基于TAS2R38探究黄连“苦寒败胃”的生物基础 被引量:4

目的基于苦味受体38(bitter taste receptor,TAS2R38)探讨久服、误服黄连导致“苦寒败胃”的生物基础。方法ICR小鼠随机分为5组,分别为空白对照组、黄连低(0.5 g/kg)/中(1.5 g/kg)/高(4.5 g/kg)剂量组和苯甲地那铵组(0.225 g/kg),每组1... 目的基于苦味受体38(bitter taste receptor,TAS2R38)探讨久服、误服黄连导致“苦寒败胃”的生物基础。方法ICR小鼠随机分为5组,分别为空白对照组、黄连低(0.5 g/kg)/中(1.5 g/kg)/高(4.5 g/kg)剂量组和苯甲地那铵组(0.225 g/kg),每组12只。各组小鼠分别灌胃给予相应的药物,持续8周。于第2、4、8周处理动物,RT-qPCR检测小鼠胃肠TAS2R38 mRNA表达水平;观察小鼠胃排空、肠推进;ELISA测定小鼠胃泌素和胆囊收缩素;HE染色评估胃肠黏膜损伤程度。结果与空白对照组相比:苯甲地那铵和黄连均显著增加TAS2R38 mRNA表达水平,其中黄连组呈剂量依赖性(P<0.01);苯甲地那铵延缓胃排空(P<0.05),黄连在第2周时促进胃排空(P<0.01),但在第4、8周则延缓胃排空,特别是中、高剂量组(P<0.05);苯甲地那铵与黄连均呈时间依赖性地抑制肠蠕动,尤以黄连高剂量组表现明显(P<0.01);苯甲地那铵与黄连均抑制胃泌素的分泌、促进胆囊收缩素的分泌(P<0.05,P<0.01);HE染色显示苯甲地那铵和黄连均可损伤胃肠组织。结论长期使用黄连可通过激动TAS2R38,影响小鼠胃肠动力,调节胃肠激素分泌,加重胃肠组织损伤,可能是其“苦寒败胃”的生物基础之一。 展开更多

探究襄阳地区大学生TAS2R38基因型频率及其与食物偏好的关联,更好地了解影响大学生饮食消费结构的因素.以湖北文理学院2016级临床医学专业学生作为受试者,采用阈值法对160名大学生PTC尝味能力进行测试,以PTC尝味能力(分14个等级)评价其... 探究襄阳地区大学生TAS2R38基因型频率及其与食物偏好的关联,更好地了解影响大学生饮食消费结构的因素.以湖北文理学院2016级临床医学专业学生作为受试者,采用阈值法对160名大学生PTC尝味能力进行测试,以PTC尝味能力(分14个等级)评价其基因型.对饮食偏好(分为2个等级)采用自编问卷进行调查,运用Hardy-Weinberg定律计算基因频率检验群体遗传平衡性,同时采用交叉表卡方检验分析比对基因型与食物偏好的关联性,显著性检验设置为P=0.05.结果 表明:有效数据160份,得出显性基因T和隐性基因t频率分别为0.55和0.45.交叉表卡方检验分析TAS2R38基因型与茶、咖啡摄入和甘蓝性蔬菜、甜水果、酸水果、香菜和茴香苗的摄食均无关,差异不具有统计学意义(P值分别为0.963、0.571、0.929、0.353、0.106、0.067、0.305).对油性食物和肉类食物PTC味盲者比PTC品尝者存在更大的摄入量,差异具有统计学意义(P=0.028和P=0.03). 展开更多

The supertaster phenomenon is an alt... This study examines a common phenomenon that is greatly ignored by the clinical community for numerous reasons. Many people for a multitude of reasons experience taste alterations. The supertaster phenomenon is an alteration of taste that requires more investigation. In this study, a proband was examined for subjective reports of a taste alteration to determine its nature through a medical history examination and interview as well as any recollections of the taste disorder in her life. Through this examination, it was found that medical history examination and interview of the proband that many members of her nuclear family showed traits of the same taste disorder or in the case of one family member being a suspect for the taste alteration and one member not showing any signs because of genetic diversity as a half-sibling. Taste disorders are heritable, have multiple health and mental health consequences, influence life choices including mate choice, avoidance behaviors, social choices, alcohol use/abuse, smoking, food choices, and more. More awareness is needed in the research and clinical community into taste alterations as well as calls for future research from neuroscience, biomedical science, life science, and allied science community to investigate taste alterations. 展开更多
