Effect of vernalization on tuberization and flowering in the Tibetan turnip is associated with changes in the expression of FLC homologues 被引量:9

The turnip(Brassica rapa var. rapa) is a biennial crop that is planted in late summer/early fall and forms fleshy tubers for food in temperate regions. The harvested tubers then overwinter and are planted again the next spring for flowering and seeds. FLOWERING LOCUS C(FLC) is a MADS-box transcription factor that acts as a major repressor of floral transition by suppressing the flowering promoters FT and SOC1. Here we show that vernalization effectively represses tuber formation and promotes flowering in Tibetan turnip. We functionally characterized four FLC homologues(BrrFLC1,FLC2, FLC3, and FLC5), and found that BrrFLC2 and BrrFLC1 play a major role in repressing flowering in turnip and in transgenic Arabidopsis. In contrast, tuber formation was correlated with BrrFLC1 expression in the hypocotyl and was repressed under cold treatment following the quantitative downregulation of BrrFLC1. Grafting experiments of non-vernalized and vernalized turnips revealed that vernalization independently suppressed tuberization in the tuber or hypocotyl of the rootstock or scion, which occurred in parallel with the reduction in BrrFLC1 activity. Together, our results demonstrate that the Tibetan turnip is highly responsive to cold exposure, which is associated with the expression levels of BrrFLC genes. 展开更多

Early tuberization,a short period from stolon occurrence to tuber formation,is one of the major characteristics of a good early-maturing potato cultivar,while the regulatory mechanism of tuberization time(TT)is still unclear.In this study,two tetraploid cultivars,Zhongshu 3(Z3)and Zhongshu 18(Z18),with short and long TT respectively,were examined to reveal regulatory genes related to TT using RNA sequencing of tissue samples taken during stolon occurrence,stolon swelling and tuber formation.Cluster analysis showed that the gene expression patterns at the stolon swelling and tuber formation stages were significantly different from those at stolon occurrence in both Z3 and Z18.Therefore,we screened the differentially expressed genes(DEGs)at stolon swelling and tuber formation and compared them to those at stolon occurrence.A total of 3085 DEGs were specifically identified and analyzed according to Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment,and DEGs involved in starch and sucrose metabolism,plant hormone signal transduction and plant circadian clock were significantly enriched.The circadian clock genes were significantly differentially expressed between Z3 and Z18,revealing an important role for the plant circadian pathway in regulating TT.Furthermore,three candidate genes,StGI,StPRR and StEFM involved in circadian regulation and significantly differentially expressed between Z3 and Z18,were discovered and confirmed by qRT-PCR.The results provide valuable information for understanding the regulatory mechanisms of potato TT and represent a step toward breeding early-maturing potato cultivars. 展开更多

Phytohormones, auxins in particular, play an important role in plant development and productivity. Earlier data showed positive impact of exogenous auxin on potato (Solanum tuberosum L.) tuberization. The aim of this study was to generate potato plants with increased auxin level predominantly in tubers. To this end, a pBinB33-tms1 vector was constructed harboring the Agrobacterium auxin biosynthesis gene tms1 fused to tuber-specific promoter of the class I patatin gene (B33-promoter) of potato. Among numerous independently generated B33:tms1 lines, those without visible differences from control were selected for detailed studies. In the majority of transgenic lines, tms1 gene transcription was detected, mostly in tubers rather than in shoots. Indoleacetic acid (IAA) content in tubers and the auxin tuber-to-shoot ratio were increased in tms1-expressing transformants. The organ-specific increase in auxin synthesis in B33:tms1-transformants accelerated and intensified the process of tuber formation, reduced the dose of carbohydrate supply required for in vitro tuberization, and decreased the photoperiodic dependence of tuber initiation. Overall, a positive correlation was observed between tms1 expression, IAA content in tubers, and stimulation of tuber formation. The revealed properties of B33:tms1 transformants imply an important role for auxin in potato tuberization and offer prospects to magnify potato productivity by a moderate organ-specific enhancement of auxin content. 展开更多

This editorial discusses a case report recently published in the World Journal of Clinical Cases.The report describes the clinical presentation,imaging,diagnosis,and treatment of a patient with tuberous sclerosis complex(TSC)combined with primary lymphedema(PLE).Additionally,it retrospectively analyzes the data of 16 previously reported cases of children with TSC combined with PLE to summarize the epidemiology,genetic diagnosis,and current main treatments of these patients.The report also speculates on the pathological and physiological mechanisms underlying TSC combined with PLE.TSC combined with PLE is rare;therefore,the report provides a theoretical basis for understanding the pathophysiological mechanisms and treatment options for patients with TSC and PLE.Comprehensive clinical management of TSC is essential due to the diverse and multiorgan nature of its manifestations,often requiring a multidisciplinary approach for newly diagnosed cases. 展开更多

Ceratocystis fimbriata is a hazardous fungal pathogen that causes black rot disease in sweet potato roots,leading to devastating losses during sweet potato growth and storage.All methods for managing C.fimbriata on sweet potato based on synthetic fungicides,biocontrol agents,coatings,and natural compounds are reviewed for the first time.The advantages and disadvantages of the different methods are discussed,and their efficacies are compared.Bacillus strains and the synthetic fungicides carbendazim,flutriafol,liguoling,Stadium?,thiabendazole,and thiophanate methyl show the strongest inhibitory effects.Bacillus strains possess an advantage over synthetic fungicides due to the detrimental impact of the latter on human health.The application of filamentous fungi and yeasts as biocontrol agents for C.fimbriata management has not yet been investigated.Coatings exhibit limited protection,and no attempts have been made to combine coatings with nanoparticles or essential oils.Although the natural compounds perillaldehyde and nerol are inexpensive and non-toxic,they are of low efficacy.All management strategies have been used on postharvest sweet potato roots and leaves.However,there is no approach to controlling C.fimbriata in roots during plant growth.Even though several methods for controlling C.fimbriata on sweet potato have been reported in recent years,numerous knowledge gaps impede the development of effective and safe approaches. 展开更多

Eye depth is an important agronomic trait affecting tubers'appearance,quality,and processing suitability.Hence,cultivating varieties with uniform shapes and shallow eye depth are important goals for potato breeding.In this study,based on the primary mapping of the tuber eyedepth locus using a small primary-segregating population,a large secondary-segregating population with 2100 individuals was used to map the eye-depth locus further.A major quantitative trait locus for eye-depth on chromosome 10 was identified(designated qEyd10.1)using BSAseq and traditional QTL mapping methods.The qEyd10.1 could explain 55.0%of the eye depth phenotypic variation and was further narrowed to a 309.10 kb interval using recombinant analysis.To predict candidate genes,tissue sectioning and RNA-seq of the specific tuber tissues were performed.Genes encoding members of the peroxidase superfamily with likely roles in indole acetic acid regulation were considered the most promising candidates.These results will facilitate marker-assisted selection for the shallow-eye trait in potato breeding and provide a solid basis for eye-depth gene cloning and the analysis of tuber eye-depth regulatory mechanisms. 展开更多

Objective Our previous studies established that microRNA(miR)-451 from human umbilical cord mesenchymal stem cell-derived exosomes(hUC-MSC-Exos)alleviates acute lung injury(ALI).This study aims to elucidate the mechanisms by which miR-451 in hUC-MSC-Exos reduces ALI by modulating macrophage autophagy.Methods Exosomes were isolated from hUC-MSCs.Severe burn-induced ALI rat models were treated with hUC-MSC-Exos carrying the miR-451 inhibitor.Hematoxylin-eosin staining evaluated inflammatory injury.Enzyme-linked immunosorbnent assay measured lipopolysaccharide(LPS),tumor necrosis factor-α,and interleukin-1βlevels.qRT-PCR detected miR-451 and tuberous sclerosis complex 1(TSC1)expressions.The regulatory role of miR-451 on TSC1 was determined using a dual-luciferase reporter system.Western blotting determined TSC1 and proteins related to the mammalian target of rapamycin(mTOR)pathway and autophagy.Immunofluorescence analysis was conducted to examine exosomes phagocytosis in alveolar macrophages and autophagy level.Results hUC-MSC-Exos with miR-451 inhibitor reduced burn-induced ALI and promoted macrophage autophagy.MiR-451 could be transferred from hUC-MSCs to alveolar macrophages via exosomes and directly targeted TSC1.Inhibiting miR-451 in hUC-MSC-Exos elevated TSC1 expression and inactivated the mTOR pathway in alveolar macrophages.Silencing TSC1 activated mTOR signaling and inhibited autophagy,while TSC1 knockdown reversed the autophagy from the miR-451 inhibitor-induced.Conclusion miR-451 from hUC-MSC exosomes improves ALI by suppressing alveolar macrophage autophagy through modulation of the TSC1/mTOR pathway,providing a potential therapeutic strategy for ALI. 展开更多

BACKGROUND Tuberous sclerosis complex(TSC)and primary lymphedema(PLE)are both rare diseases,and it is even rarer for both to occur in the same patient.In this work,we have provided a detailed description of a patient's clinical presentation,imaging findings,and treatment.And a retrospective analysis was conducted on 14 published relevant case reports.CASE SUMMARY A 16-year-old male came to our hospital for treatment due to right lower limb swelling.This swelling is already present from birth.The patient’s memory had been progressively declining.Seizures had occurred 1 year prior at an unknown frequency.The patient was diagnosed with TSC combined with PLE through multimodal imaging examination:Computed tomography,magnetic resonance imaging,and lymphoscintigraphy.The patient underwent liposuction.The swelling of the patient's right lower limb significantly improved after surgery.Epilepsy did not occur.after taking antiepileptic drugs and sirolimus.CONCLUSION TSC with PLE is a rare and systemic disease.Imaging can detect lesions of this disease,which are important for diagnosis and treatment. 展开更多

Effect of Inorganic and Organic Fertilizers on Growth and Yield of Two Industrial Potato Varieties (Asterix and Courage) in Bangladesh

Potato is one of the most important vegetable crops, which contributes more than half of the total vegetable production in Bangladesh. Four field experiments were conducted in two different locations in Bangladesh to develop integrated nutrient management practices to produce quality potato seed in industrial processing varieties Asterix and Courage. For the inorganic trial, Factorial Randomized Complete Block Design (RCBD) including 2 potato varieties, 5 treatments with 4 replications, and in the organic fertilizer trial, Factorial RCBD including 2 potato varieties, 6 treatments with 4 replications were used. In the inorganic fertilizer trail, the highest yield was obtained in the variety Asterix due to Nitrogen, Phosphorus, Potassium, and Sulfur (NPKS) plus Magnesium treated plot in Domar BADC farm and due to NPKS plus Boron, Zinc, and Magnesium treated plots in Kashimpur Farm. In the case of variety Courage, the highest yield was found in the treatment of NPKS plus Zinc in Domar BADC farm while in Kashimpur farm, NPKS plus Mg treated plots had the highest yield of potato variety-Courage. In the organic fertilizer trail, the highest tuber yield per hill was obtained by applying the government-approved commercial brand Northern organic fertilizer in variety Asterix and by organic fertilizer brand Chook Chook in variety Courage. Parameters such as days to tuber initiation, number of stems per hill, plant height, and number of tubers per hill were found statistically different among the treatments and between the two varieties. Treatments namely Northern organic fertilizer and Cowdung combined with mustard oil cake performed better considering standard grade tuber yield (grade A and B) compared to other treatments. Hence, the combination of NPKS MgZn and either Northern organic or Chook Chook or Cowdung plus mustard oil cake could be used to grow the varieties Asterix and Courage. 展开更多

This study aimed to evaluate the nutritional and sensory characteristics of penne-type pasta produced from locally sourced cereals, tubers, and legumes. To achieve this, we formulated four different types of pasta with varying levels of incorporation ranging from 10 to 50% cereals such as millet, fonio, and rice, tubers such as manioc and sweet potato, and legumes such as cowpea. The results showed that the incorporation of local products considerably improved the nutrient content of penne without being rejected by consumers. Sensory analysis showed that the best results were obtained with mixtures of sweet potato, cowpea, and wheat, as well as with rice, manioc, cowpea, and sample wheat. These results are of great importance to manufacturers in a world where developing nutritious and attractive food products is a crucial challenge. 展开更多

The tropical roots and tuber crops (R and T) consist of cassava, sweet potato, yams, elephant foot yam, taro, tannia and a couple of minor tuber crops. Tropical tuber crops are the third most important food crops after cereals and pulses. These crops play a crucial role in providing food and nutritional security to the rural masses in Africa, Latin America and parts of Asia. Cassava and sweet potato rank among the top 10 food crops produced in developing countries and contribute to about 6% of world’s dietary calories. In India, tropical tubers are grown mostly in states like Odisha, West Bengal, Andhra Pradesh, Kerala, Tamil Nadu, Bihar and North East covering 4.5% of the total area under vegetables with 5.7% of the total vegetable production. Low income farmers and most undernourished households in India depend on R and T crops. They value these crops for their high calorie and stable yields under conditions in which other crops may fail. Genetic diversity of major tropical tuber crops provides options for its diverse usage in “multicuisine”, “nutrition”, “feed”, health care” and renewable resources of processing firms. Water productivity is also higher, for example, to produce 1 kg of tubers, sweet potatoes require water (383 l), taro (606 l), yam (343 l), as compared to rice (1673 l), maize (1222 l), wheat (1827 l), etc. Tuber crops are one of most efficient producers of dry matter and edible energy. These crops have potential to reverse soil degradation. Cassava is already recognized as drought tolerant followed by yam. Tubers of taro, elephant foot yam and yams can be stored under zero energy conditions for 6 - 7 months unlike temperate potatoes in cold storage. The natural climatic resilience as well as inherent nutrition attributes, has been explored further. Such studies made these crops more robust to changing climate and as the source of nutrient enriched food bowls. Sweet potato variety Bhu Krishna is the first purple flesh variety in India containing (90 mg/100 g anthocyanin) coupled with starch 22% - 25%. Similarly, the orange flesh Bhu Sona containing beta carotene (14 mg/100 g) coupled with high starch 22% - 24% is the first variety having both high beta carotene and high starch. These varieties are tolerant to salinity and Bhu Krishna is resistant to weevil. Likewise, developed improved taro varieties resistant to biotic blight stress, tolerant to salinity and are enriched with micro nutrients. These improved varieties are used to breed the superior types further. All these climate resilient, nutritionally enriched varieties will have a greater impact on supplementing rainbow food for all. These can address the issues like “food insecurity”, “malnutrition”, diet related issue of mass consumers across the world. The analysis of innovations on enhancing adaptability, nutritional quality with a management matrix would help in strengthening future programme. 展开更多

The aim of this study is to produce large quantities of yam tubers in the field from seed tubers derived from vitroplants regenerated from aerial stem cuttings and farmers’ tuber seeds. Seed tubers from ten yam cultivars, including five from Dioscorea alata (cDa083, cDa053, cDa115, cDa150 and cDa266) and five from Dioscorea cayenensis-rotundata (cDr015, cDr027, cDr150, cDr206 and cDr148) with distinct agro-morphological characteristics, were used as material. A completely randomized block design with 3 replications was used. In each block, all cultivars and seed types were represented. When put into cultivation, the tubers produced by the vitroplants all germinated and gave 100% healthy plants, compared with 86% to 100% healthy plants for the seed tubers produced by the farmers. Yields per hectare ranged from 10 to 25 tonnes for seed tubers produced by in vitro plants, and from 4 to 9 tons for seed tubers produced by farmers. It is currently one of the minor plants of high socio-economic interest. One of the limitations to promoting this species in Burkina Faso is the lack of varieties that meet consumers’ demands. Implementing a breeding program is an important step toward achieving this goal. Such a program is based on the variability of agronomical traits of interest within evaluated germplasm. This study aimed to assess the level of two germplasms variability of S. rotundifolius from Ghana and Burkina Faso for traits related to vegetative development, cycle, and yield. Agromorphological characterization of 174 accessions, including 116 from Ghana and 58 from Burkina Faso was carried out in Randomised Complete Block Design with three replications. The characterization was made on the basis of fifteen (15) quantitative traits related to the canopy and leaf size, the cycle, and the yield. Analysis of variance revealed significant differences between accessions within each germplasm for all the evaluated traits. The analysis of the structuration of this agromorphological variability allowed organizing the accessions into different groups. These results could lead to the identification of accessions within each germplasm for breeding purposes or future research on genotype-environment interactions. 展开更多

This study aimed to assess the microbiological quality of tiger nut (Cyperus esculentus L.) products sold in Ouagadougou and Bobo Dioulasso. The microbial load of tubers and tiger nut milk was determined using ISO 4833-1 (2003) standard (total mesophilic aerobic flora), NF ISO 7954 (1998) standard (yeasts and molds), and ISO 4832 (2006) standard (total coliforms, thermotolerant coliforms). The results showed that tiger nut tubers had an average microbial load between 4.86 ± 7.03 × 10<sup>6</sup> UFC/g and 1.82 ± 2.30 × 10<sup>7</sup> UFC/g for total mesophilic aerobic flora, from 4.34 ± 10.6 × 10<sup>5</sup> UFC/g to 3.52 ± 4.54 × 10<sup>6</sup> UFC/g for yeasts and molds, from 2.23 ± 2.25 × 10<sup>5</sup> UFC/g to 1.41 ± 2.16 × 10<sup>5</sup> UFC/g for total coliforms and from 1.83 ± 2.03 × 10<sup>5</sup> UFC/g to 7.0 ± 10.8 × 10<sup>4</sup> CFU/g for thermotolerant coliforms. For tiger nut milk samples, the average microbial load varied from 3.48 ± 2.98 × 10<sup>6</sup> CFU/g to 2.80 ± 5.69 × 107 CFU/g for total mesophilic aerobic flora, from 5.00 ± 7.21 × 10<sup>4</sup> CFU/g to 1.88 ± 3.31 × 10<sup>5</sup> CFU/g for yeasts and molds, from 4.58 ± 10.4 × 10<sup>4</sup> CFU/g to 6.31 ± 9.17 × 10<sup>5</sup> CFU/g for total coliforms and 7.00 ± 7.00 × 10<sup>3</sup> CFU/g to 2.87 ± 5.86 × 10<sup>5</sup> CFU/g for thermotolerant coliforms. This study revealed that the tubers and tiger nut milk sold in Ouagadougou and Bobo Dioulasso had a high microbial load which could lead to the degradation of these products and food poisoning for consumers. 展开更多

[Method] Potato tubers of Favorita and Zhongshu No.3 were irradiated with 5, 10, 20, 30, 50 and 70 Gy of cobalt-60 γ-ray respectively, to explore the mutagenic effect of different doses of γ-ray heavy ion irradiation on potato. [Result] The results showed that the emergence rate, seedling rate, plant growth, yield and commodity of potato varied after irradiated with different doses of cobalt-60 γ-ray; 10Gy of cobalt-60 irradiation played a positive effect on the growth and development of potato, while high doses played a negative effect on the growth and development of potato. After cobalt-60 irradiation, the botanical traits of M1 gen- eration varied greatly, resulting in a number of beneficial mutation traits; some traits of M2 and M3 generations became stably inherited. [Conclusion] Cobalt-60 irradiation breeding is conducive to the variety improvement and germplasm innovation as an effective means of genetic improvement for potato. 展开更多
