Dominant intermediate Charcot-Marie-Tooth disease type C（DI-CMTC） is a dominantly inherited neuropathy that has been classified primarily based on motor conduction velocity tests but is now known to involve axonal a...Dominant intermediate Charcot-Marie-Tooth disease type C（DI-CMTC） is a dominantly inherited neuropathy that has been classified primarily based on motor conduction velocity tests but is now known to involve axonal and demyelination features.DI-CMTC is linked to tyrosyl-t RNA synthetase（YARS）-associated neuropathies,which are caused by E196 K and G41 R missense mutations and a single de novo deletion（153-156 del VKQV）.It is well-established that these YARS mutations induce neuronal dysfunction,morphological symptoms involving axonal degeneration,and impaired motor performance.The present study is the first to describe a novel mouse model of YARS-mutation-induced neuropathy involving a neuron-specific promoter with a deleted mitochondrial targeting sequence that inhibits the expression of YARS protein in the mitochondria.An adenovirus vector system and in vivo techniques were utilized to express YARS fusion proteins with a Flag-tag in the spinal cord,peripheral axons,and dorsal root ganglia.Following transfection of YARS-expressing viruses,the distributions of wild-type（WT） YARS and E196 K mutant proteins were compared in all expressed regions; G41 R was not expressed.The proportion of Flag/green fluorescent protein（GFP） double-positive signaling in the E196 K mutant-type mice did not significantly differ from that of WT mice in dorsal root ganglion neurons.All adenovirus genes,and even the empty vector without the YARS gene,exhibited GFP-positive signaling in the ventral horn of the spinal cord because GFP in an adenovirus vector is driven by a cytomegalovirus promoter.The present study demonstrated that anatomical differences in tissue can lead to dissimilar expressions of YARS genes.Thus,use of this novel animal model will provide data regarding distributional defects between mutant and WT genes in neurons,the DICMTC phenotype,and potential treatment approaches for this disease.展开更多

It is emphatically discussed whether there shall be any allowance in general average for losses,damages or expenses incurred in respect of damage to the environment or in consequence of the escape or release of pollutant substances from the property involved in the common maritime adventure prescribed in York-Antwerp Rules 1994. Some amendments about the stated aspects are intentionally suggested in the CMC and Beijing Adjustment Rules of General Axierage.展开更多
This paper is an attempt to document life of male sex workers as transgender in Pakistan and to describe their cultural practices with reference to ritual performs on different occasion. Qualitative research methods c...This paper is an attempt to document life of male sex workers as transgender in Pakistan and to describe their cultural practices with reference to ritual performs on different occasion. Qualitative research methods combing in-depth interviews, FGDs, and observation were employed to gather information. An effort has been made to investigate the cultural life of the transgender and the pattern of their social interaction within the community as well as with the outsiders. Analysis based on thematic description of the themes generated after reviewing data. The research process revealed that males who were engaged in the sex industry have two types of sociological division: first, young and having male identity with potential to transform in transgender;while other transgender plays dual roles: a) within sex worker community as transgender women, b) male outside the transgender community. These males fulfill their socio-psychological and economic needs through entrance in the transgender community. Most of them consider themselves as having feminine soul and masculine body. The male engaging in sex work is a complex phenomenon with reference to their identity and performance of cultural rituals. An exploratory research study needs to be conducted to unfold the notion of dual identity which male sex workers have.展开更多
Objective:This study aimed to determine the risk factors associated with oral polio vaccine(OPV)refusal.Methodology:A case-control study was conducted in Rahim Yar Khan(RYK)District,Pakistan.A case was defined as‘‘a...Objective:This study aimed to determine the risk factors associated with oral polio vaccine(OPV)refusal.Methodology:A case-control study was conducted in Rahim Yar Khan(RYK)District,Pakistan.A case was defined as‘‘any child aged<5 years who was enrolled in the microplans of RYK District as part of the National Immunization Days program in January 2017 and whose parents or guardians refused to receive OPV for these eligible children.”The age-and sex-matched controls(1:1)were obtained from the same locality.Ratios were calculated,and odds ratios(ORs)were determined at 95%confidence interval(CI)and p<0.05.Results:Among the 110 children,64(58%)were male(male to female ratio,1.4:1).The mean age was 29.5 months(range,0.26–60 months).Seventy-four(67.2%)children were living in urban areas and 29(26.3%)children were living in peri-urban and rural areas,while seven(6.3%)were considered nomads.A total of 72(65.2%)children were Punjabi speaking,33(30%)were Saraiki speaking,2(3.7%)were Urdu speaking,and 2(1.82%)were Balochi speaking.Repeated campaigns(n=91,82.7%)was the major reason for OPV refusal.Living at a distance>1 km from a healthcare facility was significantly associated(OR,2.5;95%CI,1.4–4.4;p<0.05)with OPV refusal,while prior visits by healthcare workers(OR,0.11;95%CI,0.06–0.22;p<0.05)and mother’education above primary level(OR,0.42;95%CI,0.24–0.73;p<0.05)were inversely associated with OPV refusal.Conclusion:On the basis of our recommendations,door-to-door social mobilization visits by healthcare workers to alleviate fear,misconception,and fatigue of repeated campaigns and strict accountability mechanism were developed by district government at pre-and intra-campaign levels.展开更多
