The total internal reflection(TIR)behavior of interface shear waves is crucial for ensuring the reliability of dielectric elastomer(DE)devices.However,due to the complex force-electric coupling and large deformation o...The total internal reflection(TIR)behavior of interface shear waves is crucial for ensuring the reliability of dielectric elastomer(DE)devices.However,due to the complex force-electric coupling and large deformation of DEs,the TIR behavior of shear waves in heterogeneous force-electric interface models is still unclear.This study modeled an elastic/DE bi-material interface to analyze the trajectory of out-of-plane shear waves.Employing Dorfmann and Ogden’s nonlinear electroelastic framework and the related linear small incremental motion theory,a method has been developed to control the TIR behavior of interface shear waves.It has been found that the TIR behavior is significantly influenced by the strain-stiffening effect induced by biasing fields.Consequently,a biasing field principle involving preset electric displacement and pre-stretch has been proposed for TIR occurrence.By controlling the pre-stretch and preset electric displacement,active regulation of TIR behavior can be achieved.These results suggest a potential method for achieving autonomous energy shielding to improve the reliability of DE devices.展开更多

Although abundant research on the anisotropy of van der Waals(vd W)materials has been published,we undertake an in-depth study of their optical properties as they have an important guiding role for light control in tw...Although abundant research on the anisotropy of van der Waals(vd W)materials has been published,we undertake an in-depth study of their optical properties as they have an important guiding role for light control in two-dimensional(2D)nanospace.As an example,we study the reflectance of few-layered black phosphorus(BP)in the total internal reflection(TIR)mode in detail.We demonstrate that its optical anisotropy can be changed on a large scale by varying the incident angles,polarization states,and the in-plane rotation angles of the BP samples.Theoretical analysis indicates that the phenomena observed are common to all the atom-thick biaxial crystals,so these conclusions can be widely applied to other anisotropic 2D materials.This research furthers the current understanding of the properties of BP more comprehensively,and provides guidance for developing new optoelectronic applications,especially when BP and other atom-thick biaxial crystals are integrated with TIR devices.展开更多

In order to obtain this parameter, second order nonlinear spectroscopic techniques including second harmonic generation (SHG) and sum frequency generation-vibrational spec- troscopy (SFG-VS) have been successfully applied through analysis of the nonlinear signal from various polarizations. In some SHG and SFG-VS experiments, total internal reflection (TIR) configuration has been adopted to get enhanced signals. However, the reports on the detailed procedure of the polarization analysis and the calculation of the orientation angle of interracial molecules under TIR configuration are still very few. In this paper, we mea- sured the orientation angles of two molecules at the hexadecane-water interface under TIR and Non-TIR experimental configurations. The results measured from polarization analysis in TIR configuration consist with those obtained from Non-TIR configuration. This work demonstrates the feasibility and accuracy of polarization analysis in the determination of the orientation angle of molecules at the interfaces under TIR-SHG configuration.展开更多

The pH dependence of synchronous fluorescence signal at the interface was analyzed. Both unprotonated (TPPS4-) and diprotonated (H2TPPS2-) forms of TPPS were observed at the interface. But the interface favored the adsorption of. The apparent estimated pKa2 value shifted from 5.00 in the bulk solution to 2.7 at the interface. In this paper,we examine the tiny polarization rotation effect in total internal reflection due to the spin–orbit interaction of light.We find that the tiny polarization rotation rate will induce a geometric phase gr...In this paper,we examine the tiny polarization rotation effect in total internal reflection due to the spin–orbit interaction of light.We find that the tiny polarization rotation rate will induce a geometric phase gradient,which can be regarded as the physical origin of photonic spin Hall effect.We demonstrate that the spin-dependent splitting in position space is related to the polarization rotation in momentum space,while the spin-dependent splitting in momentum space is attributed to the polarization rotation in position space.Furthermore,we introduce a quantum weak measurement to determine the tiny polarization rotation rate.The rotation rate in momentum space is obtained with 118 nm,which manifests itself as a spatial shift,and the rotation rate in position space is achieved with 38 μrad∕λ,which manifests itself as an angular shift.The investigation of the polarization rotation characteristics will provide insights into the photonic spin Hall effect and will enable us to better understand the spin–orbit interaction of light.展开更多

Optical fibers, core components of global communication infrastructure, are capable of transmitting data over long distances with minimal loss through principles like total internal reflection. This study explores single-mode and multi-mode fiber designs, providing an overview of key parameters such as core diameter, refractive index profile, and numerical aperture. Mathematical modeling using Maxwell’s equations plays a central role in optimizing fiber performance, helping engineers mitigate challenges like attenuation and dispersion. The paper also discusses advanced techniques, including dense wavelength division multiplexing (DWDM), which enables terabit-per-second data rates. Case studies in practical applications, such as fiber-to-the-home (FTTH) networks and transoceanic cables, highlight the impact of optimized designs on network performance. The dynamics of GLUT4 storage vesicles (GSVs) remains to be explored and it is unclear how GSVs are arranged based on their mobility. We examined this issue in 3T3-L1 cells via investigating the three-dimensional mobility of single GSV labeled with EGFP-fused GLUT4. A thin layer of cytosol right adjacent to the plasma membrane was illuminated and successively imaged at 5 Hz under a total internal reflection fluorescence microscope with a penetration depth of 136 nm. Employing single particle tracking, the three-dimensional subpixel displacement of single GSV was tracked at a spatial precision of 22 nm. Both the mean square displacement and the diffusion coefficient were calculated for each vesicle. Tracking results revealed that vesicles moved as if restricted within a cage that has a mean radius of 160 nm, suggesting the presence of some intracellular tethering matrix. By constructing the histogram of the diffusion coefficients of GSVs, we observed a smooth distribution instead of the existence of distinct groups. The result indicates that GSVs are dynamically retained in a continuous and wide range of mobility rather than into separate classes.展开更多

Drawbacks include smearing artefacts and a lack of dynamic information ...The sciatic functional index(SFI) is a popular parameter for peripheral nerve evaluation that relies on footprints obtained with ink and paper. Drawbacks include smearing artefacts and a lack of dynamic information during measurement. Modern applications use digitized systems that can deliver results with less analytical effort and fewer mice. However, the systems are expensive(€40,000). This study aimed to evaluate the applicability and precision of a self-made, low-cost infrared system for evaluating SFI in mice. Mice were subjected to unilateral sciatic nerve crush injury(crush group; n = 7) and sham operation(sham group; n = 4). They were evaluated on the day before surgery, the 2nd, 4th and 6th days after injury, and then every day up to the 23rd day after injury. We compared two SFI evaluation methods, i.e., conventional inkand-paper SFI(C-SFI) and our infrared system(I-SFI). Our apparatus visualized footprints with totally internally reflected infrared light(950 nm) and a camera that can only detect this wavelength. Additionally we performed an analysis with the ladder beam walking test(LBWT) as a reference test. I-SFI assessment reduced the standard deviation by about 33 percent, from 11.6 to 7.8, and cut the variance around the baseline to 21 percent. The system thus requires fewer measurement repetitions and fewer animals, and cuts the cost of keeping the animals. The apparatus cost €321 to build. Our results show that the process of obtaining the SFI can be made more precise via digitization with a self-made, low-cost infrared system.展开更多

A laser sheet illuminates the contact interface, and the transmitted laser sheet is projected onto a screen. Then the contact information is acquired from the screen by a camera. An improved Otsu method is proposed to process the data of experimental images. It can compute the threshold of the overall image and filter out all the pixels one by one. Through analyzing the experimental results, we describe the relationship between the real contact area and the positive pressure during a continuous loading process, at different loading rates, with the polymethyl methacrylate(PMMA)material. A hysteresis phenomenon in the relationship between the real contact area and the positive pressure is found and explained.展开更多
TIRF microscopy has provided a means to view mobile granules within 100 nm in size in two dimensions.However quantitative analysis of the position and motion of those granules requires an appropriate tracking method.I...TIRF microscopy has provided a means to view mobile granules within 100 nm in size in two dimensions.However quantitative analysis of the position and motion of those granules requires an appropriate tracking method.In this paper,we present a new tracking algorithm combined with the unique features of TIRF.Firstly a fluorescence correction procedure was processed to solve the problem of fluorescence bleaching over time.Mobile granules were then segmented from a time-lapse image stack by an adaptive background subtraction method.Kalman filter was introduced to estimate and track the granules that allowed reducing searching range and hence greater reliability in tracking process.After the tracked granules were located in x-y plane,the z-position was indirectly inferred from the changes in their intensities.In the experiments the algorithm was applied in tracking GLUT4 vesicles in living adipose cells.The results indicate that the algorithm has achieved robust estimation and tracking of the vesicles in three dimensions.展开更多

Herein, a robust single particle counting-based assay has been developed for the high-sensitive determination of T4 PNK activity through only a simple one-step reaction. Taking benefit of the exceptional space-confined enzymatic property of T4 PNK towards DNA substrates on a single nanoparticle,the T4 PNK activity can be precisely determined by counting the fluorescence-positive nanoparticles in a digital manner with a total internal reflection fluorescent microscope(TIRFM). Due to the featured spatial-confined enzymatic property of T4 PNK and the single particle counting-based signal readout, T4PNK can be effectively differentiated from other interfering enzymes. This facile strategy has been also successfully applied to screen T4 PNK inhibitor and accurately determine T4 PNK activity in complex biological samples, paving a potential avenue for the digital analysis of biomarkers.展开更多

This is becaus...In a mixture of colloidal particles and polymer molecules, the particles may experience an attractive "depletion force" if the size of the polymer molecule is larger than the interparticle separation. This is because individual polymer molecules experience less conformational entropy if they stay between the particles than they escape the inter-particle space, which results in an osmotic pressure imbalance inside and outside the gap and leads to interparticle attraction. This depletion force has been the subject of several studies since the 1980s, but the direct measurement of this force is still experimentally challenging as it requires the detection of energy variations of the order of kBT and beyond. We present here our results for applying total internal reflection microscopy (TIRM) to directly measure the interaction between a free-moving particle and a flat surface in solutions consisting of small water-soluble organic molecules or polymeric surfactants. Our results indicate that stable nanobubbles (ca. 150 nm) exist free in the above aqueous solutions. More importantly, the existence of such nanobubbles induces an attraction between the spherical particle and flat surface. Using TIRM, we are able to directly measure such weak interaction with a range up to 100 nm. Furthermore, we demonstrate that by employing thermo^sensitive microgel particles as a depleting agent, we are able to quantitatively measure and reversibly control kBT-scale depletion attraction as function of solution pH.展开更多

The layer is a part of the laser dye solution, which is in optical contact with the bottom of optical cylindrical cell, the shape of which determines the geometric shape of the exciting layer. Homogeneous excitation of this layer by the second harmonic of the Nd+: YAG (λ = 532 nm) laser is realized. Circular, plane-directed radiation, with a small, vertical, divergence was obtained from the edges of the excited layer. Is investigated experimentally the spectral and spatial characteristics of radiation. Excitation of the layer was performed from the side of the cuvette bottom. It turned out that within the concentrations of the dye in the solution from 0.12 to 0.03 wt%, the following processes are observed: 1) Plane-directed radiation, with a small vertical divergence, uniform in intensity, around the optical cell in the plane of the luminescent layer;2) An increase in the amplitude of radiation pulses with a decrease in the concentration of the dye in the solution;3) Shifting of the maximum of the emission spectrum to the short-wave region, significant narrowing of the radiation spectrum and decrease of the vertical divergence of radiation.展开更多

文摘This paper examines the design and optimization of optical fibers for high-speed data transmission, emphasizing advancements that maximize efficiency in modern communication networks. Optical fibers, core components of global communication infrastructure, are capable of transmitting data over long distances with minimal loss through principles like total internal reflection. This study explores single-mode and multi-mode fiber designs, providing an overview of key parameters such as core diameter, refractive index profile, and numerical aperture. Mathematical modeling using Maxwell’s equations plays a central role in optimizing fiber performance, helping engineers mitigate challenges like attenuation and dispersion. The paper also discusses advanced techniques, including dense wavelength division multiplexing (DWDM), which enables terabit-per-second data rates. Case studies in practical applications, such as fiber-to-the-home (FTTH) networks and transoceanic cables, highlight the impact of optimized designs on network performance. Looking forward, innovations in photonic crystal fibers and hollow-core fibers are expected to drive further improvements, enabling ultra-high-speed data transmission. The paper concludes by underscoring the significance of continuous research and development to address challenges in optical fiber technology and support the increasing demands of global communication systems.
文摘Glucose transporter 4 (GLUT4) is responsible for insulin-stimulated glucose transporting into the insulin-sensitive fat and muscle cells. The dynamics of GLUT4 storage vesicles (GSVs) remains to be explored and it is unclear how GSVs are arranged based on their mobility. We examined this issue in 3T3-L1 cells via investigating the three-dimensional mobility of single GSV labeled with EGFP-fused GLUT4. A thin layer of cytosol right adjacent to the plasma membrane was illuminated and successively imaged at 5 Hz under a total internal reflection fluorescence microscope with a penetration depth of 136 nm. Employing single particle tracking, the three-dimensional subpixel displacement of single GSV was tracked at a spatial precision of 22 nm. Both the mean square displacement and the diffusion coefficient were calculated for each vesicle. Tracking results revealed that vesicles moved as if restricted within a cage that has a mean radius of 160 nm, suggesting the presence of some intracellular tethering matrix. By constructing the histogram of the diffusion coefficients of GSVs, we observed a smooth distribution instead of the existence of distinct groups. The result indicates that GSVs are dynamically retained in a continuous and wide range of mobility rather than into separate classes.
文摘The sciatic functional index(SFI) is a popular parameter for peripheral nerve evaluation that relies on footprints obtained with ink and paper. Drawbacks include smearing artefacts and a lack of dynamic information during measurement. Modern applications use digitized systems that can deliver results with less analytical effort and fewer mice. However, the systems are expensive(€40,000). This study aimed to evaluate the applicability and precision of a self-made, low-cost infrared system for evaluating SFI in mice. Mice were subjected to unilateral sciatic nerve crush injury(crush group; n = 7) and sham operation(sham group; n = 4). They were evaluated on the day before surgery, the 2nd, 4th and 6th days after injury, and then every day up to the 23rd day after injury. We compared two SFI evaluation methods, i.e., conventional inkand-paper SFI(C-SFI) and our infrared system(I-SFI). Our apparatus visualized footprints with totally internally reflected infrared light(950 nm) and a camera that can only detect this wavelength. Additionally we performed an analysis with the ladder beam walking test(LBWT) as a reference test. I-SFI assessment reduced the standard deviation by about 33 percent, from 11.6 to 7.8, and cut the variance around the baseline to 21 percent. The system thus requires fewer measurement repetitions and fewer animals, and cuts the cost of keeping the animals. The apparatus cost €321 to build. Our results show that the process of obtaining the SFI can be made more precise via digitization with a self-made, low-cost infrared system.

文摘An experimental method of measuring the real contact area of transparent blocks based on the principle of total internal reflection is presented, intending to support the investigation of friction characteristics, heat conduction, and energy dissipation at the contact interface. A laser sheet illuminates the contact interface, and the transmitted laser sheet is projected onto a screen. Then the contact information is acquired from the screen by a camera. An improved Otsu method is proposed to process the data of experimental images. It can compute the threshold of the overall image and filter out all the pixels one by one. Through analyzing the experimental results, we describe the relationship between the real contact area and the positive pressure during a continuous loading process, at different loading rates, with the polymethyl methacrylate(PMMA)material. A hysteresis phenomenon in the relationship between the real contact area and the positive pressure is found and explained.

文摘TIRF microscopy has provided a means to view mobile granules within 100 nm in size in two dimensions.However quantitative analysis of the position and motion of those granules requires an appropriate tracking method.In this paper,we present a new tracking algorithm combined with the unique features of TIRF.Firstly a fluorescence correction procedure was processed to solve the problem of fluorescence bleaching over time.Mobile granules were then segmented from a time-lapse image stack by an adaptive background subtraction method.Kalman filter was introduced to estimate and track the granules that allowed reducing searching range and hence greater reliability in tracking process.After the tracked granules were located in x-y plane,the z-position was indirectly inferred from the changes in their intensities.In the experiments the algorithm was applied in tracking GLUT4 vesicles in living adipose cells.The results indicate that the algorithm has achieved robust estimation and tracking of the vesicles in three dimensions.

文摘T4 polynucleotide kinase(T4 PNK) is a pivotal enzyme for DNA replication, recombination, and DNA damage repair. Herein, a robust single particle counting-based assay has been developed for the high-sensitive determination of T4 PNK activity through only a simple one-step reaction. Taking benefit of the exceptional space-confined enzymatic property of T4 PNK towards DNA substrates on a single nanoparticle,the T4 PNK activity can be precisely determined by counting the fluorescence-positive nanoparticles in a digital manner with a total internal reflection fluorescent microscope(TIRFM). Due to the featured spatial-confined enzymatic property of T4 PNK and the single particle counting-based signal readout, T4PNK can be effectively differentiated from other interfering enzymes. This facile strategy has been also successfully applied to screen T4 PNK inhibitor and accurately determine T4 PNK activity in complex biological samples, paving a potential avenue for the digital analysis of biomarkers.

文摘In a mixture of colloidal particles and polymer molecules, the particles may experience an attractive "depletion force" if the size of the polymer molecule is larger than the interparticle separation. This is because individual polymer molecules experience less conformational entropy if they stay between the particles than they escape the inter-particle space, which results in an osmotic pressure imbalance inside and outside the gap and leads to interparticle attraction. This depletion force has been the subject of several studies since the 1980s, but the direct measurement of this force is still experimentally challenging as it requires the detection of energy variations of the order of kBT and beyond. We present here our results for applying total internal reflection microscopy (TIRM) to directly measure the interaction between a free-moving particle and a flat surface in solutions consisting of small water-soluble organic molecules or polymeric surfactants. Our results indicate that stable nanobubbles (ca. 150 nm) exist free in the above aqueous solutions. More importantly, the existence of such nanobubbles induces an attraction between the spherical particle and flat surface. Using TIRM, we are able to directly measure such weak interaction with a range up to 100 nm. Furthermore, we demonstrate that by employing thermo^sensitive microgel particles as a depleting agent, we are able to quantitatively measure and reversibly control kBT-scale depletion attraction as function of solution pH.

文摘Optical microscopy promises researchers to soe most tiny substances directly.However,the resolution of conventional microscopy is resticted by the diffraction limit.This makes it a challenge to observe subcellular processes happened in nanoscale.The development of super-resolution microscopy provides a solution to this challenge.Here,we briefly review several commonly used super-resolution techniques,explicating their basic principles and applications in biological science,especially in neuroscience.In addition,characteristics and limitations of each techrique are compared to provide a guidance for biologists to choose the most suitable tool.
文摘The spectral-spatial characteristics of the lateral radiation of a circular-shaped Rhodamine 6G solution layer were investigated. The layer is a part of the laser dye solution, which is in optical contact with the bottom of optical cylindrical cell, the shape of which determines the geometric shape of the exciting layer. Homogeneous excitation of this layer by the second harmonic of the Nd+: YAG (λ = 532 nm) laser is realized. Circular, plane-directed radiation, with a small, vertical, divergence was obtained from the edges of the excited layer. Is investigated experimentally the spectral and spatial characteristics of radiation. Excitation of the layer was performed from the side of the cuvette bottom. It turned out that within the concentrations of the dye in the solution from 0.12 to 0.03 wt%, the following processes are observed: 1) Plane-directed radiation, with a small vertical divergence, uniform in intensity, around the optical cell in the plane of the luminescent layer;2) An increase in the amplitude of radiation pulses with a decrease in the concentration of the dye in the solution;3) Shifting of the maximum of the emission spectrum to the short-wave region, significant narrowing of the radiation spectrum and decrease of the vertical divergence of radiation.
文摘Polymer-coated particles and surfaces have widespread applications in various industries ranging from manufacturing to biomedicine.A better understanding of the stability mechanisms underlying these coatings can inspire the design of novel polymer structures and help tune their functions.This can be achieved by quantifying the particle-particle and particle-surface interactions.This paper reviews several common experimental techniques utilized to measure the interactions between polymer-coated particles and surfaces quantitatively.These techniques include atomic force microscopy(AFM),total internal reflection microscopy(TIRM),optical tweezers(OT),and quartz crystal microbalance with dissipation monitoring(QCM-D).The examples of each measuring technique were categorized based on the types of polymer coatings and their associated factors.Additionally,this review demonstrates experimental measurements of interactions involving biological objects.By understanding the fundamental particle-particle and particle-surface interactions,researchers can gain valuable insights to guide the design and functional optimization of polymer-based materials and systems across various applications.
