Antimicrobial activity against periodontopathogenic bacteria,antioxidant and cytotoxic effects of various extracts from endemic Thermopsis turcica

Objective:To investigate the in vitro antimicrobial potential of Thermopsis turcica Kit Tan,Vural&Kckdk against periodoutopathogenie bacteria,its antioxidant activity and cytotoxic effect on various cancer cell lines.Methods:In vitro antimicrobial activities of elhanol.methanol,ethyl acetate(ElAc,n-hexane and water extracts of Thermopsis turcica herb against periodontopathogenic bacteria,Aggregatibacter actinnmycelemconilans ATCC 29523 and Porphyromonas gingivalis ATCC 33277 were tested by agar well diffusion,minimum inhibitory concentration(MIC)and minimal bactericidal concentration(MBC),Antioxidant properties of the extracts were evaluatod by 1,1-diphenyl-2-picryl-hydrazyl radical scavenging activity and p-carotene bleaching methods.Amounts of phenolic contents of the extracts were also analysed by using the Folin-Ciocalteu reagent.Additionally,cytotoxic activity of the extracts on androgcn-insensilivc prostate cancer,androgen—sensitive prostate cancer,chronic myelogenous leukemia and acute promyelocytic leukemia bunian cancer cell lines were determined by 3-4,5-dimelhylthiazol-2-yh-2,5-diphenyltclrazolium bromide assay.Human gingival fibroblast cells were used as a control.Results:Our data showed that ELAc extract had the highest antimicrobial effect on Aggregatibacter actinomycetemitans(MIC:1.562 mg/ml_MHC:3.124 mg/ml.)and Porph yromonas gingiralis(MIC:0.781 mg/mL,MBC:1.562 mg/mL).In antioxidant assays.ElAc extract exhibited also the highest radical scavenging activity[IC_(50)=(30.0±0.3)μg/ml.]and the highest inhibition[(74.35±0.30)%]|against lineloic acide oxidation.The amount of phenolic content of it was also the highest[(l62.5±l.2)μg/mg gallic acid].In cytotoxic assay,only etbanol[IC_(50)=(80.00±1.21)μg/ml.]and EtAc extract[IC_(50)=(70.0±0.9)μg/ml]were toxic on acute promyeloeytic leukemia cells at 20—100μg/mL.P<0.05>.However,no toxic effect was observed on human gingival fibroblast cells.Cunclusions:According to our findings,owing to its antioxidant and cytotoxic potential,EtAc exlrael might include anticancer agents for acute promyelocytic leukemia. 展开更多

The aim of this study was to determine the factors controlling the inte... Thermopsis turcica is an endemic species of wild legume in Türkiye. The plant produces flower pistils with unusual morphological properties. The aim of this study was to determine the factors controlling the interesting feature of this plant. Particularly the controlling features of the flower meristem and the pistils that mutations in two genes and helps occurring the number of flower organs and can cause abnormalities, This study has attempted to explore the controlling feature of Thermopsis turcica, the young bud Arabidopsis CLV2, and CRC gene homologs were isolated and analyzed in different tissues. First, with total RNA isolated from the first thread degenerated primers synthesized cDNA expressions, second with a partial length cDNA cloning process made and the replicated set of analyses. The full-length cDNA of genes related to RACE analysis was carried out with the sequencing process. The real-time PCR method was applied for gene functional analyses with buds, sepals, petals and stamen and relative expression analysis of genes in the tissue of pistils. Full-length cDNA sequence for the gene CLV2 2557 bp, the encoded protein 716 amino acids, CRC 901 for full-length cDNA sequence of the gene—bp, the encoded protein 174 identified as amino acids. Alignment and phylogenetic analyses of CLV2 and CRC genes were found to be similar to other types of legumes in homologous sets. Real-time (RT-qPCR) PCR results relative to study courses with genes. 展开更多

披针叶黄华(Thermopsis lanceolata)是我国西部地区早春重要野生蜜源植物,也是一种重要的固沙植物,然而对其繁殖特性的研究甚少。本文在系统调查披针叶黄华的访花昆虫基础上,确定其主要传粉昆虫种类、访花行为、传粉过程以及日活动规律... 披针叶黄华(Thermopsis lanceolata)是我国西部地区早春重要野生蜜源植物,也是一种重要的固沙植物,然而对其繁殖特性的研究甚少。本文在系统调查披针叶黄华的访花昆虫基础上,确定其主要传粉昆虫种类、访花行为、传粉过程以及日活动规律,以期揭示主要访花者行为对其有性繁殖的影响。作者在内蒙古毛乌素沙地设置1个10m×10m的样方,于2010和2011年在披针叶黄华盛花期,采用目测、拍照和摄像等方式对传粉昆虫进行观测,记录样方内主要访花昆虫种类、数量、访花行为及日活动规律。研究表明,大和切叶蜂(Megachilejaponica)和戎拟孔蜂(Hoplitisprinceps)是披针叶黄华的主要传粉者,但两种昆虫的访花频率存在显著差异;晴天时,大和切叶蜂在9:00-13:00和16:00-18:00出现两个活动高峰,而戎拟孔蜂只在11:30-16:30出现1个活动高峰,两种蜂的访花活动高峰期存在互补关系。大和切叶蜂访花同时具有盗蜜行为,但其盗蜜行为对披针叶黄华的结籽率没有显著影响。根据种群数量、访花频率综合判断,大和切叶蜂是披针叶黄华优势传粉蜂。 展开更多

水是种子萌发的关键因素,种子萌发及幼苗生长直接影响着干旱区植被的分布和多样性。以铃铛刺(Halimodendron halodendron)和披针叶野决明(Thermopsis lanceolata)种子为研究材料,用不同浓度PEG6000模拟干旱胁迫,探讨干旱胁迫对铃铛刺和... 水是种子萌发的关键因素,种子萌发及幼苗生长直接影响着干旱区植被的分布和多样性。以铃铛刺(Halimodendron halodendron)和披针叶野决明(Thermopsis lanceolata)种子为研究材料,用不同浓度PEG6000模拟干旱胁迫,探讨干旱胁迫对铃铛刺和披针叶野决明种子萌发的影响。结果表明:不同浓度处理均降低了铃铛刺和披针叶野决明种子的发芽率、发芽势、日相对发芽率和平均发芽速率,延缓了种子萌发进程,胚芽和胚根生长也受到抑制。不同浓度处理中,12%是相对高萌发率的浓度,其种子的发芽率、发芽势、日相对发芽率、平均发芽速率、胚根及胚芽长均明显高于其他处理,与对照相近。干旱胁迫对铃铛刺和披针叶野决明种子萌发影响不完全一样,如对胚芽和胚根生长的抑制作用,铃铛刺种子胚芽和胚根作用相近,披针叶野决明则对胚根作用较胚芽明显。 展开更多
