The fruits of Paulownia catalpifolia Gong Tong are used as a Chinese folk herbal medicine for the treatment of enteritis, tonsillitis, bronchitis, and dysentery, etc. Our previous study has identified new C-geranylated flavanones with obvious anti-proliferative effects in lung cancer A549 cells. In the present study, a new C-geranylated flavone, paucatalinone C(1) and five known C-geranylated flavanones(2-6) were isolated. In addition, a total of 34 C-geranylated flavonoids were detected by HPLC-DAD-ESI-MS/MS coupling techniques from the CH_2Cl_2 extract of P. catalpifolia. Futhermore, anti-aging effects of isolated compounds were evaluated in vitro with premature senescent 2BS cells induced by H_2O_2. Phytochemical results indicated that P. catalpifolia was a natural resource of abundant C-geranylated flavonoids. Diplacone(3) and paucatalinone A(5) were the potent anti-aging agents in the premature senescent 2BS cells induced by H_2O_2 and the C-geranyl substituent may be an important factor because of its lipophilic character. 展开更多

Aging and age-related ailments have emerged as critical challenges and great burdens within the global contemporary society.Addressing these concerns is an imperative task,with the aims of postponing the aging process and finding effective treatments for age-related degenerative diseases.Recent investigations have highlighted the significant roles of nicotinamide adenine dinucleotide(NAD+)in the realm of anti-aging.It has been empirically evidenced that supplementation with nicotinamide mononucleotide(NMN)can elevate NAD+levels in the body,thereby ameliorating certain age-related degenerative diseases.The principal anti-aging mechanisms of NMN essentially lie in its impact on cellular energy metabolism,inhibition of cell apoptosis,modulation of immune function,and preservation of genomic stability,which collectively contribute to the deferral of the aging process.This paper critically reviews and evaluates existing research on the anti-aging mechanisms of NMN,elucidates the inherent limitations of current research,and proposes novel avenues for anti-aging investigations. 展开更多

Objective:To explore the anti-aging effects of chlorogenic acid(CGA)and the underlying mechanisms based on a Caenorhabditis elegans(C.elegans)model.Methods:The anti-agingactivityofCGAwasstudied basedon thebodylength,exercisebehavior,lipofuscin content,antioxidative stress ability,swallowing frequency,body-bending frequency,and head-swinging ability of C.elegans.Through DAF-16 nuclear translocation and SOD-3-GFP fluorescence experiments,the effects of CGA on ROS levels,antioxidant enzyme activities,MDA content,mutant-strain lifespan,and anti-aging molecular signaling pathways were explored,as well as the underlying mechanisms.Results:CGA improved multiple indices of the nematode:body length was increased(all P<0.001),head-swing frequency and body-bending frequency were increased(all P<0.05),nematode longevity was prolonged(P=0.0021),lipofuscin deposition in nematodes was slowed down(all P<0.001),the chemotaxis index was improved(P=0.0012),ROS levels were reduced(all P<.001),and SOD activity and MDA content were reduced(SOD:P=0.0017 between the low-concentration group and the control group,P<.001 between the high-concentration and medium-concentration groups and the control group;MDA:P=0.0135 between the low-concentration group and the control group,and P<0.001 between the high-concentration and medium-concentration groups and the control group).In addition,CGA also activated the DAF-16 transcription factor,promoted DAF-16 nuclear translocation under oxidative stress conditions(both P<0.001 between the high-concentration and medium-concentration groups and the control group),and increased SOD-3 gene expression in nematodes(all P<0.001).Conclusion:CGA plays an anti-aging role in C.elegans.The underlying mechanisms include activation of the insulin/IGF-1 signaling pathway and enhancement of DAF-16 activity.This study lays a foundation for further research into the anti-aging effects of CGA. 展开更多

To evaluate the potential anti-aging ability of Eucommia ulmoides,four characteristic components(chlorogenic acid,geniposidic acid,aucubin,quercetin)were selected to assess their effects on H_(2)O_(2)-induced oxidative damage model of human umbilical vein endothelial cell(HUVEC).Oxidative damage indexes,inflammatory factors,cell cycle,cell apoptosis,cell senescence,and their related proteins were analyzed by methyl thiazolyl tetrazolium(MTT)assay,enzyme-linked immunosorbent assay(ELISA),propidium iodide(PI)staining,annexin V-FITC/PI double staining,SAβ-galactosidase staining,and western blotting(WB).The results showed that H_(2)O_(2)-induced cell growth inhibition rate decreased as supplementation with characteristic components when compared to H_(2)O_(2) group.Meanwhile,the contents of antioxidant indexes(reactive oxygen species,lactate dehydrogenase,molondialdehyde,superoxide dismutase,glutathione),inflammatory factors(nuclear factor kappa-B,intercellular cell adhesion molecule-1,vascular cell adhesion protein 1),and functional factors(NO,Endothelin-1)in characteristic components treated groups improved if comparison with H_(2)O_(2) group,suggesting the characteristic components of E.ulmoides could alleviate H_(2)O_(2)-induced oxidative damage.Moreover,cell cycle,cell apoptosis,cell senescence,and their related proteins under characteristic components treatment exhibited a better effect than under H_(2)O_(2) treatment,implying the characteristic components could participate in anti-aging via multiple pathways.These results manifested that the characteristic components of E.ulmoides posses the capacity of anti-aging,which provided a basis for investigating the anti-aging ability of E.ulmoides itself. 展开更多

Background: The time-related decline in regenerative capacity and organ homeostasis is a major feature of aging. Rehmannia glutinosa and Astragalus membranaceus have been used as traditional Chinese herbal medicines for enhanced immunity and prolonged life. However, the mechanism by which this herbal medicine slows aging is unknown. In this study, we investigated the mechanism of the herbal anti-aging effect.Methods: Mice were fed diets supplemented with R. glutinosa or A. membranaceus for 10 months; the control group was fed a standard diet. The phenotypes were evaluated using a grading score system and survival analysis. The percentages of the senescence phenotypes of hematopoietic stem cells(HSCs) were determined by fluorescence-activated cell sorting analysis. The function and the mechanism of HSCs were analyzed by clonogenic assay and the real-time polymerase chain reaction.Results: The anti-aging effect of R. glutinosa is due to the enhanced function of HSCs. Mice fed with R. glutinosa displayed characteristics of a slowed aging process,including decreased senescence and increased rate of survival. Flow cytometry analysis showed decreased numbers of Lin–Sca1^+c-kit–(LSK) cells, long-term HSCs(LT-HSCs) and short-term HSCs(ST-HSCs) in the R. glutinosa group. In vitro, clonogenic assays showed increased self-renewal ability of LT-HSCs from the R. glutinosa group as well as maintaining LSK quiescence through upregulated p18 expression. The R. glutinosa group also showed decreased reactive oxygen species levels and the percentage of β-gal^+ cells through downregulation of the cellular senescence-associated protein p53 and p16.Conclusion: Rehmannia glutinosa exerts anti-aging effects by maintaining the quiescence and decreasing the senescence of HSCs. 展开更多

OBJECTIVE Behavior research and urinary metabolomics method were applied to evaluate the anti-aging effects of Scutellaria baicalensis Georgi extract(SBG)in D-galactose-induced rats.METHODS Fifty rats were randomly divided into five groups(n=10in each group).Group 1served as vehicle control with injection of saline(vehicle control group),and the other groups of rats received daily subcutaneously injected with D-galactose(aged model group)at dose of 100mg·kg-1 for ten weeks,respectively.At the same time,rats in groups 3-5were intragastrically administered SBG 〔extracted twice with 60%(V/V)ethanol〕at doses of 50,100 and 200mg·kg-1 for ten weeks,and the rats of groups 1 and 2 were administrated an equal volume of the vehicle.At the tenth week,the learning and memory abilities were examined by Morris water maze.The urine was collected using metabolic cages and analyzed by high-resolution 1HNMR spectroscopy combined with multivariate statistical analyses.Principal component analysis(PCA)was utilized to classify and reveal the differences between the model group and control group.Then,the concentration of these differences was analyzed with t-test to determine whether SBG was possible to influence the metabolic pattern induced by D-galactose.RESULTS Compared with the vehicle control group,the D-galactose-treated aged model group markedly spent longer time(P<0.05)in finding the platform on days 3-5 in the spatial learning acquisition training of Morris water maze test.However,the escape latency was significantly reduced(P<0.05)by long-term administration of SBG(50,100 and 200mg·kg-1)compared with the D-galactose-treated aged model group on days 3-5.In the probe test,the D-galactose-treated aged model group made fewer(P<0.05)platform crossings and distance travelled in target quadrant(P<0.05)than the vehicle control group,and the SBG at doses of 50,100 and 200mg·kg-1 treatments groups could significantly increase(P<0.05)the number of times of crossing over the platform site.The SBG at doses of100 and 200mg·kg-1 treatments groups could significantly increase(P<0.05)the distance travelled in target quadrant compared with the D-galactose-treated aged model group.In addition,the significant difference in metabolic profiling was observed from model group compared with drug-dose group by using PCA,indicating the recovery effect of SBG on D-galactose induced aging rats.Some significantly changed metabolites like glycine,glucose and hexadecanoic acid have been identified.These biochemical changes are related to the the disturbance in aimno acid metabolism,energy metabolism and glycometabolism,which are helpful to further understanding the D-galactose induced aging rats and the therapeutic mechanism of SBG.CONCLUSION These results demonstrate that SBG extract has protective effect on the D-galactose-induced aging in rats. 展开更多

The anti-aging effects of a cream containing L-carnosine and ginseng root extract to provide a reference for the evaluation and development of anti-aging cosmetics were tested. Sixty healthy females, aged 35-60,were grouped into test sample and placebo groups. Skin elasticity, gloss, and wrinkles of the participants were analyzed before use and after use for 8 weeks. Instrument measurement results showed that after 8 weeks,skin elasticity, gloss, and wrinkles in the members of the test sample group obviously improved relative to those of the placebo group. Clinical effect assessment results showed that subjects did not experience adverse skin reactions, and their skin gloss showed obvious improvement. Self-assessment results showed that 87%subjects are satisfied with the test sample. Cream containing L-carnosine and ginseng root extract has a certain anti-aging effect. 展开更多

Aging is a complex biological process characterized by nine hallmarks,including genomic instability,mitochondrial dysfunction,and chronic inflammation,which collectively drive the progression of age-related chronic non-communicable diseases.Phytonutrients,a class of bioactive secondary metabolites abundant in plants,have emerged as a promising research focus for intervening in the aging process due to their multifaceted biological activities.This review systematically elaborates on the molecular mechanisms,key signaling pathways,specifically SIRT1,Nrf2/ARE,and AMPK/mTOR,and the synergistic anti-aging effects of four typical phytonutrient categories:polyphenols(e.g.,resveratrol,quercetin),carotenoids(e.g.,lycopene,astaxanthin),sulfur compounds(e.g.,α-lipoic acid,ergothioneine),and phytoestrogens(e.g.,soybean isoflavones).The evidence indicates that these compounds combat aging through a multidimensional network involving direct antioxidant actions,free radical scavenging,metal chelation,promotion of autophagy,and modulation of inflammatory and epigenetic pathways.Crucially,the review highlights that synergistic interactions between different phytonutrients can significantly enhance their efficacy beyond the effect of any single compound.The aim is to consolidate the anti-aging evidence of phytonutrients and address the current translational challenges,such as bioavailability and a lack of robust human trials,thereby providing a comprehensive theoretical framework for developing effective,diet-centered strategies to promote healthy aging and reduce the global burden of non-communicable diseases. 展开更多

Population aging is one of the common challenges in the current world.As people age,the body’s tissues including cells,and molecules inevitably degrade,and their functions gradually decline,causing various age-related diseases like Alzheimer’s disease,osteoporosis,low immunity,glucose and lipid metabolism disorders,and cardiovascular diseases.With the continuous increase of the elderly population,the pressure on the medical industry is increasing.To lower the burden on the medical industry and increase the average age of the elderly,it is vital to explore effective anti-aging materials.Ginseng Radix et Rhizoma(Renshen),as a traditional and precious Chinese medicinal herb,is known as the“king of all herbs”.It is famous for its effects of“tonifying Qi,restoring pulse”(helping with the generation of Qi(the fundamental,vital energy that continuously flows within the body)and the circulation of blood)and strengthening the body,nourishing the spleen and lungs,generating fluids and nourishing blood,calming the mind and improving intelligence.Recently,its anti-aging effect has received increasing attention from modern scientific research.This study summarizes the pharmacological effects of the main active ingredients of Renshen(ginsenosides,polysaccharides,etc.)on resisting aging,including preventing neuroaging,suppressing skin aging,mitigating ovarian aging,inhibiting osteoporosis and arthritis,enhancing the immune system of the elderly,protecting the cardiovascular system,resisting aging-induced fatigue and exerting the anti-tumor effects.Through network pharmacology and molecular docking,the anti-aging active ingredients of Renshen were screened,and the key targets and pathways of anti-aging active ingredients in Renshen were determined.Using network pharmacology,totally 106 drug targets and 3,479 disease targets were screened,and 79 common targets between aging and Renshen were identified.Three core targets were identified in the PPI network,including TNF,AKT1,and IL-1β.Molecular docking was used to obtain further verification.This study emphasizes the potential of Renshen as a source of anti-aging activity,which can be developed into a novel drug for the treatment of age-related diseases. 展开更多

Sulfonylureas are widely used oral anti-diabetic drugs.However,its long-term usage effects on patients’lifespan remain controversial,with no reports of influence on animal longevity.Hence,the anti-aging effects of chlorpropamide along with glimepiride,glibenclamide,and tolbutamide were studied with special emphasis on the interaction of chlorpropamide with mitochondrial ATP-sensitive K+(mito K-ATP)channels and mitochondrial complex II.Chlorpropamide delayed aging in Caenorhabditis elegans,human lung fibroblast MRC-5 cells and reduced doxorubicin-induced senescence in both MRC-5 cells and mice.In addition,the mitochondrial membrane potential and ATP levels were significantly increased in chlorpropamide-treated worms,which is consistent with the function of its reported targets,mito K-ATP channels.Increased levels of mitochondrial reactive oxygen species(mt ROS)were observed in chlorpropamide-treated worms.Moreover,the lifespan extension by chlorpropamide required complex II and increased mt ROS levels,indicating that chlorpropamide acts on complex II directly or indirectly via mito K-ATP to increase the production of mt ROS as a pro-longevity signal.This study provides mechanistic insight into the anti-aging effects of sulfonylureas in C.elegans. 展开更多

Dear Editor,The moderate level of reactive oxygen species(ROS)contributes to cellular functions such as proliferation,differentiation,and infection resistance,but the excessive level of ROS causes oxidative damage which underlies the basic mechanism for aging and geriatric diseases.1 Therefore,precisely managing cellular ROS levels,meaning to keep redox homeostasis properly,stands as an aim for health and longevity.Actually,this aim is challenging,as it asks not only to restrain excessive ROS accumulation at the right time and right place but also to guarantee proper ROS levels for fitting physiological requirements.Traditional ways for eliminating ROS put more attention on the efficiency and identified a variety of drugs and enzymes with antioxidant properties,known as enzymatic and non-enzymatic ROS scavengers,respectively.However,how to accurately manage the integrative effect of ROS scavengers in vivo remains a problem.Actually,the effectiveness of non-enzymatic antioxidants in vivo is controversial,explained mostly by the uncertainty to reach a given organ with the appropriate concentration or to work there persistently.2 As to enzymatic ROS scavengers,although their overexpression in a given place can be achieved,this mode is often unwieldy or harmful to intrinsic physiologic functions in cases.3 For this reason,we attempt to develop new ways for in vivo antioxidation,for establishing a high-quality balance between efficiency and safety.As the consequence,we innovated a gene expression system that can limit the excessive ROS accumulation but keep the physiological level of ROS. 展开更多

The effects of styrene-butadiene-styrene(SBS)pre-swelling/extraction process and the incorporation of C9 petroleum resin on the anti-aging performance of modified asphalt were systematically evaluated by characterizing the physical indexes,chemical compositions and rheological parameters.The experimental results show that the SBS pre-swelling/extraction process and the incorporation of C9 petroleum resin improve the dispersion performance of SBS in asphalt as well as the strength of SBS polymer network structures,and the synergistic effects decrease the volatilization degree of asphalt lightweight components and the degradation rate of SBS during the aging process.The anti-aging performance of SBS modified asphalt(SBSMA)was significantly enhanced by SBS pre-swelling/extraction process compounded with the incorporation of C9 petroleum resin,and the anti-aging effect was gradually enhanced with the increase of C9 petroleum resin content. 展开更多

Electronically Conductive Metal−Organic Framework With Photoelectric and Photothermal Effect as a Stable Cathode for High-Temperature Photo-Assisted Zn/Sn-Air Battery

Rechargeable Zn/Sn-air batteries have received considerable attention as promising energy storage devices.However,the electrochemical performance of these batteries is significantly constrained by the sluggish electrocatalytic reaction kinetics at the cathode.The integration of light energy into Zn/Sn-air batteries is a promising strategy for enhancing their performance.However,the photothermal and photoelectric effects generate heat in the battery under prolonged solar irradiation,leading to air cathode instability.This paper presents the first design and synthesis of Ni_(2)-1,5-diamino-4,8-dihydroxyanthraquinone(Ni_(2)DDA),an electronically conductiveπ-d conjugated metal-organic framework(MOF).Ni_(2)DDA exhibits both photoelectric and photothermal effects,with an optical band gap of~1.14 eV.Under illumination,Ni_(2)DDA achieves excellent oxygen evolution reaction performance(with an overpotential of 245 mV vs.reversible hydrogen electrode at 10 mA cm^(−2))and photothermal stability.These properties result from the synergy between the photoelectric and photothermal effects of Ni_(2)DDA.Upon integration into Zn/Sn-air batteries,Ni_(2)DDA ensures excellent cycling stability under light and exhibits remarkable performance in high-temperature environments up to 80℃.This study experimentally confirms the stable operation of photo-assisted Zn/Sn-air batteries under high-temperature conditions for the first time and provides novel insights into the application of electronically conductive MOFs in photoelectrocatalysis and photothermal catalysis. 展开更多

Beam-tracking simulations have been extensively utilized in the study of collective beam instabilities in circular accelerators.Traditionally,many simulation codes have relied on central processing unit(CPU)-based methods,tracking on a single CPU core,or parallelizing the computation across multiple cores via the message passing interface(MPI).Although these approaches work well for single-bunch tracking,scaling them to multiple bunches significantly increases the computational load,which often necessitates the use of a dedicated multi-CPU cluster.To address this challenge,alternative methods leveraging General-Purpose computing on Graphics Processing Units(GPGPU)have been proposed,enabling tracking studies on a standalone desktop personal computer(PC).However,frequent CPU-GPU interactions,including data transfers and synchronization operations during tracking,can introduce communication overheads,potentially reducing the overall effectiveness of GPU-based computations.In this study,we propose a novel approach that eliminates this overhead by performing the entire tracking simulation process exclusively on the GPU,thereby enabling the simultaneous processing of all bunches and their macro-particles.Specifically,we introduce MBTRACK2-CUDA,a Compute Unified Device Architecture(CUDA)ported version of MBTRACK2,which facilitates efficient tracking of single-and multi-bunch collective effects by leveraging the full GPU-resident computation. 展开更多

Synergistic antibacterial effect and mechanism of benzalkonium chloride and polymyxin B against Pseudomonas aeruginosa

Benzalkonium chloride(BAC)is widely employed as a broad-spectrum biocide and has emerged as a significant environmental pollutant.Polymyxin B(PB)serves as the last-line defense for the treatment of Gram-negative pathogens.Previous studies reported that BAC-adapted Pseudomonas aeruginosa increased the tolerance to PB.Herein,we present the novel finding that the combination of BAC and PB exhibited synergistic antibacterial effects against P.aeruginosa.Time-killing assay demonstrated a significant reduction in bacterial cell viability.Scanning electron microscopy,zeta potential analysis,hydrophobicity measurements,and fluorescence probe analyses collectively revealed severe disruption of the cell envelope and membrane potential induced by the combination of BAC and PB.Transcriptomic analysis revealed that the BAC-PB combination notably downreg-ulated the expression of genes involved in lipid A modification and cell envelope production,including phoPQ,pmrAB,bamABCDE,lptABCDEG,lolB,yidC,and murJ.Additionally,the combination group exhibited augmented production of reactive oxygen species and diminished ATP synthesis.The expression of the genes associated with substance metabolism and energy generation was significantly impeded.This study provides significant implica-tions for the interactions of biocides and antibiotics on Gram-negative pathogens,while also addressing antibiotic resistance and developing the external treatment strategy for Pseudomonas-infected wounds and burns. 展开更多

AIM:To evaluate and compare alterations in the effective lens position(ELP)and refractive outcomes among three distinct intraocular lens(IOL)types.METHODS:Patients with cataracts were enrolled and allocated to 3 groups:Group A(implanted with the SN6CWS),Group B(implanted with the MI60),and Group C(implanted with the Aspira-aA).ELP measurements were obtained with swept-source optical coherence tomography(SS-OCT)at 1d,1wk,1mo,and 3mo postoperatively.Subjective refraction assessments were conducted at 1wk,1mo,and 3mo following surgery.RESULTS:The study included 189 eyes of 150 cataract patients(66 males).There were 77 eyes in Group A,55 eyes in Group B,and 57 eyes in Group C.The root mean square of the ELP(ELPRMS)within the initial 3mo was significantly lower for Group A than for Groups B and C.Refractive changes within Group A were not significant across the time points of 1wk,1mo,and 3mo.Conversely,both Group B and Group C demonstrated statistically significant shifts toward hyperopia from 1wk to 3mo postsurgery.CONCLUSION:Among the three IOLs examined,the SN6CWS IOL showes the greatest stability during the first 3mo postoperatively.Between 1wk and 3mo after surgery,notable hyperopic shifts are evident in eyes implanted with the MI60 and Aspira-aA IOLs,whereas refractive outcomes remain relatively constant in eyes implanted with SN6CWS IOLs. 展开更多
