目的旨在通过生物信息学分析结合实验验证,探讨PCBP1(Poly(rC)-Binding Protein 1)在胃癌组织中的表达特征及其临床意义,对其与铁死亡主要调控因子STUB1(STIP1 Homology and U-Box Containing Protein 1)的关系研究。并通过免疫组化实...目的旨在通过生物信息学分析结合实验验证,探讨PCBP1(Poly(rC)-Binding Protein 1)在胃癌组织中的表达特征及其临床意义,对其与铁死亡主要调控因子STUB1(STIP1 Homology and U-Box Containing Protein 1)的关系研究。并通过免疫组化实验验证PCBP1与STUB1在胃癌中的表达模式及其与临床病理特征的关系。为新型胃癌靶向治疗策略提供重要的理论支撑和潜在的干预靶点。方法从TIMER 2.0在线分析网站中获取PCBP1在胃癌及癌旁组织的基因表达数据。利用癌症基因组图谱(The Cancer Genome Atlas,TCGA)数据库中胃癌数据(STAD)进行KEGG通路富集分析,并揭示其潜在作用机制;在铁死亡调控通路中找出主要调控因子STUB1。随后,采用免疫组化法检测33例胃癌组织及对应癌旁组织中PCBP1和STUB1的表达情况。将所收集的病例依据不同的分化程度、年龄、性别、肿瘤浸润深度、TNM分期及病理学形态进行分组,观察二者的阳性表达率,采用χ^(2)检验分析两者之间及其与临床和病理特征间的相关性,进一步探讨PCBP1和STUB1与胃癌恶性程度的关系。结果免疫组化结果显示PCBP1在癌组织中的阳性表达率为69.7%,明显高于癌旁组织中的阳性表达率48.5%;STUB1在癌组织中的阳性表达率为39.4%,低于癌旁组织中的阳性表达率54.5%,两者差异均有统计学意义(P<0.05)。PCBP1的阳性表达率与肿瘤分化程度、淋巴结转移及Lauren分型有相关性(P<0.05);与患者的年龄、性别、浸润深度、临床分期、神经浸润、脉管侵犯无相关性(P>0.05)。STUB1的阳性表达率与肿瘤分化程度、浸润深度、淋巴结转移及Lauren分型有相关性(P<0.05),但与患者的年龄、性别、临床分期、神经浸润、脉管侵犯无相关性(P>0.05)。PCBP1(癌)与STUB1(癌)的Spearman相关系数为-0.413,P为0.017,表明两者之间存在显著的负相关性。结论PCBP1可通过调控铁死亡通路中主要调控因子STUB1参与胃癌的恶性进展。为胃癌的分子机制探索及潜在治疗靶点提供了新的理论依据。展开更多

The simulation and experimental results verify that the test circuit with a radial stub is better than that without. As an example, a C-band AlGaN/GaN HEMT microwave power device is tested with the designed circuit and fixture. With a 5.4GHz microwave input signal,the maximum gain is 8.75dB,and the maximum output power is 33.2dBm.展开更多

A sudden decrease in antenna loading accompanying the L-mode to H-mode transition typically occurs on a timescale of a few milliseconds, as does the increase in loading at the H- to L-mode transition. Therefore, it is necessary to match dynamically in the transmission line between the generator output and the antenna input connections[1]. A new type of stub tuner being developed utilizes the difference in radio-frequency wavelengths between gas and liquid due to different relative dielectric constants. The impedance matching can be adjusted in realtime in an attempt to track the variations in the antenna loading. Since there are no mechanically moving parts in the short ends of stub, the change can be more convenient and safe, moreover, it can withstand higher voltage without breakdown. This system device will be applied in the HT-7 superconductor Tokamak ICRF experiment.展开更多
To realize cross-wedge rolling of shaft parts without a stub bar in a short process,an axial closed-open-type cross-wedge rolling technique was proposed.Based on the strain characteristics in the rolling,evaluation in...To realize cross-wedge rolling of shaft parts without a stub bar in a short process,an axial closed-open-type cross-wedge rolling technique was proposed.Based on the strain characteristics in the rolling,evaluation indices of deformation uniformity were provided,and the DEFORM-3D software was adopted to conduct numerical simulations of the rolling process.The metal flow and strain distribution in all stages of the rolling process were analyzed.It is shown that the strain value of the rolled piece close to the end is relatively high while the overall strain distribution is uniform in the rolling process.When the percentage reduction in area is smaller,the fluctuation range of the equivalent strain will be lower and the overall uniformity of the rolled piece will be better.A variable angle wedge was implemented to make metal flow inward and eliminate concavity.Finally,rolling experiment was performed,which indicate that the shape of the rolled piece obtained is consistent with the simulation results.Concavity value in the rolling is decreased by 92%as compared to conventional open rolling.The research results lay a theoretical basis for realizing closed-open-type cross-wedge rolling without a stub bar.展开更多
This paper studies the contribution of CFRP(carbon fiber-reinforced polymer)to the mechanical behavior of high strength concrete-filled square steel tube(HCFST)under biaxial eccentric compression.The new type of compo...This paper studies the contribution of CFRP(carbon fiber-reinforced polymer)to the mechanical behavior of high strength concrete-filled square steel tube(HCFST)under biaxial eccentric compression.The new type of composite member is composed of an inner CFRP tube and an outer steel tube with concrete filled in the two tubes.The finite element analysis was made by ABAQUS on the behavior of high strength concrete filled square steel tubular columns with inner CFRP circular tube subjected to bi-axial eccentric loading.The results obtained from the finite element analysis were verified with the experimental results.In addition,the load-deflection curves in the whole process were calculated and analyzed,which can be divided into three segments:Elastic phase,plastic phase,descending phase.Based on the load-deflection curves,the stresses analysis on the core concrete,CFRP tube and steel tube were conducted.The confinement effect of the CFRP tube improves the ductility of HCFST-CFRP stub column.CFRP ratio and eccentricity affect the ultimate bearing capacity of HCFST stub column.Finally,a calculation formula of ultimate bearing capacity was proposed in the paper.展开更多
