Jadomycin is an antibiotic that has shown activities against bacteria, yeasts and fungi as well as cytotoxic properties to cancer cells. Because of the wide range of its inhibitory actions, jadomycin shows promise as a novel antibiotic and cancer treatment drug. Streptomyces venezuelae are aerobic bacteria that produce jadomycin and the size of bacterial population can significantly affect the yield of jadomycin. Therefore, the bacterial population must be accurately measured in order to standardize the reproducibility of jadomycin production process. In this study, a dehydrogenase activity measurement test, using triphenyl tetrazolium chloride (TTC), was used to measure the dehydrogenase activity of Streptomyces venezuelae during growth in maltose-yeast extract-malt extract (MYM) broth. The aims were to evaluate the effectiveness of the test for measuring microbial growth and to study the effects of the test conditions (incubation time, incubation temperature and medium pH) on triphenyl formazan (TF) yield. The results showed that the TF yield was highly correlated to the optical density. The highest TF yield was observed at a pH of 6 at all incubation times and temperature. Lower TF yields were obtained at higher temperature (40 and 50oC) compared to those obtained at lower temperatures (22 and 30oC). The difference between the yields obtained at 22oC and 30oC were not significant. The differences between incubation time were also not significant. The recommended test conditions are an incubation time of 1 hour at a temperature of 30oC and a pH of 6 followed by three extractions using methanol. 展开更多

The corrosion inhibition of cold rolled steel(CRS) in 7.0 mol·L^(-1) H_2 SO_4 solution by red tetrazolium(RTZ) was carefully investigated using both experimental procedures and theoretical techniques. The results show that RTZ acts as an effective inhibitor for the corrosion of CRS in 7.0 mol·L^(-1) H_2SO_4, and the maximum inhibition efficiency is higher than 95% with a RTZ concentration of 2.0 mmol·L^(-1). The adsorption of RTZ on CRS surface follows Langmuir isotherm. RTZ effectively retards both cathodic and anodic reactions, and acts as a mixed-type inhibitor. EIS exhibits two capacitive loops, and their resistances increase drastically in the presence of RTZ. SEM and AFM confirm that the addition of RTZ could significantly retard the corrosion of CRS surface. A series of characterizations like FTIR, RS, XRD and XPS reveal that the corrosion CRS surface is composed of the corrosion products of iron sulfates, iron oxides and iron hydroxide, as well as inhibitor. Theoretical results of quantum chemical calculation and molecular dynamics(MD) indicate that the adsorption center of RTZ+(organic cationic part of RTZ) mainly relies on its tetrazole ring,and the adsorption of RTZ+on Fe(001) surface is in a nearly flat orientation mode. 展开更多

4-Hydroxyphenylpyruvate dioxygenase(EC 1.13.11.27,HPPD)is currently one of the popular targets of herbicide research,and herbicides targeting it have shown promising results in treating resistant weeds.However,the lon... 4-Hydroxyphenylpyruvate dioxygenase(EC 1.13.11.27,HPPD)is currently one of the popular targets of herbicide research,and herbicides targeting it have shown promising results in treating resistant weeds.However,the longterm application of HPPD inhibitors with in their classical scaffolds inevitably leads to the development of drug resistance.To further delay the underlying development of weed resistance to HPPD inhibitors,we successfully screened HPPD inhibitor pharmacophore with novel chelate structures using computer-aided drug molecular design and obtained 2-trifluoromethyltetrazolium nicotinamide derivatives through active fragment splicing.Among these derivatives,6-(3,3-diethyl-1-methylureido)-N-(1-methyl-1H-tetrazol-5-yl)-2-(trifluoromethyl)nicotinamide(compound 31),exhibited the most promising in vitro enzyme inhibitory activity,with IC_(50) value of 0.072μM,which was approximately five times better than that of the control agent mesotrione(IC_(50)?0.363μM).The crystal structure of the AtHPPD complexed with compound 31(2.0Å)and the binding energy calculations of the representative compounds revealed several important interactions of the ligand binding to the target protein,which explained the superior enzyme inhibitory activity of the compounds at the molecular level.In addition,compounds 7 and 31 possessed 100%inhibition against the five target weeds at the tested dosage and both were more effective against Setaria viridis than mesotrione.In general,it is promising to design novel HPPD inhibitors by developing novel chelating structures,and compounds 7 or 31 could be used as the leads for further development of valuable HPPD inhibitors. 展开更多

A new complex has been synthesized by the reaction of CuBr and 1,4-bis(2-allyl-2 H-tetrazol-5-yl)benzene. The crystal structure of the resulting complex, [Cu3(1,4-bis(2-allyl-2 H-tetrazol-5-yl)benzene)Br3]n(1), has been determined by single-crystal X-ray diffraction analysis. Complex 1 is of monoclinic system, space group P21/c with a = 10.983(7), b = 6.294(4), c = 17.606(10) ?, β = 102.465(6)°, V = 1188.4(3) ?3 and Mr = 577.52. Structural analyses revealed complex 1 shows a three-dimensional(3 D) framework. The thermal stability and luminescent properties of the complex are also investigated. 展开更多

The structure consisted of anion part and a 2,3,5-(triphenyl) tetrazolium cation as counter ions. The copper complex has a distorted tetrahedral geometry, the CI(1)-Cu-CI(2) [124.33 (2°)] and Cl(3)-Cu-Cl(4) [131.03 (2°)] angles are significantly larger than the ideal tetrahedral. The purpose of this study was to verify the efficiency of methods of pre-conditioning and concentrations of tetrazolium solution in the evaluation of physiological quality of Stizolobium aterrimum seeds. Commercial seeds have passed through the process of accelerated aging for composing lots of 0, 24, 48, 96 and 144 hours, and were submitted to the pre-conditioning: 1) Scarification with sandpaper number 100 and immersion in water at room temperature for 18 hours, 2) Immersion in water at 95°C and maintained in the same water outside the heating at 25°C for 18 hours. After preconditioning, the seed coats were removed and immersed in embryos tetrazolium salt at 0.25%, 0.5% and 1% for 150 minutes and maintained in the dark at 25°C for color development. To compare the results obtained in the tetrazolium test, we performed germination test, first counting and germination speed index. The method of scarification with sandpaper and later soaked in water for 18 hours at 25°C, the most efficient way in preconditioning of seeds of Stizolobium aterrimum and concentration of 1.0% tetrazolium solution for 150 minutes at 25°C, allows evaluating the quality of seed lots from this species. 展开更多

The tetrazolium salt 2-(4-Iodophenyl) -3-( 4-nitrophenyl ) -5-phenyltetrazolium chloride (INT) was used as a tool fi)r estimating the activity of the electron transport system (ETS) in activated sludge in a 40 L sequencing batch reactor (SBR) and domestie sewage as the organic substrate. The activity of INT-ETS during one SBR cycle, and the effeet of the ammonia concentration and the concentration of organic matter influent on the INT-ETS activity were investigated. The results show that: the use of INT is reliable in estimating of biological activity of activated sludge of SBR system; Biological activity of organic matter biodegradation, nitrification and denitrification process in SBR system reduce orderly. Obviously, INT-ETS activity reduces from 232.59 rny/(g · h) to 190. 65 rag/( g ·h) at first and then decreases to 113.88 my/( g · h) when influent concentration of COD and NH4+-N is 300 my/L and 40 mg/L respectively. In addition, various influent Nitrogen (NH4+-N are 14.5 mg/L and 42.0 my/L) and organic shock loading (COD are 293 mg/L and 685 my/L) experimenntions cure prove that operational conditions have no obvious effect on INT-ETS variation rule. 【目的】旨在筛选波罗蜜(Artocarpus heterophyllus Lam.)花粉离体萌发的最佳培养基配方,并评估快速染色法检测花粉生活力的适用性。【方法】以‘四季波罗蜜’花粉为材料,通过单因素试验和正交试验测定花粉萌发率,优化花粉培养基成分;... 【目的】旨在筛选波罗蜜(Artocarpus heterophyllus Lam.)花粉离体萌发的最佳培养基配方,并评估快速染色法检测花粉生活力的适用性。【方法】以‘四季波罗蜜’花粉为材料,通过单因素试验和正交试验测定花粉萌发率,优化花粉培养基成分;比较不同染色法测定波罗蜜花粉生活力的准确性。【结果】蔗糖、H_(3)BO_(3)、CaCl_(2)和MgSO_(4)对波罗蜜花粉萌发率影响显著,其主次顺序为CaCl_(2)>蔗糖>H_(3)BO_(3)>MgSO_(4)。最佳培养基配方为5%蔗糖+150 mg/L H_(3)BO_(3)+100 mg/L CaCl_(2)+150 mg/L MgSO_(4),萌发率达72.39%,显著高于其他处理;27℃为最适萌发温度;二苯基四氮唑溴盐染色法(MTT法)(10 min染色,活力62.99%)与氯化三苯四氮唑染色法(TTC法)(45 min染色,活力78.58%)均可用于花粉生活力快速检测,但MTT法效率更高且对比度更佳,而醋酸洋红染色法和碘-碘化钾法(I2-KI法)不适用。【结论】研究结果为波罗蜜杂交育种中花粉生活力检测及人工授粉技术提供了理论依据与实践支持。 展开更多

以大肠杆菌作为研究对象,建立一种5-cyano-2,3-ditolyl tetrazolium chloride(CTC)染色结合流式细胞仪(CTC-FCM)的方法,以选择性检测水环境中具有代谢活性的细菌总数.该方法的原理是细菌与具有氧化还原性的染料CTC发生反应,形成红色荧... 以大肠杆菌作为研究对象,建立一种5-cyano-2,3-ditolyl tetrazolium chloride(CTC)染色结合流式细胞仪(CTC-FCM)的方法,以选择性检测水环境中具有代谢活性的细菌总数.该方法的原理是细菌与具有氧化还原性的染料CTC发生反应,形成红色荧光物质,被流式细胞仪特异性识别进而可选择性检测活性菌.研究结果表明,CTC染色的最佳反应条件为:CTC浓度为2mmol·L-1、37℃避光孵育3h.该方法最低检测限为103个·mL-1.通过比较培养法和CTC-FCM方法检测热灭活后的大肠杆菌,结果表明CTC-FCM方法可准确区分活性菌和灭活菌,且与培养法之间具有较好的线性关系(R2=0.9465).应用CTC-FCM方法检测实际样品,结果显示该方法与培养法之间有较好的线性关系(R2=0.8121).本研究建立的CTC-FCM方法可满足饮用水水质标准需求,且检测时间比平板培养法缩短20~40h,可以用于环境水样中活性细菌总数检测. 展开更多

目的:比较不同来源生脉注射液体外细胞毒性的差别,寻找造成差别的主要原因。方法:将不同来源生脉注射液原液以及不同浓度的稀释液分别与L929细胞接触培养,通过倒置相差显微镜观察其形态,采用四甲基偶氮唑盐比色法(MTT法)量化细胞毒性,... 目的:比较不同来源生脉注射液体外细胞毒性的差别,寻找造成差别的主要原因。方法:将不同来源生脉注射液原液以及不同浓度的稀释液分别与L929细胞接触培养,通过倒置相差显微镜观察其形态,采用四甲基偶氮唑盐比色法(MTT法)量化细胞毒性,计算相对增殖率,并进行细胞毒性评价。结果:8个厂家生产的生脉注射液原液的细胞毒性分级均为4级,为重度毒性。样品A的EC50最高,说明其细胞毒性较其他样品高。在生脉注射液所添加的辅料中,当聚氧乙烯脱水山梨醇单油酸酯(吐温80)浓度为处方量的0.5%时,其细胞毒性分级为3~4级。将吐温80稀释至0.062 5%时,其细胞毒性分级为1级,基本无细胞毒性。8个厂家生产的生脉注射液原液中吐温80的含量不一致,有些厂家的含量大于0.5%。结论:不同厂家生产的生脉注射液由于原料产地、生产条件不同等原因,其细胞毒性存在较大的差别。吐温80作为中药注射剂的辅料,在高于0.062 5%的浓度下存在细胞毒性。生脉注射液中吐温80的含量与细胞毒性可能呈一定的正相关性。 展开更多

目的和方法:选用正常大鼠肺泡巨噬细胞(AM),在高糖及高糖+高胰岛素环境下培养,用卡介苗(BCG)、干扰素a-2b(IFNα-2b)或两者联合活化,检测其贴壁率、四唑氮兰(NBT)还原功能、NO和TNF-α释放量并观... 目的和方法:选用正常大鼠肺泡巨噬细胞(AM),在高糖及高糖+高胰岛素环境下培养,用卡介苗(BCG)、干扰素a-2b(IFNα-2b)或两者联合活化,检测其贴壁率、四唑氮兰(NBT)还原功能、NO和TNF-α释放量并观察其超微结构改变。结果:高糖及高糖+高胰岛素环境下活化AM贴壁延迟(P<0.01),NBT还原功能明显被抑制(P<0.01);BCG和IFNα-2b+BCG活化AM时,其NO和TNFα释放量均明显低于对照组(P<0.01);IFNα-2b活化AM时,其NO和TNF-α释放量与对照组无明显差别(P>0.05);细胞表面伪足减少、变短,胞质内高尔基氏体、粗面内质网减少。结论:在短期内高糖及高糖+高胰岛素环境均抑制AM吞噬功能及改变超微结构,从而使糖尿病个体易发生肺部感染。 展开更多
