[Objectives]To explore the mechanism of Mongolian medicine Valeriana officinalis L.on liver cancer by network pharmacology.[Methods]The HERB database of V.officinalis L.was searched,and the Uniprot database was used t...[Objectives]To explore the mechanism of Mongolian medicine Valeriana officinalis L.on liver cancer by network pharmacology.[Methods]The HERB database of V.officinalis L.was searched,and the Uniprot database was used to normalize and standardize the targets.Liver cancer targets were searched through GeneCards,DISGENET,and other databases.Venny website was used to obtain the intersection target of valerian active ingredients and liver cancer disease.The protein-protein interaction(PPI)network of the intersection targets was analyzed by STRING database,and the PPI network was constructed by Cytoscape software.The David database was used for GO functional annotation and KEGG pathway enrichment analysis to obtain the relevant pathways in the treatment of liver cancer with Mongolian medicine V.officinalis.The corresponding chemical components,targets and pathways of liver cancer were imported into Cytoscape software to construct the network topology of"chemical component-disease-target-pathway".According to the analysis results,the potential of the active components in V.officinalis as a therapeutic drug for liver cancer was evaluated,and the correlation between the results of network pharmacology analysis and clinical treatment of liver cancer was discussed,which provided a reference for clinical application.[Results]A total of 13 kinds of chemical components and 108 drug disease intersection target genes were screened,and the core genes acting on diseases were caffeic acid,perillyl acetate,(+)-alpha-Terpineol,eucalyptol,etc.;GO functional enrichment analysis involved 389 items of biological processes,62 items of cellular components and 120 items of molecular functions.Enrichment analysis of KEGG signaling pathways screened out chemical carcinogenesis-receptor activation,cancer pathways,prolactin signaling pathways,proteoglycans in cancer,EGFR tyrosine kinase inhibitor resistance and other signaling pathways.[Conclusions]The mechanism of Mongolian medicine V.officinalis on liver cancer was studied by network pharmacology.It was found that it can inhibit the proliferation of liver cancer cells from multiple targets and pathways.This is expected to provide a theoretical basis for further basic experimental research.展开更多
Valeriana officinalis L.is a plant from the Caprifoliaceae family,which is widely distributed in various parts of the world,especially in Europe and Asia.All species of Valeriana are distinguished by their ability to s...Valeriana officinalis L.is a plant from the Caprifoliaceae family,which is widely distributed in various parts of the world,especially in Europe and Asia.All species of Valeriana are distinguished by their ability to synthesize essen-tial oil,which has a powerful effect on the physiological and mental aspects of the human body.The aim was to study the qualitative and quantitative composition of essential oil from valerian roots,collected in different coun-tries,using the gas chromatography method,and to establish marker compounds for valerian species.13 samples of commercial roots with rhizomes of V.officinalis from nine countries of the world and a sample of Valeriana pratensis and Valeriana stolonifera growing in Ukraine were selected for the study.The essential oil was obtained from dried valerian roots by the distillation method described in the European Pharmacopoeia.To determine the component composition of the essential oils of the selected samples,the methods of gas chromatography with a mass spectrometric detector and capillary gas chromatography were used.The commercial samples of V.offici-nalis from different countries yielded 0.21%–1.03%of essential oil.Only six of 13 samples contained essential oil in an amount that satisfies the requirement of the European Pharmacopoeia standard(not less than 4 mL/kg).150 compounds were identified in the essential oils of 13 samples of V.officinalis essential oils.The range and average content and coefficients of variation of the identified compounds were determined.The principal com-pounds of V.officinalis essential oils were bornyl acetate(1.6%–27.1%),valeranone(0.5%–17.9%),valerenal(0%–14.7%),camphene(0%–14.6%),α-fenchene(0%–10.6%),and valerenic acid(0%–8.5%).The samples of V.pratensis and V.stolonifera yielded rather high levels of essential oil(1.18%and 0.93%,respectively).Three chemotypes of V.officinalis samples were determined-bornyl acetate/valerenal,valeranone,and isovaleric acid.The composition of the three essential oils compared was rather similar.Based on the study results,we propose the following marker compounds for V.officinalis consistently present in all 16 examined samples:bornyl acetate(1.6%–27.1%),limonene(0.2–2.3),and valeranone(0.5%–17.9%).The study of samples from Ukraine indicates the prospects of using these species of the genus Valeriana with the aim of expanding the raw material base and creating potential herbal preparations with a sedative effect,which are extremely necessary for the population of the country in the war and post-war periods.展开更多

Previous studies have shown that Valeriana jatamansi Jones extract exhibits clear anxiolytic effects, but it is unclear about the mechanism underlying regulation of the HPA axis dysfunction in these anxiolytic effects. OBJECTIVE: To observe the effects of Valeriana jatamansi Jones (Zhizhu Xiang) extract on HPA axis function in a rat model of anxiety, and to compare these effects with positive control estazolam. DESIGN, TIME AND SETTING: Randomized, controlled, animal experiment was performed at Chengdu University of Traditional Chinese Medicine, China, between February and September in 2006. MATERIALS: Estazolam was purchased from Shanghai Jiufu Pharmaceutical, China; Valeriana jatamansiJones was purchased from the Lotus Pond Market for Chinese Herbal Medicine in Chengdu. It consisted of iridoids and flavonoid components. METHODS: A total of 72 Sprague Dawley rats, aged 2 months, were randomly assigned to 6 groups low-, medium-, and high-dose Valerianajatamansi Jones groups intragastrically injected with 0.3, 0.6, and 0.9 g/kg per day Valerianajatamansi Jones extract, respectively; estazolam group intragastrically injected with 1.5 mg/kg per day estazolam; model and normal groups administered 5 mL physiological saline. Anxiety was established in all groups, except the normal group, through the use of elevated plus-maze test at 7 days following drug administration. MAIN OUTCOME MEASURES: Blood β-endorphin and corticosterone levels were determined using enzyme-linked immunosorbent assay following treatment with ValerianajatamansiJones extract. Expressions of HPA axis-related genes were measured by cDNA microarray. RESULTS: Blood β-endorphin and corticosterone levels were significantly greater in the model group than in the normal group. Compared with the model group, levels decreased with Valeriana jatamansi Jones extract or estazolam treatment, particularly in the low-dose Valeriana jatamansi Jones group (P〈 0.01). cDNA microarray results demonstrated that corticotropin-releasing hormone and Orexin, which are associated with HPA axis function, were differentially expressed; expression increased in the model group, but decreased in rats treated with Valerianajatamansi Jones extract. CONCLUSION: Valerianajatamansi Jones extract plays a role in regulating HPA axis function in a rat model of anxiety, and this effect was superior to estazolam.展开更多

The highest mean shoot length (3.71 cm) was achieved when media was fortified with BAP 2 mg/L in combination with NAA 1 mg/L. The highest mean leaf number i.e. 6.00 was observed when BAP was used alone at 2 mg/L. Average root length (0.77 cm) was recorded when BAP 1.5 mg/L along with NAA 0.5 mg/L was used. Maximum mean root numbers 2.57 were obtained when BAP and NAA were used at equal concentrations i.e. 1.5 mg/L. Observed data demonstrated that BAP up to 1 mg/L, 1.5 mg/L and 2 mg/L promotes shoot length, leaf number and leaf growth when used along with NAA at 0 mg/L, 0.5 mg/L and 1 mg/L. However lower quantities of both NAA (0 mg/L, 0.5 mg/L) and BAP (1 mg/L and 1.5 mg/L) produced significantly higher root length of Valeriana jatamansi Jones but the higher concentrations of plant growth hormones BAP (2 mg/L, 3 mg/L) and NAA (1 mg/L, 1.5 mg/L) were found unfavorable for increase in root length but the root number increases at higher concentration of NAA (1 mg/L and 1.5 mg/L).展开更多
Background:Inflammation and damage to neurons and other cells in the nervous system can cause disorders of the central nervous system.Microglial cells are activated by pathogen infection and injury to release nitric o...Background:Inflammation and damage to neurons and other cells in the nervous system can cause disorders of the central nervous system.Microglial cells are activated by pathogen infection and injury to release nitric oxide.Valerian(Valeriana officinalis)has been used as a sedative for the treatment of neurological diseases.This study evaluated inflammation of microglial cells and tumor necrosis factorαand induced nitric oxide synthetase gene expression influenced by valerian extract.Methods:Microglial cells were isolated from mice.Lipopolysaccharide(1 ng/mL)was used to induce inflammation and nitric oxide production in cells for an hour.The inflamed cells were then treated with different concentrations(0.1,0.5,2.5,20,and 50μL/mL)of valerian alcoholic extract for 1 and 24 h.nitric oxide production and tumor necrosis factorαand induced nitric oxide synthetase gene expression were determine by Griess assay and real-time polymerase chain reaction,respectively.Results:The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay showed no toxicity in several concentrations of the valerian extract.In addition,concentrations from 0.1 to 2.5μL/mL significantly reduced inflammation and nitric oxide production in mouse microglial cells to levels observed in control samples.Furthermore,tumor necrosis factorαand induced nitric oxide synthetase gene expression decreased when 2.5μL/mL extract was used.Conclusion:Based on these results,it can be concluded that 2.5μL/mL valerian alcoholic extract is effective as a candidate alternative medicine for reducing inflammation and nitric oxide production and consequently,the inflammatory symptoms of neurodegeneration.展开更多
[Objectives]To investigate the inhibitory effect and possible mechanism of essential oil from Valerianae Jatamansi Rhizoma et Radix on microglia activation induced by lipopolysaccharide(LPS).[Methods]The LPS-induced m...[Objectives]To investigate the inhibitory effect and possible mechanism of essential oil from Valerianae Jatamansi Rhizoma et Radix on microglia activation induced by lipopolysaccharide(LPS).[Methods]The LPS-induced microglia activation model was established and treated with different doses of essential oil from Valerianae Jatamansi Rhizoma et Radix.MTT assay was used to detect cell viability,ELISA to detect IL-6 secretion,RT-PCR to detect mRNA expression levels of IL-6,IL-1β,NF-κB,and IκBα,Western blotting to detect the protein expression of IL-6,IL-1β,NF-κB,IκBα,and their phosphorylated products.[Results]Compared with the normal control group,the model group showed increased IL-6 content(P<0.01),upregulated mRNA and protein levels of IL-6,IL-1β,and NF-κB(P<0.01),and elevated ratio of P-IκBα/IκBα(P<0.05).Compared with the model group,4 and 2μg/L essential oil from Valerianae Jatamansi Rhizoma et Radix reduced the content of IL-6(P<0.05),while 4,2,and 1μg/L essential oil from Valerianae Jatamansi Rhizoma et Radix down-regulated the mRNA and protein levels of IL-6,IL-1β,and NF-κB to varying degrees(P<0.05 or P<0.01),up-regulate the mRNA expression of IκBα(P<0.05 or P<0.01),and decreased the ratio of P-IκBα/IκBα(P<0.05 or P<0.01).[Conclusions]Essential oil from Valerianae Jatamansi Rhizoma et Radix can inhibit LPS-induced microglia activation,and its mechanism may be related to the inhibition of the NF-κB/IκBαsignaling pathway.展开更多
Objectives:To gather updated information on the traditional uses,phytochemistry,and pharmacological activities of Valeriana jatamansi Jones.Background:V.jatamansi is indigenous to the Himalayas immensely used as tradi...Objectives:To gather updated information on the traditional uses,phytochemistry,and pharmacological activities of Valeriana jatamansi Jones.Background:V.jatamansi is indigenous to the Himalayas immensely used as traditional folk remedy.It contains a variety of secondary metabolites that give it a high medicinal and therapeutic values.For the purpose of collecting relevant information on V.jatamansi,a comprehensive literature search was conducted.Based on ethnobotanical sources and historical records,traditional uses were compiled.Scientific articles and databases were consulted for phytochemical data.In order to assess the plant’s therapeutic properties,pharmacological studies were reviewed.Summary:The botanical is known for its ethnobotanical uses ranging from sedative and anxiolytic effects to treating insomnia,epilepsy,and gastrointestinal disorders.Indigenous healing practices rely heavily on its traditional uses.Based on the phytochemical analysis,it contains a diverse array of bioactive compounds,including sesquiterpenoids and iridoids.Valerenic acid and jatamansinol,among other compounds,contribute to its therapeutic potential.A broad spectrum of pharmacological activities is found including anxiety-relieving,sedative,anti-inflammatory,neuroprotective,and antimicrobial effects.Outlook:Based on the evidence supporting ethnomedicinal uses of V.jatamansi,its immense biopotential and multiple pharmacological roles,standard clinical trials are required to evaluate its therapeutic potential.展开更多

METHODS: Inducing rat model of reversible middle cerebral artery occlusion(MCAO)using Koizumi’s intraluminal suture occlusion method.48 male rats were divided into 5 groups randomly, pseudo operation group, MCAO group, saline control group,VOL group.2 hours after MCAO, we took gastric gavage with VOL and saline, 8 hour per time, and took out of brain to test C Fos,C Jun expression immunohistochemically at the 5th day after operation.RESULTS: There was no positive cell in each hippocampus zone of normal group;we observed C Fos, C Jun positive cells in each Hippocampus zone after MCAO;Density of C Fos,C Jun positive cells of VOL group were apparently lower than that of simple ischemia group. CONCLUSION: VOL can relieve histopathological lesions after cerebral ischemia and promote protection function of rat through inhibiting the expression of C Fos,C Jun expression.展开更多
Objective:To find out the antibacterial activity of Valeriana jatamansi(V.jatamansi)rhizomes against the extended-spectrumβ-lactamases(ESBLs)producing isolates of Enterobacteriaceae family.Methods:Confirmation of ESB...Objective:To find out the antibacterial activity of Valeriana jatamansi(V.jatamansi)rhizomes against the extended-spectrumβ-lactamases(ESBLs)producing isolates of Enterobacteriaceae family.Methods:Confirmation of ESBLs producing Escherichia coli,Enterobacter aerogenes,Klebsiella pneumoniae and Hafnia alvei isolated from urinary tract infections was performed by double disc diffusion assay.Antimicrobial susceptibility of all ESBLs producing isolates was determined by disc diffusion method following guidelines of Clinical and Laboratory Standards Institute.Successive extraction of rhizomes of V.jatamansi was performed with hexane,chloroform and methanol using Soxhelt apparatus.These extracts were tested against the ESBLs producing isolates using well diffusion method.Results:Hexane extract showed significant results as compared to chloroform and methanol extracts with the maximum zone of inhibition(21 mm)while ciprofloxacin and amikacin were used as standard drugs.Conclusions:Findings of the study suggested that hexane extract of V.jatamansi can be used in combination with other antibiotics as alternative treatment for urinary tract infections caused by ESBLs producing strains of Enterobacteriaceae.展开更多

However lower quantities of both NAA (0 mg/L, 0.5 mg/L) and BAP (1 mg/L and 1.5 mg/L) produced significantly higher root length of Valeriana jatamansi Jones but the higher concentrations of plant growth hormones BAP (2 mg/L, 3 mg/L) and NAA (1 mg/L, 1.5 mg/L) were found unfavorable for increase in root length but the root number increases at higher concentration of NAA (1 mg/L and 1.5 mg/L).
文摘Background:Inflammation and damage to neurons and other cells in the nervous system can cause disorders of the central nervous system.Microglial cells are activated by pathogen infection and injury to release nitric oxide.Valerian(Valeriana officinalis)has been used as a sedative for the treatment of neurological diseases.This study evaluated inflammation of microglial cells and tumor necrosis factorαand induced nitric oxide synthetase gene expression influenced by valerian extract.Methods:Microglial cells were isolated from mice.Lipopolysaccharide(1 ng/mL)was used to induce inflammation and nitric oxide production in cells for an hour.The inflamed cells were then treated with different concentrations(0.1,0.5,2.5,20,and 50μL/mL)of valerian alcoholic extract for 1 and 24 h.nitric oxide production and tumor necrosis factorαand induced nitric oxide synthetase gene expression were determine by Griess assay and real-time polymerase chain reaction,respectively.Results:The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay showed no toxicity in several concentrations of the valerian extract.In addition,concentrations from 0.1 to 2.5μL/mL significantly reduced inflammation and nitric oxide production in mouse microglial cells to levels observed in control samples.Furthermore,tumor necrosis factorαand induced nitric oxide synthetase gene expression decreased when 2.5μL/mL extract was used.Conclusion:Based on these results,it can be concluded that 2.5μL/mL valerian alcoholic extract is effective as a candidate alternative medicine for reducing inflammation and nitric oxide production and consequently,the inflammatory symptoms of neurodegeneration.

文摘[Objectives]To investigate the inhibitory effect and possible mechanism of essential oil from Valerianae Jatamansi Rhizoma et Radix on microglia activation induced by lipopolysaccharide(LPS).[Methods]The LPS-induced microglia activation model was established and treated with different doses of essential oil from Valerianae Jatamansi Rhizoma et Radix.MTT assay was used to detect cell viability,ELISA to detect IL-6 secretion,RT-PCR to detect mRNA expression levels of IL-6,IL-1β,NF-κB,and IκBα,Western blotting to detect the protein expression of IL-6,IL-1β,NF-κB,IκBα,and their phosphorylated products.[Results]Compared with the normal control group,the model group showed increased IL-6 content(P<0.01),upregulated mRNA and protein levels of IL-6,IL-1β,and NF-κB(P<0.01),and elevated ratio of P-IκBα/IκBα(P<0.05).Compared with the model group,4 and 2μg/L essential oil from Valerianae Jatamansi Rhizoma et Radix reduced the content of IL-6(P<0.05),while 4,2,and 1μg/L essential oil from Valerianae Jatamansi Rhizoma et Radix down-regulated the mRNA and protein levels of IL-6,IL-1β,and NF-κB to varying degrees(P<0.05 or P<0.01),up-regulate the mRNA expression of IκBα(P<0.05 or P<0.01),and decreased the ratio of P-IκBα/IκBα(P<0.05 or P<0.01).[Conclusions]Essential oil from Valerianae Jatamansi Rhizoma et Radix can inhibit LPS-induced microglia activation,and its mechanism may be related to the inhibition of the NF-κB/IκBαsignaling pathway.
文摘Objectives:To gather updated information on the traditional uses,phytochemistry,and pharmacological activities of Valeriana jatamansi Jones.Background:V.jatamansi is indigenous to the Himalayas immensely used as traditional folk remedy.It contains a variety of secondary metabolites that give it a high medicinal and therapeutic values.For the purpose of collecting relevant information on V.jatamansi,a comprehensive literature search was conducted.Based on ethnobotanical sources and historical records,traditional uses were compiled.Scientific articles and databases were consulted for phytochemical data.In order to assess the plant’s therapeutic properties,pharmacological studies were reviewed.Summary:The botanical is known for its ethnobotanical uses ranging from sedative and anxiolytic effects to treating insomnia,epilepsy,and gastrointestinal disorders.Indigenous healing practices rely heavily on its traditional uses.Based on the phytochemical analysis,it contains a diverse array of bioactive compounds,including sesquiterpenoids and iridoids.Valerenic acid and jatamansinol,among other compounds,contribute to its therapeutic potential.A broad spectrum of pharmacological activities is found including anxiety-relieving,sedative,anti-inflammatory,neuroprotective,and antimicrobial effects.Outlook:Based on the evidence supporting ethnomedicinal uses of V.jatamansi,its immense biopotential and multiple pharmacological roles,standard clinical trials are required to evaluate its therapeutic potential.
文摘Valeriana eichle riana (C. A. Muller) Graebner pollen are described and documented, used optical and scanning eletronic microscopy. We found that the current descriptions available goes this pollen disagree with the general pollinic pattern of the genera. Consequently, we conclude that the previous work in this area have taxonomical errors leading to inappropiate paleoenvironmental interpretations.
文摘AIM: To study influence of Valeriana officinalis var. latifolia(VOL) on expression of C Fos,C Jun after focal cerebral ischemia. METHODS: Inducing rat model of reversible middle cerebral artery occlusion(MCAO)using Koizumi’s intraluminal suture occlusion method.48 male rats were divided into 5 groups randomly, pseudo operation group, MCAO group, saline control group,VOL group.2 hours after MCAO, we took gastric gavage with VOL and saline, 8 hour per time, and took out of brain to test C Fos,C Jun expression immunohistochemically at the 5th day after operation.RESULTS: There was no positive cell in each hippocampus zone of normal group;we observed C Fos, C Jun positive cells in each Hippocampus zone after MCAO;Density of C Fos,C Jun positive cells of VOL group were apparently lower than that of simple ischemia group. CONCLUSION: VOL can relieve histopathological lesions after cerebral ischemia and promote protection function of rat through inhibiting the expression of C Fos,C Jun expression.
文摘Objective:To find out the antibacterial activity of Valeriana jatamansi(V.jatamansi)rhizomes against the extended-spectrumβ-lactamases(ESBLs)producing isolates of Enterobacteriaceae family.Methods:Confirmation of ESBLs producing Escherichia coli,Enterobacter aerogenes,Klebsiella pneumoniae and Hafnia alvei isolated from urinary tract infections was performed by double disc diffusion assay.Antimicrobial susceptibility of all ESBLs producing isolates was determined by disc diffusion method following guidelines of Clinical and Laboratory Standards Institute.Successive extraction of rhizomes of V.jatamansi was performed with hexane,chloroform and methanol using Soxhelt apparatus.These extracts were tested against the ESBLs producing isolates using well diffusion method.Results:Hexane extract showed significant results as compared to chloroform and methanol extracts with the maximum zone of inhibition(21 mm)while ciprofloxacin and amikacin were used as standard drugs.Conclusions:Findings of the study suggested that hexane extract of V.jatamansi can be used in combination with other antibiotics as alternative treatment for urinary tract infections caused by ESBLs producing strains of Enterobacteriaceae.
