With a great difference in therapeutic effects of Mahuang(MH, the stems of Ephedra sinica) and Mahuanggen(MHG, the roots of Ephedra sinica), chemical differences between MH and MHG should be investigated. In the present study, gas chromatography-mass spectrometry(GC-MS)-based plant metabolomics was employed to compare volatile oil profiles of MH and MHG. The antioxidant activities of volatile oils from MH and MHG were also compared. 32 differential chemical markers were identified according to the variable importance in the projection(VIP) value of orthogonal partial least squares discriminant analysis(OPLS-DA) and P value of Mann-Whitney test. Among them, chemical markers of tetramethylpyrazine(TMP) and α-terpineol were quantified. Their contents were much higher in most MH samples compared with MHG. The antioxidant assay demonstrated that MH had significantly higher free radical-scavenging activity than MHG. Although MH and MHG derived from the same medicinal plant, there was much difference in their volatile oil profiles. MH samples had significantly higher content of two reported pharmacologically important chemical markers of TMP and α-terpineol, which may account for their different antioxidant activities. 展开更多

Rosa x alba is cultivated almost exclusively for the production of aromatic water and fruits widely used as ingredients in some traditional food preparations sold commercially. Results of the proximate and elemental analyses revealed that flowers exhibited higher moisture content while the dry matter content was higher in leaves.Total fat and ash was higher in fruits when compared with leaves and flowers. All studied organs were found to be rich in essential mineral such as K,Ca,P and Mg.Qualitative and quantitative analysis of the volatile oils revealed different composition patterns between organs.Linalool and geraniol were found as major constituents of the leaf oil whereas 2-phenethyl alcohol and eugenol were the major constituents of the floral oil.In contrast, fruits showed very distinct composition and alkanes/alkenes were found to have the major contribution. The present composition could justify the traditional use of Rosa x alba which could be considered as a potential source of essential minerals and volatile constituents. 展开更多

The BZ oilfield in the Bohai Sea is a rare, highly volatile reservoir with fractures in the metamorphic rocks of buried hills.Clarifying the mechanism of gas injection for improving oil recovery and determining the optimal way to deploy injection-production well networks are critical issues that must be urgently addressed for efficient oilfield development.Experimental research on the mixed-phase displacement mechanism through gas injection into indoor formation fluids was conducted to guide the efficient development of gas injection in oil fields. We established a model of dual-medium reservoir composition and researched the deployment strategy for a three-dimensional well network for gas injection development. The coupling relationship between key influencing factors of the well network and fracture development was also quantitatively analyzed. The results show that the solubility of the associated gas and strong volatile oil system injected into the BZ oilfield is high. This high solubility demonstrates a mixed-phase displacement mechanism involving intermediate hydrocarbons, dissolution and condensation of medium components, and coexistence of extraction processes.Injecting gas and crude oil can achieve a favorable mixing effect when the local formation pressure is greater than 35.79 MPa.Associated gas reinjection is recommended to supplement energy for developing the highly volatile oil reservoirs in the fractured buried hills of the BZ oilfield. This recommendation involves fully utilizing the structural position and gravity-assisted oil displacement mechanism to deploy an injection-production well network.Gas injection points should be constructed at the top of high areas, and oil production points should be placed at the middle and lower parts of low areas. This approach forms a spatial threedimensional well network.By adopting high inclination well development, the oil production well forms a 45°angle with the fracture direction, which increases the drainage area and enhances single-well production capacity. The optimal injection-production well spacing along the fracture direction is approximately 1000 m,while the reasonable well spacing in the vertical fracture direction is approximately 800 m.The research results were applied to the development practice of the buried hills in the BZ oilfield, which achieved favorable development results. These outcomes provide a valuable reference for the formulation of development plans and efficient gas injection development in similar oil and gas fields in buried hills. 展开更多

This study aimed to evaluate the antidepressant effects of the combined administration of total saikosaponins(SSA)and volatile oil of Cyperi Rhizoma(VO)using a mouse depression model induced by chronic restraint plus mild stress(CRMS),and to compare the effects with the traditional antidepressant fluoxetine.Male Kunming mice were subjected to 14-day CRMS modeling and then randomly divided into four groups:the combined treatment group(intraperitoneal injection of SSA 3.5 mg·kg^(-1)+VO 35 mg·kg^(-1)),the fluoxetine treatment group(20 mg·kg^(-1)),the normal saline treatment group, and the non-model group.Drugs were administered continuously for 14 days.Depressive-like behaviors were assessed using the Forced Swimming Test(FST),Tail Suspension Test(TST),and Open-Field Test(OFT).The results showed that the absolute immobility time of mice in the CRMS model group was significantly prolonged in FST and TST.Combined administration of SSA and VO significantly improved depressive-like behaviors, restoring the absolute immobility time in FST and TST to levels close to the control group, with efficacy comparable to fluoxetine. This study confirms that the combination of SSA and VO exhibits antidepressant effects equivalent to fluoxetine in the CRMS model, providing experimental evidence for the further clinical development of this traditional Chinese medicine(TCM)compatibility. 展开更多

Microcapsules of volatile oil containing Herba Schizonepetae(VOHS) were prepared by emulsion solvent diffusion method to improve the drug loading and reduce the amount of pharmaceutical excipients.Orthogonal assay was applied to optimize the preparation condition of microcapsulation, and the results illustrated that the ratio of ethyl cellulose(EC) to VOHS influenced the property of VOHS microcapsule significantly.GC-MS analysis indicated that some volatile components with low concentration in VOHS were lost after microencapsulation. The microcapsules prepared with optimum condition had good fluidity, and the holes on the surface of the microcapsules contributed to the release of VOHS.The particles of the microcapsule conformed to a normal distribution with the diameter of 45—220 μm.In the simulated intestinal fluid containing 0.2% sodium dodecyl sulfate, pulegone in VOHS microcapsule showed a certain degree of slow release.Compared with β-cyclodextrin method, the microencapsulation used in the present work could reduce the amount of excipients and increase the drug loading. It was beneficial to reduce the dose of Chinese medicines containing volatile oils. 展开更多

[Objective] This study was to explore the effect of three precursors on the accumulation of principal volatile oil constituents in tissue culture plantlets of Atractylodes lancea (Thunb.) DC. with the aim to provide references for the improvement of artificial cultivated A. lancea quality. [Method] Three precursors were added into the MS rooting medium for A. lancea tissue culture plantlets and the volatile oil was extracted by ultrasonication after cultured for several days. The content of atractylon, atractylol, β-eucalyptol and atractydin in the volatile oil were determined by using gas chromatography method. [Result] The addition of xylose, isoprene and tetrahydrofuran impacted the growth indicators, yield of volatile oil and relative percentage content of the four constituents of A. lancea tissue culture plantlets. In the 6 g/L xylose optimized medium, the atractylon and β-eucalyptol content reached up to 4.23% and 56.34%, respectively, 1.41% and 1.66% higher than the control; although the addition of isoprene into medium raised the atractylon content, the accumulation of total volatile oil was inhibited that it decreased by 23.67%, 31.06% and 7.10% to the control; for the tetrahydrofuran optimized medium, the content of atractylon, atractylol and atractydin all increased, and the total volatile oil content increased by 49.97% to the control when the concentration of tetrahydrofuran was 0.07 g/L. [Conclusion] The addition of xylose and tetrahydrofuran promoted the accumulation of principal constituents of the volatile oil, whereas the addition of isoprene inhibited the accumulation. 展开更多

A study was conducted to explore the mechanism that emissions of volatile organic compounds(VOC) from heated needles and twigs(200°C,within 15 min) of Pinus pumila affect fire behaviours using the technology of Thermal Desorption-Gas Chromatography-Mass Spectrometry(TD-GC-MS).The results indicated that the main components of VOC from heated needles and twigs are terpenoids. Most of these terpenoids are monoterpenes.Terpenoids account for 72.93% for the needles and 92.40% for the twigs of the total VOC,and their emission ratios are 61.200 μg·g-1 and 217.060 μg·g-1 respectively.Heated twigs can emit more terpenoids than heated needles because twigs had more volatile oils than needles. In actual fires, these large amounts of terpenoid emissions, especially the monoterpene emissions, have strong effects on fire behaviors that are not only in the initial stage but also in the fast propagation stage of fires. These flammable gases are capable of causing violent combustion and creating crown fires. In addition,if these gases accumulate in an uneven geographical area, there will be a possible for eruptive fires and/or fires flashover to occur. 展开更多

Active volatile components in drug pair(DP)Herba Ephedrae-Ramulus Cinnamomi(HE-RC),single drug HE and RC were analyzed by gas chromatography/mass spectrometry(GC/MS),chemometric resolution method(CRM)and overall volume integration.By means of CRM,the two-dimensional data obtained from GC-MS instruments were resolved into a pure chromatogram and a mass spectrum of each chemical compound. In total,97,62,and 78 volatile chemical components in volatile oil of HE,RC,and DP HE-RC,were respectively determined qualitatively and quantitatively, accounting for 90.08%,91.62%,and 89.76% total contents of volatile oil of HE,RC,and DP HE-RC respectively. It is further demonstrated that the numbers of volatile components of DP HE-RC are almost the sum of those of two single drugs, but some relative contents of them are changed. Some new components, such as 1,6-dimethylhepta-1,3,5-triene,tetracyclo[4.2.1.1(2,5).0(9,10)]deca-3,7-diene,globulol and(E,E)-6,10,14-trimethyl-5,9,13-pentadecatrien-2-one are found in DP HE-RC because of chemical reactions and physical changes during decoction. 展开更多

OBJECTIVE: Ulcerative colitis is a chronically recurrent inflammatory bowel disease of unknown origin. In the present study, the effect of ginger (rhizome of Zingiber officinale Roscoe) volatile oil on a rat model of colitis was evaluated. METHODS: Volatile oil of ginger with doses of 100, 200, and 400 mg/kg, prednisolone (4 mg/kg), or vehicle were administered orally to groups of male Wistar rats (n = 6) for 5 d. Animals were randomly divided into 6 groups, each group consisting of 6 rats. Colitis was induced by intracolonic instillation of 2 mL of 4% (v/v) acetic acid solution. All rats were sacrificed 24 h later and the tissue injuries were assessed macroscopically and histopathologically. RESULTS: Ginger volatile oil with all doses reduced colon weight/length ratio (P 〈 0.01) and the effects were similar to the reference drugs. Higher oral doses of volatile oil (200 and 400 mg/kg) reduced ulcer severity (P 〈 0.05 and P 〈 0.01), ulcer area (P 〈 0.01) and ulcer index (P 〈 0.01). On the other hand, evaluation of microscopic scores showed that the dose of 400 mg/kg of volatile oil was effective to reduce inflammation severity (P 〈 0.01) and inflammation extent (P 〈 0.05) compared to the control group. CONCLUSION: It is concluded that ginger volatile oil could effectively reduce symptoms of experimental colitis in a dose-dependent manner. 展开更多

This study was designed to investigate the anti-inflammatory effects of volatile oil of Platycladus orientalis(L.)Franco leaves(VOPF)and the underlying molecular mechanisms by using the non-infectious inflammation rat models and infectious inflammation mouse models.Ear swelling and intraperitoneal capillary permeability in mice, and carrageenan-induced toe swelling and cotton ball-induced granuloma in rats were used to reveal anti-inflammatory effects of VOPF.Moreover, the lipopolysaccharide(LPS)-induced mouse model of acute lung injury was used to explore the anti-inflammatory mechanism of VOPF.The results showed that VOPF could significantly inhibit auricular swelling, intraperitoneal capillary permeability in mice, and reduce granuloma swelling and paw swelling in rats.Furthermore,it significantly alleviated the pathological damage of the lung tissue. In addition,VOPF could reduce the contents of IL-1β and TNF-αand increase the content of IL-10 in the serum. It had little effect on the expression of p65 but reduced the phosphorylation level of p65 and IκB in NF-κB pathway. In conclusion,VOPF has anti-inflammatory effects and the mechanisms involve the down-regulation of the phosphorylation levels of p65 and IκB and blockage of the NF-κB signaling pathway. 展开更多

OBJECTIVE: To analyze the essential oils from flowers, leaves, stems, roots, and fruits of Chaihu(Radix Bupleuri Chinensis).METHODS: We extracted essential oils from different parts of Chaihu(Radix Bupleuri Chinensis) using a steam distillation method. The essential oils were analyzed by gas chromatography-mass spectrometry(GC-MS). Data were collected in full scan mode(m/z 60-600). Volatile components were identified based on their retention indices and by comparing their mass spectra with those in the National Institute of Standards and Technology 2005 database assisted by tandem mass spectrometry information. The relative content of each constituent wasdetermined by area normalization.RESULTS: We identified 111 components, of which12 were common to all 5 parts, 30 were found only in roots, 14 were found only in flowers, 6 were found only in leaves, 4 were found only in stems, and 17 were found only in fruits.CONCLUSION: Our results show that the stems, flowers, leaves, and fruits of Chaihu(Radix Bupleuri Chinensis)contain a high concentration of essential oils, and that the exact composition of the essential oils differs among the plant parts. To develop new medicines and make full use of the Chaihu(Radix Bupleuri Chinensis)resource, it is important to characterize the essential oils from different parts of the plant. In future research, it will be important to determine the pharmacological effects of the various components and the essential oil mixtures. 展开更多

Objective To explore the pharmacodynamic material basis and mechanism of action of volatile oil from Chuanxiong(Chuanxiong Rhizoma)-Suhexiang(Styrax)-Bingpian(Borneolum)(hereinafter referred to as C-S-B volatile oil)in treating angina pectoris based on network pharmacology and to detect its protective effects against rat myocardial damage. Methods Gas chromatography-mass spectrometry(GC-MS)was used to determine the constituents of volatile oils from Chuanxiong(Chuanxiong Rhizoma),Suhexiang(Styrax),and Bingpian(Borneolum),and the targets of the three main constituents were found predicted and screened using the PharmMapper server, and Gene Cards and Coo LGe N databases. The STRING database and Cytoscape software were used to draw the protein-protein interaction(PPI)network,RStudio software was used to analyze Gene Ontology(GO)and Kyoto Encyclopedia of Genome and Genome(KEGG)pathways, and Cytoscape software was used to construct the component-target-pathwaydisease network. The rat model of myocardial injury was established by intraperitoneal injection of a large dose of isoprenaline hydrochloride. After continuous intervention with C-S-B volatile oil for 14 d,the ejection fraction(EF)and short axis shortening rate(FS)of the left ventricle were detected. The indices of myocardial damage were detected after hematoxylin-eosin(HE)staining.Results Fifteen volatile oil components from the C-S-B formula were identified. There are 470 targets of these volatile oil components and 401 angina-related genes. There are 28 core targets, including CHRM4,ADRA1 A,FGFR1,CHRM2,CYP2 A6,CHRM5,CHRM1,CHRM3,HDAC2,and MPO,etc..The results of the KEGG analysis indicated that the C-S-B formula probably interferes with the following pathways:neuroactive ligand-receptor interactions, calcium signaling, cytochrome P450 metabolism of heteropoietin, among others. The results of animal experiments showed that the C-S-B formula essential oil could significantly improve the following myocardial indices in rats with myocardial injury:EF,FS,left ventricular end-systolic diameter(LVIDs),left ventricular end-diastolic diameter(LVIDd),and stroke volume(SV),and all the differences were statistically significant(P<0.01).Conclusion The mechanism of action of volatile oil components in the C-S-B formula in treating angina pectoris was analyzed using multi-component,multi-target and multi-pathway systems, which has laid a foundation for further revealing its mechanism of action.Animal experiments have shown that the volatile oil of the C-S-B formula can improve EF,FS,and other indices of myocardial damage in a rat model, thus relieving myocardial damage caused by heart hyperactivity, improving cardiac function, and protecting against myocardial damage. 展开更多
