Under field conditions, these stresses occur more frequently in combination than alone, which magnifies corresponding detrimental effects on the growth and productivity of agriculturally important crops. Plant responses to such abiotic stresses are quite complex and manifested in a range of developmental, molecular, and physiological modifications that lead either to stress sensitivity or tolerance/resistance. Maize (Zea mays L.) is known for its sensitivity to abiotic stresses, which often results in substantial loss in crop productivity. Bioaugmentation with plant growth-promoting rhizobacteria (PGPR) has the potential to mitigate the adverse effects of drought and heat stresses on plants. Hence, this is considered a promising and eco-friendly strategy to ensure sustainable and long-term maize production under adverse climatic conditions. These microorganisms possess various plant growth-promoting (PGP) characteristics that can induce drought and heat tolerance in maize plants by directly or indirectly influencing molecular, metabolic, and physiological stress responses of plants. This review aims to assess the current knowledge regarding the ability of PGPR to induce drought and heat stress tolerance in maize plants. Furthermore, the drought and heat stress-induced expression of drought and heat stress response genes for this crop is discussed with the mechanisms through which PGPR alter maize stress response gene expression.展开更多

Cd stress significantly inhibited root and shoot growth, and affected the global proteome in rice roots and leaves, which induced or upregulated the expression of corresponding proteins in rice roots and leaves when rice seedlings were exposed to 0.1 or 1.0 mmol/L Cd. The Cd-induced proteins are involved in chelation and compartmentation of Cd, elimination of active oxygen free radicals, detoxification of toxic substances, degradation of denatured proteins or inactivated enzymes, regulation of physiologic metabolism and induction of pathogenesis-related proteins. Comparing the Cd-induced proteins between the two vadeties, the β-glucosidase and pathogenesis-related protein family 10 proteins were more drastically induced by Cd stress in roots and leaves of Aizaizhan, and the UDP-glucose protein transglucosylase and translational elongation factor Tu were induced by 0.1 mmol/L Cd stress in roots of Shanyou 63. This may be one of the important mechanisms for higher tolerance to Cd stress in Shanyou 63 than in Aizaizhan.展开更多

Although Nudix hydrolases are involved in plant detoxification processes in response to abiotic and biotic stresses, the biological functions of Nudix hydrolases remain largely unclear in grapevine. In the present study, a total of 25 putative grapevine Nudix hydrolases(VvNUDXs) were identified by bioinformatics analysis and classified into eight subfamilies based to their preferred substrates. Both tandem and segmental duplications were responsible for the evolution and expansion of the NUDX gene family in grapevine. To investigate the regulatory roles of VvNUDX genes during growth and development, as well as in response to abiotic and biotic stresses in grapevine, the expression patterns were revealed in publicly available microarray data. The spatial and temporal expression patterns of the VvNUDX genes indicated that they might play important roles in multiple developmental processes. Transcriptome and qRT-PCR analyses showed that ten VvNUDX genes were specifically expressed in grapevine berries, suggesting potential roles in grapevine berry development. Expression and phylogenetic analyses demonstrated that VvNUDX1 and VvNUDX3 might be involved in terpenoid biosynthesis in grapevine. Furthermore, most VvNUDX genes active toward the ADP-ribose/NADH showed different patterns in response to various abiotic and biotic stresses, such as salinity and drought, as well as different types of biotic treatments, such as Erysiphe necator,Bois Noir phytoplasma and leaf-roll-associated virus-3(GLRaV-3). These results indicated that VvNUDX genes were associated with plant detoxification processes in response to abiotic and biotic stresses, and regulate the disease immunity and resistance pathways. The information obtained here may provide good opportunities to explore the physiological functions of VvNUDX genes in berry development and stress response networks in grapevine.展开更多

文摘Objective:To explore the relationship between anesthetic depth and surgical stress response in minimally invasive cardiothoracic surgery. Methods:A total of 89 patients who underwent thoracoscopic minimally invasive cardiothoracic surgery in our hospital from June 2024 to December 2024 were selected as the research objects. They were divided into the light anesthesia group(n=45)and the deep anesthesia group(n=44).The vital signs at different intraoperative nodes and perioperative stress status of the two groups were compared. Results:Before lesion resection and after surgery, the mean arterial pressure and heart rate of the deep anesthesia group were lower than those of the light anesthesia group, with statistically significant differences. Conclusion:In thoracoscopic minimally invasive cardiothoracic surgery, deep anesthesia can effectively control the patient’s surgical stress response, but the postoperative awakening time is longer;patients under light anesthesia have a shorter awakening time, but the intraoperative stress response is obvious.
文摘BACKGROUND Although the 2021 Chinese Clinical Practice Guidelines for Enhanced Recovery after Surgery(ERAS)provide recommendations for ERAS in gastrointestinal surgery, the clinical application of standard ERAS nursing models is challenging due to the variety of diseases involved in gastrointestinal surgery and the com-plex factors contributing to patient stress responses. Moreover, stress responses are more severe in older adult patients. Therefore, precision medicine is required to improve the quality of nursing care and promote postoperative recovery in gastrointestinal surgery.and demonstrate nursing benefits through clinical practice.METHODS This randomized clinical trial first established an evidence-based nursing ERAS protocol in older adult patients based on literature related to perioperative nursing measures for gastrointestinal surgery stress response.Next,392 older adult patients who underwent gastrointestinal surgery and were admitted to our hospital between December 2021 and June 2023 were categorized into two groups to receive evidence-based(study group)or conventional(control group)ERAS nursing models, respectively.Intraoperative physiological parameters during surgery and postoperative recovery indicators were compared between the groups.RESULTS Among 64 domestic and international studies, the stress responses of older adult patients mainly included emotional anxiety, sleep disorders, gastrointestinal discomfort, physical weakness, pain, and swelling. The appropriate nursing interventions included comprehensive psychological counseling,pre-and postoperative nutritional support, temperature control, pain management, and rehabilitation training.Compared with the control group, the study group showed lower heart rate, mean arterial pressure, blood glucose level, and adrenaline level;shorter duration of drainage tube placement, time to first flatus, time to first ambulation, and postoperative hospital stay;lower anxiety scores on postoperative day 3;and lower incidences of postoperative infection, obstruction, poor wound healing, and gastrointestinal reactions were lower in the study group(all P<0.05).CONCLUSION The evidence-based nursing measures targeting stress responses based on the conventional ERAS nursing model resulted in stable intraoperative physiological parameters during surgery, promoted postoperative recovery, and reduced the incidence of complications.

文摘In yeast, the stress-responsive protein Whi2 interacts with phosphatase Psr1 to form a complex that regulates cell growth, reproduction, infection, and the stress response. However, the roles of Whi2 and Psr1 in Fusarium graminearum remain unclear. In this study,we identified homologous genes of WHI2 and PSR1 in F.graminearum and evaluated their functions by constructing deletion mutants.By comparing the responses of the mutants to different stressors,we found that FgWHI2 and FgPSR1 were involved in responding to osmotic, cell wall and cell membrane stresses, while also affecting the sexual and asexual reproduction in F.graminearum.Our studies demonstrated that FgWHI2 and FgPSR1 regulate the biosynthesis of ergosterol and the transcriptional level of FgCYP51C,which is a CYP51 paralogues unique to Fusarium species. This study also found that the deoxynivalenol(DON)production of FgWHI2 and FgPSR1 deletion mutants was reduced by≥90%and DON production was positively correlated with the transcriptional levels of FgWHI2 and FgPSR1.In addition,we observed that FgWHI2 and FgPSR1 were involved in regulating the sensitivity of F.graminearum to chlorothalonil, fluazinam, azoxystrobin, phenamacril, and oligomycin. This study revealed cross-resistance between chlorothalonil and fluazinam.Meanwhile, chlorothalonil and fluazinam inhibited DON biosynthesis by altering the normal expression of FgWhi2 and FgPsr1.Interestingly,the subcellular localization of FgWhi2 and FgPsr1 was significantly altered after treatment with chlorothalonil and fluazinam, with increased co-localization.Collectively,these findings indicate that FgWHI2 and FgPSR1 play crucial roles in stress response mechanisms, reproductive processes, secondary metabolite synthesis, and fungicide sensitivity in F.graminearum.
文摘BACKGROUND Hepatocellular carcinoma(HCC)is a common and frequently encountered malignancy in clinical practice.Patients who lack understanding of the disease and surgical treatment are prone to fear, depression, and other negative emotions, which further aggravate psychological stress. As such, less stimulating and minimally invasive surgical modalities, such as ultrasound-guided interventions, should be adopted to alleviate or eliminate negative perioperative psychological states, which can be evaluated using validated tools such as the Hamilton Anxiety Scale(HAM-A)and Hamilton Depression Scale(HAM-D).AIM To investigate the impact of ultrasound-guided surgery on stress, complications, and recovery in patients with HCC and comorbid anxiety/depression.METHODS Ninety patients with primary small HCC and comorbid anxiety/depression were randomly divided into 2 groups according to treatment(n=45 each):Experimental(ultrasound-guided intervention);and control(conventional laparoscopic hepatectomy). The HAM-A and HAM-D were used to assess psychological states before and 1 week after surgery.C-reactive protein(CRP),vascular endothelial growth factor(VEGF),and superoxide dismutase(SOD)levels,in addition to specific liver-function indicators, complication rates, and postoperative metrics were measured.Recurrence rates were monitored for 6 months.RESULTS There were no significant differences in preoperative HAM-A and HAM-D scores between the 2 groups(P>0.05);however,scores in the study group were significantly lower postoperatively(P<0.05).On postoperative day 1,there were no significant differences in serum levels of CRP,VEGF,or SOD between the groups(P>0.05),whereas the levels in the intervention group were significantly lower than those in the control group on days 3 and 7(P<0.05).The incidence of postoperative complications in the study group(6.66%)was significantly lower than that in the control group[17.78%(P<0.05)].The study group also had a significantly shorter time to first flatus, oral intake, and postoperative hospital stay(P<0.05).Postoperative serum alanine aminotransferase, aspartate aminotransferase, and total bilirubin levels decreased in both groups compared with preoperative levels, although with significantly lower values in the study group(P<0.05)but no differences preoperatively(P>0.05).There was no statistical difference in tumor recurrence rates between the 2 groups during the six-month follow-up(P>0.05).CONCLUSION Ultrasound-guided intervention for patients with primary small HCC and anxiety/depression effectively improves negative emotional states, reduce stress responses, decreases postoperative complications, promotes recovery, and enhances quality of life.

文摘We examined the possible multiple defects induced by acute and prolonged exposure to high levels of manganese(Mn) solution by monitoring the endpoints of lifespan, development, reproduction, and stress response. Our data suggest that acute exposure(6 h) to Mn did not cause severe defects of life span, development, and reproduction, similarly,no significant defect could be found in animals exposed to a low concentration of Mn(2.5 μmol/L) for 48 h.In contrast, prolonged exposure(48 h) to high Mn concentrations(75 and 200 μmol/L) resulted in significant defects of life span, development, and reproduction,as well as the increase of the percentage of population with hsp-16.2::gfp expression indicating the obvious induction of stress responses in exposed animals. Moreover, prolonged exposure(48 h) to high concentrations(75 and 200 μmol/L) of Mn decreased the expression levels of antioxidant genes of sod-1,sod-2,sod-3,and sod-4 compared to control. Therefore, prolonged exposure to high concentrations of Mn will induce the severe defects of life span, development, and reproduction in nematodes possibly by affecting the stress response and expression of antioxidant genes in Caenorhabditis elegans.

文摘The molecular response of wheat(Triticum aestivum L.,cv.Yangmai 13) seedlings to heavy metal(Cd,Hg) and 1,2,4-trichlorobenzene(TCB) stresses were examined by two-dimensional gel electrophoresis, image analysis, and peptide mass fingerprinting. The results showed inhibitions of root and shoot growth by Cd,Hg,and TCB.These stresses led to water deficit and lipid phosphorylation in the seedling which also promoted protein phophorylation in the leaves.Hg stress inhibited protein synthesis while Cd and TCB stresses induced or up-regulated more proteins in the leaves. Most of these induced proteins played important roles in the biochemical reactions involved in tolerance of wheat to Cd and TCB stresses. The primary functions of Cd-and TCB-induced proteins included methionine metabolism,Rubisco modification, protein phosphorylation regulation, protein configuration protection,H+ transmembrane transportation and also the synthesis of ethylene, defense substances and cell wall compounds.
文摘BACKGROUND Thoracic surgery for radical resection of lung tumor requires deep anesthesia which can lead to an adverse inflammatory response, loss of hemodynamic stability, and decreased immune function.Herein,we evaluated the feasibility and benefits of ultrasound-guided paravertebral nerve block anesthesia,in combination with general anesthesia, for thoracic surgery for lung cancer. The block was performed by diffusion of anesthetic drugs along the paravertebral space to achieve unilateral multi-segment intercostal nerve and dorsal branch nerve block.AIM To evaluate the application of ultrasound-guided paravertebral nerve block anesthesia for lung cancer surgery to inform practice.METHODS The analysis was based on 140 patients who underwent thoracic surgery for lung cancer at our hospital between January 2018 and May 2020.Patients were randomly allocated to the peripheral+general anesthesia(observation)group(n=74)or to the general anesthesia(control)group(n=66).Patients in the observation group received ultrasound-guided paravertebral nerve block anesthesia combined with general anesthesia, with those in the control group receiving an epidural block combined with general anesthesia.Measured outcomes included the operative and anesthesia times,as well as the mean arterial pressure(MAP),heart rate(HR),and blood oxygen saturation(SpO;)measured before surgery,15 min after anesthesia(T1),after intubation,5 min after skin incision, and before extubation(T4).RESULTS The dose of intra-operative use of remifentanil and propofol and the postoperative use of sufentanil was lower in the observation group(1.48±0.43 mg,760.50±92.28 mg,and 72.50±16.62 mg,respectively)than control group(P<0.05).At the four time points of measurement(T1 through T4),MAP and HR values were higher in the observation than control group(MAP,90.20±9.15 mmHg,85.50±7.22 mmHg,88.59±8.15 mmHg,and 90.02±10.02 mmHg,respectively;and HR,72.39±8.22 beats/min,69.03±9.03 beats/min,70.12±8.11 beats/min,and 71.24±9.01 beats/min,respectively;P<0.05).There was no difference in SpO;between the two groups(P>0.05).Postoperative levels of epinephrine, norepinephrine, and dopamine used were significantly lower in the observation than control group(210.20±40.41 pg/mL,230.30±65.58 pg/mL,and 54.49±13.32 pg/mL,respectively;P<0.05).Similarly,the postoperative tumor necrosis factor-αand interleukin-6 levels were lower in the observation(2.43±0.44 pg/mL and 170.03±35.54 pg/mL,respectively)than control group(P<0.05).There was no significant difference in the incidence of adverse reactions between the two groups(P>0.05).CONCLUSION Ultrasound-guided paravertebral nerve block anesthesia improved the stress and hemodynamic response in patients undergoing thoracic surgery for lung cancer, with no increase in the rate of adverse events.
文摘Drought and heat are major environmental stresses that continually influence plant growth and development. Furthermore, the drought and heat stress-induced expression of drought and heat stress response genes for this crop is discussed with the mechanisms through which PGPR alter maize stress response gene expression.

文摘A proteomic approach including two-dimensional electrophoresis and mass spectrometric (MALDI-TOF MS) analyses was used to investigate the responses to cadmium (Cd) stress in seedlings of rice (Oryza sativa L.) varieties Shanyou 63 and Aizaizhan. This may be one of the important mechanisms for higher tolerance to Cd stress in Shanyou 63 than in Aizaizhan.

文摘The homeodomain-leucine zipper(HD-Zip)family has been shown to perform amultitude of functions during plant development and stress responses;however, the familymembers and functions have not been identified in apple(Malus×domestica). In this study,83 HD-Zips(MdHDZs)were identified in the apple genome. They were assembled into four subgroups according to the classification in Arabidopsis, where MdHDZs in the same subgroup had similar gene structures and conserved protein motifs.Putative cis-element analysis of MdHDZs promoter regions uncovered numerous elements related to the response of stress and plant hormones. In addition, twelve transcripts of the MdHDZs showed different expression patterns under salt, drought, low temperature and ABA stresses by quantitative reverse transcription-PCR(qRT-PCR)assay.To further explore the function of MdHDZs in apple,MdHDZ3 was selected to verify its function under salt, low temperature and ABA stresses;and genetic transformation was used to obtain MdHDZ3 transgenic apple calli. The results demonstrated that MdHDZ3 increased sensitivity to salt, low temperature and abscisic acid in apple calli, suggesting that MdHDZ3 plays an important role in response to stresses.Subcellular localization and three-dimensional structural analysis revealed that MdHDZ3 was a nuclear-localized protein.Taken together, these findings provide potential information for further identification of HD-Zip proteins in apple.

文摘The Arabidopsis thaliana RPD3-type histone deacetylases have been known to form conserved SIN3-type histone deacetylase complexes, but whether they form other types of complexes is unknown. Here,we perform affinity purification followed by mass spectrometry and demonstrate that the Arabidopsis RPD3-type histone deacetylases HDA6 and HDA19 interact with several previously uncharacterized proteins, thereby forming three types of plant-specific histone deacetylase complexes, which we named SANT,ESANT,and ARID.RNA-seq indicates that the newly identified components function together with HDA6 and HDA19 and coregulate the expression of a number of genes.HDA6 and HDA19 were previously thought to repress gene transcription by histone deacetylation. We find that the histone deacetylase complexes can repress gene expression via both histone deacetylation-dependent and-independent mechanisms. In the mutants of histone deacetylase complexes, the expression of a number of stressinduced genes is up-regulated,and several mutants of the histone deacetylase complexes show severe retardation in growth.Considering that growth retardation is thought to be a trade-off for an increase in stress tolerance,we infer that the histone deacetylase complexes identified in this study prevent overexpression of stress-induced genes and thereby ensure normal growth of plants under nonstress conditions.

文摘As abscisic acid(ABA)receptor, the pyrabactin resistance 1-like(PYR/PYL)protein(named PYL for simplicity)plays an important part to unveil the signal transduction of ABA and its regulatory mechanisms.Glycyrrhiza uralensis,a drought-tolerant medicinal plant,is a good model for the mechanism analysis of ABA response and active compound biosynthesis. However, knowledge about PYL family in G.uralensis remains largely unknown.Here,10 PYLs were identified in G.uralensis genome.Characterization analysis indicated that PYLs in G.uralensis(Gu PYLs)are relatively conserved.Phylogenetic analysis showed that Gu PYL1-3 belongs to subfamily I,Gu PYL4-6 and Gu PYL10 belong to subfamily II and Gu PYL7-9 belongs to subfamily III.In addition, transcriptome data presented various expression levels of Gu PYLs under different exogenous ABA stresses. The expression pattern of Gu PYLs was verified by Quantitative real-time polymerase chain reaction(q RT-PCR).The study proved that Gu PYL4,Gu PYL5,Gu PYL8 and Gu PYL9 genes are significantly up-regulated by ABA stress and the response process is dynamic. This study paves the way for elucidating the regulation mechanism of ABA signal to secondary metabolites and improving the cultivation and quality of G.uralensis using agricultural strategies.
文摘BACKGROUND Radical surgery is a common treatment for patients with gastric cancer;however,it can lead to postoperative complications and intestinal barrier dysfunction.Ultrasound-guided quadratus lumborum block is often used for postoperative analgesia, but its effects on stress response and intestinal barrier function are not well understood.AIM To investigate the effects of an ultrasound-guided quadratus lumborum block on stress response and intestinal barrier function in patients undergoing radical surgery for gastric cancer.METHODS A total of 100 patients undergoing radical surgery for gastric cancer were randomly categorized into observation and control groups.Plasma adrenaline and cortisol levels, intestinal mucosal barrier indexes, and complication rates were compared between the two groups before, during, and 1 day after surgery.RESULTS The observation group had significantly lower plasma adrenaline and cortisol levels during surgery and at 1 day postoperatively than that of the control group(P<0.05).Additionally,intestinal barrier indexes(endotoxin and D-dimer)at 1 day postoperatively were significantly lower in the observation group than in the control group(P<0.05).CONCLUSION Ultrasound-guided quadratus lumborum block could reduce stress response, protect intestinal barrier function, and decrease the incidence of complications in patients undergoing radical surgery for gastric cancer. This technique has the potential for clinical applications.

文摘Nudix hydrolases are widely distributed across all classes of organisms and provide the potential capacity to hydrolyze a wide range of organic pyrophosphates. The information obtained here may provide good opportunities to explore the physiological functions of VvNUDX genes in berry development and stress response networks in grapevine.
