Structure identification for compound I separated and purified from taxoids-produced endophytic fungi (Alternaria. alternata var. taxi 1011)

Many endophytic fungi can produce some physio-logical active compounds, which are same to or analog to those isolated from their hosts. Producing physiological active com-pounds through microbial fermentation can give a new way to resolve resource limitation and to find out alternative source. Through the methods of organic solvent extraction, thin layer chromatography (TLC) and column chromatography, compound I was isolated, purified from the liquid fermentation metabolites of the taxoids-produced endophytic fungi (Alternaria. alternata var. taxi 1011 Y. Xiang et LU An-guo) that was screened from the bark of Taxus. cuspidata Sieb.et Zucc.. Compound I was identified as one kind of taxoids type III, based on the analyzing results by using the methods of ultraviolet spectroscopy (UV), infrared spectroscopy (IR), mass spectrometry (MS) and nuclear magnetic resonance spectroscopy (NMR). Some natural products, such as traditional Chinese medicines(TCMs), contain compounds with anticancer activity and have attracted a great interest in recent years as alternative anticancer therapies. A quick and convenient assay for screening antimicrotubule compounds in which in vitro microdialysis/high-performance liquid chromatography (HPLC) is used to monitor the binding of the compounds extracted from TCM Taxus cuspidata Siebold & Zucc(Taxus) to microtubules is reported. It was observed that the extract of Taxus contains at least five compounds which have affinity interaction with microtubules by biological fingerprinting analysis, and they were identified as the taxoids of taxol, baccatin III, 10-deacetylbaccatin Ⅲ(10-DAB), cephalomannine and 7-epi-10-deacetyltaxol (7-epi-10-DAT) based on the comparison of their high-performance liquid chromatographic/mass spectrometric and UV spectra with those of the standard samples, both assembly-promoting and disassembly-inhibiting characteristics of those compounds were evaluated. It was observed that baccatin Ⅲ and 10-DAB bound to microtubules and the binding degrees were influenced by GTP. Competitive binding behavior of taxol with other four taxoids to microtubules was also investigated. 展开更多

We have studied the fragmentation behavior of positive and negative ions of taxoi and 6/8/6 type taxoids, the influence of different substituents on fragmentation, and the correlativity between fragmentation patterns and structure by MS/MS technique with different ionization methods such as FAB-MS, ESI-MS, ete. We have also investigated in detail the fragmentation of various molecular-related ions, such as [M+H]+, [M+Na]+ and [M-H] ions, and the formation pathways of characteristic fragment ions. It has been found that there exists some competing reaction between the loss of C-13 side chain and decomposition by loss of acetic acid. In addition, by comparing CID spectra obtained with low- and high-energy collision, it is seen that CID-MS/MS with low-energy collision is more suitable for the study of the structural analysis of small molecules and drug metabolites. The experimental results demonstrate that MS/MS spectra can reflect more effectively the slight difference of structure between the related 展开更多

An oxetane ring can be constructed from 5α-acyloxy-Δ4(20)-taxoids. Hie facile intramolecular acyl migration from 5- to 20-position under slightly basic conditions enabled the construction of the oxetane ring in a convenient short cut, whereas the acyl migration from 2- to 20-position left the 2-hydroxyl accessible to a later benzoylation. An unexpected five-mem-bered 4-O, 20- O sulfite ring was formed in the attempted construction of the oxetane ring with 5α-triflate as a leaving group. After the construction of the oxetane ring, treatment with strong base LiHMDS and acetyl chloride gave the expected 4-O-acetate while treatment with acetic anhydride and DMAP gave a 4-O-acetoacetate. 展开更多

红豆杉中紫杉醇及其衍生物的含量显著地受到多种因素的影响,对这些影响因子的研究有利于红豆杉产业化栽培基地经济效益的实现。根据以往的研究,综述了影响红豆杉中紫杉醇及其衍生物含量的主要因子,包括品种、环境、季节、年龄、性别、... 红豆杉中紫杉醇及其衍生物的含量显著地受到多种因素的影响,对这些影响因子的研究有利于红豆杉产业化栽培基地经济效益的实现。根据以往的研究,综述了影响红豆杉中紫杉醇及其衍生物含量的主要因子,包括品种、环境、季节、年龄、性别、组织部位、储藏方式、激素和代谢旁路抑制剂等不同的方面。曼地亚红豆杉是美国食品与药物管理局筛选到的最适宜栽培的品种,目前市场上50%以上的10-脱酰基巴卡丁Ⅲ来自于欧洲紫杉。阴湿环境生长的红豆杉紫杉醇含量较高,冬季一般是紫杉醇及其衍生物总含量最高的季节,年龄的增加有利于紫杉醇含量的提高,性别不同的红豆杉紫杉醇的含量没有显著差异,根和皮部的紫杉醇含量较高,阴干和低温保藏能有效地减缓紫杉醇的分解速度,一些激素和代谢旁路抑制剂的施用可以明显地提高红豆杉中紫杉醇及其衍生物的含量。并进一步提出了未来急需深入研究的重点:(1)优良品系的筛选和栽培,尤其是对生长速度最快和在中国分布最广的南方红豆杉;(2)土壤中各种成分,尤其是稀土元素对紫杉醇及其衍生物含量的影响和机理;(3)定点与大尺度实验相结合研究宏观生态因子对含量的影响和机理;(4)激素和代谢旁路抑制剂对含量的影响和机理;(5)调节宏观生态因子、土壤成分、激素和代谢旁路抑制剂的配伍进一步提高产量并应用于生产实践。 展开更多

多西他赛和顺铂同步放化疗治疗局部晚期鼻咽癌临床观察 被引量:10

目的观察局部晚期鼻咽癌同步放化疗的疗效及毒副反应。方法 70例局部晚期鼻咽癌,采用静脉化疗2周期,化疗方案:多西他赛75 mg/m2,分于第1,8天给药;顺铂(DDP)25 mg/(m2.d)d1-3天,21天为1周期。同时给予同步放疗,鼻咽部原发灶给予6 MV-X线... 目的观察局部晚期鼻咽癌同步放化疗的疗效及毒副反应。方法 70例局部晚期鼻咽癌,采用静脉化疗2周期,化疗方案:多西他赛75 mg/m2,分于第1,8天给药;顺铂(DDP)25 mg/(m2.d)d1-3天,21天为1周期。同时给予同步放疗,鼻咽部原发灶给予6 MV-X线和15 MV-X线DT 70 Gy^78 Gy/7-8周。结果 70例中CR 20例,PR 37例,总有效率81.4%。1年、3年生存率分别为98.4%、83.9%。主要毒副反应为骨髓抑制、急性口腔黏膜炎、恶心呕吐、皮肤反应。结论多西他赛和顺铂同步放化疗治疗局部晚期鼻咽癌疗效较好,毒副反应可以耐受。 展开更多

目的探讨白蛋白结合型紫杉醇联合卡铂和贝伐单抗作为一线方案治疗非小细胞肺癌(NSCLC)患者的疗效和安全性。方法选取NSCLC患者共120例。采用随机数字表法分为对照组(n=60),研究组(n=60),分别行紫杉醇+顺铂;白蛋白结合型紫杉醇+卡铂+贝... 目的探讨白蛋白结合型紫杉醇联合卡铂和贝伐单抗作为一线方案治疗非小细胞肺癌(NSCLC)患者的疗效和安全性。方法选取NSCLC患者共120例。采用随机数字表法分为对照组(n=60),研究组(n=60),分别行紫杉醇+顺铂;白蛋白结合型紫杉醇+卡铂+贝伐单抗治疗。主要终结点为安全性和患者耐受性,次要终结点为无进展生存期(PFS)、总反应率(ORR)和临床获益率(CBR)。结果对照组PFS为5.7个月,CBR为76%,ORR为67%,相比较于对照组,研究组PFS为8.9个月,CBR为92%,ORR为87%。对照组中不良事件为Ⅲ度骨髓抑制(53.4%),Ⅲ度消化道反应(40.1%),而研究组中不良反应为Ⅲ度骨髓抑制(46.9%)和Ⅱ度消化道反应(20.1%)。结论对比于传统铂类化疗方案,白蛋白结合型紫杉醇联合卡铂和贝伐单抗作为NSCLC患者一线治疗安全有效。 展开更多
