小麦转录因子TaMYB5-like转录自激活能力及其参与生理型花粉败育过程表达模式研究

化学杂交剂SQ-1诱导的小麦花粉败育是一个复杂的生理代谢过程,TaMYB5-like被发现参与此过程。为了解TaMYB5-like基因在SQ-1诱导小麦生理型花粉败育过程中的作用,以SQ-1诱导的小麦生理型雄性不育系PHYMS-1376和对照可育系CK-1376四分体... 化学杂交剂SQ-1诱导的小麦花粉败育是一个复杂的生理代谢过程,TaMYB5-like被发现参与此过程。为了解TaMYB5-like基因在SQ-1诱导小麦生理型花粉败育过程中的作用,以SQ-1诱导的小麦生理型雄性不育系PHYMS-1376和对照可育系CK-1376四分体、单核早期、单核晚期、二核期和三核期花药为试验材料,克隆TaMYB5-like基因,并对其进行序列结构、生物信息学、表达模式、亚细胞定位及转录自激活效应分析。结果发现,TaMYB5-like基因序列长5 207 bp,其cDNA长为1 133 bp,其中CDS序列长819 bp,编码272个氨基酸,含有2个SANT结构域,分子量为29.36 kDa,无信号肽,无跨膜结构,亚细胞定位在细胞核内;TaMYB5-like基因启动子涉及光响应、逆境胁迫响应、茉莉酸甲酯响应等顺式作用元件。自激活效应发现,TaMYB5-like蛋白的自激活区段位于N端,合适浓度的AbA和3-AT可以抑制BD-TaMYB5-like和BD-TaMYB5-like-N-SANT融合蛋白的自激活性;随着蛋白质氨基酸序列的截断,自激活性呈降低趋势。与CK-1376相比,PHYMS-1376四分体和单核早期花药中TaMYB5-like表达量差异不显著;花药发育到单核晚期、二核期和三核期时,TaMYB5-like表达量显著增加;CK-1376和PHYMS-1376三核期时花药内TaMYB5-like表达量均最高。 展开更多

G1-like与F/98-like进化谱系的P B2、M基因在H5N6亚型禽流感病毒重配中的竞争优势研究

为评价不同H9N2亚型病毒供体在H5N6亚型禽流感病毒(AIV)重配中的作用,本研究选取全套内部基因与S基因型H9N2 AIV高度同源的3株H5N6 AIV流行株MZ34、YB0597和JT131,利用反向遗传学技术在MZ34的8基因重组质粒基础上另外添加F/98-like来源... 为评价不同H9N2亚型病毒供体在H5N6亚型禽流感病毒(AIV)重配中的作用,本研究选取全套内部基因与S基因型H9N2 AIV高度同源的3株H5N6 AIV流行株MZ34、YB0597和JT131,利用反向遗传学技术在MZ34的8基因重组质粒基础上另外添加F/98-like来源的PB2和M质粒,即以MZ34(8)+F/98(PB2+M)的10质粒组合(Group 1)共转染细胞进行重组H5N6病毒的竞争性拯救;PCR扩增经3轮空斑纯化后PCR扩增拯救病毒的PB2和M基因并测序鉴定。同时,为排除来源于同一母本病毒的8质粒易于自我组合包装的可能干扰,又将MZ34的PB2和M质粒替换为YB0597或JT131来源的质粒,构成另外的2种10质粒组合Group 2:MZ34(6)+YB0597(PB2+M)+F/98(PB2+M)和Group 3:MZ34(6)+YB0597(PB2)+JT131(M)+F/98(PB2+M),经共染获得拯救病毒,对其PB2和M基因PCR扩增后测序鉴定。结果显示,从Group1、Group 2和Group 3中拯救获得的重组H5N6病毒中PB2基因的构成情况S∶H分别为41∶23、48∶15以及37∶19,M基因的构成情况S∶H分别为40∶24、31∶32以及25∶31,而PB2和M基因的组合情况SS∶SH∶HS∶HH分别为27∶14∶13∶10、26∶22∶5∶10以及17∶20∶8∶11。表明相较于H型的F/98-like PB2与M基因,S型的G1-like PB2基因在各10质粒转染组的拯救的H5N6病毒重配中具有更显著的竞争优势,而G1-like M基因仅在Group 1组拯救的重组病毒中表现出优势,但G1-like PB2与M基因间的竞争优势叠加效应不明显。本研究为解析S基因型H9N2 AIV作为H7N9、H10N8等新型重组流感病毒内部基因供体的相关机制提供重要参考。 展开更多

Antitumor activity is one characteristic function of some certain antimicrobial peptides(AMPs)found in recent years.In the present study,we attempted to detect potential anticancer activity of a recombinant piscidin 5-like from Larimichthys crocea(r Lc-P5L)which owned widely antibacterial and strong antiparasitic activity in vitro.The light microscope observation indicated r Lc-P5L was of antitumor activity to He La cells,293 T cells and L929 cells.MTT assay showed the toxic sensitivity of r Lc-P5L to three tumor cell strains was 293 T>L929>He La.Scanning electron microscope(SEM)results showed r Lc-P5L behaved like a lytic peptide to cause damage on cells membrane of L929 cells by forming globular clusters,even pores at 60μmol/L,or degrading membrane to make it completely lose cytoskeleton structure at 80μmol/L;r Lc-P5L treatment also resulted in DNA degradation.Fluorescence observation results indicated r Lc-P5L could cause L929 cells at least two obvious changes:one is nucleus,nuclear chromatin condensed in the margin,nuclear volume became smaller and shrank to be out of shape,or lysed to be debris;the other is cytoskeleton,they became disordered and polarized to make cells atrophic shapes,or even lysed to be debris.In summary,r Lc-P5L owned potential anticancer activity causing membrane structure damage and genome DNA degradation.Interestingly,treatment with different concentration of r Lc-P5L seemingly caused the similar but different changes,whether it indeed gave rise to cancer cells diverse death way,the further studies should be performed,and the detailed mechanisms were still need further explored. 展开更多

在对华东地区家养水禽中流感病毒的带毒状况的流行病学监测过程中,从表观健康家鸭体内分离到一株H5N1亚型禽流感病毒A/duck/Shandong/009/2008(简称Dk/SD/009/08)。为了解该毒株的基因组构成,对该分离株进行全基因测序。测序结果显示:... 在对华东地区家养水禽中流感病毒的带毒状况的流行病学监测过程中,从表观健康家鸭体内分离到一株H5N1亚型禽流感病毒A/duck/Shandong/009/2008(简称Dk/SD/009/08)。为了解该毒株的基因组构成,对该分离株进行全基因测序。测序结果显示:该毒株HA裂解位点处的氨基酸序列为PLRERRRK-R/GL,符合高致病性禽流感病毒的分子特征,且参照H5N1国际统一命名准则,Dk/SD/009/08的HA基因属于2.3.4进化支。BLAST结果显示,HA、NA、NP及NS基因均与H5N1亚型病毒的核苷酸一致性最高,而RNA聚合酶基因(PB2、PB1、PA)及M基因则与H9N2亚型病毒的亲缘关系最近,故推测该分离株可能是一株天然重组病毒;遗传进化分析进一步表明,流行于华南地区鹌鹑中的G1-like H9N2亚型病毒可能为该分离株提供部分的内部基因。 展开更多

为了解我国部分地区猪繁殖与呼吸综合征病毒(PRRSV)流行毒株的遗传变异情况和流行趋势,试验利用RT-PCR方法对2015年、2016年全国超过20个省份的3 078份疑似PRRS病料进行检测,并扩增部分阳性病料的ORF5基因进行测序和遗传变异分析。结... 为了解我国部分地区猪繁殖与呼吸综合征病毒(PRRSV)流行毒株的遗传变异情况和流行趋势,试验利用RT-PCR方法对2015年、2016年全国超过20个省份的3 078份疑似PRRS病料进行检测,并扩增部分阳性病料的ORF5基因进行测序和遗传变异分析。结果表明:PRRSV阳性样本共889份,阳性率为28.88%。扩增得到177个ORF5的基因序列,通过序列分析发现,我国流行的PRRSV主要为美洲型,其中90份与高致病性PRRSV(HP-PRRSV)高度同源,53份与经典PRRSV高度同源,34份与NADC30-like PRRSV高度同源,其中14份流行毒株的GP5蛋白糖基化位点N34发生缺失。说明监测地区主要流行HP-PRRSV,同时NADC30-like PRRSV已经形成一个相对独立的分支在我国流行,提示有必要加强PRRSV变异毒株的流行和变异的监测,同时对其致病性等生物学特性进行深入研究。 展开更多

This locus, qTGW3, e... Grain size and shape are important determinants of grain weight and yield in rice. Here, we report a new major quantitative trait locus (QTL), qTGW3, that controls grain size and weight in rice. This locus, qTGW3, encodes OsSK41 (also known as OsGSK5), a member of the GLYCOGEN SYNTHASE KINASE 3/SHAGGY-like family. Rice near-isogenic lines carrying the loss-of-function allele of OsSK41 have increased grain length and weight. We demonstrate that OsSK41 interacts with and phosphorylates AUXIN RESPONSE FACTOR 4 (OsARF4). Co-expression of OsSK41 with OsARF4 increases the accumulation of OsARF4 in rice protoplasts. Loss of function of OsARF4 results in larger rice grains. RNA-sequencing analysis suggests that OsARF4 and OsSK41 repress the expression of a common set of downstream genes, including some auxin-responsive genes, during rice grain development. The loss-of-function form of OsSK41 at qTGW3 represents a rare allele that has not been extensively utilized in rice breeding. Suppression of OsSK41 function by either targeted gene editing or QTL pyramiding enhances rice grain size and weight. Thus, our study reveals the important role of OsSK41 in rice grain development and provides new candidate genes for genetic improvement of grain yield in rice and perhaps in other cereal crops. 展开更多

Atherosclerosis is the major complication of diabetes and has become a major issue in the provision of medical care.In particular the economic burden is growing at an alarming rate in parallel with the increasing worldwide prevalence of diabetes.The major disturbance of lipid metabolism in diabetes relates to the effect of insulin on fat metabolism.Raised triglycerides being the hallmark of uncontrolled diabetes,i.e.,in the presence of hyperglycaemia.The explosion of type 2 diabetes has generated increasing interest on the aetiology ofatherosclerosis in diabetic patients.The importance of the atherogenic properties of triglyceride rich lipoproteins has only recently been recognised by the majority of diabetologists and cardiologists even though experimental evidence has been strong for many years.In the post-prandial phase 50% of triglyceride rich lipoproteins come from chylomicrons produced in the intestine.Recent evidence has secured the chylomicron as a major player in the atherogenic process.In diabetes chylomicron production is increased through disturbance in cholesterol absorption,in particular Neimann Pick C1-like1 activity is increased as is intestinal synthesis of cholesterol through 3-hydroxy-3-methyl glutaryl co enzyme A reductase.ATP binding cassette proteins G5 and G8 which regulate cholesterol in the intestine is reduced leading to chylomicronaemia.The chylomicron particle itself is atherogenic but the increase in the triglyceride-rich lipoproteins lead to an atherogenic low density lipoprotein and low high density lipoprotein.The various steps in the absorption process and the disturbance in chylomicron synthesis are discussed. 展开更多

AIM:To investigate the interaction of interleukin-23 receptor(IL23R)(rs1004819 and rs2201841),autophagy-related 16-like 1(ATG16L1)(rs2241880), caspase recruitment domain-containing protein 15 (CARD15)genes,and IBD5 locus in Crohn's disease(CD) patients. METHODS:A total of 315 unrelated subjects with CD and 314 healthy controls were genotyped.Interactions and specific genotype combinations of a total of eight variants were tested.The variants of IBD5locus(IGR2198a_1 rs11739135 and IGR2096a_1 rs12521868),CARD15(R702W rs2066845 and L1007fs rs2066847),ATG16L1(rs2241880)and IL23R (rs1004819,rs2201841)genes were genotyped by PCR-RFLP,the G908R(rs2066844)in CARD15 was determined by direct sequencing. RESULTS:The association of ATG16L1 T300A with CD was confirmed[P=0.004,odds ratio(OR)=1.69, 95%CI:1.19-2.41],and both IL23R variants were found to represent significant risk for the disease(P= 0.008,OR=2.05,95%CI:1.20-3.50 for rs1004819 AA;P<0.001,OR=2.97,95%CI:1.65-5.33 for rs2201841 CC).Logistic regression analysis of pairwise interaction of the inflammatory bowel disease (IBD)loci indicated that IL23R,ATG16L1,CARD15 and IBD5(IGR2198a_1)contribute independently to disease risk.We also analysed the specific combina- tions by pair of individual ATG16L1,IL23R rs1004819, rs2201841,IGR2198a_1,IGR2096a_1 and CARD15 genotypes for disease risk influence.In almost all cases,the combined risk of susceptibility pairs was higher in patients carrying two different risk-associated gene variants together than individuals with just one polymorphism.The highest OR was found for IL23R rs2201841 homozygous genotype with combination of positive CARD15 status(P<0.001,OR=9.15,95% CI:2.05-40.74). The sea anemone species, Aulactinia veratra (Phylum Cnidaria) contains different types of mucin genes. In the intertidal zone, A. veratra is found to be exposed to air during the low tide and produces large quantities of mucus as an external covering. The relation between low tide and mucus secretion is still unclear, and what is the role of mucin during arial exposure is not yet investigated. This study hypothesised that the mucin genes in A. veratra would have significantly high expression in response to aerial exposure. Therefore, the aim of current study was to examine and analyses the response of A. veratra mucins in response to an experiment involving three hours of aerial exposure. To achieve this, aim the RNA-sequencing and bioinformatics analyses were used to examine the expression profile of A. veratra mucin genes in response to aerial exposure. The generated results have shown that, Mucin4-like and mucin5B-like were up-regulated in response to the three hours of aerial exposure in A. veratra. This finding shows a significant role of mucin5B-like and mucin4-like genes in response to air stress at low tide. The data generated from this study could be used in conjunction with future mucin gene studies of sea anemones and other cnidarians to compare A. veratra mucin gene expression results across time, and to extend our understanding of mucin stress response in this phylum. 展开更多

AC in rice grains is mainly controlled by different alleles of the Waxy(Wx) gene. The AP2/EREBP transcription factor OsEBP89 interacts with the MYC-like protein OsBP5 to synergistically regulate the expression of Wx.Here, we determined that the GLYCOGEN SYNTHASE KINASE 5(OsGSK5, also named SHAGGY-like kinase 41 [OsSK41]) inhibits the transcriptional activation activity of OsEBP89 in rice grains during amylose biosynthesis. The loss of OsSK41 function enhanced Wx expression and increased AC in rice grains. By contrast, the loss of function of OsEBP89 reduced Wx expression and decreased AC in rice grains. OsSK41 interacts with OsEBP89 and phosphorylates four of its sites(Thr-28,Thr-30, Ser-238, and Thr-257), which makes OsEBP89 unstable and attenuates its interaction with OsBP5. Wx promoter activity was relatively weak when regulated by the phosphomimicvariantOsEBP89E–OsBP5but relatively strong when regulated by the nonphosphorylatable variant OsEBP89A–OsBP5.Therefore, OsSK41-mediated phosphorylation of OsEBP89 represents an additional layer of complexity in the regulation of amylose biosynthesis during rice grain development. In addition, our findings provide four possible sites for regulating rice grain AC via precise gene editing. 展开更多

In addition to full-length VPPs, several truncated forms are expressed, but their biological functions are unknown. In this study, we functionally characterized maize vacuole H^+-PPase 5(ZmVPP5), a truncated VPP in the maize genome. Although ZmVPP5 shares high sequence similarity with ZmVPP1 ZmVPP5 lacks the complete structure of the conserved proton transport and the inorganic pyrophosphatase-related domain. Phylogenetic analysis suggests that ZmVPP5 might be derived from an incomplete gene duplication event. ZmVPP5 is expressed in multiple tissues, and ZmVPP5 was detected in the plasma membrane, vacuole membrane and nuclei of maize cells. The overexpression of ZmVPP5 in yeast cells caused a hypersensitivity to salt stress. Transgenic maize lines with overexpressed ZmVPP5 also exhibited the salt hypersensitivity phenotype. A yeast two-hybrid analysis identified the ZmBag6 like protein as a putative ZmVPP5-interacting protein. The results of bimolecular luminescence complementation(Bi LC)assay suggest an interaction between ZmBag6-like protein and ZmVPP5 in vivo. Overall, this study suggests that ZmVPP5 might act as a VPP antagonist and participate in the cellular response to salt stress. Our study of ZmVPP5 has expanded the understanding of the origin and functions of truncated forms of plant VPPs. 展开更多
