Parkinson's disease(PD)is a debilitating and progressive neurodegenerative disorder with complex pathology and multiple membrane barriers that hinder drug delivery,resulting in the absence of ideal therapeutic dru...Parkinson's disease(PD)is a debilitating and progressive neurodegenerative disorder with complex pathology and multiple membrane barriers that hinder drug delivery,resulting in the absence of ideal therapeutic drugs with minimal side effects[1].Recently,natural medicine has garnered significant attention due to its remarkable efficacy and limited side effects.Icariin,a naturally occurring flavonoid,exhibits excellent potential as a therapeutic agent for neurodegenerative disease[2].However,its clinical application is limited by poor water solubility,low bioavailability,and high clearance rates.Here,an aqueous formulation of icariin/hydroxypropyl-β-cyclodextrin(HP-β-CD)supramolecular inclusion complex was obtained to optimize icariin properties.Furthermore,icariin cyclodextrin supramolecular inclusion complex based thermosensitive(icariin gels)was innovated.At lower temperatures,it remained in a liquid phase with high fluidity.Upon reaching the gelation temperature,it underwent a transition to a gel phase with significantly reduced fluidity,which may be suitable for the design of an intranasal delivery system for PD treatment.The innovative approach capitalizes on the exceptional characteristics of HP-β-CD,which was utilized to synergize with nasal delivery for targeted brain delivery and with icariin for PD treatment[3,4].展开更多

Methods QURC-HP-β-CD inclusion complexes were prepared by the grinding method. The equilibrium inclusion constants and thermodynamic parameters were determinated by phase solubility analysis. Dissolution tests were performed to study the dissolution rate of inclusion complexes. The formation of inclusion complexes was confirmed by differential scanning calorimetry ( DSC), infrared spectroscopy (IR) , powder X-ray diffractometry (PXRD) and scanning electron microscopy (SEM). Results The aqueous solubility of quercetin was greatly increased ( about 37 folds) by inclusion technique, and the initial dissolution rate was markedly improved (10 folds) in the first 5 min. The results of DSC and SEM photographs showed that quercetin crystal disappeared in inclusion complexes, which indicated the formation of new phase. FT-IR spectra showed that the carbonyl quercetin crystal grinding method. absorption band of quercetin was shifted. PXRD showed that the diffraction peak of disappeared. In our previous study,a novel organic selenium compound Eb was synthesized and found to have significant antitumor activity with much less toxicity compared with the leading compound Ebselen.Unfortunately,Eb was pract...In our previous study,a novel organic selenium compound Eb was synthesized and found to have significant antitumor activity with much less toxicity compared with the leading compound Ebselen.Unfortunately,Eb was practically insoluble in water (2.57 μg/mL) and had very low oral bioavailability,thus its clinical application was greatly limited.In the present study,the inclusion complex of Eb with 2-hydroxypropyl-β-cyclodextrin (HP-βCD) was prepared and pharmacokinetics of Eb and the inclusion complex were investigated.The water solubility of Eb was dramatically enhanced by inclusion with HP-βCD,which reached 8.4 mg/mL.The pharmacokinetic study showed that the elimination half-life (t 1/2β) of Eb was between 22 h and 30 h and the distribution half-life (t 1/2α) of Eb was 1 h.The results indicated that Eb was rapidly distributed to tissues but slowly eliminated in rats.The absolute bioavailability of Eb/HP-βCD inclusion complex solution through the oral route was 28.3%,and it was 1552% that of Eb in its pure form.In summary,the absorption of Eb in the Eb/HP-βCD inclusion complex was better and faster than that of Eb in its pure form.展开更多

Basing on programmed-heating procedure and weight-temperature analysis, as well as the application of Satava-Sestak's, Ozawa's and Kissinger's methods, the mechanism and kinetics of thermal dissociation of this inclusion complex were studied. An additional mass loss is found at 170-180℃. The mechanism of thermal dissociation of CD-MCP is dominated by a one-dimensional random nucleation and subsequent growth process (A2/3). The activation energy Es and the pre-exponential factor AS for the process are 102.14 kJ/mol and 3.63×10^10s^-1, respectively. The triblock polymer was prepared by reversible addition-fragmentation...The effect of temperature-responsive solution behavior of PNIPAM-b-PPEOMA-b-PNIPAM on its inclusion complexation with α-cyclodextrin was studied. The triblock polymer was prepared by reversible addition-fragmentation chain transfer (RAFT) polymerization and formed inclusion complexes (ICs) after selective threading of the PEO segment of the triblock polymer through the cavities of α-CD units. For comparison, PPEOMA homopolymer was prepared and the inclusion complexation with α-CD was also studied. The ICs were prepared with a-CD when the polymer was in different conformations by changing the temperature, and the formed ICs were characterized by XRD, ^1H-NMR, TGA and DSC. The solutions of the ICs show temperature-responsive clear/turbid transition or fluidic emulsion/gel transition depending on the concentration of the α-CD added, and the stoichiometry determined by ^1H-NMR and TGA indicates that the stoichiometry of EO to α-CD of the resulted ICs increases with increasing of temperature.展开更多

Supramolecular hydrogels based on ...A series of monomethoxy poly(ethylene glycol) (mPEG) grafted copolyglutamates (PmPEGs) were synthesized through ring-opening polymerization (ROP) followed by click chemistry. Supramolecular hydrogels based on polymer inclusion complexes (ICs) between PmPEG and α-cyclodextriri (α-CD) were prepared in aqueous solution. The rheological measurements indicated their gelation properties were affected by several factors including the mPEG length, graft density and the sample concentration. These hydrogels displayed thermo-sensitive gel-sol transition under appropriate conditions due to the reversible mPEG and α-CD inclusion complexation. These hydrogels also showed pH-sensitive behavior due to the deprotonate of carboxylic acid side groups. The ICs formation of a channel-type crystalline structure induced gelation mechanism was verified by various techniques. In vitro cytotoxicity assays demonstrated that the supramolecular hydrogels are nontoxic and cytocompatible.展开更多

The objective of this paper is to prepare vinpocetine(VIN)inclusion complex and evaluate its brain targeting effect after intranasal administration.In the present study,VIN inclusion complex was prepared in order to i...The objective of this paper is to prepare vinpocetine(VIN)inclusion complex and evaluate its brain targeting effect after intranasal administration.In the present study,VIN inclusion complex was prepared in order to increase its solubility.Stability constant(Kc)was used for host selection.Factors influencing properties of the inclusion complex was investigated.Formation of the inclusion complex was identified by solubility study and DSC analysis.The brain targeting effect of the complex after intranasal administration was studied in rats.It was demonstrated that properties of the inclusion complex was mainly influenced by cyclodextrin type,organic acids type,system pH and host/vip molar ratio.Multiple component complexes can be formed by the addition of citric acid,with solubility improved for more than 23 times.Furthermore,In vivo study revealed that after intranasal administration,the absolute bioavailability of vinpocetine inclusion complex was 88%.Compared with intravenous injection,significant brain targeting effect was achieved after intranasal delivery,with brain targeting index 1.67.In conclusion,by intranasal administration of VIN inclusion complex,a fast onset of action and good brain targeting effect can be achieved.Intranasal route is a promising approach for the treatment of CNS diseases.展开更多

The solid Supramolecular complexes of β-cyclodextrin (β-CD) with ethylenediamine 1, diethylenetriamine 2 and triethylamine 3 were obtained and characterized using elemental analysis, powder X-ray diffraction, infr...The solid Supramolecular complexes of β-cyclodextrin (β-CD) with ethylenediamine 1, diethylenetriamine 2 and triethylamine 3 were obtained and characterized using elemental analysis, powder X-ray diffraction, infrared spectroscopy, thermogravimetric analysis, and ^1H nuclear magnetic resonance spectroscopy. Based on the results of elemental analysis and ^1H NMR, the vip-host stoichiometries of the three solid complexes were determined to be 5:2 for 1-β-CD, hl for 2-β-CD, and 1:3 for 3-β-CD. The yields were relative to the molar volume ratio of vip to β-CD cavity, and increased in the order: 1-β-CD〈2-3-CD〈3-β-CD. X-ray diffraction patterns of the inclusion complexes gave very good exhibitions not only in location of diffraction peaks but also in shape and diffraction intensity of the peaks due to the intermolecular complexations between β-CD and the vips. The formation of host-vip inclusion complexes exhibited obviously enhanced phase change temperatures of the complexed vips such as 1 and 3. The H-5 protons located at the narrower rim inside the CD cavity experienced a higher shift upon inclusion while all other protons experienced lower shifts.展开更多

One of the major problems of paclitaxel applied to therapy is its extremely low solubility in water. In addition, paclitaxel is administered as a slow infusion in a vehicle consisting of Cremophor EL ( polyoxyethylated castor oil ). However, Cremophor has been observed to cause severe, occasionally fatal hypersensitivity reactions in animals and humans. Therefore, paclitaxel therapy includes a prophylactic regimen of antihistamines and corticosteroids , along with a prolonged infusion time to reduce the severity and incidence of hypersensitivity reactions. The inclusion complexes (ICs) formed between the PCL polymers and α-CD were characterized by ^1H-NMR, DSC, TGA, WAXD, and FT-1R, respectively. Both branch ann number and molecular weight of the PCL polymers have apparent effect on the stoichiometry (CL:CD, mol:mol) of these ICs. All these analytical results indicate that the branch arms of the PCL polymers are incorporated into the hydrophobic α-CD cavities and their original crystalline properties are completely suppressed. Moreover, the inclusion complexation between two-ann linear or four-ann star-shaped PCL polymers and α-CD not only enhances the thermal stability of the vip PCL polymers but also improves that of α-CD.展开更多

However, its use in additive field is limited by its low aqueous solubility and chemical stability. So trace amounts of VD<sub>3</sub> was wrapped in β-CD molecule by the method of saturated aqueous vacuum drying, in order to improve its stability, uniformity and solubility in food and feed additive. The inclusion complex was characterized by NMR, IR techniques and compared with original VD<sub>3</sub> in the aspect of stability and bioavailability. Results of orthogonal design experiments show that the optimum technology of inclusion is that the feed ratio of β-CD to VD<sub>3</sub> is 15:1, being stirred for 5 hours at 80°C. Dispersion of VD<sub>3</sub> in the inclusion complex is more uniform, while stability and absorption rate of inclusion complex are significantly higher than original VD<sub>3</sub>.展开更多

These hydrogels displayed thermo-sensitive gel-sol transition under appropriate conditions due to the reversible mPEG and α-CD inclusion complexation. These hydrogels also showed pH-sensitive behavior due to the deprotonate of carboxylic acid side groups. The ICs formation of a channel-type crystalline structure induced gelation mechanism was verified by various techniques. In vitro cytotoxicity assays demonstrated that the supramolecular hydrogels are nontoxic and cytocompatible.

文摘The objective of this paper is to prepare vinpocetine(VIN)inclusion complex and evaluate its brain targeting effect after intranasal administration.In the present study,VIN inclusion complex was prepared in order to increase its solubility.Stability constant(Kc)was used for host selection.Factors influencing properties of the inclusion complex was investigated.Formation of the inclusion complex was identified by solubility study and DSC analysis.The brain targeting effect of the complex after intranasal administration was studied in rats.It was demonstrated that properties of the inclusion complex was mainly influenced by cyclodextrin type,organic acids type,system pH and host/vip molar ratio.Multiple component complexes can be formed by the addition of citric acid,with solubility improved for more than 23 times.Furthermore,In vivo study revealed that after intranasal administration,the absolute bioavailability of vinpocetine inclusion complex was 88%.Compared with intravenous injection,significant brain targeting effect was achieved after intranasal delivery,with brain targeting index 1.67.In conclusion,by intranasal administration of VIN inclusion complex,a fast onset of action and good brain targeting effect can be achieved.Intranasal route is a promising approach for the treatment of CNS diseases.

文摘Inclusion complex of Orange II withβ-Cyclodextrin(β-CD)and the anti-photolysis effect under UV-light were investigated.The molar ratio of inclusion complex ofβ-Cyclodextrin and OrangeⅡis 1∶1.The formation constant K=1.236×10^(3)L/mol was determined by the UV and Fluorescence spectra respectively,which was quite in accordance with the calculation with a modified Benesi-Hildbrand equation.The inclusion complex was characterized by the IR spectra and the molar ratio of inclusion complex is 1∶1 too.The formation constant K=1.266×10^(3)L/mol was determined by 1 H NMR analysis and was nearly the same by UV and fluorescence spectra.The photocatalytic decolorization rate of OrangeⅡsolutions containingβ-CD and TiO_(2)was smaller by 51.9%than that of the OrangeⅡsolutions only containing TiO_(2),while in the case of direct photolysis of OrangeⅡsolutions,β-CD can lower the photolysis rate by 48.1%under UV-light.This result indicatesβ-CD can inhibit the photolysis and photocatalytic decolorization of OrangeⅡunder UV-light.Theβ-CD inclusion complex was found to be persistent to UV-light photolysis.

The crystalline inclusion complex between β-CD and lowmolecular weight polypropylene (PP) was obtained and investigated. α-CD and γ-CD did not form crystalline inclusioncomplexes with PP. The FTIR spectra, TGA, X-ray diffraction spectra were studied, ~1H-NMR spectra and ^(13)C CP/MAS NMR spectra were used to characterize the crystalline inclusion complexes.
文摘The solid Supramolecular complexes of β-cyclodextrin (β-CD) with ethylenediamine 1, diethylenetriamine 2 and triethylamine 3 were obtained and characterized using elemental analysis, powder X-ray diffraction, infrared spectroscopy, thermogravimetric analysis, and ^1H nuclear magnetic resonance spectroscopy. Based on the results of elemental analysis and ^1H NMR, the vip-host stoichiometries of the three solid complexes were determined to be 5:2 for 1-β-CD, hl for 2-β-CD, and 1:3 for 3-β-CD. The yields were relative to the molar volume ratio of vip to β-CD cavity, and increased in the order: 1-β-CD〈2-3-CD〈3-β-CD. X-ray diffraction patterns of the inclusion complexes gave very good exhibitions not only in location of diffraction peaks but also in shape and diffraction intensity of the peaks due to the intermolecular complexations between β-CD and the vips. The formation of host-vip inclusion complexes exhibited obviously enhanced phase change temperatures of the complexed vips such as 1 and 3. The H-5 protons located at the narrower rim inside the CD cavity experienced a higher shift upon inclusion while all other protons experienced lower shifts.
文摘Introduction Paclitaxel (as Taxol) is a kind of diterpenoid natural product extracted from Chinese yew. It has been reported to have high anti-tumor effects, such as the activity against oophorama, mammary cancer, encephaloma, cervical carcinoma, and non- small-cell lung carcinoma. One of the major problems of paclitaxel applied to therapy is its extremely low solubility in water. In addition, paclitaxel is administered as a slow infusion in a vehicle consisting of Cremophor EL ( polyoxyethylated castor oil ). However, Cremophor has been observed to cause severe, occasionally fatal hypersensitivity reactions in animals and humans. Therefore, paclitaxel therapy includes a prophylactic regimen of antihistamines and corticosteroids , along with a prolonged infusion time to reduce the severity and incidence of hypersensitivity reactions. Because of the reasons mentioned above, currently its preparation needs to be improved further.

It crystallized in the monoclinic system with space group P 2 1/ n , a =1.734 7(2) nm, b =1.903 2(2) nm, c =1.972 9(2) nm , β =94.74(1)°, V =6.491 2(14) nm 3, Z =4. The toluene molecule was included in the macrocycle cavity. The calixanene is of a pinched cone conformation.

文摘A novel brideed bis(β-cvclodextrin), 4, 4'-diaminodiphenyl ether-bfiged-bis (6-aimino-6-deoxv-β-cyclodextrin) 3, has been synthesized and its inclusion complexation behavior with three linear'vip dyes (AR, NR and MB ) has been investigated by. means of fluorescence spectrometry. The-result obtained demonstrated that the novel bridged bis(β-cyclodextnn) showed much higher affinities towards vip dyes than native β-cyclodextrin.

文摘Both four-ann star-shaped poly(ε-caprolactone) (4sPCL) and two-ann linear PCL (2LPCL) were synthesized and their inclusion complexation with α-cyclodextrin (α-CD) were studied. The inclusion complexes (ICs) formed between the PCL polymers and α-CD were characterized by ^1H-NMR, DSC, TGA, WAXD, and FT-1R, respectively. Both branch ann number and molecular weight of the PCL polymers have apparent effect on the stoichiometry (CL:CD, mol:mol) of these ICs. All these analytical results indicate that the branch arms of the PCL polymers are incorporated into the hydrophobic α-CD cavities and their original crystalline properties are completely suppressed. Moreover, the inclusion complexation between two-ann linear or four-ann star-shaped PCL polymers and α-CD not only enhances the thermal stability of the vip PCL polymers but also improves that of α-CD.
文摘The inclusion complex of β-cyclodextrin with m-nitrophenoxyacetic acid was studied by single crystal X-ray diffraction,2D NMR spectroscopy and semi-empirical methods AMI.The crystallographic study shows that two β-cyclodextrins are held together by hydrogen bonds to form head-to-head dimers.The disordered vip molecule adjusts itself to attain the most stable accommodation into the cavity in which the nitro group is located at the dimer interface while the carboxyl group is buried in the primary hydroxyl groups of β-cyclodextrin.The vip inside the cavity is disordered over two sites and exhibits mobility.Moreover,2D NMR spectroscopy and theoretical study show the same inclusion behavior.In comparison to the inclusion complex of β-cyclodextrin with p-nitrophenoxyacetic acid,the host-vip stoichiometries are different,i.e.,2:1 for m-nitrophenoxyacetic acid and 1:1 for p-nitrophenoxyacetic acid,while the inclusion orientation and the packing pattern of the host are similar in both complexes.
文摘Vitamin D is responsible for enhancing intestinal absorption of calcium, iron, magnesium, phosphate and zinc by involving in the metabolism. However, its use in additive field is limited by its low aqueous solubility and chemical stability. So trace amounts of VD<sub>3</sub> was wrapped in β-CD molecule by the method of saturated aqueous vacuum drying, in order to improve its stability, uniformity and solubility in food and feed additive. The inclusion complex was characterized by NMR, IR techniques and compared with original VD<sub>3</sub> in the aspect of stability and bioavailability. Results of orthogonal design experiments show that the optimum technology of inclusion is that the feed ratio of β-CD to VD<sub>3</sub> is 15:1, being stirred for 5 hours at 80°C. Dispersion of VD<sub>3</sub> in the inclusion complex is more uniform, while stability and absorption rate of inclusion complex are significantly higher than original VD<sub>3</sub>.
