文摘Overweight and obesity are significant public health concerns worldwide due to their association with many chronic health conditions. This has resulted in the development of various interventions focused on weight loss to reduce the associated health burden.Physical activity is an important lifestyle behavior associated with enhanced health.Evidence supports that many of the benefits of physical activity are realized independent of initial weight status or whether weight loss is achieved, with some benefits additive to what is achieved with weight loss alone. These benefits include enhanced cardiometabolic, brain, cognitive and psychological health, and others. Moreover,in adults with overweight or obesity, physical activity has independent effects on cardiorespiratory fitness, muscular strength, physical function, and mobility. There are also benefits to body composition, with physical activity improving the quality of key tissues, such as skeletal muscle, which may not occur with diet-induced weight loss. Therefore, physical activity is an important public health target for adults with overweight or obesity to provide a wide range of health benefits that extend beyond those of weight loss alone. However, physical activity recommendations and programming efforts should consider the unique characteristics of adults with overweight or obesity to be most effective, and should support a focus on mobility, physical function, and other health outcomes.

文摘Children with autism often exhibit abnormalities in body weight, but the underlying mechanism remains unclear.SH3 and multiple ankyrin repeat domains protein 3(SHANK3),a scaffold protein of the postsynaptic density, has been reported to be associated with autism. This study aimed to investigate whether and how SHANK3 influences body weight in the hypothalamic neuronal regulation of energy homeostasis.Adeno-associated viruses 9(AAV9)carrying CMV-Cre and Agrp-Cre were stereotactically injected to restore SHANK3 expression in the arcuate nucleus(ARC)and agouti-related peptide(AgRP)neurons,respectively.Agrp-Cre mice were injected with AAV9-p38αflox/flox to overexpress p38α.Activated p38αwas generated by mutating both D176A and F327S in p38α.Inactivated p38αwas constructed by mutating both T180A and Y182F in p38α.Metabolic analysis, immunoblotting, histological analysis, the glucose tolerance test, the insulin tolerance test, and body fat mass analysis were applied to investigate the underlying mechanisms by which SHANK3 regulates body weight. We reveal that SHANK3 regulates body weight via the p38αsignaling pathway in the AgRP neurons of the hypothalamus.Shank3 knockout(Shank3−/−)mice exhibit resistance to diet-induced obesity.Shank3 re-expression in the ARC or AgRP neurons increases body weight in Shank3 knock-in mice with an inverted allele(SKO).Overexpression or activation of p38αin AgRP neurons elicits resistance to diet-induced obesity.Inactivated p38αin AgRP neurons abolished the resistance to diet-induced obesity due to SHANK3 deficiency. Our findings suggest that the SHANK3-p38αsiganling pathway in AgRP neurons regulates body weight balance in autism, revealing a promising therapeutic target for obesity in children with autism.
文摘Purpose:This study examined potential differences in strength, muscle morphology, and motor unit(MU)behavior of the abductor digiti minimi(ADM)between normal-fat(NF)and over-fat(OF)males.Methods:Dual-energy X-ray absorptiometry assessed percent body fat(%BF).Ultrasonography determined muscle cross-sectional area(CSA),echo intensity(EI),and subcutaneous fat(s FAT).MU behavior was assessed during isometric muscle actions at 50%of maximal voluntary contraction(MVC)by analyzing the y-intercepts and slopes for the MU action potential amplitude(MUAPAMP)vs.recruitment threshold(RT)relationships, the A and B terms for the mean firing rate(MFR)vs.RT relationships, and normalized electromyographic amplitude(N-EMGRMS).MU firing times and waveforms were validated with reconstruct-and-test and spike trigger average procedures. Results:%BF was greater for OF(25.70%±5.40%)than NF(16.50%±2.20%;p<0.001).MVC was greater for NF(27.13±7.16)N than OF([19.89±4.96]N;p=0.014).CSA was greater for NF(2.48±0.39)cm^(2)than OF([1.95±0.47]cm^(2);p=0.011).The y-intercepts for the MUAPAMPvs.RT relationships were greater for NF(0.283±0.254)m V than OF([-0.221±0.659]m V;p=0.004).The B terms for the MFR vs.RT relationships were greater for NF(-0.024±0.003)pps/%MVC than OF([-0.031±0.009]pps/%MVC;p=0.038).N-EMGRMSwas similar between groups(p=0.463).Conclusion:Maximal strength, muscle size, and MU recruitment and firing rate patterns for a non-weight bearing muscle differed between normal-fat and over-fat males.

文摘Pelagic appendicularians, known for their rapid bloom capability, are important secondary producers and play essential roles in marine ecosystems. To assess the environmental adaptation of the most common appendicularian species Oikopleura dioica in Jiaozhou Bay,Shandong,China, its annual variations in abundance and body size were investigated monthly at 12 stations in 2011,and the relationship between somatic growth and gonadal development was compared with previously published laboratory data.Oikopleura dioica exhibited distinct seasonal abundance patterns, peaking in summer and autumn, particularly in the inner bay, reflecting its adaptation to the eutrophic conditions and high food availability.Compared to laboratory results,O.dioica in Jiaozhou Bay exhibited shorter body lengths, smaller gonad volumes, and earlier maturation at reduced body sizes, despite high natural chlorophyll-a concentration, suggesting that factors beyond food availability, such as resource competition and environmental stressors, may limit its growth in natural settings.Correlation analysis indicated that the somatic growth of natural O.dioica was primarily influenced by temperature, while gonadal development was related to chlorophyll-a concentration, and its population abundance was linked to food availability and the abundance of other zooplankton, such as copepods and jellyfish. It was suggested that the growth and recruitment of O.dioica were constrained by resource limitations in natural environments, further exacerbated by interactions with other zooplankton, including predation and competition. This study provides valuable data on the mechanisms underlying bio-environmental interactions and their spatiotemporal variations, offering critical insights into the ecological roles of appendicularians in marine ecosystems.

文摘AIM:To investigate the causal effect of obesity on cataract risk and explores the potential mediating roles of metabolites using Mendelian randomization(MR).METHODS:Summary-level data from large-scale genome-wide association studies to examine the relationship between obesity and cataract were utilized.Obesity-related traits, including body mass index(BMI),waist-to-hip ratio(WHR),and waist circumference(WC).A two-sample MR approach was employed to assess the causal effect of obesity on cataract risk, while potential mediators were identified from suitable genome-wide association studies(GWAS)datasets.Additionally,a metabolic pathway analysis was conducted.RESULTS:An increase of 1 standard deviation(SD)in BMI,WHR,and WC was associated with a significantly higher risk of cataract(BMI:odds ratio(OR)1.0017,95%confidence interval(CI):1.0001-1.0032,P=0.0320;WHR:OR 1.0029,95%CI:1.0006-1.0051,P=0.0129;WC:OR 1.0020,95%CI:1.0001-1.0038,P=0.0390].These associations remained robust after adjusting for confounding factors in multivariable MR analysis.Furthermore,a two-step MR analysis identified eight potential metabolic mediators, with one mediator showing a significant causal role in the relationship between obesity and cataract.CONCLUSION:This work highlights the importance of addressing obesity as a modifiable risk factor for cataracts, particularly through metabolic pathways.
文摘Self-tracking(ST)has progressively increased since the turn of the millennium.Today, every smartphone is equipped with pre-installed“health apps”that cannot be deleted individually. With relatively inexpensive external sensors, such as wristbands, belts, and watches,a variety of parameters are monitored to calculate further data.Direct networking via smartphones allows users to share such data on social networks. The sociological implications of this behaviour, which can also be employed for self-optimisation,have been summarised.Selected ethical and philosophical issues—such as data security and epistemic justice have also been explored in detail. However, the phenomenological and philosophical question of how ST impacts the original perception of one’s own body has been considered quite sparingly thus far.The experience of“being oneself”—the subjective sensing and feeling of the bodily person in self-perception and in interpersonal encounters within the intersubjective bodily resonance space—remains under-explored.Extensive ST can lead to an embodiment of the body,as the supposedly“objective”body data of ST can dictate the primary bodily sensing and feeling. This results in a widening gap between the physical body and the animated body.In my view, this corresponds to a development analogous to the currently prevailing reductionist philosophy of mind, which offers strong materialistic simplifications(the brain generates consciousness solely through neuronal activities),where bodily experience or phenomenal consciousness through the subjective experiential content of mental processes(“qualia”)no longer plays a role.
文摘Background:Penile augmentation through injectable substances is becoming increasingly common.A growing number of aesthetic clinics are developing penile enlargement procedures using various injectable materials.Although these procedures are now performed in more controlled and medically supervised environments, their long-term outcomes remain poorly understood. The promotion of such medical treatments contributes to an increasing interest among adult males in self-injection as a method to alleviate psychological distress associated with penile size concerns. At the same time, access to injectable substances through unofficial or unregulated sources has become increasingly easy.Tor our knowledge,we report the first documented case of self-injection with Garamycin®(gentamicin)cream, contributing to the literature on the often multidisciplinary management of penile enlargement injections,a field still lacking well-established guidelines.Case Description:This case report describes a young patient who self-injected Garamycin®into the penis for the purpose of enlargement. He presented to our urology department with worsening symptoms, including severe and poorly tolerated pain. His primary request was prompt relief of pain while preserving,as much as possible, the aesthetic appearance and functional integrity of his penis. This case required a multi-stage surgical approach to salvage the penis and preserve both its structural integrity and functional outcome. Conclusions:To our knowledge, this case report documents the first reported instance of Garamycin®injection performed for the purpose of penile enlargement. It provides insight into the clinical course of such penile cream injections, demonstrates that a two-stage scrotal flap can achieve both functional and aesthetic outcomes, and highlights the importance of comprehensive management particularly addressing the traumatic impact of penile deformity secondary to inflammation and/or infection,as well as the body dysmorphic concerns often associated with these cases.
文摘Overweight and obesity has been a major public health problem globally. It was estimated that more than 2.1 billion adults were affected by overweight or obese in 2021 worldwide, about one fifth of whom lived in China^([1]).By 2050,the country is forecast to remain the one with the largest population of overweight and obese globally^([1]),if no effective strategies were applied on overweight/obesity control.

文摘Parkinson’s disease(PD)is a progressive age-related neurodegenerative disorder clinically defined by motor symptoms and pathologically by the loss of dopaminergic(DA)neurons in the substantia nigra pars compacta. These neurons are characterized by the presence of the cytoplasmic pigment neuromelanin(NM),and their degeneration is closely associated with the accumulation ofα-synuclein(α-syn)into intraneuronal inclusions known as Lewy bodies(LBs),which represent a neuropathological hallmark of PD.
文摘A model for dynamic frictionless contact between a viscoelastic body and foundation is considered. The viscoelastic constitutive law is assumed to be nonlinear and the contact is modelled with the normal compliance condition. We obtain the well-posedness using nonlinear semigroup theory arguments. Moreover, the exponential stability result of the solution is shown by using the energy method to produce a suitable Lyapunov function.
