Effects of the dietary Withania somnifera on stressrelief and hepatoprotection in darkbarbel catfish(Pelteobagrus vachelli)

Withania somnifera(L.)Dunal(WS),or Ashwagandha,is used clinically against arthritis,anxiety and insomnia etc,however,its potential in aquafeeds is largely unexplored.We investigated the stress-alleviation and hepatoprotective mechanisms of dietary WS supplementation in adult female darkbarbel catfish.Fish were fed with WS dietary for 5 weeks,subjected to a physical stressor(saline injection),and sampled.Brain were processed for RNA-seq to identify differentially expressed genes(DEGs)linked to stress;serum were assayed for stress-and immune-related biochemical markers;livers were examined for lipid content(Oil Red O staining)and proliferation(PCNA immunos-taining).Network pharmacology were employed to construct a"compound-target"net-work:The bioactive compounds of WS were retrieved from ChEBI and PCIDB,probable targets predicted via Swiss Target Prediction,and the“WS-ingredient-target”regulatory network was constructed using Cytoscape;key ligand-protein interactions were validated by molecular docking.Findings were replicated in juvenile fish and in SG3 cells challenged with Withanolide A.Physical stress evoked 1451 DEGs in control brains,whereas WS-supplemented reduced this to 397,indicating marked mitigation of the oxid-ative-and immune-stress response.Network pharmacology and molecular docking pre-dicted that 13 bioactive compounds in WS,especially Withanolide A and Withanolide J,can interact with targets such as MAPK8,CRHR1,NR3C2,and IARS1,being associated with stress and liver function regulation respectively.Moreover,it was proved that Withanolide A could significantly modulate stress response in juveniles and enhanced pro-liferation potency of SG3 cells.Intriguingly,in theadult female darkbarbel catfish,dietary WS lowered serum cortisol,glucose,ROS,TNF-α,and IL-6(P<0.05),up-regulated hepatocyte PCNA expression,and allievated stress-induced liver injury.These findings provide novel insights and vital basis for elucidating the mechanisms whereby herbs,including WS act in stress-relief and hepatoprotection in darkbarbel catfish.And provide a framework for advancing traditional Chinese medicine(TCM)applications in aquaculture. 展开更多

Background:Muscle is the complex and heterogeneous tissue,which comprises the primary edible part of the trunk of fish and mammals.Previous studies have shown that dietary isoleucine(Ile)exerts beneficial effects on growth in aquatic animals.However,there were limited studies regarding the benefits of Ile on fish muscle and their effects on flesh quality and muscle growth.Thus,this study was conducted to explore whether dietary Ile had affected flesh quality and muscle growth in hybrid bagrid catfish(Pelteobagrus vachelli♀×Leiocassis longirostris♂).Methods:A total of 630 hybrid fish,with an initial average body weight of 33.11±0.09 g,were randomly allotted into seven experimental groups with three replicates each,and respectively fed seven diets with 5.0,7.5,10.0,12.5,15.0,17.5,and 20.0 g Ile/kg diets for 8 weeks.Results:In the present study,we demonstrated that Ile significantly:(1)increased muscle protein and lipid contents and the frequency distribution of myofibers with≤20μm and≥50μm of diameter;(2)improved pH value,shear force,cathepsin B and L activities,hydroxyproline content,resilience,cohesiveness,and decreased cooking loss,lactate content,hardness,springiness,gumminess,and chewiness;(3)decreased reactive oxygen species(ROS),malondialdehyde(MDA),and protein carbonyl(PC)contents,GCLC and Keap1 mRNA levels,and up-regulated CuZnSOD,CAT,GPX1a,GST,and Nrf2 mRNA levels;(4)up-regulated the insulin-like growth factor 1,2(IGF-1,IGF-2),insulin-like growth factor 1 receptor(IGF-1R),proliferating cell nuclear antigen(PCNA),Myf5,Myod,Myog,Mrf4,and MyHC mRNA levels,and decreased MSTN mRNAlevel;(5)increased muscle protein deposition by activating AKT-TOR-S6K1 and AKT-FOXO3a signaling pathways.Conclusion:These results revealed that dietary Ile improved flesh quality,which might be due to increasing nutritional content,physicochemical,texture parameters,and antioxidant ability;promoting muscle growth by affecting myocytes hyperplasia and hypertrophy,and muscle protein deposition associated with protein synthesis and degradation signaling pathways.Finally,the quadratic regression analysis of chewiness,ROS,and protein contents against dietary Ile levels suggested that the optimal dietary Ile levels for hybrid bagrid catfish was estimated to be 14.19,12.36,and 12.78 g/kg diet,corresponding to 36.59,31.87,and 32.96 g/kg dietary protein,respectively. 展开更多

Based on pure lines of yellow catfish,super-males and physiological females of yellow catfish were selected by GMT technique,hormonal sex reversal method and test-crossing to establish the germplasm bank of YY physiological females( YY♀). The propagation system of male yellow catfish( XY♂) was established based on the combination of super-males( YY♂) and pure-line female( XX♀) for large-scale production of pure-line yellow catfish males,which effectively overcome the disadvantages in the production of common fingerlings and male fingerlings using low-grade fish with slow growth and small size at the bottom of the fish grader as broodfish,such as progeny germplasm degeneration,loss of growth vigor and reduction of breeding benefits. The technical route of breeding pure-line YY physiological females( YY♀) and YY super-males( YY♂) laid the foundation for large-scale production of environmentally friendly yellow catfish males with pure germplasm and strong stress resistance,and provided an efficient,stable,healthy,environmentally friendly,energy-saving and income-increasing approach for sustainable development of yellow catfish breeding industry. 展开更多

Androgenesis was the especial zoogamy that the germ plasma of offspring was from the agnate. In this study the eggs of Yellow catfish (Pelteobagrus fulvidraco) were irradiated by UV suspending in the synthetic ovarian fluid (OF), and the total dosage of UV irradiation was 220 mJ·cm^-2. Diploid could be induced by heat shock (40℃, 2 min, 3 min) with different time period after fertilization (15-37 min). The result showed that heat shock with 2 min was better than 3 min; there were two apices of induction in 17-21 min and 27-31 min after fertilization. The highest hatching rate was 3.30% at 29 min after fertilization, and the difference between two apices of induction was unobvious. 展开更多

[Objectives] This study was conducted to study early reproduction techniques of yellow catfish(Pelteobagrus fulvidraco) in Jianghan Plain. [Methods] Parents of P. fulvidraco were cultured by the methods of artificial temperature control and nutrition enhancement. [Results] After culture, the body weight, gonad weight, maturity coefficient and egg diameter of P. fulvidraco increased gradually. Up to April 30, the gonad weight of the female parents of P. fulvidraco cultured in greenhouses was significantly higher than that of the parents under natural conditions(P<0.05), 54% higher than that of the parents naturally cultured at the same period;the maturity coefficient of the parents cultured under strengthened conditions was also significantly higher than that of the parents under natural conditions at the same period(P<0.05);and the ovaries of the strengthened parents were orange yellow and full of eggs, and the number of eggs per fish was relatively high. In early May, the parents cultured under strengthened conditions were induced to spawn artificially. The average induced spawning rate and fertilization rate reached 86.7% and 71.6%, respectively, reaching the respective normal level. The gonad maturity of P. fulvidraco cultured under strengthened conditions was 15 d earlier than that under natural conditions, resulting in mass production of early-propagated larvae of P. fulvidraco. [Conclusions] In Jianghan plain area, artificial propagation can be achieved in early May by means of artificial temperature control and strengthened culture, which is 20 d ahead of natural conditions. 展开更多

该研究旨在探明十二烷基苯磺酸钠(SDBS)对黄颡鱼(Pelteobagrus fulvidraco)胚胎发育毒性和仔鱼神经毒性的影响,科学评估水体中SDBS污染的环境生态风险。将受精后4 h的黄颡鱼胚胎暴露于质量浓度为0、0.2、0.4、0.6、0.8 mg·L^(-1)的... 该研究旨在探明十二烷基苯磺酸钠(SDBS)对黄颡鱼(Pelteobagrus fulvidraco)胚胎发育毒性和仔鱼神经毒性的影响,科学评估水体中SDBS污染的环境生态风险。将受精后4 h的黄颡鱼胚胎暴露于质量浓度为0、0.2、0.4、0.6、0.8 mg·L^(-1)的SDBS中,观察胚胎孵化累计死亡率、畸形率;并通过检测5日龄仔鱼脑组织中乙酰胆碱酯酶活性及相关基因的转录水平、5-羟色胺及相关基因的转录水平,探讨其神经毒性的潜在机制。结果显示,与对照相比,暴露处理的黄颡鱼胚胎孵化畸形率提高4.5~14.5百分点(P<0.05),死亡率提高7.8~23.4百分点(P<0.05)。与对照相比,暴露处理正常仔鱼和畸形仔鱼的乙酰胆碱酯酶活性分别显著降低12.0%~50.5%和15.0%~53.9%(P<0.05),暴露处理仔鱼乙酰胆碱酯酶基因mRNA转录水平下降;暴露处理正常仔鱼和畸形仔鱼5-羟色胺含量分别显著降低2.6%~9.9%和3.0%~11.1%(P<0.05),暴露处理仔鱼5-羟色胺基因的转录水平降低。上述结果表明,十二烷基苯磺酸钠暴露显著提高了黄颡鱼胚胎孵化的死亡率和畸形率;SDBS通过抑制乙酰胆碱酯酶活性和5-羟色胺含量水平影响神经传导,抑制神经递质相关基因表达,从而对黄颡鱼产生胚胎发育毒性及神经毒性作用。 展开更多

Feeding time changed the diurnal composition of intestinal microbiota in Pelteobagrus fulvidraco

Although the effects of diet on intestinal microbiotas have been well investigated,the effect of feeding time on the rhythm of gut microbiota remains uncertain.A 9-week feeding trial on Pelteobagrus fulvidraco was conducted.Two triplicate groups of fish were fed identically either at 8:00(day)or at 20:00(night).After 9 weeks,three fish from each cage were sacrificed every 4 h for 24 h.The intestinal contents were analyzed using high-throughput sequencing.Three phyla(Fusobacteria,Proteobacteria,and Verrucomicrobia),and three genera(Cetobacterium,Akkermansia,and Clostridium)were dominant at every time point.Moreover,33.02%of detected amplicon sequence variants showed cyclical variations in fish fed at 20:00,and 3.47%in fish fed at 8:00.Among the top 20 genera,17 showed cyclical variations in the 20:00 group,and only two in the 8:00 group.Rhodobacter,Ralstonia,Mycobacterium,Lactococcus,Halomonas,Akkermansia,and Acinetobacter contributed more to the predicted functional pathways.Moreover,144 and 38 pathways exhibited daily cyclical fluctuations in fish fed at 20:00 and 8:00,respectively.Overall,certain intestinal microorganisms showed a circadian rhythm in P.fulvidraco,and this circadian rhythm was affected by shifted feeding times. 展开更多

Glass catfish(Kryptopterus vitreolus)are notable in the aquarium trade for their highly transparent body pattern.This transparency is due to the loss of most reflective iridophores and light-absorbing melanophores in the main body,although certain black and silver pigments remain in the face and head.To date,however,the molecular mechanisms underlying this transparent phenotype remain largely unknown.To explore the genetic basis of this transparency,we constructed a chromosome-level haplotypic genome assembly for the glass catfish,encompassing 32 chromosomes and 23344 protein-coding genes,using PacBio and Hi-C sequencing technologies and standard assembly and annotation pipelines.Analysis revealed a premature stop codon in the putative albinism-related tyrp1b gene,encoding tyrosinase-related protein 1,rendering it a nonfunctional pseudogene.Notably,a synteny comparison with over 30 other fish species identified the loss of the endothelin-3(edn3b)gene in the glass catfish genome.To investigate the role of edn3b,we generated edn3b^(−/−)mutant zebrafish,which exhibited a remarkable reduction in black pigments in body surface stripes compared to wild-type zebrafish.These findings indicate that edn3b loss contributes to the transparent phenotype of the glass catfish.Our high-quality chromosome-scale genome assembly and identification of key genes provide important molecular insights into the transparent phenotype of glass catfish.These findings not only enhance our understanding of the molecular mechanisms underlying transparency in glass catfish,but also offer a valuable genetic resource for further research on pigmentation in various animal species. 展开更多

为研究饲料中添加黄芪多糖(Astragalus polysaccharides, APS)对黄颡鱼(Pelteobagrus fulvidraco)生长、抗氧化及肠道健康的影响,本实验以初始体质量为(28.10±0.51) g的黄颡鱼为研究对象,分别饲喂添加0 mg/kg(CK组)、300 mg/kg(A组... 为研究饲料中添加黄芪多糖(Astragalus polysaccharides, APS)对黄颡鱼(Pelteobagrus fulvidraco)生长、抗氧化及肠道健康的影响,本实验以初始体质量为(28.10±0.51) g的黄颡鱼为研究对象,分别饲喂添加0 mg/kg(CK组)、300 mg/kg(A组)、600 mg/kg(B组)、900 mg/kg(C组)、1 200 mg/kg(D组)、1 500 mg/kg(E组)黄芪多糖的饲料,饲养56 d后测定生长性能指标、血清生化指标、抗氧化指标、肝脏肠道消化酶活性及观察肠道结构。结果显示:与CK组相比,除A组外,其余各实验组的摄食量、特定生长率显著提高,饲料系数显著降低;实验组中黄颡鱼血清总胆固醇含量随APS添加量增加显著降低,总蛋白和白蛋白水平呈剂量依赖性上升;实验组中黄颡鱼消化酶活性及抗氧化能力则呈现为先增强后减弱的趋势,其中D组黄颡鱼肠道过氧化氢酶活性与肝脏丙二醛含量的变化幅度最为显著。肠道形态学分析表明,肠道切片各实验组中D组黄颡鱼绒毛长度和肌层厚度提高程度最显著。结果表明,在黄颡鱼饲料中添加300~1 500 mg/kg的APS能够提高其生长性能和抗氧化能力及改善肠道健康,黄颡鱼饲料中APS的适宜添加量为1 200 mg/kg。 展开更多

混养黄颡鱼对大口黑鲈营养品质、肠道菌群及养殖微生态环境的调控效应

【目的】探讨混养黄颡鱼对大口黑鲈生长性能、营养品质、肠道菌群及养殖微生态环境的影响,筛选出适宜的混养比例,为开展大口黑鲈的绿色健康养殖提供参考依据。【方法】设大口黑鲈单养组(C组)及低密度混养组(L组,大口黑鲈∶黄颡鱼=4∶1)... 【目的】探讨混养黄颡鱼对大口黑鲈生长性能、营养品质、肠道菌群及养殖微生态环境的影响,筛选出适宜的混养比例,为开展大口黑鲈的绿色健康养殖提供参考依据。【方法】设大口黑鲈单养组(C组)及低密度混养组(L组,大口黑鲈∶黄颡鱼=4∶1)、中密度混养组(M组,大口黑鲈∶黄颡鱼=2∶1)和高密度混养组(H组,大口黑鲈∶黄颡鱼=1∶1),养殖60 d后,分别测定大口黑鲈的生长性能、肌肉营养品质、肠道菌群及养殖水体微生物群落特征。【结果】M组大口黑鲈的特定生长率(1.69%/d)和肥满度(1.52)最高,而饲料系数(1.08)最低。M组和H组大口黑鲈肌肉的粗蛋白含量显著高于C组大口黑鲈(P<0.05,下同);M组大口黑鲈肌肉的必需氨基酸和鲜味氨基酸含量最高,均显著高于C组大口黑鲈;M组大口黑鲈肌肉的硬度和胶着性显著高于C组大口黑鲈,而土腥味物质(土腥素和2-甲基异莰醇)含量显著低于C组大口黑鲈。混养黄颡鱼对大口黑鲈肠道菌群结构产生明显影响,与C组大口黑鲈相比,M组大口黑鲈肠道中的芽孢杆菌门和假单胞菌门相对丰度显著升高,放线菌门相对丰度则显著降低;鞘氨醇单胞菌属相对丰度随黄颡鱼混养密度的升高而显著上升,且与肌肉粗蛋白含量呈显著正相关。此外,混养黄颡鱼导致大口黑鲈养殖水体微生物群落结构发生明显变化,M组养殖水体中的马赛菌属和微小杆菌属相对丰度显著高于C组,且微小杆菌属相对丰度与绿藻门密度呈极显著正相关(P<0.01),与大口黑鲈肠道菌群中的鞘氨醇单胞菌属相对丰度呈显著正相关。【结论】大口黑鲈按2∶1的比例与黄颡鱼进行混养,能实现养殖生态重塑,不仅显著提高了大口黑鲈的生长性能、改善了肌肉营养品质,还优化了大口黑鲈肠道菌群结构及养殖微生物环境,实现生长—品质—环境协同提升,是一种理想的生态养殖模式。 展开更多

我国水产养殖业规模持续扩大,但传统鮰鱼称量方法存在效率低、误差大、鱼体应激损伤严重等问题。提出了一种基于机器视觉与多传感器协同的鮰鱼无人称量系统,融合目标检测算法与多源信息测量技术。通过改进的YOLO 11模型(HY-YOLO 11),引... 我国水产养殖业规模持续扩大,但传统鮰鱼称量方法存在效率低、误差大、鱼体应激损伤严重等问题。提出了一种基于机器视觉与多传感器协同的鮰鱼无人称量系统,融合目标检测算法与多源信息测量技术。通过改进的YOLO 11模型(HY-YOLO 11),引入可变形卷积(DCNv3)与空间增强注意力模块(SEAM),有效解决动态场景下鱼群粘连、笼体形变及光照波动干扰问题。系统集成浮式监测平台、工业相机与拉力传感器,结合自适应吊装机构与浮力补偿机制,实现鱼群总质量与数量的同步精准测量。实验结果表明,HY-YOLO 11模型在密集遮挡场景下的平均检测精度(mAP50)达94.8%,较基线模型提升3.7个百分点,鱼群计数平均绝对误差(MAE)为0.56条,均质量计算相对误差低于4%,单次称量平均耗时58 s。 展开更多

本文结合安徽阜阳太和县黄颡鱼陆基圆池循环水养殖实践,总结了养殖系统构建、放养前准备、种苗放养和饲养管理等关键技术环节,并分析了该模式养殖实践的经济效益。陆基圆池循环水养殖技术包括选择水源充沛、交通方便、电力稳定的区域建... 本文结合安徽阜阳太和县黄颡鱼陆基圆池循环水养殖实践,总结了养殖系统构建、放养前准备、种苗放养和饲养管理等关键技术环节,并分析了该模式养殖实践的经济效益。陆基圆池循环水养殖技术包括选择水源充沛、交通方便、电力稳定的区域建设陆基圆池循环水养殖系统(陆基圆形养殖池、进排水系统、配套设备、养殖尾水处理系统和水质在线监测系统);放养前进行全面的系统通风和消毒;选择来源正规、体健无虫的鱼种;养殖过程中,根据鱼体情况进行精准投喂;每日使用水质在线监测设备检测池中水体水温、溶解氧等理化指标(溶解氧≥5.5 mg/L,pH 7.0~8.0,氨氮含量<0.4 mg/L,亚硝酸盐含量<0.1 mg/L);定期处理微滤机及三级沉淀池中的固体废弃物;定期使用中草药或化学药剂对养殖水体进行杀虫、消毒处理。养殖实践表明,经过200 d的陆基圆池养殖,黄颡鱼平均饵料系数为1.50,平均存活率为89.67%;平均每池黄颡鱼产量为1222.33 kg,产值为30558元,净利润为4524元。综合表明,该技术模式具有安装灵活、占地面积小、水资源循环利用等优势,符合现代渔业绿色发展方向,同时可获得较佳的经济效益,适宜在生产中推广应用。 展开更多

黄颡鱼生长相关SSR_(S)多态性与性状关联分析

本研究选取黄颡鱼(Pelteobagrus fulvidraco)生长激素(growth hormone,gh)基因微卫星位点gh-GA和生长性状QTL定位点区域内微卫星pfu11克隆序列pfu11-CA位点,分析同一批次繁殖的12月龄、体质量1 g左右的300尾黄颡鱼的遗传多样性和主要生... 本研究选取黄颡鱼(Pelteobagrus fulvidraco)生长激素(growth hormone,gh)基因微卫星位点gh-GA和生长性状QTL定位点区域内微卫星pfu11克隆序列pfu11-CA位点,分析同一批次繁殖的12月龄、体质量1 g左右的300尾黄颡鱼的遗传多样性和主要生长性状的关联情况。结果表明,位点gh-GA在267尾样本中检测到的等位基因数(N_(a))为6,观测杂合度(H_(o))为0.801,期望杂合度(H_(e))为0.749,多态信息含量(PIC)为0.708;位点pfu11-CA共有292尾样本获得分型结果,N_(a)为5,H_(o)为0.592,He为0.623,PIC为0.548。这两个位点均为高度多态,符合哈代温伯格平衡,不存在连锁不平衡现象。性状关联分析结果表明,gh-GA位点DE基因型的全长显著高于DF和CE基因型,DE基因型的体长显著高于AF、DF和CE基因型。pfu11-CA位点BC基因型的全长和体长均极显著高于DE基因型,AC和BC基因型的全长和体长均显著高于AE和DE,AC和BC基因型的体质量均显著高于DE。gh-GA位点的DE基因型,以及pfu11-CA位点的AC和BC基因型为优势基因型,这两个位点可作为生长性状的候选位点。本研究可为开展黄颡鱼生长相关遗传标记研究提供可靠的分子遗传学工具。 展开更多

采用无菌水(CK组)、3.0 mg/mL绿原酸(chlorogenic acid,CGA)溶液(CGA组)、1.0 mg/mL壳聚糖(chitosan,CTS)溶液(CTS组)、1.0 mg/mL CTS和3.0 mg/mL CGA混合溶液(C+组)浸泡鮰鱼片后低温贮藏,通过对比不同贮藏时间鮰鱼片蒸煮损失率及熟制... 采用无菌水(CK组)、3.0 mg/mL绿原酸(chlorogenic acid,CGA)溶液(CGA组)、1.0 mg/mL壳聚糖(chitosan,CTS)溶液(CTS组)、1.0 mg/mL CTS和3.0 mg/mL CGA混合溶液(C+组)浸泡鮰鱼片后低温贮藏,通过对比不同贮藏时间鮰鱼片蒸煮损失率及熟制后鱼肉剪切力、微观结构、水分分布、挥发性成分等指标的变化情况,分析CTS-CGA复合保鲜剂对冷藏鮰鱼片食用品质的影响。结果表明:随着贮藏时间的延长,鮰鱼片蒸煮损失率上升,熟制后剪切力下降、肌肉纤维由有序变为无序,鱼肉中水分流动性和相对含量逐渐下降。贮藏6 d后,CK组不良风味物质相对含量显著高于C+组;其中,CK组己醛和三甲胺相对含量分别为40.11%和8.87%;C+组己醛相对含量仅为5.33%,未检出三甲胺,CTS-CGA复合保鲜剂可有效抑制不良风味的产生。综上,CTS-CGA复合保鲜剂可有效延缓鮰鱼片质地劣变,延缓鮰鱼片食用品质下降。 展开更多

为分析鮰鱼在低温贮运期间品质变化的规律并确定其优势腐败菌,该研究将鮰鱼肉块分别置于4和10℃条件下冷藏,通过测定感官指标、质构特性、色度、pH值、菌落总数(total viable count, TVC)及挥发性盐基氮(total volatile basic nitrogen,... 为分析鮰鱼在低温贮运期间品质变化的规律并确定其优势腐败菌,该研究将鮰鱼肉块分别置于4和10℃条件下冷藏,通过测定感官指标、质构特性、色度、pH值、菌落总数(total viable count, TVC)及挥发性盐基氮(total volatile basic nitrogen, TVB-N)含量等指标,系统分析其品质变化规律。同时,采用高通量测序和16S rDNA技术鉴定分析鮰鱼中的微生物组成,并对优势腐败菌的致腐能力进行评估。结果表明,冷藏期间鮰鱼的pH值呈现先下降后上升的趋势,硬度、咀嚼度及红绿值a^(*)均呈下降趋势,且TVB-N含量与TVC呈正相关。其中4℃条件下鮰鱼的腐败速率较慢,感官品质保持较好,微生物分析显示,相对丰度较高的气单胞菌属(Aeromonas)(28.74%)和希瓦氏菌属(Shewanella)(6.34%)致腐能力也最强,是鮰鱼冷藏期间的优势腐败菌。综上所述,鮰鱼4℃条件下冷藏时的新鲜度和品质保持较好,气单胞菌属和希瓦氏菌属是导致鮰鱼腐败的主要菌属。该研究为基于菌群结构演替规律的淡水鱼品质控制技术开发提供了一定的理论依据和数据支持。 展开更多

利用40个黄颡鱼微卫星分子标记对吉林省月亮湖野生黄颡鱼(YL,30尾)、四川野生黄颡鱼(SC,30尾)、黑龙江省哈尔滨市松花江段黄颡鱼(SH,18尾)、湖北省荆州市长湖野生黄颡鱼子代(YY,30尾)及其亲本(QQ,30尾)、天津市换新国家级原良种场黄颡鱼... 利用40个黄颡鱼微卫星分子标记对吉林省月亮湖野生黄颡鱼(YL,30尾)、四川野生黄颡鱼(SC,30尾)、黑龙江省哈尔滨市松花江段黄颡鱼(SH,18尾)、湖北省荆州市长湖野生黄颡鱼子代(YY,30尾)及其亲本(QQ,30尾)、天津市换新国家级原良种场黄颡鱼1号(TJ,30尾)6个黄颡鱼群体的观测杂合度(Ho)、期望杂合度(He)、多态信息含量(PIC)和有效等位基因数(Ae)等进行了遗传检测,根据基因频率计算遗传相似系数和Nei氏标准遗传距离,x2检验估计Hardy-Weinberg平衡,采用近交系数(FST)和基因流(Nm)分析群体的遗传分化及其来源。同时,使用PHYLIP3.63软件绘制基于Nei氏标准遗传距离的UPGMA进化树。6个群体共检测到5042个扩增片段,长度在150—650bp,40个基因座扩增出等位基因数从1—8个不等,共计143个等位基因,平均每个基因座扩增得到3.575个等位基因。结果显示:(1)6个黄颡鱼群体的多态性指标均适中,PIC在0.00—0.79之间,平均值为0.40、0.37、0.35、0.39、0.35和0.19,有效等位基因数(Ae)1.00—5.44个不等,平均有效等位基因数依次为2.05、2.30、1.93、2.07、1.93和1.47,无偏期望杂合度(He)在0.00—0.83之间,平均值为0.45、0.46、0.36、0.46、0.41和0.23;(2)遗传相似系数YY与QQ遗传相似度最高(0.9947),TJ与SC遗传相似系数最低(0.4846),聚类结果与地理分布呈一定相关性。 展开更多

春夏季投喂率与夏季间歇性断食对杂交黄颡鱼“黄优1号”生长、存活和健康指标的影响 被引量:1

为探究杂交黄颡鱼更加绿色高效的投喂策略,研究以杂交黄颡鱼“黄优1号”为试验对象,在湖北地区池塘网箱中开展了春季不同投喂率[0、0.5%、表观饱食(2.1%)]、夏季不同投喂率[1%、2%、表观饱食(3.9%)]和不同间歇性断食投喂(3d饱食1d断食,... 为探究杂交黄颡鱼更加绿色高效的投喂策略,研究以杂交黄颡鱼“黄优1号”为试验对象,在湖北地区池塘网箱中开展了春季不同投喂率[0、0.5%、表观饱食(2.1%)]、夏季不同投喂率[1%、2%、表观饱食(3.9%)]和不同间歇性断食投喂(3d饱食1d断食, 7d饱食1d断食)对杂交黄颡鱼生长、存活和肝功能、肠道屏蔽及免疫功能等健康指标的影响。研究结果显示,在春季(4月12日—5月3日),随投喂率增加,生长速度显著提高,存活率显著降低, 0.5%投喂率下杂交黄颡鱼的健康指标显著优于0%投喂率组和表观饱食投喂组。在夏季(6月1日—7月13日),随投喂率增加,生长速度显著上升,到达表观饱食投喂后, 3d饱食1d断食的间歇性断食方案显著降低生长速度, 7d饱食1d断食的间歇性断食方案与表观饱食投喂的生长速度无显著差异。此外,夏季研究结果进一步显示,不同投喂率组和不同间歇性断食方案组之间的存活率无显著性差异, 2%投喂率下的健康指标显著优于1%投喂率组、表观饱食组、3d饱食1d断食组和7d饱食1d断食组。综合上述研究结果和生产实践需求,建议在春季水温15—26℃波动时50 g左右杂交黄颡鱼的投喂率为0.5%,该投喂率既能保证及时恢复体质,又能显著降低春季投喂恢复期的死亡风险;建议夏季水温26—32℃时10 g左右杂交黄颡鱼采用7d饱食1d断食的投喂策略,既能保证最快生长速度,又能节省约14%总饲料投入量。 展开更多
