Optimized culture conditions for gonocytes provide an opportunity for their study and in vitro manipulation for potential application in reproductive technologies. Using six experiments in a step-wise design, we examined the effects of several culture conditions on the maintenance, proliferation, and colony formation of porcine gonocytes. Testis cells from neonatal piglets were cultured for 7 d in DMEM supplemented with 10% fetal bovine serum. The examined culture conditions included using different cell seeding densities, gonocyte proportions, incubation temperatures, sampling strategies, and medium changing regimens. Results: Confluency of cel s was optimal(>90% by ~6 d) when 3.0 × 104 testis cel s/cm2 containing ~40% gonocytes were used. Incubating the cel s at 35 °C or 37 °C resulted in similar cel number and viability at confluency, but incubation at 35 °C resulted in a delayed confluency. In the first 2 d of culture, gonocytes remained mostly floating in the medium and gradual y settled over the next 5 d. Consequently, not changing the medium for 7 d(as opposed to changing it every 2 d) led to a significant increase in the number of gonocyte colonies by reducing the loss of "floating gonocytes".Conclusion: We found that gonocytes require the presence of a critical minimum number of somatic cel s for settlement, and can proliferate and form growing colonies even in a basic medium. Large numbers of viable gonocytes remain floating in the medium for several days. The optimized culture conditions in the present study included seeding with 3.0 × 104 testis cel s/cm2 containing ~40% gonocytes, incubating at 37 °C, and without changing the medium in the first week, which can result in improved colony formation of porcine gonocytes.展开更多

We have established a multi-generation cultivation system for zooplankton ruth which future experiments on the biological effects of pollutants in marine and estuary environments can be better performed. A population of calanoid copepod, Sehmakeria poplesia, was collected in December 2003 and maintained in a static system through all stages (eggs to adults). The population ex- hinted an average developmental time of 13.6 d in conditions corresponding to the natural environment (water temperature 20~C salinity 15). A series of experiments were performed to examine copepod egg production and hatching success as functions of food type and feeding concentration. Results in our study showed that Isoehrysis galbana was more favored for the reproduction of cope- pods than Phaeodactylum tricornutum, and 10xl0%ellsmL-1 was the most practical algae concentration. We have demonstrated that the Schmakeria poplesia population can be maintained in the laboratory through multiple generations. In addition, methods to control egg production through changes in food concentration have been established, making it feasible to control the start date of exposure experiments or the timing of the collection of oftspring to initiate a new generation.展开更多

文摘[Objective]The aim was to study the effective factors on culturing Prunus salicina cv.Zaoshi embryos in vitro.[Method]Different age, culture medium and GA3 which affected culturing of prumssalicina embryos were discussed.[Result]The result showed that on the part of 26-41 d,after the blooming period, the embryos remained tiny and retained endosperms and showed no signs of change after having cultured for three generations. On the part of 48 d,after the blooming period, the endosperms had disappeared, the embryos kept growing until they filled the seed cavity;when they were planted on the MS culture medium, their survival rate reached 77%,in its first generation, the response of embryos was discernible. On the part of 65 d,after the blooming period,4.5% of their embryos grew into shoots on the MS culture medium;with the age of embryos growing, the survival rate of shoots increased until it reached 26% when the fruits went into ripeness;the embryos produced calli in their first generation of culturing. On the part of 65-83 d,after the blooming period, the embryos produced calli through more than 2-3 generations. On the part of 88 d,after the blooming period, the survival rate of shoots on the WPM culturing base doubled compared with that on the MS culturing base;on the same culture medium, the embryos were inhibited from growing into shoots when BA,KT or 2,4-D was added on to the culture medium. The survival rate of shoots was increased remarkably when the seeds were treated in 1 000 mg/L GA3.[Conclusion] This study provided experimental basis for the establishment of Prunus salicina cv.Zaoshi, embryos rescue techniques and cross breading.

文摘Background: Gonocytes give rise to spermatogonial stem cells, and thereby play an essential role in establishing spermatogenesis. The optimized culture conditions in the present study included seeding with 3.0 × 104 testis cel s/cm2 containing ~40% gonocytes, incubating at 37 °C, and without changing the medium in the first week, which can result in improved colony formation of porcine gonocytes.

文摘Human mesenchymal stem cells, such as human adipose-derived stem cells(hASCs),are typically cultured on a two-dimensional(2 D)monolayer material surface,on which 2 D culturing methods are easily performed and time-saving.However,hASCs usually suffer from decreased pluripotency and differentiation ability when cultured with a 2 D monolayer culturing method compared to hASCs cultured with a three-dimensional(3 D)culturing method, such as suspension culture. In this study,we evaluated whether the pluripotency and differentiation ability of hASCs can be reversibly changed during sequential cultivation with 2 D and 3 D culturing processes. The hASCs cultivated with a 3 D culturing process after 2 D culture showed at least 2-fold enhanced pluripotency(Sox2,Nanog,and OCT4)compared with that of hASCs cultured with the 2 D culture process alone.Furthermore,hASCs obtained from the 3 D culture process expressed increased levels of differentiation markers of chondrocytes and osteoblasts compared with hASCs obtained from the 2 D culture process when hASCs were induced to differentiate. However, their pluripotency and differentiation ability were extensively reduced when hASCs were shifted from 3 D culture to 2 D culture and vice versa, which indicates that hASCs show reversibility in terms of their pluripotency and differentiation ability depending on their environment in 2 D and 3 D culture. The reversibility of pluripotency and differentiation ability were found to last for at least 5 passages in culture during the alternative and sequential culture of cells with 2 D and 3 D culturing processes. Our study revealed the importance of the culture microenvironment in maintaining the pluripotency and differentiation ability of hASCs,which may reduce the effects of the aging process in hASCs.We discuss whether the environment of stem cell culture(i.e.,2 D or 3 D cultivation)can affect stem cell fate in terms of pluripotency and differentiation reversibility.

文摘Crustacean zooplankton form the keystone link between primary producers and fish stocks in marine and estuary, ecosystems. In addition, methods to control egg production through changes in food concentration have been established, making it feasible to control the start date of exposure experiments or the timing of the collection of oftspring to initiate a new generation.
