By gas chromatogram, six crude oils fingerprinting distributed in four oilfields and four oil platforms were analyzed and the corre- sponding normal paraffin hydrocarbon ( including pristane and phytane) concentration was obtained by the internal standard methed. The normal paraffin hydrocarbon distribution patterns of six crude oils were built and compared. The cluster analysis on the normal paraffin hydrocarbon concentration was conducted for classification and some ratios of oils were used for oils comparison. The results indicated: there was a clear difference within different crude oils in different oil fields and a small difference between the crude oils in the same oil platform. The normal paraffin hydrocarbon distribution pattern and ratios, as well as the cluster analysis on the nomad paraffin hydrocarbon concentration can have a better differentiation result for the crude oils with small difference than the original gas chromatogram. 展开更多

To evaluate the quality of Polyporus umbellatus,we established a simple,repeatable and reliable method based on high performance liquid chromatography(HPLC)for specific chromatogram and fingerprint analysis,which was applied to analyze samples of medicinal materials and decoction pieces collected from different regions.Finally,ten characteristic peaks were designated in the specific chromatograms and applied to the authenticate identification of P.umbellatus samples.Nine common peaks were designated in the fingerprints,and then the similarities between 32 batches of samples were calculated.Among them,eight compounds were identified by HPLC-APCI-IT-TOF-MS^(n),four of which were identified in specific chromatograms and four in fingerprints.In the present study,we,for the first time,combined HPLC specific chromatograms and fingerprints for the species identification and quality evaluation of P.umbellatus.Collectively,our findings provided a new method for establishing a comprehensive quality standard of P.umbellatus. 展开更多

DNA barcoding and HPLC specific chromatogram were used to identify three kinds of Plumeria flowers respectively. DNAs extracted from the three Plumeria species were amplified by PCR with universal primers, and the psbA-trnH region was selected. All the amplified products were sequenced and the results were analyzed by MEGA 5.0. Chemometric methods including principal components analysis and hierarchical clustering analysis were conducted on the SAS 9.0 software to demonstrate the variability among samples. In conclusion, the psbA-trnH of all samples were successfully amplified from total DNA and sequenced. These three varieties of Plumeria can be differentiated by the psbA-trnH region and clustered into three groups respectively through building neighbor joining tree, which conformed to their germplasm origins. However, it was hard to distinguish them by HPLC specific chromatograms combined with chemometrics analysis. These indicated that DNA barcoding was a promising and reliable tool for the identification of three kinds of Plumeria flowers compared to HPLC specific chromatogram generally used. It could be treated as a powerful complementary method for traditional authentication, especially for those varieties which are difficult to be identified by conventional chromatography. 展开更多

The fingerprint chromatogram of 10 batches of samples of Tetracera asiatica was determined for similarity comparison. [Results] It established HPLC fingerprint chromatogram of Tetracera asiatica produced in Guangxi,determined a total of 13 common fingerprint peaks. [Objectives]This study aimed to establish HPLC chromatograms of decoction pieces,standard decoction and formula granules of Spica Prunellae.[Methods]The chromatographic conditions were as follows:column,SHISEIDO CAPCELL PAK C18 MGII column(4.6 mm×250 mm,5μm);mobile phase,acetonitrile-0.1%phosphoric acid;gradient elution;detection wavelength,280 nm;flow rate,1.0 mL/min;and column temperature,30℃.The correlation between the decoction pieces,standard decoction and formula granules of Spica Prunellae was analyzed by specific chromatograms.[Results]The fingerprint chromatograms of decoction pieces,standard decoction and formula granules of Spica Prunellae showed five common peaks,with good correlation.Among the five common peaks,four of them were tanshinol,protocatechuic acid,caffeic acid and rosmarinic acid.[Conclusions]The main chemical constituents of decoction pieces,standard decoction and formula granules of Spica Prunellae are basically the same.The HPLC specific chromatogram established can be used for the quality control of Spica Prunellae formula granules. 展开更多

Sweet from different habitats, in order to provide experimental basis for the quality control of A. indicum and the spectrum-efficacy relationship of petroleum ether.[Methods] The HPLC chromatogram of petroleum ether was obtained using Agilent SB-C 18 (4.6 mm×250 mm, 5 μm) and acetine-0.1% phosphoric acid solution for gradient elution at temperature of 30 ℃, flow rate of 1.0 mL/min and detection wavelength of 205 nm.[Results] The typical HPLC chromatogram of petroleum ether in A. indicum was preliminarily established, with 12 characteristic peaks. It was verified by A. indicum from different habitats, characterized by reproducibility and stability.[Conclusions] Typical HPLC chromatogram provides a stable new method for the quality control of petroleum ether in A. indicum . 在尊重中药配方颗粒发生发展的客观规律前提下,国家相关部门在总结中药配方颗粒质量标准发展历程中的经验教训基础上,逐步形成中药配方颗粒国家药品标准的研究制定思路,以及中药配方颗粒国家标准的形成机制,对中药质量标准的制定路径具... 在尊重中药配方颗粒发生发展的客观规律前提下,国家相关部门在总结中药配方颗粒质量标准发展历程中的经验教训基础上,逐步形成中药配方颗粒国家药品标准的研究制定思路,以及中药配方颗粒国家标准的形成机制,对中药质量标准的制定路径具有一定的参考意义。结合已发布标准的总体情况和具体品种案例,笔者总结了中药配方颗粒国家标准的研究理念,并重点对已发布国家标准的技术特点进行深入分析,包括标准汤剂的引入、中药整体质量控制、全过程质量控制等方面的创新应用;同时总结了在中药配方颗粒国家标准的研究与制定过程中对药材源头和过程控制等存在的行业共性问题力求一并解决,鼓励新型技术手段在中药配方颗粒领域示范应用。最后结合文献分析,对目前国家标准存在的不足进行探讨,并对中药配方颗粒国家标准进一步完善提出了相关建议。通过整体分析,有助于全面了解中药配方颗粒国家标准的技术特点,为中药质量控制方法的科学探索与实践提供参考。 展开更多

目的:优化檀香油薄层色谱鉴别方法,运用气相色谱法建立檀香油特征图谱。方法:在《中华人民共和国药典》2020年版檀香油项下薄层色谱鉴别基础上,以正己烷-丙酮(16:4)为展开剂,优化薄层色谱条件。采用Agilengt HP-5毛细管色谱柱(30 m×... 目的:优化檀香油薄层色谱鉴别方法,运用气相色谱法建立檀香油特征图谱。方法:在《中华人民共和国药典》2020年版檀香油项下薄层色谱鉴别基础上,以正己烷-丙酮(16:4)为展开剂,优化薄层色谱条件。采用Agilengt HP-5毛细管色谱柱(30 m×0.32 mm,0.25μm);火焰离子化检测器,进样口温度为280℃;检测器温度为250℃;载气为氮气;流速为0.5 mL·min^(-1);程序升温(初始110℃,5℃·min^(-1)升至160℃;1℃·min^(-1)升至166℃,保持5 min;0.5℃·min^(-1)升至167℃,保持5 min;0.5℃·min^(-1)升至168℃,保持2 min;10℃·min^(-1)升至230℃,保持5 min)。进样量为1μL,分流比为20:1,建立檀香油特征图谱。结果:优化后薄层色谱斑点清晰、分离度好、专属性强、耐用性良好、重复性良好。特征图谱选取了檀香油对照品色谱图中分离度、峰形较好的7个峰为特征峰,所建立方法专属性强,稳定性、重复性良好,方法可行。结论:建立了檀香油薄层色谱鉴别和特征图谱方法,可实现对檀香油质量的有效控制,为其质量评价提供参考。 展开更多

建立了气相色谱-质谱-选择离子监测(GC-MS-SIM)法同时测定五味子及其制剂中6种木脂素(五味子醇甲、五味子醇乙、五味子甲素、五味子乙素、五味子丙素和安五脂素)的方法;同时建立了这6种木脂素的特征图谱。供试品用乙醇超声处理;分离采用... 建立了气相色谱-质谱-选择离子监测(GC-MS-SIM)法同时测定五味子及其制剂中6种木脂素(五味子醇甲、五味子醇乙、五味子甲素、五味子乙素、五味子丙素和安五脂素)的方法;同时建立了这6种木脂素的特征图谱。供试品用乙醇超声处理;分离采用TG-5MS色谱柱(30m×0.25mm×0.25μm);质谱分析同时扫描6种木脂素的基峰离子和分子离子,包含质荷比为137.1、328.3、384.2、400.3、416.3以及432.3的6类离子。采用国家药典委员会发布的中药色谱指纹图谱相似度评价系统(2012版)计算特征图谱的相似度,采用SPSS21.0软件进行聚类分析,用外标法测定木脂素的含量。结合相似度、聚类分析和木脂素含量用于五味子及其制剂的质量评价。在这6种木脂素的保留时间内供试品中检出了7种物质,该条件下未知物的质谱图与五味子乙素一致。在一定浓度范围内,6种木脂素的选择离子峰面积与浓度之间具有良好的线性关系,相关系数均大于等于0.9990。6种木脂素的平均加标回收率在93.9%~109.2%,相对标准偏差(n=5)在1.4%~4.8%。该方法用于供试品中6种木脂素含量测定的相对标准偏差(n=4)为1.3%~5.5%。特征图谱的相似度和聚类分析的结果表明,7批制剂中的五味子均为北五味子,药典收录的4批制剂和北五味子中五味子醇甲的含量均高于限量值。方法与高效液相色谱法(HPLC)和液相色谱-串联质谱(LC-MS)相比,具有6种木脂素的灵敏度相近,特征图谱简单、干扰物质少、选择性高等特点。该方法适用于五味子及其制剂中五味子种类的判断和6种木脂素的定量分析。 展开更多

特征图谱研究现状及脉络的文献计量学分析

目的总结特征图谱的研究现状及热点,为后续开展相关研究提供参考。方法采用计算机检索中国知网、万方、维普、Web of Science等数据库中有关特征图谱的研究文献,检索时限为自建库起至2024年7月25日。采用CiteSpace 6.3.R2软件对纳入文... 目的总结特征图谱的研究现状及热点,为后续开展相关研究提供参考。方法采用计算机检索中国知网、万方、维普、Web of Science等数据库中有关特征图谱的研究文献,检索时限为自建库起至2024年7月25日。采用CiteSpace 6.3.R2软件对纳入文献的年发文量、研究类型、关键词进行可视化分析。结果共纳入1006篇中文文献,年发文量整体呈上升趋势;英文文献63篇,由于数量较少,故未做分析。特征图谱相关中文文献刊发期刊153种,99.01%为研究型论文(996篇),主要涉及中药与方剂、化学、中医与中西医结合等。刊文量排名前3的期刊分别为《药物分析杂志》(112篇)、《中国医院药学杂志》(49篇)、《中药材》(41篇);引用频次排名前3的文献分别为《GC-MS法建立广藿香挥发油指纹特征图谱研究》(131次)、《指纹图谱、特征图谱技术在中药材和中成药中的应用》(105次)、《从HPLC特征图谱分析姜在炮制过程中的化学成分变化》(57次)。关键词出现频次排名前3的分别为特征图谱(681次)、含量测定(127次)、指纹图谱(101次),关键词聚类涉及11个标签,且突现强度较高的关键词主要有标准汤剂(8.33)、含量测定(7.29)、质量评价(5.36)等。中药质量控制及成分含量测定为特征图谱研究的重点内容,研究热点为基于特征图谱的方剂质量控制及评价。结论建议未来将特征图谱广泛用于经典名方、标准汤剂等方剂的质量评价,以保障中药的质量。 展开更多

目的:建立芪葵方基准样品的高效液相色谱(HPLC)指纹图谱及多指标成分含量测定方法。方法:根据中药饮片临床煎煮规范,制备15批芪葵方基准样品,并建立基准样品的HPLC指纹图谱,明确指纹图谱中峰归属、相似度范围以及干膏率和指标性成分含... 目的:建立芪葵方基准样品的高效液相色谱(HPLC)指纹图谱及多指标成分含量测定方法。方法:根据中药饮片临床煎煮规范,制备15批芪葵方基准样品,并建立基准样品的HPLC指纹图谱,明确指纹图谱中峰归属、相似度范围以及干膏率和指标性成分含量范围,建立芪葵方基准样品的质量评价体系。结果:15批基准样品共标定24个共有峰,且相似度大于0.96。基准样品中8种指标性成分含量分别为毛蕊异黄酮葡萄糖苷0.0327~0.0603 mg/mL、芦丁0.0418~0.0644 mg/mL、金丝桃苷0.4976~0.7845 mg/mL、2,3,5,4’-四羟基二苯乙烯-2-O-β-D-葡萄糖苷0.1124~0.2097 mg/mL、异槲皮苷0.3387~0.5420 mg/mL、棉花皮素-8-O-葡萄糖醛酸1.0336~1.5524 mg/mL、毛蕊异黄酮0.0094~0.0163 mg/mL、芒柄花黄素0.0025~0.0046 mg/mL。15批基准样品的干膏率范围为27.78%~33.86%,平均干膏率为30.81%。结论:芪葵方基准样品质量评价方法科学可行且质量稳定,可作为芪葵方制剂开发工艺研究以及生产过程质量控制的依据。 展开更多

目的建立冰球子药材的质量标准。方法通过性状鉴定、显微鉴定筛选有鉴别意义的特征;检查18批药材样品的水分、总灰分及醇溶性浸出物;采用中药指纹图谱相似度评价系统生成高效液相色谱(HPLC)指纹图谱,确定共有峰并计算相似度;结合层次聚... 目的建立冰球子药材的质量标准。方法通过性状鉴定、显微鉴定筛选有鉴别意义的特征;检查18批药材样品的水分、总灰分及醇溶性浸出物;采用中药指纹图谱相似度评价系统生成高效液相色谱(HPLC)指纹图谱,确定共有峰并计算相似度;结合层次聚类分析(HCA)和正交偏最小二乘法判别分析(OPLS-DA)对药材样品进行化学模式识别。采用HPLC法测定不同基原药材样品中特征性成分的含量,并比较。结果筛选出药材样品性状鉴别和显微鉴别的专属鉴别特征;拟订药材水分不高于15.0%,总灰分限度不得过5.0%,醇溶性浸出物不得少于9.0%。18批药材样品的HPLC指纹图谱中确定了6个共有峰,峰1为2-O-葡萄糖基白及苷,峰3为1,4-二[4-(葡萄糖氧)苄基]-2-异丁基苹果酸酯,峰6为3,3′-二羟基-5-甲氧基联苄;相似度均大于0.90;HCA和OPLS-DA筛选出2个特征性成分。2-O-葡萄糖基白及苷和1,4-二[4-(葡萄糖氧)苄基]-2-异丁基苹果酸酯质量浓度分别在24.44~610.88μg/mL及23.76~593.96μg/mL范围内与峰面积线性关系良好(r=0.9999,n=6);精密度、稳定性、重复性试验结果的RSD均小于2.0%;平均加样回收率分别为94.80%和99.80%,RSD均小于2.0%(n=6)。基原为独蒜兰的药材样品中2-O-葡萄糖基白及苷和1,4-二[4-(葡萄糖氧)苄基]-2-异丁基苹果酸酯的含量均高于基原为云南独蒜兰的冰球子药材样品,两种药材中前成分含量无显著差异(P>0.05),而后成分含量存在显著差异(P<0.01)。结论该研究中所建冰球子质量标准结果准确可靠,能用于较全面地评价冰球子的质量,为其质量控制提供参考。 展开更多

石决明(皱纹盘鲍)药材、饮片及标准汤剂的物相变化及量质传递分析 被引量:3

目的:通过分析石决明(皱纹盘鲍)药材-饮片-标准汤剂的物相及组成变化,探索其量质传递过程,为有效控制其质量提供参考。方法:收集20批不同产地的石决明(皱纹盘鲍)药材,制备煅制饮片和标准汤剂,分别测定三者CaCO_(3)含量并计算出膏率和转... 目的:通过分析石决明(皱纹盘鲍)药材-饮片-标准汤剂的物相及组成变化,探索其量质传递过程,为有效控制其质量提供参考。方法:收集20批不同产地的石决明(皱纹盘鲍)药材,制备煅制饮片和标准汤剂,分别测定三者CaCO_(3)含量并计算出膏率和转移率;采用X射线荧光光谱法(XRF)研究三者的元素组成及其相对含量的变化;采用X射线衍射法(XRD)研究三者的物相组成变化并建立各自的XRD特征图谱;采用傅里叶变换红外光谱法(FTIR)研究三者的化学组成及含量变化并建立各自的FTIR特征图谱,同时结合层次聚类分析(HCA)、主成分分析(PCA)及正交偏最小二乘法-判别分析(OPLS-DA)找寻三者的共同及差异性特征,探寻标准汤剂制备过程中的量质传递关系。结果:20批石决明药材、饮片、标准汤剂CaCO_(3)质量分数分别为93.87%~98.95%、96.02%~99.97%、38.29%~51.96%,标准汤剂出膏率为1.71%~2.37%,饮片-标准汤剂的CaCO_(3)转移率为0.68%~1.27%;XRF测定结果表明,石决明药材、饮片所含元素种类及其相对含量具有较高相似度,饮片、标准汤剂所含元素种类虽无明显差异,但相对含量差异明显,主要体现在Ca元素相对含量的减少和Na元素相对含量的增加;XRD测定结果表明,石决明药材主要含文石(aragonite:PDF#99-0013)和方解石(calcite:PDF#99-0022)2种晶型的CaCO_(3);而饮片仅含方解石(calcite:PDF#99-0022)一种晶型的CaCO_(3);标准汤剂主要含方解石晶型的CaCO_(3)和钠长石晶型(natrite:PDF#37-0451)的Na_(2)CO_(3);FTIR测定结果表明,石决明药材中存在O-H、C-H、C=O、HCO_(3)-及CO_(3)^(2-)基团的内部振动,饮片及标准汤剂中中主要存在O-H、HCO_(3)-及CO_(3)^(2-)基团的振动。结论:石决明(皱纹盘鲍)药材-饮片的变化主要是煅制后CaCO_(3)含量的增加、CaCO_(3)文石晶型向方解石晶型的转变及有机成分的消失,饮片-标准汤剂的变化主要是CaCO_(3)含量的减少及Na_(2)CO_(3)相对含量的增加。该研究建立的方法可用于贝壳类药材-饮片-标准汤剂的质量控制,为贝壳类中药配方颗粒质量控制研究提供新思路。 展开更多
