The development of DKD cannot be fully explained by a single mechanism.Traditional Chinese Medicine(TCM) has been applied extensively for the treatment of DKD in China. However, studying the mechanism of DKD using theories and methods that are appropriate for TCM characteristics and searching for theoretical bases for TCM clinical application are topics that still need to be explored and researched. Activation of the transforming growth factor(TGF)-β1/Smad and PI3 K/Akt/mTOR signaling pathways functions as a self-protection mechanism against renal microinflammation in DKD. However, the persistent abnormal overactivation of reactions causes secondary cell dysfunction, cell apoptosis, increased extracellular matrix(ECM) secretion, and eventually renal fibrosis. During this process, the dysregulation of self-balance among a variety of signaling pathways and the loss of self-feedback regulatory mecha-nisms downstream of these signaling pathways are critical causes of the occurrence and development of DKD. TCM may both inhibit the expression or activation of "hyperactive" signaling pathways(NFB, Smad3, and PI3 K/Akt/mTOR) and increase the expression or activation of "deficient" signaling pathways(Smad7 and PTEN) to restore balance to cells with an abnormal pathophysiological status and achieve the goal of DKD treatment.展开更多

The incidence rate of ETM, plasmic ET, serum superoxide dismutase (SOD) activity, malonyldialdehyde (MDA), complement C3 and C-reactive protein (CRP) were studied respectively.RESULTS The ET level of group A (35.92ng/L±8.30ng/L) was significantly reduced after 7 days of treatment (P<0.05) in contrast to that of group B (47.8ng/L±11.62ng/L), so did the level of MDA and CRP. But the SOD activity and C3 level in group A increased significantly (P<0.05).CONCLUSION YCHCQ decoction combined with EST had a beneficial effect for ETM in acute cholangitis.INTRODUCTIONEndotoxemia (ETM) is one of the most important physiopathologic causes of acute cholangitis and it is the trigger of cytokines and inflammatory factors. In recent studies it has been found that Yinchenhao Chengqi (YCHCQ) decoction has a beneficial effect on ETM in acute cholangitis. With the development of endoscopic surgery, endoscopic sphincterotomy (EST) has become an effective replacement for some operations in the treatment of acute cholangitis[1]. The effect of YCHCQ decoction combined with EST on ET, oxygen free radical and complement C3 was observed in order to find out its therapeutic mechanism.展开更多

We defined generalized relations and generalized reasoning, introduced the concept of steady multilateral systems with two non-compatibility relations, and discussed its energy properties. Later based on the intervention principle in image mathematics of TCMath and treated the research object of the image mathematics as a steady multilateral system, it has been proved that the mathematical intervening principle is true. The kernel of this paper is the existence and reasoning of the non-compatibility relations in steady multilateral systems, and it accords with the oriental thinking model.展开更多

文摘Background:Wenqing Yin(WQY)is a classic prescription used to treat skin diseases like atopic dermatitis(AD)in China, and the aim of this study is to investigate the therapeutic effects and molecular mechanisms of WQY on AD.Methods:The DNFB-induced mouse models of AD were established to investigate the therapeutic effects of WQY on AD.The symptoms of AD in the ears and backs of the mice were assessed, while inflammatory factors in the ear were quantified using quantitative real-time-polymerase chain reaction(qRT-PCR),and the percentages of CD4^(+)and CD8^(+)cells in the spleen were analyzed through flow cytometry. The compounds in WQY were identified using ultra-performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS)analysis and the key targets and pathways of WQY to treat AD were predicted by network pharmacology.Subsequently, the key genes were tested and verified by qRT-PCR,and the potential active components and target proteins were verified by molecular docking. Results:WQY relieved the AD symptoms and histopathological injuries in the ear and back skin of mice with AD.Meanwhile,WQY significantly reduced the levels of inflammatory factors IL-6 and IL-1βin ear tissue,as well as the ratio of CD4^(+)/CD8^(+)cells in spleen.Additionally,a total of 142 compounds were identified from the water extract of WQY by UPLC-Orbitrap-MS/MS.39 key targets related to AD were screened out by network pharmacology methods. The KEGG analysis indicated that the effects of WQY were primarily mediated through pathways associated with Toll-like receptor signaling and T cell receptor signaling. Moreover, the results of qRT-PCR demonstrated that WQY significantly reduced the mRNA expressions of IL-4,IL-10,GATA3 and FOXP3,and molecular docking simulation verified that the active components of WQY had excellent binding abilities with IL-4,IL-10,GATA3 and FOXP3 proteins. Conclusion:The present study demonstrated that WQY effectively relieved AD symptoms in mice, decreased the inflammatory factors levels, regulated the balance of CD4^(+)and CD8^(+)cells,and the mechanism may be associated with the suppression of Th2 and Treg cell immune responses.

文摘BACKGROUND Hypertension is a significant global health concern and serves as a critical risk factor for cardiovascular diseases, stroke, and renal failure.Traditional Chinese Medicine(TCM)has been utilized for an extended period to address hypertension, with the syndrome known as Yin deficiency and Yang hyperactivity syndrome(YDYHS)frequently observed in individuals with elevated blood pressure. This syndrome is characterized by symptoms including dizziness, tinnitus, irritability, and insomnia, which are thought to result from an imbalance between Yin,representing the cooling and nourishing aspects, and Yang, denoting the active and warming aspects of the body.Despite the prevalent application of TCM in clinical settings, the metabolic mechanisms underlying the YDYHS in the context of hypertension remain inadequately elucidated.AIM To explore the differential plasma metabolites and associated pathways in hypertensive patients with YDYHS using gas chromatography-mass spectrometry(GC-MS)to elucidate the distinctive blood metabolite pattern in this patient population.METHODS GC-MS was used to analyze plasma samples from 51 hypertensive patients with YDYHS and 20 healthy controls.Chemometric methods, including principal component analysis and partial least squares discriminant analysis, were employed to identify potential biochemical patterns.Simultaneously, the high-quality Kyoto Encyclopedia of Genes and Genomes metabolic pathways database was used to identify associated metabolic pathways.Using variable importance in projection and receiver operating characteristic curve analyses, potential biomarkers were extracted to assess their clinical utility.RESULTS Metabolomic profiling of hypertensive patients with YDYHS identified 20 potential biomarkers(4-hydroxybenzoic acid,pectin,1,2,3-trihydroxybenzene,D-ribose,3-hydroxybutyric acid, quinic acid,L-lysine,oleic acid,2-hydroxybutyric acid, linoleic acid, citric acid,alpha-tocopherol,D-glucuronic acid,glycerol,N-acetyl-L-aspartic acid, betamannosylglycerate, indolelactic acid,L-glutamic acid,D-maltose,L-aspartic acid)and four metabolic pathways(linoleic acid metabolism;alanine, aspartate and glutamate metabolism;D-glutamine and D-glutamate metabolism;arginine biosynthesis). The identified differential metabolites may serve as potential biomarkers for distinguishing hypertensive patients with YDYHS from healthy controls. The area under the curve values ranged from 0.750 to 0.866.Receiver operating characteristic curve analysis showed that these differential metabolites can effectively classify hypertensive patients with YDYHS and healthy individuals.CONCLUSION The metabolomic analysis revealed a distinct blood metabolite pattern in hypertensive patients with YDYHS compared to the healthy control group, highlighting the potential role of the identified 20 biomarkers and four metabolic pathways in these patients. These findings may serve as an important material basis for understanding the occurrence and development of the disease, providing a scientific foundation for future clinical diagnosis.

文摘As an important branch of traditional medicine,Zhuang Medicine is renowned for its unique prescriptions and techniques in clinical treatment, with significant therapeutic effects and widespread popularity. This article introduces Professor Fang Gang’s experience in treating obesity-type polycystic ovary syndrome(PCOS)with modified Zhuang Medicine Mao Xia Yin.Professor Fang Gang proposes that the etiology and pathogenesis of obesity-type polycystic ovary syndrome(PCOS)primarily stem from spleen deficiency, which impairs the transport of water and dampness. This dysfunction leads to the accumulation of dampness that transforms into phlegm.Over time, the stagnation of phlegm and dampness generates internal heat, resulting in the formation of“dampness toxin,”“phlegm toxin,”and“heat toxin.”These pathological toxins obstruct the circulation of Qi and blood, causing stasis and the emergence of“stasis toxin.”The buildup of these toxins ultimately blocks the three channels and two pathways, disrupting the harmony among the three Qi of heaven, earth, and man. This disharmony culminates in impaired uterine gland function. Therefore, the treatment mainly focuses on tonifying deficiency, detoxifying, and regulating Qi.

文摘BACKGROUND Helicobacter pylori(H.pylori)infection is widely considered to be a major risk factor for gastric cancer, contributing to its development through the Correa cascade.Yin Yang 1(YY1)is a transcription factor that acts as a promoter or suppressor of cancer progression. However, the role of YY1 in the inflammatory transformation associated with H.pylori-induced gastric cancer remains unclear.AIM To explore the expression of YY1 in gastric cancer and its impact on cancer progression with H.pylori infection.METHODS H.pylori bacteria were cocultured with GSE1 cells,AGS cells, and SGC7901 cells,as well as in infected and xenograft mouse models.Expression of YY1,members of the Janus kinase 2(JAK2)/signal transducer and activator of transcription 3(STAT3)signaling pathway, and epithelial-mesenchymal transition(EMT)-related proteins in gastric cancer was examined using Western blotting, quantitative realtime polymerase chain reaction, and immunohistochemistry.Survival analysis was performed using the Kaplan-Meier method with the log-rank test. The role of YY1 in gastric cancer cell proliferation was further evaluated through in vitro and in vivo assays.RESULTS YY1 was highly expressed in gastric cancer tissues and cells.Kaplan-Meier survival curves indicated that high YY1 expression correlated with a poor prognosis.YY1 expression showed a gradually increasing trend in H.pyloriinduced gastritis and gastric tumors. In vivo and in vitro experiments demonstrated that H.pylori infection promoted phosphorylation of JAK2 and STAT3,thereby activating the EMT pathway,in which YY1 played a key role.YY1 and JAK2 interaction was validated by chromatin immunoprecipitation.YY1 knockdown or pharmacological inhibition reversed EMT and suppressed gastric cancer cell proliferation and metastasis.CONCLUSION These results suggest that YY1 plays an important role in progression of H.pylori-induced gastric cancer by activating EMT.

文摘The persistence of covalently closed circular DNA(cccDNA)in hepatitis B virus(HBV)-infected hepatocytes remains a major obstacle to effective antiviral treatment.Understanding the molecular mechanisms regulating HBV cccDNA transcription is essential for developing novel therapeutic strategies. In this study,we investigated the role of RNA binding motif protein 25(RBM25)in HBV replication, focusing on its interaction with cccDNA and its regulation of host transcription factors. The results demonstrated that RBM25 knockdown markedly inhibited HBV replication, reducing levels of HBV DNA,hepatitis B e antigen(HBeAg),hepatitis B surface antigen(HBsAg),HBV RNA,and L-HBs in HBV-replicating and infected cell models.Consistent results were observed in a mouse model hydrodynamically injected with 1.2HBV plasmid.Conversely,RBM25 overexpression significantly enhanced HBV replication.Mechanistically,RBM25 promoted HBV promoter activities by binding to cccDNA through its RE/RD and PWI domains. This effect was mediated by increased Yin Yang 1(YY1)expression,which enhanced acetylation of cccDNA-bound histones, promoting HBV transcription.Furthermore,RBM25 expression was upregulated and translocated to the nucleus following core protein expression and accumulation, while overexpression of RBM25 promoted core protein degradation. In conclusion, this study demonstrates that RBM25 is a novel host factor that enhances HBV replication by upregulating YY1-dependent transcriptional activation of cccDNA.It also reveales a reciprocal regulatory mechanism between the HBV core protein and RBM25,which helps sustain HBV replication.

文摘The manuscript Yin Shu(The Book of Pulling),excavated from Zhangjiashan Han Tomb No.247,is the earliest surviving text on therapeutic exercise known as Dao Yin(lit.guiding and pulling).Discovered in 1983,this Dao Yin text, together with the drawings of 44 figures performing“guiding and pulling”exercises found in the Mawangdui Han Tomb in 1974,are of great significance to the study of the early history of Dao Yin.Prior to these discoveries, researchers into Dao Yin relied mainly on material found in the Dao Zang(the Daoist Canon),compiled in 1145.This led to their conclusion that Dao Yin was essentially Daoist. The development of Dao Yin reached its zenith during the Sui Dynasty(581–618 CE),when it became one of the three medical departments at the imperial medical education institution. As part of the medical reform of the second Sui Emperor,Yang Di,Dao Yin became the treatment of choice, and the employment of a large number of Dao Yin specialists to the Sui court transformed the state medical service. The compilation of Zhu Bing Yuan Hou Lun(Treatise on the Origins and Manifestations of Various Diseases)under Yang Di’s decree, incorporated an abundance of resources on Dao Yin,enabling physicians to potentially“prescribe”Dao Yin to their patients.Situating both Yin Shu and Zhu Bing Yuan Hou Lun in their social and historical contexts, this article analyses their editorial treatments, examines their different objectives, styles, and readerships, and compares the various exercises described in the two texts. It emphasizes the fact that over a period of nearly a thousand years, from the late Warring States(475–221 BCE)to the Sui and Tang periods,Dao Yin was an important medical practice, culminating in its institutionalization by the Sui government.

文摘The pathogenic mechanism of diabetic kidney disease(DKD) is complex. TCM may both inhibit the expression or activation of "hyperactive" signaling pathways(NFB, Smad3, and PI3 K/Akt/mTOR) and increase the expression or activation of "deficient" signaling pathways(Smad7 and PTEN) to restore balance to cells with an abnormal pathophysiological status and achieve the goal of DKD treatment.
文摘AIM To evaluate the therapeutic mechanism of Yinchenhao Chengqi (YCHCQ) decoction (containing mainly Herba Artemisia capillaris) combined with endoscopic sphincterotomy (EST) for endotoxemia (ETM) in acute cholangitis.METHODS Twenty-one cases of acute cholangitis with ETM were divided randomly into two groups: group A, 10 patients treated with YCHCQ decoction combined with EST, group B, 11 patients treated with EST. The effect of YCHCQ decoction combined with EST on ET, oxygen free radical and complement C3 was observed in order to find out its therapeutic mechanism.
文摘By using mathematical reasoning, this paper demonstrates the mathematical intervening principle: “Virtual disease is to fill his mother but real disease is to rush down his son” (虚则补其母, 实则泄其子) and “Strong inhibition of the same time, support the weak” (抑强扶弱) based on “Yin Yang Wu Xing” Theory in image mathematics of Traditional Chinese Mathematics (TCMath). The kernel of this paper is the existence and reasoning of the non-compatibility relations in steady multilateral systems, and it accords with the oriental thinking model.
