Objective This study aimed to explore the mechanism of obtaining yang from yin in Yougui pill against aging based on network pharmacology and molecular docking technology.Methods The active components and targets of Yougui Pill were obtained by searching the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP)database and the Bioinformatics Analysis Tool for Molecular mechANism of Traditional Chinese Medicine(BATMEN-TCM)database,and kidney deficiency syndrome-related targets were obtained in the Symptom Mapping(SymMap)Database,a traditional Chinese medicine(TCM)syndrome correlation database.The protein–protein interaction(PPI)network was constructed by using the STRING11.5 database.Then,we used CytoScape3.9.0 software to construct the network of TCM–active components–potential targets,and the core TCM components and core targets of Yougui Pill for the treatment of kidney deficiency were obtained.The function analysis of Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)signaling pathway enrichment analysis were performed in the Database for Annotation,Visualization,and Integrated Discovery(DAVID).Finally,preliminary verification was performed with the help of molecular docking technology.Results A total of 147 active components of 9 drugs of Yougui Pill(Fuzi[Aconiti Lateralis Radix Praeparata],Shudihaung[Rehmanniae Radix Praeparata],Gouqi[Lycii Fructus],Shanyao[Rhizoma Dioscoreae],Shanzhuyu[Corni Fructus],Tusizi[Cuscutae Semen],Danggui[Angelicae Sinensis Radix],Duzhong[Eucommiae Cortex],Lujiaojiao[Cervi Cornus Colla])were obtained,corresponding to 233 targets.A total of 2,235targets related to kidney deficiency syndrome and 43 potential therapeutic targets were obtained after the intersection.The core TCM components mainly included quercetin,kaempferol,diosgenin,β-carotene,etc.The core targets involved Trp53(Tp53),Akt1,Pparg,Nr3c1,App,Casp8,Mapk1,Cav1,and Ctnnb1.A total of 27biological processes,10 cellular components,and 11 molecular functions were obtained by gene function enrichment analysis,mainly related to the regulation of gene expression,cell apoptosis and proliferation,and the response to estrogen.A total of 51 KEGG signaling pathways,mainly involving a variety of cancer pathways,apoptosis pathways,longevity regulation pathways,etc.Conclusion Yougui Pill can play a role in preventing and treating kidney deficiency syndrome through multiple targets and pathways. 展开更多

Preliminary Study on the Differentiation of Bone Marrow Mesenchymal Stem Cells into Chondrocytes Induced by TGF-β1, Yougui Yin Drug-Containing Serum and Mechanical Stimulation

Objective: Bone marrow mesenchymal stem cells (BMSCs) have the potential to differentiate into chondrocytes, it is the ideal seed cells for treating cartilage injury. The purpose of this study was to investigate the inducible factors that stimulate the differentiation of bone marrow mesenchymal stem cells into chondrocytes. Methods: Rat BMSCs were isolated and subcultured, and then inoculated on PLGA scaffolds after successfully identificating of BMSCs. They were divided into 5 groups. Blank serum group, TGF-β1 + Yougui Yin drug-containing serum induction group, TGF-β1 + mechanical stimulation induction group, Yougui Yin drug-containing serum + mechanical stimulation induction group, TGF-β1 + Yougui Yin drug-containing serum + mechanical stimulation induction group, 4 weeks later, the adhesion status of cells and scaffolds was observed by Scanning Electron Microscopy, and then chondrocyte induction was performed on different treatment groups. After induction, the expression of Aggrecan and Collagen II was detected by immunofluorescence, and the mRNA expression of Aggrecan and Collagen II was detected by qPCR. Results: BMSCs were identified as rat bone marrow mesenchymal stem cells by immunofluorescence CD34 and CD44 cells. Scanning Electron Microscopy observed that BMSCs grew well on PLGA. Compared with PLGA group, the scaffold space of BMSCS-PLGA composite was significantly reduced, and the growth of cell adhesion scaffold was significantly increased. Compared with the blank serum group, the expression of Aggrecan in TGF-β1 + Yougui Yin drug-containing serum group and Yougui Yin drug-containing serum + mechanical stimulation group was significantly increased. The expression of Collagen II in TGF-β1 + Yougui Yin medicated serum group, Yougui Yin medicated serum + mechanical stimulation group, TGF-β1 + mechanical stimulation group and TGF-β1 + Yougui Yin medicated serum + mechanical stimulation group were significantly increased;PCR results showed that compared with blank serum group, the expression of Aggrecan in other groups was significantly increased, except for Collagen II expression in Yougui Yin drug-containing serum + mechanical stimulation group. Conclusion: BMSCs can differentiate into chondrocytes on PLGA scaffold. Compared with blank serum group, BMSCs can be induced to differentiate into chondrocytes in all induction groups, among all induction groups, TGF-β1 + Yougui-yin drug-containing serum group and TGF-β1 + mechanical stimulation group were more significant, and the difference was statistically significant. 展开更多

OBJECTIVE:To examine,dissect,and understand the molecular mechanisms and combinatorial effects of Zuogui(左归丸,ZGP)and Yougui pills(右归丸,YGP)in 4-vinyl cyclohexene diepoxide(4-VCD)-induced Perimenopausal syndrome(PMS).METHODS:Using the 4-VCD-induced PMS mouse model,uterine and ovary index were measured,and serum sex steroidal hormone levels were evaluated after treatment with ZGP,YGP,ZGP+YGP,estradiol valerate(EV),and Gengnian An(更年安,GNA).Histopathological examinations,ingredient-target network predictions,western blotting,and real-time quantitative polymerase chain reaction(RT-q PCR)analyses were performed to ascertain the possible pharmacological effects and molecular mechanisms of ZYP and YGP.RESULTS:Treatment with ZGP and YGP remarkably improves estrous cyclicity and prevents pathological damage to the uterus.Also,altered sex hormones,including AMH,E2,FSH,LH,P,and T,were restored to normal levels after ZGP and YGP administration.Ingredient-target network analysis showed that the 5 ingredients common to the ZGP and YGP formula modulate 53 targets shared with PMS.Pathwayenrichment analysis further predicted that ZGY and YGP likely regulate of apoptosis and other essential pathways during PMS.In-vivo studies showed that ZGP and YGP suppress PMS via modulating apoptosis through decreasing Caspase-3 and Bcl-2-associated X(Bax)levels and increasing B-cell lymphoma-2(Bcl-2)/Bax and Bcl-2 levels.Importantly,ZGP+YGP treatment modulation effects were somewhat or significantly better compared to ZGP or YGP alone treatment.CONCLUSION:ZGP and YGP represent novel anti-PMS agents whose effects involve restoring altered hormonal levels,protecting the uterus,and regulating apoptosis. 展开更多

Systematic review of Yougui pills combined with levothyroxine sodium in the treatment of hypothyroidism

BACKGROUND Yougui pills have long been used to treat hypothyroidism,usually in combination with levothyroxine sodium in clinical treatment,while their clinical efficacy and safety are still controversial when compared to levothyroxine treatment alone.AIM To explore the clinical efficacy and safety of Yougui pills combined with levothyroxine sodium in the treatment of hypothyroidism.METHODS This meta-analysis was performed in accordance with the PRISMA guidelines.Randomized controlled trials on Yougui pills in the treatment of hypothyroidism published from 2008 to May 2021 were searched in a total of 8 databases(4 databases in Chinese and 4 databases in English).The quality of the included studies was evaluated according to the Cochrane risk assessment tool.Weighted mean difference(WMD)was used for continuous variables,and relative risk(RR)was used for binary variables.Data were extracted,and the meta-analysis was conducted with the statistical software of Stata15.0 and RevMan5.0.RESULTS A total of 140 articles were retrieved,and 9 of them were finally included,with a total sample size of 936 cases.The main meta-analysis results are as follows:(1)The group of Yougui pills combined with levothyroxine sodium had a significantly higher overall response rate than the group of levothyroxine sodium(RR=1.20,95%CI 1.12,1.28,P<0.00001);(2)Yougui pills combined with levothyroxine sodium achieved significantly better efficacy than levothyroxine sodium alone in alleviating adverse symptoms[standard mean difference(SMD)=-1.10,95%CI:-1.37,-0.84,P<0.00001];(3)The level of thyrotropin stimulating hormone in the group of Yougui pills combined with levothyroxine sodium was significantly lower than in the control group of levothyroxine sodium(WMD=-1.38,95%CI:-2.10,-0.67,P=0.00001);(4)The level of free triiodothyronine in the group of Yougui pills combined with levothyroxine sodium was higher than that in the control group of levothyroxine sodium(WMD=0.41,95%CI:0.03,0.79,P=0.03);(5)The level of free thyroxine in the group of Yougui pills combined with levothyroxine sodium was significantly higher than that in the control group of levothyroxine sodium(SMD=0.83,95%CI:0.44,1.22,P≤0.0001);and(6)The adverse reactions in the group of Yougui pills combined with levothyroxine sodium were significantly less than those in the control group of levothyroxine sodium(RR=0.33,95%CI:0.20,-0.53,P<0.00001).CONCLUSION In the treatment of hypothyroidism,the combination of Yougui pills with levothyroxine sodium may be better than levothyroxine sodium treatment alone. 展开更多

Long-term glucocorticoid use may result in sustained suppression of one or more secreted components from the hypothalamo-pituitary-adrenal axis, and often results in apoptosis. Yougui Pill(YGP), a 10-component traditional Chinese herbal medicine, has been shown to be clinically effective for glucocorticoid-induced suppression of the hypothalamo-pituitary-adrenal axis. However, the pharmacological and molecular mechanisms remain unclear. We hypothesized that YGP would exert an anti-apoptosis effect on dexamethasone-treated anterior pituitary cells. In vivo experiments showed that YGP significantly reduced the number of apoptotic cells, down-regulated m RNA expression of cytochrome c, caspase-3, and caspase-9, and up-regulated m RNA expression of Bcl-2. These findings suggest that YGP reduced glucocorticoid-induced apoptosis in rat anterior pituitary cells by regulating the mitochondria-mediated apoptosis pathway. 展开更多

Network pharmacological study of Zuogui pill and Yougui pill for the treatment of postmenopausal osteoporosis

Background:Postmenopausal osteoporosis(PMO)is the most common primary osteoporosis in older women.This condition imposes a huge economic and medical burden on society.Aside from western medicine,traditional Chinese medicine is also widely used for the treatment of PMO,especially in Asian countries.Zuogui pill(ZGP)and Yougui pill(YGP)are classical formulas for the treatment of PMO with potent clinical effects.This study aimed to explore the potential active compounds,potential targets,and potential mechanisms of ZGP and YGP for the treatment of PMO.Methods:Compounds from ZGP and YGP were collected from three online databases,and these compounds were screened by oral bioavailability and drug-likeness.Their corresponding targets were determined from the Medicine Systems Pharmacology Database and Analysis platform.The corresponding targets for PMO were obtained from two disease databases.The target protein-protein interaction was identified through the STRING platforms and the core targets were ascertained by analyzing the protein-protein interaction network by using Cytoscape software.PMO-related genes were identified via Gene Ontology and Kyoto Encyclopedia of Genes and Genomes analyses to identify specific biological processes,cellular components,molecular functions and key signalling pathways of ZGP and YGP for PMO treatment.Results:A total of 68 compounds were screened from ZGP with 168 putative potential target genes,whereas 99 compounds were screened from YGP with 214 potential target genes associated with PMO.Most of the core components included quercetin and kaempferol,and most of the core targets were TP53,AKT1,MAPK1,JUN,TNF,HSP90AA1,APP,IL6,VEGFA,RELA,MAPK8,NR3C1,EGFR,CXCL8,ESR1,FOS and MAPK14.Gene Ontology enrichment and Kyoto Encyclopedia of Genes and Genomes pathway analyses revealed that ZGP and YGP treat PMO primarily via regulation of bone formation and resorption,anti-inflammatory immunity and hormonal regulation.Conclusion:Our data provided insights into the mechanisms,by which ZGP and YGP treat PMO.This study points out a direction for further research into ZGP and YGP monomers and pathways and offers a new clinical treatment option for non-traditional Chinese medicine clinicians in the treatment of PMO. 展开更多

目的分析右归丸、涤痰汤化裁治疗多囊卵巢综合征对内分泌激素代谢和胰岛素抵抗的影响。方法选取2020年3月—2024年3月医院收治的多囊卵巢综合征患者102例为研究对象,51例对照组采用单纯西药治疗,51例观察组在上述基础上予以右归丸、涤... 目的分析右归丸、涤痰汤化裁治疗多囊卵巢综合征对内分泌激素代谢和胰岛素抵抗的影响。方法选取2020年3月—2024年3月医院收治的多囊卵巢综合征患者102例为研究对象,51例对照组采用单纯西药治疗,51例观察组在上述基础上予以右归丸、涤痰汤化裁治疗,对比两组临床效果、性激素指标、卵巢及月经情况、糖代谢指标情况。结果观察组与对照组总有效率对比,98.04%高于78.43%,差异有统计学意义(P<0.05)。治疗后,两组血清睾酮(Serum Testosterone,T)、黄体生成素(Luteinizing Hormone,LH)水平较前降低,雌二醇(Estradiol,E_(2))、泌乳素(Prolactin,PRL)、卵泡刺激素(Follicle-Stimulating Hormone,FSH)以及性激素结合球蛋白(Sex Hormone-Binding Globulin,SHBG)较前升高(P<0.05),且与对照组比较,观察组降低/升高更为显著(P<0.05)。治疗后两组卵巢体积、卵泡数目均减少(P<0.05),且与对照组比较,观察组减少更为显著(P<0.05);观察组月经恢复率96.08%,高于对照组的74.51%(P<0.05);观察组卵巢恢复率96.08%,高于对照组的76.47%(P<0.05)。治疗后两组空腹血糖(Fasting Blood Glucose,FBG)、空腹胰岛素(Fasting Insulin,FINS)、2 h血糖(2-hour Plasma Glucose,2 hPG)、性激素结合球蛋白(Sex Hormone-Binding Globulin,SHBG)水平均有所降低(P<0.05),与对照组比较,观察组FBG、FINS、2 hPG以及胰岛素抵抗指数(HOMA-IR)水平降低更为明显(P<0.05)。结论被诊断为多囊卵巢综合征的患者使用药物、右归丸、涤痰汤化裁联合治疗后,治疗效果显著,可有效改善患者的激素水平,改善其糖代谢指标,促进患者恢复。 展开更多

目的基于中医全息诊疗理论探讨灸法联合右归丸加减治疗肾阳虚体质型更年期妇女亚健康状态的临床效果。方法选择肾阳虚体质型更年期妇女亚健康状态患者142例,依据治疗方法的不同分为对照组与观察组,各71例,对照组给予右归丸加减治疗,观... 目的基于中医全息诊疗理论探讨灸法联合右归丸加减治疗肾阳虚体质型更年期妇女亚健康状态的临床效果。方法选择肾阳虚体质型更年期妇女亚健康状态患者142例,依据治疗方法的不同分为对照组与观察组,各71例,对照组给予右归丸加减治疗,观察组在对照组基础上给予全息经络灸法治疗,2组均持续治疗8周。结果观察组总有效率(97.18%,69/71)高于对照组(83.10%,59/71)(P<0.05)。治疗后,2组中医证候各项评分、血清促卵泡激素(FSH)、促黄体生成素(LH)、脱氧吡啶啉(D-Pyr)、Ⅰ型前胶原氨基端前肽(s-PINP)水平均低于治疗前(P<0.05),观察组低于对照组(P<0.05);2组社会领域、心理领域、生理领域、环境领域各项评分、血清雌二醇(E2)、孕激素(P)、5-羟色胺(5-HT)、γ-氨基丁酸(GABA)水平均高于治疗前(P<0.05),观察组高于对照组(P<0.05);对照组复常率(46.48%,33/71)低于观察组(78.87%,56/71)(P<0.05)。结论基于中医全息诊疗理论灸法联合右归丸加减可稳定肾阳虚体质型更年期妇女亚健康状态患者体内激素水平,有效调节神经体质表达,改善骨代谢水平及临床症状,提高患者生存质量、复常率,临床疗效显著。 展开更多

为探究右归丸(YGP)对过氧化氢(H_(2)O_(2))诱导的人卵巢颗粒细胞株(KGN)氧化损伤的作用及机制。通过超高效液相色谱-四极杆静电场轨道阱质谱(UHPLC-QE-MS)分析给予YGP低、高剂量后SD大鼠的入血成分,选择右归丸高剂量组含药血清作为细胞... 为探究右归丸(YGP)对过氧化氢(H_(2)O_(2))诱导的人卵巢颗粒细胞株(KGN)氧化损伤的作用及机制。通过超高效液相色谱-四极杆静电场轨道阱质谱(UHPLC-QE-MS)分析给予YGP低、高剂量后SD大鼠的入血成分,选择右归丸高剂量组含药血清作为细胞模型的药物。采用200μmol·L^(-1)H_(2)O_(2)刺激KGN,构建氧化损伤模型。将其分为正常组,模型组,YGP低、中、高剂量组,在沉默或过表达Serpina3k基础上进一步验证药效。采用TUNEL染色检测细胞凋亡,酶联免疫吸附测定(ELISA)法检测KGN分泌雌二醇(E_(2))、17β-E_(2)水平,蛋白免疫印迹(Western blot)法检测激肽释放酶结合蛋白(Serpina3k)及Kelch样ECH相关蛋白1(Keap1)/核因子E_(2)相关因子2(Nrf2)信号通路相关蛋白的表达。结果显示,与模型组比较,右归丸各剂量不仅可显著降低颗粒细胞凋亡,上调E_(2)、17β-E_(2)分泌水平,还可显著上调Serpina3k/Nrf2通路。进一步研究发现,Serpina3k过表达后不仅可增强YGP的治疗效应,还增加Nrf2的表达,抑制Keap1的表达;而干扰Serpina3k后,YGP的治疗作用被部分逆转,同时YGP对Keap1/Nrf2信号通路的调控作用也被部分削弱。结果表明,YGP改善氧化应激诱导颗粒细胞损伤的机制可能与其上调Serpina3k表达,影响Keap1/Nrf2信号通路的传导有关。该研究揭示了YGP对颗粒细胞保护的作用机制,为其临床应用提供了一定的理论依据。 展开更多

目的研究二仙汤合右归汤治疗卵巢早衰闭经的临床效果。方法卵巢早衰闭经患者62例,随机数字表法分为A组(31例)与B组(31例)。A组进行序贯疗法,B组联合二仙汤合右归汤治疗,比较两组疗效,中医证候评分、症状自评量表(SCL-90)积分、Kupperma... 目的研究二仙汤合右归汤治疗卵巢早衰闭经的临床效果。方法卵巢早衰闭经患者62例,随机数字表法分为A组(31例)与B组(31例)。A组进行序贯疗法,B组联合二仙汤合右归汤治疗,比较两组疗效,中医证候评分、症状自评量表(SCL-90)积分、Kupperman评分、子宫三径线之和、子宫内膜厚度、外周血磷脂酰肌醇3激酶(PI3K)、丝氨酸苏氨酸蛋白激酶(AKT)、调节性T细胞(Treg)、辅助性T细胞(Th17)、血清抗苗勒管激素(AMH)、抑制素B(INHB)水平,治疗期间不良反应发生情况。结果治疗3个月后,B组总有效率高于A组;两组治疗3个月后中医证候评分、SCL-90积分、Kupperman评分、外周血PI3K、AKT、Th17水平与治疗前比较,降低,与A组比较,B组更低;两组治疗3个月后子宫三径线之和、血清AMH、外周血INHB、Treg水平与治疗前比较,升高,与A组比较,B组更高;两组治疗3个月后子宫内膜厚度与治疗前比较,增厚,与A组比较,B组更厚(P<0.05)。B组与A组治疗期间总不良反应发生率接近(P>0.05)。结论二仙汤合右归汤可帮助调控卵巢早衰闭经患者体内控P13K/AKT信号通路,改善其临床症状、卵巢功能及免疫功能,缓解患者子宫萎缩情况及病情严重程度,降低机体炎症反应,提高患者精神心理健康状况,临床疗效较好,同时不会增加不良反应的发生概率。 展开更多

目的:探究右归丸与金匮肾气丸对肾阳虚小鼠免疫器官和精子质量作用效果的差异。方法:昆明雄性小鼠28只(死亡1只)随机分为4组,其中正常组6只,模型组、金匮组和右归组各7只。除正常组外,其余各组给予氢化可的松灌胃、播放警报音频和强迫游... 目的:探究右归丸与金匮肾气丸对肾阳虚小鼠免疫器官和精子质量作用效果的差异。方法:昆明雄性小鼠28只(死亡1只)随机分为4组,其中正常组6只,模型组、金匮组和右归组各7只。除正常组外,其余各组给予氢化可的松灌胃、播放警报音频和强迫游泳3种方法进行复合建模4 d,同时金匮组(0.02 g/10 g)、右归组(0.018 g/10 g)给予相应药物混悬液灌胃。实验期间每天观察并记录各组小鼠体质量、体温以及精神情况,第5天进行耐力耐寒实验,测定小鼠尾部负重在冰水中游泳存活时间,随后处死,取其胸腺和脾脏计算脏器指数,并取附睾使用血细胞计数板计数法测精子质量。结果:造模给药4 d后,模型组与给药组均出现肾阳虚症状,与正常组比较,模型组小鼠胸腺指数、脾脏指数、精子存活率及个数均降低,差异有统计学意义(P<0.05);与模型组相比,右归组小鼠精子活动度0级、Ⅰ级和Ⅲ级、精子存活率及个数明显升高,差异有统计学意义(P<0.05);与金匮组相比,右归组小鼠精子活动度Ⅰ级和Ⅲ级、精子存活率明显升高,差异有统计学意义(P<0.01);各组小鼠冰水负重游泳死亡时间比较,差异无统计学意义(P>0.05)。结论:右归丸与金匮肾气丸对肾阳虚的治疗均有疗效,但右归丸较金匮肾气丸在机体免疫与精子质量方面的疗效更为突出。 展开更多

Objective:To study the effects of Zuogui Pill(左归丸,ZGP)and Yougui Pill(右归丸,YGP)on the expressions of brain-derived neurotrophic factor(BDNF)and cyclic adenosine monophosphate(cAMP)/protein kinase A(PKA)signaling of axonal regeneration in the Lewis rats with experimental autoimmune encephalomyelitis(EAE),in order to explore the possible mechanism of ZGP and YGP on promoting axonal regeneration.Methods:The rats were randomly divided into normal control(NC),model(MO),prednisone acetate(PA),ZGP and YGP groups.The EAE model of rat was established by injecting antigen containing myelin basic protein(MBP)68-86.The brain and spinal cord were harvested on the 14th and 28th day postimmunization(PI),the protein and mRNA expression of BDNF and PKA in the brain and spinal cord of rats were detected by Western blot analysis and real-time quantitative polymerase chain reaction(PCR),and the cAMP levels were detected by using enzyme-immunoassay method.Results:(1)On the 28th day PI,the mRNA expression of BDNF in brain white matter and spinal cord of rats in ZGP and YGP groups were up-regulated,especially in YGP group(P〈0.05 or P〈0.01).(2)On the 14th day PI,the cAMP levels in brain white matters significantly increased in PA and YGP groups compared with MO group(P〈0.05 or P〈0.01),and the cAMP level in YGP group was higher than that in ZGP group(P〈0.05).The cAMP level in spinal cord also significantly increased in YGP group compared with MO,PA and ZGP groups,respectively(P〈0.01).(3)On the 14th day PI,the PKA expression in spinal cord of rats in ZGP group was significantly decreased compared with MO and YGP groups,respectively(P〈0.05).(4)On the 28th day PI,there was a positive correlation between cAMP and PKA expression in the brain white matter of YGP rats.Conclusions:The results suggest that ZGP and YGP may promote axonal regeneration by modulating cAMP/PKA signal transduction pathway,but the targets of molecular mechanism of ZGP may be different from those of YGP. 展开更多

Objective:To study the molecular mechanism of Zuogui Pill(左归丸,ZGP) and Yougui Pill(右归丸,YGP) on axonal regeneration in rats with experimental autoimmune encephalomyelitis(EAE).Methods: EAE rat model was established by bilateral rear pedes subcutaneous injection of antigen made by mixing myelin basic protein(MBP) and complete Freud's adjuvant(CFA) in the volume ratio of 1:1.The pathological changes of axonal injury and regeneration in the brain and the spinal cord were observed on the 14th(the acute stage) and the 28th day(the remission stage) after modeling,with hematoxylin-eosin(HE) staining,silver stain, and immunohistochemical staining.The rats treated with prednisone acetate were taken as controls.Results: Observation under the light microscope with HE staining showed a sleeve-like change in rats' cerebrospinal parenchyma with inflammatory cell infiltration around the small vessels and neuronic denaturation,while silver staining showed excessive tumefaction and abscission of axon,and immunohistochemical analysis showed decreasing of nerve growth factor(NGF) expression at the acute stage of EAE,which was even more remarkable at the remission stage,showing significant difference as compared with the normal control(P〈0.05). And the expressions of Nogo A,an axon growth inhibitor,and its receptor(Nogo-66 receptor,Ng R) were significantly higher than those in the normal control at the acute stage(P〈0.01).However,after the intervention of ZGP and YGP,the pathological changes and axon damage in rats' brain and spinal cord were much more alleviated,and the NGF expression was significantly higher than that in the model group at the acute stage (P〈0.05).The expression of NGF was even stronger during the remission stage,and a better effect was shown by YGP.As for Nogo A and Ng R expressions,they were significantly lower than those in the model group at the acute stage(P〈0.05),but a better effect was shown by ZGP.Conclusions:ZGP and YGP can prevent axonal injury and promote the axonal regeneration in rats of EAE,and the possible mechanism is to increase the expression of NGF and reduce the expression of Nogo A and its receptor.However,some differences are observed between the two Chinese preparations in their acting times and points,which provides a certain basis for revealing the modern connotation of the Chinese medicine theory on tonifying Shen(肾)-yin and Shen-yang. 展开更多

探讨右归饮治疗糖皮质激素诱导的骨坏死的药理作用及机制。使用地塞米松(20 mg·kg-1,隔日肌肉注射1次,持续8周)建立大鼠糖皮质激素性股骨头坏死(GA-ONFH)模型,将所有大鼠随机分为对照组、模型组以及右归饮低、高剂量(1.5、3.0 g... 探讨右归饮治疗糖皮质激素诱导的骨坏死的药理作用及机制。使用地塞米松(20 mg·kg-1,隔日肌肉注射1次,持续8周)建立大鼠糖皮质激素性股骨头坏死(GA-ONFH)模型,将所有大鼠随机分为对照组、模型组以及右归饮低、高剂量(1.5、3.0 g·kg-1)组。造模结束后用右归饮灌胃干预,随后取股骨标本。使用苏木精-伊红(HE)染色观察大鼠股骨头修复情况,免疫荧光检测股骨头内骨髓间充质干细胞(BMSCs)成脂分化情况。同时,在体外进行地塞米松(1μmol·L^(-1))诱导大鼠BMSCs实验,观察右归饮含药血清对BMSCs成脂分化的影响。动物实验结果显示,与模型组相比,右归饮各剂量可显著改善股骨头内部骨密度(BMD)、骨体积占比(BV/TV)以及骨小梁厚度(Tb.Th),显著减轻GA-ONFH大鼠坏死样改变以及股骨头内脂肪细胞浸润,显著减少股骨头内部过氧化物酶体增殖物激活受体γ(PPARγ)表达水平从而减少BMSCs的成脂分化。体外实验结果显示,右归饮含药血清可显著减轻地塞米松诱导的BMSCs的成脂分化,降低PPARγ表达水平。此外,PPARγ的过表达削弱了右归饮含药血清对BMSCs成脂分化的抑制效果,表明PPARγ在右归饮抑制BMSCs脂肪生成中发挥着关键作用。综上,右归饮可通过降低PPARγ表达水平减轻BMSCs的成脂分化发挥治疗糖皮质激素诱导的骨坏死的作用。 展开更多
