Mango(Mangifera indica L.)is one of the main economic crops in Hainan,China,prized for its distinctive flavor and high nutritional value.It is also rich in health-promoting antioxidants such as vitamin C and flavonoid...Mango(Mangifera indica L.)is one of the main economic crops in Hainan,China,prized for its distinctive flavor and high nutritional value.It is also rich in health-promoting antioxidants such as vitamin C and flavonoids.Enhanced ultraviolet-B(UV—B)radiation,a growing global environmental concern,alters plant antioxidant systems,with increased flavonoid accumulation as a common adaptive response.However,its effects on mango fruit remain largely unexplored.To investigate the antioxidant responses of mango to enhanced UV-B radiation and identify key responsive flavonoid compounds and regulatory genes,we exposed‘Tainong 1’mango fruits growing under natural light to 96 kJ·m^(-2)·d^(-1)of UV-B radiation to simulate high UV-B conditions.Treated fruits were smaller in size and had a pulp of a more intense yellow colour.Further,malondialdehyde content in treated fruits was higher during the phase of rapid fruit enlargement.Additionally,treated fruits showed increased sugar-acid ratios,total phenol,total flavonoid,carotenoid,and ascorbic acid contents.Furthermore,they showed significantly enhanced antioxidant activity,as measured by the FRAP,ABTS,and DPPH assays.Extensive targeted metabolomic-analysis identified flavonoids as the largest category of compounds differentially expressed in treated and control groups.Quantitative metabolomics of flavonoids identified hyperoside,quercimeritrin,and(-)-catechin gallate as the key flavonoid metabolites responsive to UV-B treatment.Transcriptome analysis revealed an enrichment of the flavonoid biosynthesis pathway,with most associated differentially expressed genes showing upregulation.Furthermore,qRT-PCR analysis confirmed that the expression of the genes MiCHS7,MiCHI1,MiCHI2,MiFLS,MiF3H2,and MiF3H3 correlated with changes in key flavonoid metabolites.Indeed,correlation analysis indicated that MiCHS7,MiCHI1,MiFLS,and MiF3H3 are potential key genes involved in flavonoid accumulation under UV-B treatment.Thus,our study provides a theoretical basis for breeding for new resilient varieties and developing UV-B-resistant mango cultivation techniques.展开更多

On the basis of single-factor experiment,Box-Behnken response surface methodology was employed for selecting the optimum extraction process. [Results]The optimum extraction conditions of total flavonoids were as follows: 75% of ethanol concentration,1∶ 25 of ratio of material to liquid,31 min of ultrasonic time. Under these conditions,the extraction rate of total flavonoids was 1. 115%.[Conclusions] The extraction process obtained by response surface methodology was stable,reasonable,accurate and reliable. Rat nerve growth factor and total flavonoids from hawthorn leaf contribute to the recovery of neurological function after spinal cord injury,including traumatic,non-traumatic spinal cord injuries.However,it remains ch...Rat nerve growth factor and total flavonoids from hawthorn leaf contribute to the recovery of neurological function after spinal cord injury,including traumatic,non-traumatic spinal cord injuries.However,it remains challenging to efficiently deliver nerve growth factor and total flavonoids from hawthorn leaf to spinal cord injury sites,ensure their sustained release,and minimize further damage.In the present study,we chose a biocompatible and biodegradable gelatin as the substrate,which was crosslinked with the natural biological crosslinker genipin to form a gelatin-genipin hydrogel carrier for the slow release of nerve growth factor and total flavonoids from hawthorn leaf in spinal cord injury sites.The prepared gelatin-genipin hydrogel had good injectable properties and photothermal effects.Furthermore,when the hydrogel with 2%genipin,200 ng/mL nerve growth factor,and 320μg/mL total flavonoids from hawthorn leaf was combined with near infrared irradiation,there was a slow release of total flavonoids from hawthorn leaf and nerve growth factor,reduced oxidative stress,an improved inflammatory microenvironment,and accelerated angiogenesis and axonal regeneration via inhibition of the nuclear factor kappa-B signaling pathway,thereby promoting recovery from spinal cord injury.Collectively,our results indicate that this new hydrogel may improve the prognosis of spinal cord injury,and may represent a new strategy for treating spinal cord injury.展开更多

Amyotrophic lateral sclerosis is characterized by the progressive loss of motor neurons.Early-stage axonal dysfunction,rather than central nervous system injury,plays a key role in the disease process.However,the mole...Amyotrophic lateral sclerosis is characterized by the progressive loss of motor neurons.Early-stage axonal dysfunction,rather than central nervous system injury,plays a key role in the disease process.However,the molecular mechanisms underlying this dysfunction remain unclear.To investigate the relationship between peripheral immune dysregulation and axonal dysfunction in amyotrophic lateral sclerosis,we recruited 372 patients within the first 12 months of sporadic amyotrophic lateral sclerosis onset between January 2018 and May 2024.We collected peripheral immune markers at baseline,including total leukocytes,lymphocytes,monocytes,neutrophils,basophils,eosinophils,and platelets.We also calculated four derived ratios:neutrophil-to-lymphocyte ratio,platelet-to-lymphocyte ratio,lymphocyte-to-monocyte ratio,and systemic immune inflammation index.Multivariate analysis,adjusted for confounding factors,revealed that higher counts of total leukocytes and neutrophils,as well as higher neutrophil-related ratios,including the neutrophil to lymphocyte ratio and the systemic immune inflammation index,were significantly correlated with higher compound muscle action potential scores.Stratified analyses revealed that these associations varied by age and sex.Furthermore,mediation analysis demonstrated that axonal dysfunction plays a significant role in the relationship between immune markers and disease progression.These findings emphasize the critical role that peripheral immune dysregulation plays in amyotrophic lateral sclerosis progression by mediating peripheral nerve injury,particularly in the early stages of the disease.This study highlights the importance of the peripheral nervous system in the early stages of amyotrophic lateral sclerosis and provides new insights into disease mechanisms and potential therapeutic targets.展开更多
[Objectives]Based on spectrophotometry,a method for determining the total flavonoid content in milk powder was established by optimizing sample pretreatment conditions,and method validation was performed.[Methods]Tota...[Objectives]Based on spectrophotometry,a method for determining the total flavonoid content in milk powder was established by optimizing sample pretreatment conditions,and method validation was performed.[Methods]Total flavonoids in milk powder were extracted with ethanol using 45℃ water bath ultrasonication for 60 min.Flavonoids contain a benzopyran ring structure and react with aluminum nitrate in a potassium acetate environment to form a yellow complex.The absorbance of this complex was measured at 420 nm for quantitative determination.[Results]The method exhibited a good linear relationship within the range of 0.2-1.0 mg.The limit of detection(LOD)was 0.05 g/100 g,and the limit of quantification(LOQ)was 0.3 g/100 g.When the spiked amount of total flavonoids was in the range of 0.3% to 1.0%,the spike recovery rates were 99.3% to 103.4%,and the relative standard deviations(RSD s)were less than 3.0%.[Conclusions]This method offers advantages such as rapidness,accuracy,good stability,and high sensitivity,and can be used for the detection of total flavonoid content in milk powder.展开更多
Retrieval analysis in total knee arthroplasty(TKA)has been little studied in the literature.A narrative review of the literature to understand the current importance of retrieval analysis in TKA has been conducted.On ...Retrieval analysis in total knee arthroplasty(TKA)has been little studied in the literature.A narrative review of the literature to understand the current importance of retrieval analysis in TKA has been conducted.On August 27,2024,a literature search was performed in PubMed using“TKA retrieval analysis”as keywords.A total of 160 articles were found,of which only 19 were analyzed because they were directly related to the subject of this article.Rotating-platform(mobile-bearing)TKA has no surface damage advantage over fixed-bearing TKA.TKAs with central locking mechanisms are more prone to debond from the cement mantle.No major wear of the polyethylene(PE)component in TKA using oxidized zirconium components occurs.Femoral components of cobalt-chromium roughen more than oxidized zirconium femoral components.The use of a polished tibial tray over an unpolished design is advised.At short-run assessment(15 months on average),antioxidant-stabilized highly crosslinked PE components are not clinically different in surface damage,density of crosslinking,or oxidation compared to standard remelted highly crosslinked PE components.A correlation between implant position and PE component surface damage has been reported.It shows the importance of optimizing component position to reduce PE component damage.Contemporary knee tumor megaendoprostheses show notable volumetric metal wear originated at the rotating hinge.Retrieval analysis in TKA renders relevant data on how different prosthetic designs described in the literature perform.Such information can help to improve future prosthetic designs to increase prosthetic survival.展开更多
Objective:To perform phytochemical screening,estimate total phenolics,flavonoids and to evaluate antioxidant potential of Moringa peregrina(M.peregrina) leaves.Methods:The dried powdered leaves of M.peregrina(150 g) w...Objective:To perform phytochemical screening,estimate total phenolics,flavonoids and to evaluate antioxidant potential of Moringa peregrina(M.peregrina) leaves.Methods:The dried powdered leaves of M.peregrina(150 g) were extracted exhaustively by Soxhlet with ethanol and then fractionated into hexane,chloroform,ethy alacetate and methanol.All the prepared extracts were also analyzed by gas chromatography-mass spectrometry to identify and characterize the chemical compounds present in the crude extracts.Folin- Ciocalteu reagent and aluminium chloride colorimetric methods were used to estimate total phenolic and flavonoid content of extracts.Hydrogen peroxide and 1,1 diphenyl-2-picrylhydrazyl were used to determine in vitro antioxidant activity.Results:Phytochemical analysis of ethanol extract showed presence of major classes of phytochemicals.Gas chromatography-mass spectrometry results revealed presence of 19 phytoconstituents in hexane extract,6 in ethyl acetate and 7 compounds in methanolic extract.Methanol extract was found to contain the highest phenolic content and flavonoids.In vitro antioxidant activities of all crude extracts were significant and comparable with the standard ascorbic acid.Conclusions:Results of this study show that the leaves of M.peregrina are the rich source of phenolic compounds that can play an important role in preventing the progression of many diseases.展开更多

In silico analysis, the OsMRP15 (LOC_Os06g06440), an orthologous gene of mazie anthocyanin transporter ZmMRP3, has been identified in rice. The OsMRP15 contained a 4425bp open reading frame (ORF) encoding a 1475 amino acid protein, belonging to a MRP subfamily of ABC transporters, and has a high sequence identity, very similar protein structure, and the same arrangement of domains to ZmMRP3, but the genomic structure of OsMRP15 was significant difference with ZmMRP3. The prediction promoter of OsMRP15 has many presumed anthocyanin regulatory sites. The phylogenetic analysis of MRPs in rice, mazie and Arabidopsis showed that OsMRP15 and ZmMRP3 belonged to the same subbranch. The expression pattern indicated that OsMRP15 was co-expression with two anthocyanin transcription factors. These analysis results implied that as an ortholog of ZmMRP3, the function of OsMRP15 was possibly as a membrane-bound transporter required for vacuolar uptake of anthocyanins in rice.展开更多

And t...Objective: In this study, one of the objectives was to investigate the total flavonoid contents of Fupenzi (R. chingii Hu.) obtained from different regions of China and to evaluate their anatioxidant activities. And the second objective of this study was to develop a validated HPLC method for chromatographic fingerprints of the samples extracts of Fupenzi. Method: The total flavonoid contents were determined by a colorimetric method and the antioxidant activity was determined spectrophotometrically by DPPH and ABTS radical scavenging assays. The chromatographic fingerprint was developed by high-performance liquid chromatography coupled with diode array detection for the control of Fupenzi. Results: A significant correlation between antioxidant activity and the total flavonoid content was observed for the DPPH assay (r2 = 0.758, ρ = 0.004) and the ABTS assay (r2 = 0.788, ρ = 0.002). Under the optimized chromatographic conditions, the validated method was successfully applied to assessment of chemical fingerprinting of 12 batches of FPZ collected from different regions of China. Comparisons of the chromatograms showed that 15 characteristic peaks could be selected as markers for identification and evaluation of Fupenzi. In addition, the proposed method was also successfully applied to simultaneous determination of five compounds (including puerarin, rutin, hyperin, quercetin and kaempferol) in these samples. Conclusions: The qualitative and quantitative analysis described in this paper could be used for identification and evaluation of Fupenzi.展开更多

The compounds in the mixture were clearly identified by comparing their HPLC-DAD ultraviolet spectra, retention times, and MS data with those of corresponding reference compounds. All calibration curves showed good linearity (r2 > 0.9998) within the test ranges. The LOD, LOQ, specificity, precision, and accuracy for the method were validated. The results demonstrated that this analytical approach is ideal for the determination of bioactive compounds, such as flavonoids, and that it constructed a basis for the comprehensive evaluation of the quality of Salix bordensis Turcz.展开更多

Methods The static and dynamic adsorption/desorption methods were used, and the separation and purification process was evaluated by measuring the concentration of total flavonoid in the fractions with UV spectrophotometer. Results The SP70 macroporous resin was the most effective compared with other macroporous resins. The optimum conditions were screened, which were 0.5 g· mL^- 1 corresponding to crude drug for concentration of extract, pH 5 - 6, and appended 60 times the volume of the resin bed (BV) with the adsorption speed 2 BV·h^-1, and the volume of aq. 70% (V/V) ethanol as eluant was 5 BV with desorption speed 2 BV·h^-1. By this method, the final contents of total flavonoids exceeded 80%. Conclusion The SP70 macroporous resin is the most effective one for large-scale isolation and purification of flavonoids from Radix Pueraria, which meets industrial needs.展开更多

TFE (300 mg/kg) was administered once daily to male Sprague-Dowley rats by gavage for fifteen days. The total cytochrome P450 content and its main isoforms CYP1A2, CYP3A4 and CYP2E1 activities in rat liver microsomes were detected. The activity of CYP1A2 was measured by fluorometry and the activities of CYP3A4 and CYP2E1 were determined by measuring the amount of methanal and p-aminophenol formed using UV/Vis spectrophotometer, respectively. Administration of TFE significantly increased the total CYP450 content and activities ofCYP1A2, CYP3A4 and CYP2E1 in rat liver microsomes, compared with the control group. Particularly, the activities of CYP1A2 and CYP2E1 were enhanced significantly (P〈0.01). TFE induced the increase in total CYP450 content and its main isoforms CYP1A2, CYP3A4 and CYP2E1 activities in rat liver microsomes.展开更多

Suaeda salsa an annual herb of Chenopodiaceae Suaeda well accepted as both a folk medicinal and food plant.In the current study,the extraction of total flavonoids from S.salsa and the in vitro antioxidant,hypoglycemic...Suaeda salsa an annual herb of Chenopodiaceae Suaeda well accepted as both a folk medicinal and food plant.In the current study,the extraction of total flavonoids from S.salsa and the in vitro antioxidant,hypoglycemic and hypolipidemic activities of the extracts were investigated.The total flavonoids was extracted by refluxing with ethanol and the selection and optimization of main experimental parameters were carried out using single factor experiments combined with central composite design-response surface methodology(CCD-RSM).The total flavonoids extracted from S.salsa were tested for hydroxyl and oxygen radical scavenging activity,and inhibitory activities againstα-amylase and lipase in vitro.Results revealed that the optimal conditions for the extraction of total flavonoids were as following:liquid-tosolid ratio,31.40:1;ethanol concentration,83.12%;reflux temperature,90℃,and reflux time,128.40 min.Under these optimized conditions,the predicted and actual values of total flavonoids yield were 0.896%and 0.932%,respectively.The in vitro antioxidant tests revealed that the total flavonoids could scavenge hydroxyl and oxygen radicals and showed inhibitory activity againstα-amylase and lipase.The optimal extraction conditions obtained in this experiment are simple to perform and have good reproducibility.The results have demonstrated that the total flavonoids from S.salsa can be used as a potential source of natural antioxidant,hypoglycemic and hypolipidemic ingredients,which provides new pharmacological basis for the development of S.salsa.展开更多
Objective:To prepare various crude extracts using different polarities of solvent and to quantitatively evaluate their total phenol,flavonoids contents and phytochemical screening of Thymus vulgaris collected from Al ...Objective:To prepare various crude extracts using different polarities of solvent and to quantitatively evaluate their total phenol,flavonoids contents and phytochemical screening of Thymus vulgaris collected from Al Jabal Al Akhdar,Nizwa.Sultanate of Oman.Methods:The leave sample was extracted with methanol and evaporated.Then it was defatted with water and extracted with different polarities organic solvents with increasing polarities.The prepare hexane,chloroform,ethyl acetate,hutanol and methanol crude extracts were used for their evaluation of total phenol,flavonoids contents and phytochemical screening study.The established conventional metliods were used for quantitative determination of total phenol,flavonoids contents and phytochemical screening.Results:Phytochemical screening for various crude extracts were tested and shown positive result for flavonoids,saponins and steroids compounds.The result for total phenol content was the highest in hutanol and the lowest in methanol crude extract whereas the total flavonoids contents was the highest in methanol and the lowest hexane crude extract.Conclusions:The crude extracts from locally grown Thymus vulgaris showed high concentration of flavonoids and it could be used as antibiotics for different curable and uncurable diseases.展开更多

[Methods] The ultrasonic extraction method was adopted and orthogonal design was carried out to study the effects of ethanol concentration( A),ultrasonic time( B),the solid-to-liquid ratio( C) on the extraction of flavonoids from leaves of P. ramosissimus. The Microplate Reader was used to measure the content of flavonoids in leaves of P. ramosissimus. in different extraction conditions,P. ramosissimus. and the optimal extraction conditions were used to measure the content of flavonoids from leaves of P. ramosissimus.[Results]Effects of the ultrasonic extraction method on the extraction rate of flavonoids from leaves of P. ramosissimus: extraction times >ethanol concentration > Ultrasound time > solid-liquid ratio. The screened optimal extraction process was: ethanol concentration of 65%,the solid-to-liquid ratio of 1∶ 25,ultrasonic time of 2 h and extraction times Under this condition,the extraction rate of total flavonoids in leaves of P. ramosissimus. was 2. 56%. 文摘Mango(Mangifera indica L.)is one of the main economic crops in Hainan,China,prized for its distinctive flavor and high nutritional value.It is also rich in health-promoting antioxidants such as vitamin C and flavonoids.Enhanced ultraviolet-B(UV—B)radiation,a growing global environmental concern,alters plant antioxidant systems,with increased flavonoid accumulation as a common adaptive response.However,its effects on mango fruit remain largely unexplored.To investigate the antioxidant responses of mango to enhanced UV-B radiation and identify key responsive flavonoid compounds and regulatory genes,we exposed‘Tainong 1’mango fruits growing under natural light to 96 kJ·m^(-2)·d^(-1)of UV-B radiation to simulate high UV-B conditions.Treated fruits were smaller in size and had a pulp of a more intense yellow colour.Further,malondialdehyde content in treated fruits was higher during the phase of rapid fruit enlargement.Additionally,treated fruits showed increased sugar-acid ratios,total phenol,total flavonoid,carotenoid,and ascorbic acid contents.Furthermore,they showed significantly enhanced antioxidant activity,as measured by the FRAP,ABTS,and DPPH assays.Extensive targeted metabolomic-analysis identified flavonoids as the largest category of compounds differentially expressed in treated and control groups.Quantitative metabolomics of flavonoids identified hyperoside,quercimeritrin,and(-)-catechin gallate as the key flavonoid metabolites responsive to UV-B treatment.Transcriptome analysis revealed an enrichment of the flavonoid biosynthesis pathway,with most associated differentially expressed genes showing upregulation.Furthermore,qRT-PCR analysis confirmed that the expression of the genes MiCHS7,MiCHI1,MiCHI2,MiFLS,MiF3H2,and MiF3H3 correlated with changes in key flavonoid metabolites.Indeed,correlation analysis indicated that MiCHS7,MiCHI1,MiFLS,and MiF3H3 are potential key genes involved in flavonoid accumulation under UV-B treatment.Thus,our study provides a theoretical basis for breeding for new resilient varieties and developing UV-B-resistant mango cultivation techniques.

文摘Background:The study aimed to investigate the protective effect and mechanism of total flavonoids of Scutellaria baicalensis(TFSB)on acute myocardial ischemia(AMI)rats by using functional metabonomics.Methods:Rats were divided into the Control,Model,AMI positive control(Propranolol hydrochloride,30 mg/kg),low dose TFSB(50 mg/kg),and high dose TFSB(100 mg/kg)groups.Rats received the corresponding treatment by intragastric administration once daily for 10 consecutive days.Electrocardiogram,myocardial enzyme,triphenyltetrazolium chloride staining,hematoxylin-eosin,and enzyme-linked immunosorbent assay were performed to evaluate the protective effect of TFSB on AMI rats.Then,the UHPLC-Q-Orbitrap MS method based on serum metabolomics was utilised to search for metabolic biomarkers and metabolic pathways.Subsequently,Western blot and RT-PCR techniques were employed to identify the respective genes and proteins.Results:Pharmacodynamics revealed that TFSB could ameliorate AMI in rats.The results of the metabolomics analysis indicated that the alterations in metabolic profile observed in rats with AMI were partially improved by treatment with TFSB.Moreover,the mRNA expression levels of 5-lipoxygenase(5-LOX)and 15-lipoxygenase(15-LOX)and the protein expression levels of 5-LOX,15-LOX,interleukin-1β(IL-1β),and NF-κB p65 were reduced following treatment with TFSB.Conclusion:The potential treatment of TFSB in AMI may be ascribed to its ability to regulate arachidonic acid metabolism.

文摘[Objectives]To analyze the chemical components of Meconopsis integrifolia(Maxim.)Franch total flavonoids(MITF).[Methods]The chemical components of MITF were identified by UPLC-Q-Exactive Orbitrap MS.The column was performed using ACQUITY UPLC HSS T3(100 mm×2.1 mm,1.8μm)with 0.1%formic acid water(A)-acetonitrile(B)with gradient elution at 30℃,injection volume of 2μL,and flow rate of 0.3 mL/min.Electrospray ion source adopted positive and negative ion detection mode with scanning range m/z 100-1000.[Results]A total of 93 compounds were identified from MITF,including 57 flavonoids,25 phenolic acids,9 alkaloids and 2 others.Among them,dimethoxytaxifolin was identified as a possible new compound by SciFinder search,and 67 compounds were first identified in M.integrifolia.[Conclusions]This study provides a scientific foundation for clarifying the material basis of the efficacy of M.integrifolia and improving the quality standards.

文摘[Objectives] Response surface analysis methodology was used to optimize the extraction technology of total flavonoids from Mallotus apelta leaves. [Methods] The total flavonoids were extracted using ultrasonic assisted ethanol extraction,and extraction rate of total flavonoids was taken as evaluation index. On the basis of single-factor experiment,Box-Behnken response surface methodology was employed for selecting the optimum extraction process. [Results]The optimum extraction conditions of total flavonoids were as follows: 75% of ethanol concentration,1∶ 25 of ratio of material to liquid,31 min of ultrasonic time. Under these conditions,the extraction rate of total flavonoids was 1. 115%.[Conclusions] The extraction process obtained by response surface methodology was stable,reasonable,accurate and reliable. It was a feasible method to extract the total flavonoids from M. apelta leaves.

文摘Objective:To study the effect of total flavonoids of onion treatment on oxidative stress and cell apoptosis in the retinal tissue of diabetic model rats.Methods:Male SD rats were selected as experimental animals and randomly divided into control group, diabetes group, low-dosage total flavonoids group (total flavonoids L group) and high-dose total flavonoids (total flavonoids H group), intraperitoneal injection of streptozotocin was adopted to make diabetes model, and the total flavonoids L group and H group received lavage of 100 mg/kg and 200 mg/kg total flavonoids of onion for intervention. After 8 weeks of intervention, retinal tissue was collected and the degree of oxidative stress and cell apoptosis were determined.Results:P2X7, ROS, -OH, O2-, H2O2, FADD, p-JNK, caspase-8 and caspase-3 levels in retinal tissue of diabetes group were significantly higher than those of control group while Mn-SOD, CAT, GSH, AsA, NGF, BDNF and CNTF levels were significantly lower than those of control group;P2X7, ROS, -OH, O2-, H2O2, FADD, p-JNK, caspase-8 and caspase-3 levels in retinal tissue of total flavonoids L group and total flavonoids H group were significantly lower than those of diabetes group while Mn-SOD, CAT, GSH, AsA, NGF, BDNF and CNTF levels were significantly higher than those of diabetes group;P2X7, ROS, -OH, O2-, H2O2, FADD, p-JNK, caspase-8 and caspase-3 levels in retinal tissue of total flavonoids H group were significantly lower than those of total flavonoids L group while Mn-SOD, CAT, GSH, AsA, NGF, BDNF and CNTF levels were significantly higher than those of total flavonoids L group.Conclusion:Total flavonoids of onion have inhibiting effect on the oxidative stress and cell apoptosis in retinal tissue of diabetic model rats.

文摘Rat nerve growth factor and total flavonoids from hawthorn leaf contribute to the recovery of neurological function after spinal cord injury,including traumatic,non-traumatic spinal cord injuries.However,it remains challenging to efficiently deliver nerve growth factor and total flavonoids from hawthorn leaf to spinal cord injury sites,ensure their sustained release,and minimize further damage.In the present study,we chose a biocompatible and biodegradable gelatin as the substrate,which was crosslinked with the natural biological crosslinker genipin to form a gelatin-genipin hydrogel carrier for the slow release of nerve growth factor and total flavonoids from hawthorn leaf in spinal cord injury sites.The prepared gelatin-genipin hydrogel had good injectable properties and photothermal effects.Furthermore,when the hydrogel with 2%genipin,200 ng/mL nerve growth factor,and 320μg/mL total flavonoids from hawthorn leaf was combined with near infrared irradiation,there was a slow release of total flavonoids from hawthorn leaf and nerve growth factor,reduced oxidative stress,an improved inflammatory microenvironment,and accelerated angiogenesis and axonal regeneration via inhibition of the nuclear factor kappa-B signaling pathway,thereby promoting recovery from spinal cord injury.Collectively,our results indicate that this new hydrogel may improve the prognosis of spinal cord injury,and may represent a new strategy for treating spinal cord injury.

文摘Amyotrophic lateral sclerosis is characterized by the progressive loss of motor neurons.Early-stage axonal dysfunction,rather than central nervous system injury,plays a key role in the disease process.However,the molecular mechanisms underlying this dysfunction remain unclear.To investigate the relationship between peripheral immune dysregulation and axonal dysfunction in amyotrophic lateral sclerosis,we recruited 372 patients within the first 12 months of sporadic amyotrophic lateral sclerosis onset between January 2018 and May 2024.We collected peripheral immune markers at baseline,including total leukocytes,lymphocytes,monocytes,neutrophils,basophils,eosinophils,and platelets.We also calculated four derived ratios:neutrophil-to-lymphocyte ratio,platelet-to-lymphocyte ratio,lymphocyte-to-monocyte ratio,and systemic immune inflammation index.Multivariate analysis,adjusted for confounding factors,revealed that higher counts of total leukocytes and neutrophils,as well as higher neutrophil-related ratios,including the neutrophil to lymphocyte ratio and the systemic immune inflammation index,were significantly correlated with higher compound muscle action potential scores.Stratified analyses revealed that these associations varied by age and sex.Furthermore,mediation analysis demonstrated that axonal dysfunction plays a significant role in the relationship between immune markers and disease progression.These findings emphasize the critical role that peripheral immune dysregulation plays in amyotrophic lateral sclerosis progression by mediating peripheral nerve injury,particularly in the early stages of the disease.This study highlights the importance of the peripheral nervous system in the early stages of amyotrophic lateral sclerosis and provides new insights into disease mechanisms and potential therapeutic targets.

文摘[Objectives]Based on spectrophotometry,a method for determining the total flavonoid content in milk powder was established by optimizing sample pretreatment conditions,and method validation was performed.[Methods]Total flavonoids in milk powder were extracted with ethanol using 45℃ water bath ultrasonication for 60 min.Flavonoids contain a benzopyran ring structure and react with aluminum nitrate in a potassium acetate environment to form a yellow complex.The absorbance of this complex was measured at 420 nm for quantitative determination.[Results]The method exhibited a good linear relationship within the range of 0.2-1.0 mg.The limit of detection(LOD)was 0.05 g/100 g,and the limit of quantification(LOQ)was 0.3 g/100 g.When the spiked amount of total flavonoids was in the range of 0.3% to 1.0%,the spike recovery rates were 99.3% to 103.4%,and the relative standard deviations(RSD s)were less than 3.0%.[Conclusions]This method offers advantages such as rapidness,accuracy,good stability,and high sensitivity,and can be used for the detection of total flavonoid content in milk powder.
文摘Retrieval analysis in total knee arthroplasty(TKA)has been little studied in the literature.A narrative review of the literature to understand the current importance of retrieval analysis in TKA has been conducted.On August 27,2024,a literature search was performed in PubMed using“TKA retrieval analysis”as keywords.A total of 160 articles were found,of which only 19 were analyzed because they were directly related to the subject of this article.Rotating-platform(mobile-bearing)TKA has no surface damage advantage over fixed-bearing TKA.TKAs with central locking mechanisms are more prone to debond from the cement mantle.No major wear of the polyethylene(PE)component in TKA using oxidized zirconium components occurs.Femoral components of cobalt-chromium roughen more than oxidized zirconium femoral components.The use of a polished tibial tray over an unpolished design is advised.At short-run assessment(15 months on average),antioxidant-stabilized highly crosslinked PE components are not clinically different in surface damage,density of crosslinking,or oxidation compared to standard remelted highly crosslinked PE components.A correlation between implant position and PE component surface damage has been reported.It shows the importance of optimizing component position to reduce PE component damage.Contemporary knee tumor megaendoprostheses show notable volumetric metal wear originated at the rotating hinge.Retrieval analysis in TKA renders relevant data on how different prosthetic designs described in the literature perform.Such information can help to improve future prosthetic designs to increase prosthetic survival.

文摘Objective:To perform phytochemical screening,estimate total phenolics,flavonoids and to evaluate antioxidant potential of Moringa peregrina(M.peregrina) leaves.Methods:The dried powdered leaves of M.peregrina(150 g) were extracted exhaustively by Soxhlet with ethanol and then fractionated into hexane,chloroform,ethy alacetate and methanol.All the prepared extracts were also analyzed by gas chromatography-mass spectrometry to identify and characterize the chemical compounds present in the crude extracts.Folin- Ciocalteu reagent and aluminium chloride colorimetric methods were used to estimate total phenolic and flavonoid content of extracts.Hydrogen peroxide and 1,1 diphenyl-2-picrylhydrazyl were used to determine in vitro antioxidant activity.Results:Phytochemical analysis of ethanol extract showed presence of major classes of phytochemicals.Gas chromatography-mass spectrometry results revealed presence of 19 phytoconstituents in hexane extract,6 in ethyl acetate and 7 compounds in methanolic extract.Methanol extract was found to contain the highest phenolic content and flavonoids.In vitro antioxidant activities of all crude extracts were significant and comparable with the standard ascorbic acid.Conclusions:Results of this study show that the leaves of M.peregrina are the rich source of phenolic compounds that can play an important role in preventing the progression of many diseases.
文摘There are many studies on enzymatic pathways of anthocyanin biosynthesis, but little is known about the anthocyanins transport in Oryze sativa. In silico analysis, the OsMRP15 (LOC_Os06g06440), an orthologous gene of mazie anthocyanin transporter ZmMRP3, has been identified in rice. The OsMRP15 contained a 4425bp open reading frame (ORF) encoding a 1475 amino acid protein, belonging to a MRP subfamily of ABC transporters, and has a high sequence identity, very similar protein structure, and the same arrangement of domains to ZmMRP3, but the genomic structure of OsMRP15 was significant difference with ZmMRP3. The prediction promoter of OsMRP15 has many presumed anthocyanin regulatory sites. The phylogenetic analysis of MRPs in rice, mazie and Arabidopsis showed that OsMRP15 and ZmMRP3 belonged to the same subbranch. The expression pattern indicated that OsMRP15 was co-expression with two anthocyanin transcription factors. These analysis results implied that as an ortholog of ZmMRP3, the function of OsMRP15 was possibly as a membrane-bound transporter required for vacuolar uptake of anthocyanins in rice.
文摘Objective: In this study, one of the objectives was to investigate the total flavonoid contents of Fupenzi (R. chingii Hu.) obtained from different regions of China and to evaluate their anatioxidant activities. And the second objective of this study was to develop a validated HPLC method for chromatographic fingerprints of the samples extracts of Fupenzi. Method: The total flavonoid contents were determined by a colorimetric method and the antioxidant activity was determined spectrophotometrically by DPPH and ABTS radical scavenging assays. The chromatographic fingerprint was developed by high-performance liquid chromatography coupled with diode array detection for the control of Fupenzi. Results: A significant correlation between antioxidant activity and the total flavonoid content was observed for the DPPH assay (r2 = 0.758, ρ = 0.004) and the ABTS assay (r2 = 0.788, ρ = 0.002). Under the optimized chromatographic conditions, the validated method was successfully applied to assessment of chemical fingerprinting of 12 batches of FPZ collected from different regions of China. Comparisons of the chromatograms showed that 15 characteristic peaks could be selected as markers for identification and evaluation of Fupenzi. In addition, the proposed method was also successfully applied to simultaneous determination of five compounds (including puerarin, rutin, hyperin, quercetin and kaempferol) in these samples. Conclusions: The qualitative and quantitative analysis described in this paper could be used for identification and evaluation of Fupenzi.
文摘The qualitative characterization and quantitative analysis of five bioactive flavonoids in Salix bordensis Turcz. were achieved via reversed-phase high-performance liquid chromatography coupled with diode array detection and tandem mass spectrometry, by using an Agilent ZORBAX SB-C18 HPLC column with a gradient elution of 0.3% (v/v) formic acid in water and methanol as the mobile phase. The compounds in the mixture were clearly identified by comparing their HPLC-DAD ultraviolet spectra, retention times, and MS data with those of corresponding reference compounds. All calibration curves showed good linearity (r2 > 0.9998) within the test ranges. The LOD, LOQ, specificity, precision, and accuracy for the method were validated. The results demonstrated that this analytical approach is ideal for the determination of bioactive compounds, such as flavonoids, and that it constructed a basis for the comprehensive evaluation of the quality of Salix bordensis Turcz.

文摘Aim To screen the optimum macroporous resin and conditions for the isolation and purification of flavonoids from Radix Puerariae. Methods The static and dynamic adsorption/desorption methods were used, and the separation and purification process was evaluated by measuring the concentration of total flavonoid in the fractions with UV spectrophotometer. Results The SP70 macroporous resin was the most effective compared with other macroporous resins. The optimum conditions were screened, which were 0.5 g· mL^- 1 corresponding to crude drug for concentration of extract, pH 5 - 6, and appended 60 times the volume of the resin bed (BV) with the adsorption speed 2 BV·h^-1, and the volume of aq. 70% (V/V) ethanol as eluant was 5 BV with desorption speed 2 BV·h^-1. By this method, the final contents of total flavonoids exceeded 80%. Conclusion The SP70 macroporous resin is the most effective one for large-scale isolation and purification of flavonoids from Radix Pueraria, which meets industrial needs.

文摘To assess the potential effect of total flavonoids of epimedium (TFE) on cytochrome P450 and activity of its main isoforms in rat liver microsomes. TFE (300 mg/kg) was administered once daily to male Sprague-Dowley rats by gavage for fifteen days. The total cytochrome P450 content and its main isoforms CYP1A2, CYP3A4 and CYP2E1 activities in rat liver microsomes were detected. The activity of CYP1A2 was measured by fluorometry and the activities of CYP3A4 and CYP2E1 were determined by measuring the amount of methanal and p-aminophenol formed using UV/Vis spectrophotometer, respectively. Administration of TFE significantly increased the total CYP450 content and activities ofCYP1A2, CYP3A4 and CYP2E1 in rat liver microsomes, compared with the control group. Particularly, the activities of CYP1A2 and CYP2E1 were enhanced significantly (P〈0.01). TFE induced the increase in total CYP450 content and its main isoforms CYP1A2, CYP3A4 and CYP2E1 activities in rat liver microsomes.

文摘Objective:To detect the in vitro total phenolics,flavonoids contents and antioxidant activity of essential oil,various organic extracts from the leaves of tropical medicinal plant Tetrastigma from Sabah.Methods:The dry powder leaves of Tetrastigma were extracted with different organic solvent such as hexane.ethyl acetate,chloroform,butanol and aqueous methanol.The total phenolic and total flavonoids contents of the essential oil and various organic extracts such as hexane,ethyl acetate,chloroform,butanol and aqueous ethanol were determined by Folin-Ciocalteu method and the assayed antioxidant activity was determined in vitro models such as antioxidant capacity by radical scavenging activity usingα,α-diphenyl-β-picrylhydrazyl (DPPH) method.Results:The total phenolic contents of the essential oil and different extracts as gallic acid equivalents were found to be highest in methanol extract(386.22 mg/g) followed by ethyl acetate(190.89 mg/g).chloroform(175.89 mg/g).hexane(173.44 mg/g).and Imtanol extract (131.72 mg/g) and the phenolic contents not detected in essential oil.The antioxidant capacity of the essential oil and different extracts as ascorbic acid standard was in the order of methanol extract】ethyl acetate extract】chloroform】butanol】hexane extract also the antioxidant activity was not detected in essential oil.Conclusions:The findings show that the extent of antioxidant activity of the essential oil and all extracts are in accordance with the amount of phenolics present in that extract.Leaves of Tetrastigma being rich in phenolics may provide a good source of antioxidant.
文摘Suaeda salsa an annual herb of Chenopodiaceae Suaeda well accepted as both a folk medicinal and food plant.In the current study,the extraction of total flavonoids from S.salsa and the in vitro antioxidant,hypoglycemic and hypolipidemic activities of the extracts were investigated.The total flavonoids was extracted by refluxing with ethanol and the selection and optimization of main experimental parameters were carried out using single factor experiments combined with central composite design-response surface methodology(CCD-RSM).The total flavonoids extracted from S.salsa were tested for hydroxyl and oxygen radical scavenging activity,and inhibitory activities againstα-amylase and lipase in vitro.Results revealed that the optimal conditions for the extraction of total flavonoids were as following:liquid-tosolid ratio,31.40:1;ethanol concentration,83.12%;reflux temperature,90℃,and reflux time,128.40 min.Under these optimized conditions,the predicted and actual values of total flavonoids yield were 0.896%and 0.932%,respectively.The in vitro antioxidant tests revealed that the total flavonoids could scavenge hydroxyl and oxygen radicals and showed inhibitory activity againstα-amylase and lipase.The optimal extraction conditions obtained in this experiment are simple to perform and have good reproducibility.The results have demonstrated that the total flavonoids from S.salsa can be used as a potential source of natural antioxidant,hypoglycemic and hypolipidemic ingredients,which provides new pharmacological basis for the development of S.salsa.

文摘Objective:To prepare various crude extracts using different polarities of solvent and to quantitatively evaluate their total phenol,flavonoids contents and phytochemical screening of Thymus vulgaris collected from Al Jabal Al Akhdar,Nizwa.Sultanate of Oman.Methods:The leave sample was extracted with methanol and evaporated.Then it was defatted with water and extracted with different polarities organic solvents with increasing polarities.The prepare hexane,chloroform,ethyl acetate,hutanol and methanol crude extracts were used for their evaluation of total phenol,flavonoids contents and phytochemical screening study.The established conventional metliods were used for quantitative determination of total phenol,flavonoids contents and phytochemical screening.Results:Phytochemical screening for various crude extracts were tested and shown positive result for flavonoids,saponins and steroids compounds.The result for total phenol content was the highest in hutanol and the lowest in methanol crude extract whereas the total flavonoids contents was the highest in methanol and the lowest hexane crude extract.Conclusions:The crude extracts from locally grown Thymus vulgaris showed high concentration of flavonoids and it could be used as antibiotics for different curable and uncurable diseases.

文摘[Objectives] To screen the optimal extraction conditions of flavonoids from leaves of Paliurus ramosissimus( Lour.) Poir.,measure the content of flavonoids in leaves of P. ramosissimus. using the Microplate Reader,and provide experimental basis for the development of medicinal resources of P. ramosissimus. [Methods] The ultrasonic extraction method was adopted and orthogonal design was carried out to study the effects of ethanol concentration( A),ultrasonic time( B),the solid-to-liquid ratio( C) on the extraction of flavonoids from leaves of P. ramosissimus. The Microplate Reader was used to measure the content of flavonoids in leaves of P. ramosissimus. in different extraction conditions,P. ramosissimus. and the optimal extraction conditions were used to measure the content of flavonoids from leaves of P. ramosissimus.[Results]Effects of the ultrasonic extraction method on the extraction rate of flavonoids from leaves of P. ramosissimus: extraction times >ethanol concentration > Ultrasound time > solid-liquid ratio. The screened optimal extraction process was: ethanol concentration of 65%,the solid-to-liquid ratio of 1∶ 25,ultrasonic time of 2 h and extraction times Under this condition,the extraction rate of total flavonoids in leaves of P. ramosissimus. was 2. 56%. [Conclusions] This method is simple and feasibility,reliable,and suitable for rapid measurement with large samples.
