OBJECTIVE:To compare the efficacy of Tongxie Yaofang(痛泻要方,TXYF)versus pinaverium bromide for the treatment of diarrheal irritable bowel syndrome(IBSD)with liver depression and spleen deficiency.METHODS:A search of...OBJECTIVE:To compare the efficacy of Tongxie Yaofang(痛泻要方,TXYF)versus pinaverium bromide for the treatment of diarrheal irritable bowel syndrome(IBSD)with liver depression and spleen deficiency.METHODS:A search of the China National Knowledge Infrastructure Database,China Science and Technology Journal Database,Wanfang Database,Pub Med,EMBASE,Cochrane Library and Web of Science databases was conducted from database creation to the 28th of September 2022.Meta-analysis was performed using Revman 5.4 software.RESULTS:Twelve randomized controlled trials comprising 1058 patients were included in the analysis.TXYF was more efficacious in improving the clinical symptoms of patients.Comparison of the efficacy rate,Traditional Chinese Medicine Evidence Score(TCMPES),self-rating anxiety scale anxiety score,self-rating depression scale depression score,adverse reaction rate and calcitonin gene-related peptide(CGRP)levels showed that TXYF was significantly superior to pinaverium bromide treatment(P<0.05);there was no statistically significant difference in the plasma vasoactive peptide level between the two groups(P>0.05).Funnel plots of the total efficacy rate and TCM-PES were asymmetrically distributed,suggesting publication bias.The GRADEpro Guideline Development Tool was also used to evaluate the studies,and the evidence rating of the included studies was not high.CONCLUSIONS:Although the results of this study suggest that TXYF may be superior for the treatment of IBS-D with liver depression and spleen deficiency,as compared to pinaverium bromide,definitive conclusions are unable to be drawn at this time due to flaws in the overall design of the included trials.Therefore,more rigorous trials are needed to determine the benefit and safety of TXYF.展开更多
Objective:To predict the mechanism of Tongxie Yaofang in the treatment of colorectal cancer(CRC).Methods:The active compounds and targets of Tongxie Yaofang were screened by the TCMSP database,and the MalaCards databa...Objective:To predict the mechanism of Tongxie Yaofang in the treatment of colorectal cancer(CRC).Methods:The active compounds and targets of Tongxie Yaofang were screened by the TCMSP database,and the MalaCards database used“Colorectal cancer”as the key word to search for CRC targets.After the two database were mapped,the overlapping targets are merged.In David 6.8 database,the GO function and KEGG pathway of intersection targets were analyzed.Cytoscape 3.7.2 was used to construct the networks of“Active compounds-Targets”and“Herbs-Active compounds-CRC-Targets”,and the core compounds and the key targets of Tongxie Yaofang in treating CRC were selected through Network Analysis.THE HUMAN PROTEIN ATLAS database was used to verify the protein expression of key genes.Using AutoDock Vina and Python scripts,the core compounds are interfaced with the key target protein receptors.Results:TCMSP database indicated that there were 34 active compounds and 114 targets of Tongxie Yaofang,while MalaCards database indicated that there were 838 targets closely related to CRC,and 52 cross targets were obtained from the two database.Go enrichment analysis showed that 1410 items were involved in the cross targets(P<0.05),and the biological process(BP)was mainly concentrated in Lipid metabolism,hormone reaction,proliferation and apoptosis of cancer cells;Cell composition(CC)mainly affected the mitochondrial membrane potential and endoplasmic reticulum stress of CRC cells;while Molecular Function(MF)mainly involves oxidoreductase activity,protein kinase activity,etc.KEGG pathway enrichment analysis indicated that 104 signaling pathways were involved in the cross targets(P<0.05)including PI3K/Akt,TNF,TLR and so on.THE HUMAN PROTEIN ATLAS database showed that PTGS2,PTGS1,and ESR1 showed stronger positive protein expression in CRC tissues than normal intestinal tissues.The combination of Wogonin,Kaempferol and beta-sitosterol with PTGS2,PTGS1 and ESR1 were lower than≤−5.0 kJ/mol that showing good affinity.Conclusion:The Tongxie Yaofang in treating CRC involves many signal pathways and biological processes,and the main active components including Wogonin,Kaempferol,beta-sitosterol,and the key targets PTGS2,PTGS1,ESR1,may be one of the important mechanisms.展开更多

Background:Ulcerative colitis(UC)is a diffuse nonspecific intestinal inflammation.Spleen-kidney Yang deficiency combined with liver stagnation is the most common symptom.Sishen Pills-Tongxie Yaofang(SSP-TXYF)is a trad...Background:Ulcerative colitis(UC)is a diffuse nonspecific intestinal inflammation.Spleen-kidney Yang deficiency combined with liver stagnation is the most common symptom.Sishen Pills-Tongxie Yaofang(SSP-TXYF)is a traditional Chinese medicine(TCM)that is widely used in the treatment of this symptom.However,its pharmacological mechanism and active components remain unclear.Objective:This study elucidated the potential mechanism and active components of SSP-TXYF in the treatment of UC from the perspective of TCM syndrome.Methods:Metascape,STRING,and Cytoscape were used to explore the SSP-TXYF-compound-target-UC network and biological enrichment pathways,so as to screen the active compounds,key targets,and pathways of SSP-TXYF.Through the construction of a rat model with UC,the key targets and active components were verified after SSP-TXYF administration.Results:A total of 77 effective active chemical components,208 potential targets,and 5 core target genes were screened out.Gene Ontology biological process items and Kyoto Encyclopedia of Genes and Genomes signaling pathways showed that SSP-TXYF played a role in regulating nerve-endocrine,cell proliferation and apoptosis,and immune-related pathways.The main compounds and the target protein exhibited a good binding ability in molecular docking.The results of animal experiments showed that SSP-TXYF could improve UC through IL-6,AKT1,PTGS2,CASP3,and JUN,and nobiletin and wogonin were identified as the main active components.Conclusions:This study suggests that nobiletin and wogonin are the main components of SSP-TXYF in the treatment of UC,which provides effective therapeutic targets and drugs for future clinical treatment of UC.展开更多
