Winter wheat (Triticum aestivum) cultivars Dongnongdongmai 1 with strong cold/freezing tolerance and Jimai 22 with weak cold/freezing tolerance were used for investigating the difference of microstructure and ultrastructure between leaves of two cultivars under low temperature stress (5℃ and -15 ℃) using optical and electron microscope. The results showed that there was no obvious difference between leaves of Dongnongdongmai 1 and Jimai 22 in microstructure. However, the difference between those leaves was distinct in ultrastructure. The grana lamella and stroma lamella were stacked regularly and arranged parallelly along the long axis of chloroplasts in cv. Dongnongdongmai 1, while the arrangement directions of thylakoids in Jimai 22's leaves were so irregular as to form various angles with the long axis of chloroplasts. At -15℃, the mitochondrias were swelled to be round and the structure of cristaes became blurry in both cultivars' leaves, while some cristaes of Jimai 22 disappeared. Freezing of gait is a significant and debilitating motor symptom often observed in individuals with Parkinson's disease.Resting-state functional magnetic resonance imaging,along with its multi-level feature indices,has provided a fresh perspective and valuable insight into the study of freezing of gait in Parkinson's disease.It has been revealed that Parkinson's disease is accompanied by widespread irregularities in inherent brain network activity.However,the effective integration of the multi-level indices of resting-state functional magnetic resonance imaging into clinical settings for the diagnosis of freezing of gait in Parkinson's disease remains a challenge.Although previous studies have demonstrated that radiomics can extract optimal features as biomarkers to identify or predict diseases,a knowledge gap still exists in the field of freezing of gait in Parkinson's disease.This cross-sectional study aimed to evaluate the ability of radiomics features based on multi-level indices of resting-state functional magnetic resonance imaging,along with clinical features,to distinguish between Parkinson's disease patients with and without freezing of gait.We recruited 28 patients with Parkinson's disease who had freezing of gait(15 men and 13 women,average age 63 years)and 30 patients with Parkinson's disease who had no freezing of gait(16 men and 14 women,average age 64 years).Magnetic resonance imaging scans were obtained using a 3.0T scanner to extract the mean amplitude of low-frequency fluctuations,mean regional homogeneity,and degree centrality.Neurological and clinical characteristics were also evaluated.We used the least absolute shrinkage and selection operator algorithm to extract features and established feedforward neural network models based solely on resting-state functional magnetic resonance imaging indicators.We then performed predictive analysis of three distinct groups based on resting-state functional magnetic resonance imaging indicators indicators combined with clinical features.Subsequently,we conducted 100 additional five-fold cross-validations to determine the most effective model for each classification task and evaluated the performance of the model using the area under the receiver operating characteristic curve.The results showed that when differentiating patients with Parkinson's disease who had freezing of gait from those who did not have freezing of gait,or from healthy controls,the models using only the mean regional homogeneity values achieved the highest area under the receiver operating characteristic curve values of 0.750(with an accuracy of 70.9%)and 0.759(with an accuracy of 65.3%),respectively.When classifying patients with Parkinson's disease who had freezing of gait from those who had no freezing of gait,the model using the mean amplitude of low-frequency fluctuation values combined with two clinical features achieved the highest area under the receiver operating characteristic curve of 0.847(with an accuracy of 74.3%).The most significant features for patients with Parkinson's disease who had freezing of gait were amplitude of low-frequency fluctuation alterations in the left parahippocampal gyrus and two clinical characteristics:Montreal Cognitive Assessment and Hamilton Depression Scale scores.Our findings suggest that radiomics features derived from resting-state functional magnetic resonance imaging indices and clinical information can serve as valuable indices for the identification of freezing of gait in Parkinson's disease. 展开更多

Sudden temperature drops cause soils in natural environments to freeze unidirectionally,resulting in soil expansion and deformation that can lead to damage to engineering structures.The impact of temperature-induced freezing on deformation and solute migration in saline soils,especially under extended freezing,is not well understood due to the lack of knowledge regarding the microscopic mechanisms involved.This study investigated the expansion,deformation,and water-salt migration in chlorinated saline soils,materials commonly used for canal foundations in cold and arid regions,under different roof temperatures and soil compaction levels through unidirectional freezing experiments.The microscopic structures of saline soils were observed using scanning electron microscopy(SEM)and optical microscopy.A quantitative analysis of the microstructural data was conducted before and after freezing to elucidate the microscopic mechanisms of water-salt migration and deformation.The results indicate that soil swelling is enhanced by elevated roof temperatures approaching the soil's freezing point and soil compaction,which prolongs the duration and accelerates the rate of water-salt migration.The unidirectional freezing altered the microstructure of saline soils due to the continuous temperature gradients,leading to four distinct zones:natural frozen zone,peak frozen zone,gradual frozen zone,and unfrozen zone,each exhibiting significant changes in pore types and fractal dimensions.Vacuum suction at the colder end of the soil structure facilitates the upward migration of salt and water,which subsequently undergoes crystallization.This process expands the internal pore structure and causes swelling.The findings provide a theoretical basis for understanding the evolution of soil microstructure in cold and arid regions and for the management of saline soil engineering. 展开更多

Cold air is one key factor affecting the freezing process of ice tanks.The volume of fluid method is employed to simulate the freezing process of water in an ice tank with cold air inlets.The temperature field distribution in the ice tank is calculated.The temperature field at different typical instants are compared.The main characteristics of the freezing process in an ice tank driven by cold air are analyzed.The influence of the number of cold air inlets on the freezing process is investigated.The numerical results show that with increasing the number of cold air inlets,the temperature field becomes more uniform.The inlet velocity of cold air has a great influence on the generation of ice.This work can serve for the design of the ice tank and control of the freezing process. 展开更多

Role of CaCl_(2) in Cold Acclimation-Induced Freezing Resistance of Populus tomentosa Cuttings 被引量:2

Results indicated that 10?mmol·L -1 of CaCl 2 has greater effect than other concentrations on the enhancement of freezing resistance, and the optimum time of pretreatment was 5?d. In addition, cuttings used for cold acclimation at -3℃ were pretreated with or without 10?mmol·L -1 of CaCl 2, 3?mmol·L -1 of Ca 2+ chelator EGTA, 0 05?mmol·L -1 of CaM antagonist CPZ or 0 1?mmol·L -1 of Ca 2+ channel inhibitor LaCl 3 The changes in CaM and freezing resistance of all cuttings were investigated. The results showed that cold acclimation at -3℃ increased CaM content and decreased the minimum temperature for 100% survival. The CaCl 2 pretreatment enhanced the effect of cold acclimation and obviously increased CaM content and decreased the minimum temperature for 100% survival, but this effect was strongly inhibited by the EGTA, CPZ or LaCl 3 It is concluded that the effect of CaCl 2 on freezing resistance is associated with its concentration and time of pretreatment, Ca 2+ CaM may be involved in the induction of freezing resistance of the cuttings. 展开更多

Results showed that cold acclimation increased the contents of total soluble protein and CaM, and freezing resistance of seedlings, which could be strongly reduced by the pretreatments of EGTA CPZ and LaCl 3 Cold acclimation combined with CaCl 2 pretreatment enhanced the effect of cold acclimation on freezing resistance, and obviously increased the contents of total soluble protein and CaM, reduced the declining degree of the contents of total soluble protein and CaM caused by chilling stress as compared with cold acclimation, augmented the increase in the level of total soluble protein and CaM during the recovery periods. Further analysis found that an increase in total soluble protein content during cold acclimation with or without CaCl 2 pretreatment mainly resulted from the increase in content of heat stable protein in total soluble protein. The changes in the contents of total soluble protein and RNA, the activity of RNase in leaves and branches of Populus tomentosa cuttings at various periods (viz: cold acclimation, deacclimation, chilling stress and the recovery after chilling stress), and the survival rate and the freezing resistance of cuttings during cold acclimation at -3℃ were investigated. Results showed that cold acclimation not only increased the contents of total soluble protein and RNA, the survival rates and the freezing resistance of cuttings, decreased the activity of RNase, but also reduced the declining degree of total soluble protein and RNA contents, and the increasing level of RNase caused by chilling stress as compared with the controls. In addition, cold acclimation augmented the increase in the level of total soluble protein and RNA, and facilitated the decrease of RNase during the recovery periods. Further analysis found that the DNA content of all treatments kept relative stability at various periods. The changes in total soluble protein, RNA and RNase were closely related to the freezing resistance of cuttings. It appears that the increase of RNA content caused by cold acclimation induced decrease of RNase activity may be involved in the accumulation of total soluble protein and the induction of freezing resistance of cuttings. 展开更多

Fur ther analysis found that the concentrations of total soluble sugar in branches i ncreased greater than that in leaves during both cold acclimation with or withou t the pretreatment of saccharose. Moreover, an increase of the concentrations of total soluble sugar in branches and leaves was closely related to the freezing tolerance of seedlings. Potato(Solanum tuberosum)is a globally important staple crop.However,cultivated potato varieties are highly sensitive to low temperatures.The molecular mechanisms underlying freezing resistance in potatoes remain poorly understood.Through comparative metabolome and transcriptome analyses of freezing-tolerant(CM,Solanum commersonii)and freezing-sensitive(DM,DM1-3516R44)varieties,we identified a coldinduced UDP-glycosyltransferase gene,ScUGT73B4,which is associated with the accumulation of glycosylated flavonoids in wild CM varieties.Overexpression of ScUGT73B4 led to increased accumulation of glycosylated flavonoids and enhanced antioxidant capacity,resulting in improved freezing tolerance in potato plantlets.These findings reveal a UDP-glycosyltransferase in the flavonoid pathway and offer a potential valuable genetic resource for breeding potatoes with improved freezing tolerance. 展开更多

Cold acclimation is associated with many metabolic changes that lead to an increase of freezing tolerance. In order to investigate the biochemical process of cold acclimation in Ammopiptanthus mongolicus, seedlings were acclimated at 2℃ under 16-h photoperiod (150 μmol·m^-2·s^-1 photosynthetically active radiation) for 14 d. Freezing tolerance in seedlings increased after 14 d of cold-hardening. Contents of protein, proline and solute carbohydrate in cotyledon increased after cold acclimation. Patterns of isozymes of superoxide dismutase (SOD), peroxidase, catalase and polyphenol oxidase (PPO) were investigated. The activities of SOD, peroxidase and PPO in cold acclimated plants were increased during cold-hardening. We deduced that compatible solutes and antioxidant enzymes play important roles in development of freezing tolerance during cold acclimation in this evergreen woody plant. 展开更多

Soil aggregate stability,as an important indicator of soil functions,may be affected by seasonal freezing and thawing(SFT)and land use in high cold and wet regions.Therefore,comprehensive understanding the effects of SFT on aggregate stability in orchards during winter and spring is crucial to develop appropriate management strategies that can effectively alleviate the degradation of soil quality to ensure sustainable development of orchard ecosystems.To determine the mechanism of degradation in orchard soil quality,the effects of SFT on the stability of water-stable aggregates were examined in apple-pear orchards(Pyrus ussuriensis var.ovoidea)of four different ages(11,25,40,and 63 yr)on 0 to 5%slopes before freezing and after thawing from October 2015 to June 2016 in Longjing City,Yanbian Prefecture,Northeast China,involving a comparison of planted versus adjacent uncultivated lands(control).Soil samples were collected to investigate water-stable aggregate stability in three incremental soil layers(0–20,20–40 and 40–60 cm).In the same samples,iron oxide,organic matter,and clay contents of the soil were also determined.Results showed that the destructive influences of SFT on water-stable aggregates were more pronounced with the increased orchards ages,and SFT exerted severe effects on water-stable aggregates of older orchards(40 and 63 yr)than juvenile orchards.Undergoing SFT,the soil instability index and the percentage of aggregate destruction increased by mean 0.15 mm and 1.86%,the degree of aggregation decreased by mean 1.32%,and the erosion resistance weakened,which consequently led to aggregate stability decreased.In addition,soil free,amorphous,and crystalline iron oxide as well as soil organic matter and clay contents are all important factors affecting the stability of water-stable aggregates,and their changes in their contents were consistent with those in the stability of water-stable aggregates.The results of this study suggest that long-term planting fruit trees can exacerbate the damaging effects of SFT on aggregate stability and further soil erosion increases and nutrient losses in an orchard,which hider sustainable use of soil and the productivity orchards. 展开更多

Objectives:Cold-acclimated organisms accumulate low molecular weight organic solutes such as sugar alcohols and soluble sugars.This study aimed to compare the efficacy of five sugar alcohols and 14 soluble sugars in stabilizing proteins under freezing,freeze-drying,and air-drying stresses.Materials and methods:Glucose-6-Phosphate Dehydrogenase(G6PD)was used as the model protein.G6PD solutions with or without sugar alcohols and or sugars were subjected to freezing,freeze-drying,and air-drying stresses.The recovery of G6PD activity was measured to evaluate the protective efficacy of these compounds.Results:Without stabilizers,freezing G6PD at-20℃ or-80℃ reduced enzyme activity by around 24%,while freeze-drying or air-drying reduced activity by 90%-95%.Among the five sugar alcohols tested,pinitol,quebrachitol and sorbitol stabilized G6PD,whereas mannitol and myo-inositol destabilized it.Among 14 soluble sugars,trehalose and raffinose showed slightly lower enzyme recovery after repeated freeze-thaw cycles at-20℃.Most soluble sugars(except arabinose and xylose)protected G6PD during freeze-drying,with di-,tri-,and oligosaccharides generally outperforming monosaccharides.During air-drying,lactose was ineffective,while arabinose,galactose,and xylose were detrimental.Conclusion:The study highlights the diverse mechanisms of sugar alcohols and sugars in protein stabilization under stress,offering insights for formulating stable protein-and cell-based drugs. 展开更多

Based on comprehensive observations of 20 wire icing events during winter from 2019 to 2021,we investigated the characteristics of the icing properties,the atmospheric boundary layer structure,the raindrop size distribution,and their associated effects on the ice accretion mechanism in the mountainous region of Southwest China.The maximum ice weight was positively correlated with the duration of ice accretion in the mountainous area.The duration of precipitation accounted for less than 20%of the icing period in the mountainous area,with solid-phase hydrometeors being predominant.Icing events,dominated by freezing rain(FR)and mixed rain–graupel(more than 70%),were characterized by glaze or highdensity mixed icing.The relationship between the melting energy and refreezing energy reflected the distribution characteristics of the proportion of FR under mixed-phase precipitation.The intensity of the warm layer and the dominant precipitation phase significantly affected the variations in the microphysical properties of FR.The melting of large dry snowflakes significantly contributed to FR in the mountainous areas,resulting in smaller generalized intercepts and larger mass-weighted mean diameters in the presence of a stronger warm layer.Under a weaker warm layer,the value of the massweighted mean diameter was significantly smaller because of the inability of large solid particles to melt.Finally,FR in the mountainous area dominated the ice weight during the rapid ice accumulation period.A numerical simulation of FR icing on wires effectively revealed the evolution of disaster-causing icing in mountainous areas. 展开更多

Subtropical evergreen broad-leaved trees are usually vulnerable to freezing stress,while hexaploid wild Camellia oleifera shows strong freezing tolerance.As a valuable genetic resource of woody oil crop C.oleifera,wild C.oleifera can serve as a case for studying the molecular bases of adaptive evolution to freezing stress.Here,47 wild C.oleifera from 11 natural distribution sites in China and 4 relative species of C.oleifera were selected for genome sequencing.“Min Temperature of Coldest Month”(BIO6)had the highest comprehensive contribution to wild C.oleifera distribution.The population genetic structure of wild C.oleifera could be divided into two groups:in cold winter(BIO6≤0℃)and warm winter(BIO6>0℃)areas.Wild C.oleifera in cold winter areas might have experienced stronger selection pressures and population bottlenecks with lower N_(e) than those in warm winter areas.155 singlenucleotide polymorphisms(SNPs)were significantly correlated with the key bioclimatic variables(106 SNPs significantly correlated with BIO6).Twenty key SNPs and 15 key copy number variation regions(CNVRs)were found with genotype differentiation>50%between the two groups of wild C.oleifera.Key SNPs in cis-regulatory elements might affect the expression of key genes associated with freezing tolerance,and they were also found within a CNVR suggesting interactions between them.Some key CNVRs in the exon regions were closely related to the differentially expressed genes under freezing stress.The findings suggest that rich SNPs and CNVRs in polyploid trees may contribute to the adaptive evolution to freezing stress. 展开更多

Non-freezing cold injury is a prevalent cause of peripheral nerve damage, but its pathogenic mechanism is poorly understood, and treatment remains inadequate. Glucocorticoids have anti-inflammatory and lipid peroxidation-inhibiting properties. We therefore examined whether dexamethasone, a synthetic glucocorticoid compound, would alleviate early-stage non-freezing cold injury of the sciatic nerve. We established Wistar rat models of non-freezing cold injury by exposing the left sciatic nerve to cold(3–5°C) for 2 hours, then administered dexamethasone(3 mg/kg intraperitoneally) to half of the models. One day after injury, the concentration of Evans blue tracer in the injured sciatic nerve of rats that received dexamethasone was notably lower than that in the injured sciatic nerve of rats that did not receive dexamethasone; neither Evans blue dye nor capillary stenosis was observed in the endoneurium, but myelinated nerve fibers were markedly degenerated in the injured sciatic nerve of animals that received dexamethasone. After dexamethasone administration, however, endoneurial vasculopathy was markedly improved, although damage to the myelinated nerve fiber was not alleviated. These findings suggest that dexamethasone protects the blood-nerve barrier, but its benefit in non-freezing cold injury is limited to the vascular system. 展开更多

Ammopiptanthus mongolicus, the only freezing tolerant evergreen broad-leaved shrub, local species of the Alashan desert, northwest sand area of China, can survive -30℃ or even lower temperature in winter. In the present study, the secondary products phenolics in A. mongolicus cotyledons were determined to study the effects of phenolics on cold tolerance. Cytochemical localization of phenolics in cotyledon cells was observed by electron microscopy and the content of phenolic compounds was assayed by spectrophotometric measurement. The results showed that the freezing tolerance ofA. mongolicus seedlings increased after acclimation at 2℃ for 14 days, which accompanied the increase of the content of phenolic compounds in cotyledons. Cytochemical observation showed that phenolic deposits were mainly localized in vacuoles and in close proximity to tonoplast, and also in the cytoplasm. The amount and the size of phenolics droplets increased obviously in cytoplasm and vacuoles after cold acclimation, predominantly aggregated along membranes of vacuoles and tonoplast. No phenolic deposits were found in cell walls. As hydrogen- or elec- tron-donating agents, phenolics may protect plant cells against reactive oxygen species formed during chilling or freezing stress and improve the freezing tolerance of cold-acclimated A. mongolicus seedlings. 展开更多

Current practice of underground artificial ground freezing(AGF)typically involves huge refrigeration systems of large economic and environmental costs.In this study,a novel AGF technique is proposed deploying available cold wind in cold regions.This is achieved by a static heat transfer device called thermosyphon equipped with an air insulation layer.A refrigeration unit can be optionally integrated to meet additional cooling requirements.The introduction of air insulation isolates the thermosyphon from ground zones where freezing is not needed,resulting in:(1)steering the cooling resources(cold wind or refrigeration)towards zones of interest;and(2)minimizing refrigeration load.This design is demonstrated using well-validated mathematical models from our previous work based on two-phase enthalpy method of the ground coupled with a thermal resistance network for the thermosyphon.Two Canadian mines are considered:the Cigar Lake Mine and the Giant Mine.The results show that our proposed design can speed the freezing time by 30%at the Giant Mine and by two months at the Cigar Lake Mine.Further,a cooling load of 2.4 GWh can be saved at the Cigar Lake Mine.Overall,this study provides mining practitioners with sustainable solutions of underground AGF. 展开更多

5-Aminolevulinic acid(ALA)is a natural plant growth regulator that promotes plant freezing tolerance.The WRKY family consists of plantspecific transcription factors(TFs)associated with abiotic stress responses.Up to n... 5-Aminolevulinic acid(ALA)is a natural plant growth regulator that promotes plant freezing tolerance.The WRKY family consists of plantspecific transcription factors(TFs)associated with abiotic stress responses.Up to now,whether WRKYs are involved in ALA-induced plant freezing tolerance and the underlying mechanism is not clear.In this study,we found that pretreatment with 50 mg·L^(-1) ALA one week earlier significantly increased the freezing tolerance of nectarine(Prunus persica var.nectarina)pistils with higher antioxidant enzyme activity and osmotic solutes when the floral twigs were stressed by-3℃ for 6 h.ALA also enhanced the expression of PpWRKY18,PpCBF1,PpCOR1,and several genes encoding antioxidant enzymes(such as superoxide dismutase,peroxidase,and catalase)and pyrroline-5-carboxylate synthase(P5CS).When PpWRKY18 was overexpressed in tobacco,the transgenic plants exhibited greater freezing tolerance,which was further promoted by exogenous ALA.Y2H,Pull-down,BiFC,and LCI analyses revealed that PpWRKY18 interacts with PpCBF1,promoting the latter transcriptional activity.Additionally,Y1H experiments showed that PpWRKY18 directly binds to the promoter of PpPOD41 while PpCBF1 binds to the promoters of PpP5CS1 and PpCOR1,activating the target gene expressions.Furthermore,we established a yeast library using the promoter of PpWRKY18 as the bait to screen the upstream regulatory factors.By library screening,Y1H,DLR,and EMSA,we found that PpC3H37,a zinc finger protein,was responsive to chilling and ALA treatment,and as a transcription factor,it activated PpWRKY18 expression by directly binding to the promoter.Taken together,we reveal a regulatory network where ALA induces upregulation of PpC3H37 expression,which positively regulates PpWRKY18 expression.Subsequently,the regulatory pathway diverges into two branches.The first is CBF-dependent,where PpCBF1 interacts with PpWRKY18,binding the promoters of PpP5CS1 and PpCOR1.The second is CBF-independent,where PpWRKY18 directly binds the promoter of PpPOD41 to upregulate the gene expression and increase the antioxidant enzyme activity and freezing tolerance.These findings provide a novel insight of the mechanism of ALA in regulating the cold hardiness of nectarine pistil. 展开更多

The ability of plants to tolerate cold is a complex process.When temperatures drop or freeze,plant tissues can develop ice,which dehydrates the cells.However,plants can protect themselves by preventing ice formation.This intricate response to cold stress is regulated by hormones,photoperiod,light,and various factors,in addition to genetic influences.In autumn,plants undergo morphological,physiological,biochemical,and molecular changes to prepare for the low temperatures of winter.Understanding cellular stress responses is crucial for genetic manipulation aimed at enhancing cold resistance.Early autumn frosts or late spring chills can cause significant damage to plants,making it essential to adapt in autumn to survive winter conditions.While the general process of acclimatization is similar across many plant species,variations exist depending on the specific type of plant and regional conditions.Different plant organs exhibit varying degrees of damage from cold stress,and by applying agricultural principles,potential damage can be largely controlled.Timely reinforcement and stress prevention can minimize cold-related damage.Research has shown that in temperate climates,low temperatures restrict plant growth and yield.However,the intricate structural systems involved remain poorly understood.Over the past decade,studies have focused on the molecular mechanisms that enable plants to adapt to and resist cold stress.The gene signaling system is believed to play a crucial role in cold adaptation,and researchers have prioritized this area in their investigations.This study critically examines plant responses to cold stress through physiological adaptations,including calcium signaling dynamics,membrane lipid modifications,and adjustments in antioxidant systems.These mechanisms activate downstream gene expression and molecular functions,leading to key resistance strategies.Additionally,we explore the regulatory roles of endogenous phytohormones and secondary metabolites in cold stress responses.This review aims to enhance our foundational understanding of the mechanisms behind plant cold adaptation. 展开更多
