Toxocariasis is a helminthic zoonosis due to the presence in the human body of larvae of Toxocara sp., roundworms of the Ascaridae family. Less than 50 cases of central involvement related to toxocarasis have been reported in immunocompetent individuals. This involvement can result in epilepsy, meningoencephalitis, myelitis or encephalopathy. The standard treatment is albendazole at a dosage of 10 to 15 mg/kg/day. The duration of treatment varies greatly depending on the clinical cases reported, ranging from 5 days to several weeks in the case of severe forms. We report a case of myelitis due to Toxocara canis in a 14-year-old patient admitted for gait disorders. The laboratory assessment shows isolated hypereosinophilia at 8000 elements per mm3. Medullary magnetic resonance imaging (MRI) showed an intradural process of inflammatory and infectious appearance extended between T10 and L1 levels, hypointense in T1, hyperintense in T2, and homogeneous. Parasitological analysis of the stools noted the presence of high concentrations of Toxocara canis. Serology by ELISA (enzyme-linked immunosorbent assay) is strongly positive for toxocariasis, and western blot confirms the presence of antibodies directed against Toxocara larvae. Treatment with albendazole 400 mg × 2/day for 10 days associated with corticosteroid therapy (prednisone 50 mg/day for 5 days) allowed the disappearance of pain in 8 days, normalization of eosinophilia and improvement of walking. 展开更多

We evaluated thirteen eyes of twelve patients diagnosed clinically and serologically with Toxocara optic neuropathy. Eleven patients had unilateral involvement and one patient had bilateral optic neuropathy. Eight patients (66.7%) had a possible infection source to Toxocara. Six patients (50%) had painless acute optic neuropathy. Ten eyes had asymmetric, sectorial optic disc edema with peripapillary infiltration and three eyes had diffuse optic disc edema. Eosinophilia was noted in five patients (41.7%) and optic nerve enhancement was observed in eight of eleven eyes (72.7%) with available orbit magnetic resonance imaging (MRI). Mean visual acuity significantly improved following treatment [mean logarithmic of the minimum angle of resolution (IogMAR) 0.94±0.56 at baseline and 0.47±0.59 at the final (P=0.02)]. Asymmetric optic disc edema with a peripapillary lesion and a history of raw meat ingestion were important clues for diagnosing Toxocara optic neuropathy. Additionally, Toxocara IgG enzymelinked immunosorbent assay (ELISA) test and evaluating eosinophil may be helpful for diagnosis. 展开更多

Arginine kinase in Toxocara canis:Exon-intron organization,functional analysis of site-directed mutants and evaluation of putative enzyme inhibitors

Objective: To determine exon/intron organization of the Toxocara canis(T. canis) AK(TCAK) and to test green and black tea and several other chemicals against the activity of recombinant TCAK in the guanidino-specific region by site-directed mutants. Methods: Amplification of genomic DNA fragments containing introns was carried out by PCRs. The open-reading frame(1 200 bp) of TCAK(wild type) was cloned into the BamH 1/SalI site of pM AL-c2X. The maltose-binding protein-TCAK fusion protein was expressed in Escherichia coli TB1 cells. The purity of the expressed enzyme was verified by SDS-PAGE. Mutations were introduced into the guanidino-specific region and other areas of pM AL/TCAK by PCR. Enzyme activity was measured with an NADH-linked assay at 25℃ for the forward reaction(phosphagen synthesis). Results: Arginine kinase in T. canis has a seven-exon/six-intron gene structure. The lengths of the introns ranged from 542 bp to 2 500 bp. All introns begin with gt and end with ag. Furthermore, we measured the enzyme activity of site-directed mutants of the recombinant TCAK. The K_m value of the mutant(Alanine to Serine) decreased indicating a higher affinity for substrate arginine than the wild-type. The K_m value of the mutant(Serine to Glycine) increased to 0.19 mM. The Km value(0.19 mM) of the double mutant(Alanine-Serine to Serine-Glycine) was slightly greater than in the wild-type(0.12 mM). In addition, several other chemicals were tested; including plant extract Azadiracta indica(A. indica), an aminoglycoside antibiotic(aminosidine), a citrus flavonoid glycoside(rutin) and a commercially available catechin mixture against TCAK. Green and black tea(1:10 dilution) produced 15% and 25% inhibition of TCAK, respectively. The extract of A. indica produced 5% inhibition of TCAK. Moreover, green and black tea produced a non-competitive type of inhibition and A. indica produced a mixed-type of inhibition on TCAK. Conclusions: Arginine kinase in T. canis has a seven-exon/six-intron gene structure. However, further studies are needed to identify a specific compound within the extract causing the inhibitory effect and also to determine the molecular mechanisms behind inhibition of arginine kinase in T. canis. 展开更多

Toxocara canis(T.canis)is one of the most important zoonotic parasites of dogs.The aim of the present study was to perform an in vitro analysis of the effect of Milbemycin oxime on T.canis eggs following exposure to a concentration gradient of the drug and to determine the inflammatory reaction produced by the infective T.canis larvae in mice.The present study was undertaken using the model nematode,T.canis,to investigate the effect of Milbemycin oxime on T.canis eggs and larvae.T.canis eggs were exposed to a concentration gradient of Milbemycin oxime in vitro,the higher concentration of Milbemycin oxime was,the lower percentage of infective stage larvae was.Light micrographs showed that Milbemycin oxime induced eggs dissolved and eggshell broken.Histological analyses of mice that stained with hematoxylin and eosin(H&E)showed in lower dosing(10-7 and 10-8 g·mL-1)drug-treated groups,atrophy of alveolar space and interalveolar septum thickening appeared,inflammatory infiltrates accompanied with erythrocytes around blood vessels and bronchioles presented.In higher dosing(10-6,10-5 and 10-4 g·mL-1)drug-treated groups,low-grade or no pathological changes occurred,indicating that Milbemycin oxime could obviously decrease the inflammatory reaction produced by the infection of T.canis larvae in vivo. 展开更多

The frequency of the infective larvae of Toxocara canis, the common parasite of the dog was assessed in brain portions of male Albino mice ingested two doses: 200 mg/Kg and 400 mg/Kg of potassium dichromate-hexavalent (VI) as an indicator. Only 2,000 ova of the parasite were inoculated orally to twenty seven healthy young male mice. Frequency of the parasite was tested 13 weeks post-inoculation. Using bio-statistical analysis (one way analysis of variance) the frequency of detected larvae of both doses in the brain portions of the experimental animals showed an insignificant difference (P〉 0.05) in comparison with control. The results indicate the safety of small and mild intoxication doses of the potassium dichromate on distribution of Toxocara canis in brain portions of mice. 展开更多

The average age at diagnosis of ocular Toxocariasis is 7.5 years, ranging from 2 to 31 years. It constitutes 1%-2% of uveitis in children. Diagnosis is based upon the clinical features observed in a young patient and confirmed by the presence of specific Ig G in the serum or aqueous humor by Enzyme-linked immunosorbent assay test. We report a case of Presumed Toxocara infection in 45-year-old male which is unique in presentation with multifocal granulomata in retina. Our Pub Med search could not produce case with similar presentation. Probably this is the first reported case of multifocal granulomata in presumed ocular Toxocara in any age 展开更多

Background Toxocara canis,the most prevalent helminth in dogs and other canines,is one of the socioeconomically important zoonotic parasites,particularly affecting pediatric and adolescent populations in impoverished communities.However,limited information is available regarding the proteomes of female and male adult T.canis.To address this knowledge gap,we performed a comprehensive proteomic analysis to identify the proteins with differential abundance(PDAs)and gender-specifically expressed proteins between the two sexes adult T.canis.Methods The comparative proteomic analysis was carried out by the Orbitrap mass spectrometry(MS)with asymmetric track lossless(Astral)analyzer.The difference analysis was conducted using t-test and the proteins verification was achieved through parallel reaction monitoring(PRM).The potential biological functions of identified adult T.canis proteins and PDAs were predicted by Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)databases.The domain,transcription factor and subcellular localization of the identified proteins and PDAs were analyzed by InterPro,AnimalTFDB 4.0 and Cell-mPLOC 2.0 databases,respectively.Results A total of 8565 somatic proteins of adult T.canis were identified.Compared to male adult,682 up-regulated PDAs and 844 down-regulated PDAs were identified in female adult with P-values<0.05 and|log2FC|>1,including 139 proteins exclusively expressed in female and 272 proteins exclusively expressed in male.The GO annotation analysis using all PDAs revealed that the main biological processes,cellular components and molecular functions corresponded to aminoglycan metabolic process,extracellular region and protein tyrosine phosphatase activity,respectively.The KEGG analysis using all PDAs showed that the pathways were mainly associated with adipocytokine signaling pathway,proximal tubule bicarbonate reclamation and PPAR signaling pathway.Conclusions This study reveals the differential protein expression between female and male adult T.canis,providing valuable resource for developing the novel intervention strategies against T.canis infection in humans and animals,especially from the perspective of sexual development and reproduction. 展开更多

弓首蛔虫病是一种被严重忽视的人畜共患寄生虫病,呈世界性分布,主要由弓首蛔虫的感染性幼虫在人及多种动物体内移行所致。本研究以犬弓首蛔虫组织移行的第1个环节“侵入宿主”为切入点,首先用5%健康小鼠血清体外刺激孵化的犬弓首蛔虫感... 弓首蛔虫病是一种被严重忽视的人畜共患寄生虫病,呈世界性分布,主要由弓首蛔虫的感染性幼虫在人及多种动物体内移行所致。本研究以犬弓首蛔虫组织移行的第1个环节“侵入宿主”为切入点,首先用5%健康小鼠血清体外刺激孵化的犬弓首蛔虫感染性幼虫,然后提取虫体样本的总RNA进行小RNA测序,最后开展处理组与未处理组的虫体微RNA(microRNA, miRNA)差异表达分析及靶基因预测。结果显示:在4个处理组样本和4个未处理组样本中共检测到972个miRNAs,其中血清刺激造成15个miRNAs的转录水平发生显著变化;靶基因预测发现,这些转录水平发生显著变化的miRNAs靶向宿主免疫细胞黏附与迁移、嗅觉转导等相关基因,在寄生虫与宿主互作中发挥作用。本研究鉴定到一组具有宿主血清反应性的犬弓首蛔虫感染性幼虫miRNAs,为其在感染宿主、免疫调控以及致病中的具体作用研究奠定了基础。 展开更多

犬弓首蛔虫超氧化物歧化酶的免疫保护性分析

为探究犬弓首蛔虫(Toxocara canis,T.canis)超氧化物歧化酶(superoxide dismutase,SOD)对小鼠的免疫保护效果,选取75只6周龄昆明系小鼠随机分为5组进行免疫保护试验,分别于试验0、14、28 d对每只小鼠皮下注射50、75和125 mg/L的Tc-SOD 0... 为探究犬弓首蛔虫(Toxocara canis,T.canis)超氧化物歧化酶(superoxide dismutase,SOD)对小鼠的免疫保护效果,选取75只6周龄昆明系小鼠随机分为5组进行免疫保护试验,分别于试验0、14、28 d对每只小鼠皮下注射50、75和125 mg/L的Tc-SOD 0.1 mL,空白组不做任何处理,PBS组注射等量的无菌PBS。共免疫3次,每次免疫间隔14 d;第3次免疫后14 d,经口感染3000个/只感染性虫卵。在每次免疫前、第3次免疫后14 d和攻虫后7 d对小鼠尾静脉采血并分离血清,采用间接ELISA法检测血清中特异性IgG抗体;通过CCK-8和qRT-PCR法分析小鼠脾淋巴细胞的增殖能力及Th1(IFN-γ,IL-2)和Th2(IL-4,IL-10)型细胞因子mRNA的表达水平;计算攻虫后7 d的幼虫减虫率并采用苏木精-伊红(H&E)染色观察肝脏和肺脏的病理变化。结果显示,与PBS组相比,Tc-SOD免疫组小鼠血清IgG抗体水平随着免疫次数的增加和时间延长而升高,攻虫感染后仍维持在较高水平,差异极显著(P<0.001)。三免后14 d,IFN-γ、IL-2、IL-4和IL-10的mRNA表达水平均极显著增高(P<0.01),其中IL-4和IL-10显著高于IFN-γ和IL-2(P<0.05),表现为Th2型为主的免疫反应。小鼠脾淋巴细胞的增殖能力显著增强(P<0.01)。50、75和125 mg/L的Tc-SOD免疫组的幼虫减虫率分别为18.7%、24.9%、37.6%,其中125 mg/L Tc-SOD组差异显著(P<0.05),具有较好的免疫保护效果。H&E染色结果显示,3个Tc-SOD免疫组均缓解了小鼠肝脏和肺脏的炎性细胞浸润(中性粒细胞、嗜酸粒细胞、巨噬细胞)和出血性病变。上述研究结果表明,Tc-SOD可诱导小鼠产生体液免疫和细胞免疫应答,以Th2型免疫反应为主,并对T.canis的感染具有一定程度的免疫保护作用。 展开更多

弓首蛔虫病是一种严重危害动物和人健康的全球性线虫病,虫卵是该病的主要感染源。为研究弓首蛔虫卵的体外发育形态学变化,本研究采用彩色显微图像智能识别系统对体外培养的弓首蛔虫3个常见种:犬弓首蛔虫、猫弓首蛔虫和犊弓首蛔虫虫卵进... 弓首蛔虫病是一种严重危害动物和人健康的全球性线虫病,虫卵是该病的主要感染源。为研究弓首蛔虫卵的体外发育形态学变化,本研究采用彩色显微图像智能识别系统对体外培养的弓首蛔虫3个常见种:犬弓首蛔虫、猫弓首蛔虫和犊弓首蛔虫虫卵进行连续21 d的观察和记录。结果表明:1)犬弓首蛔虫和犊弓首蛔虫虫卵存在12个体外发育阶段,包括1细胞期、2细胞期、3细胞期、4细胞期、早期桑葚期、晚期桑葚期、囊胚期、肠胚期、1期幼虫前期、1期幼虫期、2期幼虫前期和2期幼虫期,猫弓首蛔虫卵因5细胞期的出现而存在13个体外发育阶段。2)比较观察发现,弓首蛔虫卵在培养24 h后出现卵裂,进入2~5细胞期,随后虫卵第3~5天陆续进入桑葚期、囊胚期、肠胚期;猫弓首蛔虫卵第6天出现1期幼虫(2.2%),第7天出现感染性2期幼虫(2.1%);犬弓首蛔虫卵第7天出现1期幼虫(22.5%),第8天出现感染性2期幼虫(10.7%);犊弓首蛔虫卵第6天出现1期幼虫(1.9%),第9天出现感染性2期幼虫(0.9%);由此可见,猫弓首蛔虫卵是弓首属发育速度最快的虫卵。以上研究结果补充和完善了弓首蛔虫卵体外发育的形态学基础信息。 展开更多

为了调查陕西省榆林地区犬弓首蛔虫的流行情况,并研究其基因变异、遗传多样性及种系发育关系,试验采用饱和盐水漂浮法对采自该地区的402份犬粪便进行虫卵检查,利用IBM SPSS Statistics 22软件对虫卵检测数据进行统计和分析,并采用χ^(2... 为了调查陕西省榆林地区犬弓首蛔虫的流行情况,并研究其基因变异、遗传多样性及种系发育关系,试验采用饱和盐水漂浮法对采自该地区的402份犬粪便进行虫卵检查,利用IBM SPSS Statistics 22软件对虫卵检测数据进行统计和分析,并采用χ^(2)检验分析各地区、各类型犬感染犬弓首蛔虫的差异性;PCR扩增犬弓首蛔虫的cox1、18S rRNA基因序列并测序;运用Clustal X与DNAStar软件分析2个基因的碱基组成与遗传多样性;运用Clustal X及PhyML 3.0软件分析犬弓首蛔虫的种系发育关系。结果表明:陕西省榆林地区犬对弓首蛔虫的总感染率为20.65%,其中宠物犬的感染率为8.42%,而流浪犬的感染率为33.00%,各地区、各类型犬感染犬弓首蛔虫的情况存在一定差异;cox1基因序列长度为1044 bp,A+T含量为61.5%~62.2%,G+C含量为37.8%~38.5%,种内差异为0~1.3%,种间差异为9.87%~14.37%;18S rRNA基因序列长度为598 bp,A+T含量为50.0%~50.5%,G+C含量为49.5%~50.0%,种内差异为0~1.4%,种间差异为0.33%~6.68%。在2个种系发育树中,犬弓首蛔虫均互相掺杂在一起共同形成每个种系发育树的一个分支。说明犬弓首蛔虫在榆林地区的流行情况较为严重,各个体之间虽存在较低程度的基因变异和遗传多样性,但具有较近的亲缘关系。 展开更多

为探究犬弓首蛔虫(Toxocara canis,T.canis)超氧化物歧化酶(Superoxide Dismutase, SOD)的分子特性,根据GenBank中登录的Tc-sod基因序列(GenBank:AAB00227.1),以T.canisc DNA为模板对Tc-sod全长基因进行扩增,并进行序列分析和多重序列比... 为探究犬弓首蛔虫(Toxocara canis,T.canis)超氧化物歧化酶(Superoxide Dismutase, SOD)的分子特性,根据GenBank中登录的Tc-sod基因序列(GenBank:AAB00227.1),以T.canisc DNA为模板对Tc-sod全长基因进行扩增,并进行序列分析和多重序列比对;同时构建Tc-sod/pET-32a原核表达载体,经ITPG诱导表达,对重组蛋白进行纯化并制备多克隆抗体.结果显示Tc-sod全长基因为573 bp,共编码190个氨基酸;多重序列分析表明Tc-SOD的氨基酸序列与曼氏血吸虫、异尖线虫、美洲板口线虫、马来丝虫、犬钩口线虫均具有SOD-Cu保守结构域.种系发育分析发现Tc-SOD与马来丝虫进化关系较近. SDS-PAGE结果显示重组蛋白Tc-SOD的大小约为37 ku,以可溶性形式表达;利用Ni-NTA亲和层析柱纯化蛋白,以70 mmol/L咪唑进行洗脱时可获得高纯度的目的蛋白;将纯化后的重组蛋白免疫新西兰大白兔制备多克隆抗体,间接ELISA检测显示抗体滴度>1∶320 000,表明重组蛋白的免疫原性较好;Western Blot结果显示抗体能与Tc-SOD蛋白特异性结合,表明抗体特异性高. 展开更多

【目的】对弓首属犬弓首蛔虫、猫弓首蛔虫、马来西亚弓首蛔虫、牛弓首蛔虫和狮弓属狮弓蛔虫线粒体DNA(mtDNA)中烟酰胺脱氢酶亚基I(nad1)基因部分序列(pnad1)进行比较研究,分析它们之间的序列差异和种群遗传关系。【方法】先用同位素(γ3... 【目的】对弓首属犬弓首蛔虫、猫弓首蛔虫、马来西亚弓首蛔虫、牛弓首蛔虫和狮弓属狮弓蛔虫线粒体DNA(mtDNA)中烟酰胺脱氢酶亚基I(nad1)基因部分序列(pnad1)进行比较研究,分析它们之间的序列差异和种群遗传关系。【方法】先用同位素(γ33P)标记上下游引物,通过PCR方法扩增出单个虫体线粒体pnad1片段,然后采用单链构象多态性(SSCP)分析技术对pnad1片段进行多态性研究,最后挑选出代表性样品进行测序,并利用基因分析软件DNAStar(版本5.01)对序列进行分析比较。【结果】犬弓首蛔虫种内pnad1基因序列差异为0.3%~1.9%,与猫弓首蛔虫、马来亚弓首蛔虫、牛弓首蛔虫和狮弓蛔虫的种间序列差异分别为11.0%~14.1%、10.7%~12.4%、12.6%~13.7%和17.5%~18.2%。猫弓首蛔虫种内pnad1基因序列差异为2.8%,与马来西亚弓首蛔虫、牛弓首蛔虫和狮弓蛔虫的种间序列差异分别是12.0%~13.0%、15.1%和18.6%~21.2%。马来西亚弓首蛔虫种内pnad1基因序列差异为0~0.3%,与牛弓首蛔虫和狮弓蛔虫的种间序列差异分别是12.4%~12.8%和18.6%~19.0%。【结论】犬弓首蛔虫、猫弓首蛔虫和马来西亚弓首蛔虫不同地方虫株的pnad1序列都有一定差异(0~2.8%)。但种间pnad1的序列差异(10.7%~21.2%)明显高于种内的序列差异,说明nad1基因可以作为弓首蛔虫和狮弓蛔虫分子分类及种群遗传关系研究的遗传标记。 展开更多

目的比较犬、猫常见蛔虫的线粒体DNA(mtDNA)中烟酰胺脱氢酶亚基Ⅳ基因(nad4)部分序列(pnad4),以期找出它们之间的序列差异和种群遗传关系。方法抽提61个弓首属蛔虫(包括犬弓首蛔虫、猫弓首蛔虫、马来西亚弓首蛔虫和犊弓首蛔虫)和狮弓蛔... 目的比较犬、猫常见蛔虫的线粒体DNA(mtDNA)中烟酰胺脱氢酶亚基Ⅳ基因(nad4)部分序列(pnad4),以期找出它们之间的序列差异和种群遗传关系。方法抽提61个弓首属蛔虫(包括犬弓首蛔虫、猫弓首蛔虫、马来西亚弓首蛔虫和犊弓首蛔虫)和狮弓蛔虫虫体的总DNA,用PCR方法扩增mtDNApnad4,然后用SSCP技术和DNA测序对序列进行分析研究。结果犬弓首蛔虫种群内pnad4的序列差异为1.17%,与猫弓首蛔虫、马来西亚弓首蛔虫、牛弓首蛔虫和狮弓蛔虫种间的平均序列差异分别为13.92%、16.48%、15.12%和20.32%。猫弓首蛔虫种群内pnad4的序列差异为1.00%,与马来西亚弓首蛔虫、牛弓首蛔虫、狮弓蛔虫平均序列差异分别是17.90%、13.55%和18.50%。马来西亚弓首蛔虫种群内pnad4序列差异为0.13%,与牛弓首蛔虫、狮弓蛔虫序列差异分别为13.50%和20.60%。结论犬弓首蛔虫不同地方虫株的pnad4有一定差异,猫弓首蛔虫种群内差异较小,马来西亚弓首蛔虫种群内差异很小。弓首属各虫种之间的差异以及与狮弓蛔虫的差异都较大。马来西亚弓首蛔虫pnad4序列与其它虫种差异都较大,证明它确为一独立种。pnad4序列是弓首蛔虫和狮弓蛔虫理想的种特异的遗传标记。 展开更多

本文报道犬弓首线虫幼虫体外培养、排泄分泌抗原(TES抗原)的制备和分析结果。采用自行研制的幼虫分离器分离人工孵化的犬弓首线虫第二期幼虫,用Eagle's基础培养基培养幼虫,并用饱和硫酸铵盐析法从培养液中提取TES抗原均取得满意结... 本文报道犬弓首线虫幼虫体外培养、排泄分泌抗原(TES抗原)的制备和分析结果。采用自行研制的幼虫分离器分离人工孵化的犬弓首线虫第二期幼虫,用Eagle's基础培养基培养幼虫,并用饱和硫酸铵盐析法从培养液中提取TES抗原均取得满意结果。用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE)分析显示此抗原,可见到7条蛋白质区带。免疫双扩散表明本抗原与本虫感染的免血清(1:32)作用呈阳性反应。免疫电泳显示至少含有四条沉淀线。转移电泳免疫识别显示本抗原含有5~7个血清学抗原成份。 展开更多
