In the present study, the antigenic regions of fi ve outer membrane proteins(OMPs), including zinc metalloprotease, prolyl oligopeptidase, thermolysin, collagenase and chondroitin AC lyase, were bioinformatically analyzed, fused together, and then expressed as a recombinant fusion protein in Escherichia coli. The expressed protein of 95.6 k Da, as estimated by 10% sodium dodecyl sulfate-polyacrylamide gel electrophoresis, was consistent with the molecular weight deduced from the amino acid sequence. The purifi ed recombinant protein was used to vaccinate the grass carp, C tenopharyngodon idella. Following vaccination of the fi sh their Ig M antibody levels were examined, as was the expression of I g M, Ig D and Ig Z immunoglobulin genes and other genes such as MHC Iα and MHC I I β, which are also involved in adaptive immunity. Interleukin genes( IL), including I L- 1β, IL- 8 and I L- 10, and type I and type II interferon(I FN) genes were also examined. At 3 and 4 weeks post-vaccination(wpv), signifi cant increases in Ig M antibody levels were observed in the fi sh vaccinated with the recombinant fusion protein, and an increase in the expression levels of I g M, Ig D and Ig Z genes was also detected following the vaccinations, thus indicating that an adaptive immune response was induced by the vaccinations. Early increases in the expression levels of IL and IFN genes were also observed in the vaccinated fi sh. At four wpv, the fi sh were challenged with F. column a re, and the vaccinated fi sh showed a good level of protection against this pathogen, with 39% relative percent survival(RPS) compared with the control group. It can be concluded, therefore, that the fi ve OMPs, in the form of a recombinant fusion protein vaccine, induced an immune response in fi sh and protection against F. columnare.展开更多

Isomahanine was found to have the strongest activity against F. columnare (isolate ALM-00-173) and S. iniae (isolate LA94-426) based on 24-h 50% inhibition concentration (IC50) and minimum inhibition concentration (MIC). Although compound (7), a nicotinamide isolated from Amyris texana had the lowest MIC (2.8 ± 0 mg/L) of any of the test compounds against F. columnare, the 24-h IC50 of 14.8 ± 0.6 mg/L was higher than that of isomahanine and subsequently the 24-h IC50 RDC values for (7) were almost a magnitude of order higher than those obtained for isomahanine. Isomahanine also had the strongest activity against S. iniae, with a 24-h IC50 of 1.3 ± 0.1 mg/L and MIC of 3.5 ± 0 mg/L, respectively.展开更多

Using a recently developed gene deletion strategy, two genes that encode the Glyco hydro_19 domain (GH19 domain) containing proteins, ghd-1 and ghd-2, were deleted separately and together from the F. columnare G4 wild type strain. Surprisingly, the single-, Aghd-1 and Aghd-2, and double-gene mutants, Aghd-1 Aghd-2, all had rhizoid and non-rhizoid colony morphotypes, which we named Aghd-1, Aghd-2, Aghd-1 Aghd-2, and NAghd-1, NAghd-2, and NAghd-1 Aghd-2. However, chitin utilization was not detected in either these mutants or in the wild type. Instead, skimmed milk degradation was observed for the mutants and the wild type; the non-rhizoid strain NAghd-2 exhibited higher degradation activity as revealed by the larger transparent circle on the skimmed milk plate. Using zebrafish as the model organism, we found that non-rhizoid mutants had higher LDs0 values and were less virulent because zebrafish infected with these survived longer. Transcriptome analysis between the non-rhizoid and rhizoid colony morphotypes of each mutant, i.e., NAghd-1 versus (vs) Aghd-1, NAghd-2 vs Aghd-2, and NAghd-1 Aghd-2 vs Aghd-1 Aghd-2, revealed a large number of differentially expressed genes, among which 39 genes were common in three of the pairs compared. Although most of these genes encode hypothetical proteins, a few molecules such as phage tail protein, rhs element Vgr protein, thiol-activated cytolysin, and TonB-dependent outer membrane receptor precursor, expression of which was down-regulated in non-rhizoid mutants but up-regulated in rhizoid mutants, may play a role F. columnare virulence.展开更多

In this study, we constructed two artificial selectable markers (chloramphenicol and spectinomycin resistance) for gene transfer in F. columnare. These two new artificial selectable markers, which were created by placing the chloramphenicol or spectinomycin resistance gene under the control of the native acs regulatory region of F. columnare, were functional in both F. columnare and Escherichia coli. The integrative/conjugative plasmids constructed by using these markers were introduced into F. columnare G4 via electroporation or conjugation. The integrated plasmid DNA was confirmed by Southern blotting and PCR analysis. These two markers can be employed in future investigations into gene deletion and the pathogenicity of virulence factors in F. columnare.展开更多
Columnaris (caused by Flavobacterium columnare) is one of the most common bacterial diseases affecting the pond-raised channel catfish (Ictalurus punctatus) in the southeastern United States of America resulting in an...Columnaris (caused by Flavobacterium columnare) is one of the most common bacterial diseases affecting the pond-raised channel catfish (Ictalurus punctatus) in the southeastern United States of America resulting in annual losses of millions of dollars. As part of our continuing effort to discover environmentally benign compounds for the control of columnaris disease, acyl derivatives of phloroglucinol were synthesized and tested against F. columnare using a rapid bioassay. Among the analogs that were tested, diacyl analogs showed very high antibacterial activity against F. columnare in the laboratory bioassay. Diisovaleryl and diisobutyryl analogs were found to have the strongest activity against F. columnare (isolate ALM-00-173) based on 24-h 50% inhibitory concentration (IC50) and minimum inhibitory concentration (MIC). Diisovaleryl and diisobutyryl analogs had IC50 values 0.82 mg/L and 0.80 mg/L, respectively, whereas the drug control florfenicol had an IC50 value of 0.81 mg/L. Diisovaleryl and diisobutyryl analogs also had 24-h relative-to- drug-control IC50 values around 1.0 indicating activities similar to florfenicol, which is included in medicated feed and is one of the current management approach for columnaris.展开更多

Materials and Methods: A research was performed in 1-inch long tilapia stimulated its immunity by infection with Flavobacterium columnare, VETSV01. Pathogenesis of 0.2 mL Flavobacterium columnare, VETSV01 in the range of 10^2-10^8 mL-1 fish^-1 was injected into its intraperitoneal injection and intramuscular injection and suitable concentrations were screened to obtain for further experiment in a late restage. Based on data of minimal concentration, 50% mortality rate was caused and it was found the concentration of 108 mL^-1 fish^-1 caused 100% mortality rate and no lethal rate was found in other concentrations. Afterwards, efficacy of bio-floc sediment on survival rate of Tilapia infected with Flavobacterium columnare, VETSV01 was examined. Completely randomized design was employed and experiments were divided into 4 treatment: (1) contxol-commercial feed only, (2) commercial feed attached with dried bio-floc, (3) commercial feed attached with bio-floc and beta-glucan and (4) commercial feed attached with bio-floc and nucleotide. Experiment design was performed as 3 replicates with 10 fishes/replicates in 24 inch glass aquarium. Analysis of difference was performed and determination of mean differences of mortality rate mad relative percent survival was done through Duncan's Multiple Range Test (p 〈 0.05). Results and Discussions: It was demonstrated feeding tilapia with commercial feed in combination with dried bio-floc with or without supplementation of 2 immuno-stimulants could lead to 100% survival rate and 100% relative percent survival rate and it was statistically significant different (p 〈 0.05) compared to control group. Bio-floc product supplemented with beta glucan or nucleotide could effectively stimulate immunity against Flavobacterium columnare, VETSV01 and such high effective performance is viewed as a promising potential product to be further developed and practically employed for sake of aquaculture industry in the near future.展开更多
Flavobacterium columnare is a Gram-negative bacterium that causes columnaris disease in freshwater fish worldwide.Many studies have focused on the identification of protective antigens to aid in the development of nov...Flavobacterium columnare is a Gram-negative bacterium that causes columnaris disease in freshwater fish worldwide.Many studies have focused on the identification of protective antigens to aid in the development of novel vaccines against the disease.In this study,an immunoblotting approach was employed to identify immunogenic outer membrane proteins(OMPs)from F.columnare in two-dimensional electrophoresis(2-DE)map gels using antibacterial sera obtained from grass carp(Ctenopharyngodon idella),and anti-grass carp-recombinant Ig(rIg)monoclonal antibodies.Five unique immunogenic proteins,including the gliding motility lipoprotein GldJ(GldJ),hypothetical protein FCOL_13420(Fco1),lipoprotein(Lip),F0F1 ATP synthase subunit beta(F0f1)and outer membrane efflux protein precursor(Omep),were characterized.Over-expression of these proteins in Escherichia coli DE3,and their immunogenicity and protective efficacy were evaluated in grass carp.The relative percent survival(RPS)of the groups immunized separately with recombinant GldJ,Lip and Omep was 72%,64%and 68%,respectively when compared to control fish.Up-regulation of immuno-related genes and specific antibodies were detected in immunized fish and sera of immunized fish inhibited the growth of F.columnare.The results suggest that GldJ,Lip and Omep are major protective antigens and may be considered as novel candidates in the development of vaccines against columnaris disease in fish.展开更多
Control of the columnar to equiaxed transition(CET)is a major challenge in additively manufacturedβtitanium alloys.In this work,the promotion of CET was successfully achieved through in-situ fabrication of Ti-5Cu(wt....Control of the columnar to equiaxed transition(CET)is a major challenge in additively manufacturedβtitanium alloys.In this work,the promotion of CET was successfully achieved through in-situ fabrication of Ti-5Cu(wt.%)alloys with additions of 5,15,and 25 wt.%Nb using elemental Ti,Cu,and Nb powders by employing laser powder bed fusion(LPBF).The alloy containing 5 wt.%Nb consisted ofαlamellae,Ti2 Cu precipitates,and unmeltedβ-Nb inclusions,whereas the 25 wt.%Nb alloy consisted of equiaxedβgrains,ωprecipitates,and Ti2 Cu precipitates at the grain boundaries.In terms of mechanical proper-ties,despite the presence of Nb inclusions and liquation cracks in the 5 wt.%Nb alloy,it showed a yield strength of 1051±40 MPa and an elongation of 5.2%±1.3%.Both the strength and ductility decreased with increasing Nb content,e.g.,the 25 wt.%Nb alloy exhibited a yield strength of 808±53 MPa and an elongation of 1.6%±0.2%.As the Nb content increased from 5 to 25 wt.%,the Young’s modulus decreased from 110 to 65 GPa.The 25 wt.%Nb alloy showed a high ratio of hardness to Young’s mod-ulus(H/E)and yield pressure(H3/E2).However,due to its brittle nature,the material manifested high wear rates.These findings provide a basis for the future development of novel low-modulus isotropicβ-titanium alloys using LPBF.展开更多
The pure Mg with columnar crystals was prepared by directional solidification,and the effect of process parameters on the crystal orientation and tensile properties was studied.Moreover,the microstructure evolution du...The pure Mg with columnar crystals was prepared by directional solidification,and the effect of process parameters on the crystal orientation and tensile properties was studied.Moreover,the microstructure evolution during tensile deformation was analyzed by electron backscatter diffraction(EBSD)technology.Furthermore,the slip within adjacent grains and grain boundary strain were discussed using the bicrystal model theory.The results show that the microstructure of the pure Mg at a pulling rate of 200μm/s is columnar polycrystal with growth orientation concentrated in<022ˉ5>,and no transverse grain boundaries can be seen.In addition,the Schmid factors(SFs)of basalslips in columnar crystals are higher than 0.43 under tensile stress.Moreover,the geometric compatibility factor of slip systems on both sides of grain boundaries is greater than 0.7,showing good strain coordination ability of grain boundaries.Therefore,the elongation of the directionally solidified pure Mg is as high as 53%at room temperature.展开更多
The careful observation and evaluation of the squamocolumnar junction(SCJ)are essential requirements in colposcopy procedures.While traditional colposcopes with visible light offer high resolution and sensitivity,the ...The careful observation and evaluation of the squamocolumnar junction(SCJ)are essential requirements in colposcopy procedures.While traditional colposcopes with visible light offer high resolution and sensitivity,the presence of glare patterns on the cervix limits the visualization of deeper tissue features.Cross-polarization imaging,on the other hand,overcomes this limitation by effectively removing glare and providing greater optical penetration depth to noninvasively examine subsurface tissue structures.In this study,we present the potential of polarized light in enhancing diagnostics through cross-polarization imaging for the identification and contrast enhancement of squamous and columnar epithelium(CE)in cases of cervical ectropion.Our results demonstrate that polarized light colposcopy offers valuable diagnostic information that complements traditional colposcopy.By effectively reducing glare and improving visibility during cervical examinations,polarized light colposcopy proves to be a useful tool.Furthermore,we introduce a novel method that practically enhances the contrast ratio(CR)between columnar and squamous epithelium(SE)in colposcopic images.This method significantly increases the contrast between these tissue types,facilitating clearer differentiation and improving diagnostic accuracy.Notably,the combination of the cross-polarization imaging technique with our proposed algorithm enables the clear observation of the SCJ boundary.These findings emphasize the potential of our approach in enhancing the accuracy and effectiveness of polarized colposcopy for evaluating cervical tissue.展开更多
The role of mantle plume in the final stages of rifting of the East Gondwana crustal fragments remains equivocal with only limited evidence so far reported from the southern part of Peninsular India.Here,we report for...The role of mantle plume in the final stages of rifting of the East Gondwana crustal fragments remains equivocal with only limited evidence so far reported from the southern part of Peninsular India.Here,we report for the first time a suite of columnar basalts from the Mesoarchean Coorg Block in the Southern Granulite Terrain(SGT)of India and characterize these rocks through field,petrological,geo-chemical,and isotope geochronological studies.The basalts show porphyritic texture with phenocrysts of pyroxene and plagioclase embedded in fine groundmass.Geochemical data reveal tholeiitic flood basalt affinity with affinities of plume-related magmatism.The zircon U-Pb data of the rocks yield a weighted mean age of 137 Ma,thus corresponding to the Valanginian Age of the Early Cretaceous Period.We suggest the possible geochemical affinity of the studied rocks Kerguelen plume basalts which provide new insights into magmatism associated with the final stages of East Gondwana rifting.展开更多

The use of water resources for energy generation has become increasingly prevalent,encompassing the conversion of kinetic energy from streams,tides,and waves into renewable electrical power.Water energy sources offer ...The use of water resources for energy generation has become increasingly prevalent,encompassing the conversion of kinetic energy from streams,tides,and waves into renewable electrical power.Water energy sources offer numerous benefits,including widespread availability,stability,and the absence of carbon dioxide and other greenhouse gas emissions,making them a clean and environmentally friendly form of energy.In this work,we develop a droplet-based liquid-solid triboelectric nanogenerator(LS-TENG)using sophisticatedly designed inflatable columnar structures with inner and outer dual-electrodes.This device can be utilized to harvest both the internal droplet-rolling mechanical energy and the external droplet-falling mechanical energy,capable of being assembled into various structures for versatile applications.The design incorporates a combined structure of both internal and external TENG to optimize output performance via multiple energy harvesting strategies.The internal structure features a dual-electrode columnar-shaped LS-TENG,designed to harvest fluid kinetic energy from water droplets.By leveraging the back-and-forth motion of a small amount of water within the air column,mechanical energy can be readily collected,achieving a maximum mass power density of 9.02 W·Kg^(−1)and an energy conversion efficiency of 10.358%.The external component is a droplet-based LS-TENG,which utilizes a double-layer capacitor switch effect elucidated with an equivalent circuit model.Remarkably,without the need for pre-charging,a single droplet can generate over 140 V of high voltage,achieving a maximum power density of 7.35 W·m^(−2)and an energy conversion efficiency of 22.058%.The combined LS-TENG with a sophisticated inflatable columnar structure can simultaneously collect multiple types of energy with high efficacy,exhibiting great significance in potential applications such as TENG aeration rollers,inflatable lifejacket,wind energy harvesting,TENG tents,and green houses.展开更多
Rock masses are often exposed to dynamic loads such as earthquakes and mechanical disturbances in practical engineering scenarios.The existence of underground caverns and weak geological structures like columnar joint...Rock masses are often exposed to dynamic loads such as earthquakes and mechanical disturbances in practical engineering scenarios.The existence of underground caverns and weak geological structures like columnar jointed rock masses(CJRMs)and interlayer shear weakness zones(ISWZs)with inferior mechanical properties,significantly undermines the overall structural stability.To tackle the dynamic loading issues in the process of constructing subterranean caverns,a programmable modeling approach was utilized to reconstruct a large-scale underground cavern model incorporating ISWZs and columnar joints(CJs).By conducting dynamic simulations with varying load orientations,the analyses focused on the failure patterns,deformation characteristics,and acoustic emission activity within the caverns.Results revealed that the failure modes of the underground caverns under dynamic loading were predominantly tensile failures.Under X-direction loading,the failed elements were mainly distributed parallel to the CJs,while under Y-direction loading,they were distributed parallel to the transverse weak structural planes.Furthermore,the dynamic stability of the overall structure varied with the number of caverns.The dual-cavern model demonstrated the highest stability under X-direction loading,while the single-cavern model was the least stable.Under Y-direction loading,the cavern stability increased with the number of caverns.Importantly,different weak structures affected the dynamic response of caverns in different ways;the CJRMs were the primary contributors to structural failure,while ISWZs could mitigate the rock mass failure induced by CJs.The findings could offer valuable insights for the dynamic stability analysis of caverns containing CJRMs and ISWZs.展开更多

The detaching and floating of fragments of dendrite arms are the prelude of the transition when the solute boundary layer in front of the solid-liquid interface is thin. And the dendrite triangular tip is the fracture sensitive zone. When the conditions are suitable, new dendrites can sprout and grow up. As in other vertebrate CD3 molecules, the deduced amino acid sequences of mandarin fish CD3γ/δ and CD3ε contained conserved residues and motifs, such as cysteine residues and CXXC and immunoreceptor tyrosine-based activation motifs. However, mandarin fish CD3γ/δ and CD3ε showed some differences to their mammalian counterparts, specifically the absence of a negatively charged residue in the transmembrane region of CD3γ/δ. Additionally, while an N-glycosylation site was present in CD3c, the site was not observed in CD3γ/δ. The CD3γ/δ and CD3ε subunit sequences contain six and five exons, respectively, consistent with homologues from Atlantic salmon, Salmo salar. Phylogenetic analysis also revealed that CD3γ/δ and CD3ε in mandarin fish are closely related to their counterparts in Acanthopterygian fish. Real-time PCR showed CD3γ/δ and CD3ε were expressed mainly in the thymus and spleen in normal healthy fish and, to a lesser extent, in mucosal-associated lymphoid tissues, such as the intestine and gills. When lymphocytes isolated from head kidney were treated with the mitogens phytohemagglutinin, concanavalin, and polyriboinosinic polyribocytidylic acid, mRNA expression levels of CD3γ/δ and CD3ε were significantly elevated within 12 h of treatment. This indicated the presence of T lymphocytes in the head kidney of teleost fish, and also the recognition of mitogens by the lymphocytes. Mandarin fish infected with the bacterial pathogen Flavobacterium columnare also showed an increase in the expression of CD3γ/δ and CD3ε mR_NA, indicating that CD3γ/δ and CD3ε lymphocytes are involved in the immune response of this species.展开更多

A select screening of fish pathogens, freshwater and human isolates identified several other species that responded similarly to exogenous protein. The optimal protein concentration was species specific;S. marcescens at 5% milk protein, Aeromonas sp. at 2% - 3%, Flavobacterium columnare at 1% and Pseudomonas aeruginosa at 0.1% - 0.4%. Media supplemented with milk protein also increased the cell counts in biofilm as well as the protein incorporated into the biofilm matrix. These data suggest that relatively high concentrations of exogenous protein may serve as an environmental trigger for biofilm formation, particularly for pathogenic bacteria exposed to relatively high concentrations of protein in bodily fluids and mucosal surfaces.展开更多

However, it is still controversial o...The West Junggar of the western Central Asian Orogenic Belt is one of the typical regions in the term of ocean subduction, contraction and continental growth in the Late Paleozoic. However, it is still controversial on the exact time of ocean-continent transition so far. This study investigates rhyolites with columnar joint in the West Junggar for the first time.Based on zircon U-Pb dating, we determined that the ages of the newly-discovered rhyolites are between 303.6 and 294.5 Ma, belonging to Late Carboniferous–Early Permian, which is the oldest rhyolite with columnar joint preserved in the world at present. Geochemical results show that the characteristics of the major element compositions include a high content of SiO_2(75.78–79.20 wt%) and a moderate content of Al_2O_3(12.21–13.19 wt%). The total alkali content(K_2O +Na_2O) is 6.14–8.05 wt%, among which K_2O is 2.09–4.72 wt% and the rate of K_2O/Na_2O is 0.38–3.05. Over-based minerals such as Ne, Lc, and Ac do not appear. The contents of TiO_2(0.09–0.24 wt%), CaO(0.15–0.99 wt%) and MgO(0.06–0.18 wt%) are low. A/CNK=0.91–1.68, A/NK=1.06–1.76, and as such, these are associated with the quasi-aluminum-weak peraluminous high potassium calc-alkaline and some calc-alkaline magma series. These rhyolites show a significant negative Eu anomaly with relative enrichment of LREE and LILE(Rb, Ba, Th, U, K) and depletion of Sr, HREE and HFSE(Nb, Ta, Ti, P). These rhyolites also have the characteristics of an A2-type granite, similar to the Miaoergou batholith,which indicates they both were affected by post-orogenic extension. Combining petrological, zircon U-Pb dating and geochemical characteristics of the rhyolites, we conclude that the specific time of ocean-continent transition of the West Junggar is the Late Carboniferous–Early Permian.展开更多

Furthermore, a program to convert the AUTOCAD model into 3DEC (3-dimensional distinct element code) model was developed, and a numerical model was established in 3DEC. Moreover, the results of triaxial compression tests of columnar jointed rock masses were simulated numerically. The REV (representative element volume) scale was studied, and the result shows that the REV size is 3 m × 3 m. The proposed approach, the established model and the numerical simulation were applied to study the macro-mechanical properties and the equivalent strength parameters of the columnar jointed rock mass. The numerical simulation results are in good accordance with the in-situ test results.展开更多

文摘Flavobacterium columnare causes columnaris disease in freshwater fi sh. In the present study, the antigenic regions of fi ve outer membrane proteins(OMPs), including zinc metalloprotease, prolyl oligopeptidase, thermolysin, collagenase and chondroitin AC lyase, were bioinformatically analyzed, fused together, and then expressed as a recombinant fusion protein in Escherichia coli. The expressed protein of 95.6 k Da, as estimated by 10% sodium dodecyl sulfate-polyacrylamide gel electrophoresis, was consistent with the molecular weight deduced from the amino acid sequence. The purifi ed recombinant protein was used to vaccinate the grass carp, C tenopharyngodon idella. Following vaccination of the fi sh their Ig M antibody levels were examined, as was the expression of I g M, Ig D and Ig Z immunoglobulin genes and other genes such as MHC Iα and MHC I I β, which are also involved in adaptive immunity. Interleukin genes( IL), including I L- 1β, IL- 8 and I L- 10, and type I and type II interferon(I FN) genes were also examined. At 3 and 4 weeks post-vaccination(wpv), signifi cant increases in Ig M antibody levels were observed in the fi sh vaccinated with the recombinant fusion protein, and an increase in the expression levels of I g M, Ig D and Ig Z genes was also detected following the vaccinations, thus indicating that an adaptive immune response was induced by the vaccinations. Early increases in the expression levels of IL and IFN genes were also observed in the vaccinated fi sh. At four wpv, the fi sh were challenged with F. column a re, and the vaccinated fi sh showed a good level of protection against this pathogen, with 39% relative percent survival(RPS) compared with the control group. It can be concluded, therefore, that the fi ve OMPs, in the form of a recombinant fusion protein vaccine, induced an immune response in fi sh and protection against F. columnare.
文摘From the ethyl acetate extract of Murraya koenegii (Rutaceae) leaves, isomahanine (1) and mahanine (2) were isolated that showed antibacterial activity towards Flavobacterium columnare and Streptococcus iniae which caused columnaris disease and streptococcosis respectively. Isomahanine was found to have the strongest activity against F. columnare (isolate ALM-00-173) and S. iniae (isolate LA94-426) based on 24-h 50% inhibition concentration (IC50) and minimum inhibition concentration (MIC). Although compound (7), a nicotinamide isolated from Amyris texana had the lowest MIC (2.8 ± 0 mg/L) of any of the test compounds against F. columnare, the 24-h IC50 of 14.8 ± 0.6 mg/L was higher than that of isomahanine and subsequently the 24-h IC50 RDC values for (7) were almost a magnitude of order higher than those obtained for isomahanine. Isomahanine also had the strongest activity against S. iniae, with a 24-h IC50 of 1.3 ± 0.1 mg/L and MIC of 3.5 ± 0 mg/L, respectively.

文摘Flavobacterium columnare is the pathogenic agent of columnaris disease in aquaculture. Using a recently developed gene deletion strategy, two genes that encode the Glyco hydro_19 domain (GH19 domain) containing proteins, ghd-1 and ghd-2, were deleted separately and together from the F. columnare G4 wild type strain. Surprisingly, the single-, Aghd-1 and Aghd-2, and double-gene mutants, Aghd-1 Aghd-2, all had rhizoid and non-rhizoid colony morphotypes, which we named Aghd-1, Aghd-2, Aghd-1 Aghd-2, and NAghd-1, NAghd-2, and NAghd-1 Aghd-2. However, chitin utilization was not detected in either these mutants or in the wild type. Instead, skimmed milk degradation was observed for the mutants and the wild type; the non-rhizoid strain NAghd-2 exhibited higher degradation activity as revealed by the larger transparent circle on the skimmed milk plate. Using zebrafish as the model organism, we found that non-rhizoid mutants had higher LDs0 values and were less virulent because zebrafish infected with these survived longer. Transcriptome analysis between the non-rhizoid and rhizoid colony morphotypes of each mutant, i.e., NAghd-1 versus (vs) Aghd-1, NAghd-2 vs Aghd-2, and NAghd-1 Aghd-2 vs Aghd-1 Aghd-2, revealed a large number of differentially expressed genes, among which 39 genes were common in three of the pairs compared. Although most of these genes encode hypothetical proteins, a few molecules such as phage tail protein, rhs element Vgr protein, thiol-activated cytolysin, and TonB-dependent outer membrane receptor precursor, expression of which was down-regulated in non-rhizoid mutants but up-regulated in rhizoid mutants, may play a role F. columnare virulence.

文摘Flavobacterium columnare, the etiological agent of colunmaris disease, is one of the most important and widespread bacterial pathogens of freshwater fish. In this study, we constructed two artificial selectable markers (chloramphenicol and spectinomycin resistance) for gene transfer in F. columnare. These two new artificial selectable markers, which were created by placing the chloramphenicol or spectinomycin resistance gene under the control of the native acs regulatory region of F. columnare, were functional in both F. columnare and Escherichia coli. The integrative/conjugative plasmids constructed by using these markers were introduced into F. columnare G4 via electroporation or conjugation. The integrated plasmid DNA was confirmed by Southern blotting and PCR analysis. These two markers can be employed in future investigations into gene deletion and the pathogenicity of virulence factors in F. columnare.
文摘Columnaris (caused by Flavobacterium columnare) is one of the most common bacterial diseases affecting the pond-raised channel catfish (Ictalurus punctatus) in the southeastern United States of America resulting in annual losses of millions of dollars. As part of our continuing effort to discover environmentally benign compounds for the control of columnaris disease, acyl derivatives of phloroglucinol were synthesized and tested against F. columnare using a rapid bioassay. Among the analogs that were tested, diacyl analogs showed very high antibacterial activity against F. columnare in the laboratory bioassay. Diisovaleryl and diisobutyryl analogs were found to have the strongest activity against F. columnare (isolate ALM-00-173) based on 24-h 50% inhibitory concentration (IC50) and minimum inhibitory concentration (MIC). Diisovaleryl and diisobutyryl analogs had IC50 values 0.82 mg/L and 0.80 mg/L, respectively, whereas the drug control florfenicol had an IC50 value of 0.81 mg/L. Diisovaleryl and diisobutyryl analogs also had 24-h relative-to- drug-control IC50 values around 1.0 indicating activities similar to florfenicol, which is included in medicated feed and is one of the current management approach for columnaris.
文摘Objective: The aim of the research was to investigate efficacy of dried bio-floc supplemented with immuno-stimulants as beta-glucan and nucleotide on mortality rate and relative percent survival (RPS) of tilapia infected with Flavobacterium columnare, VETSV01. Materials and Methods: A research was performed in 1-inch long tilapia stimulated its immunity by infection with Flavobacterium columnare, VETSV01. Pathogenesis of 0.2 mL Flavobacterium columnare, VETSV01 in the range of 10^2-10^8 mL-1 fish^-1 was injected into its intraperitoneal injection and intramuscular injection and suitable concentrations were screened to obtain for further experiment in a late restage. Based on data of minimal concentration, 50% mortality rate was caused and it was found the concentration of 108 mL^-1 fish^-1 caused 100% mortality rate and no lethal rate was found in other concentrations. Afterwards, efficacy of bio-floc sediment on survival rate of Tilapia infected with Flavobacterium columnare, VETSV01 was examined. Completely randomized design was employed and experiments were divided into 4 treatment: (1) contxol-commercial feed only, (2) commercial feed attached with dried bio-floc, (3) commercial feed attached with bio-floc and beta-glucan and (4) commercial feed attached with bio-floc and nucleotide. Experiment design was performed as 3 replicates with 10 fishes/replicates in 24 inch glass aquarium. Analysis of difference was performed and determination of mean differences of mortality rate mad relative percent survival was done through Duncan's Multiple Range Test (p 〈 0.05). Results and Discussions: It was demonstrated feeding tilapia with commercial feed in combination with dried bio-floc with or without supplementation of 2 immuno-stimulants could lead to 100% survival rate and 100% relative percent survival rate and it was statistically significant different (p 〈 0.05) compared to control group. Bio-floc product supplemented with beta glucan or nucleotide could effectively stimulate immunity against Flavobacterium columnare, VETSV01 and such high effective performance is viewed as a promising potential product to be further developed and practically employed for sake of aquaculture industry in the near future.
文摘Flavobacterium columnare is a Gram-negative bacterium that causes columnaris disease in freshwater fish worldwide.Many studies have focused on the identification of protective antigens to aid in the development of novel vaccines against the disease.In this study,an immunoblotting approach was employed to identify immunogenic outer membrane proteins(OMPs)from F.columnare in two-dimensional electrophoresis(2-DE)map gels using antibacterial sera obtained from grass carp(Ctenopharyngodon idella),and anti-grass carp-recombinant Ig(rIg)monoclonal antibodies.Five unique immunogenic proteins,including the gliding motility lipoprotein GldJ(GldJ),hypothetical protein FCOL_13420(Fco1),lipoprotein(Lip),F0F1 ATP synthase subunit beta(F0f1)and outer membrane efflux protein precursor(Omep),were characterized.Over-expression of these proteins in Escherichia coli DE3,and their immunogenicity and protective efficacy were evaluated in grass carp.The relative percent survival(RPS)of the groups immunized separately with recombinant GldJ,Lip and Omep was 72%,64%and 68%,respectively when compared to control fish.Up-regulation of immuno-related genes and specific antibodies were detected in immunized fish and sera of immunized fish inhibited the growth of F.columnare.The results suggest that GldJ,Lip and Omep are major protective antigens and may be considered as novel candidates in the development of vaccines against columnaris disease in fish.

文摘Control of the columnar to equiaxed transition(CET)is a major challenge in additively manufacturedβtitanium alloys.In this work,the promotion of CET was successfully achieved through in-situ fabrication of Ti-5Cu(wt.%)alloys with additions of 5,15,and 25 wt.%Nb using elemental Ti,Cu,and Nb powders by employing laser powder bed fusion(LPBF).The alloy containing 5 wt.%Nb consisted ofαlamellae,Ti2 Cu precipitates,and unmeltedβ-Nb inclusions,whereas the 25 wt.%Nb alloy consisted of equiaxedβgrains,ωprecipitates,and Ti2 Cu precipitates at the grain boundaries.In terms of mechanical proper-ties,despite the presence of Nb inclusions and liquation cracks in the 5 wt.%Nb alloy,it showed a yield strength of 1051±40 MPa and an elongation of 5.2%±1.3%.Both the strength and ductility decreased with increasing Nb content,e.g.,the 25 wt.%Nb alloy exhibited a yield strength of 808±53 MPa and an elongation of 1.6%±0.2%.As the Nb content increased from 5 to 25 wt.%,the Young’s modulus decreased from 110 to 65 GPa.The 25 wt.%Nb alloy showed a high ratio of hardness to Young’s mod-ulus(H/E)and yield pressure(H3/E2).However,due to its brittle nature,the material manifested high wear rates.These findings provide a basis for the future development of novel low-modulus isotropicβ-titanium alloys using LPBF.

文摘The pure Mg with columnar crystals was prepared by directional solidification,and the effect of process parameters on the crystal orientation and tensile properties was studied.Moreover,the microstructure evolution during tensile deformation was analyzed by electron backscatter diffraction(EBSD)technology.Furthermore,the slip within adjacent grains and grain boundary strain were discussed using the bicrystal model theory.The results show that the microstructure of the pure Mg at a pulling rate of 200μm/s is columnar polycrystal with growth orientation concentrated in<022ˉ5>,and no transverse grain boundaries can be seen.In addition,the Schmid factors(SFs)of basalslips in columnar crystals are higher than 0.43 under tensile stress.Moreover,the geometric compatibility factor of slip systems on both sides of grain boundaries is greater than 0.7,showing good strain coordination ability of grain boundaries.Therefore,the elongation of the directionally solidified pure Mg is as high as 53%at room temperature.
文摘The careful observation and evaluation of the squamocolumnar junction(SCJ)are essential requirements in colposcopy procedures.While traditional colposcopes with visible light offer high resolution and sensitivity,the presence of glare patterns on the cervix limits the visualization of deeper tissue features.Cross-polarization imaging,on the other hand,overcomes this limitation by effectively removing glare and providing greater optical penetration depth to noninvasively examine subsurface tissue structures.In this study,we present the potential of polarized light in enhancing diagnostics through cross-polarization imaging for the identification and contrast enhancement of squamous and columnar epithelium(CE)in cases of cervical ectropion.Our results demonstrate that polarized light colposcopy offers valuable diagnostic information that complements traditional colposcopy.By effectively reducing glare and improving visibility during cervical examinations,polarized light colposcopy proves to be a useful tool.Furthermore,we introduce a novel method that practically enhances the contrast ratio(CR)between columnar and squamous epithelium(SE)in colposcopic images.This method significantly increases the contrast between these tissue types,facilitating clearer differentiation and improving diagnostic accuracy.Notably,the combination of the cross-polarization imaging technique with our proposed algorithm enables the clear observation of the SCJ boundary.These findings emphasize the potential of our approach in enhancing the accuracy and effectiveness of polarized colposcopy for evaluating cervical tissue.

文摘The role of mantle plume in the final stages of rifting of the East Gondwana crustal fragments remains equivocal with only limited evidence so far reported from the southern part of Peninsular India.Here,we report for the first time a suite of columnar basalts from the Mesoarchean Coorg Block in the Southern Granulite Terrain(SGT)of India and characterize these rocks through field,petrological,geo-chemical,and isotope geochronological studies.The basalts show porphyritic texture with phenocrysts of pyroxene and plagioclase embedded in fine groundmass.Geochemical data reveal tholeiitic flood basalt affinity with affinities of plume-related magmatism.The zircon U-Pb data of the rocks yield a weighted mean age of 137 Ma,thus corresponding to the Valanginian Age of the Early Cretaceous Period.We suggest the possible geochemical affinity of the studied rocks Kerguelen plume basalts which provide new insights into magmatism associated with the final stages of East Gondwana rifting.

文摘The special columnar jointed structure endows rocks with significant anisotropy,accurately grasping the strength and deformation properties of a columnar jointed rock mass(CJRM)under complex geological conditions is crucial for related engineering safety.Combined with the irregular jointed networks observed in the field,artificial irregular CJRM(ICJRM)samples with various inclination angles were prepared for triaxial tests.The results showed that the increase in confining pressure can enhance the ability of the ICJRM to resist deformation and failure,and reduce the deformation and strength anisotropic degrees.Considering the field stress situation,the engineering parts with an inclination angle of 30°−45°need to be taken seriously.Four typical failure modes were identified,and the sample with an inclination angle of 15°showed the same failure behavior as the field CJRM.Traditional and improved joint factor methods were used to establish empirical relationships for predicting the strength and deformation of CJRM under triaxial stress.Since the improved joint factor method can reflect the unique structure of CJRM,the predictive ability of the empirical relationship based on the improved method is better than that based on the traditional joint factor method.

文摘The use of water resources for energy generation has become increasingly prevalent,encompassing the conversion of kinetic energy from streams,tides,and waves into renewable electrical power.Water energy sources offer numerous benefits,including widespread availability,stability,and the absence of carbon dioxide and other greenhouse gas emissions,making them a clean and environmentally friendly form of energy.In this work,we develop a droplet-based liquid-solid triboelectric nanogenerator(LS-TENG)using sophisticatedly designed inflatable columnar structures with inner and outer dual-electrodes.This device can be utilized to harvest both the internal droplet-rolling mechanical energy and the external droplet-falling mechanical energy,capable of being assembled into various structures for versatile applications.The design incorporates a combined structure of both internal and external TENG to optimize output performance via multiple energy harvesting strategies.The internal structure features a dual-electrode columnar-shaped LS-TENG,designed to harvest fluid kinetic energy from water droplets.By leveraging the back-and-forth motion of a small amount of water within the air column,mechanical energy can be readily collected,achieving a maximum mass power density of 9.02 W·Kg^(−1)and an energy conversion efficiency of 10.358%.The external component is a droplet-based LS-TENG,which utilizes a double-layer capacitor switch effect elucidated with an equivalent circuit model.Remarkably,without the need for pre-charging,a single droplet can generate over 140 V of high voltage,achieving a maximum power density of 7.35 W·m^(−2)and an energy conversion efficiency of 22.058%.The combined LS-TENG with a sophisticated inflatable columnar structure can simultaneously collect multiple types of energy with high efficacy,exhibiting great significance in potential applications such as TENG aeration rollers,inflatable lifejacket,wind energy harvesting,TENG tents,and green houses.

文摘Rock masses are often exposed to dynamic loads such as earthquakes and mechanical disturbances in practical engineering scenarios.The existence of underground caverns and weak geological structures like columnar jointed rock masses(CJRMs)and interlayer shear weakness zones(ISWZs)with inferior mechanical properties,significantly undermines the overall structural stability.To tackle the dynamic loading issues in the process of constructing subterranean caverns,a programmable modeling approach was utilized to reconstruct a large-scale underground cavern model incorporating ISWZs and columnar joints(CJs).By conducting dynamic simulations with varying load orientations,the analyses focused on the failure patterns,deformation characteristics,and acoustic emission activity within the caverns.Results revealed that the failure modes of the underground caverns under dynamic loading were predominantly tensile failures.Under X-direction loading,the failed elements were mainly distributed parallel to the CJs,while under Y-direction loading,they were distributed parallel to the transverse weak structural planes.Furthermore,the dynamic stability of the overall structure varied with the number of caverns.The dual-cavern model demonstrated the highest stability under X-direction loading,while the single-cavern model was the least stable.Under Y-direction loading,the cavern stability increased with the number of caverns.Importantly,different weak structures affected the dynamic response of caverns in different ways;the CJRMs were the primary contributors to structural failure,while ISWZs could mitigate the rock mass failure induced by CJs.The findings could offer valuable insights for the dynamic stability analysis of caverns containing CJRMs and ISWZs.

文摘Directional solidification of Al-15% (mass fraction) Cu alloy was investigated by in situ and real time radiography which was performed by Shanghai synchrotron radiation facility (SSRF). The imaging results reveal that columnar to equiaxed transition (CET) is provoked by external thermal disturbance. The detaching and floating of fragments of dendrite arms are the prelude of the transition when the solute boundary layer in front of the solid-liquid interface is thin. And the dendrite triangular tip is the fracture sensitive zone. When the conditions are suitable, new dendrites can sprout and grow up. This kind of dendrite has no obvious stem and is named anaxial columnar dendrites.

文摘The genomic and cDNA sequences of the CD3γ/δ and CD3ε homologues in the mandarin fish, Siniperca chuatsi, were determined. As in other vertebrate CD3 molecules, the deduced amino acid sequences of mandarin fish CD3γ/δ and CD3ε contained conserved residues and motifs, such as cysteine residues and CXXC and immunoreceptor tyrosine-based activation motifs. However, mandarin fish CD3γ/δ and CD3ε showed some differences to their mammalian counterparts, specifically the absence of a negatively charged residue in the transmembrane region of CD3γ/δ. Additionally, while an N-glycosylation site was present in CD3c, the site was not observed in CD3γ/δ. The CD3γ/δ and CD3ε subunit sequences contain six and five exons, respectively, consistent with homologues from Atlantic salmon, Salmo salar. Phylogenetic analysis also revealed that CD3γ/δ and CD3ε in mandarin fish are closely related to their counterparts in Acanthopterygian fish. Real-time PCR showed CD3γ/δ and CD3ε were expressed mainly in the thymus and spleen in normal healthy fish and, to a lesser extent, in mucosal-associated lymphoid tissues, such as the intestine and gills. When lymphocytes isolated from head kidney were treated with the mitogens phytohemagglutinin, concanavalin, and polyriboinosinic polyribocytidylic acid, mRNA expression levels of CD3γ/δ and CD3ε were significantly elevated within 12 h of treatment. This indicated the presence of T lymphocytes in the head kidney of teleost fish, and also the recognition of mitogens by the lymphocytes. Mandarin fish infected with the bacterial pathogen Flavobacterium columnare also showed an increase in the expression of CD3γ/δ and CD3ε mR_NA, indicating that CD3γ/δ and CD3ε lymphocytes are involved in the immune response of this species.
文摘A screening of environmental conditions that would elicit robust biofilm in a collection of Serratia marcescens isolated from soil revealed that exogenous milk protein increased biofilm productivity up to ten-fold. A select screening of fish pathogens, freshwater and human isolates identified several other species that responded similarly to exogenous protein. The optimal protein concentration was species specific;S. marcescens at 5% milk protein, Aeromonas sp. at 2% - 3%, Flavobacterium columnare at 1% and Pseudomonas aeruginosa at 0.1% - 0.4%. Media supplemented with milk protein also increased the cell counts in biofilm as well as the protein incorporated into the biofilm matrix. These data suggest that relatively high concentrations of exogenous protein may serve as an environmental trigger for biofilm formation, particularly for pathogenic bacteria exposed to relatively high concentrations of protein in bodily fluids and mucosal surfaces.

文摘The West Junggar of the western Central Asian Orogenic Belt is one of the typical regions in the term of ocean subduction, contraction and continental growth in the Late Paleozoic. However, it is still controversial on the exact time of ocean-continent transition so far. This study investigates rhyolites with columnar joint in the West Junggar for the first time.Based on zircon U-Pb dating, we determined that the ages of the newly-discovered rhyolites are between 303.6 and 294.5 Ma, belonging to Late Carboniferous–Early Permian, which is the oldest rhyolite with columnar joint preserved in the world at present. Geochemical results show that the characteristics of the major element compositions include a high content of SiO_2(75.78–79.20 wt%) and a moderate content of Al_2O_3(12.21–13.19 wt%). The total alkali content(K_2O +Na_2O) is 6.14–8.05 wt%, among which K_2O is 2.09–4.72 wt% and the rate of K_2O/Na_2O is 0.38–3.05. Over-based minerals such as Ne, Lc, and Ac do not appear. The contents of TiO_2(0.09–0.24 wt%), CaO(0.15–0.99 wt%) and MgO(0.06–0.18 wt%) are low. A/CNK=0.91–1.68, A/NK=1.06–1.76, and as such, these are associated with the quasi-aluminum-weak peraluminous high potassium calc-alkaline and some calc-alkaline magma series. These rhyolites show a significant negative Eu anomaly with relative enrichment of LREE and LILE(Rb, Ba, Th, U, K) and depletion of Sr, HREE and HFSE(Nb, Ta, Ti, P). These rhyolites also have the characteristics of an A2-type granite, similar to the Miaoergou batholith,which indicates they both were affected by post-orogenic extension. Combining petrological, zircon U-Pb dating and geochemical characteristics of the rhyolites, we conclude that the specific time of ocean-continent transition of the West Junggar is the Late Carboniferous–Early Permian.

文摘From the geological structure of the columnar jointed rock mass, a visual model was established in software AUTOCAD by programming based on the algorithm of the Voronoi diagram. Furthermore, a program to convert the AUTOCAD model into 3DEC (3-dimensional distinct element code) model was developed, and a numerical model was established in 3DEC. Moreover, the results of triaxial compression tests of columnar jointed rock masses were simulated numerically. The REV (representative element volume) scale was studied, and the result shows that the REV size is 3 m × 3 m. The proposed approach, the established model and the numerical simulation were applied to study the macro-mechanical properties and the equivalent strength parameters of the columnar jointed rock mass. The numerical simulation results are in good accordance with the in-situ test results.
