Complex trimalleolar ankle fractures are a major orthopaedic challenge,with an incidence of 4.22 per 10000 person-years in the United States and an annual cost of 3.4 billion dollars.This review synthesizes current evidence on diagnostic protocols and management strategies,highlighting optimal approaches and emerging trends.Initial care emphasizes soft tissue assessment,often guided by the Tscherne classification,and fracture classification systems.External fixation may be required in open injuries,while early open reduction and internal fixation within six days is linked to improved outcomes.Minimally invasive techniques for the lateral malleolus,including intramedullary nailing and locking plates,are effective,while medial malleolus fractures are commonly managed with screw fixation or tension-band wiring.Posterior malleolus fragments involving more than 25%of the articular surface usually warrant fixation.Alternatives to syndesmotic screws,such as cortical buttons or high-strength sutures,reduce the need for secondary procedures.Arthroscopic-assisted open reduction and internal fixation benefits younger,active patients by enabling concurrent management of intra-articular and ligamentous injuries.Postoperative care prioritizes early weight-bearing and validated functional scores.Despite advances,complications remain common,and further research is needed to refine surgical strategies and improve outcomes. 展开更多

目的本研究旨在探讨弥漫大B细胞淋巴瘤(DLBCL)中主要组织相容性复合体(MHC)Ⅰ、Ⅱ表达及其与临床病理特征及预后的相关性。方法收集新疆医科大学第一附属医院病理科DLBCL 130例,采用免疫组化EnVision法检测MHC-Ⅰ、MHC-Ⅱ蛋白表达情况,... 目的本研究旨在探讨弥漫大B细胞淋巴瘤(DLBCL)中主要组织相容性复合体(MHC)Ⅰ、Ⅱ表达及其与临床病理特征及预后的相关性。方法收集新疆医科大学第一附属医院病理科DLBCL 130例,采用免疫组化EnVision法检测MHC-Ⅰ、MHC-Ⅱ蛋白表达情况,分析二者相关性及其与DLBCL临床病理特征和预后的关系。结果(1)免疫组化染色结果显示,DLBCL肿瘤细胞中MHC-Ⅰ、MHC-Ⅱ低表达率分别为57.6%(68/118)和40.3%(48/119)。(2)MHC-Ⅰ低表达与Ann-Arbor分期、PS评分、发生部位相关(P<0.05),MHC-Ⅱ低表达与年龄、结外受累病灶相关(P<0.05),MHC-Ⅰ/MHC-Ⅱ共高表达与骨髓不易受累相关(P<0.05)。(3)Kaplan-Meier单因素生存结果显示MHC-Ⅱ低表达组预后差(P<0.05),MHC-Ⅰ对生存无显著影响(P>0.05),多因素COX回归分析显示LDH、PS评分、IPI评分是影响DLBCL患者的独立危险因素。结论MHC-Ⅰ和MHC-Ⅱ在DLBCL中低表达,与Ann-Arbor分期、PS评分、发生部位、年龄、结外受累病灶相关。MHC-Ⅱ低表达的DLBCL患者预后较差。 展开更多

目的 探讨复方浙贝颗粒(CZBG)基于双微体同源基因2(MDM2)/p53信号通路调节主要组织相容性复合体Ⅱ类(MHC-Ⅱ)逆转白血病多药耐药的作用机制。方法 选取人急性髓系白血病细胞(KG-1a、MOLM-13细胞),将KG-1a、MOLM-13细胞分别分为CZBG组、... 目的 探讨复方浙贝颗粒(CZBG)基于双微体同源基因2(MDM2)/p53信号通路调节主要组织相容性复合体Ⅱ类(MHC-Ⅱ)逆转白血病多药耐药的作用机制。方法 选取人急性髓系白血病细胞(KG-1a、MOLM-13细胞),将KG-1a、MOLM-13细胞分别分为CZBG组、空白对照组。空白对照组予完全培养基培养,CZBG组予含不同浓度的CZBG完全培养基培养。CCK-8法检测细胞增殖,Western blotting法检测MDM2、p53蛋白表达,流式细胞术检测MHC-Ⅱ表达。结果 干预24、48、72 h,CZBG对KG-1a、MOLM-13细胞增殖均具有抑制作用。其中CZBG在24、48、72 h时对KG-1a细胞的半数抑制浓度(IC50)为(2.276±0.218)、(1.254±0.104)、(1.040±0.108)mg/mL,对MOLM-13细胞的IC50为(2.809±0.121)、(1.585±0.103)、(1.194±0.105)mg/mL。应用2.2 mg/mL CZBG干预KG-1a细胞24 h时,与空白对照组比较,CZBG组MDM2蛋白相对表达量低(P<0.05),p53蛋白相对表达量高(P<0.05)。用2.8 mg/mL CZBG干预MOLM-13细胞24 h时,与空白对照组比较,CZBG组MDM2蛋白表达低(P<0.05),p53蛋白相对表达量高(P<0.05)。KG-1a、MOLM-13细胞分别经CZBG处理24 h,CZBG组细胞表面的MHC-Ⅱ表达高(P<0.05)。结论 CZBG逆转白血病多药耐药的作用机制可能与通过MDM2/p53通路调节白血病细胞表面MHC-Ⅱ表达,干预免疫逃逸有关。 展开更多

鸡MHC基因家族鉴定与分析

本研究旨在对鸡主要组织相容复合物(Major Histocompatibility Complex,MHC)基因家族进行生物信息学分析,明确鸡感染马立克氏病毒(Marek's Disease Virus,MDV)和注射羊红细胞(Sheep Red Blood Cells,SRBC)后MHC基因家族成员的表达... 本研究旨在对鸡主要组织相容复合物(Major Histocompatibility Complex,MHC)基因家族进行生物信息学分析,明确鸡感染马立克氏病毒(Marek's Disease Virus,MDV)和注射羊红细胞(Sheep Red Blood Cells,SRBC)后MHC基因家族成员的表达模式。通过生物信息学工具构建跨物种系统进化树,分析鸡MHC蛋白的理化性质、亚细胞定位、染色体分布、保守结构域、共线性、顺式作用元件,并构建蛋白质互作网络,进行蛋白质三维结构建模。进一步分析鸡感染MDV和SRBC免疫应答后鸡脾脏中MHC基因的表达变化。研究共鉴定出49个鸡MHC基因家族成员,鸡MHC基因均密集分布于第16号染色体的特定区域,与日本鹌鹑亲缘关系最近。共线性分析发现其中存在2次串联重复和片段重复事件。蛋白特性分析发现MHC基因编码的蛋白具有1~2个跨膜结构域,均含有免疫球蛋白超家族保守结构域,且主要定位于内质网和质膜。顺式元件分析发现MHC基因启动子区富含C/EBPα、NF-1等免疫相关调控位点,蛋白质互作网络分析发现BF2为枢纽节点。三维结构建模显示经典MHC I闭合槽结构适配内源肽段呈递,II类开放槽结构呈递外源肽段,而非经典基因(如MHCY)与BG基因可能通过非经典途径调控免疫应答。正选择分析发现鸟类特有的BG基因受到自然和人工的选择压力。转录组数据表明鸡感染MDV后大部分MHC基因的表达量上调。qRT-PCR分析表明鸡在SRBC免疫应答后,MHC基因(BF1、BF2、BLB2和DMB2)表达显著上调。 展开更多

【目的】进一步了解中国花鲈适应性免疫机制在病害防治中的作用。【方法】本研究通过荧光定量PCR(RT-PCR)和快速扩增c DNA末端(RACE)技术克隆获得了免疫球蛋白M重链(IgMH)和主要组织兼容性复合体MHC Ⅱβ基因的全长;通过实时荧光定量PCR... 【目的】进一步了解中国花鲈适应性免疫机制在病害防治中的作用。【方法】本研究通过荧光定量PCR(RT-PCR)和快速扩增c DNA末端(RACE)技术克隆获得了免疫球蛋白M重链(IgMH)和主要组织兼容性复合体MHC Ⅱβ基因的全长;通过实时荧光定量PCR(RT-qPCR)检测其在中国花鲈各组织的分布情况以及LPS、Poly(I:C)刺激及迟缓爱德华氏菌人工感染后其对应mRNA水平的表达变化情况;实验构建了IgMH-pET21d、IgMCH1-2-pET21d和MHCⅡβ-pET21d原核重组表达质粒,进行重组蛋白表达,并通过分子筛层析技术纯化中国花鲈IgMH、IgMCH1-2和MHCⅡβ重组蛋白,进而制备了抗中国花鲈IgMH的多克隆抗体。【结果】中国花鲈IgMH和MHCⅡβ的cDNA全长分别为1977 bp和1242 bp;二者在鳃、脾脏和头肾等免疫相关组织中对应mRNA表达量较高;LPS、Poly(I:C)刺激及迟缓爱德华氏菌人工感染中国花鲈导致鳃、脾脏和头肾中这两个基因的表达水平发生显著变化,表明IgMH和MHCⅡβ均参与了中国花鲈的抗感染免疫反应;此外,抗中国花鲈IgM的多克隆抗体能与中国花鲈全血清发生强烈反应,与鳜全血清弱反应,与大口黑鲈和草鱼血清不发生反应,推测其反应强度反映了物种间亲缘关系的远近。【结论】本研究首次克隆了中国花鲈的IgMH和MHCⅡβ基因全长,表达了IgMH和MHCⅡβ原核重组蛋白,制备了抗中国花鲈IgM的多克隆抗体,揭示了IgMH和MHCⅡβ参与中国花鲈的抗感染免疫应答,为深入研究中国花鲈的免疫调控机制和疾病防控策略奠定了基础。 展开更多

To expand the study on the structures and biological activities of the anthracyclines anticancer drugs and reduce their toxic side effects,the new anthraquinone derivatives,9‑pyridylanthrahydrazone(9‑PAH)and 9,10‑bispyridylanthrahydrazone(9,10‑PAH)were designed and synthesized.Utilizing 9‑PAH and 9,10‑PAH as promising anticancer ligands,their respective copper complexes,namely[Cu(L1)Cl_(2)]Cl(1)and{[Cu_(4)(μ_(2)‑Cl)_(3)Cl_(4)(9,10‑PAH)_(2)(DMSO)_(2)]Cl_(2)}_(n)(2),were subsequently synthesized,where the new ligand L1 is formed by coupling two 9‑PAH ligands in the coordination reaction.The chemical and crystal structures of 1 and 2 were elucidated by IR,MS,elemental analysis,and single‑crystal X‑ray diffraction.Complex 1 forms a mononuclear structure.L1 coordinates with Cu through its three N atoms,together with two Cl atoms,to form a five‑coordinated square pyramidal geometry.Complex 2 constitutes a polymeric structure,wherein each structural unit centrosymmetrically encompasses two five‑coordinated binuclear copper complexes(Cu1,Cu2)of 9,10‑PAH,with similar square pyramidal geometry.A chlorine atom(Cl_(2)),located at the symmetry center,bridges Cu1 and Cu1A to connect the two binuclear copper structures.Meanwhile,the two five‑coordinated Cu2 atoms symmetrically bridge the adjacent structural units via one coordinated Cl atom,respectively,thus forming a 1D chain‑like polymeric structure.In vitro anticancer activity assessments revealed that 1 and 2 showed significant cytotoxicity even higher than cisplatin.Specifically,the IC_(50)values of 2 against HeLa‑229 and SK‑OV‑3 cancer cell lines were determined to be(5.92±0.32)μmol·L^(-1)and(6.48±0.39)μmol·L^(-1),respectively.2 could also block the proliferation of HeLa‑229 cells in S phase and significantly induce cell apoptosis.In addition,fluorescence quenching competition experiments suggested that 2 might interact with DNA by an intercalative binding mode,offering insights into its underlying anticancer mechanism.CCDC:2388918,1;2388919,2. 展开更多

High-resolution seeing through complex scattering media such as turbid water,biological tissues,and mist is a significant challenge because the strong scattering scrambles the light paths and forms the scattering wall.We propose an active polarized iterative optimization approach for high-resolution imaging through complex scattering media.By acquiring a series of sub-polarized images,we can capture the diverse pattern-illuminated images with various high-frequency component information caused by the Brownian motion of complex scattering materials,which are processed using the common-mode rejection of polarization characteristics to extract target information from scattering medium information.Following that,our computational reconstruction technique employs an iterative optimization algorithm that commences with patternilluminated Fourier ptychography for reconstructing the high-resolution scene.It is extremely important that our approach for high-resolution imaging through complex scattering media is not limited by priori information and optical memory effect.The proposed approach is suitable for not only dynamic but also static scattering media,which may find applications in the biomedicine field,such as skin abnormalities,non-invasive blood flow,and superficial tumors. 展开更多

A complex system is inherently high-dimensional.Recent studies indicate that,even without complete knowledge of its evolutionary dynamics,the future behavior of such a system can be predicted using time-series data(data-driven prediction).This suggests that the essential dynamics of a complex system can be captured through a low-dimensional representation.Virus evolution and climate change are two examples of complex,time-varying systems.In this article,we show that mutations in the spike protein provide valuable data for predicting SARS-CoV-2 variants,forecasting the possible emergence of the new macro-lineage Q in the near future.Our analysis also demonstrates that carbon dioxide concentration is a reliable indicator for predicting the evolution of the climate system,extending global surface air temperature(GSAT)forecasts through 2500. 展开更多

Image-maps,a hybrid design with satellite images as background and map symbols uploaded,aim to combine the advantages of maps’high interpretation efficiency and satellite images’realism.The usability of image-maps is influenced by the representations of background images and map symbols.Many researchers explored the optimizations for background images and symbolization techniques for symbols to reduce the complexity of image-maps and improve the usability.However,little literature was found for the optimum amount of symbol loading.This study focuses on the effects of background image complexity and map symbol load on the usability(i.e.,effectiveness and efficiency)of image-maps.Experiments were conducted by user studies via eye-tracking equipment and an online questionnaire survey.Experimental data sets included image-maps with ten levels of map symbol load in ten areas.Forty volunteers took part in the target searching experiments.It has been found that the usability,i.e.,average time viewed(efficiency)and average revisits(effectiveness)of targets recorded,is influenced by the complexity of background images,a peak exists for optimum symbol load for an image-map.The optimum levels for symbol load for different image-maps also have a peak when the complexity of the background image/image map increases.The complexity of background images serves as a guideline for optimum map symbol load in image-map design.This study enhanced user experience by optimizing visual clarity and managing cognitive load.Understanding how these factors interact can help create adaptive maps that maintain clarity and usability,guiding AI algorithms to adjust symbol density based on user context.This research establishes the practices for map design,making cartographic tools more innovative and more user-centric. 展开更多

The Jiuyishan granitic complex,located in the Nanling Range,South China,is composed of five granitic plutons(Xuehuading,Jinjiling,Pangxiemu,Shaziling and Xishan).Zircon U-Pb dating of four plutons(Jinjiling,Pangxiemu,... The Jiuyishan granitic complex,located in the Nanling Range,South China,is composed of five granitic plutons(Xuehuading,Jinjiling,Pangxiemu,Shaziling and Xishan).Zircon U-Pb dating of four plutons(Jinjiling,Pangxiemu,Shaziling and Xishan)yielded similar ages of approximately 153 Ma,indicating indistinguishable ages within error.Three plutons except the Shaziling pluton,have consistentε_(Nd)(t)(-7.8 to-5.8)andε_(Hf)(t)(-9.1 to-2.2)values,which are similar to those of the lower crustal granulitic metasedimentary and meta-igneous rocks in South China.Compared to other three plutons,the Shaziling pluton has consistentε_(Nd)(t)(-7.4 to-6.8)andε_(Hf)(t)(-7.5 to-4.7)values and shows similar source,but the Shaziling mafic microgranular enclaves(MMEs)show variableε_(Hf)(t)(-14.2 to 4.8)values,indicating a remarkable mantle magma injection of the Shaziling pluton.Zircon Ce/Sm-Yb/Gd,whole-rock CaO-P_(2)O_(5)and CaO-TiO_(2)linear trends reveal that from the Xishan to the Shaziling and from the Jinjiling to the Pangxiemu granites,they experienced apatite and titanite fractionation,respectively.Zircon Th,U,Nb,Ta,Hf,Ti,Y,P and rare earth element(REE)contents and whole-rock Sr,Ba and Rb contents also show that the Shaziling,Xishan,Jinjiling and Pangxiemu granites followed a discontinuous evolutionary series,but the Pangxiemu granites exhibit highly evolved nature.Four main controlling factors of W-Sn and rare metal mineralization in granitic rocks were discussed,and we found that the mineralization in Jiuyishan granitic complex was mainly controlled by the fractionation degree and crystallization temperature,but were rarely affected by oxygen fugacity and mantle material input.The Pangxiemu granites show particularly higher Rb and Ta contents than the other three plutons,implying that the ore deposits developed in the Jiuyishan Complex were directly related to the most evolved Pangxiemu pluton,with the occurrence of Rb and Ta as the most likely rare metal mineralization in the Jiuyishan District.A crystal mush model is proposed to interpret the petrogenetic and mineralizing processes of the Jiuyishan granitic complex. 展开更多

A phenylphenothiazine anchored Tb(Ⅲ)-cyclen complex PTP-Cy-Tb for hypochlorite ion(ClO^(-))detection has been designed and prepared.PTP-Cy-Tb shows a weak Tb-based emission with AIE-characteristics in aqueous solutions.After addition of ClO^(-),the fluorescence of PTP-Cy-Tb gives a large enhancement for oxidization the thioether to sulfoxide group.The detection limit of PTP-Cy-Tb toward ClO^(-)is as low as 8.85 nmol/L.The sensing mechanism was detailedly investigated by time of flight mass spectrometer(TOF-MS),Fourier transform infrared spectroscopy(FT-IR)and density functional theory(DFT)calculation.In addition,PTP-Cy-Tb has been successfully used for on-site and real-time detection of ClO^(-)in real water samples by using the smartphone-based visualization method and test strips. 展开更多

Metal complexes hold significant promise in tumor diagnosis and treatment.However,their potential applications in photodynamic therapy(PDT)are hindered by issues such as poor photostability,low yield of reactive oxygen species(ROS),and aggregation-induced ROS quenching.To address these challenges,we present a molecular self-assembly strategy utilizing aggregation-induced emission(AIE)conjugates for metal complexes.As a proof of concept,we synthesized a mitochondrial-targeting cyclometalated Ir(Ⅲ)photosensitizer Ir-TPE.This approach significantly enhances the photodynamic effect while mitigating the dark toxicity associated with AIE groups.Ir-TPE readily self-assembles into nanoaggregates in aqueous solution,leading to a significant production of ROS upon light irradiation.Photoirradiated Ir-TPE triggers multiple modes of death by excessively accumulating ROS in the mitochondria,resulting in mitochondrial DNA damage.This damage can lead to ferroptosis and autophagy,two forms of cell death that are highly cytotoxic to cancer cells.The aggregation-enhanced photodynamic effect of Ir-TPE significantly enhances the production of ROS,leading to a more pronounced cytotoxic effect.In vitro and in vivo experiments demonstrate this aggregation-enhanced PDT approach achieves effective in situ tumor eradication.This study not only addresses the limitations of metal complexes in terms of low ROS production due to aggregation but also highlights the potential of this strategy for enhancing ROS production in PDT. 展开更多

Background:Enzyme fragility remains a major challenge in research and applications.Free enzymes are highly unstable,inactivated by heat,acid,alkali,or organic solvents,and often lose activity even under optimal storage conditions.Limiting their use in cosmetics.Few commercial products combine acids and enzymes effectively.Objective:To investigate the physicochemical properties,in vitro exfoliation efficacy,and effects on facial skin parameters of a supramolecular acid-enzyme complex(SAE)composed of mandelic acid(MAN),betaine(BET),and composite enzymes(CE;papain and bromelain),thereby establishing a theoretical foundation for cosmetic applications.Methods:The supramolecular structure was characterized using Fourier transform infrared(FTIR)spectroscopy and proton nuclear magnetic resonance(1H NMR)spectroscopy.Dissolution experiments were conducted to compare the solubility of SAE and CE in aqueous solutions.Enzymatic activity assays evaluated the stabilizing effect of supramolecular deep eutectic technology on enzymes.In vitro exfoliation tests assessed acid-enzyme synergy in keratin removal.A 4-week clinical trial evaluated the efficacy of a 2%SAE essence aqueous solution on facial skin parameters.Results:Dissolution experiments confirmed that supramolecular deep eutectic technology significantly improved enzyme solubility.Enzymatic activity tests demonstrated that this technology effectively preserved protease activity,substantially enhancing its practical applicability.Furthermore,in vitro exfoliation efficacy tests revealed that this technology strengthened the synergistic interaction between acids and enzymes and exhibited superior stratum corneum-removing capability of the SAE.In clinical evaluations of efficacy,after 7 days of using the essence containing SAE,the formulation significantly enhanced cheek gloss(+8.08%),while reducing comedones volume(-16.25%).after 28 days,significantly enhanced cheek hydration(+25.0%,SCH),gloss(+15.93%),and smoothness(−7.78%SEsm),while reducing TEWL(−6.86%),sebum(−15.54%),roughness(+16.24%SEr),and pore metrics(volume:−39.98%;count:−30.64%),and decreased comedones(blackheads:−70.33%;Whiteheads:−52.42%;all p<0.05).Conclusion:The supramolecular acid-enzyme complex demonstrates enhanced stability,improved solubility,and superior exfoliation efficacy compared to free enzymes.Clinical results further confirm its multifunctional benefits,including enhancing skin hydration,sebum regulation,barrier repair,pore refinement,and comedolytic effects.This study provides both theoretical and practical foundations for developing stable acid-enzyme combinations in dermatological applications. 展开更多

Viral infectious diseases,characterized by their intricate nature and wide-ranging diversity,pose substantial challenges in the domain of data management.The vast volume of data generated by these diseases,spanning from the molecular mechanisms within cells to large-scale epidemiological patterns,has surpassed the capabilities of traditional analytical methods.In the era of artificial intelligence(AI)and big data,there is an urgent necessity for the optimization of these analytical methods to more effectively handle and utilize the information.Despite the rapid accumulation of data associated with viral infections,the lack of a comprehensive framework for integrating,selecting,and analyzing these datasets has left numerous researchers uncertain about which data to select,how to access it,and how to utilize it most effectively in their research.This review endeavors to fill these gaps by exploring the multifaceted nature of viral infectious diseases and summarizing relevant data across multiple levels,from the molecular details of pathogens to broad epidemiological trends.The scope extends from the micro-scale to the macro-scale,encompassing pathogens,hosts,and vectors.In addition to data summarization,this review thoroughly investigates various dataset sources.It also traces the historical evolution of data collection in the field of viral infectious diseases,highlighting the progress achieved over time.Simultaneously,it evaluates the current limitations that impede data utilization.Furthermore,we propose strategies to surmount these challenges,focusing on the development and application of advanced computational techniques,AI-driven models,and enhanced data integration practices.By providing a comprehensive synthesis of existing knowledge,this review is designed to guide future research and contribute to more informed approaches in the surveillance,prevention,and control of viral infectious diseases,particularly within the context of the expanding big-data landscape. 展开更多

Two novel lanthanide complexes,[Sm_(2)(BA)_(6)(4-OH-terpy)_(2)]·2H_(2)O·2EtOH(1)and[Pr_(2)(BA)_(6)(4-OH-terpy)_(2)(H_(2)O)_(2)]·HBA·H_(2)O(2),where HBA=benzoic acid,4-OH-terpy=4-hydroxy-2,2'∶6',2″-terpyridine,were successfully synthesized using ultrasonic dissolution and the conventional solution method with two mixed ligands HBA and 4-OH-terpy.During the synthesis,4-OH-terpy was involved in the reaction as a neutral ligand,while HBA,in its deprotonated form(BA-),coordinated with the lanthanide ions as an acidic ligand.The crystal structures of these two complexes were precisely determined by single-crystal X-ray diffraction.Elemental analysis,infrared and Raman spectroscopy,and powder X-ray diffraction techniques were also employed to further explore the physicochemical properties of the two complexes.The single-crystal X-ray diffraction data indicate that,despite their structural differences,both complexes belong to the triclinic crystal system P1 space group.The central lanthanide ions have the same coordination number but exhibit different coordination environments.To comprehensively evaluate the thermal stability of these two complexes,comprehensive tests including thermogravimetric analysis,differential thermogravimetric analysis,differential scanning calorimetry,Fourier transform infrared spectroscopy,and mass spectrometry were conducted.Meanwhile,an in-depth investigation was conducted into the 3D infrared stacked images and mass spectra of the gases emitted from the complexes.In addition,studies of the fluorescence properties of complex1 showed that it exhibited fluorescence emission matching the Sm^(3+)characteristic transition. 展开更多
