Objective The protein interacting with C kinase 1(PICK1)plays a critical role in vesicle trafficking,and its deficiency in sperm cells results in abnormal vesicle trafficking from Golgi to acrosome,which eventually di...Objective The protein interacting with C kinase 1(PICK1)plays a critical role in vesicle trafficking,and its deficiency in sperm cells results in abnormal vesicle trafficking from Golgi to acrosome,which eventually disrupts acrosome formation and leads to male infertility.Methods An azoospermia sample was filtered,and the laboratory detection and clinical phenotype indicated typical azoospermia in the patient.We sequenced all of the exons in the PICK1 gene and found that there was a novel homozygous variant in the PICK1 gene,c.364delA(p.Lys122SerfsX8),and this protein structure truncating variant seriously affected the biological function.Then we constructed a PICK1 knockout mouse model using clustered regularly interspaced short palindromic repeat cutting technology(CRISPRc).Results The sperm from PICK1 knockout mice showed acrosome and nucleus abnormalities,as well as dysfunctional mitochondrial sheath formation.Both the total sperm and motility sperm counts were decreased in the PICK1 knockout mice compared to wild-type mice.Moreover,the mitochondrial dysfunction was verified in the mice.These defects in the male PICK1 knockout mice may have eventually led to complete infertility.Conclusion The c.364delA novel variant in the PICK1 gene associated with clinical infertility,and pathogenic variants in the PICK1 may cause azoospermia or asthenospermia by impairing mitochondrial function in both mice and humans.展开更多

A total of 393 potholes(368 fluvial and 25 marine potholes)were studied at seven different sites in both the fluvial and marine environments.Diverse bedrock properties and large-scale delivery of tools and grinders re...A total of 393 potholes(368 fluvial and 25 marine potholes)were studied at seven different sites in both the fluvial and marine environments.Diverse bedrock properties and large-scale delivery of tools and grinders regulate the dynamic growth,truncation,and amalgamation of potholes.Therefore,the principal objectives of the study are(i)to examine the relationship between the growth of potholes and substrate lithological with structural characteristics(applying geospatial and Schmidt hammer for rock strength analysis)and(ii)to measure the morphology,and size of tools and grinders,processes of truncation and amalgamation in hydro-geomorphic environment using various indices and field techniques.The result showed that large potholes are stretched in the direction of lineament axes and roughly parallel to the river flow direction.Here,the steady growth of pothole depth-diameter is controlled by active bedrock structures,tools,or grinders,and monsoonal high-velocity bank full discharge.Consequently,the deepening and widening of potholes are relatively slow at Bindu,Deuli,and marine beach Neil Island due to fewer structures and little supply of tools or grinders.In small stretches,(Damodar,Subarnarekha,and Rarhu)canyons and gorge-like features(bedrock incision)are formed at Rajrappa,Bhakuyadi,and Guridih sites due to cyclic truncation and amalgamation.Truncation and amalgamation processes restrict the vertical depth threshold value of potholes within 3m,particularly at Rajrappa,Bhakuyadi,and Guridih sites.Scientific study of the pothole's dynamic growth is greatly necessary for the different environmental engineering and river hydraulic projects like excavation,dredging,and dam or barrage construction.Successively,it is essential to compute the cost of rock excavation or dredging,primarily for the mechanical strength of the bedrock river channel and its stability.展开更多
While variable regions of immunoglobulins are extensively diversified by V(D)J recombination and somatic hypermutation in vertebrates,the constant regions of immunoglobulin heavy chains also utilize certain mechanisms...While variable regions of immunoglobulins are extensively diversified by V(D)J recombination and somatic hypermutation in vertebrates,the constant regions of immunoglobulin heavy chains also utilize certain mechanisms to produce diversity,including class switch recombination(CSR),subclass differentiation,and alternative expression of the same gene.Many species of birds,reptiles,and amphibians express a truncated isoform of immunoglobulin Y(IgY),termed IgY(ΔFc),which lacks theυCH3 andυCH4 domains.In Anseriformes,IgY(ΔFc)arises from alternative transcriptional termination sites within the sameυgene,whereas in some turtles,intact IgY and IgY(ΔFc)are encoded by distinct genes.Different from the previously reported IgY(ΔFc)variants,this study identified a truncated IgY in the snake Elaphe taeniura,characterized by the loss of only a portion of the CH4 domain.Western blotting and liquid chromatographytandem mass spectrometry confirmed that this truncated IgY is generated by post-translational cleavage at N338 within the IgY heavy chain constant(CH)region.Furthermore,both human and snake asparaginyl endopeptidase were shown to cleave snake IgY in vitro.These findings reveal a novel mechanism for the production of shortened IgY forms,demonstrating that the immunoglobulin CH region undergoes diversification through distinct strategies across vertebrates.展开更多
The count of one column for high-dimensional datasets, i.e., the number of records containing this column, has been widely used in nuinerous applications such as analyzing popular spots based on check-in location info...The count of one column for high-dimensional datasets, i.e., the number of records containing this column, has been widely used in nuinerous applications such as analyzing popular spots based on check-in location information and mining valuable items from shopping records. However, this poses a privacy threat when directly publishing this information. Differential privacy (DP), as a notable paradigm for strong privacy guarantees, is thereby adopted to publish all column counts. Prior studies have verified that truncating records or grouping columns can effectively improve the accuracy of published results. To leverage the advantages of the two techniques, we combine these studies to further boost the accuracy of published results. However, the traditional penalty function, which measures the error imported by a given pair of parameters including truncating length and group size, is so sensitive that the derived parameters deviate from the optimal parameters significantly. To output preferable parameters, we first design a smart penalty function that is less sensitive than the traditional function. Moreover, a two-phase selection method is proposed to compute these parameters efficiently, together with the improvement in accuracy. Extensive experiments on a broad spectrum of real-world datasets validate the effectiveness of our proposals.展开更多
Alzheimer's disease is characterized by two major neuropathological hallmarks—the extracellularβ-amyloid plaques and intracellular neurofibrillary tangles consisting of aggregated and hyperphosphorylated Tau pro...Alzheimer's disease is characterized by two major neuropathological hallmarks—the extracellularβ-amyloid plaques and intracellular neurofibrillary tangles consisting of aggregated and hyperphosphorylated Tau protein.Recent studies suggest that dysregulation of the microtubuleassociated protein Tau,especially specific proteolysis,could be a driving force for Alzheimer's disease neurodegeneration.Tau physiologically promotes the assembly and stabilization of microtubules,whereas specific truncated fragments are sufficient to induce abnormal hyperphosphorylation and aggregate into toxic oligomers,resulting in them gaining prion-like characteristics.In addition,Tau truncations cause extensive impairments to neural and glial cell functions and animal cognition and behavior in a fragment-dependent manner.This review summarizes over 60 proteolytic cleavage sites and their corresponding truncated fragments,investigates the role of specific truncations in physiological and pathological states of Alzheimer's disease,and summarizes the latest applications of strategies targeting Tau fragments in the diagnosis and treatment of Alzheimer's disease.展开更多

This paper investigates the hydrodynamic characteristics of floating truncated cylinders undergoing horizontal and vertical motions due to earthquake excitations in the finite water depth.The governing equation of the...This paper investigates the hydrodynamic characteristics of floating truncated cylinders undergoing horizontal and vertical motions due to earthquake excitations in the finite water depth.The governing equation of the hydrodynamic pressure acting on the cylinder is derived based on the radiation theory with the inviscid and incompressible assumptions.The governing equation is solved by using the method of separating variables and analytical solutions are obtained by assigning reasonable boundary conditions.The analytical result is validated by a numerical model using the exact artificial boundary simulation of the infinite water.The main variation and distribution characteristics of the hydrodynamic pressure acting on the side and bottom of the cylinder are analyzed for different combinations of wide-height and immersion ratios.The added mass coefficient of the cylinder is calculated by integrating the hydrodynamic pressure and simplified formulas are proposed for engineering applications.The calculation results show that the simplified formulas are in good agreement with the analytical solutions.展开更多
Landslides are a type of natural disaster that can cause substantial harm to humanity.Monitoring and predicting the initiation of potential landslides is critical to avoiding losses due to disasters and economic activ...Landslides are a type of natural disaster that can cause substantial harm to humanity.Monitoring and predicting the initiation of potential landslides is critical to avoiding losses due to disasters and economic activities.The impact of the controlled-source audio-frequency magnetotelluric method on investigating landslide surfaces is assessed through numerical simulations with a finite element approach.A Dirichlet boundary condition is selected to match the truncated boundary,resulting in a remarkable improvement in simulation efficiency.Rederivation of the formulas for a layered medium adept to the controlled-source audiofrequency magnetotelluric method is necessary to determine the electromagnetic field at any location along the truncated boundary.After the reliability evaluation of the new codes,a landslide model with a slide surface is designed,and the characteristics of its electromagnetic field and the apparent resistivity are studied.Instead of the total electromagnetic field,which is strongly infl uenced by topography variation,the apparent resistivity should be used for sliding surface detection.The normalized pure anomalous electromagnetic field may also be employed to quickly assess the detectability of the sliding surface.Overall,this study demonstrates that the controlled-source audio-frequency magnetotelluric method can be employed for investigating landslides,and recommends survey parameters,including configuration,frequency range,and length of survey line in landslide exploration.展开更多

CRISPR/Cas9 technology is a powerful genome manipulation tool in insects.However,little is known about whether mRNA and protein of a target gene are completely cleared in homozygous mutants.This study generated homozy...CRISPR/Cas9 technology is a powerful genome manipulation tool in insects.However,little is known about whether mRNA and protein of a target gene are completely cleared in homozygous mutants.This study generated homozygous mutants of the insulin receptor gene 2(NlInR2)in the brown planthopper(Nilaparvata lugens)using CRISPR/Cas9 genome editing.Both frameshift mutants,E5_D17 and E6_I7,differentiated towards long wings,but there were differences in wing morphology,with E5_D17 showing wing deformities.Subsequent investigations revealed the presence of residual expression of NlInR2 mRNA in both mutants,as well as the occurrence of spliceosomes featuring exon skipping splicing in E5_D17.Additionally,the E5_D17 exhibited the detection of N-terminally truncated NlInR2 protein.RNA interference experiments indicated that the knockdown of NlInR2 expression in the E5_D17 mutant line increased the proportion of wing deformities from 11.1 to 65.6%,suggesting that the residual NlInR2 mRNA of the E5_D17 mutant might have retained some genetic functions.Our results imply that systematic characterization of residual protein expression or function in CRISPR/Cas9-generated mutant lines is necessary for phenotypic interpretation.展开更多

This study presents the design of a modified attributed control chart based on a double sampling(DS)np chart applied in combination with generalized multiple dependent state(GMDS)sampling to monitor the mean life of t...This study presents the design of a modified attributed control chart based on a double sampling(DS)np chart applied in combination with generalized multiple dependent state(GMDS)sampling to monitor the mean life of the product based on the time truncated life test employing theWeibull distribution.The control chart developed supports the examination of the mean lifespan variation for a particular product in the process of manufacturing.Three control limit levels are used:the warning control limit,inner control limit,and outer control limit.Together,they enhance the capability for variation detection.A genetic algorithm can be used for optimization during the in-control process,whereby the optimal parameters can be established for the proposed control chart.The control chart performance is assessed using the average run length,while the influence of the model parameters upon the control chart solution is assessed via sensitivity analysis based on an orthogonal experimental design withmultiple linear regression.A comparative study was conducted based on the out-of-control average run length,in which the developed control chart offered greater sensitivity in the detection of process shifts while making use of smaller samples on average than is the case for existing control charts.Finally,to exhibit the utility of the developed control chart,this paper presents its application using simulated data with parameters drawn from the real set of data.展开更多
Trastuzumab resistance is one of the causes of poor prognosis in patients with human epidermal growth factor receptor 2(HER2)-positive(HER2+)breast cancer(BC).The truncated isoform of dopamine-and cAMPregulated phosph...Trastuzumab resistance is one of the causes of poor prognosis in patients with human epidermal growth factor receptor 2(HER2)-positive(HER2+)breast cancer(BC).The truncated isoform of dopamine-and cAMPregulated phosphoprotein(t-DARPP)has been reported to be involved in trastuzumab therapy resistance and promoting tumor progression.To evaluate the t-DARPP expression in BC,paired tumors and surrounding normal tissues were analyzed by real-time polymerase chain reaction and confirmed higher DARPP-32 kDa family mRNA expression in HER2+BC tumor tissues.We established 2 patient-derived xenografts(PDX)mice models to test the efficacy of trastuzumab,named model 1(non-responder)and model 2(responder).t-DARPP and p95-HER2 protein-protein interactions were detected in PDX tumor tissue from non-responders using Förster resonance energy transfer assays.Instead,there is no response from the responder.Furthermore,mechanistic studies using transwell and western blot assays demonstrated that t-DARPP could upregulate epithelial-mesenchymal transition signaling proteins,enhance p95-HER2 expression and promote cell migration.We found that quercetin effectively reduced t-DARPP expression in HER2+BC cells.In t-DARPP ShRNA-suppressed cells,quercetin synergistically enhanced trastuzumab-induced apoptotic cell death and G2/M phase arrest.In conclusion,the combination of quercetin and trastuzumab treatment by targeting t-DARPP in HER2+BC patients has the potential as a biomarker for mitigating drug resistance.展开更多

Phospholipase A1(PLA1)is a kind of specific phospholipid hydrolase widely used in food,medical,textile.However,limitations in its expression and enzymatic activity have prompted the investigation of the phospholipase-...Phospholipase A1(PLA1)is a kind of specific phospholipid hydrolase widely used in food,medical,textile.However,limitations in its expression and enzymatic activity have prompted the investigation of the phospholipase-assisting protein PlaS.In this study,we elucidate the role of PlaS in enhancing the expression and activity of PlaA1 through N-terminal truncation.Our research demonstrates that truncating the N-terminal region of PlaS effectively overcomes its inhibitory effect on host cells,resulting in improved cell growth and increased protein solubility of the protein.The yeast two-hybrid assay confirms the interaction between PlaA1 and N-terminal truncated PlaS(ΔN27 PlaS),highlighting their binding capabilities.Furthermore,in vitro studies using Biacore analysis reveal a concentration-dependent and specific binding between PlaA1 andΔN27 PlaS,exhibiting high affinity.Molecular docking analysis provides insights into the hydrogen bond interactions betweenΔN27 PlaS and PlaA1,identifying key amino acid residues crucial for their binding.Finally,the enzyme activity of PLA1 was boost to 8.4 U/mL by orthogonal test.Study significantly contributes to the understanding of the interaction mechanism between PlaS and PlaA1,offering potential strategies for enhancing PlaA1 activity through protein engineering approaches.展开更多

文摘We consider the multiplicity of solutions to a p(x)-Laplacian problem involving supercritical Sobolev growth via Ricceri’s principle.By means of truncation combined with De Giorgi iteration,we can extend the results of subcritical and critical growth to supercritical growth and obtain at least three solutions to the p(x)-Laplacian problem.
文摘In this paper,we introduce the real pairwise completely positive(RPCP)matrices with one of them is necessarily positive semidefinite while the other one is necessarily entrywise nonnegative,which has a real pairwise completely positive(RPCP)decomposition.We study the properties of RPCP matrices and give some necessary and sufficient conditions for a matrix pair to be RPCP.First,we give an equivalent decomposition for the RPCP matrices,which is different from the RPCP-decomposition and show that the matrix pair(X,X)is RPCP if and only if X is completely positive.Besides,we also prove that the RPCP matrices checking problem is equivalent to the separable completion problem.A semidefinite algorithm is also proposed for detecting whether or not a matrix pair is RPCP.The asymptotic and finite convergence of the algorithm are also discussed.If it is RPCP,we can further give a RPCP-decomposition for it;if it is not,we can obtain a certificate for this.
文摘A total of 393 potholes(368 fluvial and 25 marine potholes)were studied at seven different sites in both the fluvial and marine environments.Diverse bedrock properties and large-scale delivery of tools and grinders regulate the dynamic growth,truncation,and amalgamation of potholes.Therefore,the principal objectives of the study are(i)to examine the relationship between the growth of potholes and substrate lithological with structural characteristics(applying geospatial and Schmidt hammer for rock strength analysis)and(ii)to measure the morphology,and size of tools and grinders,processes of truncation and amalgamation in hydro-geomorphic environment using various indices and field techniques.The result showed that large potholes are stretched in the direction of lineament axes and roughly parallel to the river flow direction.Here,the steady growth of pothole depth-diameter is controlled by active bedrock structures,tools,or grinders,and monsoonal high-velocity bank full discharge.Consequently,the deepening and widening of potholes are relatively slow at Bindu,Deuli,and marine beach Neil Island due to fewer structures and little supply of tools or grinders.In small stretches,(Damodar,Subarnarekha,and Rarhu)canyons and gorge-like features(bedrock incision)are formed at Rajrappa,Bhakuyadi,and Guridih sites due to cyclic truncation and amalgamation.Truncation and amalgamation processes restrict the vertical depth threshold value of potholes within 3m,particularly at Rajrappa,Bhakuyadi,and Guridih sites.Scientific study of the pothole's dynamic growth is greatly necessary for the different environmental engineering and river hydraulic projects like excavation,dredging,and dam or barrage construction.Successively,it is essential to compute the cost of rock excavation or dredging,primarily for the mechanical strength of the bedrock river channel and its stability.

文摘While variable regions of immunoglobulins are extensively diversified by V(D)J recombination and somatic hypermutation in vertebrates,the constant regions of immunoglobulin heavy chains also utilize certain mechanisms to produce diversity,including class switch recombination(CSR),subclass differentiation,and alternative expression of the same gene.Many species of birds,reptiles,and amphibians express a truncated isoform of immunoglobulin Y(IgY),termed IgY(ΔFc),which lacks theυCH3 andυCH4 domains.In Anseriformes,IgY(ΔFc)arises from alternative transcriptional termination sites within the sameυgene,whereas in some turtles,intact IgY and IgY(ΔFc)are encoded by distinct genes.Different from the previously reported IgY(ΔFc)variants,this study identified a truncated IgY in the snake Elaphe taeniura,characterized by the loss of only a portion of the CH4 domain.Western blotting and liquid chromatographytandem mass spectrometry confirmed that this truncated IgY is generated by post-translational cleavage at N338 within the IgY heavy chain constant(CH)region.Furthermore,both human and snake asparaginyl endopeptidase were shown to cleave snake IgY in vitro.These findings reveal a novel mechanism for the production of shortened IgY forms,demonstrating that the immunoglobulin CH region undergoes diversification through distinct strategies across vertebrates.

文摘The count of one column for high-dimensional datasets, i.e., the number of records containing this column, has been widely used in nuinerous applications such as analyzing popular spots based on check-in location information and mining valuable items from shopping records. However, this poses a privacy threat when directly publishing this information. Differential privacy (DP), as a notable paradigm for strong privacy guarantees, is thereby adopted to publish all column counts. Prior studies have verified that truncating records or grouping columns can effectively improve the accuracy of published results. To leverage the advantages of the two techniques, we combine these studies to further boost the accuracy of published results. However, the traditional penalty function, which measures the error imported by a given pair of parameters including truncating length and group size, is so sensitive that the derived parameters deviate from the optimal parameters significantly. To output preferable parameters, we first design a smart penalty function that is less sensitive than the traditional function. Moreover, a two-phase selection method is proposed to compute these parameters efficiently, together with the improvement in accuracy. Extensive experiments on a broad spectrum of real-world datasets validate the effectiveness of our proposals.

文摘Alzheimer's disease is characterized by two major neuropathological hallmarks—the extracellularβ-amyloid plaques and intracellular neurofibrillary tangles consisting of aggregated and hyperphosphorylated Tau protein.Recent studies suggest that dysregulation of the microtubuleassociated protein Tau,especially specific proteolysis,could be a driving force for Alzheimer's disease neurodegeneration.Tau physiologically promotes the assembly and stabilization of microtubules,whereas specific truncated fragments are sufficient to induce abnormal hyperphosphorylation and aggregate into toxic oligomers,resulting in them gaining prion-like characteristics.In addition,Tau truncations cause extensive impairments to neural and glial cell functions and animal cognition and behavior in a fragment-dependent manner.This review summarizes over 60 proteolytic cleavage sites and their corresponding truncated fragments,investigates the role of specific truncations in physiological and pathological states of Alzheimer's disease,and summarizes the latest applications of strategies targeting Tau fragments in the diagnosis and treatment of Alzheimer's disease.

文摘This paper investigates the hydrodynamic characteristics of floating truncated cylinders undergoing horizontal and vertical motions due to earthquake excitations in the finite water depth.The governing equation of the hydrodynamic pressure acting on the cylinder is derived based on the radiation theory with the inviscid and incompressible assumptions.The governing equation is solved by using the method of separating variables and analytical solutions are obtained by assigning reasonable boundary conditions.The analytical result is validated by a numerical model using the exact artificial boundary simulation of the infinite water.The main variation and distribution characteristics of the hydrodynamic pressure acting on the side and bottom of the cylinder are analyzed for different combinations of wide-height and immersion ratios.The added mass coefficient of the cylinder is calculated by integrating the hydrodynamic pressure and simplified formulas are proposed for engineering applications.The calculation results show that the simplified formulas are in good agreement with the analytical solutions.

文摘Landslides are a type of natural disaster that can cause substantial harm to humanity.Monitoring and predicting the initiation of potential landslides is critical to avoiding losses due to disasters and economic activities.The impact of the controlled-source audio-frequency magnetotelluric method on investigating landslide surfaces is assessed through numerical simulations with a finite element approach.A Dirichlet boundary condition is selected to match the truncated boundary,resulting in a remarkable improvement in simulation efficiency.Rederivation of the formulas for a layered medium adept to the controlled-source audiofrequency magnetotelluric method is necessary to determine the electromagnetic field at any location along the truncated boundary.After the reliability evaluation of the new codes,a landslide model with a slide surface is designed,and the characteristics of its electromagnetic field and the apparent resistivity are studied.Instead of the total electromagnetic field,which is strongly infl uenced by topography variation,the apparent resistivity should be used for sliding surface detection.The normalized pure anomalous electromagnetic field may also be employed to quickly assess the detectability of the sliding surface.Overall,this study demonstrates that the controlled-source audio-frequency magnetotelluric method can be employed for investigating landslides,and recommends survey parameters,including configuration,frequency range,and length of survey line in landslide exploration.

文摘We study the nonlinear stage of modulation instability(MI)in the non-intergrable pure-quartic nonlinear Schrödinger equation where the fourth-order dispersion is modulated periodically.Using the three-mode truncation,we reveal the complex recurrence of parametric resonance(PR)breathers,where each recurrence is associated with two oscillation periods(PR period and internal oscillation period).The nonlinear stage of parametric instability admits the maximum energy exchange between the spectrum sidebands and central mode occurring outside the MI gain band.

文摘CRISPR/Cas9 technology is a powerful genome manipulation tool in insects.However,little is known about whether mRNA and protein of a target gene are completely cleared in homozygous mutants.This study generated homozygous mutants of the insulin receptor gene 2(NlInR2)in the brown planthopper(Nilaparvata lugens)using CRISPR/Cas9 genome editing.Both frameshift mutants,E5_D17 and E6_I7,differentiated towards long wings,but there were differences in wing morphology,with E5_D17 showing wing deformities.Subsequent investigations revealed the presence of residual expression of NlInR2 mRNA in both mutants,as well as the occurrence of spliceosomes featuring exon skipping splicing in E5_D17.Additionally,the E5_D17 exhibited the detection of N-terminally truncated NlInR2 protein.RNA interference experiments indicated that the knockdown of NlInR2 expression in the E5_D17 mutant line increased the proportion of wing deformities from 11.1 to 65.6%,suggesting that the residual NlInR2 mRNA of the E5_D17 mutant might have retained some genetic functions.Our results imply that systematic characterization of residual protein expression or function in CRISPR/Cas9-generated mutant lines is necessary for phenotypic interpretation.
文摘By applying the mastersymmetry of degree one to the time-independent symmetry K_(1), the fifth-order asymmetric Nizhnik-Novikov-Veselov system is derived. The variable separation solution is obtained by using the truncated Painlevé expansion with a special seed solution. New patterns of localized excitations, such as dromioff, instanton moving on a curved line, and tempo-spatial breather, are constructed. Additionally, fission or fusion solitary wave solutions are presented,graphically illustrated by several interesting examples.
文摘In this paper,we establish a new multivariate Hermite sampling series involving samples from the function itself and its mixed and non-mixed partial derivatives of arbitrary order.This multivariate form of Hermite sampling will be valid for some classes of multivariate entire functions,satisfying certain growth conditions.We will show that many known results included in Commun Korean Math Soc,2002,17:731-740,Turk J Math,2017,41:387-403 and Filomat,2020,34:3339-3347 are special cases of our results.Moreover,we estimate the truncation error of this sampling based on localized sampling without decay assumption.Illustrative examples are also presented.

文摘This study presents the design of a modified attributed control chart based on a double sampling(DS)np chart applied in combination with generalized multiple dependent state(GMDS)sampling to monitor the mean life of the product based on the time truncated life test employing theWeibull distribution.The control chart developed supports the examination of the mean lifespan variation for a particular product in the process of manufacturing.Three control limit levels are used:the warning control limit,inner control limit,and outer control limit.Together,they enhance the capability for variation detection.A genetic algorithm can be used for optimization during the in-control process,whereby the optimal parameters can be established for the proposed control chart.The control chart performance is assessed using the average run length,while the influence of the model parameters upon the control chart solution is assessed via sensitivity analysis based on an orthogonal experimental design withmultiple linear regression.A comparative study was conducted based on the out-of-control average run length,in which the developed control chart offered greater sensitivity in the detection of process shifts while making use of smaller samples on average than is the case for existing control charts.Finally,to exhibit the utility of the developed control chart,this paper presents its application using simulated data with parameters drawn from the real set of data.

文摘Trastuzumab resistance is one of the causes of poor prognosis in patients with human epidermal growth factor receptor 2(HER2)-positive(HER2+)breast cancer(BC).The truncated isoform of dopamine-and cAMPregulated phosphoprotein(t-DARPP)has been reported to be involved in trastuzumab therapy resistance and promoting tumor progression.To evaluate the t-DARPP expression in BC,paired tumors and surrounding normal tissues were analyzed by real-time polymerase chain reaction and confirmed higher DARPP-32 kDa family mRNA expression in HER2+BC tumor tissues.We established 2 patient-derived xenografts(PDX)mice models to test the efficacy of trastuzumab,named model 1(non-responder)and model 2(responder).t-DARPP and p95-HER2 protein-protein interactions were detected in PDX tumor tissue from non-responders using Förster resonance energy transfer assays.Instead,there is no response from the responder.Furthermore,mechanistic studies using transwell and western blot assays demonstrated that t-DARPP could upregulate epithelial-mesenchymal transition signaling proteins,enhance p95-HER2 expression and promote cell migration.We found that quercetin effectively reduced t-DARPP expression in HER2+BC cells.In t-DARPP ShRNA-suppressed cells,quercetin synergistically enhanced trastuzumab-induced apoptotic cell death and G2/M phase arrest.In conclusion,the combination of quercetin and trastuzumab treatment by targeting t-DARPP in HER2+BC patients has the potential as a biomarker for mitigating drug resistance.

文摘We introduce a factorized Smith method(FSM)for solving large-scale highranked T-Stein equations within the banded-plus-low-rank structure framework.To effectively reduce both computational complexity and storage requirements,we develop techniques including deflation and shift,partial truncation and compression,as well as redesign the residual computation and termination condition.Numerical examples demonstrate that the FSM outperforms the Smith method implemented with a hierarchical HODLR structured toolkit in terms of CPU time.

文摘Phospholipase A1(PLA1)is a kind of specific phospholipid hydrolase widely used in food,medical,textile.However,limitations in its expression and enzymatic activity have prompted the investigation of the phospholipase-assisting protein PlaS.In this study,we elucidate the role of PlaS in enhancing the expression and activity of PlaA1 through N-terminal truncation.Our research demonstrates that truncating the N-terminal region of PlaS effectively overcomes its inhibitory effect on host cells,resulting in improved cell growth and increased protein solubility of the protein.The yeast two-hybrid assay confirms the interaction between PlaA1 and N-terminal truncated PlaS(ΔN27 PlaS),highlighting their binding capabilities.Furthermore,in vitro studies using Biacore analysis reveal a concentration-dependent and specific binding between PlaA1 andΔN27 PlaS,exhibiting high affinity.Molecular docking analysis provides insights into the hydrogen bond interactions betweenΔN27 PlaS and PlaA1,identifying key amino acid residues crucial for their binding.Finally,the enzyme activity of PLA1 was boost to 8.4 U/mL by orthogonal test.Study significantly contributes to the understanding of the interaction mechanism between PlaS and PlaA1,offering potential strategies for enhancing PlaA1 activity through protein engineering approaches.
