AIM To study the agglutination pattern ofHelicobacter pylori coccoid and spiral forms.METHODS Assays of agglutination andagglutination inhibition were applied usingfifteen commercial lectins.RESULTS Strong agglutination was observedwith mannose-specific Concanavalin A(Con A),fucose-specific Tetragonolobus purpureas(Lotus A)and N-acetyl glucosamine-specificTriticum vulgaris(WGA)lectins.Mannose andfucose specific lectins were reactive with allstrains of H.pylori coccoids as compared to thespirals.Specific carbohydrates,glycoproteinsand mucin were shown to inhibit H.pylorilectin-agglutination reactions.Pre-treatment ofthe bacterial cells with formalin and sulphuricacid did not alter the agglutination patterns withlectins.However,sodium periodate treatment ofbacterial cells were shown to inhibitagglutination reaction with Con A,Lotus A andWGA lectins.On the contrary,enzymatictreatment of coccoids and spirals did not showmarked inhibition of H.pylori-lectinagglutination.Interestingly,heating of H.pylori cells at 60℃ for 1 hour was shown toaugment the agglutination with all of the lectinstested.CONCLUSION The considerable differences inlectin agglutination patterns seen among the twodifferentiated forms of H.pylori might beattributable to the structural changes during theevents of morphological transformation,resulting in exposing or masking some of the sugar residues on the cell surface.Possibility ofvarious sugar residues on the cell wall of thecoccoids may allow them to bind to differentcarbohydrate receptors on gastric mucus andepithelial cells.The coccoids with adherencecharacteristics like the spirals could aid in thepathogenic process of Helicobacter infection.This may probably lead to different clinicaloutcome of H.pylori associated gastroduodenaldisease. 展开更多

The presence of viable Helicobacter pylori(H. pylori) in the environment is considered to contribute to the levels of H. pylori found in the human population, which also aids to increase its genetic variability and its environment adaptability and persistence. H. pylori form biofilms both within the in vitro and in vivo envi-ronment. This represents an important attribute that assists the survival of this bacterium within environ-ments that are both hostile and adverse to prolifera-tion. It is the aim of this paper to review the ability of H. pylori to form biofilms in vivo and in vitro and to address the inherent mechanisms considered to sig-nificantly enhance its persistence within the host and in external environments. Furthermore, the dissemi-nation of H. pylori in the external environment and within in the human body and its impact upon infec-tion control shall be discussed. 展开更多

BACKGROUND Helicobacter pylori(H.pylori)is a spiral-shaped bacterium responsible for the development of chronic gastritis,gastric ulcer,gastric cancer(GC),and MALTlymphoma of the stomach.H.pylori can be present in the gastric mucosa(GM)in both spiral and coccoid forms.However,it is not known whether the severity of GM contamination by various vegetative forms of H.pylori is associated with clinical and morphological characteristics and long-term results of GC treatment.AIM To establish the features of H.pylori infection in patients with GC and their correlations with clinical and morphological characteristics of diseases and long-term results of treatment.METHODS Of 109 patients with GC were included in a prospective cohort study.H.pylori in the GM and tumor was determined by rapid urease test and by immunohistochemically using the antibody to H.pylori.The results obtained were compared with the clinical and morphological characteristics and prognosis of GC.Statistical analysis was performed using the Statistica 10.0 software.RESULTS H.pylori was detected in the adjacent to the tumor GM in 84.5%of cases,of which a high degree of contamination was noted in 50.4%of the samples.Coccoid forms of H.pylori were detected in 93.4%of infected patients,and only coccoid-in 68.9%.It was found that a high degree of GM contamination by the coccoid forms of H.pylori was observed significantly more often in diffuse type of GC(P=0.024),in poorly differentiated GC(P=0.011),in stage T3-4(P=0.04)and in N1(P=0.011).In cases of moderate and marked concentrations of H.pylori in GM,a decrease in 10-year relapse free and overall survival from 55.6%to 26.3%was observed(P=0.02 and P=0.07,respectively).The relationship between the severity of the GM contamination by the spiral-shaped forms of H.pylori and the clinical and morphological characteristics and prognosis of GC was not revealed.CONCLUSION The data obtained indicates that H.pylori may be associated not only with induction but also with the progression of GC. 展开更多

The infectivity o... Helicobacter pylori (H. pylori) can infect into the epithelial cell to cause benign or malignant disorders. Under stressful environment, a spiral form of H. pylori is transformed into a coccoid form. The infectivity of the coccoid form is still controversial. Since spiral forms are transformed into two types of coccoid forms via different mechanisms, the infectivity of the two types of coccoid forms into human gastric epithelial cell was examined. A laboratory and a clinical strain of H. pyloriv were cultured in liquid medium under different conditions to produce the two types of coccoid forms. These coccoid H. pylorisv were then co-cultured with human derived gastric epithelial cell, MKN-28. Adhesion and penetration of bacteria into MKN-28 cells were monitored by scanning-, standard transmission- and immunotransmission-electron microscopy (SEM, TEM and ITEM). We observed that both coccoid forms were able to adhere onto the surface of MKN-28 cells in agminated formation and also penetrated into the gastric epithelial cells besides the spiral form of H. pyloriv. Coccoid H. pylori is not a passive entity but can actively infect the human gastric epithelial cell. 展开更多

A 3 day old culture of H. pylori... AIM To detect antibodies against Helicobacter pylori spiral and coccoid antigens in human sera. METHODS Blood samples were collected from 278 patients with gastric diseases. A 3 day old culture of H. pylori on chocolate blood agar was used to provide spiral form. ‘Synchronous’ coccoids were cultured in (BHY) (brain heart infusion supplemented with 10% horse serum and 0 4% yeast extract) medium in a chemostat. Antigens from spiral and coccoid form were prepared using acid glycine extraction. Enzyme linked immunosorbent assay (ELISA) was performed to detect serum IgG antibodies against spiral and coccoid forms of H. pylori . RESULTS Seroprevalence of H. pylori infection was higher in patients with gastric ulcer (79%) and gastric cancer (83%) than those with non ulcer dyspepsia (NUD) (44%) and other diseases (45%) ( P <0 05). IgG antibodies against spiral and coccoid antigens were detected in 50 7% (141/278) and 49 6% (138/278) , respectively. CONCLUSION The spiral and coccoid forms of H. pylori coexist in patients infected with the bacterium. 展开更多

Makinoella Okada is a genus of coccoid green algae belonging to the family Oocystaceae with typical morphological characteristics.Reports to date included only one type species.Since the genus is rarely reported,there is a lack of taxonomic research.In this study,17 strains of this genus were collected from several different places in China,and their taxonomic studies were conducted based on morphological and molecular phylogenetic analysis.The phylogenetic results based on the analysis of ITS,chloroplast genes rbc L and tuf A showed that the genus was divided into two branches.One of the branches comprised newly collected algae.Compensatory base changes(CBCs)and hemi-compensatory base changes(hemi-CBCs)within the secondary structure of the ITS-2 confirmed the branches as two species.Compared with the type species of Makinoella tosaensis,the cell size of this new branch was only about 50%,and included 2-4 colonial cells.Therefore,based on the smaller cell size,simpler colony composition,independent phylogenetic position and CBCs and hemi-CBCs,we suggest that the new clade should be designated as a new taxon of Makinoella,namely Makinoella parva.Among the four molecular markers using in this study,the rbc L and newly introduced ITS are recommended for species separation,which can help further studies to revise the species and generic concepts of the family. 展开更多

Understanding the Competitive and Cooperative Interactions between Probiotics and Autochthonous Intestinal Bacteria

The present study combines the theory and the experimental data to predict the changes on intestinal bacterial populations during ingestion of beneficial probiotic bacteria. Our proposed model is a modified version of the Lotka-Volterra model, which takes the probiotic administration into account. Using the linear stability analysis of the model, the conditions for coexistence of the probiotics with other bacteria are established. Using the model fitted to the data of C. coccoides species and Bifidobacterium species, the effects of oral probiotics on autochthonous bacterial cultures is investigated. The estimated parameter values suggest that C. coccoides and Bifidobacterium facilitate each other during the probiotics administration, whereas they compete in the absence of the probiotics administration. This may suggest the beneficial effect of probiotic administration as it promotes the growth of C. coccoides species. The results also confirm prior studies showing that once probiotic supplementation is discontinued, the probiotic population and the promoting effect within the digestive tract will diminish. 展开更多

The fossils are described and compared with coccoid dinoflagellates. These fossils may be a coccoid life-cycle stage (vegetative cyst) of coccoid dinoflagellates. If this identification is correct, the coccoid dinoflagellates-like fossils could give a reasonable explanation of the dinoflagellate-specific biomarkers from Cambrian, H4 well, Tarim Basin. 展开更多
