongjing Zhang in the Synopsis of the Golden Chamber,represents an effective anticancer traditional Chinese medicine(TCM).In this review,it was found that DHZCP is therapeutically utilized in liver,lung,gastric,pancreatic and other cancers in clinic.Pharmacological evidence showed that its anti-tumor mechanisms mainly involve induced cell cycle arrest,apoptosis and autophagy,as well as suppressed tumor cell proliferation,obstructed angiogenesis and metastasis,enhanced immunity,and reversal of multidrug resistance.The present review provides a solid basis for the clinical application of DHZCP and may promote the wide use of TCM in clinical antitumor application. 展开更多

目的:探讨大黄灵仙方对胆囊结石豚鼠模型肠道菌群结构与功能的影响。方法:50只豚鼠按随机数字表法分为空白组15只、模型组20只、中药组15只。空白组给予豚鼠正常饲料喂养,每日予以0.9%生理盐水灌胃;模型组给予高脂致石饲料喂养,每日予以... 目的:探讨大黄灵仙方对胆囊结石豚鼠模型肠道菌群结构与功能的影响。方法:50只豚鼠按随机数字表法分为空白组15只、模型组20只、中药组15只。空白组给予豚鼠正常饲料喂养,每日予以0.9%生理盐水灌胃;模型组给予高脂致石饲料喂养,每日予以0.9%生理盐水灌胃;中药组给予高脂致石饲料喂养,每日予以1 g/m L大黄灵仙方灌胃。共干预8周。收集第8周的结肠粪便,利用16SrDNA测序分析肠道菌群的组成、多样性及功能预测。结果:(1)空白组OTUs数为4623,模型组为3970,中药组为4335;3组共有OTUs为474;(2)主要优势菌门包括Bacteroidetes、Firmicutes、Proteobacteria、Verrucomicrobia和Spirochaetes;模型组中Bacteroidetes和Verrucomicrobia增加,而Firmicutes和Spirochaetes减少,中药组Bacteroidetes、Firmicutes、Verrucomicrobia减少,但Proteobacteria和Spirochaetes增加;(3)各组间肠道菌群的多样性指数(Ace、Chao1、Shannon、Simpson)无明显差异(P>0.05);(4)模型组与空白组菌群分布差异显著(P<0.01),中药组肠道菌群部分重叠于模型组;(5)空白组中Ruminococcus、Rikenellaceae和其他属高表达,模型组Paludibacteraceae和Verrucomicrobia表现突出,中药组Alphaproteobacteria和Desulfovibrio高表达;(6)空白组肠道菌群呈厌氧和生物膜合成特征,模型组中革兰氏阴性菌增加,可移动基因原件和生物膜合成减少,而中药组则表现出革兰氏阴性和生物膜合成特征的恢复。结论:胆囊结石形成过程中肠道菌群结构变化明显,而大黄灵仙方能够有效调节菌群趋向正常稳态。 展开更多

该研究通过文献资料收集整理结合文献计量学的方法对大黄附子汤的源流、方义衍变、基原、剂量与煎服法等信息进行全面分析考证。大黄附子汤首载于东汉张仲景所撰的《金匮要略》,除《邹氏寒疫论》中记载大黄、芒硝、生附子、枳实,余均沿... 该研究通过文献资料收集整理结合文献计量学的方法对大黄附子汤的源流、方义衍变、基原、剂量与煎服法等信息进行全面分析考证。大黄附子汤首载于东汉张仲景所撰的《金匮要略》,除《邹氏寒疫论》中记载大黄、芒硝、生附子、枳实,余均沿用仲景原方,由大黄、附子(炮)、细辛组成。经考证后仲景大黄附子汤药物的剂量折合成现代剂量为大黄41.4 g、附子30 g、细辛27.6 g。以上三药的基原均与2020年版《中华人民共和国药典》一致。对煎服方法考证可知,建议三药加水1000 mL,煎至400或500 mL,均分日3次温服。古代应用涉及大多为胁痛、腹痛、便秘、寒疝等疾病,以寒积里实证为主,现代临床应用涉及泌尿、消化等多系统疾病。药理研究证实其通过调节胃肠功能、抑制肾纤维化、调节神经与免疫、减轻炎症反应等机制发挥作用。通过分析大黄附子汤的古籍记载、现行标准及临床研究,明确其关键信息,为经典名方的临床应用和新药研发提供依据。 展开更多

Objective: To investigate effects of Dahuang Zhechong Pill (大黄庶虫丸, DHZCP) on the cell cycle and the related signal pathways in vascular smooth muscle cells (VSMCs) stimulated by platelet-derived growth factor (PDGF) with the method of serum pharmacology. Methods: DNA synthesis in VSMCs was examined by detecting 5'-bromo-2'-deoxyuridine incorporation with the immunocytochemical method. The cycle of VSMCs was evaluated with flow cytometry. Expressions of cyclin D1, p27, protein kinase C α (PKC α), and phosphorylated extracellular signal regulated kinase 1/2 (ERK1/2) were quantified by Western blot method. Results: DHZCP containing serum significantly inhibited DNA synthesis of PDGF-stimulated VSMCs, arrested the cells in G1 phase, modulated the protein expressions of cyclin D1 and p27, and suppressed the activation of PKC α and ERK1/2. Conclusion: DHZCP containing serum inhibits VSMCs proliferation via modulating the expressions of cell cycle proteins to arrest the cell in G1 phase, which is attributed to, at least in part, suppressing PKC α -ERK1/2 signaling in VSMCs. 展开更多

Objective: To investigate and compare the effects and mechanisms of three functional parts of Dahuang Zhechong Pill (大黄〈庶虫〉虫丸, DHZCP), including drugs with the function of removing blood stasis and promoting blood circulation (FP- I ), drugs with the function of expelling heat and moistening dryness (FP-II), and drugs with the function of nourishing yin and replenishing blood (FP-m) of DHZCP, on platelet-derived growth factor (PDGF)-stimulated vascular smooth muscle cells (VSMCs) proliferation with the method of serum pharmacology. Methods: VSMCs proliferation of rat was assayed by measuring the cell viability with the 3-(4, 5-dimethylthiazol-2yl)-2, 5-diphenyltetrazolium bromide (Ml-I') method. DNA synthesis in VSMCs was examined by detecting 5'-bromo-2'-deoxyuridine incorporation with the immunocytochemical method. Cycle of VSMCs was evaluated with flow cytometry. Expression of cyclin D1, p27, PKC a, and phosphorylated extracollular signal regulated kinase 1/2 (ERK1/2) was quantified by the Western blotting method. Results: The FP- I and FP-m containing serum was capable of inhibiting PDGF-stimulated proliferation and DNA synthesis of VSMCs, arrested VSMCs in G1 phase, downregulated cycUn D1, and upregulated p27 expression (P〈0.01 or P〈0.05). The FP- I and FP-11I containing serum also inhibited the PDGF-induced phosphorylation of tyrosine of ERK1/2 and PKCo~ expression (P〈0.01 or P〈0.05). Conclusions: FP- I and FP-$ of DHZCP are able to inhibit VSMCs proliferation via interrupting PKC a-ERK1/2 signaling, modulating the expression of cell cycle proteins to result in arresting the cells in G1 phase. The inhibitory effect is mainly related to the function of removing blood stasis and promoting blood circulation, slightly to the function of nourishing yin and replenishing blood, but not to the function of expelling heat and moistening dryness. 展开更多

Whereasin the control group, the symptoms were not significantly improved, hemoglobin, levels of sex hormonesand creatine phosphokinase differed slightly.
