Understanding how species diverge and adapt is fundamental to unraveling biodiversity.While environmental impacts on species evolution are well-documented,the roles of intrinsic life-history traits remain underexplored.The Qinghai-Xizang Plateau,with its harsh conditions and unique biodiversity,offers a natural laboratory for such investigations.Here,we examine two sympatric small mammals—the solitary,low-dispersal plateau zokor(Eospalax baileyi)and the social,high-dispersal plateau pika(Ochotona curzoniae)—to elucidate how life-history traits shape population structures and adaptive strategies.Through whole-genome sequencing and cardiac-blood phenotype analyses,we reveal striking differences in their evolutionary trajectories.Despite enduring similar environmental pressures,plateau zokor populations exhibit pronounced genetic subdivisions,high inbreeding,and distinct local adaptations.In contrast,plateau pika populations display genetic panmixia,widespread diversity,and adaptive uniformity.Demographic inference highlights that plateau zokors experienced severe population bottlenecks and restricted gene flow during glacial periods,underscoring the impact of dispersal capacity on evolutionary outcomes.Our findings demonstrate that intrinsic biological traits,particularly dispersal ability,fundamentally influence genetic architecture,population connectivity,and local adaptation.This study not only provides empirical evidence of how life-history traits shape evolutionary dynamics but also offers a framework for integrating intrinsic and extrinsic factors in understanding biodiversity formation. 展开更多

The mound-making behavior of plateau zokors is one of the most important factors in remodeling meadow microtopography and causing soil erosion in the Yellow River source area of western China,but little is known about the effects of microtopography on particle size characteristics(PSC)of eroded sediments from the bare slopes of zokor mounds during different rainfall events.In this study,we analyzed the relationship of microtopographic features derived from laser point cloud data and PSC of eroded sediments at six simulated rainfall intensities(all lasting 60 min).The effects of microtopography on PSC of eroded sediments were studied via partial least squares regression(PLSR)and structural equation modeling(SEM).The results showed that:(1)15-20 minutes from the beginning of rainfall was the sensitive period of soil loss from the slopes,and the function relationship between the rate of sediment and runoff and rainfall intensity can better predict the development trend of soil erosion;(2)Intense erosion occurred mainly in the upper half of the zokor mound,while deposition was mainly limited to its lower half.It is suggested that diminished plateau zokor activity intensity can effectively prevent and control soil erosion;(3)The PSC of eroded sediment is dominated by silt,followed by sand,with clay being the least abundant,and the eroded sediments with a particle size of 10-20μm were sensitive and highly susceptible to rainfall erosion.This finding facilitates the understanding of the formation process of surface geomorphology and the mechanism of soil erosion;(4)The PLSR model indicates that microtopography has an extensive influence on eroded sediments during hydraulic erosion,and the SEM analysis results further confirm that the fractal dimension was the best parameter to represent the PSC of eroded sediments,whereas surface cutting degree was the dominant factor controlling the PSC of eroded sediments.These findings are crucial for predicting soil erosion in the Yellow River source area and provide a new perspective for understanding soil erosion mechanisms in alpine meadow ecosystems. 展开更多

Zokors in the genus Eospalax,which are endemic to northern and western China,are subterranean rodents that inhabit various niches,including grasslands,high-altitude meadows,forests,and farmlands.Six species in Eospalax were described a century ago but their taxonomy and phylogeny remain controversial.In this study,we performed high-depth whole-genome sequencing of 47 zokor samples,comprising all six previously described species.Genomic analyses revealed a reliable and robust phylogeny of Eospalax and supported the validity of the six named species.According to the inferred phylogenetic relationships,Eospalax first divergent into two clades in the early Pliocene(ca.4.68 million years ago(Ma)),one inhabiting the highaltitude Qinghai-Xizang(Tibet)Plateau(QTP)and adjacent regions,and the another inhabiting the lowaltitude Loess Plateau and Qinling-Daba Mountains.The most recent divergences occurred between E.baileyi and E.smithii and between E.rufescens and E.rothschildi in the late Pliocene(ca.2.09 and 2.19 Ma,respectively).We also collected specimens of zokors in the southern Hengduan Mountains(Muli County,Sichuan Province),far from the known distributions of all other zokors.Morphological and molecular analyses strongly suggested that the specimens represent a new species,formally described here as Eospalax muliensis sp.nov.The new species belongs to the high-altitude clade and diverged from closely related species(ca.4.22 Ma)shortly after the first divergence in Eospalax.Interestingly,Eospalax muliensis sp.nov.possesses more supposedly plesiomorphic characters,suggesting a possible origin of the genus in the Hengduan Mountains. 展开更多

Mounds constructed by plateau zokors,which is widely distributed in alpine meadows significantly modified plant community structure.However,the variations of plant community structure under the disturbance of plateau zokor-made mound are less concerned.Therefore,we investigated the responses of plant community on zokor-made mound of different years(1 a and 3-4 a),and compared with undisturbed sites(no mound)in an alpine meadow in the eastern Qinghai-Tibetan Plateau(QTP),China.Species richness,coverage and Simpson diversity index were all significantly reduced by the presence of zokor-made mound,but plant heights were significantly increased,particularly in grasses and sedges.Several perennial forage species showed an increased importance value and niche breadth,including Koeleria macrantha,Elymus nutans and Poa pratensis.The effect of zokor-made mound on niche overlap showed that more intense interspecific competition produced a greater utilization of environmental resources.And this interspecific niche overlap was strengthened as succession progressed.The bare mound created by zokor burrowing activities provided a colonizing opportunity for non-dominant forage species,resulting in abundant plant species and plant diversity during the succession period.We concluded that presence of zokor-made mound was conducive to regeneration and vitality of plant community in alpine meadows,thus improving their resilience to anthropogenic stress. 展开更多

Plateau zokor(Eospalax baileyi)is a subterranean rodent and seasonal breeder.During the non-breeding season,the testicles regress,leading to the arrest of spermatogenesis and loss of fertility.The identification of the specific germ cell type at which spermatogenesis is arrested,as well as potential regulatory factors during the non-breeding season,is important for understanding seasonal spermatogenesis in subterranean species.This study analyzed genes in spermatocytes of plateau zokor by referring to single-cell RNA results in mice.We discovered that spermatogenesis is arrested at the spermatocyte during the non-breeding season,which was corroborated via immunofluorescence staining results.The analysis of gene expression during different stages of meiotic prophase I has revealed that germ cell development may be arrested,starting from zygonema,during the non-breeding season.Meanwhile,we discovered that the apoptosis genes were up-regulated,leading to apoptosis in spermatocytes.To confirm that the germ cell differentiation was blocked during the non-breeding season due to a decrease in the androgen level,we used androgen receptor antagonist(flutamide)to intervene in the breeding season and found that the inner diameter of the seminiferous tubules was significantly reduced,spermatogenesis was arrested,and spermatocytes underwent apoptosis.This study revealed that spermatocytes are the terminal of germ cell differentiation in plateau zokor during the non-breeding season and that the arrest of differentiation is attributed to a decline in androgen levels.Our results complement the theoretical basis of seasonal reproduction in plateau zokor. 展开更多

Gut Microbiota Regulate Lipid Metabolism via the Bile Acid Pathway:Resistance to Hypoxia in Gansu Zokor(Eospalax cansus)

The Gansu zokor(Eospalax cansus),a subterranean rodent endemic to the Loess Plateau of China,exhibits remarkable adaptability to hypoxic environments.While gut microbiota are known to regulate lipid metabolism through bile acid(BA)pathways,this phenomenon has not been investigated in subterranean rodents exposed to hypoxia.This study employed 16SrRNA sequencing,targeted analysis of BA metabolites in colonic contents,and assessments of BA and lipid metabolites alongside molecular analyses in the liver and ileum under conditions of acute and chronic hypoxia in Gansu zokors.The results revealed that hypoxia altered the composition of gut microbiota and BA pools in Gansu zokors.Hypoxia-induced changes increased the abundance of gut microbiota associated with BA metabolism,thereby modulating lipid metabolism via farnesoid X receptor(FXR)signaling in the distal ileum and liver cells.Under acute hypoxia,FXR upregulated lipid synthesis and suppressed fatty acidβ-oxidation by downregulating the carnitine palmitoyl-transferase1A(CPT1A)expression.Conversely,during chronic hypoxia,particularly under long-term exposure,FXR reduced lipid synthesis and enhanced fatty acidβ-oxidation by upregulating acyl-CoA oxidase(ACOX1)expression.In both hypoxic conditions,FXR facilitated lipoprotein metabolism.In summary,this study elucidates that gut microbiota-mediated BA metabolic pathways contribute to the Gansu zokor’s ability to maintain lipid metabolic homeostasis and adaptation to hypoxia. 展开更多

物种多样性与生产力关系是当前生态学研究的重点之一。高原鼢鼠鼠丘作为自然形成的“扰动梯度实验平台”,为探究植物群落构建过程中物种多样性与生产力关系提供了独特视角。以青藏高原东麓高寒草甸禁牧样地中不同年限的高原鼢鼠(Myospal... 物种多样性与生产力关系是当前生态学研究的重点之一。高原鼢鼠鼠丘作为自然形成的“扰动梯度实验平台”,为探究植物群落构建过程中物种多样性与生产力关系提供了独特视角。以青藏高原东麓高寒草甸禁牧样地中不同年限的高原鼢鼠(Myospalax baileyi)鼠丘为研究对象,分析其上植被物种多样性及地上、地下和总生物量的变化格局,结合土壤理化特性解析影响鼠丘植被演替的关键因子。结果表明:(1)随着演替年限增加,土壤容重、含水率、有机碳和碳磷比在各年份间存在显著性差异,其中多年鼠丘的土壤全磷、有机碳和碳磷比均显著低于一年鼠丘。(2)地上生物量随演替年限增加而显著增大,为演替初期的5.40倍;而地下生物量和总生物量呈先降低后增加的趋势,其中总生物量在多年时达到峰值,较最低时(两年)高出156.51%。Shannon-Wiener指数和Simpson指数均呈先增长后降低的趋势,且在第二年时达到最大,比平地对照分别高出15.34%和5.54%。(3)高原鼢鼠鼠丘植被演替过程中物种多样性与生产力(地上生物量、地下生物量、总生物量)间呈负相关格局,但是该负相关关系的形成是演替阶段特异性过程的综合结果,而非单一阶段的静态关系。(4)Mantel分析和结构方程模型表明,物种多样性与生产力受土壤容重、含水量、有机碳含量和碳磷比的阶段性动态调控,其中土壤有机碳含量对总生物量的变异解释率为32.00%,土壤碳磷比对多样性指数(Shannon-Wiener指数和Simpson指数)和总生物量的变异解释率分别为57.00%和51.00%。综上所述,高原鼢鼠鼠丘植被演替过程中资源竞争加剧和空间异质性的降低会使群落生产力提高的同时降低物种多样性。该结果为高寒草甸生态恢复、生物多样性保护及生态系统功能维持提供了理论依据。 展开更多

Background:Plateau zokor inhabits in sealed burrows from 2,000 to 4,200 meters at Qinghai-Tibet Plateau.This extreme living env ironment makes it a great model to study animal adaptation to hypoxia,low temperature,and high carbon dioxide concentration.Methods:We provide an integrated resource,ZokorDB,for tissue specific regulatory network annotation for zokor.ZokorDB is based on a high-quality draft genome of a plateau zokor at 3,300 m and its transcriptional profiles in brain,heart,liver,kidney,and lung.The conserved non-coding elements of zokor are annotated by their nearest genes and upstream transcriptional factor motif binding sites.Results:ZokorDB provides a general draft gene regulatory network(GRN),Le?potential transcription factor(TF)binds to non-coding regulatory elements and regulates the expression of target genes(TG).Furthermore,we refined the GRN by incorporating matched RNA-seq and DNase-seq data from mouse ENCODE project and reconstructed five tissue-specific regulatory networks.Conclusions:A web-based,open-access database is developed for easily searching,visualizing,and downloading the annotation and data.The pipeline of non-coding region annotation for zokor will be useful for other non-model species.ZokorDB is free available at the website(bigd.big.ac.cn/zokordb/). 展开更多

All organisms respond to variation in their environments and manage environmental stress through metabolic adjustments.The plateau zokor(Myospalax baileyi) is an endemic and keystone subterranean rodent species that inhabits the Qinghai-Tibet Plateau between 2800 and 4200 m above sea level.It is a hypoxic-tolerant mammal with a high ratio of oxygen utilization that enables it to cope with its harsh surroundings.To explore the molecular mechanism of altitude acclimatization of the plateau zokor,we cloned the zokor erythropoietin(Epo) gene and used real-time PCR to compare Epo mRNA levels in zokors inhabiting 16 different altitudes.The full-length zokor Epo open reading frame was 579 bp that encoded a precursor peptide of 192 amino acids with a signal peptide of 26 residues.The Epo gene of the plateau zokor was 81%-95% homologous to that of human,mouse,rat,root vole and the Golan Heights blind mole rat,with the highest homology(95%) to species of the genus Spalax.Epo mRNA was detected mainly in the zokor kidney and spleen among 8 selected tissues.The level of Epo mRNA increased in the liver and kidney with increases in altitude.The increase in the kidney was 5 times that in the liver.Remarkably,expression of Epo mRNA in the kidney of zokors living at the highest altitude(4268 m) was 12-fold higher than that of zokors living at the lowest(2492 m) altitude.These findings provide essential information for understanding the possible role of Epo in adaptation to hypoxia in the plateau zokor. 展开更多

为探究高原鼢鼠(Eospalax baileyi)干扰对土壤生态化学计量特征与植物群落多样性的影响及其内在关联变化,设置轻度、中度和重度3个干扰强度作为干扰区,以无鼠丘草地作为非干扰区,分别调查各样地植物丰富度、多度、土壤有机碳(C)、全氮(N... 为探究高原鼢鼠(Eospalax baileyi)干扰对土壤生态化学计量特征与植物群落多样性的影响及其内在关联变化,设置轻度、中度和重度3个干扰强度作为干扰区,以无鼠丘草地作为非干扰区,分别调查各样地植物丰富度、多度、土壤有机碳(C)、全氮(N)和全磷(P)含量,分析不同干扰强度下植物群落物种多样性和土壤化学计量比的差异和变化规律,明晰高原鼢鼠干扰是否对土壤生态化学计量比和植物群落物种多样性的关系产生影响。结果表明:(1)重度干扰土壤有机碳、全氮含量显著低于其他干扰强度,轻度干扰土壤全磷含量显著低于其他干扰强度(P<0.05);(2)轻度干扰下土壤碳磷比显著高于其他干扰强度,土壤氮磷比显著高于非干扰区与重度干扰区(P<0.01),而不同干扰强度下土壤碳氮比无显著性差异;(3)随着干扰强度增加,植物群落物种丰富度指数、多样性指数和均匀度指数均呈增加趋势,且显著高于非干扰区(P<0.05);(4)物种丰富度指数与土壤碳氮比和氮磷比的关系在非干扰区表现为显著正相关,而高原鼢鼠干扰后两者的显著关系消失,当高原鼢鼠干扰达到一定程度(重度干扰)会显著影响物种多样性指数与土壤氮磷比和碳磷比的关系,而植物均匀度指数与土壤氮磷比的显著正相关关系重新得以恢复。综上,高原鼢鼠干扰显著影响了土壤有机碳、全磷和全氮含量,有利于植物物种多样性的增加,同时调控植物群落多样性与土壤养分计量比间的相互作用。 展开更多

为明确高原鼢鼠(Eospalax baileyi)鼠丘土壤化学计量比和酶活性动态变化特征,本研究基于时空互代法,在青藏高原东北缘的高寒草甸区选择不同年限的鼠丘,探究其恢复过程中植被、土壤化学计量比和酶活性变化规律。结果发现:与未受干扰样地... 为明确高原鼢鼠(Eospalax baileyi)鼠丘土壤化学计量比和酶活性动态变化特征,本研究基于时空互代法,在青藏高原东北缘的高寒草甸区选择不同年限的鼠丘,探究其恢复过程中植被、土壤化学计量比和酶活性变化规律。结果发现:与未受干扰样地相比,1年鼠丘的植被丰富度、地上和地下植物生物量显著降低(P<0.05),但随鼠丘恢复年限增加呈上升趋势;与1年鼠丘相比,多年鼠丘的土壤C∶P,N∶P以及脲酶、淀粉酶和纤维素酶活性显著增加(P<0.05),而土壤C∶N、蔗糖酶、硝酸还原酶和亚硝酸还原酶活性无显著变化;鼠丘恢复年限解释了24.4%的土壤化学计量比和76.1%的酶活性变异。鼠丘恢复过程中土壤容重、有机碳含量、全氮含量、地上和地下植物生物量的增加,提高了土壤酶活性和碳氮含量。研究增进了高原鼢鼠对草地生态系统影响的认识,为鼠害型退化高寒草地的修复提供了参考。 展开更多

为深入了解高原鼢鼠(Eosplax baileyi)高原适应性的生物学基础,应用生物信息学相关软件分析高原鼢鼠肉碱棕榈酰基转移酶1A(carnitine palmitoyl transferase 1A,CPT-1A)基因序列的同源性和趋同进化位点,利用qRT-PCR方法测定不同海拔条件... 为深入了解高原鼢鼠(Eosplax baileyi)高原适应性的生物学基础,应用生物信息学相关软件分析高原鼢鼠肉碱棕榈酰基转移酶1A(carnitine palmitoyl transferase 1A,CPT-1A)基因序列的同源性和趋同进化位点,利用qRT-PCR方法测定不同海拔条件下(3700 m和2700 m)高原鼢鼠肝脏、骨骼肌和脂肪组织中CPT-1A基因的表达水平。结果表明:高原鼢鼠CPT-1A基因与盲鼹鼠(Nannospalax galili)的同源性最高;高原鼢鼠、盲鼹鼠和裸鼹鼠(Heterocephalus glaber)3种地下鼠CPT-1A存在明显的趋同进化位点;随着海拔的升高,高原鼢鼠背部脂肪组织中CPT-1A基因表达水平显著升高;不同组织之间的比较发现,肝脏组织中CPT-1A基因表达量最高。以上结果提示,高原鼢鼠CPT-1A基因在进化过程中形成了特定的适应性突变,低氧上调高原鼢鼠脂肪组织中CPT-1A基因的表达,这可能与高原鼢鼠适应地下寒冷、低氧和高二氧化碳浓度的洞道生境有关,是高原鼢鼠适应地下洞道生境的分子机制之一。 展开更多

低氧环境下高原鼢鼠及SD大鼠肺和骨骼肌中SP‐A的表达模式

肺表面活性蛋白A(Surfactant protein A,SP‐A)是最丰富的肺表面活性蛋白,由SFTPA基因编码合成,主要在哺乳动物的肺组织中合成,也在骨骼肌等组织中表达。SP‐A具有降低肺表面张力、增强肺气体交换、调节免疫、清除病原体和降低炎症反应... 肺表面活性蛋白A(Surfactant protein A,SP‐A)是最丰富的肺表面活性蛋白,由SFTPA基因编码合成,主要在哺乳动物的肺组织中合成,也在骨骼肌等组织中表达。SP‐A具有降低肺表面张力、增强肺气体交换、调节免疫、清除病原体和降低炎症反应的功能。本研究通过对不同海拔SD大鼠(2250 m和3700 m)和高原鼢鼠(Eospalax baileyi)(2700 m和3700 m)肝、肺、骨骼肌、小肠、结肠和脂肪组织转录组测序分析后;利用qRT‐PCR法比较分析SFTPA基因在两个物种骨骼肌和肺组织中的表达水平;利用Western Blotting法比较分析SP‐A在两个物种骨骼肌、肺组织和血清中的表达水平。转录组分析发现,随着海拔升高,大鼠骨骼肌中SFTPA基因表达显著上调,但在高原鼢鼠骨骼肌中其表达没有显著差异;qRT‐PCR结果显示,随着海拔升高,生境氧含量下降,SD大鼠骨骼肌组织中SFTPA表达升高,高原鼢鼠骨骼肌组织中SFTPA表达显著降低,且SD大鼠和高原鼢鼠肺组织中SFTPA表达均显著降低;Western Blotting结果显示,随着海拔升高,生境氧含量下降,在SD大鼠和高原鼢鼠骨骼肌、血清和肺组织中SP‐A的表达水平均显著升高。总之,高海拔低氧环境下,SD大鼠和高原鼢鼠肺组织中SFTPA基因转录表达显著降低,但两个物种肺组织中SP‐A含量不仅没有下降,反而显著升高,这可能是由骨骼肌合成并分泌的SP‐A经血液转运至肺组织的结果,也是SD大鼠习服低氧环境,高原鼢鼠适应低氧环境的重要机制。 展开更多

在管涔山高桥洼林场采用多种防治措施对中华鼢鼠进行了防治,旨在提高油松幼林的保存率。实验设计10种不同处理,涉及单一和综合防治手段,以及物理、化学、生物和营林措施。结果显示,综合防治措施(尤其是包含预防措施的处理)在提高油松幼... 在管涔山高桥洼林场采用多种防治措施对中华鼢鼠进行了防治,旨在提高油松幼林的保存率。实验设计10种不同处理,涉及单一和综合防治手段,以及物理、化学、生物和营林措施。结果显示,综合防治措施(尤其是包含预防措施的处理)在提高油松幼林保存率方面效果显著,其中A4处理的平均保存率达到了99.1%。不同处理在各年份的保存率波动情况表明,综合防治措施能够提供更稳定的保护效果,尤其是预防措施在综合防治策略中具有很大的作用,并推荐在相似生态条件下采用此策略,以有效控制中华鼢鼠,保护油松幼林,促进生态平衡和可持续发展。本研究为中华鼢鼠的防治提供了科学依据,对其他地区的害鼠管理也具有参考价值。 展开更多

高原鼢鼠和高原鼠兔是青藏高原土著动物,对低氧具有很好的适应性。为了探讨在低氧环境中两者肺细叶结构的适应特征,应用体视学方法测量了肺细叶相关指标。结果发现:高原鼢鼠和高原鼠兔肺单位面积肺泡数显著高于SD大鼠,单个肺泡面积和弹... 高原鼢鼠和高原鼠兔是青藏高原土著动物,对低氧具有很好的适应性。为了探讨在低氧环境中两者肺细叶结构的适应特征,应用体视学方法测量了肺细叶相关指标。结果发现:高原鼢鼠和高原鼠兔肺单位面积肺泡数显著高于SD大鼠,单个肺泡面积和弹性纤维/肺实质比显著低于SD大鼠;高原鼢鼠肺泡隔厚度最厚,高原鼠兔最薄,且三种动物具显著差异;高原鼢鼠和高原鼠兔气-血屏障的算术平均厚度(Ta)和调和平均厚度(Th)均显著低于SD大鼠;在三个级别的微血管中,高原鼠兔中膜肌层厚度显著低于高原鼢鼠,两种高原动物均显著低于SD大鼠;高原鼢鼠和高原鼠兔的微血管密度(MVD)显著高于SD大鼠。以上结果表明,高原鼢鼠和高原鼠兔肺细叶结构特征表现出一定趋同,这些特征有利于在低氧条件下提高肺气体扩散容量;但是,肺泡隔厚度和微血管中膜肌层厚度/血管外径比又表现出明显的差异,可能是不同生境造成的。 展开更多
