Serum was collected after 7 d of treatment. The healthy group included 20 individuals. Gastric mucosal epithelial cell line GES-1 was cultured in vitro and randomly divided into the following seven groups: control, model, healthy,Western Medicine, prior treatment, low dosage, and high dosage. After 72 h of treatment with the corresponding serum, the m RNA and protein expression levels of TGF-β1, Smad3, and Smad7 were measured by reverse transcription quantitative polymerase chain reaction and western blotting, respectively.RESULTS: The m RNA expression levels of TGF-β1 and Smad3 in GES-1 cells were increased after Hp introduction, and these increased levels were reduced by the BXD-containing serum. The protein levels of p-Smad3, but not TGF-β1 or Smad3, were significantly increased in Hp-treated GES-1 cells, and treatment with the BXD-containing serum markedly decreased the protein levels. Smad7 expression was significantly enhanced following treatment with the BXD-containing serum at transcriptional and protein levels in a dose-dependent manner.CONCLUSION: BXD regulates the TGF-β/Smad signaling pathway by inhibiting the expression of TGF-β1 and Smad3, and increasing the expression of Smad7.展开更多

Methods: Twenty-four Sprague Dawley (SD) rats were randomly divided into three groups. Rats in the treatment group received GXBD (13 g crude drug/kg) for three weeks, while rats in the model control and normal control groups received equal volumes of distilled water. On the 22nd day, rats in the ischemia-reperfusion (I/R) control and GXBD-treated groups underwent 30 min occlusion of the left anterior descending (LAD) coronary artery, followed by 120 min reperfusion. Electrocardiogram was recorded, and the activities of cardiac enzymes, cytokines, and NF-кB were assessed after I/R. Results: Compared with the I/R control group, GXBD treatment restored the activity of the specific myocardial-injury marker creatine kinase (CK) and lactate dehydrogenase (LDH), and inhibited the inflammatory response involving the nuclear factor-кB (NF-кB) pathway, including down-regulation of interleukin (IL)-1β and IL-6, and up-regulation of IL-10 gene expression. Conclusion: GXBD strongly reduced myocardial impairment in our I/R model, including inhibition of NF-кB activation and inflammatory cytokine responses.展开更多

Methods:Mice were treated with different proportions of the P and C drug pair (1:1, 3:1, and 4:1) for 10 days, and subsequentlyinjected with atropine (ATR) or neostigmine (NEO). The effects of the different proportions of P and C were evaluatedbased on the alvine advance rate. In addition, we used the same modeling method used in the first experiment andadministered P: C at ratio of 3:1 and at different doses respectively (4.68 g/L, 2.34 g/L, and 1.17 g/L), and tested levelsof the gastrointestinal hormones, gastrin (GAS), vasoactive intestinal polypeptide (VIP), and somatostatin (SS) in thesmall intestinal tissue using an enzyme-linked immunosorbent assay. Results: In the groups of NEO-induced mice, P:Cat ratios of 1:1, 3:1, and 4:1 significantly reduced the alvine advance rate compared with the NEO model group (P =0.003, P = 0.012 and P = 0.021, respectively). In the groups of ATR-induced mice, only P:1 at ratio of 3:1 significantlyincreased the alvine advance rate compared with the ATR model group (P = 0.007). After exposure to P: C at ratio of 3:1and at different dose, the GAS level was lower in the low-, medium-, and high-dose NEO groups than that in the NEOmodel group (P = 0.001, P = 0.004, and P = 0.003, respectively). The VIP levels were higher in the medium-andhigh-dose NEO groups than that in the NEO model group (P = 0.004 and P = 0.002, respectively). In addition, the SSlevel increased in the NEO medium-dose group compared with that in the NEO model group (P = 0.002). The GAS levelwas higher in the ATR medium- and high-dose groups than in the ATR model group (P = 0.007 and P = 0.021,respectively). The VIP level was lower in the ATR low-, medium-, and high-dose than that in the ATR model group (P =0.001, P = 0.001, and P = 0.001, respectively). Furthermore, the SS level was lower in the ATR medium- and high-dosegroups than that in the ATR model group (P = 0.001 and P = 0.006). Conclusion: The PC drug pair bidirectionallyadjusted the NEO- and ATR-induced functional dyspepsia in mice by modulating GAS, VIP, and SS levels in theintestine.展开更多

文摘Background:The development of gastric cancer(GC)encompasses precancerous conditions like chronic atrophic gastritis(CAG)and premalignant lesions of gastric cancer(PLGC).In these situations, abnormal Notch signaling results in mucosal impairment and the initiation of cancer.Banxia Xiexin Decoction(BXD),a well-known formula in traditional Chinese medicine(TCM),shows promise in treating gastric disorders, but its mechanisms in gastric restoration remain unclear. Methods:Using MNNG-induced CAG and PLGC rat models,BXD was administered for 12 weeks.Gastric mucosal pathology was assessed via hematoxylin-eosin staining.Proliferation(Ki-67)and angiogenesis(VEGFA)markers were evaluated by immunohistochemistry.Network pharmacology identified BXD’s targets and pathways.Notch pathway components(Notch1,Jagged1,Dll4,Hes1)were analyzed via qPCR,Western blot, and immunohistochemistry. Results:BXD significantly ameliorated mucosal atrophy, glandular structural disorder, and dysplasia in CAG and PLGC rats.Network pharmacology revealed 323 overlapping targets between BXD and PLGC,with Notch signaling as a central pathway.BXD downregulated Notch1,Jagged1,Dll4,and Hes1 expression at transcriptional and protein levels, suppressed Ki-67(proliferation)and VEGFA(angiogenesis)overexpression, and restored gastric mucosal integrity. Conclusion:BXD inhibits Notch signaling, reduces aberrant proliferation and angiogenesis, and interrupts Correa’s gastric carcinogenesis cascade. This study provides mechanistic evidence supporting BXD as a TCM-based intervention for gastric precancerous lesions.

文摘Background:Liver cancer(LC)remains a leading cause of cancer-related mortality worldwide, with current treatments often limited by suboptimal efficacy and adverse effects.Banxia Houpu Decoction(BHD),a traditional Chinese herbal formula, has demonstrated potential anti-tumor properties in clinical practice. However, its precise mechanisms against LC remain unclear. This study employs network pharmacology(NP)and molecular docking(MD)approaches to systematically identify BHD’s active components and their molecular targets, aiming to elucidate its anti-LC mechanisms and provide a scientific basis for further investigation. Methods:We utilized Liquid Chromatography-Mass Spectrometry alongside the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)to identify the constituents of BHD.We identified potential targets through the utilization of TCMSP,SwissTargetPrediction,Comparative Toxicogenomics Database, and SuperPred Database.Targets linked to LC were obtained from GeneCards,OMIM,the Therapeutic Target Database, and DrugBank.A Venn diagram illustrated the intersection between component and disease targets, while a protein-protein interaction(PPI)network was developed utilizing Cytoscape 3.9.1.Primary targets were discerned through the analysis of centrality metrics, including“Degree,”“Betweenness,”and“Closeness.”The study encompassed analyses of Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathways to clarify the biological roles and pathways associated with these proteins. The essential interactions between the active constituents of BHD and the principal LC targets were investigated through MD using AutoDock software. Results:We identified 34 active components in BHD.The PPI network revealed 212 interaction targets relevant to drug-disease correlations, emphasizing key proteins including Epidermal Growth Factor Receptor(EGFR),Signal Transducer and Activator of Transcription 3(STAT3),Steroid Receptor Coactivator(SRC),Heat Shock Protein 90 Alpha Family Class A Member 1(HSP90AA1),and AKT Serine/Threonine Protein Kinase 1(AKT1).The GO analysis revealed a total of 443 biological processes,94 cellular components, and 182 molecular functions. The KEGG analysis revealed a total of 169 pathways that are involved. The results from MD revealed that the majority of binding energies fell below−7 kcal/mol,indicating strong interactions between the active compounds and their target proteins. Conclusion:Evidence suggests that BHD effectively manages LC through a synergistic mechanism encompassing various components(Magnolol,Chrysoeriol,Cerevisterol,etc.),targets(EGFR,STAT3,SRC,HSP90AA1,AKT1,etc.),and pathways(PI3K-Akt,FoxO,and Ras signaling pathways,etc.).This analysis offers a comprehensive theoretical framework for further investigative and clinical exploration.

文摘OBJECTIVE: To investigate the effect of Banxia Xiexin decoction(BXD) on Helicobacter pylori(Hp)-related peptic ulcers(PUs) and the possible mecha-nism underlying BXD actions via the transforming growth factor-β/small mothers against decapentaplegic(TGF-β/Smad) signaling pathway.METHODS: PU patients with cold-heat complex syndrome were randomly assigned to groups that received Chinese or Western medicines with 20 patients in each group. Smad7 expression was significantly enhanced following treatment with the BXD-containing serum at transcriptional and protein levels in a dose-dependent manner.CONCLUSION: BXD regulates the TGF-β/Smad signaling pathway by inhibiting the expression of TGF-β1 and Smad3, and increasing the expression of Smad7.

文摘Objective: To evaluate the regulatory effects of Banxia (Pinellia ternata, P) and Huanglian (Coptis chinensis, C) drugpair, derived from Banxiaxiexin soup, on the gastrointestinal movement of mice with functional dyspepsia. Conclusion: The PC drug pair bidirectionallyadjusted the NEO- and ATR-induced functional dyspepsia in mice by modulating GAS, VIP, and SS levels in theintestine.
