Cyclocarya paliurus(Batalin)Iljinskaja,as a unique and rare monocotyledonous plant in Southern China,is a promising and economical Chinese herbal medicine and functional food.People have conducted a number of research on C.paliurus because of its rich triterpenoids.However,no comprehensive review has illustrated the composition and pharmacological activity of triterpenoids from C.paliurus.This review summarizes 177 triterpenoids from different parts of C.paliurus.The structures of compounds were elucidated,and their biosynthesis was inferred.The biological activities of compounds and triterpenoid-rich extracts,including anti-diabetes,antihyperlipidemia,anti-inflammatory,anticancer or cytotoxicity,antioxidation,etc.,were discussed.C.paliurus can be an important and valuable supplement to the food market.This review provides a reference for the further research and application of C.paliurus triterpenoids in the fields of foods and pharmaceuticals. 展开更多

Gentianopsis barbata(G.barbata)represents a significant plant species with considerable ornamental and medicinal value in China.This investigation sought to elucidate the primary constituents within the plant and investigate their pharmacological properties.Fifty triterpenoids(1-50),including nine previously undescribed compounds(1,2,7,10,20,28,29,37,and 41)were isolated and characterized from the whole plants of G.barbata.Notably,compounds 1 and 2 exhibited the novel 3,4;9,10-diseco-24-homo-cycloartane triterpenoid skeleton.The isolated triterpenoids demonstrated substantial anti-inflammatory activity through inhibition of tumor necrosis factorα(TNF-α)and interleukin-6(IL-6)cytokine secretion in LPS-induced RAW264.7 macrophages,and hepatoprotective effects by preventing tertbutyl hydroperoxide(t-BHP)-induced oxidative injury in HepG2 cells.These results demonstrate both the presence of diverse triterpenoids in G.barbata and their therapeutic potential for inflammatory and hepatic conditions,providing scientific evidence supporting the clinical application of this traditional Mongolian medicinal plant. 展开更多

Discovery of fernane-type triterpenoids from Diaporthe discoidispora using genome mining and HSQC-based SMART technology

In this study,we employed a combination of genome mining and heteronuclear single quantum coherence(HSQC)-based small molecule accurate recognition technology(SMART)technology to search for fernane-type triterpenoids.Initially,potential endophytic fungi were identified through genome mining.Subsequently,fine fractions containing various fernane-type triterpenoids were selected using HSQC data collection and SMART prediction.These triterpenoids were then obtained through targeted isolation and identification.Finally,their antifungal activity was evaluated.As a result,three fernane-type triterpenoids,including two novel compounds,along with two new sesquiterpenes and four known compounds were isolated from one potential strain,Diaporthe discoidispora.Their structures were elucidated through analysis of high-resolution electrospray ionization mass spectrometry(HR-ESI-MS)and nuclear magnetic resonance(NMR)spectroscopic data.The absolute configurations were determined using single-crystal X-ray diffraction analysis and electron capture detector(ECD)analysis.Compound 3 exhibited moderate antifungal activity against Candida albicans CMCC 98001 and Aspergillus niger. 展开更多

Triterpenoids widely exist in nature,displaying a variety of pharmacological activities.Determining triterpenoids in different matrices,especially in biological samples holds great significance.High-performance liquid chromatography(HPLC)has become the predominant method for triterpenoids analysis due to its exceptional analytical performance.However,due to the structural similarities among botanical samples,achieving effective separation of each triterpenoid proves challenging,necessitating significant improvements in analytical methods.Additionally,triterpenoids are characterized by a lack of ultraviolet(UV)absorption groups and chromophores,along with low ionization efficiency in mass spectrometry.Consequently,routine HPLC analysis suffers from poor sensitivity.Chemical derivatization emerges as an indispensable technique in HPLC analysis to enhance its performance.Considering the structural characteristics of triterpenoids,various derivatization reagents such as acid chlorides,rhodamines,isocyanates,sulfonic esters,and amines have been employed for the derivatization analysis of triterpenoids.This review comprehensively summarized the research progress made in derivatization strategies for HPLC detection of triterpenoids.Moreover,the limitations and challenges encountered in previous studies are discussed,and future research directions are proposed to develop more effective derivatization methods. 展开更多

Five ganoderic acids, ganoderic acid C2, C6, G, B and A were simultaneously detected in rat plasma. Their structures were identified by the spectroscopic analyses(IR, ESI-MS, HR-ESI-MS, and 1D and 2D NMR) and chemical reactions. Wolf, an important medicinal and food fungus, is well known in East Asia. Due to growing market demand, long cultivation period, and consumption of pine trunk during cultivation, developing alternative methods for producing P. cocos and/or its active components is of interest. In the present study, the effects of different culture methods on biomass and accumulation of four triterpenoids were investigated. The ethanol extract of fermented mycelium(EFM) was orally administered to rats. Urine output and concentrations of electrolytes(Na^+, K^+, and Cl^-) were measured. Our results showed that mycelia grew better under continuous shaking culture condition(7.5 g DW·L^(-1)), and higher triterpenoid levels were accumulated in two-stage culture(112 mg·L-1, 2.03%). The optimal starting time of static culture for triterpenoid yield was 4^(th) d after shaking culture. Single administration of middle and high dose of EFM significantly increased urine output, Na^+ and Cl^- excretion, and Na^+/K^+ ratio. These results suggested that ethanol extract of cultured mycelia showed significant diuretic activity in rats and two-stage culture of P. cocos could be an alternative way to produce mycelia and triterpenoids. 展开更多

Cyclocarya paliurus is widely distributed in subtropical areas of China.Secondary metabolites in the leaves,including flavonoids and triterpenoids,provide protection against diseases such as hyperlipidemia,diabetes,hypertension,and hypoimmunity.In this study,343 C.paliurus leaves were collected from 32 distinct populations covering most of the main distribution,to deduce the response of flavonoids and triterpenoids to seven environmental factors.Principal component analysis was performed using seven flavonoid and six triterpenoid compounds.The results show that altitude,latitude,and longitude significantly contributed to variations in total flavonoid and triterpenoid contents.In addition,the interaction of these metabolites was extensively correlated with environment changes,and therefore should be considered during selection and cultivation. 展开更多

Betulin, oleanolic acid, and betulinic acid are naturally occurring pentacyclic triterpenoids that have significant medicinal value. Considerable amounts of these triterpenoids are available in the outer bark of white birch. In this study, we used ultrasound-assisted extraction (UAE) to extract triterpenoids from birch bark rapidly and with high efficiency. Using high performance liquid chro- matography (HPLC), three types of triterpenoids were separated and detected. We examined the differences among triterpenoids extracted from diploid versus tetra- ploid white birch. Then, we used factor analysis to screen out tetraploid white birches with comprehensively excel- lent performance. The results indicate that the optimum conditions for extraction include the use of ethanol as an extraction solvent, a solid-to-liquid ratio of 0.1 g/10 ml, ultrasonic power set at 100 W, a temperature of 60 ℃ and an extraction time of 15 min. A reversed-phase C18 col- umn (4.6 mm × 250 mm × 5 μm) with a column tem- perature of 30 ℃ and the mobile phase composed of A (acetonitrile) and B (0.1% aqueous phosphoric acid, v/v) at a flow rate of 0.5 ml/min were used, and the detection wavelength was 195 nm. No significant difference wasobserved between diploid and tetraploid white birch in terms of the content of three types of triterpenoids (at a confidence level of 0.05). As triterpenoid content, height, and DBH (diameter at breast height) are strongly interre- lated, we used factor analysis to evaluate all individuals, and we screened out six plus trees with excellent com- prehensive characters. 展开更多

Three new limonoid-type triterpenoids,namely toonasins A–C(1–3)with a rare lactam E ring,along with six known compounds(4–9)were isolated from the barks of Toona sinensis.The structures of new compounds were elucidated by interpretation of spectroscopic data,and the relative configuration of compound 1 was further characterized by X-ray crystallographic analyses.The isolated compounds were evaluated for their cytotoxic activities against five human tumor cell lines(HL-60,SMMC-7721,A-549,MCF-7 and SW480),and compounds 3 and 5 showed weak cytotoxicities. 展开更多
