Methods: From the Traditional Chinese Medicine Systems Pharmacology da...Objective: To explore the molecular mechanism of Magnoliae Flos and Xanthii Fructus herb pair for allergic rhinitis based on network pharmacology. Methods: From the Traditional Chinese Medicine Systems Pharmacology database, Uniprot database, and Gene Cards database, the relevant chemical constituents information, pharmacokinetic information and hub target of allergic rhinitis were obtained. The protein-protein interaction network was constructed by STRING online database, analyzed and showed by the Cytoscape software. The screened target information was analyzed by the Metascape database for Gene Ontology biological function and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analysis. Results: Main components of Magnoliae Flos and Xanthii Fructus herb pair, such as aloe-emodin, stigmasterol, beta-sitosterol and Yangambin, depend on the interaction of Nitric Oxide Synthase 3, Tumor Necrosis Factor, Caspase-3 and other functions involve G protein-coupled (amine) receptor activity, RNA polymerase II basic transcription factor binding, protease binding, heme binding, and integrin binding;can regulate calcium signal pathway, serotonergic synapse, Kyoto Encyclopedia of Genes and Genomes signal pathway, tryptophan inflammatory mediator regulation pathway, estrogen signal pathway alone or in combination, and play a role in the treatment of allergic rhinitis. Conclusion: Magnoliae Flos combined with Xanthii Fructus can regulate biomolecular network in multiple targets and pathways to treat allergic rhinitis.展开更多

Method: A549 human lung cancer cells were cultured and tr...Objective: To investigate the influence of Xanthii Fructuson the expression of small non-coding RNAs(snc RNA) and the malignant behaviors of lung cancer cells. Method: A549 human lung cancer cells were cultured and treated with different concentrations(5-15 mg/ml) of the ethanol extract of Xanthii Fructus. The expression of mi R-21 and pi RNA55490 was analyzed by Realtime PCR. The proliferation capacity was detected by CCK-8 assay. The anchorage-independent growth capacity was measured by soft agar colony formation experiment. The cell invasion capacity was determined by Matrigel assay. Results: The Xanthii Fructus alcohol extract down-regulated the expression of the oncogenic snc RNAmi R-21, and up-regulated the expression of the tumor suppressive snc RNA pi RNA55490. At the functional level, proliferation, anchorage-independent growth, and invasion capacities of the lung cancer cells were strongly inhibited by the extract treatment. Conclusion: The Chinese herb Xanthii Fructus inhibits the malignant behaviors of lung cancer cells, and the inhibition is largely associated with its ability to change the expression of snc RNAs.展开更多
目的建立高效毛细管电泳法(HPCE)同时测定苍耳类药材中绿原酸、咖啡酸、阿魏酸、异绿原酸A、异绿原酸C、新绿原酸和1,3-O-二咖啡酰奎宁酸含量的方法。方法以50 mmol·L^-1硼砂-水(p H 9.54)为缓冲溶液,未涂渍标准熔融石英毛细管...目的建立高效毛细管电泳法(HPCE)同时测定苍耳类药材中绿原酸、咖啡酸、阿魏酸、异绿原酸A、异绿原酸C、新绿原酸和1,3-O-二咖啡酰奎宁酸含量的方法。方法以50 mmol·L^-1硼砂-水(p H 9.54)为缓冲溶液,未涂渍标准熔融石英毛细管(75μm×64.5 cm,有效长度56 cm)为分离通道,运行电压为25 k V,检测波长为326 nm,毛细管温度为25℃,压力进样为25 k Pa×5 s。结果 7种酚酸的浓度与峰面积的线性关系良好(r〉0.999 4);加样回收率为99.85%-102.70%。用此法测定了苍耳类药材中7种酚酸的含量,得到满意结果。结论该方法简单、准确,重复性较好,可用于苍耳类药材内在质量的评价和控制。展开更多
