Objective This study aimed to investigate the effects of treatment with palosuran,a urotensin receptor blocker,on molecular changes in the corpora cavernosa(CC)in diabetic rats.Methods Streptozotocin-induced diabetic ...Objective This study aimed to investigate the effects of treatment with palosuran,a urotensin receptor blocker,on molecular changes in the corpora cavernosa(CC)in diabetic rats.Methods Streptozotocin-induced diabetic rats were treated with palosuran 300 mg/kg per day for 6 weeks.Contraction of CC induced by potassium chloride,phenylephrine,and NG-nitro-L-arginine methyl ester and relaxation of CC induced by electrical field stimulation(EFS)and sodium nitroprusside(SNP)(endothelium-dependent and endothelium-independent stimuli,respectively),and Y-27632(Rho-kinase inhibitor)were examined in organ baths.Direct contraction or relaxation induced by palosuran and urotensin-II(U-II)were also evaluated.The expression levels of nitric oxide synthetases(NOSs),RhoA,oxidative stress regulators,and U-II were analyzed by Western blotting or immunohistochemistry.Results Induction of diabetes in rats resulted in the decreased relaxant response to SNP,decreased pD2 value of SNP,attenuated relaxant response to Y-27632 as well as the decreased RhoA expression in CC.Palosuran treatment of diabetic rats reversed all of these parameters;however,it further impaired the already weakened relaxation of diabetic CC in response to EFS.Although induction of diabetes did not change U-II expression in CC significantly,palosuran treatment reduced U-II expression in diabetic CC.The expression level of nNOS was lowered in diabetic CC;however,palosuran treatment did not change the decreased the neuronal NOS expression.In vitro exposure of diabetic CC strips to palosuran produced a direct relaxant response.Conclusion Palosuran treatment did not affect the expression of NOSs or reduce nitrergic conduction induced by EFS stimulation in diabetic CC.However,while directly triggering a relaxant response,it did not induce a prominent contraction either by decreasing U-II expression,or increasing the sensitivity of CC to nitric oxide which suggested that palosuran has the potential to support erectile function.Further and comprehensive studies are required to clarify this issue.展开更多

We excised 2 mm...The objective is to develop an easier technique for regenerating corpora cavernosa tissue through transplantation of human bone marrow-derived CD133+ cells into a rat corpora cavernosa defect model. We excised 2 mm× 2 mm squares of the right corpora cavernosa of twenty-three 8-week-old male nude rats. AIginate gel sponge sheets supplemented with 1 × 10^4 CD133+ cells were then placed over the excised area of nine rats. Functional and histological evaluations were carried out 8 weeks later. The mean intracavernous pressure/mean arterial pressure ratio for the nine rats (0.34258 ± 0.0831) was significantly higher than that for eight rats with only the excision (0.0580±0.0831, P = 0.0238) and similar to that for five rats for which the penis was exposed, and there was no excision (0.37228±0.1051, P = 0.8266). Immunohistochemical analysis revealed that the nine fully treated rats had venous sinus-like structures and quantitative reverse transcription polymerase chain reaction analysis of extracts from their alginate gel sponge sheets revealed that the amounts of mRNA encoding the nerve growth factor (NGF), and vascular endothelial growth factor (VEGF) were significantly higher than those for rats treated with alginate gel sheets without cell supplementation (NGF: P= 0.0309; VEGF: P〈 0.0001). These findings show that transplantation of CD133+ cells accelerates functional and histological recovery in the corpora cavernosa defect model.展开更多

Objective:To find a potent antibiotic producer from the sponge-associated bacteria as well as to profile the important substances.Methods:Sponge collection,bacteria isolation,extraction and characterization of potent ...Objective:To find a potent antibiotic producer from the sponge-associated bacteria as well as to profile the important substances.Methods:Sponge collection,bacteria isolation,extraction and characterization of potent active compounds were carried out for this study.Results:Approximately 59 single strains of bacteria were isolated from this sponge.Totally 40 strains showed activity against Escerichia coli,Staphylococcus aureus and Vibrio eltor.The chemical separation of the potent strain Bacterium sp.Lb.10%.2.1.1.b,using n-phase column chromatography revealed 7 active fractions(7,8,9,10,11,14 and 15).The gas chromatography-mass spectrometer analysis of Fraction 7 indicated some phenolic compounds including 4-nonylphenol,methyl 3-(3,5-di-tert-butyl-4-hydroxyphenyl)propionate,acetosyringone,2,4-bis(1-phenylethyl)phenol,1,2-benzenedicarboxylic acid,mono(2-ethylhexyl)ester,tri(2-ethylhexyl)trimellitate and oleamide.Conclusions:Indeed,this is a preliminary information in profiling chemical substances,produced by Bacterium sp.Lb.10%.2.1.1.b.Further purification and structural chemical determination were needed to find a comprehensive result.展开更多

Surgical exploration and repair of the a...Introduction: Fracture of the penis is a rare uro-andrological emergency defined as the rupture of the tunica albuginea of the erect penis most often during sexual intercourse. Surgical exploration and repair of the albuginea breach as soon as possible are the standard treatment. However, in our African context, early diagnosis and management are negatively influenced by the delay in consultation due to modesty. Our objective was to describe the clinical and therapeutic aspects of the fracture of the penis seen late in the African context and to compare it with the literature review. Presentation of the case: Our aim was to report our experience in the management of late-onset fracture of the penis, after more than 48 hours of evolution, by reporting the observation of a 54-year-old patient, who had a false coitus, treated in the urology department of the Nianankoro Fomba Hospital. The physical examination revealed a deformation of the penis following the formation of a large hematoma with a characteristic eggplant appearance. Therapeutically, a suture of the albuginea was performed with simple follow-up. Conclusion: This case study shows a delay in the management of this emergency due to the reluctance of the patient to be consulted as soon as possible for reasons of modesty. A delayed emergency surgical management was the only therapeutic alternative and the coronal incision with degloving was the only way to approach the voluminous hematoma related to the delay in management.展开更多

It is...Introduction: Penile fracture is a urological emergency that occurs when the penis in an erect state suffers a blunt trauma resulting in a rupture of the tunica albuginea of either one or both corpora cavernosa. It is often caused by vigorous sexual intercourse, hence the incidence of penile fracture is under-reported. We therefore present our experience of the clinical presentation and surgical management of penile fracture. Presentation of Cases: We report three cases of penile fracture and all were diagnosed based on their clinical presentation and examination findings. The patients include two middle aged men and one young man, all with history of hearing a popping sound and experiencing sudden onset pain with detumescence of the penis. No radiological investigations were carried out. An emergency surgical repair was done for all patients. Discussion: All the patients had no urethral injury on presentation and underwent immediate surgical repair. The erectile and voiding function of each patient was preserved. Conclusion: The management of penile fracture involves early diagnosis and immediate surgical repair. Early intervention is necessary to preserve penile function.展开更多
