The Detection of THCA Using 2-Dimensional Gas Chromatography-Tandem Mass Spectrometry in Human Fingernail Clippings: Method Validation and Comparison with Head Hair

Marijuana use as well as abuse is a significant public health and public safety concern in the United States and using hair to identify marijuana users and abusers has been gaining acceptance in a number of venues including workplace, court ordered, and substance abuse treatment monitoring. After the presentation of a fully validated 2-dimensional gas chromatography-tandem mass spectrometry method for the detection of 11-nor-9-carboxy-Δ9-tetrahydrocannabinol (THCA), the chief metabolite of the main psychoactive compound in marijuana, Δ9-tetrahydrocannabinol (THC), we evaluated the usefulness of fingernail clippings as an alternative specimen type to hair by the analysis of a set of 60 matched pairs of head hair and fingernail clippings. The limit of detection was 10 fg/mg, the limit of quantitation was 20 fg/mg, and the assay was linear from 20 fg/mg to 500 fg/mg. The intra- and inter-assay imprecision and bias studies at 4 different concentrations (50, 100, 500, and 1000 fg/mg) were acceptable where all % Target observations were within 16% of their expected concentrations and all %CV calculations were less than 13.5%. THCA was detectable in more fingernail specimens (53.3%) than hair specimens (46.7%) and the mean concentrations in nails were on average 4.9 times higher than in hair (1813 fg/mg and 364 fg/mg, respectively). The THCA concentrations in hair and nail were strongly associated (r = 0.974, P < 0.01, n = 60) and the association was significant. The study demonstrated that fingernail clippings are a suitable alternative specimen type to hair to monitor for marijuana use and abuse. 展开更多

目的利用生物信息学探讨依托泊苷诱导蛋白2.4(Etoposide-induced 2.4,EI^(2)4)在甲状腺癌(Thyroid cancer,THCA)中的诊断和预后价值并进行功能分析和体外实验验证。方法利用肿瘤基因组图谱(TCGA)数据库和人类蛋白图谱(HPA)数据库分析EI^... 目的利用生物信息学探讨依托泊苷诱导蛋白2.4(Etoposide-induced 2.4,EI^(2)4)在甲状腺癌(Thyroid cancer,THCA)中的诊断和预后价值并进行功能分析和体外实验验证。方法利用肿瘤基因组图谱(TCGA)数据库和人类蛋白图谱(HPA)数据库分析EI^(2)4在THCA中的表达水平及其与临床特征的关系。采用Kaplan-Meier生存分析、Cox回归分析和受试者工作特征曲线(ROC)评价EI^(2)4对THCA的诊断及预后价值。通过基因富集分析(GSEA)探索可能的细胞机制和生物学功能,并利用蛋白互作数据库(STRING)探索EI^(2)4的互作蛋白。随后,本研究通过细胞转染、实时荧光定量PCR(RT-qPCR)、Western blot(WB)、CCK-8、克隆形成实验、Transwell实验、Oris细胞迁移实验等探讨EI^(2)4不同表达水平对THCA细胞功能的影响。结果与正常甲状腺组织相比,THCA组织中EI^(2)4 mRNA及其蛋白水平均呈高表达,且与THCA患者的总生存期(Overall survival,OS)和无进展间期(Progress free interval,PFI)等均显著相关。ROC曲线分析表明,EI^(2)4在THCA中的曲线下面积(AUC)为0.900。较高的EI^(2)4 mRNA表达与预后不佳有明显相关关系,且M分期、病理阶段和EI^(2)4的表达状态是THCA患者PFI的独立危险因素。京都基因与基因组百科全书(KEGG)和GSEA的分析显示,EI^(2)4主要通过肿瘤蛋白p53(TP53)调控细胞周期基因和细胞死亡基因的转录、间质-上皮转化因子(MET)激活蛋白酪氨酸激酶2(PTK2)信号、MET信号等途径在THCA中发挥作用。体外实验证实,EI^(2)4高表达促进THCA细胞系的增殖、侵袭和迁移能力。结论EI^(2)4在THCA组织中高表达,与THCA预后不良有关,EI^(2)4的过表达能促进THCA细胞的增殖、侵袭和迁移,提示EI^(2)4可作为THCA预后和诊断的生物标记物以及治疗潜在靶标。 展开更多

TNFRSF12A与甲状腺癌免疫细胞浸润相关性分析

目的:利用生物信息学分析,系统研究甲状腺癌患者的免疫细胞浸润情况,探索甲状腺癌关键免疫相关基因及潜在分子机制。方法:获取TCGA数据库中甲状腺癌(THCA)患者的表达谱,并从IMMPORT数据库下载免疫学相关基因列表。通过CIBERSORT工具分析... 目的:利用生物信息学分析,系统研究甲状腺癌患者的免疫细胞浸润情况,探索甲状腺癌关键免疫相关基因及潜在分子机制。方法:获取TCGA数据库中甲状腺癌(THCA)患者的表达谱,并从IMMPORT数据库下载免疫学相关基因列表。通过CIBERSORT工具分析THCA患者与正常对照组免疫细胞浸润水平的差异,利用R软件鉴定THCA中差异表达基因(DEGs),使用GEPIA2工具选出对甲状腺癌患者生存影响较大的前500个基因,与影响生存的免疫相关基因交叉,筛选影响生存的免疫相关基因。使用STRING数据库分析差异表达基因,然后通过Cytoscape软件构建PPI网络,鉴定出枢纽基因。借助cBioPortal数据库分析56种癌症中THFRSF12A(肿瘤坏死因子受体超家族成员12A)的表达,同时分析TNFRSF12A在THCA中的遗传改变,使用TIMER数据库评估TNFRSF12A表达与6种免疫细胞浸润的相关性。利用WebGestalt数据库进行功能富集分析,构建Cox比例风险模型,比较TNFRSF12A高表达组和低表达组的生存时间。结果:与正常组织相比,THCA中M0巨噬细胞(p<0.001)、M2巨噬细胞(p<0.001)、静息树突状细胞(p<0.001)等的比例显著增加,较高比例的静止树突状细胞(p=0.028)、M0巨噬细胞(p=0.018)和单核细胞(p=0.033)总生存率较差。M1巨噬细胞(p=0.024)、浆细胞(p=0.02)和CD8+T细胞(p=0.028)比例低与较差的总生存率相关。与预后相关的差异表达免疫基因包括BMP2、SEMA6B、INHBB和TNFRSF12A。PPI网络模块中,TNFRSF12A基因处于核心位置,与其他基因联系最紧密。THCA与正常组织中免疫细胞的浸润水平存在显著差异。与在正常组织中的表达相比,TNFRSF12A基因在THCA中高表达,且TNFRSF12A突变发生在5%的THCA样本中,低信使RNA(mRNA)表达是最常见的突变类型。TNFRSF12A的表达与B细胞、CD8+T细胞、CD4+T细胞、树突状细胞及中性粒细胞的浸润水平显著相关。基因富集分析结果显示,TNFRSF12A可能参与细胞因子-细胞因子受体相互作用的关键生物学过程和途径。TNFRSF12A低表达的患者通常比TNFRSF12A高表达的患者总生存时间短。结论:TNFRSF12A与甲状腺癌的免疫细胞浸润显著相关。 展开更多

目的建立尿液中四氢大麻酚(THC)、四氢大麻酸(THCA)的气相色谱串联质谱测定方法。方法尿样加入四氢大麻酸-D3(Δ9-THCA-D3)为内标,100℃水浴中氢氧化钾水解20 min、调pH,正庚烷振荡提取10 min,上层有机相吹干后经双(三甲基硅基)三氟乙酰... 目的建立尿液中四氢大麻酚(THC)、四氢大麻酸(THCA)的气相色谱串联质谱测定方法。方法尿样加入四氢大麻酸-D3(Δ9-THCA-D3)为内标,100℃水浴中氢氧化钾水解20 min、调pH,正庚烷振荡提取10 min,上层有机相吹干后经双(三甲基硅基)三氟乙酰胺(BSTFA)硅烷化70℃加热40 min衍生,利用气相色谱-串联质谱多反应监测(MRM)模式测定,内标法定量。结果本法平均加标回收率为90%~110%,相对标准偏差(RSD)为3.4%~6.3%,在0.25μg/ml^5μg/ml范围内具有良好的线性关系,线性相关系数(r)为:THC:0.9992,THCA:0.9998,最低检出浓度:THC:0.01μg/ml,THCA:0.05μg/ml。结论本法灵敏度高、准确性好,具有可靠的精密度与准确度,可用于尿样中四氢大麻酚、四氢大麻酸的同时测定。 展开更多

The in vitro study of tetracannabinolic acid(THCA)derivatives ALAM027 and ALAM108 was carried out on the following human tumor cells:T47D(breast,ductal carcinoma),PC-3(prostate,adenocarcinoma),HT 29(colorectal carcinoma),Caco-2(colon,adenocarcinoma),A549(lung,carcinoma),U87MG(human glioblastoma)and U266B1(multiple myeloma).The in vitro effects of THCA derivatives ALAM027 and ALAM108 on cell growth inhibition and IC50 values were measured using the CellTiter Glo assay.The ALAM027 compound showed good growth inhibition in all cell lines tested with the exception of U87MG cells.The ALAM108 compound also suppressed the growth of U87 MG cells but had little effect on T47D tumor cells.In vitro studies of THCA derivatives ALAM027 and ALAM108 showed antitumor activity in all cell lines tested.The difference in the activity of these compounds in relation to the T47D and U87MG tumor cells may be indicative of different functional mechanisms. 展开更多

COL4A2 enhances thyroid cancer cell proliferation through the AKT pathway

Objectives:Thyroid cancer(THCA)is the most common malignant tumor in endocrine system and the incidence has been increasing worldwide.And the number of patients dying from THCA has also gradually risen because the incidence continues to increase,so the mechanisms related to effective targets is necessary to improve the survival.This study was to preliminarily investigate the effects of the COL4A2 gene on the regulation of thyroid cancer(THCA)cell proliferation and the associated pathways.Methods:Bioinformatics analysis revealed that COL4A2 was closely associated with cancer development.COL4A2 expression in THCA tissues was analyzed using immunohistochemistry,and survival information was determined via Kaplan-Meier curves.The expression of COL4A2 and AKT pathway-related genes were analyzed using qPCR and western blot analyses.Colony formation as well as CCK-8 assays exhibited the cell proliferation level and cell activity,respectively.Downstream of COL4A2 was identified by Gene set enrichment analysis(GSEA).The effects of the COL4A2 and AKT pathways on THCA tumor growth in vivo were determined using a mouse model.Results:Bioinformatics analysis exhibited that COL4A2 plays a significant role in cancer and that the AKT pathway is downstream of COL4A2.THCA patients with high COL4A2 expression had shorter recurrence-free survival.Upregulation of COL4A2 gene expression in 2 THCA cell lines promoted tumor cell growth and activity.The use of AKT pathway blockers also restrained the growth and activity of the 2 THCA cell lines.The use of AKT pathway blockers reduced tumor volume and mass and prolonged mouse survival.Conclusions:COL4A2 can promote the growth as well as development of THCA through the AKT pathway and COL4A2 could be used as a target for THCA. 展开更多

Background:This study investigated the multifaceted role of BRCA2(breast cancer 2)in various cancer types,with a specific focus on thyroid carcinoma(THCA).Methods:Data sets were obtained from the University of California Santa Cruz database to analyze BRCA2 expression,genetic alterations,and clinical implications.Sample filtering criteria were applied,and immunohistochemistry from the Human Protein Atlas was used to validate protein expression.Correlation analyses were used to explore associations between immune-related genes,and immunological signatures were assessed using various tools.Genetic alterations in BRCA2 were analyzed using cBioPortal,and prognostic analysis involved evaluating gene expression differences at different clinical stages of THCA.Results:In patients with THCA,differences in BRCA2 expression were observed at both the mRNA and protein levels when comparing tumor and normal tissues.Correlation studies revealed associations between BRCA2 and immune-related genes,emphasizing its potential role in modulating the tumor microenvironment.Immunological signature analyses indicated distinct frequencies of tumor-infiltrating immune cell subsets in BRCA2 high versus low tumors.Moreover,genetic alterations in BRCA2,particularly the A2738S mutation in exon 18,have been identified in patients with THCA.The prognostic analysis demonstrated a significant correlation between altered BRCA2 levels and improved overall survival in patients with THCA.Additionally,BRCA2 expression was associated with prognostic factors such as stage and N.Conclusions:This study provides a holistic exploration of BRCA2 in cancer and highlights its diverse roles in expression,immune modulation,genetic alterations,and clinical prognosis.These findings underscore the potential significance of BRCA2 as a diagnostic and prognostic marker and offer valuable insights for future research and potential clinical applications in cancer management. 展开更多

Aim:To investigate the in vitro structure-activity relationship(SAR)of a range of tetrahydrocannabinolic(THCA)and cannabidiolic(CBDA)deriv­atives using the PANC-1 tumor cell line(pancreas,ductal carcinoma).Ma­terials and methods:The in vitro effects of a range of THCA and CBDA derivatives with different carbonyl group substituents were tested on the PANC-1 cells cell line using the CellTiter Glo Viability Assay(72 hours)and the XTT assay(48 hours).Results:A study of a series of THCA and CBDA derivatives containing different functional groups at the carbonyl nitrogen atom demonstrated that THCA amides have better inhibitory activ­ity,on the PANC-1 tumor cell line,than CBDA derivatives.Conclusions:THCA derivatives have better inhibitory activity than CBDA analogs with the same substituents.It is noteworthy that even a slight change in the structure of the substituent of the amide or hydrazone moiety of the mole­cule has a dramatic effect on the activity of these compounds. 展开更多
