Photoacoustic microscopy depth-of-field extension method and system based on three-dimensional continuity and sparsity deconvolution

Optical-resolution photoacoustic microscopy is a novel imaging technique that combines the advantages of optical and ultrasound imaging,enabling high-resolution visualization of biological tissues at the micrometer scale.However,the divergence of the excited Gaussian beam limits the depth-of-field of the system to less than 100μm,which hinders accurate three-dimensional imaging of living tissues and restrictsits applicability in biological research.Therefore,there is an urgent need for an effective method to enhance the depth-of-field without altering the hardware configuration.This paper presents a photoacoustic microscopy depth-of-field extension method and system based on three-dimensional continuity and sparsity deconvolution.This method utilizes a depth-varying point spread function and incorporates continuity and sparsity con-straints into the deconvolution process to mitigate the effect of background noise,enhancing the stability and accuracy of the depth-of-field extension.Experimental results using tungsten wire phantoms suggest that the depth-of-field of system can be extended to 650 pm,which is 7.2 times greater than conventional system,while improving the resolution of the defocused region by an average factor of 3.5.Furthermore,experiments on zebrafish and nude mouse ears with irregular topologies demonstrate that the proposed method successfully overcomes image blurring and the loss of structural information due to limited depth-of-field.All the results suggest that the system with higher lateral resolution and enhanced depth-of-field has significant potential for a wide range of practical biomedical applications. 展开更多

The environment of low-altitude urban airspace is complex and variable due to numerous obstacles,non-cooperative aircraft,and birds.Unmanned Aerial Vehicles(UAVs)leveraging environmental information to achieve three-dimension collision-free trajectory planning is the prerequisite to ensure airspace security.However,the timely information of surrounding situation is difficult to acquire by UAVs,which further brings security risks.As a mature technology leveraged in traditional civil aviation,the Automatic Dependent Surveillance-Broadcast(ADS-B)realizes continuous surveillance of the information of aircraft.Consequently,we leverage ADS-B for surveillance and information broadcasting,and divide the aerial airspace into multiple sub-airspaces to improve flight safety in UAV trajectory planning.In detail,we propose the secure Sub-airSpaces Planning(SSP)algorithm and Particle Swarm Optimization Rapidly-exploring Random Trees(PSO-RRT)algorithm for the UAV trajectory planning in law-altitude airspace.The performance of the proposed algorithm is verified by simulations and the results show that SSP reduces both the maximum number of UAVs in the sub-airspace and the length of the trajectory,and PSO-RRT reduces the cost of UAV trajectory in the sub-airspace. 展开更多

In recent years,notable progress has been achieved in both the hardware and algorithms of structured illumination microscopy(SIM).Nevertheless,the advancement of three-dimensional structured illumination microscopy(3DSIM)has been impeded by challenges arising from the speed and intricacy of polarization modulation.We introduce a high-speed modulation 3DSIM system,leveraging the polarizationmaintaining and modulation capabilities of a digital micromirror device(DMD)in conjunction with an electrooptic modulator.The DMD-3DSIM system yields a twofold enhancement in both lateral(133 nm)and axial(300 nm)resolution compared to wide-field imaging and can acquire a data set comprising 29 sections of 1024 pixels×1024 pixels,with 15 ms exposure time and 6.75 s per volume.The versatility of the DMD-3DSIM approach was exemplified through the imaging of various specimens,including fluorescent beads,nuclear pores,microtubules,actin filaments,and mitochondria within cells,as well as plant and animal tissues.Notably,polarized 3DSIM elucidated the orientation of actin filaments.Furthermore,the implementation of diverse deconvolution algorithms further enhances 3D resolution.The DMD-based 3DSIM system presents a rapid and reliable methodology for investigating biomedical phenomena,boasting capabilities encompassing 3D superresolution,fast temporal resolution,and polarization imaging. 展开更多

Melanosomes, isolated by centrifugal separation from culture broth of B16 melanoma cells derived from mouse, were observed by scanning electron microscopy (SEM), and by transmission electron microscopy (TEM). Some interesting structural features were found inside and outside of the melanosomes. By SEM observation, the melanosomes were ellipsoid shape, their surface was not smooth and was covered with rough substructure, 10 to 20 nm particles. By TEM, uneven structure and micro particles were observed in the melanosomes. Furthermore, three-dimensional analysis was tried by using the ultra-high voltage electron microscopy(UHVEM). Micrographs of the melanosomes were taken at various tilted angles by UHVEM, after preparing 500 nm thickness specimens stained with lead citrate. From the micrographs collected, the three-dimensional structures were reconstructed by using i-mode software. Melanin stained by lead and non stained parts was clearly observed in the reconstructed structure. Non stained parts were round, regular size, and distributed widely in the melanosomes. 展开更多

This work presents a method for the three-dimensional localization of individual shallow NV center in diamond,leveraging the near-field quenching effect of a gold tip.Our experimental setup involves the use of an atomic force microscope to precisely move the gold tip close to the NV center,while simultaneously employing a home-made confocal microscope to monitor the fluorescence of the NV center.This approach allows for lateral super-resolution,achieving a full width at half maximum(FWHM)of 38.0 nm and a location uncertainty of 0.7 nm.Additionally,we show the potential of this method for determining the depth of the NV centers.We also attempt to determine the depth of the NV centers in combination with finite-difference time-domain(FDTD)simulations.Compared to other depth determination methods,this approach allows for simultaneous lateral and longitudinal localization of individual NV centers,and holds promise for facilitating manipulation of the local environment surrounding the NV center. 展开更多

We present a threedimensional(3D)isotropic imaging of mouse brain using light-sheet fuo-rescent microscopy(LSFM)in conjumction with a multi-view imaging computation.Unlike common single view LSFM is used for mouse brain imaging,the brain tissue is 3D imaged under eight views in our study,by a home-built selective plane ilumination microscopy(SPIM).An output image containing complete structural infornation as well as significantly improved res olution(~4 times)are then computed based on these eight views of data,using a bead-guided multi-view registration and deconvolution.With superior imaging quality,the astrocyte and pyrarmidal neurons together with their subcellular nerve fbers can be clearly visualized and segmented.With further incuding other computational methods,this study can be potentially scaled up to map the conectome of whole mouse brain with a simple light.sheet microscope. 展开更多

BACKGROUND Sigmoid colon cancer faces challenges due to anatomical diversity,including variable inferior mesenteric artery(IMA)branching and tumor localization complexities,which increase intraoperative risks.AIM To comprehensively evaluate the impact of three-dimensional(3D)visualization technology on enhancing surgical precision and safety,as well as optimizing perioperative outcomes in laparoscopic sigmoid cancer resection.METHODS A prospective cohort of 106 patients(January 2023 to December 2024)undergoing laparoscopic sigmoid cancer resection was divided into the 3D(n=55)group and the control(n=51)group.The 3D group underwent preoperative enhanced computed tomography reconstruction(3D Slicer 5.2.2&Mimics 19.0).3D reconstruction visualization navigation intraoperatively guided the following key steps:Tumor location,Toldt’s space dissection,IMA ligation level selection,regional lymph node dissection,and marginal artery preservation.Outcomes included operative parameters,lymph node yield,and recovery metrics.RESULTS The 3D group demonstrated a significantly shorter operative time(172.91±20.69 minutes vs 190.29±32.29 minutes;P=0.002),reduced blood loss(31.5±11.8 mL vs 44.1±23.4 mL,P=0.001),earlier postoperative flatus(2.23±0.54 days vs 2.53±0.61 days;P=0.013),shorter hospital length of stay(13.47±1.74 days vs 16.20±7.71 days;P=0.013),shorter postoperative length of stay(8.6±2.6 days vs 10.5±4.9 days;P=0.014),and earlier postoperative exhaust time(2.23±0.54 days vs 2.53±0.61 days;P=0.013).Furthermore,the 3D group exhibited a higher mean number of lymph nodes harvested(16.91±5.74 vs 14.45±5.66;P=0.030).CONCLUSION The 3D visualization technology effectively addresses sigmoid colon anatomical complexity through surgical navigation,improving procedural safety and efficiency. 展开更多

An in-situ plasma spark sintering(SPS)apparatus,coupled with laboratory X-ray microscopy,was uti-lized to three-dimensionally investigate the dynamic evolution process of 7055 aluminum alloy during SPS process.The influences of sintering temperatures and particle morphology on the sintering kinetics were discussed in detail.It was observed that elevating the sintering temperatures enhanced both the rate of densification and the final compactness of the alloy.Furthermore,three-dimensional quantitative analysis of pore evolution indicated that greater discrepancies in powder size between neighboring par-ticles facilitated pore elimination during sintering by increasing available interstitial spaces.Mechanistic analysis rationalized these observations by attributing the enhanced sintering kinetics to the greater par-ticle size disparity,which resulted in higher necking curvature and accelerated densification.The present study therefore provides a comprehensive three-dimensional in-situ quantitative analysis on the dynamic SPS process,and is expected to advance the current comprehension of sintering mechanisms at the mi-cron scale. 展开更多

TIRF microscopy has provided a means to view mobile granules within 100 nm in size in two dimensions.However quantitative analysis of the position and motion of those granules requires an appropriate tracking method.In this paper,we present a new tracking algorithm combined with the unique features of TIRF.Firstly a fluorescence correction procedure was processed to solve the problem of fluorescence bleaching over time.Mobile granules were then segmented from a time-lapse image stack by an adaptive background subtraction method.Kalman filter was introduced to estimate and track the granules that allowed reducing searching range and hence greater reliability in tracking process.After the tracked granules were located in x-y plane,the z-position was indirectly inferred from the changes in their intensities.In the experiments the algorithm was applied in tracking GLUT4 vesicles in living adipose cells.The results indicate that the algorithm has achieved robust estimation and tracking of the vesicles in three dimensions. 展开更多

Microscopy imaging is fundamental in analyzing bacterial morphology and dynamics,offering critical insights into bacterial physiology and pathogenicity.Image segmentation techniques enable quantitative analysis of bacterial structures,facilitating precise measurement of morphological variations and population behaviors at single-cell resolution.This paper reviews advancements in bacterial image segmentation,emphasizing the shift from traditional thresholding and watershed methods to deep learning-driven approaches.Convolutional neural networks(CNNs),U-Net architectures,and three-dimensional(3D)frameworks excel at segmenting dense biofilms and resolving antibiotic-induced morphological changes.These methods combine automated feature extraction with physics-informed postprocessing.Despite progress,challenges persist in computational efficiency,cross-species generalizability,and integration with multimodal experimental workflows.Future progress will depend on improving model robustness across species and imaging modalities,integrating multimodal data for phenotype-function mapping,and developing standard pipelines that link computational tools with clinical diagnostics.These innovations will expand microbial phenotyping beyond structural analysis,enabling deeper insights into bacterial physiology and ecological interactions. 展开更多

Desulfurization of CaO–Al_(2)O_(3) particles in molten steel was observed in situ using high-temperature confocal scanning laser microscopy.The effects of the aluminum and silicon contents of molten steel on desulfurization were analyzed.When the total aluminum content in the steel increased from 6 to 1100 ppm,the CaS content in CaO–Al_(2)O_(3) particles increased from 2.1wt%to 84.84wt%after the reaction for 90 s.Furthermore,when the silicon content in the steel increased from 0.01wt%to 2.20wt%,the CaS content in CaO–Al_(2)O_(3) particles increased from 1.53wt%to 79.01wt%after the reaction for 90 s.This indicates that the increase in the aluminum and silicon contents of the steel promoted the desulfurization of CaO–Al_(2)O_(3) particles.A kinetic model was established to predict the CaO–Al_(2)O_(3) particles composition,and the diffusion coefficient of sulfur in CaO–Al_(2)O_(3) particles was 9.375×10^(−10)m^(2)·s^(−1) at 1600℃,which provided a new method for the calculation of diffusion coefficient. 展开更多

Objective Aim to create a three-dimension(3D) reconstruction of acupuncture needling at Sidu(四渎 TE 9).Methods First,the skin,subcutaneous tissue,muscles,the blood vessels and nerves near TE 9 were defined in anatomical terms.Second,the above anatomical structures including the dissected muscles associated with at TE 9,the underlying blood vessels and nerves were reconstructed in 3D.Third,the process of acupuncturing TE 9 on a virtual human body was simulated using the VOXEL-MAN technology.Results Both the local anatomy of the point and the process of acupuncture needling at TE 9 were reconstructed.Different layers of tissues were visualized during the process of needling,as the needle was inserted and manipulated at the point.Conclusions Virtual 3D reconstruction can contribute to research on the anatomical structure of acupoints and the visualization of the acupoints.We recommend that it be fully utilized in acupuncture research. 展开更多

By Using (scanning) transmission electron microscopy, localized-corrosion morphology variations of the AA7055 AIZn(Cu)Mg alloy with different thermal processes and their underlying microscopic causes were investigated systematically. Our study shows that the corrosion resistance of the nanoscale precipitates varies with their structure type and Cu-content. Just like the Al-matrix, the early-stage precipitates are corrosion resistant, as compared with the ηp/η-precipitates without high Cu-content. With a high Cu-content, however, the η-precipitates become most corrosion resistant among all phases involved. The turbulence mechanism plays an important part in the mixing process and momentum transfer of turbulence. A three-dimensional Prandtl mixing length tidal model has been developed to simulate tidal flows and water quality. The eddy viscosities and diffusivities are computed from the Prandtl mixing length model. In order to model the water quality of an estuary or coastal area many interdependent processes need to be simulated. These may be conveniently separated into three main groups: transport and mixing processes, biochemical interaction of water quality variables and the utilization and re-cycling of nutrients by living matter. The model simulates full oxygen and nutrient balance, primary productivity and the transport, reaction mechanism and fate of pollutants over tidal time-scales. The model is applied to numerical simulation of tidal flows and water quality in Dalian Bay. The model has been calibrated against a limited data set of historical water quality observations and in general demonstrates excellent agreement with all available data. 展开更多

The traditional two-dimensional model exhibits limitations when one attempts to model the de- tailed two-phase flow inside the columns. The present paper presents a three-dimensional computational fluid dy- namics (CFD) model to simulate the two-phase flow in a representative unit of the column. The unit consists of an CFD calculations on column packed with Flexipak 1Y were implemented within the volume of fluid (VOF) mathe- matical framework. The CFD model was validated by comparing the calculated thickness of liquid film with the available experimental data. Special attention was given to quantitative analysis of the effects of gravity on the hy- drodynamics. Fluctuations in the liquid mass flow rate and the calculated pressure drop loss were found to be quali- tatively in agreement with the experimental observations. 展开更多

Knowledge of the microscopic structure, including three-dimensional (3-D) size and orientation of the precipitates, is essential to fully understand the mechanical properties of the magnesium alloys and designing the alloys with better performance. Analytical TEM with high spatial resolution offers the simultaneous measurements of 3-D size, structure, orientation, composition of the precipitates from one typical sample along an established crystallographic axis. Besides popular Burgers orientation relationship (OR), other ORs such as Pitsch-Schrader OR, Crawley OR, Potter OR and a new OR with the form of [0001]α 1.0° from [311]γ and (1120)α 2.0° from (033)γ between the magnesium matrix and the precipitate γ-MglTAl 12 are identified by TEM imaging and diffraction techniques. As a case study, the thicknesses of the individual precipitates with Burgers OR are further measured to be 100 200 nm through both electron energy-loss spectroscopy and x-ray energy dispersive spectroscopy combining differential x-ray absorption and extrapolation, which are in agreement with the overall 3-D size statistic distribution results obtained through analysing various samples along various directions. Furthermore, the fabricated wedge-shape structure provides a platform on which to study the dependence of the interfacial strain on the variation of the thickness. 展开更多

An appropriate Monte Carlo method was developed to simulate the three-dimensional normal grain growth more completely. Comparative investigation on the three-dimensional and the cross-sectional characteristics of normal grain growth was done. It was found that the time exponent of grain growth determined from cross-section exhibits the same rule of increasing slowly with time and approaching the theoretical value n = 0.5 of steadygrain growth as the three-dimensional (3-D) system. From change of the number of grains per unit area with timemeasured in cross-section, the state of 3-D normal grain growth may be predicted. The gtain size distribution incross-section is different from that in 3-D system and can not express the evolution characteristic of the 3-D distribution. Furthermore, there exists statistical connection between the topological parameters in cross-section and thosein three-dimensions. 展开更多

The nearly analytic discrete(NAD)method is a kind of finite difference method with advantages of high accuracy and stability.Previous studies have investigated the NAD method for simulating wave propagation in the time-domain.This study applies the NAD method to solving three-dimensional(3D)acoustic wave equations in the frequency-domain.This forward modeling approach is then used as the“engine”for implementing 3D frequency-domain full waveform inversion(FWI).In the numerical modeling experiments,synthetic examples are first given to show the superiority of the NAD method in forward modeling compared with traditional finite difference methods.Synthetic 3D frequency-domain FWI experiments are then carried out to examine the effectiveness of the proposed methods.The inversion results show that the NAD method is more suitable than traditional methods,in terms of computational cost and stability,for 3D frequency-domain FWI,and represents an effective approach for inversion of subsurface model structures. 展开更多
