文摘Downregulation of the inwardly rectifying potassium channel Kir4.1 is a key step for inducing retinal Müller cell activation and interaction with other glial cells, which is involved in retinal ganglion cell apoptosis in glaucoma.Modulation of Kir4.1 expression in Müller cells may therefore be a potential strategy for attenuating retinal ganglion cell damage in glaucoma. In this study,we identified seven predicted phosphorylation sites in Kir4.1 and constructed lentiviral expression systems expressing Kir4.1 mutated at each site to prevent phosphorylation.Following this,we treated Müller glial cells in vitro and in vivo with the m Glu R I agonist DHPG to induce Kir4.1 or Kir4.1 Tyr^(9)Asp overexpression. We found that both Kir4.1 and Kir4.1 Tyr^(9)Asp overexpression inhibited activation of Müller glial cells.Subsequently,we established a rat model of chronic ocular hypertension by injecting microbeads into the anterior chamber and overexpressed Kir4.1 or Kir4.1 Tyr^(9)Asp in the eye,and observed similar results in Müller cells in vivo as those seen in vitro. Both Kir4.1 and Kir4.1 Tyr^(9)Asp overexpression inhibited Müller cell activation, regulated the balance of Bax/Bcl-2,and reduced the m RNA and protein levels of pro-inflammatory factors, including interleukin-1βand tumor necrosis factor-α.Furthermore,we investigated the regulatory effects of Kir4.1 and Kir4.1 Tyr^(9)Asp overexpression on the release of pro-inflammatory factors in a co-culture system of Müller glial cells and microglia. In this co-culture system,we observed elevated adenosine triphosphate concentrations in activated Müller cells, increased levels of translocator protein(a marker of microglial activation),and elevated interleukin-1βm RNA and protein levels in microglia induced by activated Müller cells. These changes could be reversed by Kir4.1 and Kir4.1 Tyr^(9)Asp overexpression in Müller cells.Kir4.1 overexpression, but not Kir4.1 Tyr^(9)Asp overexpression, reduced the number of proliferative and migratory microglia induced by activated Müller cells.Collectively, these results suggest that the tyrosine residue at position nine in Kir4.1 may serve as a functional modulation site in the retina in an experimental model of glaucoma.Kir4.1 and Kir4.1 Tyr^(9)Asp overexpression attenuated Müller cell activation, reduced ATP/P2X receptor–mediated interactions between glial cells, inhibited microglial activation, and decreased the synthesis and release of pro-inflammatory factors, consequently ameliorating retinal ganglion cell apoptosis in glaucoma.

文摘The hot compression deformation behavior of Mg-6Zn-1Mn-0.5Ca(ZM61-0.5Ca)and Mg-6Zn-1Mn-2Sn-0.5Ca(ZMT612-0.5Ca)alloys was investigated at deformation temperatures ranging from 250℃to 400℃and strain rates varying from 0.001 s^(-1) to 1 s^(-1).The results show that the addition of Sn promotes dynamic recrystallization(DRX),and CaMgSn phases can act as nucleation sites during the compression deformation.Flow stress increases with increasing the strain rate and decreasing the temperature. Both the ZM61-0.5Ca and ZMT612-0.5Ca alloys exhibit obvious DRX characteristics.CaMgSn phases can effectively inhibit dislocation motion with the addition of Sn,thus increasing the peak fl ow stress of the alloy. The addition of Sn increases the hot deformation activation energy of the ZM61-0.5Ca alloy from 199.654 kJ/mol to 276.649 kJ/mol,thus improving the thermal stability of the alloy. For the ZMT612-0.5Ca alloy, the optimal hot deformation parameters are determined to be a deformation temperature range of 350–400℃and a strain rate range of 0.001–0.01 s^(-1).

文摘This study investigates prescribed-time position tracking control for electromagnetic satellite formations subject to model uncertainties and external disturbances.Using the Clohessy-Wiltshire equations as the relative motion dynamics model,a prescribed time output feedback control strategy is proposed.A prescribed-time extended state observer is designed to estimate the relative velocity and external disturbances. The disturbance estimates are then used as the feedforward component of the controller.Building on this framework,a novel prescribed-time active disturbance rejection control strategy for position tracking is developed via a backstepping control design. The convergence of the extended state observer and the stability of the closed-loop system are rigorously analyzed using Lyapunov stability theory.Numerical simulations are performed to validate the effectiveness of the proposed controller.

文摘To elucidate the effect of calcite-regulated activated carbon(AC)structure on low-temperature denitrification performance of SCR catalysts, this work prepared a series of Mn-Ce/De-AC-xCaCO_(3)(x is the calcite content in coal)catalysts were prepared by the incipient wetness impregnation method, followed by acid washing to remove calcium-containing minerals.Comprehensive characterization and low-temperature denitrification tests revealed that calcite-induced structural modulation of coal-derived AC significantly enhances catalytic activity.Specifically,NO conversion increased from 88.3%of Mn-Ce/De-AC to 91.7%of Mn-Ce/De-AC-1CaCO_(3)(210℃).The improved SCR denitrification activity results from the enhancement of physicochemical properties including higher Mn^(4+)content and Ce^(4+)/Ce^(3+)ratio,an abundance of chemisorbed oxygen and acidic sites, which could strengthen the SCR reaction pathways(richer NH_(3)activated species and bidentate nitrate active species). Therefore,NO removal is enhanced.
文摘BACKGROUND:Hemiplegia,a prevalent stroke-related condition,is often studied for motor dysfunction;however, spasticity remains under-researched.Abnormal muscle tone significantly hinders hemiplegic patients’walking recovery.OBJECTIVE:To determine whether early suspension-protected training with a personal assistant machine for stroke patients enhances walking ability and prevents muscle spasms.METHODS:Thirty-two early-stage stroke patients from Shenzhen University General Hospital and the China Rehabilitation Research Center were randomly assigned to the experimental group(n=16)and the control group(n=16).Both groups underwent 4 weeks of gait training under the suspension protection system for 30 minutes daily,5 days a week. The experimental group used the personal assistant machine during training.Three-dimensional gait analysis(using the Cortex motion capture system),Brunnstrom staging,Fugl-Meyer Assessment for lower limb motor function,Fugl-Meyer balance function, and the modified Ashworth Scale were evaluated within 1 week before the intervention and after 4 weeks of intervention.RESULTS AND CONCLUSION:After the 4-week intervention, all outcome measures showed significant changes in each group. The experimental group had a small but significant increase in the modified Ashworth Scale score(P<0.05,d=|0.15|),while the control group had a large significant increase(P<0.05,d=|1.48|).The experimental group demonstrated greater improvements in walking speed(16.5 to 38.44 cm/s,P<0.05,d=|4.01|),step frequency(46.44 to 64.94 steps/min,P<0.05,d=|2.32|),stride length(15.50 to 29.81 cm,P<0.05,d=|3.44|),and peak hip and knee flexion(d=|1.82|to|2.17|).After treatment, the experimental group showed significantly greater improvements than the control group in walking speed(38.44 vs.26.63 cm/s,P<0.05,d=|2.75|),stride length, peak hip and knee flexion(d=|1.31|to|1.45|),step frequency(64.94 vs.59.38 steps/min,P<0.05,d=|0.85|),and a reduced support phase(bilateral:24.31%vs.28.38%,P<0.05,d=|0.88|;non-paretic:66.19%vs.70.13%,P<0.05,d=|0.94|).For early hemiplegia, personal assistant machine-assisted gait training under the suspension protection system helps establish a correct gait pattern, prevents muscle spasms, and improves motor function.
文摘Passiflora incarnata L.,commonly known as passionflower,is traditionally cultivated as an ornamental plant but has demonstrated diverse therapeutic potential. Its pharmacological effects are attributed to bioactive compounds such as flavonoids and alkaloids, which influence multiple biological pathways. This review aims to summarise and critically analyse recent findings on the pharmacological properties of Passiflora incarnata L.,focusing on its neuropsychiatric, antioxidant, antimicrobial, and anticancer activities.A targeted literature search was conducted in PubMed,Scopus,Web of Science, and Google Scholar for peer-reviewed publications between 2000 to 2025.Relevant articles were screened, and a more appropriate article related to the objective of the review was selected. Some classical papers are also cited as per the requirement of the topic.Passiflora incarnata L.showed multifunctional medicinal properties with various applications in neuropsychiatry, oxidative stress management, antimicrobial agent, and as an anticancer agent. The U.S.Food and Drug Administration categorizes passionflower extracts as“generally recognized as safe”.However, most evidence remains preclinical, with methodological variation limiting generalisation.Standardised formulation, robust clinical trials, and in-depth in vivo studies are essential to establish its therapeutic relevance and safety in modern medicine.

文摘Aerodynamic research on road cars was reviewed in this work under the thread of reducing drag, with the awareness that this may succeed in effectively decreasing the carbon footprint of transportation.First,a selection of studies was presented to focus on the most important aerodynamic features of the flow around realistic car body shapes. Then, the discussion was organized around three pillars related to passive flow control, active flow control and active aerodynamics. Both experimental and numerical investigations were included to provide a comprehensive overview.A clear distinction was made between simplified and realistic car models,as well as production vehicles(within the limits of restricted access information). Moreover,a short essay was dedicated to electric vehicles, for which aerodynamics matters, especially at highway speeds.Last, the impact of aerodynamic principles on the design of current and future vehicle fleet was assessed, honestly admitting that recent market trends must be reversed to turn decarbonization goals into reality and damp the effects of global warming.
文摘Origanum elongatum(OE)is an aromatic, medicinal plant endemic to Morocco that is widely used in traditional medicine due to its biological properties. This study aimed to elucidate the chemical composition of the essential oil(EO)obtained from O.elongatum(OEEO)at three stages of its life cycle, including vegetative stage(OEEO-VS),flowering stage(OEEO-FS),and post-flowering(OEEO-PFS),as well as to evaluate its biological and antiradical characteristics. The chemical analysis of the essential oil was conducted using gas chromatography-mass spectrometry(GC-MS).The antibacterial activity was evaluated in vitro through distinct methodologies, namely, disc diffusion and microatmosphere assay;subsequently, the minimum inhibitory concentration(MIC)was then determined. The antioxidant potential was also measured by using the DPPH and FRAP assays. The GC-MS revealed the predominant of p-cymene(26.83%_31.45%),γ-terpinene(8.46%_26.95%),thymol(13%_29.54%),and carvacrol(20.25%_37.26%),in all three samples, with notable variations according to the phenological stage of the samples. The EOs extracted at three phenological stages demonstrated notable antibacterial efficacy against all the phytopathogen tested. The MICs for Erwinia amylovora exhibited a range of 6.25 and 250μg/mL.However,for Agrobacterium tumefaciens C58 and Allorhizobium vitis S4,the MICs spanned 125 and 250μg/mL.In the DPPH test, the IC50 values were 168.25±1.14,147.01±0.78,and 132.01±2.06μg/mL for EOs derived from the vegetative, flowering, and post-flowering period, respectively. In the FRAP test, the EC50 values were 164.22±1.04,215.73±1.48,and 184.06±0.95μg/mL for the same stages. The findings offer promising prospects for the phytochemical development, demonstrating how the phenological stage significantly influences the therapeutic and biotechnological potential of O.elongatum.This has the potential to open up new avenues of research in the pharmaceutical, agronomic, and environmental fields.

文摘To develop herbicides with a novel mechanism of action,a series of 1,3,4-oxadiazolpyridine derivatives were designed and synthesized based on active substructure splicing and structure optimization. These derivatives(5aa-5bd)were characterized by their melting points,^(1)H and^(13)C nuclear magnetic resonance spectroscopy, and high-resolution mass spectrometry. The configuration of 5 al was determined using single-crystal X-ray diffraction.Additionally,5 al exhibited excellent herbicidal activity at a dosage of 75 g/hm^(2),showing an EC 50 of 4.03 g/hm^(2)against both E.crus-galli and quinclorac-resistant E.crus-galli.At a dosage of 375 g/hm 2,5 al was safe for application on rice and sorghum and showed low toxicity(>200μg/g)towards Apis mellifera. After treatment with 5 al,the lamellae of the chloroplast grana of barnyard grass leaves were stacked disorderly and arranged loosely, and some thylakoids were broken,as observed by transmission electron microscopy.Transcriptomics analysis of E.crus-galli revealed that 5 al affects the defense response, membranes, plasma membranes, and chloroplasts of differentially expressed genes, which alter membrane permeability and energy metabolism, potentially leading to plant death. Thus,we successfully developed a novel molecular scaffold with a new mechanism of action that exhibits herbicidal activity against resistant E.crus-galli.Therefore,further development of lead herbicides based on this scaffold is required.
文摘Rationale:This case report describes a couple with recurrent fertilization failure despite undergoing multiple cycles of intracytoplasmic sperm injection(ICSI).The principal clinical concern was suspected oocyte activation deficiency(OAD),in which fertilization is impeded due to the oocyte’s inability to initiate embryogenesis, commonly attributed to inadequate intracellular calcium(Ca^(2+))release following sperm injection.Patient concerns:The couple repeatedly experienced complete or near-complete fertilization failure in previous ICSI cycles, raising suspicion of an underlying oocyte activation defect.Diagnosis:Based on the repeated absence of fertilization post-ICSI and clinical history,a diagnosis of suspected OAD leading to recurrent ICSI fertilization failure was considered.Interventions:Artificial oocyte activation(AOA)using the calcium ionophore A23187 was performed. After ICSI,unfertilized oocytes were exposed to the ionophore to induce Ca^(2+)influx,simulating physiological calcium oscillations essential for oocyte activation. The efficacy of intervention was evaluated through subsequent embryonic development, morphological grading, and chromosomal integrity.Outcomes:Following AOA treatment, successful oocyte activation occurred, resulting in the formation of high-grade embryos with normal developmental progression.Chromosomal analysis revealed no detectable abnormalities, indicating genomic stability.Lessons:Calcium ionophore–mediated AOA may serve as an effective adjunct in cases of recurrent ICSI failure attributed to OAD.This case highlights the importance of individualized therapeutic strategies in assisted reproduction;however, further research is needed to refine protocols, validate broader clinical efficacy, and assess long-term safety, including potential epigenetic risks.
文摘BACKGROUND Postoperative delirium(POD)is a common and serious complication in surgical patients, particularly older adults.Alterations in cholinergic function have been implicated in its pathophysiology.AIM To evaluate the association between preoperative serum cholinesterase(ChE)activity—specifically butyrylcholinesterase(BuChE)and acetylcholinesterase(AChE)—and the risk of POD in adult surgical patients in a meta-analysis.METHODS A systematic search was conducted in PubMed,EMBASE,and Web of Science up to March 28,2025 for studies reporting preoperative serum BuChE or AChE activity in relation to subsequent POD incidence.Standardized mean differences(SMDs)and odds ratios(ORs)with 95%confidence intervals(CIs)were pooled using random-effects models.Subgroup and sensitivity analyses were performed based on follow-up duration and analytic models.RESULTS Thirteen studies(n=2730 patients)were included.Patients who developed POD had significantly lower preoperative BuChE activity than those who did not(SMD=-0.28;95%CI:-0.39 to-0.16;I^(2)=18%).Higher BuChE activity was associated with a reduced risk of POD(OR per 100 unit increment=0.97;95%CI:0.95-0.99;I2=0%).In contrast, pooled AChE activity did not differ significantly between POD and non-POD groups(SMD=-0.25;95%CI:-0.53 to 0.03;P=0.08;I^(2)=80%),and the ORs per 1 unit increment in AChE activity were not statistically significant(OR=0.98;95%CI:0.95-1.01).CONCLUSION Lower preoperative serum BuChE activity is associated with an increased risk of POD in adults undergoing surgery.BuChE activity may serve as a potential preoperative biomarker for POD risk stratification.

文摘Conductive materials(CM)can improve methane production(MP)efficiency in many methanogenic systems. However, several types of CM exist, and there are uncertainties regarding whether they all improve MP efficiency to the same extent and modulate microbial communities in a similar way.To investigate that, different microbial enrichments with and without activated carbon(AC),magnetite(Mag),and zeolites(Zeo)(at 0.5 g/L)were developed.MP profiles and microbial composition changes were compared among enrichments. The behavior of all enrichments was different, although the initial inoculum sludge was the same.Lag phase duration was lower in AC enrichment, while the complete conversion of butyrate to methane was faster in Mag enrichment.Syntrophomonas was the most abundant bacterial genus in all enrichments, but changes in the methanogenic community were evident.Acetoclastic methanogens were more diverse in Mag enrichment, with microorganisms assigned to Methanosarcina and Methanothrix gener1,but Methanothrix was the only acetoclastic methanogen in the other enrichments. On the other hand, different species of hydrogenotrophic methanogens prevailed in distinct enrichments. The metatranscriptomics results revealed that the dominant mechanism of interspecies electron transfer in the AC enrichment utilized hydrogen as the electron carrier, and no evidences of direct interspecies electron transfer(DIET)could be found. These results showed how different CM modulate microbial communities and affect MP efficiency through mechanisms that do not necessarily involve DIET or mediation via CM.

文摘Guggulsterone(GS)is a bioactive compound primarily extracted from the oleo-gum resin of plants in the Commiphora and Boswellia genera.Modern pharmacological studies have demonstrated that GS possesses a broad spectrum of biological activities, with notable therapeutic potential in inflammatory disorders, neurodegenerative conditions, diabetes mellitus, and various cancers. In this review,we systematically analyzed relevant literature published up to 2024 from the CNKI,Web of Science,ScienceDirect, and PubMed databases to summarize the current understanding of GS's pharmacological effects, toxicity profile, and pharmacokinetic properties. The findings indicate that GS exerts potent antioxidant,anti-inflammatory,anticancer,antiviral,antidepressant,lipid-lowering,and cardiovascular protective effects, primarily through modulation of key signaling pathways such as the Janus kinase(JAK)-signal transducer and activator of transcription 3(STAT3),nuclear factor erythroid 2-related factor 2(Nrf2)/heme oxygenase-1(HO-1),Nrf2/Keap1,nuclear factor kappa-B(NF-κB),AMPK,phosphoinositide 3-kinase(PI3K)/protein kinase B(Akt),and mitogen-activated protein kinase(MAPK)/activator protein-1(AP-1).Additionally,GS may help overcome limitations associated with conventional chemotherapy by modulating drug resistance via regulation of p-glycoprotein activity.Following hepatic metabolism mediated by cytochrome P450 enzymes,GS does not appear to cause significant adverse effects. This review provides a comprehensive synthesis of the sources, pharmacological actions, safety, pharmacokinetics, and potential applications of GS.Future research should focus on structural modification of GS,development of novel formulations, and exploration of synergistic combinations with other therapeutic agents to broaden its clinical utility.

文摘Activation pruning reduces neural network complexity by eliminating low-importance neuron activations, yet identifying the critical pruning threshold—beyond which accuracy rapidly deteriorates—remains computationally expensive and typically requires exhaustive search. We introduce a thermodynamics-inspired framework that treats activation distributions as energy-filtered physical systems and employs the free energy of activations as a principled evaluation metric.Phase-transition-like phenomena in the free-energy profile—such as extrema, inflection points, and curvature changes—yield reliable estimates of the critical pruning threshold, providing a theoretically grounded means of predicting sharp accuracy degradation. To further enhance efficiency,we propose a renormalized free energy technique that approximates full-evaluation free energy using only the activation distribution of the unpruned network. This eliminates repeated forward passes, dramatically reducing computational overhead and achieving speedups of up to 550×for MLPs.Extensive experiments across diverse vision architectures(MLP,CNN,ResNet,MobileNet,Vision Transformer)and text models(LSTM,BERT,ELECTRA,T5,GPT-2)on multiple datasets validate the generality, robustness, and computational efficiency of our approach.Overall, this work establishes a theoretically grounded and practically effective framework for activation pruning, bridging the gap between analytical understanding and efficient deployment of sparse neural networks.

文摘BACKGROUND Major depressive disorder(MDD)and obesity(OB)are bidirectionally comorbid conditions with common neurobiological underpinnings. However, the neurocognitive mechanisms of their comorbidity remain poorly understood.AIM To examine regional abnormalities in spontaneous brain activity among patients with MDD-OB comorbidity.METHODS This study adopted a regional homogeneity(ReHo)analysis of resting-state functional magnetic resonance imaging. The study included 149 hospital patients divided into four groups:Patients experiencing their first episode of drug-naive MDD with OB,patients with MDD without OB,and age-and sex-matched healthy individuals with and without OB.Whole-brain ReHo analysis was conducted using SPM12 software and RESTplus toolkits, with group comparisons via ANOVA and post-hoc tests.Correlations between ReHo values and behavioral measures were examined.RESULTS ANOVA revealed significant whole-brain ReHo differences among the four groups in four key regions:The left middle temporal gyrus(MTG.L),right cuneus, left precuneus, and left thalamus.Post-hoc analyses confirmed pairwise differences between all groups across these regions(P<0.05).OB was associated with ReHo alterations in the MTG.L,right cuneus, and left thalamus, whereas abnormalities in the precuneus suggested synergistic pathological mechanisms between MDD and OB.Statistically significant correlations were found between the drive and fun-seeking dimensions of the behavioral activation system,as well as behavioral inhibition and the corresponding ReHo values.CONCLUSION Our findings provide novel evidence for the neuroadaptive mechanisms underlying the MDD-OB comorbidity.Further validation could lead to personalized interventions targeting MTG.L hyperactivity and targeting healthy food cues.
