目的研究肉桂内生真菌Xylaria sp.DYR-1-4的次级代谢产物及其抗肉桂枝枯病菌的活性。方法采用现代色谱分离手段,包括反相中压层析柱色谱和半制备高效液相方法分离纯化Xylaria sp.DYR-1-4菌株的次级代谢产物;采用现代波谱技术包括紫外光... 目的研究肉桂内生真菌Xylaria sp.DYR-1-4的次级代谢产物及其抗肉桂枝枯病菌的活性。方法采用现代色谱分离手段,包括反相中压层析柱色谱和半制备高效液相方法分离纯化Xylaria sp.DYR-1-4菌株的次级代谢产物;采用现代波谱技术包括紫外光谱(UV)、核磁共振光谱(NMR)、质谱(MS)等技术鉴定其结构;采用微量肉汤稀释法体外测定抗肉桂枝枯病菌的活性。结果从肉桂内生真菌Xylaria sp.DYR-1-4中分离鉴定得11个化合物,分别为Xylariaopyrone I(1)、3-(4-methoxy-2-oxo-2H-pyran-6-yl)propanoic acid(2)、nodulisporipyrones B(3)、nodulisporipyrones C(4)、nodulisporipyrones A(5)、4R,6S,9S-环(D-4-OH-L-Pro-L-Leu)(6)、环(4-OH-Pro-Phe)(7)、5-carboxymellein(8)、(2S)-butylitaconic acid(9)、邻苯二甲酸二丁酯(10)和亚油酸(11)。其中化合物4对肉桂枝枯病菌具有较强抑制作用,IC50值为(3.29±0.11)μg/mL,其他化合物表现出中等强度抑制活性(IC50<60μg/mL)。构效关系分析表明,化合物3、4和5的差别主要集中于侧链上C-8位-OH的存在与否及其相对构型,其抗枝枯病菌活性表现为化合物4>3>5,说明当侧链上C-8位存在-OH时,其抗枝枯病活性更强,且R-OH比S-OH活性更强。结论α-吡喃酮衍生物类化合物可能是炭角菌Xylaria sp.DYR-1-4的抗肉桂枝枯病菌主要特征性成分,这为进一步寻找新型环保的生物农药提供新思路。 展开更多

对海洋真菌Xylaria sp.(No.2508)生产xyloketal B的摇瓶发酵过程进行了初步优化。结果表明,Xylaria sp.(No.2508)在培养120h时xyloketal B产量最高,当培养168 h后菌体进入衰亡期且xyloketal B迅速降解,因此Xylaria sp.(No.2508)生产xylo... 对海洋真菌Xylaria sp.(No.2508)生产xyloketal B的摇瓶发酵过程进行了初步优化。结果表明,Xylaria sp.(No.2508)在培养120h时xyloketal B产量最高,当培养168 h后菌体进入衰亡期且xyloketal B迅速降解,因此Xylaria sp.(No.2508)生产xyloketal B的最适发酵周期为120h;Xylaria sp.(No.2508)合成xyloketal B的过程及用乙酸乙酯萃取xyloketal B的过程对pH非常敏感,其中最适xyloketal B生产的发酵液初始pH为5.7,利用乙酸乙酯萃取xyloketal B时的发酵液最适pH为3.0;最适接种量为10%(υ/υ);接种前发酵液中加入10g/L的大孔树脂XAD-16可以降低某些代谢产物对xyloketal B合成的抑制作用,从而使产量提高2倍左右,而菌体进入稳定期以后加入大孔树脂XAD-16对产物产量没有影响。 展开更多

Two new tetralone derivatives named xylariol A (1) and B (2) were isolated from the EtOAc extract of the culture broth of Xylaria hypoxylon AT-028 (Xylariaceae), and their structures were elucidated by MS, 1R, 1D- and 2D-NMR spectra, and by X-ray diffraction analysis. 1 and 2 showed moderate cytotoxic activities against Hep G2 cells in the in vitro cytotoxic assay with IC50 values of 22.3 and 21.2 μg/mL, respectively. 展开更多

[ Objective ] The paper was to explore the antimicmbial activities of extracts from Xylaria striata via submerged culture. [ Methods ] The antibacterial activities of X. striata extracts against 10 kinds of bacteria were determined using Oxford cup method and mJcroplate assay, and the antifungal activities against nine kinds of pathogenic fungi were determined using mycelial growth rate method. [ Results ] The inhibition zone diameters of the mycelium extract against tested bacte- ria ranged from 13.0 to 17.5 mm, and that of the fermentation broth extract ranged from 10.5 to 12.7 ram. Enterobacter cloacae was the most sensitive to both my- celium extract and fermentation broth extract, with the inhibition zone diameters of 17.1 and 12.7 mm and the minimum inhibitory concentrations of 2.5 and 5 mg/mL, respectively. The inhibitory effects of the mycelium extract and the fermentation broth extract against Fusarium graminearum were the greatest, and the in- hibition rates were 89.88% and 69.53%, respectively. [ Conclusion] X. striata has potential antimicrobial activity and deserves to be further studied. 展开更多

The fungus Xylaria sp.KYJ-15 was isolated from Illigera celebica.Based on the one strain many compounds(OSMAC)strategy,the strain was fermented on potato and rice solid media,respectively.As a result,two novel steroids,xylarsteroids A(1)and B(2),which are the first examples of C28-steroid with an unusualβ-andγ-lactone ring,respectively,along with two new dihydroisocoumarin glycosides,xylarglycosides A(3)and B(4),were identified.Their structures were elucidated by spectroscopic methods,X-ray diffraction and electronic circular dichroism(ECD)experiments.All isolated compounds were evaluated for cytotoxicity,DPPH radical scavenging activity,acetylcholinesterase inhibitory and antimicrobial effect.Compound 1 exhibited potent AChE inhibitory activity with an IC50 value of 2.61±0.05μmol·L^(−1).Theβ-lactone ring unit of 1 is critical for its AChE inhibitory activity.The finding was further confirmed through exploring the interaction of 1 with AChE by molecular docking.In addition,both compounds 1 and 2 exhibited obvious antibacterial activity against Bacillus subtilis with a minimum inhibitory concentration(MIC)of 2μg·mL^(−1).Compounds 3 and 4 exhibited antibacterial activities against Staphylococcus aureus with MICs of 4 and 2μg·mL^(−1),respectively,which also exhibited DPPH radical scavenging activity comparable to the positive control with IC50 values of 9.2±0.03 and 13.3±0.01μmol·L^(−1),respectively. 展开更多

Three previously undescribed cytochalasins,named xylariasins A‒C(1‒3),together with six known ones(4‒9)were iso-lated from Xylaria sp.CFL5,an endophytic fungus of Cephalotaxus fortunei.The chemical structures of all new compounds were elucidated on the basis of extensive spectroscopic data analyses and electronic circular dichroism calculation,as well as optical rotation calculation.Biological activities of compounds 1,4‒9 were evaluated,including cytotoxic,LAG3/MHC II binding inhibition and LAG3/FGL1 binding inhibition activities.Compounds 6 and 9 possessed cytotoxicity against AGS cells at 5μM,with inhibition rates of 94%and 64%,respectively.In addition,all tested isolates,except compound 6,exhibited obvious inhibitory activity against the interaction of both LAG3/MHC II and LAG3/FGL1.Compounds 1,5,7,and 8 inhibited LAG3/MHC II with IC50 values ranging from 2.37 to 4.74μM.Meanwhile,the IC50 values of compounds 1,7,and 8 against LAG3/FGL1 were 11.78,4.39,and 7.45μM,respectively. 展开更多

Bioactive compounds from the wood-decay fungus Xylaria cf.longipes SWUF08-81,cultivated in three different culture media(GM,YM and PDB),were isolated.Their structures and stereochemistry were deduced from spectroscopic and MS data analysis,together with quantum chemical calculations of 13C NMR chemical shifts and electronic circular dichroism(ECD)spectra.Five undescribed polyketides including dibenzofuran(1),mellein(2),dihydroisocoumarin(15),and two pyrans(16,17),together with twenty-three compounds were determined.Compounds 18 and 20 were significantly toxic against cancer cell lines(HCT116,HT29,MCF-7 and HeLa)based on the MTT assay.Quantification by HPLC showed that 18 was produced three-fold higher in the broth of PDB than YM.These studies showed that the production of different compounds were primarily dependent on nutrition sources and it has given a starting point for the growth optimization conditions for the scaling up of bioactive compounds production. 展开更多

Two new cytochalasans,xylariaides A(1)and B(2),were isolated and identified from a soil fungus Xylaria sp.Y01.Their structures were unambiguously determined by extensive spectroscopic methods including high resolution electro-spray ionization mass spectrometry,ultraviolet radiation,infrared spectroscopy,and 1D/2D NMR,as well as in-depth quantum chemical calculations of gauge-including atomic orbital(GIAO)^(13)C NMR chemical shifts,electronic circular dichroism(ECD),and spin-spin coupling constants.The unprecedented core structure with a 5/6/5/3 fused tetracyclic ring system further enriches the scaffold types of cytochalasans. 展开更多

Owing to their potential applications,as well as their structural diversity,the discovery of novel secondary metabolites from insect-associated fungi has been of interest to researchers in recent years.The aim of this study was therefore to estimate the diversity of fungi associated with fungus-growing termites and bioprospecting these for potential secondary metabolites.In total,18 fungal species were isolated and described from the gut and comb of Macrotermes barneyi based on 18S ribosomal DNA gene sequence analysis.Antimicrobial activity assays were carried out on all the known fungi,and nine isolates were recorded as active against pathogenic fungi.Xylaria escharoidea,the best performing isolate,was grown at laboratory scale and 4,8-dihydroxy-3,4-dihydronaphthalen-l(2H)was isolated and characterized.The minimum inhibitory concentration of this isolated compound against tested pathogenic organisms was found to be 6.25 fig.In addition,molecular docking studies have revealed that 4,8-dihydroxy-3,4-dihydronaphthalen-l(2H)is a prominent antibacterial agent with a marked interaction with key residues on protein A(agrAc)that regulates the accessory gene.The findings of this study support the drug discovery of antimicrobial properties in insect-associated fungi,which may lead to novel secondary metabolites. 展开更多

Objective:To isolate,purify and characterize extracellular protease produced by Xylaria psidii(X.psidii)KT30.Methods:In the present study,the extracellular protease secreted by X.psidii KT30 was isolated and purified by using three steps of protein purification,then the purified protease was characterized by applying qualitative and quantitative enzymatic assays.Results:Extracellular protease with molecular mass 71 kDa has been purified successfully by applying diethylaminoethanol-Sepharose followed by sephadex SG75 with its final specific protease activity of 0.091 IU/mg.Protease was the most active at temperature 60℃ and pH 7.The activity of enzyme was abolished mostly by phenylmethanesulfonyl fluoride,showing it is family of serine protease.Conclusions:Extracellular serine protease produced by X.psidii KT30 with good biochemical properties displayed some promising results for its further application in field of biotechnology or medicine. 展开更多
