Objective Magnetoencephalography(MEG),a non-invasive neuroimaging technique,meticulously captures the magnetic fields emanating from brain electrical activity.Compared with MEG based on superconducting quantum interfe...Objective Magnetoencephalography(MEG),a non-invasive neuroimaging technique,meticulously captures the magnetic fields emanating from brain electrical activity.Compared with MEG based on superconducting quantum interference devices(SQUID),MEG based on optically pump magnetometer(OPM)has the advantages of higher sensitivity,better spatial resolution and lower cost.However,most of the current studies are clinical studies,and there is a lack of animal studies on MEG based on OPM technology.Pain,a multifaceted sensory and emotional phenomenon,induces intricate alterations in brain activity,exhibiting notable sex differences.Despite clinical revelations of pain-related neuronal activity through MEG,specific properties remain elusive,and comprehensive laboratory studies on pain-associated brain activity alterations are lacking.The aim of this study was to investigate the effects of inflammatory pain(induced by Complete Freund’s Adjuvant(CFA))on brain activity in a rat model using the MEG technique,to analysis changes in brain activity during pain perception,and to explore sex differences in pain-related MEG signaling.Methods This study utilized adult male and female Sprague-Dawley rats.Inflammatory pain was induced via intraplantar injection of CFA(100μl,50%in saline)in the left hind paw,with control groups receiving saline.Pain behavior was assessed using von Frey filaments at baseline and 1 h post-injection.For MEG recording,anesthetized rats had an OPM positioned on their head within a magnetic shield,undergoing two 15-minute sessions:a 5-minute baseline followed by a 10-minute mechanical stimulation phase.Data analysis included artifact removal and time-frequency analysis of spontaneous brain activity using accumulated spectrograms,generating spectrograms focused on the 4-30 Hz frequency range.Results MEG recordings in anesthetized rats during resting states and hind paw mechanical stimulation were compared,before and after saline/CFA injections.Mechanical stimulation elevated alpha activity in both male and female rats pre-and post-saline/CFA injections.Saline/CFA injections augmented average power in both sexes compared to pre-injection states.Remarkably,female rats exhibited higher average spectral power 1 h after CFA injection than after saline injection during resting states.Furthermore,despite comparable pain thresholds measured by classical pain behavioral tests post-CFA treatment,female rats displayed higher average power than males in the resting state after CFA injection.Conclusion These results imply an enhanced perception of inflammatory pain in female rats compared to their male counterparts.Our study exhibits sex differences in alpha activities following CFA injection,highlighting heightened brain alpha activity in female rats during acute inflammatory pain in the resting state.Our study provides a method for OPM-based MEG recordings to be used to study brain activity in anaesthetized animals.In addition,the findings of this study contribute to a deeper understanding of pain-related neural activity and pain sex differences.展开更多
Soil microbial communities are key factors in maintaining ecosystem multifunctionality(EMF).However,the distribution patterns of bacterial diversity and how the different bacterial taxa and their diversity dimensions ...Soil microbial communities are key factors in maintaining ecosystem multifunctionality(EMF).However,the distribution patterns of bacterial diversity and how the different bacterial taxa and their diversity dimensions affect EMF remain largely unknown.Here,we investigated variation in three measures of diversity(alpha diversity,community composition and network complexity)among rare,intermediate,and abundant taxa across a latitudinal gradient spanning five forest plots in Yunnan Province,China and examined their contributions on EMF.We aimed to characterize the diversity distributions of bacterial groups across latitudes and to assess the differences in the mechanisms underlying their contributions to EMF.We found that multifaceted diversity(i.e.,diversity assessed by the three different metrics)of rare,intermediate,and abundant bacteria generally decreased with increasing latitude.More importantly,we found that rare bacterial taxa tended to be more diverse,but they contributed less to EMF than intermediate or abundant bacteria.Among the three dimensions of diversity we assessed,only community composition significantly affected EMF across all locations,while alpha diversity had a negative effect,and network complexity showed no significant impact.Our study further emphasizes the importance of intermediate and abundant bacterial taxa as well as community composition to EMF and provides a theoretical basis for investigating the mechanisms by which belowground microorganisms drive EMF along a latitudinal gradient.展开更多

Tianxiangdan(TXD),a traditional Chinese herbal remedy,demonstrates efficacy in mitigating myocardial ischemia-reperfusion(I/R)-induced damage.This study employed network pharmacology to evaluate the therapeutic target...Tianxiangdan(TXD),a traditional Chinese herbal remedy,demonstrates efficacy in mitigating myocardial ischemia-reperfusion(I/R)-induced damage.This study employed network pharmacology to evaluate the therapeutic targets and mechanisms of TXD in treating I/R.Highperformance liquid chromatography-mass spectrometry(HPLC-MS)identified 86 compounds in TXD.Network pharmacological analysis predicted potential target genes and their modes of action.Cardiac function,ischaemic ST changes,lactate dehydrogenase(LDH),malondialdehyde(MDA),superoxide dismutase(SOD)activity,myocardial fiber,and infarct size were assessed using in vivo and in vitro I/R injury models.Estrogen receptor alpha(ERα)protein expression and estradiol(E2)levels were measured to confirm TXD's impact on estrogen levels and ERαexpression.To examine if TXD reduces I/R injury through ERα,an AZD group(300 nmol·L^(-1)AZD9496 and 15%TXD serum)was compared to a TXD group(15%TXD serum).The study hypothesized that TXD upregulates the ERα-mediated iron metamorphosis pathway.I/R injury-induced ferroptosis was identified using a Fer-1 group(1.0μmol·L^(-1)Fer-1 and 15%TXD serum)to elucidate the potential association between ferroptosis and ERαproteins.A DCFH-DA probe detected reactive oxygen species(ROS)and Fe^(2+),while Western blotting assessed target protein expression.Both in vitro and in vivo experiments demonstrated that TXD attenuated I/R injury by reducing elevated ST-segment levels,improving cardiac injury biomarkers(LDH,MDA,and SOD),alleviating pathological features,and preventing I/R-induced loss of cell viability in vitro.The effects and mechanisms of TXD on I/R injury-associated ferroptosis were investigated using I/R-induced H9c2 cells.The TXD group showed significantly decreased ROS and Fe^(2+)levels,while the AZ group(treated with AZD9496)exhibited increased levels.The TXD group demonstrated enhanced expression of ERαand glutathione peroxidase 4(GPX4),with reduced levels of P53 protein and ferritinheavy polypeptide 1(FTH1).The AZ group exhibited contrasting effects on these expression levels.The literature indicated a novel connection between ERαand ferroptosis.TXD activates the ERαsignaling pathway,promoting protection against I/R-induced myocardial cell ferroptosis.This study provides evidence supporting TXD use for myocardial ischemia treatment,particularly in older female patients who may benefit from its therapeutic outcomes.展开更多
AIM:To investigate how angles kappa and alpha affect postoperative visual quality in patients with multifocal intraocular lens(mIOLs)implantation.METHODS:Retrospective cases series.A total of 46 patients(46 eyes)who u...AIM:To investigate how angles kappa and alpha affect postoperative visual quality in patients with multifocal intraocular lens(mIOLs)implantation.METHODS:Retrospective cases series.A total of 46 patients(46 eyes)who underwent phacoemulsification were subsumed.The correlation between Preoperative angles kappa and alpha,wave-front aberrations and objective visual quality of cornea,internal,and total eye after surgery were analyzed using iTrace.RESULTS:The magnitude of angle kappa was negatively correlated with internal and total modulation transfer function(MTF)at 3 mm;the magnitude of angle kappa was positively correlated with astigmatism,trefoil,higher-order aberrations(HOAs)of both internal and total eye at 3 mm.The magnitude of angle alpha was negatively correlated with total MTF and total Strehl ratio at 3 mm.The magnitude of angle alpha was positively correlated with corneal coma at 5 mm,internal astigmatism at both 3 mm and 5 mm,and total spherical aberration(SA)at 3 mm.Multivariate linear regression analysis showed that,among candidate independent variables(kappa,alpha,astigmatism,SA,coma,trefoil,and HOAs),astigmatism is the only independent factor for altering corneal MTF at 3 mm and 5 mm;astigmatism and HOAs emerged as independent factors for altering internal MTF at 3 mm and 5 mm,and total MTF at 3 mm;astigmatism,SA and HOAs emerged as independent factors for altering total MTF at 5 mm.CONCLUSION:With greater preoperative angle kappa or angle alpha,patients who accept mIOL implantation tend to have larger internal astigmatism and HOAs,which resulting in poor visual quality,especially those with small pupil size.展开更多
Today,noise control is of critical importance.In order to provide sound insulation,parameters such as Sound Transmission Loss(STL)and sound absorption coefficient are measured in environments such as impedance tubes,A...Today,noise control is of critical importance.In order to provide sound insulation,parameters such as Sound Transmission Loss(STL)and sound absorption coefficient are measured in environments such as impedance tubes,Alpha Cabin and echo chambers.However,the low number of accredited acoustic test rooms in Turkey and the high-test costs cause these tests to be performed in limited numbers.In this direction,test box similar to the Alpha Cabin designed aims to both reduce costs and perform tests in a healthy way using natural and recyclable materials,and to prevent damage to test devices caused by hard materials.In this study,samples with STL values above 30 dB at 500–8000 Hz.were selected and tested in the designed system.As a result,it was seen that the data were close to each other.The highest value was obtained as 49.13 dB at 4000 Hz.in a 2 cm thick gypsum board,gypsum and concave walnut shell sample(moving surface L_(1)).This situation provides an important contribution in terms of sound insulation by using natural and recyclable materials and the proposed test box,meeting the experimental criteria at low cost and in the field of noise control.展开更多
BACKGROUND Hepatocellular carcinoma(HCC)is a global health concern,representing the second most common cause of malignancy-related mortality in the world.The primary cause of HCC in the United States is chronic infect...BACKGROUND Hepatocellular carcinoma(HCC)is a global health concern,representing the second most common cause of malignancy-related mortality in the world.The primary cause of HCC in the United States is chronic infection with the hepatitis C virus(HCV).Clinical observations have established sex-based differences in HCV infection with the disease progressing more severely and more rapidly in males and postmenopausal females compared to premenopausal females,suggesting that estrogens and their receptors may play an important role in hepatic defenses and development of HCV-mediated HCC.However,the precise mechanism of estrogen protection and their effects on inflammation is poorly understood.AIM To determine whether estrogen receptor(ER)expression is correlated with the expression of tumor necrosis factor-alpha(TNF-α)in males and females with HCV-associated diseases.METHODS The role of ERs in modulating innate immune responses was investigated using human liver tissues with HCV/cirrhosis and HCV/HCC.Messenger RNA(mRNA)and protein(nuclear and cytoplasmic)expression were measured for all markers of interest and compared to normal human liver tissue samples.RESULTS ERβwas reported for the first time to have a greater mRNA expression than ERαin normal liver(P≤0.001).In addition,ERβmRNA expression was found to be decreased in diseased livers(P≤0.05),while TNF-αexpression was increased(P≤0.0001).Upon stratifying by sex within each disease group,ESR1 was found to be negatively correlated with ESR2 in females with HCV/cirrhosis(r=-0.84,P≤0.001),whereas males with HCV/cirrhosis were found to have a significant positive correlation(r=0.57,P≤0.05).ESR2 mRNA expression had a significant positive correlation with TNF-αin both HCV/cirrhosis(r=0.61,P≤0.001)and HCV/HCC patients(r=0.45,P≤0.05).CONCLUSION All together,these findings indicate that changes in ERβand TNF-αexpression are associated with worsening disease,and may be part of the sex-dependent factors in HCC pathogenesis.展开更多
AIM:To explore the effect of Alpha angle and Kappa angle before multifocal intraocular lenses(MIOLs)implantation on postoperative visual quality of patients.METHODS:Before and 3mo after cataract surgery,Alpha angle an...AIM:To explore the effect of Alpha angle and Kappa angle before multifocal intraocular lenses(MIOLs)implantation on postoperative visual quality of patients.METHODS:Before and 3mo after cataract surgery,Alpha angle and Kappa angle were collected using IOL Master 700,iTrace,and Pentacam for clinical observation.Postoperative visual quality indicators,including high-order aberrations(HOA),modulation transfer function(MTF)and point spread function(PSF),were collected using iTrace.multiple linear regression analysis was used to analyze the correlation of the Kappa angle and the Alpha angle with age,axial length(AL),anterior chamber depth(ACD),keratometry(K),lens thickness(LT)and corneal white to white distance(WTW).Pearson correlation coefficient was used to analyze the correlation between Alpha angle and Kappa angle;Bland Altman analysis was used to evaluate the consistency of pairwise detection results of three instruments.RESULTS:The Alpha angle was modeled as Alpha=2.230+0.003×age-0.036×AL-0.025×K-0.058×WTW and the Kappa angle was modeled as Kappa=0.685+0.003×age-0.013×K-0.061×WTW.The correlation between the total Alpha angle and Kappa angle of the three instruments was weakly positive(r=0.291,P=0.000).Comparing the measurement of Alpha angle and Kappa angle using three instruments,only IOL Master 700 and iTrace showed good consistency in measuring Kappa angle(P=0.4254).After 3mo of surgery,the Alpha angle and Kappa angle significantly decreased(P=0.011,0.018;P=0.008,0.036).△Kappa=1.136-0.021×AL-0.013×K.Kappa angle could positively predict HOA(β=0.18,P=0.000),MTF(β=0.171,P=0.000),PSF(β=0.088,P=0.000),Alpha angle cannot(P>0.05).CONCLUSION:The patients with older age,flatter K and shorter WTW should be alert to the possibility of larger Alpha angle and Kappa angle.Alpha angle should also consider the factor of AL.When selecting patients with MIOLs implantation,there is no need to consider the Alpha angle.Careful consideration should be given to the Kappa angle,and the preoperative standard of<0.5 mm can refer to△Kappa=1.136-0.021×AL-0.013×K and be appropriately relaxed.展开更多

Peroxisome proliferator-activated receptor alpha is a member of the nuclear hormone receptor superfamily and functions as a transcription factor involved in regulating cellular metabolism.Previous studies have shown t...Peroxisome proliferator-activated receptor alpha is a member of the nuclear hormone receptor superfamily and functions as a transcription factor involved in regulating cellular metabolism.Previous studies have shown that PPARαplays a key role in the onset and progression of neurodegenerative diseases.Consequently,peroxisome proliferator-activated receptor alpha agonists have garnered increasing attention as potential treatments for neurological disorders.This review aims to clarify the research progress regarding peroxisome proliferator-activated receptor alpha in nervous system diseases.Peroxisome proliferator-activated receptor alpha is present in all cell types within adult mouse and adult neural tissues.Although it is conventionally believed to be primarily localized in the nucleus,its function may be regulated by a dynamic balance between cytoplasmic and nuclear shuttling.Both endogenous and exogenous peroxisome proliferator-activated receptor alpha agonists bind to the peroxisome proliferator-activated response element to exert their biological effects.Peroxisome proliferator-activated receptor alpha plays a significant therapeutic role in neurodegenerative diseases.For instance,peroxisome proliferator-activated receptor alpha agonist gemfibrozil has been shown to reduce levels of soluble and insoluble amyloid-beta in the hippocampus of Alzheimer's disease mouse models through the autophagy-lysosomal pathway.Additionally,peroxisome proliferator-activated receptor alpha is essential for the normal development and functional maintenance of the substantia nigra,and it can mitigate motor dysfunction in Parkinson's disease mouse models.Furthermore,peroxisome proliferator-activated receptor alpha has been found to reduce neuroinflammation and oxidative stress in various neurological diseases.In summary,peroxisome proliferator-activated receptor alpha plays a crucial role in the onset and progression of multiple nervous system diseases,and peroxisome proliferator-activated receptor alpha agonists hold promise as new therapeutic agents for the treatment of neurodegenerative diseases,providing new options for patient care.展开更多

The accumulation of^(222)Rn and^(220)Rn progeny in poorly ventilated environments poses the risk of natural radiation exposure to the public.A previous study indicated that satisfactory results in determining the^(222...The accumulation of^(222)Rn and^(220)Rn progeny in poorly ventilated environments poses the risk of natural radiation exposure to the public.A previous study indicated that satisfactory results in determining the^(222)Rn and^(220)Rn progeny concentrations by measuring the total alpha counts at five time intervals within 560 min should be expected only in the case of high progeny concentrations in air.To complete the measurement within a relatively short period and adapt it for simultaneous measurements at comparatively lower^(222)Rn and^(220)Rn progeny concentrations,a novel mathematical model was proposed based on the radioactive decay law.This model employs a nonlinear fitting method to distinguish nuclides with overlapping spectra by utilizing the alpha particle counts of non-overlapping spectra within consecutive measurement cycles to obtain the concentrations of^(222)Rn and^(220)Rn progeny in air.Several verification experiments were conducted using an alpha spectrometer.The experimental results demonstrate that the concentrations of^(222)Rn and^(220)Rn progeny calculated by the new method align more closely with the actual circumstances than those calculated by the total count method,and their relative uncertainties are all within±16%.Furthermore,the measurement time was reduced to 90 min,representing an acceleration of 84%.The improved capability of the new method in distinguishing alpha particles with similar energies emitted from ^(218)Po and^(212)Bi,both approximately 6 MeV,contributed to realizing more accurate results.The proposed method has the potential advantage of measuring relatively low concentrations of^(222)Rn and^(220)Rn progeny in air more quickly via air filtration.展开更多

文摘Soil microbial communities are key factors in maintaining ecosystem multifunctionality(EMF).However,the distribution patterns of bacterial diversity and how the different bacterial taxa and their diversity dimensions affect EMF remain largely unknown.Here,we investigated variation in three measures of diversity(alpha diversity,community composition and network complexity)among rare,intermediate,and abundant taxa across a latitudinal gradient spanning five forest plots in Yunnan Province,China and examined their contributions on EMF.We aimed to characterize the diversity distributions of bacterial groups across latitudes and to assess the differences in the mechanisms underlying their contributions to EMF.We found that multifaceted diversity(i.e.,diversity assessed by the three different metrics)of rare,intermediate,and abundant bacteria generally decreased with increasing latitude.More importantly,we found that rare bacterial taxa tended to be more diverse,but they contributed less to EMF than intermediate or abundant bacteria.Among the three dimensions of diversity we assessed,only community composition significantly affected EMF across all locations,while alpha diversity had a negative effect,and network complexity showed no significant impact.Our study further emphasizes the importance of intermediate and abundant bacterial taxa as well as community composition to EMF and provides a theoretical basis for investigating the mechanisms by which belowground microorganisms drive EMF along a latitudinal gradient.

文摘Hereditary alpha tryptasemia was first described in 2016 and is the most common(up to 72%)cause of elevated serum basal tryptase(TPS).The clinical presentation of this condition,which is caused by copy number gains in the TPSAB1 gene encoding serumαTPS,is variable for each patient.Some patients are asymptomatic,whereas in others,especially those with increased mast cell activation,it has been associated with a higher risk of anaphylaxis.Better characterization of this entity is important to identify atrisk patients and to develop new treatment strategies.This review provided an overview of hereditary alpha tryptasemia and increased awareness of this condition by discussing the current information in the literature.

文摘Tianxiangdan(TXD),a traditional Chinese herbal remedy,demonstrates efficacy in mitigating myocardial ischemia-reperfusion(I/R)-induced damage.This study employed network pharmacology to evaluate the therapeutic targets and mechanisms of TXD in treating I/R.Highperformance liquid chromatography-mass spectrometry(HPLC-MS)identified 86 compounds in TXD.Network pharmacological analysis predicted potential target genes and their modes of action.Cardiac function,ischaemic ST changes,lactate dehydrogenase(LDH),malondialdehyde(MDA),superoxide dismutase(SOD)activity,myocardial fiber,and infarct size were assessed using in vivo and in vitro I/R injury models.Estrogen receptor alpha(ERα)protein expression and estradiol(E2)levels were measured to confirm TXD's impact on estrogen levels and ERαexpression.To examine if TXD reduces I/R injury through ERα,an AZD group(300 nmol·L^(-1)AZD9496 and 15%TXD serum)was compared to a TXD group(15%TXD serum).The study hypothesized that TXD upregulates the ERα-mediated iron metamorphosis pathway.I/R injury-induced ferroptosis was identified using a Fer-1 group(1.0μmol·L^(-1)Fer-1 and 15%TXD serum)to elucidate the potential association between ferroptosis and ERαproteins.A DCFH-DA probe detected reactive oxygen species(ROS)and Fe^(2+),while Western blotting assessed target protein expression.Both in vitro and in vivo experiments demonstrated that TXD attenuated I/R injury by reducing elevated ST-segment levels,improving cardiac injury biomarkers(LDH,MDA,and SOD),alleviating pathological features,and preventing I/R-induced loss of cell viability in vitro.The effects and mechanisms of TXD on I/R injury-associated ferroptosis were investigated using I/R-induced H9c2 cells.The TXD group showed significantly decreased ROS and Fe^(2+)levels,while the AZ group(treated with AZD9496)exhibited increased levels.The TXD group demonstrated enhanced expression of ERαand glutathione peroxidase 4(GPX4),with reduced levels of P53 protein and ferritinheavy polypeptide 1(FTH1).The AZ group exhibited contrasting effects on these expression levels.The literature indicated a novel connection between ERαand ferroptosis.TXD activates the ERαsignaling pathway,promoting protection against I/R-induced myocardial cell ferroptosis.This study provides evidence supporting TXD use for myocardial ischemia treatment,particularly in older female patients who may benefit from its therapeutic outcomes.
文摘AIM:To investigate how angles kappa and alpha affect postoperative visual quality in patients with multifocal intraocular lens(mIOLs)implantation.METHODS:Retrospective cases series.A total of 46 patients(46 eyes)who underwent phacoemulsification were subsumed.The correlation between Preoperative angles kappa and alpha,wave-front aberrations and objective visual quality of cornea,internal,and total eye after surgery were analyzed using iTrace.RESULTS:The magnitude of angle kappa was negatively correlated with internal and total modulation transfer function(MTF)at 3 mm;the magnitude of angle kappa was positively correlated with astigmatism,trefoil,higher-order aberrations(HOAs)of both internal and total eye at 3 mm.The magnitude of angle alpha was negatively correlated with total MTF and total Strehl ratio at 3 mm.The magnitude of angle alpha was positively correlated with corneal coma at 5 mm,internal astigmatism at both 3 mm and 5 mm,and total spherical aberration(SA)at 3 mm.Multivariate linear regression analysis showed that,among candidate independent variables(kappa,alpha,astigmatism,SA,coma,trefoil,and HOAs),astigmatism is the only independent factor for altering corneal MTF at 3 mm and 5 mm;astigmatism and HOAs emerged as independent factors for altering internal MTF at 3 mm and 5 mm,and total MTF at 3 mm;astigmatism,SA and HOAs emerged as independent factors for altering total MTF at 5 mm.CONCLUSION:With greater preoperative angle kappa or angle alpha,patients who accept mIOL implantation tend to have larger internal astigmatism and HOAs,which resulting in poor visual quality,especially those with small pupil size.
文摘Today,noise control is of critical importance.In order to provide sound insulation,parameters such as Sound Transmission Loss(STL)and sound absorption coefficient are measured in environments such as impedance tubes,Alpha Cabin and echo chambers.However,the low number of accredited acoustic test rooms in Turkey and the high-test costs cause these tests to be performed in limited numbers.In this direction,test box similar to the Alpha Cabin designed aims to both reduce costs and perform tests in a healthy way using natural and recyclable materials,and to prevent damage to test devices caused by hard materials.In this study,samples with STL values above 30 dB at 500–8000 Hz.were selected and tested in the designed system.As a result,it was seen that the data were close to each other.The highest value was obtained as 49.13 dB at 4000 Hz.in a 2 cm thick gypsum board,gypsum and concave walnut shell sample(moving surface L_(1)).This situation provides an important contribution in terms of sound insulation by using natural and recyclable materials and the proposed test box,meeting the experimental criteria at low cost and in the field of noise control.

文摘BACKGROUND Hepatocellular carcinoma(HCC)is a global health concern,representing the second most common cause of malignancy-related mortality in the world.The primary cause of HCC in the United States is chronic infection with the hepatitis C virus(HCV).Clinical observations have established sex-based differences in HCV infection with the disease progressing more severely and more rapidly in males and postmenopausal females compared to premenopausal females,suggesting that estrogens and their receptors may play an important role in hepatic defenses and development of HCV-mediated HCC.However,the precise mechanism of estrogen protection and their effects on inflammation is poorly understood.AIM To determine whether estrogen receptor(ER)expression is correlated with the expression of tumor necrosis factor-alpha(TNF-α)in males and females with HCV-associated diseases.METHODS The role of ERs in modulating innate immune responses was investigated using human liver tissues with HCV/cirrhosis and HCV/HCC.Messenger RNA(mRNA)and protein(nuclear and cytoplasmic)expression were measured for all markers of interest and compared to normal human liver tissue samples.RESULTS ERβwas reported for the first time to have a greater mRNA expression than ERαin normal liver(P≤0.001).In addition,ERβmRNA expression was found to be decreased in diseased livers(P≤0.05),while TNF-αexpression was increased(P≤0.0001).Upon stratifying by sex within each disease group,ESR1 was found to be negatively correlated with ESR2 in females with HCV/cirrhosis(r=-0.84,P≤0.001),whereas males with HCV/cirrhosis were found to have a significant positive correlation(r=0.57,P≤0.05).ESR2 mRNA expression had a significant positive correlation with TNF-αin both HCV/cirrhosis(r=0.61,P≤0.001)and HCV/HCC patients(r=0.45,P≤0.05).CONCLUSION All together,these findings indicate that changes in ERβand TNF-αexpression are associated with worsening disease,and may be part of the sex-dependent factors in HCC pathogenesis.

文摘AIM:To explore the effect of Alpha angle and Kappa angle before multifocal intraocular lenses(MIOLs)implantation on postoperative visual quality of patients.METHODS:Before and 3mo after cataract surgery,Alpha angle and Kappa angle were collected using IOL Master 700,iTrace,and Pentacam for clinical observation.Postoperative visual quality indicators,including high-order aberrations(HOA),modulation transfer function(MTF)and point spread function(PSF),were collected using iTrace.multiple linear regression analysis was used to analyze the correlation of the Kappa angle and the Alpha angle with age,axial length(AL),anterior chamber depth(ACD),keratometry(K),lens thickness(LT)and corneal white to white distance(WTW).Pearson correlation coefficient was used to analyze the correlation between Alpha angle and Kappa angle;Bland Altman analysis was used to evaluate the consistency of pairwise detection results of three instruments.RESULTS:The Alpha angle was modeled as Alpha=2.230+0.003×age-0.036×AL-0.025×K-0.058×WTW and the Kappa angle was modeled as Kappa=0.685+0.003×age-0.013×K-0.061×WTW.The correlation between the total Alpha angle and Kappa angle of the three instruments was weakly positive(r=0.291,P=0.000).Comparing the measurement of Alpha angle and Kappa angle using three instruments,only IOL Master 700 and iTrace showed good consistency in measuring Kappa angle(P=0.4254).After 3mo of surgery,the Alpha angle and Kappa angle significantly decreased(P=0.011,0.018;P=0.008,0.036).△Kappa=1.136-0.021×AL-0.013×K.Kappa angle could positively predict HOA(β=0.18,P=0.000),MTF(β=0.171,P=0.000),PSF(β=0.088,P=0.000),Alpha angle cannot(P>0.05).CONCLUSION:The patients with older age,flatter K and shorter WTW should be alert to the possibility of larger Alpha angle and Kappa angle.Alpha angle should also consider the factor of AL.When selecting patients with MIOLs implantation,there is no need to consider the Alpha angle.Careful consideration should be given to the Kappa angle,and the preoperative standard of<0.5 mm can refer to△Kappa=1.136-0.021×AL-0.013×K and be appropriately relaxed.

文摘Parkinson's disease(PD)is the second most common neurodegenerative disorder.The clinical manifestations of PD include motor symptoms,such as bradykinesia,resting tremor,rigidity,and nonmotor symptoms,which include disturbances in sleep,gastrointestinal function,and olfaction.PD misdiagnosis rates have been reported to reach approximately 30%,partly owing to the heterogeneity of parkinsonism with non-PD pathologies,and the differential diagnosis of PD from neurodegenerative diseases such as multiple systemic atrophy(MSA)and progressive supranuclear palsy poses another unmet need.

文摘Peroxisome proliferator-activated receptor alpha is a member of the nuclear hormone receptor superfamily and functions as a transcription factor involved in regulating cellular metabolism.Previous studies have shown that PPARαplays a key role in the onset and progression of neurodegenerative diseases.Consequently,peroxisome proliferator-activated receptor alpha agonists have garnered increasing attention as potential treatments for neurological disorders.This review aims to clarify the research progress regarding peroxisome proliferator-activated receptor alpha in nervous system diseases.Peroxisome proliferator-activated receptor alpha is present in all cell types within adult mouse and adult neural tissues.Although it is conventionally believed to be primarily localized in the nucleus,its function may be regulated by a dynamic balance between cytoplasmic and nuclear shuttling.Both endogenous and exogenous peroxisome proliferator-activated receptor alpha agonists bind to the peroxisome proliferator-activated response element to exert their biological effects.Peroxisome proliferator-activated receptor alpha plays a significant therapeutic role in neurodegenerative diseases.For instance,peroxisome proliferator-activated receptor alpha agonist gemfibrozil has been shown to reduce levels of soluble and insoluble amyloid-beta in the hippocampus of Alzheimer's disease mouse models through the autophagy-lysosomal pathway.Additionally,peroxisome proliferator-activated receptor alpha is essential for the normal development and functional maintenance of the substantia nigra,and it can mitigate motor dysfunction in Parkinson's disease mouse models.Furthermore,peroxisome proliferator-activated receptor alpha has been found to reduce neuroinflammation and oxidative stress in various neurological diseases.In summary,peroxisome proliferator-activated receptor alpha plays a crucial role in the onset and progression of multiple nervous system diseases,and peroxisome proliferator-activated receptor alpha agonists hold promise as new therapeutic agents for the treatment of neurodegenerative diseases,providing new options for patient care.

文摘Alpha-synuclein and Parkinson's disease:Neuronal damage and inflammation caused by the aggregation of alpha-synuclein(α-syn)are central to a group of disorders known as synucleopathies,which includes Parkinson's disease(PD),dementia with Lewy bodies,and multiple system atrophy,among others.PD,the most common synucleinopathy,is the second most prevalent neurodegenerative disease after Alzheimer's disease,and it is the fastest growing.Its primary hallmark is the degeneration of dopaminergic neurons in the substantia nigra pars compacta,disrupting the communication with the striatum.

文摘The accumulation of^(222)Rn and^(220)Rn progeny in poorly ventilated environments poses the risk of natural radiation exposure to the public.A previous study indicated that satisfactory results in determining the^(222)Rn and^(220)Rn progeny concentrations by measuring the total alpha counts at five time intervals within 560 min should be expected only in the case of high progeny concentrations in air.To complete the measurement within a relatively short period and adapt it for simultaneous measurements at comparatively lower^(222)Rn and^(220)Rn progeny concentrations,a novel mathematical model was proposed based on the radioactive decay law.This model employs a nonlinear fitting method to distinguish nuclides with overlapping spectra by utilizing the alpha particle counts of non-overlapping spectra within consecutive measurement cycles to obtain the concentrations of^(222)Rn and^(220)Rn progeny in air.Several verification experiments were conducted using an alpha spectrometer.The experimental results demonstrate that the concentrations of^(222)Rn and^(220)Rn progeny calculated by the new method align more closely with the actual circumstances than those calculated by the total count method,and their relative uncertainties are all within±16%.Furthermore,the measurement time was reduced to 90 min,representing an acceleration of 84%.The improved capability of the new method in distinguishing alpha particles with similar energies emitted from ^(218)Po and^(212)Bi,both approximately 6 MeV,contributed to realizing more accurate results.The proposed method has the potential advantage of measuring relatively low concentrations of^(222)Rn and^(220)Rn progeny in air more quickly via air filtration.
