Tumor necrosis factor alpha-induced protein 3:A key biomarker for response to ursodeoxycholic acid in primary biliary cholangitis

BACKGROUND The pathogenesis of primary biliary cholangitis(PBC)remains unclear.Ursodeoxycholic acid(UDCA)is the only first-line clinical treatment,but approximately 40%of patients exhibit a poor response.AIM To identify novel biomarkers for PBC to predict the efficacy of UDCA and enhance treatment.METHODS Microarray expression profiling datasets were downloaded from the Gene Expression Omnibus and analyzed to identify differentially expressed genes between PBC patients and healthy controls.Immunohistochemistry was performed to validate key genes in liver tissues of the participants.Logistic regression was employed to evaluate prognostic risk factors,receiver operating characteristic curves were used to assess predictive performance,and correlations between key genes and clinicopathological characteristics were analyzed.RESULTS By bioinformatic analysis,13 genes primarily associated with the progression of PBC were identified,and tumor necrosis factor alpha-induced protein 3(TNFAIP3)was selected for further investigation.Then expression of TNFAIP3 in PBC patients was significantly elevated compared to healthy controls on immunohistochemistry(P<0.0001).Multivariate Cox regression analysis indicated that both TNFAIP3 and fatigue were independent risk factors for response to UDCA in PBC patients(P<0.05).The area under the curve for TNFAIP3 and fatigue were 0.691 and 0.704,respectively,while their combination showed a significantly higher area under the curve of 0.848.The expression of TNFAIP3 was also correlated with age,albumin,total bilirubin,alkaline phosphatase and splenomegaly(P<0.05).CONCLUSION TNFAIP3 and fatigue are independent risk factors for response to UDCA in Chinese patients with PBC.TNFAIP3 may be a potential biomarker or therapeutic target for PBC.These findings offer new insights into the pathogenesis of PBC. 展开更多

The prediction of nuclear cross-section data is crucial, especially in the absence of experimental data or in the difficulty of these experimental data. Nickel(Ni) is an important material in fusion and fission reactor technologies, the production of radionuclides in nuclear medicine,and many other fields. In this study, the excitation functions for^(60,62) Ni(a,n),^(60,61) Ni(a,2 n),^(58,64) Ni(a,p), and ^(nat)Ni(a,x) reactions have been investigated by using preequilibrium reaction models. The calculations of the excitation functions for the reactions are used with the geometry-dependent hybrid model in ALICE/ASH code and the two-component exciton model in TALYS 1.8 code. The obtained results are compared to each other, and the experimental data are taken from the EXFOR database. 展开更多

The molten salt reactor(MSR) has received much recent attention. The presence of beryllium and the mixing of actinides with light nuclei in the fuel salt result in a relatively strong neutron source that can affect the surveillance at subcritical and transient characteristics during operation. In this study, we predict the inherent neutron sources based on a MSR model. The calculation shows that in the fresh core, the inherent neutron sources are mainly from alpha-induced neutrons. After power operation, the inherent neutron sources become remarkably stronger due to photoneutrons. Although being an insignificant part in the total neutron population during operation, the inherent neutron sources can be used as the installed neutron source after shutdown. If the MSR has continuously operated at full power(2 MWt) for 10 days,then there would be no need for the installed source within80 days after shutdown. After operating constantly for500 days, the installed neutron source can be eliminated within 2 years after shutdown. 展开更多

Objective: The results of a previous study showed that a clear dysregulation was evident in the global gene expression of the BCL11A-suppressed B-lymphoma cells. In this study, the bone morphogenetic protein receptor, type II(BMPR2), E1 A binding protein p300(EP300), transforming growth factor-β2(TGFβ2), and tumor necrosis factor, and alpha-induced protein 3(TNFAIP3) gene expression patterns in B-cell malignancies were studied. Methods: The relative expression levels of BMPR2, EP300, TGFβ2, and TNFAIP3 mRNA in B-lymphoma cell lines, myeloid cell lines, as well as in cells from healthy volunteers, were determined by real-time quantitative reverse transcriptpolymerase chain reaction(qRT-PCR) with SYBR Green Dye. Glyceraldehyde-3-phosphate dehydrogenase(GAPDH) was used as reference. Results: The expression level of TGFβ2 mRNA in B-lymphoma cell lines was significantly higher than those in the cells from the healthy control(P<0.05). However, the expression level of TNFAIP3 mRNA in B-malignant cells was significantly lower than that of the healthy control(P<0.05). The expression levels of BMPR2 and EP300 mRNA showed no significant difference between B-malignant cell lines and the healthy group(P>0.05). In B-lymphoma cell lines, correlation analyses revealed that the expression of BMPR2 and TNFAIP3(r=0.882, P=0.04) had significant positive relation. The expression levels of BMPR2, EP300, and TNFAIP3 mRNA in cell lines from myeloid leukemia were significantly lower than those in the cells from the healthy control(P<0.05). The expression levels of TGFβ2 mRNA showed no significant difference between myeloid leukemia cell lines and the healthy control or B-malignant cell lines(P>0.05). The expression levels of BMPR2, EP300, and TNFAIP3 mRNA in B-lymphoma cells were significantly higher than those of the myeloid leukemia cells(P<0.05).Conclusion: Different expression patterns of BMPR2, EP300, TGFβ2, and TNFAIP3 genes in B-lymphoma cells exist. 展开更多

Objective: To study the correlation of the abnormal expression of hexokinase-Ⅱ (HK-Ⅱ) and tumor necrosis factor alpha-induced protein 3 (TNFAIP3) in diffuse large B-cell lymphoma (DLBCL) with the malignant features of tumor cells. Methods: DLBCL patients who underwent surgical resection in our hospital between March 2016 and March 2018 were selected as the DLBCL group, and the patients who underwent surgical resection during the same period and were pathologically confirmed as reactive hyperplasia of lymph nodes were selected as the control group. The lesions were collected to measure the expression levels of HK-Ⅱ, TNFAIP3, proliferation genes and invasion genes. Results: HK-Ⅱ mRNA expression level in the lesions of the DLBCL group was significantly higher than that of the control group while TNFAIP3 mRNA expression level was significantly lower than that of the control group;HK-Ⅱ mRNA expression levels in the lesions of DLBCL group of patients with lymphoma stage ⅡI-IV and lymphoma group B were significantly higher than those of the patients with lymphoma stage I-Ⅱ and group A while TNFAIP3 mRNA expression levels were significantly lower than those of the patients with lymphoma stage I-Ⅱ and lymphoma group A;CyclinD2, PDE4B, BCL2, XIAP, CCL5, CXCR4 and MMP26 mRNA expression levels in the lesions of the DLBCL group were significantly higher than those of the control group, positively correlated with HK-Ⅱ and negatively correlated with TNFAIP3 while Caspase-3 and TIMP4 mRNA expression levels were significantly lower than those of the control group, negatively correlated with HK-Ⅱ and positively correlated with TNFAIP3. Conclusion: The high expression of HK-Ⅱ and the low expression of TNFAIP3 in DLBCL are closely related to the pathological process of tumor as well as the proliferation and invasion of tumor cells. 展开更多

Methods: A total of 68 patients who were diagnosed with diffuse large B cell lymphoma in Guangzhou 421 Hospital of PLA between August 2014 and July 2017 were selected as the DLBCL group of the study, and 34 patients who accepted lymph node biopsy due to lymphadenectasis and were diagnosed with reactive lymphoid hyperplasias in Guangzhou 421 Hospital of PLA during the same period were selected as the control group. The expression levels of TNFAIP3, PDE4B, apoptosis genes and invasion genes in lymph node tissue were determined. Results: TNFAIP3, PTEN, PTPL1 and Bax protein expression in lesion tissue of DLBCL group were significantly lower than those of control group whereas PDE4B, c-myc, AURKA, AURKB, PLK1, Ets-1, β-catenin, MMP7, MMP9 and CD44 protein expression were significantly higher than those of control group;PTEN, PTPL1 and Bax protein expression in DLBCL lesions were positively correlated with TNFAIP3 protein expression and negatively correlated with PDE4B protein expression;c-myc, AURKA, AURKB, PLK1, Ets-1, β-catenin, MMP7, MMP9 and CD44 protein expression were negatively correlated with TNFAIP3 protein expression and positively correlated with PDE4B protein expression. Conclusion: The low expression of TNFAIP3 and the high expression of PDE4B in diffuse large B cell lymphoma can antagonize tumor cell apoptosis and promote tumor cell invasion. 展开更多
