It may progress to severe rigidity, dystonia, and quadriparesis. Combination therapy of high-dose thiamine and biotin helps to control the symptoms and prevent progression of the disease. Methods: This retrospective, cross-sectional study was conducted at King Fahad Medical City in Riyadh, Saudi Arabia, to investigate the demographic, clinical features, treatment response, outcomes, and predictive factors of BTBGD in the pediatric population. Results: Twenty-five records of pediatric patients diagnosed with BTBGD were included in the study. The most common symptoms observed at presentation were ataxia in 13 patients (52%), followed by developmental regression in 11 patients (44%), and seizures in 7 patients (28%). Statistically significant associations were found between patient’s age of presentation, seizures at presentation, lactate level and their health outcomes. Multivariate logistic regression analysis revealed significant differences in patient outcomes (prognosis) based on their age at presentation, seizures, and lactate levels (p Conclusion: This study reported BTBGD in 25 pediatric patients in Saudi Arabia. Age at presentation, seizures, and lactate levels were found to be significantly associated with patient health outcomes. Increasing public awareness of the condition, particularly among parents and pediatricians, is imperative. Food is a critical environmental factor that influences animal survival,especially for small passerines due to their high mass-specific metabolic rates.Basal metabolic rate(BMR)reflects the energy expended by endother...Food is a critical environmental factor that influences animal survival,especially for small passerines due to their high mass-specific metabolic rates.Basal metabolic rate(BMR)reflects the energy expended by endothermic animals for basic physiological processes and constitutes a major part of their daily energy budget.Some birds have been shown to employ compensatory mechanisms during food shortages,temporarily reducing these selfmaintenance expenditures without using hypothermia.However,the mechanisms of BMR adjustment remain unexplored.In the present study,we assessed the phenotypic variation in basal thermogenesis of Eurasian Tree Sparrows(Passer montanus)by comparing a control group to groups fasted for 6,12,18,and 24 h.We focused on the correlation between a reduction in energy metabolism and the alterations of cellular metabolic activities,mitochondrial substrate supply,and changes in serum thyroid hormones during fasting.Our data indicated that fasting groups had significantly lower body mass,BMR,body temperature,and body fat content.Furthermore,fasting groups had significantly lower glycogen levels,mitochondrial state 4 respiration and cytochrome c oxidase(CCO)activity in the liver,and CCO activity in pectoral muscle.The levels of avian uncoupling protein(avUCP)m RNA were significantly reduced,while the levels of myostatin protein in pectoral muscle were significantly increased in the fasting groups.Furthermore,the groups subjected to fasting exhibited significantly lower levels of serum glucose,triglyceride,thyroxine(T_(4)),and triiodothyronine(T_(3)).Positive correlations were observed between the following pairs of variables:log BMR and log body mass,log body mass and log body fat,log BMR and log state 4 respiration in the liver,log BMR and log CCO activity in the liver and muscle,log BMR and log av-UCP m RNA expression,whereas a negative correlation was observed between log BMR and log myostatin level.In addition,a positive correlation was also detected between log T_(3) and each of the following:log BMR,state 4 respiration,and log CCO activity in the liver.Our results suggested that decreased metabolic thermogenesis via down-regulation in cellular aerobic capacity of organs and serum thyroid hormones may be an important survival strategy for fasting Tree Sparrows to reduce energy expenditure.展开更多
Models that predict a forest stand’s evolution are essential for developing plans for sustainable management.A simple mathematical framework was developed that con-siders the individual tree and stand basal area unde...Models that predict a forest stand’s evolution are essential for developing plans for sustainable management.A simple mathematical framework was developed that con-siders the individual tree and stand basal area under random resource competition and is based on two assumptions:(1)a sigmoid-type stochastic process governs tree and stand basal area dynamics of living and dying trees,and(2)the total area that a tree may potentially occupy determines the number of trees per hectare.The most effective method to satisfy these requirements is formalizing each tree diameter and potentially occupied area using Gompertz-type stochastic differential equations governed by fixed and mixed-effect parameters.Data from permanent experimental plots from long-term Lithuania experiments were used to construct the tree and stand basal area models.The new models were relatively unbiased for live trees of all species,including silver birch(Betula pen-dula Roth)and downy birch(Betula pubescens Ehrh.),[spruce(Picea abies),and pine(Pinus sylvestris)].Less reliable predic-tions were made for the basal area of dying trees.Pines gave the highest accuracy prediction of mean basal area among all live trees.The mean basal area prediction for all dying trees was lower than that for live trees.Among all species,pine also had the best average basal area prediction accuracy for live trees.Newly developed basal area growth and yield models can be recommended despite their complex formulation and implementation challenges,particularly in situations when data is scarce.This is because the newly observed plot provides sufficient information to calibrate random effects.展开更多
BACKGROUND Basal cell carcinoma(BCC)is the most prevalent skin cancer,characterized by indolent growth and low metastatic rates.When metastatic BCC(mBCC)does occur,it most commonly involves lymph nodes,lungs,and bones...BACKGROUND Basal cell carcinoma(BCC)is the most prevalent skin cancer,characterized by indolent growth and low metastatic rates.When metastatic BCC(mBCC)does occur,it most commonly involves lymph nodes,lungs,and bones,with metastases to other sites being exceptionally rare.This case reports the first documented instance of mBCC to the stomach,highlighting the importance of considering atypical metastatic sites,and the challenges associated with diagnosis and management.CASE SUMMARY A 52-year-old male with recurrent BCC and known mBCC to the bone presented with progressive dysphagia,cranial neuropathies and generalized weakness.He had been treated with immune checkpoint inhibitors,with intermittent therapy modifications due to treatment related toxicities.His past history was notable for malignant perineural invasion,radiotherapy for osseous metastases,immune checkpoint inhibitor-induced enteritis,and osteonecrosis of the jaw.Gastroscopy revealed subepithelial gastric lesions,and biopsies confirmed mBCC-a previously unreported site of disease dissemination.A multidisciplinary team involving gastroenterology,oncology,neurology,and palliative care guided his management.Given his declining functional status and poor prognosis,he was ultimately transitioned to hospice care.CONCLUSION Clinicians should consider atypical metastatic sites in advanced BCC.A multidisciplinary approach remains essential for timely diagnosis and coordinated management.展开更多

However,it is still not well understood how the BG maintains wakefulness and suppresses sleep to achieve al these fundamental functions until genetical y engineered systems developed these years. Significant research efforts have recently been directed at developing genetic-molecular tools to achieve reversible and cell-type specific in vivo silencing or activation of neurons in behaving animals. Optogenetic tools can be used both to specifically activate or inhibit neurons of interest and identify functional synaptic connectivity between specific neuronal populations,both in vivo and in brain slices. Another recently developed system by Roth and colleagues permits the selective and ″remote″ manipulation(activation and silencing) of neuronal activity via all 3 major GPCR signaling pathways(G_i,G_s and G_q). These so-called ″ designer receptors exclusively activated by designer drugs″(DREADD) involve mutant GPCRs that do not respond to their endogenous ligands but are responsive to otherwise inert biological compounds. Recently,we demonstrated the essential roles and the neural pathways of the neurons expressing adenosine A_(2A) receptors or dopamine D_1 receptors in the BG for sleep-wake regulation using the genetically engineered systems including optogenetics and DREADD. BACKGROUNDβ-ketothiolase deficiency(β-KTD)is an inherited disease,and insufficient attention has been paid to imageology due to its lower morbidity.Therefore,few lesions outside the basal ganglia have been found bef...BACKGROUNDβ-ketothiolase deficiency(β-KTD)is an inherited disease,and insufficient attention has been paid to imageology due to its lower morbidity.Therefore,few lesions outside the basal ganglia have been found before,and the persistent pathological changes have rarely been reported.CASE SUMMARY A 10-mo-old Chinese female patient with a free previous medical history but with poor physical and athletic development had received the haemophilus influenzae vaccine and then developed a low fever 2 d prior.She was initially diagnosed with severe brain injury,central respiratory failure,metabolic acidosis complicated with respiratory alkalosis,hyper-IgE,etc.With further examination,a definite diagnosis ofβ-KTD was made.Symptomatic treatment was adopted.Ten days later,the dyspnea was improved evidently and the ventilator was removed,but there were still obvious abnormalities on magnetic resonance imaging(MRI).The lesions mainly invaded the corpus striatum but were not limited to the basal ganglia.Then,the patient’s disease improved and discharged approximately 1 mo later,and the abnormal lesions on MRI had partially improved.However,for about 1 year,the residual irreversible lesions were observed on MRI,the mental and physical development of the patient was obviously regressive,and extra rehabilitation training was needed.CONCLUSION The case highlights the critical importance of one view that the range of lesions in some patients may be more extensive than previously thought in someβ-KTD patients.In addition to biochemical tests,genetic tests and magnetic resonance imaging are not only conducive to quickly diagnosingβ-KTD but also to partially evaluating the short-and long-term outcomes.Moreover,more attention should be paid to the two mutations(c.478 C>G;c.951 C>T)that may be associated with severeβ-KTD.展开更多

Then, the correlations between LDBT and other agronomic traits of rice were analyzed. Two QTLs con- trolling LDBT were mapped on chromosome 6 and 8, respectively, qLDBT6 was mapped between RM3 and RM3628 markers on chromosome 6 with a LOD value of 3.23, contribution rate of 6.18% and additive effect of 17.74%. qLDBT8 was mapped between RM1376-RM4085 markers on chromosome 8 with a LOD value 2.95, contribution rate of 8.00% and additive effect of 20.09%. This domain probably had a relationship with the sterility of rice. The LDBT of RILs had extremely signifi- cant positive correlations with flag leaf width, 1 000-grain weight, grain width, grain length and grain thickness, but no significant correlations with other agronomic traits, such as plant height, heading date, tiller number, flag leaf length, average and the maximum temperature during heading period or rice.展开更多
In recent years,there has been a significant acceleration in the thinning,calving and retreat of the Pine Island Ice Shelf(PIIS).The basal channels,results of enhanced basal melting,have the potential to significantly...In recent years,there has been a significant acceleration in the thinning,calving and retreat of the Pine Island Ice Shelf(PIIS).The basal channels,results of enhanced basal melting,have the potential to significantly impact the stability of the PIIS.In this study,we used a variety of remote sensing data,including Landsat,REMA DEM,ICESat-1 and ICESat-2 satellite altimetry observations,and Ice Bridge airborne measurements,to study the spatiotemporal changes in the basal channels from 2003 to 2020 and basal melt rate from 2010 to 2017 of the PIIS under the Eulerian framework.We found that the basal channels are highly developed in the PIIS,with a total length exceeding 450 km.Most of the basal channels are ocean-sourced or groundingline-sourced basal channels,caused by the rapid melting under the ice shelf or near the groundingline.A raised seabed prevented warm water intrusion into the eastern branch of the PIIS,resulting in a lower basal melt rate in that area.In contrast,a deepsea trough facilitates warm seawater into the mainstream and the western branch of the PIIS,resulting in a higher basal melt rate in the main-stream,and the surface elevation changes above the basal channels of the mainstream and western branch are more significant.The El Ni?o event in 2015–2016 possibly slowed down the basal melting of the PIIS by modulating wind field,surface sea temperature and depth seawater temperature.Ocean and atmospheric changes were driven by El Ni?o,which can further explain and confirm the changes in the basal melting of the PIIS.展开更多
Objective:Triple-negative breast cancer(estrogen receptor-negative,progesterone receptor-negative and Her2-negative) can be classified into two subtypes:basal and non-basal phenotype.And the basal phenotype is associa...Objective:Triple-negative breast cancer(estrogen receptor-negative,progesterone receptor-negative and Her2-negative) can be classified into two subtypes:basal and non-basal phenotype.And the basal phenotype is associated with poor outcome.The purpose of this study was to figure out the differences of clinicopathological characters and related factors of prognosis between these two subtypes.Methods:Immunohistochemical staining was performed for the CK5/6,CK17 basal markers and EGFR on biopsy samples from 40 triple-negative patients and the clinicopathology features of these samples were investigated.Results:Seventy percent of the patients were diagnosed as the basal phenotype.Compared with the non-basal phenotype,the basal phenotype lesions were significantly larger in diameter with a high nuclear grade.In the node-negative group the basal phenotype clearly showed the same clinicopathological differences.There was statistically significant concordance among all three antibodies.Conclusion:Expression of basal markers identifies a biologically and clinically distinct subgroup of TN tumors,justifying the use of basal markers to define the basal or the non-basal phenotype.It is important to help the doctor deciding the therapeutic strategy for patient with triple-negative breast cancer.展开更多

However, wheat requires less N fertilizer at seedling growth stage, and its basal N fertilizer utilization efficiency is relatively low; therefore, reducing the N application rate at the seedling stage and postponing the N fertilization period may be effective for reducing N application and increasing wheat yield and NUE. A 4-year field experiment was conducted with two cultivars under four N rates(240 kg N ha–1(N240), 180 kg N ha–1(N180), 150 kg N ha–1(N150), and 0 kg N ha–1(N0)) and three basal N application stages(seeding(L0), fourleaf stage(L4), and six-leaf stage(L6)) to investigate the effects of reducing the basal N application rate and postponing the basal N fertilization period on grain yield, NUE, and N balance in a soil-wheat system. There was no significant difference in grain yield between the N180 L4 and N240 L0(control) treatments, and the maximum N recovery efficiency and N agronomy efficiency were observed in the N180 L4 treatment. Grain yield and NUE were the highest in the L4 treatment. The leaf area index, flag leaf photosynthesis rate, flag leaf nitrate reductase and glutamine synthase activities, dry matter accumulation, and N uptake post-jointing under N180 L4 did not differ significantly from those under N240 L0. Reduced N application decreased the inorganic N content in the 0–60-cm soil layer, and the inorganic N content of the L6 treatment was higher than those of the L0 and L4 treatments at the same N level. Surplus N was low under the reduced N rates and delayed basal N application treatments. Therefore, postponing and reducing basal N fertilization could maintain a high yield and improve NUE by improving the photosynthetic production capacity, promoting N uptake and assimilation, and reducing surplus N in soil-wheat systems.展开更多

Pot trials and cylinder trials were carried out from 2002 to 2005 to study the influences of single basal application of 3 controlled-release fertilizers on the changes of soil available N, root development, senescence and lodging resistance at late growth stages. Results showed that at 30 days after fertilization, single basal application of controlled-release fertilizers coated with vegetal-substance (CRF1) and polymer materials (CRF3) increased soil available N to 12.0 and 147.9%, respectively, in comparison to split fertilization of rice-specific fertilizer (RSF1). Treatments of the two CRFs obviously benefited the development of root system, resulting in greater rice root weights with extensive distribution and higher root activity. In addition, the two CRF treatments, in comparison to RSF1, enhanced chlorophyll consents of the flag leaves to 9.5 and 15.5%, and soluble protein up to 89.7 and 108.0% respectively. Application of the two CRFs also made the base of rice stems strong and large, declined the proportion of shoot and root, increased root depth index. Though relatively low K rate, single basal application of the CRF3 coated with NH4MgPO4 could also promote the development of root system, enhance root activity and some physiological functions of flag leaves. Based on these results, it was concluded that major mechanisms for increasing rice yield by single basal application of the CRFs should be attributed to grater soil available N supply, superior development of root systems, better nutrient absorption capacity, slower senescence and enhancement of lodging resistance at late stages.展开更多
The purpose of this study is to characterize the re-epithelialization of wound healing in canine prostatic urethra and to evaluate the effect of this re-epithelialization way after two-micron laser resection of the pr...The purpose of this study is to characterize the re-epithelialization of wound healing in canine prostatic urethra and to evaluate the effect of this re-epithelialization way after two-micron laser resection of the prostate (TmLRP). TmLRP and partial bladder neck mucosa were performed in 15 healthy adult male crossbred canines. Wound specimens were harvested at 3 days, and 1, 2, 3, and 4 weeks after operation, respectively. The histopathologic characteristics were observed by hematoxylin and eosin staining. The expression of cytokeratin 14 (CK14), CK5, CK18, synaptophysin (Syn), chromogranin A (CgA), uroplakin, transforming growth factor-β1 (TGF-β1), and TGF-β type Ⅱ receptor in prostatic urethra wound were examined by immunohistochemistry and real-time polymerase chain reaction, respectively. Van Gieson staining was performed to determine the expression of collagen fibers in prostatic urethra and bladder neck would. The results showed that the re-epithelialization of the prostatic urethra resulted from the mobilization of proliferating epithelial cells from residual prostate tissue under the wound. The proliferating cells expressed CK14, CK5, but not CK18, Syn, and CgA and re-epithelialize expressed uroplakin since 3 weeks. There were enhanced TGF-β1 and TGF-β type Ⅱ receptor expression in proliferating cells and regenerated cells, which correlated with specific phases of re-epithelialization. Compared with the re-epithelialization of the bladder neck, re-epithelialization of canine prostatic urethra was faster, and the expression of collagen fibers was relatively low. In conclusion, re-epithelialization in canine prostatic urethra resulted from prostate basal cells after TmLRP and this re-epithelialization way may represent the ideal healing method from anatomic repair to functional recovery after injury.展开更多
文摘目的探究肌层浸润性膀胱癌(muscle-invasive bladder cancer,MIBC)分子亚型特异性,为不同MIBC分子亚型患者的治疗提供指导。方法基于MIBC分子分型方法中的UNC分型法,应用转录组测序数据,将48例MIBC患者分为2种分子亚型即Basal型和Luminal型,并进行差异表达分析以探究MIBC特异性的lncRNAs,并进一步探讨其分子特征和临床意义。结合单细胞质谱流式细胞术(single-cell mass cytometry,CyTOF)和镜像质谱流式细胞术(imaging mass cytometry,IMC)分析MBNL1-AS1高表达组和低表达组Basal型MIBC患者的免疫微环境异质性。结果基于分子分型法筛选发现了在MIBC特异性表达的lncRNA MBNL1-AS1,其低表达与Basal型MIBC患者的不良预后密切相关(P=0.022)。且进一步分析转录组测序数据后发现,在Basal型MIBC中,MBNL1-AS1低表达组具有去分化(P=0.008)、高干性(P=0.020)和高增殖(P=0.010)的特征。MBNL1-AS1低表达组的Basal型MIBC患者的免疫评分与NK CD56bright细胞和Treg细胞的评分呈负相关(P<0.05),而MBNL1-AS1高表达组的B细胞和CD8+T细胞相关基因的表达水平较高。高维度单细胞蛋白质组学分析结果显示,MBNL1-AS1低表达的Basal型MIBC患者表现出较高的Treg细胞亚群丰度(P=0.016)。结论MBNL1-AS1低表达组的Basal型MIBC患者具有去分化、高干性、高增殖和免疫抑制的特点。MBNL1-AS1具有作为Basal型MIBC免疫响应生物标志物的潜能。

文摘Food is a critical environmental factor that influences animal survival,especially for small passerines due to their high mass-specific metabolic rates.Basal metabolic rate(BMR)reflects the energy expended by endothermic animals for basic physiological processes and constitutes a major part of their daily energy budget.Some birds have been shown to employ compensatory mechanisms during food shortages,temporarily reducing these selfmaintenance expenditures without using hypothermia.However,the mechanisms of BMR adjustment remain unexplored.In the present study,we assessed the phenotypic variation in basal thermogenesis of Eurasian Tree Sparrows(Passer montanus)by comparing a control group to groups fasted for 6,12,18,and 24 h.We focused on the correlation between a reduction in energy metabolism and the alterations of cellular metabolic activities,mitochondrial substrate supply,and changes in serum thyroid hormones during fasting.Our data indicated that fasting groups had significantly lower body mass,BMR,body temperature,and body fat content.Furthermore,fasting groups had significantly lower glycogen levels,mitochondrial state 4 respiration and cytochrome c oxidase(CCO)activity in the liver,and CCO activity in pectoral muscle.The levels of avian uncoupling protein(avUCP)m RNA were significantly reduced,while the levels of myostatin protein in pectoral muscle were significantly increased in the fasting groups.Furthermore,the groups subjected to fasting exhibited significantly lower levels of serum glucose,triglyceride,thyroxine(T_(4)),and triiodothyronine(T_(3)).Positive correlations were observed between the following pairs of variables:log BMR and log body mass,log body mass and log body fat,log BMR and log state 4 respiration in the liver,log BMR and log CCO activity in the liver and muscle,log BMR and log av-UCP m RNA expression,whereas a negative correlation was observed between log BMR and log myostatin level.In addition,a positive correlation was also detected between log T_(3) and each of the following:log BMR,state 4 respiration,and log CCO activity in the liver.Our results suggested that decreased metabolic thermogenesis via down-regulation in cellular aerobic capacity of organs and serum thyroid hormones may be an important survival strategy for fasting Tree Sparrows to reduce energy expenditure.

文摘Models that predict a forest stand’s evolution are essential for developing plans for sustainable management.A simple mathematical framework was developed that con-siders the individual tree and stand basal area under random resource competition and is based on two assumptions:(1)a sigmoid-type stochastic process governs tree and stand basal area dynamics of living and dying trees,and(2)the total area that a tree may potentially occupy determines the number of trees per hectare.The most effective method to satisfy these requirements is formalizing each tree diameter and potentially occupied area using Gompertz-type stochastic differential equations governed by fixed and mixed-effect parameters.Data from permanent experimental plots from long-term Lithuania experiments were used to construct the tree and stand basal area models.The new models were relatively unbiased for live trees of all species,including silver birch(Betula pen-dula Roth)and downy birch(Betula pubescens Ehrh.),[spruce(Picea abies),and pine(Pinus sylvestris)].Less reliable predic-tions were made for the basal area of dying trees.Pines gave the highest accuracy prediction of mean basal area among all live trees.The mean basal area prediction for all dying trees was lower than that for live trees.Among all species,pine also had the best average basal area prediction accuracy for live trees.Newly developed basal area growth and yield models can be recommended despite their complex formulation and implementation challenges,particularly in situations when data is scarce.This is because the newly observed plot provides sufficient information to calibrate random effects.

文摘BACKGROUND Basal cell carcinoma(BCC)is the most prevalent skin cancer,characterized by indolent growth and low metastatic rates.When metastatic BCC(mBCC)does occur,it most commonly involves lymph nodes,lungs,and bones,with metastases to other sites being exceptionally rare.This case reports the first documented instance of mBCC to the stomach,highlighting the importance of considering atypical metastatic sites,and the challenges associated with diagnosis and management.CASE SUMMARY A 52-year-old male with recurrent BCC and known mBCC to the bone presented with progressive dysphagia,cranial neuropathies and generalized weakness.He had been treated with immune checkpoint inhibitors,with intermittent therapy modifications due to treatment related toxicities.His past history was notable for malignant perineural invasion,radiotherapy for osseous metastases,immune checkpoint inhibitor-induced enteritis,and osteonecrosis of the jaw.Gastroscopy revealed subepithelial gastric lesions,and biopsies confirmed mBCC-a previously unreported site of disease dissemination.A multidisciplinary team involving gastroenterology,oncology,neurology,and palliative care guided his management.Given his declining functional status and poor prognosis,he was ultimately transitioned to hospice care.CONCLUSION Clinicians should consider atypical metastatic sites in advanced BCC.A multidisciplinary approach remains essential for timely diagnosis and coordinated management.

文摘Objective:To analyze the therapeutic effect and prognosis of frameless stereotactic soft channel intracranial hematoma evacuation for severe basal ganglia hemorrhage.Methods:Clinical data of 411 patients with severe basal ganglia hemorrhage admitted to the Neurological Intensive Care Unit of Linyi People’s Hospital from January 2020 to December 2021 were collected.According to the modified Rankin Scale(mRS)score at 180 days after onset,the patients were divided into the good prognosis group and the poor prognosis group.The therapeutic effect of frameless stereotactic soft channel intracranial hematoma evacuation on severe basal ganglia hemorrhage was explored,and the influencing factors of prognosis were analyzed.Results:Multivariate Logistic regression analysis showed that the admission Glasgow Coma Scale(GCS)score was an independent protective factor for the prognosis of patients with severe basal ganglia hemorrhage,while age,preoperative hematoma volume,random blood glucose level,and mechanical ventilation were independent risk factors.Conclusion:Frameless stereotactic soft channel intracranial hematoma evacuation has a good therapeutic effect on severe basal ganglia hemorrhage.However,it is necessary to screen the patients’basic information before surgery and provide medical care based on their specific conditions to promote their rapid recovery.
文摘The basal ganglia(BG) act as a cohesive functional unit that regulates motor function,habit formation,and reward/addictive behaviors. However,it is still not well understood how the BG maintains wakefulness and suppresses sleep to achieve al these fundamental functions until genetical y engineered systems developed these years. Significant research efforts have recently been directed at developing genetic-molecular tools to achieve reversible and cell-type specific in vivo silencing or activation of neurons in behaving animals. Optogenetic tools can be used both to specifically activate or inhibit neurons of interest and identify functional synaptic connectivity between specific neuronal populations,both in vivo and in brain slices. Another recently developed system by Roth and colleagues permits the selective and ″remote″ manipulation(activation and silencing) of neuronal activity via all 3 major GPCR signaling pathways(G_i,G_s and G_q). These so-called ″ designer receptors exclusively activated by designer drugs″(DREADD) involve mutant GPCRs that do not respond to their endogenous ligands but are responsive to otherwise inert biological compounds. Recently,we demonstrated the essential roles and the neural pathways of the neurons expressing adenosine A_(2A) receptors or dopamine D_1 receptors in the BG for sleep-wake regulation using the genetically engineered systems including optogenetics and DREADD. We proposed a plausible model in which the caudate-putamen and the nucleus accumbens integrates behavioral processes with sleep/wakefulness through adenosine and dopamine receptors.
文摘BACKGROUNDβ-ketothiolase deficiency(β-KTD)is an inherited disease,and insufficient attention has been paid to imageology due to its lower morbidity.Therefore,few lesions outside the basal ganglia have been found before,and the persistent pathological changes have rarely been reported.CASE SUMMARY A 10-mo-old Chinese female patient with a free previous medical history but with poor physical and athletic development had received the haemophilus influenzae vaccine and then developed a low fever 2 d prior.She was initially diagnosed with severe brain injury,central respiratory failure,metabolic acidosis complicated with respiratory alkalosis,hyper-IgE,etc.With further examination,a definite diagnosis ofβ-KTD was made.Symptomatic treatment was adopted.Ten days later,the dyspnea was improved evidently and the ventilator was removed,but there were still obvious abnormalities on magnetic resonance imaging(MRI).The lesions mainly invaded the corpus striatum but were not limited to the basal ganglia.Then,the patient’s disease improved and discharged approximately 1 mo later,and the abnormal lesions on MRI had partially improved.However,for about 1 year,the residual irreversible lesions were observed on MRI,the mental and physical development of the patient was obviously regressive,and extra rehabilitation training was needed.CONCLUSION The case highlights the critical importance of one view that the range of lesions in some patients may be more extensive than previously thought in someβ-KTD patients.In addition to biochemical tests,genetic tests and magnetic resonance imaging are not only conducive to quickly diagnosingβ-KTD but also to partially evaluating the short-and long-term outcomes.Moreover,more attention should be paid to the two mutations(c.478 C>G;c.951 C>T)that may be associated with severeβ-KTD.
文摘A population of 240 recombinant inbred lines (RIL) was derived from a cross between Japonica rice cultivar TD70 and Indica cultivar Kasalath, and their linkage map including 141 SSR markers was used to locate the QTLs controlling the length of dehiscence at the basal part of thecae (LDBT). Then, the correlations between LDBT and other agronomic traits of rice were analyzed. Two QTLs con- trolling LDBT were mapped on chromosome 6 and 8, respectively, qLDBT6 was mapped between RM3 and RM3628 markers on chromosome 6 with a LOD value of 3.23, contribution rate of 6.18% and additive effect of 17.74%. qLDBT8 was mapped between RM1376-RM4085 markers on chromosome 8 with a LOD value 2.95, contribution rate of 8.00% and additive effect of 20.09%. This domain probably had a relationship with the sterility of rice. The LDBT of RILs had extremely signifi- cant positive correlations with flag leaf width, 1 000-grain weight, grain width, grain length and grain thickness, but no significant correlations with other agronomic traits, such as plant height, heading date, tiller number, flag leaf length, average and the maximum temperature during heading period or rice.

文摘In recent years,there has been a significant acceleration in the thinning,calving and retreat of the Pine Island Ice Shelf(PIIS).The basal channels,results of enhanced basal melting,have the potential to significantly impact the stability of the PIIS.In this study,we used a variety of remote sensing data,including Landsat,REMA DEM,ICESat-1 and ICESat-2 satellite altimetry observations,and Ice Bridge airborne measurements,to study the spatiotemporal changes in the basal channels from 2003 to 2020 and basal melt rate from 2010 to 2017 of the PIIS under the Eulerian framework.We found that the basal channels are highly developed in the PIIS,with a total length exceeding 450 km.Most of the basal channels are ocean-sourced or groundingline-sourced basal channels,caused by the rapid melting under the ice shelf or near the groundingline.A raised seabed prevented warm water intrusion into the eastern branch of the PIIS,resulting in a lower basal melt rate in that area.In contrast,a deepsea trough facilitates warm seawater into the mainstream and the western branch of the PIIS,resulting in a higher basal melt rate in the main-stream,and the surface elevation changes above the basal channels of the mainstream and western branch are more significant.The El Ni?o event in 2015–2016 possibly slowed down the basal melting of the PIIS by modulating wind field,surface sea temperature and depth seawater temperature.Ocean and atmospheric changes were driven by El Ni?o,which can further explain and confirm the changes in the basal melting of the PIIS.

文摘Objective:Triple-negative breast cancer(estrogen receptor-negative,progesterone receptor-negative and Her2-negative) can be classified into two subtypes:basal and non-basal phenotype.And the basal phenotype is associated with poor outcome.The purpose of this study was to figure out the differences of clinicopathological characters and related factors of prognosis between these two subtypes.Methods:Immunohistochemical staining was performed for the CK5/6,CK17 basal markers and EGFR on biopsy samples from 40 triple-negative patients and the clinicopathology features of these samples were investigated.Results:Seventy percent of the patients were diagnosed as the basal phenotype.Compared with the non-basal phenotype,the basal phenotype lesions were significantly larger in diameter with a high nuclear grade.In the node-negative group the basal phenotype clearly showed the same clinicopathological differences.There was statistically significant concordance among all three antibodies.Conclusion:Expression of basal markers identifies a biologically and clinically distinct subgroup of TN tumors,justifying the use of basal markers to define the basal or the non-basal phenotype.It is important to help the doctor deciding the therapeutic strategy for patient with triple-negative breast cancer.
文摘Basal cell carcinoma is the most common skin cancer mainly caused by prolonged exposure to ultraviolet rays. It is also known as rodent ulcer or basal cell epithelioma. The main mechanism suggested is prolonged exposure to high intensity ultraviolet rays, which causes DNA damage. Pigmented basal cell carcinoma is a rare variety of basal cell carcinoma. Usually, it presents as pigmented nodular mass over the nose or malar region. Other differential diagnoses of this mass, are malignant melanoma and seborrheic keratosis. Treatment of choice is surgical excision with 2 mm of margins.

文摘Excessive nitrogen(N) fertilization with a high basal N ratio in wheat can result in lower N use efficiency(NUE) and has led to environmental problems in the Yangtze River Basin, China. However, wheat requires less N fertilizer at seedling growth stage, and its basal N fertilizer utilization efficiency is relatively low; therefore, reducing the N application rate at the seedling stage and postponing the N fertilization period may be effective for reducing N application and increasing wheat yield and NUE. A 4-year field experiment was conducted with two cultivars under four N rates(240 kg N ha–1(N240), 180 kg N ha–1(N180), 150 kg N ha–1(N150), and 0 kg N ha–1(N0)) and three basal N application stages(seeding(L0), fourleaf stage(L4), and six-leaf stage(L6)) to investigate the effects of reducing the basal N application rate and postponing the basal N fertilization period on grain yield, NUE, and N balance in a soil-wheat system. There was no significant difference in grain yield between the N180 L4 and N240 L0(control) treatments, and the maximum N recovery efficiency and N agronomy efficiency were observed in the N180 L4 treatment. Grain yield and NUE were the highest in the L4 treatment. The leaf area index, flag leaf photosynthesis rate, flag leaf nitrate reductase and glutamine synthase activities, dry matter accumulation, and N uptake post-jointing under N180 L4 did not differ significantly from those under N240 L0. Reduced N application decreased the inorganic N content in the 0–60-cm soil layer, and the inorganic N content of the L6 treatment was higher than those of the L0 and L4 treatments at the same N level. Surplus N was low under the reduced N rates and delayed basal N application treatments. Therefore, postponing and reducing basal N fertilization could maintain a high yield and improve NUE by improving the photosynthetic production capacity, promoting N uptake and assimilation, and reducing surplus N in soil-wheat systems.
文摘This paper was to explore the mechanism of single basal application of controlled-release fertilizers for increasing yield of rice (Oryza sativa L.). Pot trials and cylinder trials were carried out from 2002 to 2005 to study the influences of single basal application of 3 controlled-release fertilizers on the changes of soil available N, root development, senescence and lodging resistance at late growth stages. Results showed that at 30 days after fertilization, single basal application of controlled-release fertilizers coated with vegetal-substance (CRF1) and polymer materials (CRF3) increased soil available N to 12.0 and 147.9%, respectively, in comparison to split fertilization of rice-specific fertilizer (RSF1). Treatments of the two CRFs obviously benefited the development of root system, resulting in greater rice root weights with extensive distribution and higher root activity. In addition, the two CRF treatments, in comparison to RSF1, enhanced chlorophyll consents of the flag leaves to 9.5 and 15.5%, and soluble protein up to 89.7 and 108.0% respectively. Application of the two CRFs also made the base of rice stems strong and large, declined the proportion of shoot and root, increased root depth index. Though relatively low K rate, single basal application of the CRF3 coated with NH4MgPO4 could also promote the development of root system, enhance root activity and some physiological functions of flag leaves. Based on these results, it was concluded that major mechanisms for increasing rice yield by single basal application of the CRFs should be attributed to grater soil available N supply, superior development of root systems, better nutrient absorption capacity, slower senescence and enhancement of lodging resistance at late stages.
文摘The purpose of this study is to characterize the re-epithelialization of wound healing in canine prostatic urethra and to evaluate the effect of this re-epithelialization way after two-micron laser resection of the prostate (TmLRP). TmLRP and partial bladder neck mucosa were performed in 15 healthy adult male crossbred canines. Wound specimens were harvested at 3 days, and 1, 2, 3, and 4 weeks after operation, respectively. The histopathologic characteristics were observed by hematoxylin and eosin staining. The expression of cytokeratin 14 (CK14), CK5, CK18, synaptophysin (Syn), chromogranin A (CgA), uroplakin, transforming growth factor-β1 (TGF-β1), and TGF-β type Ⅱ receptor in prostatic urethra wound were examined by immunohistochemistry and real-time polymerase chain reaction, respectively. Van Gieson staining was performed to determine the expression of collagen fibers in prostatic urethra and bladder neck would. The results showed that the re-epithelialization of the prostatic urethra resulted from the mobilization of proliferating epithelial cells from residual prostate tissue under the wound. The proliferating cells expressed CK14, CK5, but not CK18, Syn, and CgA and re-epithelialize expressed uroplakin since 3 weeks. There were enhanced TGF-β1 and TGF-β type Ⅱ receptor expression in proliferating cells and regenerated cells, which correlated with specific phases of re-epithelialization. Compared with the re-epithelialization of the bladder neck, re-epithelialization of canine prostatic urethra was faster, and the expression of collagen fibers was relatively low. In conclusion, re-epithelialization in canine prostatic urethra resulted from prostate basal cells after TmLRP and this re-epithelialization way may represent the ideal healing method from anatomic repair to functional recovery after injury.
