Each cDNA fragment of strain Shimen was...According to the previously published CSFV sequences, 18 paris of partially overlapping primers which span the entire genome of CSFV strain Shimen were designed and synthesized. Each cDNA fragment of strain Shimen was amplified by RT-PCR method from the anticoagulant blood of strain Shimen infected pig. The PCR fragments were cloned into pGEM-T vector respectively and sequenced. The results show that we have obtained the nucleotide sequence of strain Shimen. The viral RNA consists of 12 297 nucleotides including noncoding regions of 373 and 227 bases at the 5′ and 3′ end, respectively, and a single large open reading frame spanning 11 697 nucleotides in the middle, which encodes an amino acid sequence of 3 989 residues with a calculated molecular weight of 437.6×103. The precisely sequencing of 5′ and 3′ termini is undertaking.展开更多
Effects of attenuated highly pathogenic pig reproductive and respiratory syndrome(HP-PRRS)TJM-F92 strain vaccine on immune antibody level against classical swine fever(CSF)and foot-and-mouth disease(FMD)were stu...Effects of attenuated highly pathogenic pig reproductive and respiratory syndrome(HP-PRRS)TJM-F92 strain vaccine on immune antibody level against classical swine fever(CSF)and foot-and-mouth disease(FMD)were studied from October 8 to November 12 in 2014,in order to optimize vaccination program of CSF,HP-PRRS and FMD and to provide scientific guidance for animal disease control and prevention work.The results showed that attenuated HP-PRRS(TJMF92 strain)vaccine had no significant effect on immune antibody level of hog cholera lapinized virus(HCLV,ST passage cell vaccine)attenuated vaccine and FMD-O inactivated vaccines(OZK/93 strain),and single or combined use of three vaccines received good immunization effects.展开更多
Media engineering and strain improvement are critical aspects of microbial biotechnology playing a vital role in enhancing microbial productivity,and ensuring cost-effective bioprocessing.In this investigation,we opti...Media engineering and strain improvement are critical aspects of microbial biotechnology playing a vital role in enhancing microbial productivity,and ensuring cost-effective bioprocessing.In this investigation,we optimized the various medium components,nutritional condition,and fermentation parameters for the industrial production of phenoxymethylpenicillin or penicillin V(PenV).We have isolated,characterized Penicillium rubens BIONCL P45 strain which initially produced 100 mg/L of PenV.Further,optimization using production medium 4(PM4)comprising lactose,corn steep solids,sodium sulfate,calcium carbonate,and phenoxy acetic acid lead to a significant increase in production,reaching 430 mg/L.Further improvements through response surface methodology(RSM)predicted a production of 646 mg/L,which was experimentally validated at 685 mg/L.Subsequently,mutagenesis studies using UV(ultraviolet)exposure resulted in the UV-65 mutant,which demonstrated a superior performance,achieving 934 mg/L,surpassing the parental strain.These combined strategies lead to a tenfold increase in PenV titer,highlighting their effectiveness in bioprocess development and industrial-scale antibiotic production.展开更多
