However, the...Huosu Yangwei(HSYW) Formula is a traditioanl Chinese herbal medicine that has been extensively used to treat chronic atrophic gastritis, precancerous lesions of gastric cancer and advanced gastric cancer. However, the effective compounds of HSYW and its related anti-tumor mechanisms are not completely understood. In the current study, 160 ingredients of HSYW were identified and 64 effective compounds were screened by the ADMET evaluation. Furthermore, 64 effective compounds and 2579 potential targets were mapped based on public databases. Animal experiments demonstrated that HSYW significantly inhibited tumor growth in vivo. Transcriptional profiles revealed that 81 mRNAs were differentially expressed in HSYW-treated N87-bearing Balb/c mice. Network pharmacology and PPI network showed that 12 core genes acted as potential markers to evaluate the curative effects of HSYW. Bioinformatics and qRT-PCR results suggested that HSYW might regulate the mRNA expression of DNAJB4, CALD,AKR1C1, CST1, CASP1, PREX1, SOCS3 and PRDM1 against tumor growth in N87-bearing Balb/c mice.展开更多
Central composite design(CCD),together with multiple linear regression,was successfully used to optimize the electrophoretic buffer system of micellar electrokinetic capillary chromatography(MEKC) for the determinatio...Central composite design(CCD),together with multiple linear regression,was successfully used to optimize the electrophoretic buffer system of micellar electrokinetic capillary chromatography(MEKC) for the determination of albiflorin,paeoniflorin,liquiritin,and glycyrrhizic acid in the traditional Chinese medicine(TCM) prescription,Yangwei granule.Concentrations of sodium deoxycholate(SDC) and borate,and proportions of ammonia,acetonitrile,and methanol were optimized.The total resolutions of peaks between the analytes and their adjacent peaks in real samples were integrated into the evaluation index of separation efficiency.The optimum electrophoretic buffer contained 80 mmol/L SDC,20 mmol/L borate,5%(v/v) methanol,0.5%(v/v) ammonia,and 5%(v/v) acetonitrile.The correlation coefficients(R 2 ) between the peak areas and the corresponding concentrations of analytes were greater than 0.9956.The limits of detection(LODs) (S/N=3) of the analytes were 0.97-4.00μg/ml.The results indicate the superiority of CCD in optimizing the separation conditions of complex samples such as TCM prescriptions.展开更多

Before treatment and after one course treatment, blood samples from vein were obtained, the rosette rate of c3b receptor (RBCC3bRR), tumor and red cell (RRTR) and RBC immune complex (RBCICR) were measured by counting the rosette formed by sensitivated yeast or tumor cell or unsentivated yeast adhering blood cell under microscopy. The T lymphocyte subset was observed by the method of APAAP. Chemotherapy alone was used as controls (n=20). In addition, the dynamic changes of RBCC3bRR, RRTR and RBCICR of mice with tumor after treated by YWKL granules (n=30) were observed. The chemotherapy (n=30) and water (n=30) were used as controls.RESULT Clinical therapeutic effect of combination of YWKL granules and chemotherapy (the treatment group) was markedly superior to that of chemotherapy alone (the control group) (P<001). In the treatment group, the rosette rate of c3b receptor (RBCC3bRR) and the rosette rate of tumor and red cell (RRTR) were significantly increased (P<001) after treatment, the rate of RBC immune complex (RBCICR) was markedly decreased (P<001), and the ratio of CD4 to CD8 was obviously elevated (P<001). CD8 was much lower (P<001), and the ratio of CD4 to CD8 was much higher (P<001), than the control group. The RRTR rate was positively correlated with the ratio of CD4 to CD8. Experiment in mice showed on the ninth day of bearing cancer, the tumor weight in the YWKL granules alone group was much lighter than that of chemotherapy alone group or water alone group respectively, the RBC immune function of YWKL granules alone group was higher than the other two groups, and on the thirteenth day, there were no differences of RBC immune function and tumor weight among the three groups (P<005).CONCLUSION The anticancer mechanism of YWKL granules might lie in enhancing the RBC immunity and T lymphocyte immune function, and RBC immune function is correlated with T lymphocyte immune function.展开更多

Objective:To develop a quality consistency evaluation strategy for traditional Chinese medicine(TCM)granules using sugar free Yangwei Granules as a model drug,and demonstrate the effectiveness of the developed method....Objective:To develop a quality consistency evaluation strategy for traditional Chinese medicine(TCM)granules using sugar free Yangwei Granules as a model drug,and demonstrate the effectiveness of the developed method.Methods:The strategy integrates several methods including,HPLC fingerprint and physical fingerprint methods analyze the similarity in chemical and physical properties of the TCM granule samples.Nearinfrared(NIR)spectroscopy with principal components cluster analysis method is used to monitor normal operating conditions(NOC)samples accurately and to identify different types of abnormal operating conditions(AOC)samples,particularly those that deviate from the normal range.Results:The combined use of HPLC fingerprint and physical fingerprint provides insights into the chemical and physical properties of the samples.NIR spectroscopy,combined with principal components cluster analysis,achieves high accuracy in monitoring NOC samples and identifying AOC samples without misjudgment.The approach proves useful as a complementary method in cases where HPLC fingerprint and physical fingerprint alone lack sufficient resolution.Conclusion:This study establishes the feasibility and utility of the integrated approach for assessing the quality consistency of TCM granules.The strategy shows a high degree of generalization and holds significant importance for enhancing the quality control processes of TCM granules.展开更多
