Ceratocystis fimbriata is a hazardous fungal pathogen that causes black rot disease in sweet potato roots,leading to devastating losses during sweet potato growth and storage.All methods for managing C.fimbriata on sw...Ceratocystis fimbriata is a hazardous fungal pathogen that causes black rot disease in sweet potato roots,leading to devastating losses during sweet potato growth and storage.All methods for managing C.fimbriata on sweet potato based on synthetic fungicides,biocontrol agents,coatings,and natural compounds are reviewed for the first time.The advantages and disadvantages of the different methods are discussed,and their efficacies are compared.Bacillus strains and the synthetic fungicides carbendazim,flutriafol,liguoling,Stadium?,thiabendazole,and thiophanate methyl show the strongest inhibitory effects.Bacillus strains possess an advantage over synthetic fungicides due to the detrimental impact of the latter on human health.The application of filamentous fungi and yeasts as biocontrol agents for C.fimbriata management has not yet been investigated.Coatings exhibit limited protection,and no attempts have been made to combine coatings with nanoparticles or essential oils.Although the natural compounds perillaldehyde and nerol are inexpensive and non-toxic,they are of low efficacy.All management strategies have been used on postharvest sweet potato roots and leaves.However,there is no approach to controlling C.fimbriata in roots during plant growth.Even though several methods for controlling C.fimbriata on sweet potato have been reported in recent years,numerous knowledge gaps impede the development of effective and safe approaches.展开更多
Eye depth is an important agronomic trait affecting tubers'appearance,quality,and processing suitability.Hence,cultivating varieties with uniform shapes and shallow eye depth are important goals for potato breedin...Eye depth is an important agronomic trait affecting tubers'appearance,quality,and processing suitability.Hence,cultivating varieties with uniform shapes and shallow eye depth are important goals for potato breeding.In this study,based on the primary mapping of the tuber eyedepth locus using a small primary-segregating population,a large secondary-segregating population with 2100 individuals was used to map the eye-depth locus further.A major quantitative trait locus for eye-depth on chromosome 10 was identified(designated qEyd10.1)using BSAseq and traditional QTL mapping methods.The qEyd10.1 could explain 55.0%of the eye depth phenotypic variation and was further narrowed to a 309.10 kb interval using recombinant analysis.To predict candidate genes,tissue sectioning and RNA-seq of the specific tuber tissues were performed.Genes encoding members of the peroxidase superfamily with likely roles in indole acetic acid regulation were considered the most promising candidates.These results will facilitate marker-assisted selection for the shallow-eye trait in potato breeding and provide a solid basis for eye-depth gene cloning and the analysis of tuber eye-depth regulatory mechanisms.展开更多

[Method] Potato tubers of Favorita and Zhongshu No.3 were irradiated with 5, 10, 20, 30, 50 and 70 Gy of cobalt-60 γ-ray respectively, to explore the mutagenic effect of different doses of γ-ray heavy ion irradiation on potato. [Result] The results showed that the emergence rate, seedling rate, plant growth, yield and commodity of potato varied after irradiated with different doses of cobalt-60 γ-ray; 10Gy of cobalt-60 irradiation played a positive effect on the growth and development of potato, while high doses played a negative effect on the growth and development of potato. After cobalt-60 irradiation, the botanical traits of M1 gen- eration varied greatly, resulting in a number of beneficial mutation traits; some traits of M2 and M3 generations became stably inherited. [Conclusion] Cobalt-60 irradiation breeding is conducive to the variety improvement and germplasm innovation as an effective means of genetic improvement for potato.展开更多

In this study, total five sweetpotato varieties were selected, and the effects of different shapes and sizes on mechanical harvesting of sweetpotato were investigated. The results showed that the breakage rate of Xuzishu 5 was lowest, so it is more suitable for mechanical harvesting in field. At the same time, the correlations between sweetpotato shape, breakage rate and exposure rate were analyzed. It was found that there was a significant linear correlation between sweet- potato shape and breakage rate (P〈0.01); the higher the flat rate is, the higher the breakage rate is; and sweetpotato shape had no significant effect on exposure rate. Therefore, tuberous root with lower flat rate, as well as better agronomic characters should be selected in the breeding process of new sweetpotato varieties.展开更多

Based on previously reported in vitro assays, β amylase is considered as one of the key enzymes catalyzing starch breakdown, but up to date its role in starch breakdown in living cells remains unclear because the enzyme was shown often extrachloroplastic in living cells. Recently we have shown for the first time that β_amylase is predominantly immuno_localized to plastids in living cells of developing apple fruit. But it remains to know whether this model of β_amylase compartmentation is more widespread in plant living cells. The present experiment, conducted in tuberous root of sweet potato ( Ipomea batatas Lam. cv. Xushu 18) and via immunogold electron_microscopy technique, showed that β amylase visualized by gold particles was predominantly localized in plastids especially at periphery of starch granules, but the gold particles were scarcely found in other subcellular compartments, indicating that the enzyme is subcellularly compartmented in the same zone as its starch substrates. The density of gold particles (β amylase) in plastids was increasing during growing season, but the predominantly plastid_distributed pattern of β amylase in cells was shown unchanged throughout the tuberous root development. These data prove that the enzyme is compartmented in its functional sites, and so provide evidence to support the possible widespread biological function of the enzyme in catalyzing starch breakdown in plant living cells or at least in living cells of plant storage organs.展开更多

China is an important domestication center of yams,and two main yam species of Dioscorea opposita and D.alata are commonly cultivated in China.However,the differences of nutritional and medicinal characteristics betwe...China is an important domestication center of yams,and two main yam species of Dioscorea opposita and D.alata are commonly cultivated in China.However,the differences of nutritional and medicinal characteristics between the two species and their subgroups remain unclear,which would greatly affect the resource conservation and commercial utilization of yams.In this study,typical yam resources including the species of D.opposita(wild and cultivated Ruichang yam from southern China,and Tiegun yam from northern China)and two landraces of D.alata(Longyan yam and Anyuan yam from southern China)were selected as materials.Nutritional traits and medicinal characteristics were determined and analyzed respectively.The results showed that there was no significant differences in the content of most nutrients between D.opposita and D.alata,but most cultivated Ruichang yam of D.opposita showed higher levels of starch,soluble sugar,sucrose,and ascorbate in tuber than that in yam from D.alata.Moreover,an UPLC-MS method was developed for identification and determination of medicinal characteristics in the two species.The results showed that allantoin can be detected in all selected samples.Cultivated Ruichang yam of D.opposita possessed the highest allantoin content among the tested materials,and was significantly different with that in Tiegun yam and D.alata.Dioscin was not detected in D.alata.Overall,there was little difference in nutritional composition between D.opposita and D.alata,but the medicinal quality of D.opposita was better than that of D.alata.Due to the outstanding comprehensive quality,the local variety of cultivated Ruichang yam can be further developed and utilized.展开更多

This research aimed to explore the effect of processing on cardiac efficacy of alkaloids in Aconitum tubers. First, the chemical ingredients in unprocessed and processed Aconitum tubers were identified and compared by using high performance liquid chromatography time-of-flight mass spectrometry(HPLC-TOF/MS) and multivariate pattern recognition methods. Then the representative alkaloids in Aconitum tubers, aconitine, benzoylaconine, and aconine, which belong to diester-diterpenoid alkaloids,monoester-diterpenoid alkaloids, and amine-diterpenoid alkaloids, respectively, were selected for further validation of attenuated mechanism. Subsequent pharmacological experiments with aconitine, benzoylaconine,and aconine in SD rats were used to validate the effect of processing on cardiac functions. After processing the Aconitum tubers, it was found that the contents of diester-diterpenoid alkaloids were reduced, and those of monoester-diterpenoid alkaloids and amine-diterpenoid alkaloids were increased, suggesting that diesterditerpenoid alkaloids were transformed into monoester-diterpenoid alkaloids and amine-diterpenoid alkaloids.Through further decocting the aconitine in boiling water, it was confirmed that the three alkaloids could be progressively transformed. Pharmacological experiments with aconitine, benzoylaconine, and aconine in SD rats showed that aconitine at a dose of 0.01 mg/kg and aconine at a dose of 10 mg/kg enhanced the cardiac function, while benzoylaconine at a dose of 2 mg/kg weakened the cardiac function. The effect of processing is attributed to the transformation of the most toxic diester-diterpenoid alkaloids into less toxic monoesterditerpenoid alkaloids and amine-diterpenoid alkaloids.展开更多
First reported in 1976,hepatic angiomyolipoma(HAML)is a rare mesenchymal liver tumor occurring mostly in middle-aged women.Diagnosis of the liver mass is often incidental on abdominal imaging due to the frequent absen...First reported in 1976,hepatic angiomyolipoma(HAML)is a rare mesenchymal liver tumor occurring mostly in middle-aged women.Diagnosis of the liver mass is often incidental on abdominal imaging due to the frequent absence of specific symptoms.Nearly 10%of HAMLs are associated with tuberous sclerosis complex.HAML contains variable proportions of blood vessels,smooth muscle cells and adipose tissue,which renders radiological diagnosis hazardous.Cells express positivity for HMB-45 and actin,thus these tumors are integrated into the group of perivascular epithelioid cell tumors.Typically,a HAML appears on magnetic resonance imaging(or computed tomography scan)as a hypervascular solid tumor with fatty areas and with washout,and can easily be misdiagnosed as other liver tumors,particularly hepatocellular carcinoma.The therapeutic strategy is not clearly defined,but surgical resection is indicated for symptomatic patients,for tumors showing an aggressive pattern(i.e.,changes in size on imaging or high proliferation activity and atypical epithelioid pattern on liver biopsy),for large(>5 cm)biopsy-proven HAML,and if doubts remain on imaging or histology.Conservative management may be justified in other conditions,since most cases follow a benign clinical course.In summary,the correct diagnosis of HAML is challenging on imaging and relies mainly on pathological findings.展开更多

文摘A 58-year-old man presented with the chief complaint of abdominal bloating and was incidentally found to have a liver tumor.As diagnostic imaging studies could not rule out malignancy,the patient underwent partial resection of segment 3 of the liver.The lesion pathologically showed eosinophilic proliferation,in addition to immunohistochemical positivity for human melanoma black 45 and Melan-A,thereby leading to the diagnosis of a hepatic perivascular epithelioid cell tumor(PEComa).A PEComa arising from the liver is relatively rare.Moreover,the name ‘PEComa' has not yet been widely recognized,and the same disease entity has been called epithelioid angiomyolipoma(EAML),further diminishing the recognition of PEComa.In addition,PEComa imaging findings mimic those of malignant liver tumors,and clinically,this tumor tends to enlarge.Therefore,a PEComa is difficult to diagnose.We conducted a systematic review of PEComa and EAML cases and discuss the results,including findings useful for differentiating perivascular epithelioid cell tumors from malignant liver tumors.

文摘China is an important domestication center of yams,and two main yam species of Dioscorea opposita and D.alata are commonly cultivated in China.However,the differences of nutritional and medicinal characteristics between the two species and their subgroups remain unclear,which would greatly affect the resource conservation and commercial utilization of yams.In this study,typical yam resources including the species of D.opposita(wild and cultivated Ruichang yam from southern China,and Tiegun yam from northern China)and two landraces of D.alata(Longyan yam and Anyuan yam from southern China)were selected as materials.Nutritional traits and medicinal characteristics were determined and analyzed respectively.The results showed that there was no significant differences in the content of most nutrients between D.opposita and D.alata,but most cultivated Ruichang yam of D.opposita showed higher levels of starch,soluble sugar,sucrose,and ascorbate in tuber than that in yam from D.alata.Moreover,an UPLC-MS method was developed for identification and determination of medicinal characteristics in the two species.The results showed that allantoin can be detected in all selected samples.Cultivated Ruichang yam of D.opposita possessed the highest allantoin content among the tested materials,and was significantly different with that in Tiegun yam and D.alata.Dioscin was not detected in D.alata.Overall,there was little difference in nutritional composition between D.opposita and D.alata,but the medicinal quality of D.opposita was better than that of D.alata.Due to the outstanding comprehensive quality,the local variety of cultivated Ruichang yam can be further developed and utilized.

文摘According to the Chinese Pharmacopoeia 2015, only processed Aconitum tubers can be clinically applied, and the effect of processing is unclear. This research aimed to explore the effect of processing on cardiac efficacy of alkaloids in Aconitum tubers. First, the chemical ingredients in unprocessed and processed Aconitum tubers were identified and compared by using high performance liquid chromatography time-of-flight mass spectrometry(HPLC-TOF/MS) and multivariate pattern recognition methods. Then the representative alkaloids in Aconitum tubers, aconitine, benzoylaconine, and aconine, which belong to diester-diterpenoid alkaloids,monoester-diterpenoid alkaloids, and amine-diterpenoid alkaloids, respectively, were selected for further validation of attenuated mechanism. Subsequent pharmacological experiments with aconitine, benzoylaconine,and aconine in SD rats were used to validate the effect of processing on cardiac functions. After processing the Aconitum tubers, it was found that the contents of diester-diterpenoid alkaloids were reduced, and those of monoester-diterpenoid alkaloids and amine-diterpenoid alkaloids were increased, suggesting that diesterditerpenoid alkaloids were transformed into monoester-diterpenoid alkaloids and amine-diterpenoid alkaloids.Through further decocting the aconitine in boiling water, it was confirmed that the three alkaloids could be progressively transformed. Pharmacological experiments with aconitine, benzoylaconine, and aconine in SD rats showed that aconitine at a dose of 0.01 mg/kg and aconine at a dose of 10 mg/kg enhanced the cardiac function, while benzoylaconine at a dose of 2 mg/kg weakened the cardiac function. The effect of processing is attributed to the transformation of the most toxic diester-diterpenoid alkaloids into less toxic monoesterditerpenoid alkaloids and amine-diterpenoid alkaloids.
文摘First reported in 1976,hepatic angiomyolipoma(HAML)is a rare mesenchymal liver tumor occurring mostly in middle-aged women.Diagnosis of the liver mass is often incidental on abdominal imaging due to the frequent absence of specific symptoms.Nearly 10%of HAMLs are associated with tuberous sclerosis complex.HAML contains variable proportions of blood vessels,smooth muscle cells and adipose tissue,which renders radiological diagnosis hazardous.Cells express positivity for HMB-45 and actin,thus these tumors are integrated into the group of perivascular epithelioid cell tumors.Typically,a HAML appears on magnetic resonance imaging(or computed tomography scan)as a hypervascular solid tumor with fatty areas and with washout,and can easily be misdiagnosed as other liver tumors,particularly hepatocellular carcinoma.The therapeutic strategy is not clearly defined,but surgical resection is indicated for symptomatic patients,for tumors showing an aggressive pattern(i.e.,changes in size on imaging or high proliferation activity and atypical epithelioid pattern on liver biopsy),for large(>5 cm)biopsy-proven HAML,and if doubts remain on imaging or histology.Conservative management may be justified in other conditions,since most cases follow a benign clinical course.In summary,the correct diagnosis of HAML is challenging on imaging and relies mainly on pathological findings.
