Carbon Footprint and Economic Analysis of LNG-fueled Fishing Vessel Using Real Engine Performance Simulation

Analysis of the environmental and economic performance of fishing vessels has received limited attention compared with other ship types despite their notable contribution to global greenhouse gas(GHG)emissions.This study evaluates the carbon footprint(CF)and economic viability of a liquefied natural gas(LNG)-fueled fishing vessel,using real engine operation simulations to provide precise and dynamic evaluation of fuel consumption and GHG emissions.Operational profiles are obtained through the utilization of onboard monitoring systems,whereas engine performance is simulated using the 1D/0D AVL Boost^(TM)model.Life cycle assessment(LCA)is conducted to quantify the environmental impact,whereas life cycle cost assessment(LCCA)is performed to analyze the profitability of LNG as an alternative fuel.The potential impact of the future fuel price uncertainties is addressed using Monte Carlo simulations.The LCA findings indicate that LNG has the potential to reduce the CF of the vessel by 14%to 16%,in comparison to a diesel power system configuration that serves as the baseline scenario.The LCCA results further indicate that the total cost of an LNG-powered ship is lower by 9.5%-13.8%,depending on the share of LNG and pilot fuels.This finding highlights the potential of LNG to produce considerable environmental benefits while addressing economic challenges under diverse operational and fuel price conditions. 展开更多

Parkinson’s disease is characterized by synucleinopathy-associated neurodegeneration.Previous studies have shown that glucagon-like peptide-1(GLP-1)has beneficial effects in a mouse model of Parkinson’s disease induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine.However,the effect of GLP-1 on intrinsic synuclein malfunction remains unclear.In this study,we investigated the effect of Lactococcus lactis MG1363-pMG36e-GLP-1 on parkinsonism in SncaA53T transgenic mice and explored the underlying mechanisms.Our data showed that Lactococcus lactis MG1363-pMG36e-GLP-1 inhibited dopaminergic neuronal death,reduced pathological aggregation ofα-synuclein,and decreased movement disorders in SncaA53T transgenic mice.Furthermore,Lactococcus lactis MG1363-pMG36e-GLP-1 downregulated lipopolysaccharide-related inflammation,reduced cerebral activation of microglia and astrocytes,and promoted cell survival via the GLP-1 receptor/PI3K/Akt pathway in the substantia nigra.Additionally,Lactococcus lactis MG1363-pMG36e-GLP-1 decreased serum levels of pro-inflammatory molecules including lipopolysaccharide,lipopolysaccharide binding protein,interleukin-1β,and interleukin-6.Gut histopathology and western blotting further revealed that Lactococcus lactis MG1363-pMG36e-GLP-1 increased the expression of gut integrity-related proteins and reduced lipopolysaccharide-related inflammation by reversing gut dysbiosis in SncaA53T transgenic mice.Our findings showed that the beneficial effect of Lactococcus lactis MG1363-pMG36e-GLP-1 on parkinsonism traits in SncaA53T transgenic mice is mediated by microglial polarization and the reversal of dysbiosis.Collectively,our findings suggest that Lactococcus lactis MG1363-pMG36e-GLP-1 is a promising therapeutic agent for the treatment of Parkinson’s disease. 展开更多

以假俭草(Eremochloa ophiuroides(Munro)Hack.)品种‘赣北’(‘Ganbei’)基因组序列数据为基础,通过生物信息学手段挖掘和设计了假俭草染色体特异的单拷贝寡核苷酸(oligo)探针库,同时结合端粒重复序列和rDNA重复序列探针,在假俭草根尖... 以假俭草(Eremochloa ophiuroides(Munro)Hack.)品种‘赣北’(‘Ganbei’)基因组序列数据为基础,通过生物信息学手段挖掘和设计了假俭草染色体特异的单拷贝寡核苷酸(oligo)探针库,同时结合端粒重复序列和rDNA重复序列探针,在假俭草根尖细胞有丝分裂中期染色体上进行了系列荧光原位杂交(FISH)试验。结果显示:假俭草染色体核型公式为2n=2x=18(2SAT),利用5S rDNA和18S rDNA重复序列探针可识别出第5和第4号染色体,且第4号染色体18S rDNA富集区段易出现或存在断裂现象;设计的假俭草染色体特异单拷贝oligo探针可识别出第2、第6和第9号染色体,进一步结合rDNA重复序列探针结果还发现第4号染色体与第8号染色体间发生了染色体易位。综合分析认为,假俭草染色体核型存在不稳定性,且其基因组在进化过程中发生了染色体结构重排。本研究建立的基于重复序列和单拷贝oligo探针的FISH技术体系,可为假俭草染色体的准确识别和变异鉴定提供新方法,观察到的假俭草染色体核型特征可为假俭草高质量基因组图谱的构建提供细胞学证据。 展开更多

Photodynamic therapy(PDT) employs accumulation of photosensitizers(PSs) in malignant tumor tissue followed by the light-induced generation of cytotoxic reactive oxygen species to kill the tumor cells. The success of PDT depends on optimal PS dosage that is matched with the ideal power of light. This in turn depends on PS accumulation in target tissue and light administration time and period.As theranostic nanomedicine is driven by multifunctional therapeutics that aim to achieve targeted tissue delivery and image-guided therapy, fluorescent PS nanoparticle(NP)accumulation in target tissues can be ascertained through fluorescence imaging to optimize the light dose and administration parameters. In this regard, zebrafish larvae provide a unique transparent in vivo platform to monitor fluorescent PS bio-distribution and their therapeutic efficiency. Using fluorescent PS NPs with unique aggregation-induced emission characteristics, we demonstrate for the first time the real-time visualization of polymeric NP accumulation in tumor tissue and, more importantly, the best time to conduct PDT using transgenic zebrafish larvae with inducible liver hyperplasia as an example. 展开更多

目的:准确的间变性淋巴瘤激酶(anaplastic lymphoma kinase,ALK)重排检测在非小细胞肺癌(non-small cell lung cancer,NSCLC)患者ALK抑制剂(anaplastic lymphoma kinase tyrosine kinase inhibitors,ALK-TKI)靶向治疗中至关重要。目前,... 目的:准确的间变性淋巴瘤激酶(anaplastic lymphoma kinase,ALK)重排检测在非小细胞肺癌(non-small cell lung cancer,NSCLC)患者ALK抑制剂(anaplastic lymphoma kinase tyrosine kinase inhibitors,ALK-TKI)靶向治疗中至关重要。目前,医院实验室所使用的检测方法有基于逆转录PCR(reverse transcription-PCR,RT-PCR)、荧光原位杂交(fluorescence in situ hybridization,FISH)和二代测序(next generation sequencing,NGS)。本文评估了三种方法检测ALK重排的性能,为ALK的精确检测提供理论支持。方法:选取38例ALK阳性患者,分别用RT-PCR、FISH和NGS对样本进行ALK重排检测,评估三种方法在ALK检测中的一致性。再分别分析这三种ALK方法与ALK-TKI治疗效果的一致性。结果:分别用RT-PCR、FISH和NGS方法检测38例得到的ALK重排阳性率分别是89.47%、86.84%和97.37%,其中,NGS检测出3例罕见ALK重排。RT-PCR与FISH方法在检查ALK重排结果有较强的一致性(Kappa=0.623,P<0.05);NGS与RT-PCR、FISH检测ALK重排没有一致性(Kappa=-0.044和0.303)。该三组方法定义的ALK阳性病例经ALK-TKI治疗22个月的存活数,统计分析结果显示差异无统计学意义(P>0.05)。结论:与RT-PCR和FISH相比,NGS方法显示出更高的阳性检出率,特别是在罕见的ALK重排中具有更强的检测能力,因此,本研究倡导在实验条件满足的情况下采用NGS来检测ALK重排,能有效提升NSCLC患者的治疗效果和预后评估。 展开更多

Background: The zebrafish(Danio rerio) has recently been shown to be an ideal model to study bone disease including osteoporosis. The zebrafish osteoporosis model could be induced by glucocorticoid treatment with chemical staining for reflecting the level of bone mineralization. However, this methodology was unstable.Here, we developed a novel methodology to directly evaluate the bone mass and density.Methods: We generated and used the bone of transgenic zebrafish Tg(ola.sp7:nlsGFP) to evaluate the bone mass and density by measuring the areal extent and the integrated optical density(IOD) of enhanced green fluorescent protein(eGFP).This methodology was further compared with the traditional chemically stained method showing the bone mineralization. Furthermore, genes related to zebrafish osteoporosis were analyzed by quantitative reverse transcription-polymerase chain reaction(qRT-PCR).Results: Our results of new methods were consistent with those from chemically stained fish, following glucocorticoid-induction or epimedium flavonoid(FE)-rescue treatments. q RT-PCR analyses on mRNA levels revealed that glucocorticoid induces osteoporosis by downregulating the expression of osteoblast-related factors osterix,osteocalcin, and osteopontin, and upregulating the expression of osteoclast-related factor tartrate-resistant acid phosphatase. In FE-rescued fish, the expression of osteogenic factors osterix, osteocalcin, and osteopontin were increased.Conclusion: Compared to the traditional chemical staining methods, the new osteoporosis model using Tg(ola.sp7:nlsGFP) is more convenient and efficient for studying osteoporosis in vivo, and especially for high-throughput anti-osteoporosis drug screening. 展开更多

A recombinant plasmid (pcMTsGH) containing the common carp methlothionein promoter (cMT) and salmon growth hormone gene (sGH) was microinjected into common carp fertilized eggs and embryos to optimize the conditions for producing transgenic fish. Several key elements were found to have significant effects on the integration of exogenous gene. These elements include forms and doses of pcMTsGH gene microinjected, stages of embryo at which microinjection conducted and pretreatment of fertilized eggs for microinjection. The results show that, in producing transgenic fish, (1) the best time for microinjection is late onecell stage, (2) the most appropriate dose of pcMTsGH gene for microinjection is around 106~107 copies; (3) the hatching rate rises up when the gene is microinjected into the eggs whose chorions are removed, but the integration rate has no significant changes before and after the removal of egg chorions. 展开更多

Conditional expression of a target gene during zebrafish development is a powerful approach to elucidate gene functions. The tetracycline-controlled systems have been successfully used in the modulation of gene expression in mammalian cells, but few lines of zebrafish carrying these systems are currently available. In this study, we had generated a stable transgenic zebrafish line that ubiquitously expressed the second-generation of reverse Tet transactivator (rtTA-M2). Southern blotting analysis and high-throughput genome sequencing verifed that a single copy of rtTA-M2 gene had stably integrated into the zebrafish genome. After induction with doxycycline (Dox), a strong green fluorescent protein (GFP) was seen in rtTA-transgenic eggs injected with pTRE--EGFP plasmids. The fluorescent signal gradually decreased after the withdrawal of Dox and disappeared. However, leaky expression of GFP was undetectable before Dox- induction. Additionally, transgenic embryos expressing rtTA-M2 exhibited no obvious defects in morphological phenotypes, hatching behavior and expression patterns of developmental marker genes, suggesting that rtTA-M2 had little effect on the development of transgenic zebrafish. Moreover, expressed Dickkopf-1 (DKK1) in pTRE-DKKl-injected embryos led to alterations in the expression of marker genes associated with Wnt signaling. Thus, this rtTA-transgenic zebrafish can be utilized to dissect functions of genes in a temporal manner. 展开更多

Marine fish diversity in the Beibu Gulf from 2014 to 2022 was analyzed,using various methods including at-sea fisheries resource surveys,fishing port surveys,underwater survey techniques,and data from literature,books,and databases,from which 1059 fish species across 40 orders,166 families,and 503 genera were identified.Among them,Acanthuriformes displayed the highest diversity,followed by Carangiformes and Perciformes.Notably,eight alien species were found in the Beibu Gulf.The International Union for Conservation of Nature(IUCN)Red List assessment revealed 51threatened species,primarily cartilaginous fish.Taxonomic changes affected 88.70%of species due to classification adjustments,misidentifications,and junior synonymy.This study contributed new data of the gulf,including one additional order,six new families,34 new genera,and 81 new species,emphasizing the dynamic nature of marine ecosystems.Fish diversity in the gulf was relatively lower than the broader South China Sea,attributed to limited habitats and survey methods.Different survey methods,such as gill nets,trap nets,and underwater cameras,yielded varying results.Ocean currents may have transported deep-sea species into the Beibu Gulf,leading to accidental records.Research gaps exist in unexplored areas,warranting further investigation.Therefore,the Beibu Gulf hosts significant marine biodiversity,but taxonomic challenges and undiscovered species require new research and conservation efforts. 展开更多

Environmental DNA assessment of fish diversity, distribution and niche characteristics in Zhutuo spawning ground in the upper reaches of the Yangtze River

[Objective]Implementation of the Ten-Year Fishing Ban policy may alter fish diversity and niche characteristics of dominant species in spawning grounds within the National Nature Reserve for Rare and Endemic Fish in the Upper Yangtze River.This study initiated continuous monitoring of natural spawning habitats from February 2022 to assess these ecological changes.[Methods]Environmental DNA(eDNA)metabarcoding was employed to analyze fish species composition,biodiversity patterns,and niche parameters of dominant species.Water sampling followed the CEN/TS 19461 standard across five monitoring transects(ZT1-ZT5).[Results]The eDNA analysis detected 45 species of fish belonging to 38 genera,13 families,and 3 orders were detected through environmental DNA(eDNA)in this survey,including 10 species endemic to the upper reaches of the Yangtze River,such as Procypris rabaudi and Myxocyprinus asiaticus.The fish community was mainly composed of bottom-dwelling,settling ovum-producing,omnivorous fish.The variation ranges of the Chao1 index,ACE index,Shannon index,and Simpson index are 736~996,719~965,1.58~3.23,and 0.83~0.99,respectively,indicating that fish species in spawning sites are abundant and community distribution uniformity is high.All indexes are highest at ZT1 monitoring points.Cluster analysis showed that,at a certain similarity level,fish community types in spawning sites could be basically divided into two groups:ZT1,ZT3,and ZT5 clustered together,and ZT2 and ZT4 clustered together,indicating similar fish community habitats.There are 9 dominant fish species in typical deep pool habitats in the reserve,with niche widths(Bi)ranging from 1.13 to 3.87.The dominant fish species are broad and medium niche fish,such as Cyprinus carpio and Hemiculter tchangi,with the niche overlap index(Oik)of some dominant fish species reaching more than 0.95.This indicates fierce competition for resources among the fish in this spawning ground.[Conclusion]The Zhutuo spawning ground demonstrates high species richness with homogeneous community structure and intense resource competition.This study establishes an eDNA-based monitoring framework that enhances conventional survey method,providing critical baseline data for adaptive management under the fishing moratorium regime. 展开更多

Over the last few decades, dengue fever epidemics have increased in frequency and intensity worldwide, making it a major global concern for public health. Its prevention, which is essentially vector-based control, is already being compromised by reports of resistance of the main vector Aedes aegypti to insecticides. To tackle the rapid increase in insecticide resistance and outbreaks, the biological vector control is a promising approach. One of the strategies of this approach is the use of entomopathogenic fungi because of their great efficacy and their eco-friendly aspects. However, some aspects of their use, such as the low efficiency, the high cost of production and the sensitivity to various adverse conditions, need to be addressed for their successful large-scale application. Therefore, innovative technologies based on strains of transgenic fungi with improved biocontrol potentials by genetic engineering are actively pursued. Although these modified mycoinsecticides are acclaimed for their better effectiveness against target insects, the main concern remains their potential adverse effects on the environment and human health. The present review is dedicated to giving an update on recent developments in transgenic entomopathogenic fungi (TEF) for Aedes mosquito control. Future perspectives are also proposed to address the safety concerns related to the release of transgenic entomopathogenic fungi into the environment. 展开更多

We report here arsenic speciation in 1643 freshwater fish samples,representing 14 common fish species from 53 waterbodies in Alberta,Canada.Arsenic species were extracted from fish muscle tissue.Arsenic species in the extracts were separated using anion-exchange high-performance liquid chromatography(HPLC)and quantified using inductively coupled plasma mass spectrometry(ICPMS).The total arsenic concentrations in fish ranged from 2.8 to 1200μg/kg(in wet weight of sample)(mean 71±101μg/kg),which are all below the 2000μg/kg(wet weight)maximum allowable total arsenic in fish,recommended by the Ontario Ministry of the Environment.In 99.7%,or 1638 of all 1643 freshwater fish samples analyzed,arsenobetaine(AsB)was detectable,with concentrations higher than the method detection limit of 0.25μg/kg(wet weight).Dimethylarsinic acid(DMA)was detectable(concentration>0.25μg/kg)in 92.1%,or 1514 of the 1643 freshwater fish samples.Inorganic arsenate(iAs^(Ⅴ))was detectable(>0.25μg/kg)in 1119 fish(i.e.,68.1% of 1643 samples).Monomethylarsonic acid(MMA)was detectable(>0.25μg/kg)in 418 fish(25.4%of 1643 samples).The concentrations of arsenic species in the 1643 fish samples varied by as much as three orders of magnitude,ranging from below the method detection limit of 0.25μg/kg to the maximum concentrations of 380μg/kg for AsB,150μg/kg for DMA,70μg/kg for iAs^(Ⅴ),and 51μg/kg for MMA.AsB made up 46.1%±26.2% of total arsenic species.Arsenic speciation patterns varied between lake whitefish,northern pike,and walleye,the three most common types of fish analyzed.The relative proportion of DMA in northern pike was larger than in lake whitefish and walleye,and conversely,the relative proportion of iAs^(Ⅴ) was lower in northern pike.Seven unknown arsenic species were detected,and their chromatographic retention time did not match with those of available arsenic standards.At least one unknown arsenic species was detected in 33.4%,or 549 of 1643 freshwater fish samples.The concentrations of unknown arsenic species were as high as 61μg/kg.Future research is necessary to identify unknown arsenic species and to determine contributing factors to the observed arsenic species patterns and concentrations. 展开更多

Current status and diversity of fish resources in the Pinglu Canal Century Project

[Objective]The channel straightening project of the Pinglu Canal has fragmented the river course,compromising the integrity of original river course and causing ecosystem patchiness.Understanding the current status of fish resources and the characteristics of their diversity is crucial for the ecological management of the Pinglu Canal.[Methods]During the spring and autumn in 2021 and 2022,a survey of fish resources and species diversity in the Pinglu Canal was conducted using multi-mesh gill nets.A total of 125 fish species were collected,belonging to 10 orders,34 families,and 89 genera.[Results]The result showed that the Pinglu Canal contained three nationally protected Class II species,two endemic species of the Qinjiang River,three anadromous/migratory species,and eight invasive species,accounting for 2.4%,1.6%,2.4%,and 6.4%of the total species,respectively.The fish community primarily consisted of mid-and bottom-dwelling,adhesive-egg-laying,and omnivorous species.The Shannon-Wiener,Simpson,Margalef,and Pielou indices of the fish community in the Pinglu Canal ranged from 2.347 to 2.757,0.081 to 0.151,3.493 to 4.382,and 0.812 to 0.892,respectively.These indices showed relatively uniform distribution across different river reaches.[Conclusion]The result indicate that the fish community structure in the Pinglu Canal is relatively uniform.The reach from the Yujiang River to the Shaping River shows higher stability,while other river reaches experience moderate or severe disturbances.This study provides supplementary baseline data on the fish community structure in the Pinglu Canal and explores the potential impact of inter-basin connectivity on fish resources,aiming to provide a scientific basis for habitat restoration assessments after the channel straightening project. 展开更多

Captive breeding and stocking are commonly employed strategies for enhancing fisheries and conserving endangered fish species.However,hatchery-raised fish often exhibit reduced performance in the wild,displaying alterations in physiological,morphological,and behavioral traits.We tested for differences in swimming capacity and metabolic traits between wild and hatchery-reared individuals of the Spanish toothcarp(Aphanius iberus)from 2 different populations.Furthermore,we experimentally tested if these changes translated into fitness differences after their stocking into the wild.There were significant differences in swimming capacity and metabolic traits between wild and hatchery-reared individuals and also between the 2 populations.Captive-bred individuals displayed consistently lower metabolic rates than wild individuals from the same population(30-76% lower).Critical swimming speed rather differed between the 2 populations.Sex-specific differences were observed in maximum and standard metabolic rates,with wild individuals and females generally exhibiting higher values but with some exceptions.During a 3-month experiment,survival rates did not significantly differ between wild and captive-bred fish.Captive-bred individuals started smaller but exhibited rapid growth during the experiment.Initially,larger captive-bred fish had lower body conditions than their wild counterparts,but these differences progressively diminished.In summary,captive-bred individuals of this fish species showed lower metabolic rates,although the differences with wild individuals slightly depended on sex and size. 展开更多

Nutritive and Microbiological Quality of Fish Feed Formulated from Local Flours Enriched with Larvaes of Hermetia illucens

In aquaculture, feed represents the main component of production costs, and the development of this sector depends on the development of an economical feed formulation that meets the qualitative and quantitative requirements of fish. The aim of this study was to determine the nutritive and microbiological quality of fish feed formulated from local flours enriched with Hermetia illucens larvae. The raw materials used for formulation were fishmeal, corn meal, low-grade rice, soybean meal and Hermetia illucens larvae meal. Different iso-protein feed compositions were prepared with 0%, 10%, 25%, 35%, 50%, 65%, 75% and 100% incorporation of Hermetia illucens larvae meal as a substitute for fish meal. Biochemical and microbiological analyses of these flours were determined using standard methods. The results showed that incorporation of larvae meal had an influence on the biochemical characteristics ash (8.15 to 20.27%), lipid (11.55 to 24.94%), fiber (13.93 to 20.41%) and dry matter (89.65 to 91.19%) of various formulated feed. Loads of fecal Streptococci, Staphylococci, Aeromonas, yeasts and molds ranged from 2.4 to 4.9 log 10 CFU/g;3.6 to 3.9 log 10 CFU/g;2.2 to 2.7 log 10 CFU/g;2.1 to 2.3 log 10 CFU/g, respectively. The level of contamination of these flours was below the microbiological criteria applicable to animal feed. Feed formulated with 0% and 10% Hermetia illucens larvae showed the best nutritive and microbiological characteristics. These results suggest that flours enriched with Hermetia illucens larvae could be used in fish feed. 展开更多

Development of a piggy Bac transgenic system in Bactrocera dorsalis and its potential for research on olfactory molecular targets

Chemicals that modify pest behavior are developed to reduce crop damage by altering pest behavior, using specific genes within the olfactory system as molecular targets. The identification of these molecular targets in Bactrocera dorsalis, also known as the functional study of key olfactory genes, relies on CRISPR/Cas9-mediated gene knockout techniques. However, these techniques face limitations when applied to lethal genes. Transgenic technology offers a solution since it enables precise manipulation of gene expression in specific tissues or during certain developmental stages. Consequently, this study developed a piggyBac-mediated transgenic system in B. dorsalis to investigate reporter gene expression in olfactory organs, and assessed the olfactory behavior andantennal electrophysiological responses in transgenic lines. The goal was to assess the potential of this approach for future research on olfactory gene function. A universally expressed housekeeping gene from the BdorActin family was identified using the developmental transcriptome dataset. Its candidate promoter region(BdorActinA3a-1^(P–2k)) was then cloned into the piggyBac plasmid. We subsequently established two stable transgenic lines with specific TTAA insertion sites on chromosomes 4 and 5, consistent with the characteristics of piggyBac transposition. The transgenic strains exhibited essentially normal survival, with hatchability and adult lifespan unaffected, althoughthere were slight reductions in the emergence rate and oviposition capacity. The fluorescent reporter has been successfully expressed in olfactory-related organs, such as the antennae, proboscis, maxillary palp, legs, external genitalia, and brain. The antennal electrophysiological responses to representative chemicals in the transgenic lines were consistent with those of the wild type. However, some olfactory-related behaviors, such as pheromone response and mating, were significantly affected in the transgenic lines. These findings suggest that our system could potentially be applied in future olfactory research, such as driving the expression of exogenous elements that are effective in olfactory organs. However, caution is advised regarding its impact when applied to some olfactoryrelated behavioral phenotypes. 展开更多
